Abstract

本发明是申请日为2017年9月1日,申请号为201780066838.2,发明名称为视觉功能再生剂或视觉功能降低预防剂的专利申请的分案申请。
技术领域
本发明涉及视觉功能再生剂或视觉功能降低预防剂。
背景技术
视紫红质是在人和动物的视网膜中具有7次跨膜型结构(seven transmembranestructure)的光敏性的受体,已知还有源自微生物的离子通道型、离子泵型的视紫红质。
例如,非专利文献1中公开了作为离子通道型的视紫红质的通道视紫红质2(ChR2)。另外,非专利文献2中报道了,通过将突变型通道视紫红质导入视网膜神经节细胞中,而在小鼠/大鼠中再生了一定的视觉功能。

发明内容
发明要解决的问题
然而,离子通道型视紫红质的视觉功能的再生效果并不充分,存在改善的余地。
本发明是鉴于以上的实际情况而完成的,目的在于提供具有优异的视觉功能的再生功能的视觉功能再生剂或视觉功能降低预防剂。
用于解决问题的方案
本发明人等发现:融合了源自微生物的离子转运视紫红质和动物来源的G蛋白偶联型受体视紫红质这两种完全不同的视紫红质而成的嵌合蛋白实际上具有优异的视觉功能的再生功能,以至完成了本发明。更具体而言,本发明由以下的特征构成。
(1)一种视觉功能再生剂或视觉功能降低预防剂,其含有嵌合蛋白作为有效成分,所述嵌合蛋白具有微生物来源的离子转运型受体视紫红质的氨基酸序列和动物来源的G蛋白偶联型受体视紫红质的氨基酸序列。
(2)根据(1)所述的视觉功能再生剂或视觉功能降低预防剂,其中,前述嵌合蛋白是微生物来源的离子转运型受体视紫红质的氨基酸序列中的、细胞质侧的第2环路和/或细胞质侧的第3环路的氨基酸序列被置换为G蛋白偶联型受体视紫红质的细胞质侧的第2环路和/或细胞质侧的第3环路的氨基酸序列的蛋白。
(3)根据(1)或(2)所述的视觉功能再生剂或视觉功能降低预防剂,其中,前述微生物来源的离子转运型受体视紫红质为属于粘杆菌(Gloeobacter)属的微生物来源的视紫红质,且前述G蛋白偶联型受体视紫红质为牛或人来源的视紫红质。
(4)根据(1)~(3)中任一项所述的视觉功能再生剂或视觉功能降低预防剂,其中,前述嵌合蛋白具有以下的(a)~(d)的任一项所述的DNA所编码的氨基酸序列,
(a)具有编码序列号1~4的任一项所述的氨基酸序列的碱基序列的DNA
(b)具有和与编码序列号1~4的任一项所述的氨基酸序列的碱基序列互补的碱基序列在严格的条件下能够杂交的碱基序列的DNA
(c)具有编码在序列号1~4的任一项所述的氨基酸序列中置换、缺失和/或添加了1个或多个的氨基酸的氨基酸序列的碱基序列、且具有视觉功能再生功能或视觉功能降低预防功能的DNA
(d)由编码与序列号1~4的任一项所述的氨基酸序列具有90%以上的同源性的氨基酸序列的碱基序列组成、且具有视觉功能再生功能或视觉功能降低预防功能的DNA。
(5)一种视觉功能再生剂或视觉功能降低预防剂,其含有嵌入了编码(1)~(4)的任一项所述的嵌合蛋白的氨基酸序列的DNA的表达载体作为有效成分。
(6)根据(1)~(5)中任一项所述的视觉功能再生剂或视觉功能降低预防剂,其用于色素性视网膜炎的治疗或预防。
(7)一种腺相关病毒(AAV)载体或慢病毒载体,其插入了嵌合蛋白的序列,所述嵌合蛋白具有微生物来源的离子转运型受体视紫红质的氨基酸序列和动物来源的G蛋白偶联型受体视紫红质的氨基酸序列。
(8)一种腺相关病毒(AAV)载体或慢病毒载体在制造视觉功能再生或视觉功能降低预防的药物中的应用,其中,所述腺相关病毒(AAV)载体或慢病毒载体插入了嵌合蛋白的序列,所述嵌合蛋白具有微生物来源的离子转运型受体视紫红质的氨基酸序列和动物来源的G蛋白偶联型受体视紫红质的氨基酸序列。
发明的效果
