本发明涉及用于生产重组胶原样蛋白(collagen-like protein)(CLP)的新型方法。更具体地,本发明涉及迄今为止从未被观察到的在不存在V结构域的情况下形成三螺旋结构的Scl2蛋白序列的体外折叠,和涉及对于将Scl2蛋白适当地折叠成三螺旋结构而言至关重要的实际条件。
背景技术
细菌来源的胶原样蛋白(CLP)(最工业相关的是酿脓链球菌(Streptococcuspyogenes)的产物)具有相当令人感兴趣的机械特性,其类似于高等真核生物的胶原蛋白的机械特性,而不需要真核生物对应物所需的复杂成熟步骤。CLP呈现以下共同结构:彼此稳定化的两个α螺旋构成“V结构域”,其后是杆状结构性胶原结构域(CL)。在胶原结构域之后,在该蛋白的C末端通常存在膜锚(GPI样)。
如在各种出版物(Lukomski等人2002、Brodsky等人2009)中所描述的,对这种过程的当前理解是,需要V结构域以用于在体外将三个Scl2蛋白单体折叠成一个三螺旋结构(Lukomski等人揭示在没有V结构域的情况下的体内折叠)。
在若干个系统中已经尝试了胶原样蛋白的表达,这些系统包括大肠杆菌(Escherichia coli)和酿酒酵母(Saccharomyces cerevisiae)。本发明重点关注酿脓链球菌(Streptococcus pyogenes)CLP在不同宿主中的表达优化和纯化。
对于在大肠杆菌中的表达,用于此种生产所选择的构建体携带特定且必要的修饰,以便于高效地去除潜在免疫原性的V结构域:此种修饰由通常插入在V结构域和胶原序列之间的蛋白酶切割位点组成。由于这种修饰,由细菌宿主产生的蛋白必须从细胞内部分中提取,并且用特定的蛋白酶处理以去除该V结构域。必须针对切割的V结构域、全部细胞内蛋白内容物和所添加的用于处理未成熟的CLP的蛋白酶来纯化仅由胶原样结构域组成的成熟蛋白。此种工作流程极大地阻碍整个过程的成本效益,原因在于1)必须将所选择的产物从表达宿主细胞的全部内容物中分离出,和2)蛋白酶通常是昂贵的酶。
因此,本发明的目的是提供用于CLP的生产和纯化的改善的方法,该改善的方法是具有成本效益的并且是适用的,而不需要添加用于切割所述结构域的特定蛋白酶。
本发明提供解决方案以通过使用工业主力例如酵母巴斯德毕赤酵母(Pichiapastoris)来实现更具成本效益的方法。巴斯德毕赤酵母已被用作用于其它类别的胶原分子的宿主,通常是哺乳动物来源的胶原分子的宿主,如由Werten和同事最近所报道的(Biotechnology Advances 37,Issue 5,2019,Pages642-666);然而,使用巴斯德毕赤酵母以用于CLP生产还未被描述。此外,此种酵母的使用出人意料地提供了从成熟蛋白中切割V结构域的解决方案。
为了理解V结构域的存在是否会是分泌型表达为何如此低效的原因,使用最近发布的X-射线结构已经分析了此种结构域(J.Biol.Chem.289,5122-5133),通过使用公开可获得的工具即JPred(http://www.compbio.dundee.ac.uk/jpred)的手动评价来补充。已经可以鉴定V结构域的结构性决定因素:这种结构域先前已经在绝大多数科学出版物(Protein Science 2010,vol.19,pp775-785;J.Biol.Chem.,Vol.280,No.19,pp.19343-19349,以及许多其它文献)中被报道为对胶原折叠而言是必不可少的,尽管V结构域的存在对蛋白表达的影响曾经被讨论或提及。Yu等人分析了scl2细菌亚单位的不同片段(J.Biol.Chem.286,pp.18960–18968)在胶原稳定性的竞争中的作用,将观测限制于结构性区域,但也生成缺乏V结构域的片段。此处没有提及关于改善生产水平的报告:这并不出人意料,因为据认为,由于V结构域的截短而观测到的效果在分泌胶原时得以执行,这对于细菌胶原而言是目前为止未描述的过程。
