Abstract
本发明公开了一种基于微藻的胃潴留生物漂浮给药系统及其制备方法和用途。该胃潴留生物漂浮给药系统由微藻、药物及凝胶材料组成,口服后能够在胃液中长时间漂浮,延长药物胃肠道的潴留时间并缓慢释放药物,充分发挥对胃肠道和肝脏的酒精损伤保护作用。此胃潴留生物漂浮给药系统还能实现营养物质补充和肠道炎症调节作用,口服安全性良好,制备过程简单易行,克服了现有生物制剂难以在胃部长时间滞留,载药及修饰等工艺复杂,难以维持生物活性,成本高难以大规模生产等问题,在胃肠道药物递送领域具有广阔的应用前景。

技术领域
本发明属于药物制剂技术领域,具体涉及一种用于胃肠道给药的胃潴留生物漂浮系统、制备方法及其在胃肠道和肝脏酒精损伤防护、营养补充、炎症调节方面的应用。
背景技术
口服给药被认为是首选的给药途径,因为它安全、无创、低成本、患者依从性高,以及与靶向或局部治疗(如胃疾病的治疗)相关的好处。然而,胃排空限制了药物在胃中的滞留和吸收,酸性环境和蛋白酶也可能影响药物的活性,特别是生物制品。为了将药物保留在胃中,已经使用了增加粘连、改变形状、使用微针和产生浮力的策略,例如地尔硫卓、维拉帕米等的“漂浮系统”。这些漂浮系统可以通过肿胀或产生气体来降低它们的密度,从而促进它们在胃液中漂浮。与其他策略相比,浮选系统在临床上已经得到了很好的发展和广泛的应用,但主要是作为片剂设计来携带药物和保持浮力,这对吞咽困难患者并不友好,片剂在胃内产生的肿胀或气体可能会引起消化不良,甚至加重胃部疾病。
近年来,各种微生物药物载体(如细菌、血细胞和微藻)因其特殊的生物学功能而引起了相当大的兴趣。然而,目前对微生物载体的研究大多集中在静脉或肠道给药,并且很少有大规模生产的报道。原因如下:1.胃排空使这些微生物系统难以留在胃内,酸性环境和蛋白酶可能会迅速破坏它们的生物活性;2.传统的载药工艺需要用高浓度的药物长时间培养微生物,可能会影响微藻的活性,特别是对于水溶性药物,需要有机溶剂,而有机溶剂大多有毒;3.微生物的特殊修饰或多步加工会增加成本、生产难度,降低微生物作为天然药物载体的优势。
专利(CN115364182A,CN115212189A,CN103505629B,CN114983951A)公开了几种口服胃肠道药物的制备方法及其应用,尽管实现了口服输送药物,但不可避免的存在一些胃肠道潴留时间短和口服安全性等问题,其中潴留时间短会造成药物难以在胃肠道组织缓慢释放及长效留存,特别是对于部分难溶性的药物,其佐剂大多含有有机试剂等带有一定毒性的化学品,且其制剂的安全性并未报道;另一方面,胃漂浮片虽然可以提高药物的生物利用度,但其漂浮过程会膨胀或产生气体,不适用于吞咽困难患者,还可能影响正常胃消化功能;此外,微球等口服制剂的制备过程较为复杂且成本高昂,不适合规模化生产及应用。然而,迄今为止,兼具药效持久、易于吞咽、副作用小等多项功能且成本低廉的新型胃潴留药物制剂或药物复合物仍没有被开发出来,因此,相关药物及制剂的研发已成为胃肠道药物递送问题的重中之重。
发明内容
针对现有技术的不足,本发明的目的在于,提供一种用于胃肠道给药的胃潴留生物漂浮系统、制备方法及其用途。该生物胃漂浮系统克服了现有胃滞留系统不适用于吞咽困难患者,影响正常胃消化功能等缺陷,同时克服了现有生物制剂难以在胃部长时间滞留,载药及修饰等工艺复杂,难以维持生物活性,成本高难以大规模生产等问题,实现了生物载体的低成本快速载药,维持载体生物活性,胃部长时间滞留及药物缓释,增强微藻及药物的胃肠道吸收及局部治疗效果。制备流程简单、可行性高、易规模化的;此胃漂浮给药系统还可以提供少量人体所需蛋白质、不饱和脂肪酸和微量元素。