根据本发明,能够发挥优异的视觉功能的再生功能。

具体实施方式
以下对本发明的具体的实施方式进行详细地说明,但本发明不受以下的实施方式任何限定,可以在本发明的目的的范围内进行适宜变更地实施。需要说明的是,对于重复说明的部分,有时适宜省略说明,但不用于限定发明的主旨。
<视觉功能再生剂或视觉功能降低预防剂>
本发明的视觉功能再生剂或视觉功能降低预防剂含有嵌合蛋白作为有效成分,所述嵌合蛋白具有微生物来源的离子转运型受体视紫红质的氨基酸序列和动物来源的G蛋白偶联型受体视紫红质的氨基酸序列。
视紫红质在内部具有视黄醛(retinal)这样的色素,使其通过接受光而激活并将视觉信号传递至脑。由于微生物来源的离子转运型受体视紫红质即使照射光视黄醛也不会脱落,因此通过能够吸收光而重复活化,但不能像动物来源的G蛋白偶联型受体视紫红质那样激活G蛋白。相对于此,根据本发明,推测出:通过使动物来源的G蛋白偶联型受体视紫红质与能够重复使用的微生物来源的离子转运型受体视紫红质融合,从而保持微生物来源的离子转运型受体离子通道型受体视紫红质所具有的重复活化功能,且能够得到借由基于G蛋白偶联型受体的内源性的G蛋白的高的活性,可以得到优异的视觉再生效果。由此,本发明人等实际发现:微生物来源的视紫红质与动物来源的G蛋白偶联型受体是功能完全不同的类型的受体,组合这2种受体的嵌合蛋白具有优异的视觉功能的再生功能。另外,如上所述,由于得到高的活性的同时能够重复活化,因此还可以期待视觉功能的降低的预防效果(例如,抑制色素性视网膜炎等视网膜疾病的加剧)。
作为离子转运型受体视紫红质,可列举出:离子泵型受体视紫红质、离子通道型受体视紫红质。
本发明的嵌合蛋白是微生物来源的离子转运型受体视紫红质与G蛋白偶联型受体视紫红质的嵌合蛋白,具有7次跨膜型结构。本发明中,微生物来源的离子转运型受体视紫红质与G蛋白偶联型受体视紫红质的嵌合蛋白优选被设计为:以高水平具有微生物来源的离子转运型受体视紫红质的重复活化的功能和基于G蛋白偶联型受体视紫红质的G蛋白活性这两者。从该观点出发,由于以高水平维持两者的活性、特别是发挥高的视觉功能再生功能,因此本发明的嵌合蛋白优选为微生物来源的离子转运型受体视紫红质的氨基酸序列中的、细胞质侧的第2环路和/或细胞质侧的第3环路的氨基酸序列被置换为G蛋白偶联型受体视紫红质的细胞质侧的第2环路和/或细胞质侧的第3环路的氨基酸序列的蛋白。需要说明的是,“细胞质侧的第2环路”、“细胞质侧的第3环路”分别是指位于7个环路中的从N末端侧起第2位、从N末端侧起第3位的环路。
作为微生物来源的离子转运型受体视紫红质,例如可列举出:粘杆菌(Gloeobacter)属等真细菌、团藻(Volvox)属、衣藻(Chlamydomonas)属、黄藻(Guillardia)属等属于真核生物等的微生物来源的视紫红质。作为粘杆菌(Gloeobacter)属,可列举出:紫色粘杆菌(Gloeobacter violaceus)等。作为团藻(Volvox)属,可列举出团藻(Volvoxcarteri)等。作为衣藻(Chlamydomonas)属,可列举出莱茵衣藻(Chlamydomonasreinhardtii)等。作为黄藻(Guillardia)属,可列举出蓝隐藻(Guillardia theta)等。为了得到更高的视觉再生/预防效果,可以认为重要的是与G蛋白活化环路的立体结构的相容性和膜转移效率,但微生物来源的离子转运型受体视紫红质尤其与G蛋白活化环路的立体结构的相容性和膜转移效率良好,因此优选为粘杆菌(Gloeobacter)属。特别是,属于粘杆菌(Gloeobacter)属的微生物中,优选紫色粘杆菌。另外,属于粘杆菌属的微生物优选与动物来源的G蛋白偶联型受体视紫红质中的、牛或人来源的G蛋白偶联型受体视紫红质组合而融合。另外,粘杆菌(Gloeobacter)属具有在作为真细菌的大肠杆菌及作为真核生物的人的细胞中均可表达这样重要的性质,在这方面也是优选的。