按照这种假设,生成了一系列的截短:底层逻辑是维持V结构域的通常被报道为对胶原折叠是必不可少的一部分,减少它的序列以最小化对细胞内的生产机制(machinery)的任何干扰,并且因此能够在上清液中分泌显著量的蛋白。在将这些构建体克隆并引入巴斯德毕赤酵母和其他宿主中之后,出人意料地实现的是,当V结构域从原始完整序列截短时,CLP的分泌得到了极大的改进。V结构域的部分去除允许显著地增加所有宿主中的蛋白产量,出人意料的是,V结构域的完全去除不如此种结构域的部分截短那样有效。
尽管V结构域对这种过程可能有积极影响,但已发现的是它并不是折叠该蛋白的唯一因素。可以示出的是,在不存在V结构域的情况下也发生适当的折叠。所鉴定的主要因素是Scl2单体的浓度、温度、时间、pH值和盐浓度。
由于V结构域被认为对三螺旋Scl2的产生至关重要,因此从未考虑过去除该序列,这导致了以下挑战:
-V结构域大约占整个序列的三分之一,并且阻碍蛋白从巴斯德毕赤酵母宿主中运输出来。这需要包括细胞裂解的复杂下游过程,以从该细胞中去除靶蛋白。
-V结构域本身具有病原性特性,并且需要在纯化过程期间被去除。这通过蛋白酶消化来完成。蛋白酶的使用成本相当高,而且还需要在下游期间将它去除。
以下概述示出使用附接有V结构域的Scl2构建体的产物纯化所需的方法步骤:
细胞分离(离心)

发明内容
本发明描述在甲基营养型酵母巴斯德毕赤酵母或其它宿主中生产胶原样蛋白(CLP)的新型方法。相比于现有技术中已知的当前方法,此种方法的关键特点是:1)蛋白被分泌在培养上清液中,允许在低成本培养基中达到高效价(titer)(>5g/L);2)蛋白容易从所述上清液中纯化,因为在培养基中不存在复杂组分。
出人意料地,来自分泌Scl2p的巴斯德毕赤酵母培养的上清液的纯化产物示出了出乎意料的特性(profile),其与成熟的胶原样序列相容。进一步的分析示出了细胞内酶,最可能是加工蛋白酶Kex2p,在不需要任何额外的蛋白酶步骤的情况下如何能够去除V结构域蛋白序列。另外,为了修改存在于最终产物中的导致降解产物显著积累的切割位点,已对蛋白序列进行了突变,以对此种切割位点进行工程化改造并消除降解。出乎意料地,当非极性氨基酸(缬氨酸,在野生型序列中)突变为极性氨基酸(谷氨酰胺)时,获得了最有效的性能。
出人意料的发现是,在裂解前,在细胞沉淀物(pellet)的储存/冷冻期间,在不存在V结构域的情况下,重组CLP可以在培养上清液中的分泌之后被正确地折叠。与本发明一起公开了新的下游过程,其包括不具有V结构域的Scl2蛋白的生产和用于CL结构域的折叠步骤的有意整合。
因此,本发明提供用于生产重组胶原样蛋白(CLP)的新型方法,其包括以下步骤:
a)在培养基中发酵宿主细胞,所述宿主细胞表达具有与SEQ ID NO:1的氨基酸序列至少60%相同的氨基酸序列的CLP,其中所述氨基酸序列包含SEQ ID NO:1的氨基酸序列的N末端处的至少38个氨基酸的缺失,
b)在所述培养基中积累所述CLP,其中获得发酵液,
c)从所述发酵液中分离所述宿主细胞,以获得上清液,
d)将步骤c)的发酵液的上清液在不超过25℃下孵育至少1小时以用于所述CLP的折叠,
e)任选地,通过以下中的至少一种来纯化所述CLP:溶剂沉淀、切向流过滤(TFF)、离子交换色谱、反相色谱。
所述宿主细胞优选地选自细菌细胞、酵母细胞或植物细胞。优选的是,使用细菌细胞或酵母细胞。
具体实施方式
出人意料的发现是,胶原样蛋白的截短变体,包括具有截短V结构域的变体或没有任何V结构域的变体,导致增加的胶原样蛋白产量和分泌至发酵培养基中。进一步出人意料的是,在不存在V结构域的情况下,截短变体可以正确地折叠。
优选的是,所述氨基酸序列包含SEQ ID NO:1的氨基酸序列的N末端处的38至90个之间的氨基酸的缺失。这包括对N末端的V结构域(包含74个氨基酸)的完全缺失和对V结构域的至少38个氨基酸的不同截短。