本发明的第一个目的是提供了一种用于胃肠道给药的胃潴留生物漂浮系统,所述胃潴留生物漂浮给药系统包含天然微藻及需要负载的药物,以及凝胶材料增强其漂浮能力,其中,天然微藻和生物漂浮系统的尺寸均为微米级;所述系统中,药物与天然微藻之间通过空间结构,渗透压及静电吸附作用结合,所述的系统结合的检测方法为,在200-1350nm范围内吸收光谱扫描条件下,所述生物漂浮系统在440,625,680nm和对应药物的吸收峰处具有特征峰。
优选的,所述胃潴留生物漂浮给药系统还包含溶剂,胃潴留生物漂浮给药系统中,天然微藻的浓度为5mg/mL。
优选的,所述的溶剂选自无菌的磷酸盐缓冲液、超纯水、蒸馏水或生理盐水中的至少一种或多种。
优选的,所述的胃潴留生物漂浮给药系统具有胃肠道长效留存和缓慢降解性能,其中胃肠道长效留存和缓慢降解是指给药后3小时以上胃肠道内的荧光信号依然能够被检测到;优选的,其检测方法为:按照一定剂量给予禁食24小时的Balb/c白鼠灌胃,作用后荧光图像显示胃肠道内还含有药物复合物;优选的,所述的一定剂量为1-100mL/kg,更优选的,所述的一定剂量为10mL/kg;进一步,优选的,所述的作用的时间为48小时;更优选的,所述的荧光图像采用Cy5.5,激发波长605nm,发射波长615-665nm的叶绿素通道。
优选的,所述的胃潴留生物漂浮给药系统具有口服安全性,其检测方法为:按照一定剂量给予Balb/c白鼠灌胃,每日一次连续给药后,白鼠体重不变,血液学指标和肝肾功能显示正常;优选的,所述的一定剂量为1-100mL/kg,更优选的,所述的一定剂量为10mL/kg,进一步,优选的,所述的连续给药为23天。优选的,所述的胃潴留生物漂浮给药系统具有以下特征:
(1)外观呈现为黄绿色的混悬液;
(2)保留了天然微藻的生物活性;
(3)保留了天然微藻的漂浮能力;
(4)保留了天然微藻的荧光成像功能。
优选的,所述的天然微藻和药物的质量比为1:0.01-1:10。
优选的,所述的天然微藻和凝胶材料的质量比为1:0.1-1:10。

本发明的第二个目的是提供了一种基于微藻的胃潴留生物漂浮给药系统的制备方法,所述制备方法包括药物溶液、天然微藻及漂浮生物系统的制备;所述制备方法包括以下步骤:
(1).制备天然微藻和药物溶液:取微米级微藻培养离心弃去上清或使用筛网过滤去除培养基,洗涤沉淀后收集,得到新鲜天然微藻。
优选的,所述的离心的转速和时间分别为4500rpm和10min,筛网为200目。
优选的,所述的洗涤沉淀的溶液选自无菌的磷酸盐缓冲液、超纯水、蒸馏水或生理盐水中的至少一种。
优选的,所述的洗涤沉淀的次数为3-5次;
称取白藜芦醇固体,采用二甲基亚砜进行溶解,随后加水稀释得到重结晶后的白藜芦醇混悬液。
优选的,所述的稀释溶剂选自无菌的磷酸盐缓冲液、超纯水、蒸馏水或生理盐水中的至少一种。
优选的,所述的稀释溶剂与二甲基亚砜混合的体积比为1:2-1:100;

(2).制备生物漂浮系统药物复合物:在特定条件下,向白藜芦醇悬液中加入步骤(1)得到的天然微藻悬液,搅拌,然后离心并收集沉淀,洗涤3-5次,再加入果胶铋粉末,后处理得到药物复合物的悬液或固体粉末。

本发明的第三个目的是提供了一种药物组合物,所述药物组合物包含至少一种活性组分以及至少一种药学上可接受的添加剂,所述的活性组分选自上述基于微藻的胃潴留生物漂浮给药系统或上述制备方法得到的基于微藻的胃潴留生物漂浮给药系统中的任意一种。
优选的,所述的添加剂选自无菌的磷酸盐缓冲液、生理盐水、超纯水或蒸馏水中的任意一种或任意多种。
本发明的第四个目的是提供了上述基于微藻的胃潴留生物漂浮给药系统、上述制备方法获得的基于微藻的胃潴留生物漂浮给药系统在制备腹部疾病治疗相关药物的用途。

本发明的第五个目的是提供了上述药物组合物在胃肠道营养补充中用途。
优选的,所述的营养成分为微藻本身的成分,包括蛋白质、不饱和脂肪酸、类胡萝卜素、维生素,以及铁、碘、锌等多种微量元素或多糖等益生中的一种或多种。