作为动物来源的G蛋白偶联型受体视紫红质,例如可列举出:牛、人、小鼠、大鼠、猫、狗、猪、羊、马等来源的视紫红质。其中,特别优选牛或人来源的视紫红质。
更具体而言,嵌合蛋白优选具有以下的(a)~(d)的任一项所述的DNA所编码的氨基酸序列。
(a)具有编码序列号1~4的任一项所述的氨基酸序列的碱基序列的DNA
(b)具有和与编码序列号1~4的任一项所述的氨基酸序列的碱基序列互补的碱基序列在严格的条件下能够杂交的碱基序列的DNA
(c)具有编码在序列号1~4的任一项所述的氨基酸序列中置换、缺失和/或添加了1个或多个的氨基酸的氨基酸序列的碱基序列、且具有视觉功能再生功能或视觉功能降低预防功能的DNA
(d)由编码与序列号1~4的任一项所述的氨基酸序列具有90%以上的同源性的氨基酸序列的碱基序列组成、且具有视觉功能再生功能或视觉功能降低预防功能的DNA。
作为上述的G蛋白偶联型受体视紫红质的细胞质侧的第2环路,优选具有以下的(e)~(h)所述的DNA所编码的氨基酸序列。
(e)具有编码序列号5或6所述的氨基酸序列的碱基序列的DNA
(f)具有和与编码序列号5或6所述的氨基酸序列的碱基序列互补的碱基序列在严格的条件下能够杂交的碱基序列的DNA
(g)具有编码在序列号5或6所述的氨基酸序列中置换、缺失和/或添加了1个或多个的氨基酸的氨基酸序列的碱基序列的DNA
(h)由编码与序列号5或6所述的氨基酸序列具有90%以上的同源性的氨基酸序列的碱基序列组成的DNA。
作为上述的G蛋白偶联型受体视紫红质的细胞质侧的第3环路,优选具有以下的(i)~(l)所述的DNA所编码的氨基酸序列。
(i)具有编码序列号7所述的氨基酸序列的碱基序列的DNA
(j)具有和与编码序列号7所述的氨基酸序列的碱基序列互补的碱基序列在严格的条件下能够杂交的碱基序列的DNA
(k)具有编码在序列号7所述的氨基酸序列中置换、缺失和/或添加了1个或多个的氨基酸的氨基酸序列的碱基序列的DNA
(l)由编码与序列号7所述的氨基酸序列具有90%以上的同源性的氨基酸序列的碱基序列组成的DNA。
编码序列号1~4的任一项所述的氨基酸序列的碱基序列为编码本发明的嵌合蛋白的碱基序列的优选序列。编码该序列号1~4的任一项所述的氨基酸序列的碱基序列具有视觉功能再生功能或视觉功能降低预防功能。需要说明的是,本说明书中,“碱基序列具有视觉功能再生功能或视觉功能降低预防功能”是指:碱基序列所编码的多肽具有视觉功能再生功能或视觉功能降低预防功能。另外,具有编码序列号1~4的任一项所述的氨基酸序列的碱基序列的DNA中还包括:具有视觉功能再生功能或视觉功能降低预防功能的各种突变体、同系物(homologs)。具有编码序列号1~4的任一项所述的氨基酸序列的碱基序列的DNA的突变体、同系物中,包括例如:具有和编码序列号1~4的任一项所述的氨基酸序列的碱基序列在严格的条件下能够杂交的碱基序列的DNA。另外,具有编码序列号5~7的任一项所述的氨基酸序列的碱基序列的DNA的突变体、同系物中包括:具有和编码序列号5~7的任一项所述的氨基酸序列的碱基序列在严格的条件下能够杂交的碱基序列的DNA。作为“严格的条件”,例如可列举出:在通常的杂交分析缓冲液中、在40~70℃(优选为50~67℃、更优选为60~65℃)下进行反应,在盐浓度15~300mM(优选为15~150mM、更优选为15~60mM、进一步优选为30~50mM)的清洗液中进行清洗的条件。