在一个优选的实施方案中,所述氨基酸序列与SEQ ID NO:2、SEQ ID NO:3、SEQ IDNO:4、SEQ ID NO:5、SEQ ID NO:6、SEQ ID NO:7、SEQ ID NO:8或SEQ ID NO:9的氨基酸序列至少60%相同。
在一个优选的构型中,所述氨基酸序列与SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4、SEQ ID NO:5、SEQ ID NO:6、SEQ ID NO:7、SEQ ID NO:8或SEQ ID NO:9的氨基酸序列至少90%、92%、94%、96%、97%、98%、99%或100%相同,优选地至少97%,特别优选地至少98%,非常特别优选地至少99%极其优选地是100%相同。
在本发明的一个优选的实施方案中,所述CLP是来自酿脓链球菌的细菌胶原样蛋白。
相应地,本发明还涉及SEQ ID NO:2至9的多肽变体,所述多肽变体含有一个或多个插入(insertion)或缺失。优选地,该多肽含有最多5个、最多4个、最多3个或最多2个的氨基酸的插入或缺失。
在一个优选的实施方案中,步骤d)中的CLP的折叠在-80℃至25℃之间,优选在0℃至20℃之间的温度下进行。在一个优选的配置中,折叠在甘油或盐的存在下进行。
在另一优选的实施方案中,步骤d)中的CLP的折叠进行1小时至48小时之间的时间,优选地1小时至24小时之间的时间。
在另一优选的实施方案中,步骤d)中的CLP折叠以至少1mg/ml,优选地至少4mg/ml的CLP的浓度进行。
在一个优选的实施方案中,所述宿主细胞是巴斯德毕赤酵母、大肠杆菌、恶臭假单胞菌(P.putida)或谷氨酸棒状杆菌(C.glutamicum)物种的微生物,所述微生物包含根据本发明的多肽中的任意一种。

过表达通常意指,相比于起始菌株(亲本菌株)或野生型菌株,如果这是起始菌株的话,核糖核酸、蛋白(多肽)或酶的细胞内浓度或活性的增加。起始菌株(亲本菌株)意指这样的菌株,对该菌株进行了导致过表达的措施。
在过表达中,优选重组过表达的方法。这些方法包括使用体外提供的DNA分子产生微生物的所有方法。此类DNA分子包含例如启动子、表达盒、基因、等位基因、编码区等。这些通过转化法、缀合(conjugation)法、转导法或类似方法被转换(convert)至需要的微生物中。
通过测量由基因转录的mRNA的量,通过确定多肽的量,和通过确定酶活性,可以确认表达或过表达的程度。
待使用的培养基或发酵培养基必须适当地满足相应菌株的要求。在美国细菌学会(the American Society for Bacteriology)的指南“普通细菌学方法手册(Manual ofMethods for General Bacteriology)”(Washington D.C.,USA,1981)中含有各种微生物的培养基的描述。术语“培养基”(culture medium)”和术语“发酵培养基”或“培养基(medium)”是相互可交换的。
可以使用以下作为碳源:糖和碳水化合物,例如葡萄糖、蔗糖、乳糖、果糖、麦芽糖、糖蜜、来自甜菜糖或甘蔗加工的含蔗糖溶液、淀粉、淀粉水解物和纤维素;油和脂肪,例如大豆油、向日葵油、花生油和椰子脂(coconut fat);脂肪酸,例如棕榈酸、硬脂酸和亚油酸;醇,例如甘油、甲醇和乙醇;和有机酸,例如乙酸或乳酸。

另外,培养基必须含有对于生长所必要的盐,例如以金属如钠、钾、镁、钙和铁的氯化物或硫酸盐的形式的盐,例如硫酸镁或硫酸铁。最后,除上文提及的物质之外,还可以使用必需的生长物质,例如氨基酸如高丝氨酸,和维生素如硫胺素、生物素或泛酸。
可以将所述起始材料以单一批次的形式添加至培养物中,或在培养期间以合适的方式供应。