本发明的第六个目的是提供了一种用于胃潴留药物递送及酒精所致损伤防护的口服制剂,所述口服制剂的活性成分为基于微藻的胃潴留生物漂浮给药系统、上述制备方法获得的基于微藻的胃潴留生物漂浮给药系统或药物组合物中的至少一种。
优选的,所述的制剂为液体制剂或固体制剂。
优选的,所述的液体制剂选为口服混悬剂。
优选的,所述的固体制剂选自片剂、散剂、颗粒剂、胶囊剂中的至少一种。
有益效果:
1、本发明首次制备了由天然微藻、药物和凝胶材料组成的胃潴留生物漂浮系统药物复合物,克服了传统药物难以在胃部长时间滞留的缺陷,接触胃液后能够长时间的漂浮于液面,并在添加凝胶材料后实现更长时间的漂浮与胃肠道潴留,应用后能保持较高的药物释放水平和口服安全性,可实现在胃肠道组织的长效留存和缓慢降解。
2、本发明打破了传统生物载体载药过程复杂的局限性,此药物复合物中的微藻能够与白藜芦醇形成空间网格状结构,无需长时间孵育,只需要简单搅拌即可得到稳定的结构,节约成本的同时维持了微藻自身的生物活性和漂浮能力。
3、本发明的药物复合物,是一种微米级生物胃漂浮系统,相比于其他生物载体集中在肠道和静脉给药中的应用,首次实现了利用生物载体进行胃部长时间潴留的药物递送,发挥了长效的胃肠道炎症调节作用;药物复合物在接触酒精后会加速释放白藜芦醇,实现酒精触发的控制释放;同时该药物复合物易于被消化系统降解,避免在体内的长时间滞留所带来的毒副作用。

具体实施方式
下文将结合以下结合附图和实施例进一步说明本发明,但本发明不局限于以下实施例。
实施例1.SP-RSV-Bi的合成
取培养于无菌条件的螺旋藻(SP)悬液,通过40目筛网的SP在200目筛网上被收集,并用磷酸盐缓冲液洗涤3次,去除残留培养基。将100mg白藜芦醇(RSV)粉末溶解在1mL二甲基亚砜(DMSO)中,然后迅速加入9mL超纯水,得到长条状RSV悬液。采用简单混合的方法合成SP-RSV-Bi,如下:将SP与RSV悬液直接混合,获得5mg/mL SP,2mg/mL RSV的悬液,SP与RSV之间形成空间网格结构,无需额外的孵育时间即可长期保持稳定。然后加入果胶铋粉末,使其浓度为4mg/mL,即制备得到SP-RSV-Bi。制备的SP-RSV-Bi作为悬液状态存在,但在与人工胃液(SGF)接触后会迅速形成凝胶。收集药物复合物,并拍摄显微镜及扫描电镜图片,可见药物复合物在液体(超纯水)中为均匀悬液,其形态为3D螺旋状微藻与长条状白藜芦醇构成的网格状结构,且具有红色荧光成像特性,在SGF中则形成片状凝胶结构(图1)。
实施例2.载药性能验证
应用吸收光谱仪和Zeta电位仪检测实施例1合成的药物复合物的吸收光谱和Zeta电位(图2),显示SP在440、625及680nm处具有特征峰,RSV在306nm处具有特征峰。SP-RSV的吸收光谱显示其兼具SP的特征峰(440、625及680nm)和RSV的特征峰(306nm);同时,SP、RSV、SP-RSV、SP-RSV-Bi和SGF中的SP-RSV-Bi的Zeta电位均发生变化。证明RSV被成功装载在SP中。
实施例3.体外漂浮能力检测
取不同含量果胶铋的药物复合物1mL(a:4mg/mLBi,5mg/mL SP,2mg/mL RSV,b:10mg/mLBi,5mg/mL SP,2mg/mL RSV,c:25mg/mLBi,5mg/mL SP,2mg/mL RSV),加入10mL人工胃液(SGF)中,分别于1,5和24小时后拍摄照片(图3)。体外漂浮实验证明,药物复合物具有良好的漂浮能力,即使长时间接触胃酸也能维持漂浮状态。
实施例1-3结果说明,药物在口服时为悬液态,适用于吞咽困难患者,在进入胃液后形成片状凝胶结构,漂浮于胃液上,从而能长时间潴留,但其漂浮过程不会膨胀或产生气体,不会影响正常胃消化功能,并且凝胶能避免胃液对微藻的侵蚀,维持其生物活性。
实施例4.体外药物释放性能检测