序列号1~4的任一项可以作为本发明的嵌合蛋白的氨基酸序列使用。编码本发明的嵌合蛋白的氨基酸序列的DNA中还包括:具有编码在上述序列号1~4的任一项所述的的氨基酸序列中置换、缺失和/或添加了1个或多个的氨基酸的氨基酸序列的碱基序列的DNA。此处,序列号1~4的任一项中,“1个或多个”通常为50个氨基酸以内,优选为30个氨基酸以内,进一步优选为10个氨基酸以内(例如,5个氨基酸以内、3个氨基酸以内、1个氨基酸)。另外,在序列号5~7的任一项中,“1个或多个”通常为6个氨基酸以内,优选为5个氨基酸以内,进一步优选为4个氨基酸以内(例如,3个氨基酸以内、2个氨基酸以内、1个氨基酸)。在维持嵌合蛋白的视觉功能再生功能或视觉功能降低预防功能的情况下,理想的是:将待突变的氨基酸残基突变为保存有氨基酸侧链的性质的其它氨基酸。例如作为氨基酸侧链的性质,可以列举出:疏水性氨基酸(A、I、L、M、F、P、W、Y、V)、亲水性氨基酸(R、D、N、C、E、Q、G、H、K、S、T)、具有脂肪族侧链的氨基酸(G、A、V、L、I、P)、具有含有羟基的侧链的氨基酸(S、T、Y)、具有含有硫原子的侧链的氨基酸(C、M)、具有含有羧酸和酰胺的侧链的氨基酸(D、N、E、Q)、具有含有碱基的侧链的氨基酸(R、K、H)、具有含有芳香族的侧链的氨基酸(H、F、Y、W)(括号内均表示氨基酸的一个字符标记)。需要说明的是,已知的是:具有相对于某氨基酸序列通过1个或多个氨基酸残基缺失、添加和/或置换为其它氨基酸而被修饰的氨基酸序列的蛋白质维持其生物学活性(Mark,D.F.et al.,Proc.Natl.Acad.Sci.USA(1984)81,5662-5666、Zoller,M.J.&Smith,M.Nucleic Acids Research(1982)10,6487-6500、Wang,A.et al.,Science224,1431-1433、Dalbadie-McFarland,G.et al.,Proc.Natl.Acad.Sci.USA(1982)79,6409-6413)。
具有编码序列号1~4的任一项所述的氨基酸序列的碱基序列的DNA的突变体、同系物中包括:由与编码序列号1~4的任一项所述的氨基酸序列的碱基序列具有高的同源性的碱基序列组成的DNA。这样的DNA优选与编码序列号1~4的任一项所述的氨基酸序列的碱基序列具有90%以上、进一步优选具有95%以上(96%以上、97%以上、98%以上、99%以上)的同源性。另外,具有编码序列号5~7的任一项所述的氨基酸序列的碱基序列的DNA的突变体、同系物中包括:由与编码序列号5~7的任一项所述的氨基酸序列的碱基序列具有高的同源性的碱基序列组成的DNA。这样的DNA优选与编码序列号5~7的任一项所述的氨基酸序列的碱基序列具有90%以上、进一步优选具有95%以上(96%以上、97%以上、98%以上、99%以上)的同源性。氨基酸序列、碱基序列的同源性可以通过基于Karlin andAltschul的算法BLAST
(Proc.Natl.Acad.Sci.USA90:5873-5877,1993)来确定。基于该算法,开发出了被称为BLASTN、BLASTX的程序(Altschul et al.J.Mol.Biol.215:403-410,1990)。基于BLAST并通过BLASTN来解析碱基序列时,参数例如为score(得分)=100、wordlength(字长)=12。另外,基于BLAST并通过BLASTX来解析氨基酸序列时,参数例如为score=50、wordlength=3。使用BLAST和Gapped BLAST程序时,使用各程序的默认参数(default parameter)。这些解析方法的具体的方法是公知的(http://www.ncbi.nlm.nih.gov.)。