以合适的方式使用碱性化合物例如氢氧化钠、氢氧化钾、氨或氨水,或酸性化合物例如磷酸或硫酸,以用于培养物的pH控制。通常将pH调节至6.0至8.5,优选地6.5至8。对于泡沫进展(development)的控制,可以使用消泡剂,例如脂肪酸的聚乙二醇酯。对于维持质粒的稳定性,可以将合适的选择性作用物质例如抗生素添加至培养基中。发酵优选地在有氧条件下进行。为了维持所述有氧条件,将氧气或含氧气体混合物例如空气引入至培养物中。同样可以使用富含过氧化氢的液体。任选地,发酵在超大气压下进行,例如在0.03至0.2MPa的超大气压下进行。培养的温度通常为20℃至45℃,优选地为25℃至40℃,特别优选地为30℃至37℃。在分批方法或补料分批方法的情况下,优选地继续培养直至已形成足够的量以用于获得期望的有机化学化合物的措施。这一目的通常在10小时至160小时内实现。在连续方法中,更长的培养时间是有可能的。由于微生物的活性,因此发生精细化学品在该微生物的发酵培养基中和/或在该微生物的细胞中的富集(积累)。
合适的发酵培养基的实例可被找到,特别可在专利文件US 5,770,409、US 5,990,350、US 5,275,940、WO 2007/012078、US 5,827,698、WO 2009/043803、US 5,756,345或US7,138,266中找到;可以任选地对所使用菌株的要求进行适当的修改。
该方法的特征可以在于,它是选自分批方法、补料分批方法、重复补料分批方法和连续方法的方法。
该方法的特征可以进一步在于,精细化学品或含有液体或固体精细化学品的产物从含有精细化学品的发酵液中获得。
基于用存在根据本发明的启动子变体的微生物的方法或发酵方法,就选自浓度(每体积形成的化合物)、转化率(每消耗的碳源形成的化合物)、体积生产率(每体积和时间形成的化合物)和生物质特定生产率(每细胞干质量或生物干质量和时间形成的化合物,或每细胞蛋白和时间形成的化合物)、或其它过程参数、和它们的组合的参数中的一个或多个而言,根据本发明的方法或发酵方法的性能增加至少0.5%、至少1%、至少1.5%或至少2%。
由于发酵的措施,获得这样的发酵液,所述发酵液含有期望的精细化学品,优选地含有氨基酸或有机酸。
然后,提供或产生或获得含有精细化学品的以液体或固体形式的产物。
在一个优选的实施方案中,发酵液意指其中微生物在特定的温度下培养特定的时间的发酵培养基或营养培养基。发酵培养基或发酵期间使用的培养基含有确保期望的化合物的生产且通常确保生长和/或活力的所有物质或组分。
发酵完成后,所得发酵液相应地含有:
a)由微生物细胞的生长产生的微生物的生物质(细胞团块(cell mass)),
b)在发酵的过程中形成的期望的精细化学品,
c)在发酵的过程中可能形成的有机副产物,和
d)未被发酵消耗的所使用的发酵培养基的组分或起始材料的组分,例如维生素如生物素,或盐如硫酸镁。
除相应的期望的化合物之外,有机副产物还包括由发酵中使用的微生物生成的并可能分泌的物质。
将发酵液从培养器皿或发酵容器中取出,任选地收集,并且用于提供含有精细化学品的以液体或固体形式的产物。表述“获得含精细化学品的产物”也用于此。在最简单的情况下,从发酵容器中取出的含精细化学品的发酵液本身就是所获得的产物。
通过选自以下的措施中的一个或多个,
a)水的部分(>0%至<80%)至完全(100%)或几乎完全(≥80%、≥90%、≥95%、≥96%、≥97%、≥98%、≥99%)去除,
b)生物质的部分(>0%至<80%)至完全(100%)或几乎完全(≥80%、≥90%、≥95%、≥96%、≥97%、≥98%、≥99%)去除,其中在该去除之前任选地将该生物质灭活,
c)在发酵的过程中形成的有机副产物的部分(>0%至<80%)至完全(100%)或几乎完全(≥80%、≥90%、≥95%、≥96%、≥97%、≥98%、≥99%、≥99.3%、≥99.7%)去除,和
d)未被发酵消耗的所使用的发酵培养基的组分或起始材料的组分的部分(>0%)至完全(100%)或几乎完全(≥80%、≥90%、≥95%、≥96%、≥97%、≥98%、≥99%、≥99.3%、≥99.7%)去除,