取实施例1合成的药物复合物500μL于透析袋中,加入500μL超纯水或人工胃液(SGF),分别置于超纯水和SGF中,在不同时间点(1、3、5、12、13、15和17h)采样,采用吸收光谱法测定RSV浓度(图4)。12h时,将溶剂换成含有50%乙醇的相应溶剂。结果表明RSV在SGF中能够缓慢被释放,同时当加入乙醇后会迅速被释放,3h内即可释放60%以上的RSV。可见,该药物可长时间潴留并且在饮入酒精时及时释放药物,快速保护胃肠道。
实施例5.口服后体内分布和胃部潴留
禁食24h后,Balb/c小鼠首先给予200μL PBS补充胃液,然后灌胃200μL SP-RSV-Bi,分别于灌胃后0.5、1、2、6、12、24和48h麻醉小鼠,并用活体成像仪(激发波长:445-490nm,发射波长:515-575nm)拍摄荧光图像,在注射后24和48小时截取胃部再次进行体外成像(图5)。在成像后取胃内容物和十二指肠,制备成扫描电镜样本,观察材料的形态(图6)。结果表明,口服后的材料荧光始终集中于腹部,可于0-12小时内保持较高荧光强度,有较长的胃肠道分布时间,有利于药物在胃肠道组织的浓度累积;胃部荧光图像(图5)和胃内容物的扫描电镜(图6)进一步表明了SP-RSV-Bi在胃部的潴留效果。
实施例6.药物复合物对急性腹部酒精性损伤的防护作用
将8周龄雌性Balb/c小鼠随机分为正常对照组、酒精组、RSV组、SP组、SP-RSV组、SP-RSV-Bi组(每组n=5只)。禁食24h后,各组小鼠首先给予10mL/kg PBS补充胃液,随后正常对照组、SP组、RSV组、SP-RSV组、SP-RSV-Bi组小鼠分别给予10mL/kg PBS、10mL/kg SP(5mg/mL)、10mL/kg RSV(2mg/mL)、10mL/kg SP-RSV(含5mg/mL SP,2mg/mL RSV)、10mL/kgSP-RSV-Bi(含5mg/mL SP,2mg/mL RSV,4mg/mL Bi)。1h后,除正常对照组外,其余各组小鼠均灌胃7mL/kg乙醇。灌胃3h后,动物安乐死,取胃部拍照并制备病理切片,进行免疫组化染色。结果显示,酒精组小鼠胃黏膜损伤明显,表现为大面积溃疡出血,而治疗组损伤面积有效减小(图7)。H&E和免疫组化染色结果进一步表明SP联合果胶铋更有效地减少急性酒精性损伤引起的胃粘膜上皮细胞损失、水肿、炎症细胞浸润及相关炎症因子(IL-1、IL-6、MPO)上调(图8)。
实施例7.药物复合物对慢性腹部酒精性损伤的防护作用
6周龄雌性Balb/c小鼠随机分为正常对照组、酒精组、RSV组、SP组、SP-RSV组、SP-RSV-Bi组(每组n=7只)。禁食24h后,正常对照组、SP组、RSV组、SP-RSV组、SP-RSV-Bi组小鼠首先分别给予10mL/kg PBS、10mL/kg SP(5mg/mL)、10mL/kg RSV(2mg/mL)、10mL/kg SP-RSV(含5mg/mL SP,2mg/mL RSV)、10mL/kg SP-RSV-Bi(含5mg/mL SP,2mg/mL RSV,4mg/mL Bi),1h后再给予4mL/kg乙醇,并称重,每24小时给药和酒精一次,连续23天。灌胃3h后,动物安乐死,切除胃、肠和肝脏组织,拍照并制备病理切片。在连续灌胃酒精过程中,小鼠体重明显下降,甚至死亡,而RSV与SP联合使用时,可有效维持小鼠体重(图9)。慢性小鼠胃内未见明显溃疡和出血,但病理切片仍可观察到胃黏膜上皮细胞缺失及炎症细胞浸润,经SP-RSV或SP-RSV-Bi治疗后得到有效缓解(图10)。慢性小鼠除胃部外,还表现出肠道损伤。灌胃SP-RSV或SP-RSV-Bi后,酒精诱导的结肠缩短和肠胀气得到抑制,十二指肠的再生隐窝细胞和肠绒毛也得到保护(图10和图11)。慢性饮酒也会引起肝脏损伤,经油红O染色后,在一些小鼠中观察到脂肪肝现象(图10),但在SP-RSV或SP-RSV-Bi治疗组中未观察到明显的肝脏病变。主要器官(心、脾、肺、肾)的病理检查进一步显示SP-RSV-Bi的生物安全性,通过微藻的保护胃肠道作用及缓释作用可有效避免白藜芦醇的潜在副作用,此外,肺部H&E染色显示SP-RSV和SP-RSV-Bi还可以减少酒精诱导的肺部炎症浸润(图12)。