本发明中的“DNA”可以是正义链或反义链(例如,可以用作探针),其形状可以是单链或双链。另外,可以是基因组DNA,还可以是cDNA或被合成的DNA。
作为获得本发明的DNA的方法,没有特别限定,可列举出如下方法:通过由mRNA逆转录来获得cDNA的方法(例如,RT-PCR法);由基因组DNA制备的方法;利用化学合成来合成的方法;由基因组DNA文库、cDNA文库分离的方法等公知的方法(例如,参照日本特开平11-29599号公报)。
本发明的视觉功能再生剂或视觉功能降低预防剂中所使用的嵌合蛋白的制备,例如可以通过使用导入了包含编码前述的嵌合蛋白的DNA的表达载体的转化体来进行。例如,首先,在适宜的条件下培养该转化体,合成该DNA所编码的嵌合蛋白。接着,从转化体或培养液中回收所合成的蛋白,由此能够得到本发明的嵌合蛋白。
进行更具体的说明,可以通过将编码上述的嵌合蛋白的DNA插入适合的表达载体中来制作。“适合的载体”只要是在原核生物和/或真核生物的各种宿主内能够保持复制或自体繁殖(self-proliferate)的载体即可,可以根据使用目的进行适宜选择。例如,欲获得大量DNA时可以选择高拷贝数载体,欲获得多肽(嵌合蛋白)时可以选择表达载体。作为其具体例,没有特别限定,例如可列举出日本特开平11-29599号公报中记载的公知的载体。
另外,表达载体不仅能用来合成嵌合蛋白,而且还能用于本发明的视觉功能再生剂或视觉功能降低预防剂。即,本发明的视觉功能再生剂或视觉功能降低预防剂可以含有嵌入了编码上述的嵌合蛋白的氨基酸序列的DNA的表达载体作为有效成分。通过将上述表达载体直接导入人体中而能够用于视觉功能再生、视觉功能降低预防。此时的载体使用能导入人的细胞内的载体。作为上述载体,例如腺相关病毒载体(AAV载体)、慢病毒载体是适合的。
载体的导入方法可以根据载体、宿主的种类等进行适宜选择。作为其具体例,没有特别限定,例如将细菌作为宿主时,可列举出原生质体(protoplast)法、感受态(competent)法等公知的方法(例如,参照日本特开平11-29599号公报)。另外,使用表达载体作为本发明的视觉功能再生剂或视觉功能降低预防剂的有效成分时,例如,可以通过将上述的AAV载体等注射至眼内来导入。
导入表达载体的宿主只要适于表达载体且可进行转化即可,作为其具体例,没有特别限定,可列举出:细菌、酵母、动物细胞、昆虫细胞等公知的天然细胞或人工建立的细胞(参照日本特开平11-29599号公报)、或者人、小鼠等动物。对于转化体的培养,可以以能够大量且容易获得嵌合蛋白的方式、根据转化体的种类等从公知的营养培养基中适宜选择,适宜调节温度、营养培养基的pH、培养时间等来进行(例如,参照日本特开平11-29599号公报)。
作为嵌合蛋白的分离方法和纯化方法,没有特别限定,可列举出:利用溶解度的方法、利用分子量的差的方法、利用电荷的方法等公知的方法(例如,参照日本特开平11-29599号公报)。
本说明书中的“有效成分”是指以为了得到视觉功能再生效果或视觉功能降低预防效果所需的量所含有的成分,在不损害效果直至低于期望的水平的范围内,还可以含有其它成分。另外,本发明的视觉功能再生剂或视觉功能降低预防剂可以是经制剂化的物质。另外,本发明的视觉功能再生剂或视觉功能降低预防剂的给药途径可以是口服或非口服,可以根据制剂的形态等进行适宜设定。
口服给药的情况,可以制剂化成片剂、颗粒剂、细粒剂、散剂、胶囊剂等各种形状来使用,还可以含有在制剂中通常使用的粘合剂、遮盖剂、赋形剂、润滑剂、崩解剂、湿润剂之类的添加剂。另外,除了这些之外,口服给药时的制剂还可以制剂化成内服水溶液、悬浮剂、乳剂、浆液剂等液体状态,还可以制剂化成在使用时再溶解的干燥状态的物质。