从发酵液中实现期望的有机化学化合物的浓缩或纯化。以这种方式,分离出具有期望含量的所述化合物的产物。
水的部分(>0%至<80%)至完全(100%)或几乎完全(≥80%至<100%)去除(措施a))也被称为干燥。
在该方法的一个变体中,通过水、生物质、有机副产物和所使用的发酵培养基的未消耗组分的完全或几乎完全去除,成功地获得纯(≥80重量%、≥90重量%)或高纯度(≥95重量%、≥97重量%、≥99重量%)产物形式的期望的有机化学化合物,优选地胶原样蛋白。对于根据a)、b)、c)或d)的措施,在现有技术中可获得多种多样的技术指导。
在用于生产胶原样蛋白的方法的情况下,优选这样的方法,其中获得不含有发酵液的任何组分的产物。这些产物特别用于人类医学中,用于制药工业中,且用于食品工业中。
实施例
通过发酵在酵母宿主细胞巴斯德毕赤酵母中,生产了胶原样蛋白。为了在巴斯德毕赤酵母中生产来自酿脓链球菌的Scl2,使用不同的算法已经对胶原样蛋白(全长蛋白和截短变体以及无V结构域变体)的序列进行了密码子优化,并且将其克隆在巴斯德毕赤酵母的分泌载体中。所使用的序列概述于SEQ ID NO:1至SEQ ID NO:9中。对于特定序列中的每一个,按照标准方案在巴斯德毕赤酵母中转化载体,并且应用了补料分批模式的标准表达方案(Damasceno,L.M.,Huang,CJ.&Batt,C.A.Protein secretion in Pichia pastorisand advances in protein production.Appl Microbiol Biotechnol 93,31–39(2012))。在细胞培养物的上清液中,通过HPLC分析检测了Scl2p蛋白的胶原结构域。发酵后,通过离心(12000g,室温下5分钟)已经将上清液与生物质分离。
使用大肠杆菌、桥石短芽孢杆菌(B.choshinensis)或谷氨酸棒状杆菌,在类似条件下可以生产基于序列SEQ ID NO:1至序列SEQ ID NO:9的Scl2p蛋白的胶原结构域。在酵母或谷氨酸棒状杆菌中的生产的情况下,由细胞分泌胶原结构域。在这种方式中,不需要细胞裂解作为初始纯化步骤。在大肠杆菌中的生产的情况下,细胞裂解是强制的,以从该细胞中去除胶原结构域。
在桥石短芽孢杆菌(Brevibacillus choshinensis)中也表达了全长胶原样蛋白、截短变体(截短3)和无V结构域变体(基于来自酿脓链球菌的基因scl2)。因此,对应的DNA序列被克隆至对于桥石短芽孢杆菌合适的分泌载体中。用新构建的质粒转化桥石短芽孢杆菌是根据Mizukami等人2010(Curr Pharm Biotechnol 2010,13:151-258)来完成的。
使用来自Eppendorf(Hamburg,德国)的平行生物反应器系统,在33℃和pH 7下的分批培养中,分析了桥石短芽孢杆菌菌株的生产不同胶原样蛋白的能力。使用1L反应器进行了发酵。生产培养基(TM培养基,Biomed Res Int 2017,2017:5479762)含有了10g/L的葡萄糖。发酵后,通过离心已经将上清液与生物质分离,并且将该上清液用于SDSPAGE分析。对于所有三种变体,都产生了Scl2p蛋白的胶原结构域。
在谷氨酸棒状杆菌中也表达了全长胶原样蛋白和无V结构域变体(基于来自酿脓链球菌的基因scl2)。因此,对应的DNA序列与位于上游的用于蛋白分泌的信号肽一起被克隆至用于谷氨酸棒状杆菌的穿梭载体中(Biotechnology Techniques 1999,13:437-441.)。通过电穿孔的方式,例如由Ruan等人(Biotechnology Letters 2015,37:2445-2452)所描述的,用新构建的质粒转化了谷氨酸棒状杆菌菌株ATCC 13032。
