癌症是严重威胁人类健康的疾病,癌症的早期诊断和治疗可以减轻患者痛苦和精神、经济负担,争取让癌症患者早日康复。目前,大多数用于诊断与治疗的靶向药物为单克隆片段,其肿瘤穿透能力差,不能特异地被单核巨噬细胞系统捕获。因此,靶向药物的研究一直是肿瘤诊断和治疗的热点。
1971年,Folkman最早提出肿瘤生长是血管依赖性的,肿瘤血管不仅可向肿瘤提供充足的营养,同时也是肿瘤细胞恶性生长和转移的重要条件。膜联蛋白A1(annexinA1,Anxal)是膜联蛋白超家族中的一员,它参与细胞信号转导、分化及凋亡等多种重要的生命过程。Anxa1是肿瘤新生血管特异的标志物,是EGFR的底物,可以特异性调节MAPK/ERK信号传导通路,同时作为PLA2的抑制剂等参与信号转导、细胞凋亡、钙离子通道的形成等众多生理过程,其在肿瘤组织及癌前病变中表达明显高于在相应的正常组织中的表达,其与肿瘤的发生、发展、诊断及治疗密切相关。
糖模拟肽(carbohydrate mimetic peptide,CMP)是一短序列肽分子,能模拟生物大分子或细胞表面复杂的糖结构,常用于替代糖分子进行诱导免疫反应制备特异性抗体。CMP分子小,血浆清除快,在药代动力学方面极具优势,备受研究者青睐,现在也多用于筛选受体或抗体的特异性高亲和力配体。如能筛选出一种与肿瘤新生血管特异性结合的CMP,将有较好应用前景。Shingo Hatakeyama等在研究糖模拟肽的过程中,发现了一个名为IFLLWQR的七肽片段(IF7),能有效靶向肿瘤血管,而且基本不引起免疫反应。
基于IF7制备的18F-AlF-p-SCN-NODA-IF7和18F-AlF-p-SCN-NOTA-IF7两种探针在U87脑胶质瘤荷瘤小鼠Micro-PET显像中显示肿瘤部位有显像剂浓聚,但是显像剂在荷瘤小鼠体内本底过高且靶向性不佳,在此基础上,在不影响IF7与肿瘤亲和力的前提下用Gly-Gly-Gly-Arg-Asp-Asn(GGGRDN)对IF7进行修饰改良,提高了IF7的水溶性。水溶性高的药物能够减少肝、胃、肠组织的摄取,降低肝肠循环,加快血液的清除,促进药物肾脏的代谢,可获得更佳质量的显像图像。用正电子(18F)和单光子(99mTc)核素分别对GGGRDN-IF7进行标记,成功合成了18F-AlF-NODA-GGGRDN-IF7和99mTc-DTPA-GGGRDN-IF7示踪剂,通过核医学显像,U87脑胶质瘤荷瘤小鼠的肿瘤部位有明显的放射性浓聚,提高了示踪剂的显像效果,但是肿瘤对示踪剂摄取值较低。
发明内容
鉴于此,本发明的目的在于提供一种双聚体多肽、正电子分子探针及制备方法和应用。本发明提供的多肽经放射性核素标记后,能够提高其在肿瘤中的摄取量,同时能够灵敏的、快速的与肿瘤特异性结合,对肿瘤进行显像。
为了实现上述发明目的,本发明提供了一种技术方案。
本发明提供了一种双聚体多肽,具有如序列表Seq ID No.1所示的氨基酸序列。
本发明提供了一种正电子分子探针,具有X-N-G所示的结构;其中X为正电子显像核素,N为双功能螯合剂,G为权利要求1所述的双聚体多肽。
优选地,所述双功能螯合剂为与异硫氰基定位结合和/或与羟基定位结合的双功能螯合剂。
优选地,所述双功能螯合剂为p-SCN-Bn-NOTA、p-SCN-Bn-NODA、p-SCN-Bn-DOTA、DOTA-NHS、p-SCN-Bn-DTPA和p-SCN-Bn-NOTA中的一种或几种。
本发明提供了上述所述单光子或正电子分子探针的制备方法,包括以下步骤:
(1)将双功能螯合剂的溶液和双聚体多肽的溶液混合后,进行螯合反应,得到标记前体;
(2)将含标记前体的溶液与正电子显像核素溶液混合,进行螯合反应后,分离纯化,得到所述正电子分子探针。
优选地,所述双功能螯合剂的溶液和双聚体多肽的溶液的溶剂独立地为乙酸铵溶液、水、乙醇、磷酸盐缓冲液或二甲亚砜。
优选地,所述含标记前体的溶液中还包括AlCl3。
优选地,所述标记前体和AlCl3的摩尔比为1:1-3。
本发明还提供了所述正电子分子探针或所述的制备方法制备得到的正电子分子探针作为分子影像示踪剂的应用。