实施例8.药物复合物对猪腹部酒精性损伤的防护作用
将1个月龄大的Yorkshire X Landrace仔猪随机分为正常对照组、酒精组、RSV组、SP组、SP-RSV组和SP-RSV-Bi组(每组n=5只)。禁食24h后,正常对照组、SP组、RSV组、SP-RSV组、SP-RSV-Bi组仔猪分别给予2mL/kg PBS补充胃液,然后再分别对应给予2mL/kg PBS、2mL/kg SP(5mg/mL)、2mL/kg RSV(2mg/mL)、2mL/kg SP-RSV(含5mg/mL SP,2mg/mL RSV)、2mL/kg SP-RSV-Bi(含5mg/mL SP,2mg/mL RSV,4mg/mL Bi)。1h后,除正常组外,其余仔猪均灌胃1.5mL/kg乙醇。灌胃3h后取心脏血样进行血常规和血生化分析,安乐死后取胃。与小鼠模型的结果相似,SP-RSV或SP-RSV-Bi具有最佳的治疗效果,进一步证明了该漂浮生物系统的应用潜力(图13)。此外,血常规和血生化指标显示SP-RSV或SP-RSV-Bi可抑制酒精诱导的WBC、AST、BUN、CREA、CHOL、TG、HDL-C、LDL-C水平上调,并且显示出良好的生物安全性(图14),通过微藻的保护胃肠道作用及缓释作用可有效避免白藜芦醇的潜在副作用。

Claims (10)
1.一种基于微藻的胃潴留生物漂浮给药系统,其特征在于,所述胃潴留生物漂浮给药系统包含天然微藻、药物和凝胶材料,其中,天然微藻和药物的尺寸均为微米级;所述胃潴留生物漂浮给药系统中,药物与天然微藻之间通过空间结构,渗透压及静电吸附作用结合,凝胶材料具有pH响应,在酸性条件下形成凝胶。
2.根据权利要求1所述的基于微藻的胃潴留生物漂浮给药系统,其特征在于,所述基于微藻的胃潴留生物漂浮给药系统还包含溶剂,所述的溶剂选自无菌的磷酸盐缓冲液、超纯水、蒸馏水或生理盐水中的至少一种或多种。
3.根据权利要求1所述的基于微藻的胃潴留生物漂浮给药系统,其特征在于,所述的天然微藻和药物的质量比为1:0.01-1:10,所述的天然微藻和凝胶材料的质量比为1:0.1-1:10。
4.根据权利要求1所述的基于微藻的胃潴留生物漂浮给药系统,其特征在于,所述的天然微藻为螺旋藻;所述的药物为白藜芦醇;所述的凝胶材料为果胶铋。
5.一种权利要求1-4任一项所述基于微藻的胃潴留生物漂浮给药系统的制备方法,其特征在于,所述制备方法包括以下步骤:
取微米级微藻培养去除培养基,洗涤沉淀后收集,得到新鲜天然微藻;
称取白藜芦醇固体,采用二甲基亚砜进行溶解,随后稀释得到重结晶后的白藜芦醇混悬液;
向白藜芦醇混悬液中加入天然微藻,搅拌,然后离心并收集沉淀,洗涤,再加入果胶铋粉末,后处理得到药物复合物的悬液或固体粉末。
6.根据权利要求5所述的制备方法,其特征在于,所述的后处理包括干燥或药物复合物混悬液制备中的至少一种。
7.权利要求1-4任一项所述的基于微藻的胃潴留生物漂浮给药系统的用途,其特征在于,包括:
(1)制备腹部疾病相关防护药物;
(2)制备胃肠道调节药物。
8.根据权利要求7所述的用途,其特征在于,所述的腹部疾病为酒精损伤。
9.根据权利要求7所述的用途,其特征在于,所述胃肠道调节药物包括胃肠道营养补充药物。
10.一种口服制剂,所述口服制剂的活性成分包括权利要求1-4任一项所述的基于微藻的胃潴留生物漂浮给药系统以及至少一种药学上可接受的添加剂;所述的制剂为液体制剂或固体制剂。