使用来自Eppendorf(Hamburg,德国)的平行生物反应器系统,在30℃和pH 7下的补料分批培养中,分析了谷氨酸棒状杆菌菌株生产不同胶原蛋白的能力。使用1L反应器进行了发酵。生产培养基在分批阶段含有了20g/L的葡萄糖,并且用4g/L*h的葡萄糖进料运行了补料分批阶段。发酵后,通过离心已经将上清液与生物质分离,并且将该上清液用于HPLC分析。对于两种变体,都产生了Scl2p蛋白的胶原结构域。对于胶原样蛋白的截短变体,效价与全长变体相比更高。
方法步骤总结如下:
1.在酵母、大肠杆菌或棒状杆菌中的胶原样蛋白的生产
2.细胞裂解(仅针对大肠杆菌)
3.细胞分离(过滤或离心)
4.CL单链的折叠以形成三螺旋结构
5.通过溶剂沉淀和超滤的进一步的纯化
6.对纯化的CL蛋白的冷冻干燥
为了确定折叠动力学,将来自在巴斯德毕赤酵母中的生产的冷冻干燥的Scl2p蛋白的胶原结构域以40g/L的浓度溶解在去离子(DI)水中,并且在40℃下展开。将溶液分开并且进一步稀释在1至40g/L的浓度范围内。然后,在从4至30℃的范围内的不同温度下将不同的样品孵育20小时,以获得给定温度和浓度范围内的胶原样蛋白的折叠动力学。使用尺寸排阻色谱(SEC)确定折叠比率。将胶原样蛋白的温度依赖性折叠总结在图1中。
制备第二样品集,以覆盖在0.25至1小时的时间范围和4至20℃的孵育温度内的CL蛋白的时间依赖性折叠。结果总结在图2至图4中。

Claims (9)
1.用于生产重组胶原样蛋白(CLP)的方法,其包括以下步骤:
a)在培养基中发酵宿主细胞,所述宿主细胞表达具有与SEQ ID NO:1的氨基酸序列至少60%相同的氨基酸序列的CLP,其中所述氨基酸序列包含在SEQ ID NO:1的氨基酸序列的N-末端处的至少38个氨基酸的缺失,
b)在所述培养基中积累所述CLP,其中获得发酵液,
c)从所述发酵液中分离所述宿主细胞,以获得上清液,
d)将步骤c)的所述发酵液的所述上清液在不超过25℃下孵育至少1小时,以用于所述CLP的折叠,
e)任选地,通过以下中的至少一种来纯化所述CLP:溶剂沉淀、切向流过滤(TFF)、离子交换色谱、反相色谱。
2.根据权利要求1所述的方法,其中所述氨基酸序列包含在SEQ ID NO:1的氨基酸序列的N-末端处的38至90个氨基酸的缺失。
3.根据权利要求1或2所述的方法,其中所述氨基酸序列与SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4、SEQ ID NO:5、SEQ ID NO:6、SEQ ID NO:7、SEQ ID NO:8或SEQ ID NO:9的氨基酸序列至少60%相同。
4.根据前述权利要求中的任意一项所述的方法,其中所述氨基酸序列与SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4、SEQ ID NO:5、SEQ ID NO:6、SEQ ID NO:7、SEQ ID NO:8或SEQID NO:9的氨基酸序列至少90%、92%、94%、96%、97%、98%、99%或100%相同,优选地至少97%,特别优选地至少98%,非常特别优选地至少99%,极其优选地是100%相同。
5.根据前述权利要求中的任意一项所述的方法,其中所述CLP是来自酿脓链球菌(Streptococcus pyogenes)的胶原样蛋白。
6.根据前述权利要求中的任意一项所述的方法,其中步骤d)中的CLP的折叠在-80℃至25℃,优选地在0℃至20℃的温度下进行。
7.根据前述权利要求中的任意一项所述的方法,其中步骤d)中的CLP的折叠进行1小时至48小时,优选地1小时至24小时的时间。
8.根据前述权利要求中的任意一项所述的方法,其中步骤d)中的CLP的折叠以至少1mg/ml,优选地至少4mg/ml的CLP的浓度进行。
9.根据前述权利要求中的任意一项所述的方法,其中所述宿主细胞是酵母细胞,优选地是巴斯德毕赤酵母(Pichia pastoris),或细菌细胞,优选地是大肠杆菌(E.coli)、棒状杆菌(Corynebacterium)或短杆菌(Brevibactetium)。