非口服给药的情况,可以制剂化成收纳于单次给药量安瓿或多次给药量容器或管内的状态,另外,还可以含有稳定剂、缓冲剂、保存剂、等渗剂等添加剂。另外,非口服给药时的制剂还可以制成在使用时能用适合的载体(灭菌水等)进行再溶解的粉体。
作为非口服给药,可列举出:玻璃体内给药、结膜下给药、前房内给药、滴眼给药等,优选为玻璃体内给药。
以上叙述的本发明的视觉功能再生剂或视觉功能降低预防剂可以利用如上述那样的方法对人给药而用于视觉功能再生或视觉功能降低预防。
本发明中,“视觉功能再生”是指使降低了的视觉功能得以改善,可以是一部分视觉功能的再生,还可以是完全再生。另外,“视觉功能降低预防”是指防止视觉功能降低、或者抑制视觉功能降低的发展等。作为这样的视觉功能,可列举出:视力、对比敏感度、亮暗适应、色觉等。
本发明的视觉功能再生剂或视觉功能降低预防剂还可以用作通过视觉功能再生或视觉功能降低预防而可期待的用途,例如,可以用于与视觉功能降低相关的疾病的治疗或预防。作为与视觉功能降低相关的疾病,可列举出:色素性视网膜炎、老年性黄斑变性、近视性黄斑病变、黄斑营养不良、糖尿病视网膜病变、葡萄膜炎、视网膜剥离等。
<载体>
本发明包括:插入了嵌合蛋白的序列的腺相关病毒(AAV)载体或慢病毒载体,所述嵌合蛋白的序列具有微生物来源的离子转运型受体视紫红质的氨基酸序列和动物来源的G蛋白偶联型受体视紫红质的氨基酸序列。
另外,本发明包括:插入了嵌合蛋白的序列的腺相关病毒(AAV)载体或慢病毒载体在制造视觉功能再生或视觉功能降低预防的药物中的应用,所述嵌合蛋白具有微生物来源的离子转运型受体视紫红质的氨基酸序列和动物来源的G蛋白偶联型受体视紫红质的氨基酸序列。
嵌合蛋白可以使用与上述情况同样的物质。
实施例
如下所述,使用小鼠进行了有关视觉功能的实验。
(实验动物)
实验使用野生型小鼠(C57BL/6J,CLEAJapan Inc.)和色素性视网膜炎模型(rd1)小鼠(C3H/HeJ Jcls,CLEAJapan Inc.),均使用3周龄雄性小鼠。
(编码嵌合蛋白(GR/BvRh)的DNA的制作)
将从相当于紫色粘杆菌的视紫红质(Gloeobacter violaceus Rhodopsin(GR)、序列号8)的细胞质侧的第2环路的N末端的相当于第137-145位的氨基酸的序列置换为牛视紫红质(BvRh)(序列号9)的第137-145位的氨基酸相当序列,另外,将从相当于紫色粘杆菌的视紫红质的细胞质侧的第3环路的N末端的第198-206位的氨基酸的序列置换为牛视紫红质的第225-252位的氨基酸相当序列,进而将编码作为紫色粘杆菌的视紫红质的第132位的氨基酸的谷氨酸置换为谷氨酰胺的嵌合蛋白的DNA插入pCDNA3.1载体。突变体的制作利用QuicChange法进行。
(插入了嵌合蛋白的序列的腺相关病毒(AAV)载体的制作)
将EGFP或GR/BvRh基因亚克隆至AAV2穿梭质粒(shuttle plasmid),来制作AAV2-CAGGS-EGFP-WPRE-pA(EGFP的表达用的载体)和AAV2-CAGGS-GR/BvRh-WPRE-pA(嵌合蛋白表达用的载体)作为病毒表达构建体。病毒载体的包装通过将载体质粒、AAV载体质粒、腺病毒辅助质粒这3种质粒转染至HEK293细胞中来进行,病毒载体的纯化使用氯化铯法。需要说明的是,载体中“ITR”是“Inverted Terminal Repeat(反向末端重复)”的简称。“CAGGS”是指CAG启动子的区域的序列。“WPRE”是“woodchuck hepatitis virus post-transcriptionalregulatory element(土拨鼠肝炎病毒转录后调控元件)”的简称。“pA”是指肽/标签。“EGFP”是“enhanced green fluorescent protein(增强型绿色荧光蛋白)”的简称。