本发明提供的双聚体多肽,具有如序列表Seq ID No.1所示的氨基酸序列,即将两分子GGGRDN-IF7通过赖氨酸连接,形成的GGGRDN-IF7肽二聚体。该多肽经放射性核素标记后,能够提高其在肿瘤中的摄取量。
本发明还提供了一种正电子分子探针,具有X-N-G所示的结构;其中X为正电子显像核素,N为双功能螯合剂,G为上述技术方案所述的多肽,本发明以正电子核素进行标记,制备了靶向Anxal的分子探针。阻断实验证实此类显像剂与Anxal特异性结合,经正电子断层扫描显像,肿瘤可清晰显影,并进行肿瘤Anxal表达的定量分析。为人类检测及诊治肿瘤病症提供了新的医疗途径。
本发明提供了正电子分子探针的制备方法,包括以下步骤:(1)将双功能螯合剂的溶液和双聚体多肽的溶液混合后,进行螯合反应,得到标记前体;
(2)将含标记前体的溶液与正电子显像核素溶液混合,进行螯合反应后,分离纯化,得到所述正电子分子探针。本发明提供的制备工艺简单、操作方便,耗时短、标记率高,标记物稳定,便于临床、科研及药物开发的进一步应用。
本发明还提供了所述正电子分子探针作为分子影像显像剂的应用。荷瘤鼠microPET显像表明,肿瘤清晰可见,肿瘤与背景对比度高。尾静脉注射1h后,肿瘤与正常肌肉组织对所述正电子分子探针的摄取比值高,提高所述正电子分子探针与肿瘤高表达的Anxal特异性结合。阻断实验显示,在未标记Anxal靶向肽阻断下,肿瘤中放射性浓聚显著降低,该结果证实所述正电子分子探针与Anxal特异性结合。由于正电子分子探针与肿瘤血管内皮细胞高度表达的Anxal特异性结合且放射性核素标记多肽具有组织渗透迅速的优点,因此本发明所述正电子分子探针可以灵敏的、快速的与肿瘤特异性结合,进而可以快速的对肿瘤进行显像,有利于对肿瘤的诊断和分析,进而有利于肿瘤的及早发现。

具体实施方式
本发明提供了一种双聚体多肽,具有如序列表Seq ID No.1所示的氨基酸序列。
在本发明中,所述多肽由GGGRDNIFLLWQR通过赖氨酸连接实现双聚体化得到。在本发明中,所述多肽为委托上海楚肽生物科技有限公司合成。
本发明还提供了一种正电子分子探针,具有X-N-G所示的结构;其中X为正电子显像核素,N为双功能螯合剂,G为上述所述的双聚体多肽。
在本发明中,所述双功能螯合剂优选为与异硫氰基定位结合和/或与羟基定位结合的双功能螯合剂,更优选为p-SCN-Bn-NOTA、p-SCN-Bn-NODA、p-SCN-Bn-DOTA、DOTA-NHS、p-SCN-Bn-DTPA和p-SCN-Bn-NOTA中的一种或几种,进一步优选为p-SCN-Bn-NOTA。
本发明还提供了所述正电子分子探针的制备方法,包括以下步骤:
(1)将双功能螯合剂的溶液和双聚体多肽的溶液混合后,进行螯合反应,得到标记前体;

在本发明中,所述双功能螯合剂的溶液和双聚体多肽的溶液中溶剂独立地优选为乙酸铵溶液、水、乙醇、磷酸盐缓冲液或二甲亚砜(DMSO)。在本发明中,所述双功能螯合剂的溶液和和双聚体多肽的溶液的pH值独立地优选为4~6。
在本发明中,所述双功能螯合剂和双聚体多肽的摩尔比优选为1.3:1。
在本发明中,所述步骤(1)中,所述螯合反应优选为:将双功能螯合剂的溶液和双聚体多肽的溶液混合后,水浴加热至37℃搅拌过夜即得。
得到标记前体后,本发明将含标记前体的溶液与正电子显像核素溶液混合,进行螯合反应后,分离纯化,得到所述正电子分子探针。
在本发明中,所述含标记前体的溶液中溶剂优选为极性溶剂;所述极性溶剂优选为乙腈或水;所述含标记前体的溶液的pH值优选为4~6。
在本发明中,所述pH值的调节试剂优选为醋酸。在本发明中,所述含标记前体的溶液中还包括AlCl3,优选为AlCl3。在本发明中,当含标记前体的溶液中含有AlCl3时,所述标记前体和AlCl3的摩尔比为1:1-3。
在本发明中,双功能螯合剂上的配位原子能够与金属Al3+形成稳定配合物,而Al3+与溶液中F-有着很强的络合力。所以通过加入AlCl3溶液达到间接标记18F的目的。
在本发明中,所述正电子显像核素溶液中放射性核素优选为18F。在本发明中,所述正离子显像核素溶液优选由加速器生产得到。
在本发明中,所述步骤(2)中螯合反应优选在水浴加热及搅拌条件下进行。在本发明中,所述步骤(2)中,螯合反应的温度优选为50~100℃,更优选为80℃,时间优选为10~20min,,更优选为15min。
在本发明中,所述步骤(2)中螯合反应后,还包括将螯合反应体系冷却至室温。
在本发明中,所述分离纯化的固相萃取柱优选为C18柱,更优选为Sep-Pak C18柱。在本发明中,所述分离纯化的步骤优选包括依次进行的活化、上样、淋洗和洗脱;所述活化优选为依次采用无水乙醇活化和水活化;所述淋洗优选包括依次进行0.01mol/LPBS溶液淋洗和水淋洗;所述洗脱的洗脱剂优选为盐酸乙醇;所述盐酸乙醇中盐酸的浓度优选为15mol/L。在本发明中,所述洗脱后,优选还包括将洗脱液用生理盐水稀释后无菌过滤。
本发明所述的标记方法选用适宜的螯合剂或标记辅基进行正电子现象核素标记,所述方法简单快捷,标记率和放化纯均可达到满意效果,且适用于医用检测之用,便于临床推广。
本发明还提供了所述正电子分子探针作为分子影像示踪剂的应用。
下面将结合本发明中的实施例,对本发明中的技术方案进行清楚、完整地描述。显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1
本实施例所述的PET示踪剂是以双功能螯合剂p-SCN-Bn-NOTA为连接基对具有氨基酸序列GGGRDNIFLLWQR(GR13)的多肽化合物进行正电子核素18F标记,具体步骤包括:
1)标记前体p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys的制备:
将5.57mg(GGGRDN-IF7)2-Lys与1.0mg p-SCN-Bn-NOTA(摩尔比1:1.3)溶于1mLDMSO,氮气封闭后37℃水浴搅拌过夜。反应结束后,使用制备型HPLC纯化,条件如下:
制备型反相C18柱(Xbridge,19×150mm,Waters);Waters 2998二级管阵列紫外检测器;BioScan放射性检测器;Waters2545二元高压梯度泵;流动相为:A、含0.1%三氟乙酸(TFA)的乙腈溶液;B、含0.1%TFA的水溶液;
梯度洗脱:0~2分钟的5%A和95%B匀速增加到21分钟的65%A和35%B;
流速为20mL/min,检测波长为218nm。收集出峰时间为18.5min时的流份,获得标记前体。
通过分析型HPLC检测产品纯度,所述分析型HPLC的检测条件为:
反相C18柱(Luna,4.6×250mm,phenomenex);Waters1525二元HPLC液相泵;Radiomatic 610TR放射性检测器(Perkin-Elmer);Waters 2998双波长紫外检测器;流动相为:A、含0.1%三氟乙酸(TFA)的乙腈溶液;B、含0.1%TFA的水溶液;梯度洗脱:0~2分钟的5%A和95%B增加到32分钟的65%A和35%B;流速为1mL/min,检测波长为218nm;
产品纯度>95%。LC-MS:[MH]+=3713.11(m/z),这与标记前体即p-SCN-NOTA-(GGGRDN-IF7)2-Lys(C166H248N50O46S:3712.17)的计算值相吻合。
所述标记前体p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys的结构式如下:
2)18F溶液的制备:医用加速器中采用质子速轰击H2 18O得到18F溶液。采用CRC-15R活度计(CAPINTEC)测定其放射剂量为20mCi。
3)PET示踪剂18F-AlF-p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys的制备:将40μg标记前体用200μL DMSO溶解,加入6μL的2mM的AlCl3溶液和10μL的浓度为5wt%的醋酸,充分混匀后加入100μL20mCi前述新鲜制得的18F溶液,100℃反应10min。冷却,并加15mL水稀释后注入C18分离小柱(Sep-Pak C18柱),用5mL水冲洗柱子,每次冲洗后测C18柱的放射性活度,待C18柱的放射性活度变动小于3.7MBq(100μCi),用300μL 15mol/L盐酸乙醇洗脱标记产品,生理盐水稀释后无菌过滤,即得所述PET示踪剂,合成时间约20min,采用CRC-15R活度计(CAPINTEC)测定其放射剂量为11.6mCi,因此,计算其标记率(未校正)为58%。