(玻璃体注射)
对于野生型小鼠或色素性视网膜炎模型(rd1)小鼠,将美托咪定盐酸盐(0.75mg/kg)、咪达唑仑(4mg/kg)、酒石酸布托啡诺(5mg/kg)的混合液进行腹腔内给药,在全身麻醉下使用安装有32号针头的微型注射器将上述AAV载体(“AAV2-CAGGS-EGFP-WPRE-pA”或“AAV2-CAGGS-GR/BvRh-WPRE-pA”)以各1×1012vg/ml 1μl从锯齿缘(ora serrata)附近注射至玻璃体内。
(报道观察)
在注射7周后、从注射了AAV2-CAGGS-EGFP-WPRE-pA的野生型小鼠中摘取视网膜,用4%多聚甲醛固定1小时,利用荧光显微镜观察经整体制片(whole-mount)的视网膜。将其结果示于图1。图1中,GCL:神经节细胞层,INL:内核层,ONL:外核层。观察的结果是:在视网膜中可观察到绿色的荧光(例如,图1的箭头),因此确认到载体的导入、目的基因的表达得以正常进行。
(Multielectorode Array Recording(MEA):多电极阵列测定)
对色素性视网膜炎模型(rd1)小鼠注射了
AAV2-CAGGS-GR/BvRh-WPRE-pA起7周后,在全身麻醉下摘取眼球,然后将眼球静置于用95%O2和5%CO2鼓泡的Ames培养基(Sigma-Aldrich,St Louis,MO;A1420)内,摘取视网膜。对于视网膜以神经节细胞层朝下与电极接触的方式进行固定,进行光刺激(白色光、1000cd/m2、1秒)并记录了视网膜神经节细胞的细胞外电位。另外,将未注射AAV2-CAGGS-GR/BvRh-WPRE-pA的色素性视网膜炎模型(rd1)小鼠作为对照,利用同样的方法记录了视网膜神经节细胞的细胞外电位。多电极阵列测定使用MEA2100-Lite system(Multi-ChannelSystems(多通系统),Reutlingen,Germany)。将其结果示于图2。图2的(a)表示对照小鼠的图,图2中的(b)表示注射了AAV2-CAGGS-GR/BvRh-WPRE-pA的小鼠的图。需要说明的是,图2的图中,横轴表示经过时间,箭头所示的区域表示进行了光刺激的区域。
如图2所示,可知:对照在进行了光刺激的区域也未观察到任何变化,但在注射了AAV2-CAGGS-GR/BvRh-WPRE-pA的小鼠中,电位得以增加。由该结果可知,GR/BvRh对于色素性视网膜炎具有视觉功能再生效果。
另外,利用与上述同样的方法进行多电极阵列测定,得到了用光栅图显示10次视网膜神经节细胞的发射的图(图3的上半部分)、及纵轴以每1秒的发射频率进行的直方图(图3的下半部分)。以0~1秒进行光刺激。图3的(a)表示对照小鼠的图,图3的(b)表示注射了AAV2-CAGGS-GR/BvRh-WPRE-pA的小鼠的图。
如图3所示,对照小鼠中未观察到光应答,而注射了AAV2-CAGGS-GR/BvRh-WPRE-pA的小鼠中可观察到神经节细胞的发射,确认了视觉再生效果。
Claims (1)
1.一种腺相关病毒(AAV)载体或慢病毒载体,其插入了含有编码嵌合蛋白的核酸序列的核酸分子,
所述嵌合蛋白具有属于粘杆菌(Gloeobacter)属的微生物来源的离子转运型受体视紫红质的氨基酸序列和牛或人来源的G蛋白偶联型受体视紫红质的氨基酸序列,
所述嵌合蛋白具有如下氨基酸序列:微生物来源的离子转运型受体视紫红质的氨基酸序列中的、细胞质侧的第2环路和/或细胞质侧的第3环路的氨基酸序列被置换为G蛋白偶联型受体视紫红质的细胞质侧的第2环路和/或细胞质侧的第3环路的氨基酸序列。