采用分析型HPLC的检测条件对示踪剂进行检测,产品保留时间:18.5分钟(HPLC图谱见图1所示),放射化学纯度>97%。
本发明还对18F-AlF-p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys进行了血浆稳定性测试:
测试方法为:将100μCi18F-AlF-p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys与健康人血浆在37℃共同孵育10min、30min、1h、2h后,分别取50μL反应液过无菌滤膜后,采用放射高效液相色谱法测定18F-AlF-p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys在健康人血浆中10min、30min、1h、2h的体外稳定性。测试结果见图2,从图2可知:在人血清中稳定性良好,2h后出峰时间及峰型无明显改变,2h后放化纯仍>95%。
PET示踪剂18F-AlF-p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys的结构式及合成路线图如图7所示。
4)18F-AlF-p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys的油水分配系数:
将18F-AlF-p-SCN-Bn-NODA-GGGRDN-IF7用PBS稀释成37kBq(1μCi)/mL,各取0.5mL分别置于3个离心管中,每管中加入0.5mL的正辛醇并混合均匀,室温下涡旋2min,18000g离心5min。分别取上层(油层)100μL,下层(水层)100μL,置入6只计数管中,称重后于γ计数仪测定每管的放射性活度(CPM值)并分别计算油相与水相的平均CPM值,计算油水分配系数(log P)值。最终计算logP=0.21±0.04,表面所述PET示踪剂具有良好的亲水性。
5)18F-AlF-p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys的体外实验:在细胞摄取实验中,细胞在24孔板中播种过夜(每孔4×105)。用PBS(pH 7.4,0.01M)洗涤细胞3次,然后用不含胎牛血清的新鲜培养基替换。
在1mL培养基中加入370kBq(10μCi)的18F-AlF-p-SCN-NOTA-(GGGRDN-IF7)2-Lys,在37℃、5%CO2细胞培养箱中孵育10、30、60和120min。阻断组加入非放射性标记前体(1μg/孔),置于37℃、5%CO2细胞培养箱中30min之后,重复上述步骤,再用200μL NaOH(0.1M)裂解,收集裂解液,用γ计数器测定每管放射性计数(CPM值),同时测定标准源的总放射性计数(CPM值)。最终测得在U87细胞中,18F-AlF-p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys的细胞摄取随着时间的推移逐渐增加,在120min时达到3.69±0.12%AD。通过添加未标记的(GGGRDN-IF7)2-Lys,可以成功阻断肿瘤细胞对18F-AlF-p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys的摄取,在120min时肿瘤细胞摄取值为0.78±0.05%AD(如图3)。
竞争结合实验中,将U87细胞以4×105/L培养于24孔板中分为12组,每组设4平行孔,37℃、5%CO2的细胞培养箱过夜。弃去培养液后用PBS冲洗3次,将不同浓度的用DMEM培养基稀释的未标记前体(0.1nM至10μM)添加到各孔板中,再加入74kBq(2μCi)放射性标记18F-AlF-p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys于37℃孵育2h,PBS(pH 7.4,0.01M)洗涤3次,再用200μLNaOH(0.1M)裂解,收集裂解液,用γ计数仪计数。每种浓度重复实验3次。在GraphPad Prism 9.4.1软件(GraphPad software,San Diego,CA,USA)中用非线性回归法拟合曲线,计算IC50。18F-AlF-p-SCN-NOTA-(GGGRDN-IF7)2-Lys的IC50计算值为75.1nM(如图4)。
MicroPET显像
取荷人移植瘤(U87)模型鼠置于小动物PET床板上,异氟烷麻醉模型鼠,经胶带固定。模型鼠经尾静脉注射上述PET示踪剂的生理盐水溶液(1.85MBq,0.2mL)。注射后30分钟、60分钟、90分钟、120分钟,分别进行microPET显像,结果分别如图5所示。图中箭头所示为肿瘤。图中颜色由明到暗表示所述PET示踪剂摄取值由高到低。
采用二维有序子集期望最大化算法进行图像重建。在肿瘤、肌肉、肝等器官用感兴趣区(ROI)法计算放射性活度(MBq/mL),所得值除以注射剂量获得各组织对PET示踪剂摄取值(%ID/g)(假定组织密度为1g/mL)。计算结果分别如图5所示。如图5所示,注射上述PET示踪剂后30min到90min,各移植瘤均清晰可见,从图中可以看到,肿瘤与对侧相比具有良好的对比度。
所述PET示踪剂在模型鼠的肝肾中具有显著的浓聚,这表明其主要通过肝肾代谢。随着时间的延长,所述PET示踪剂在模型鼠体内的浓聚逐渐降低,显示所述PET示踪剂逐渐代谢排出体外,这表明所述PET示踪剂是安全的,不会持久的存于鼠体内,且半衰期较短,不会对模型鼠造成伤害。
如图5所示,注射后30min,肿瘤对所述PET示踪剂摄取值达最高约4.09±1.13%ID/g,随着时间的变化,摄取值逐渐降低。肾对所述PET示踪剂摄取值显著,证实所述PET示踪剂主要通过肾代谢。此外,如图4所示,肿瘤较正常组织如肌肉对所述PET示踪剂摄取显著,便于肿瘤的诊断和治疗。120min后骨中的摄取低,说明标记物在体内稳定,无脱18F的现象,此标记物可能成为一种良好的肿瘤显像剂。
特异性实验:
荷U87神经胶质瘤模型鼠预先注射未标记的靶向Anxal多肽化合物即p-SCN-Bn-NOTA-(GGGRDN-IF7)2-Lys(10mg/kg体重)30min后,然后注入所述PET示踪剂,60min后进行microPET显像。
由图5中可见,注射后60min,阻断组模型鼠移植瘤对PET示踪剂的摄取值较未阻断组显著下降约80%。这是由于未标记的靶向Anxal的多肽化合物,有效抑制肿瘤高表达的Anxa1与所述PET示踪剂的结合,导致肿瘤对所述PET示踪剂的摄取值相应降低。实验证实,所述PET示踪剂与Anxa1特异性结合。
18F-AlF-p-SCN-NOTA-(GGGRDN-IF7)2-Lys在小鼠的体内的生物分布:按照本实施例制备好标记率大于95%的18F-AlF-p-SCN-NOTA-(GGGRDN-IF7)2-Lys溶液。将12只U87荷瘤鼠随机分成4组,每组3只。经小鼠从尾静脉注射0.2mL(0.74MBq)标记化合物,分别于30min、60min、120min以及60min阻断后处死小鼠,收集肿瘤、血、心、肝、脾、肺、肾、胃、肠、肌肉、骨,称重后用γ计数器测放射性计数,并计算各脏器和组织的摄取量(%ID/g),实验结果以表示,结果显示,18F-AlF-p-SCN-NOTA-(GGGRDN-IF7)2-Lys注射后30min,肝肾有明显的放射性浓聚,120min后大部分消除(如图6和表1所示)。

以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。

1.一种双聚体多肽,其特征在于,具有如序列表Seq ID No.1所示的氨基酸序列。
2.一种正电子分子探针,其特征在于,具有X-N-G所示的结构;其中X为正电子显像核素,N为双功能螯合剂,G为权利要求1所述的双聚体多肽。
3.根据权利要求2所述的正电子分子探针,其特征在于,所述双功能螯合剂为与异硫氰基定位结合和/或与羟基定位结合的双功能螯合剂。
4.根据权利要求2所述的正电子分子探针,其特征在于,所述双功能螯合剂为p-SCN-Bn-NOTA、p-SCN-Bn-NODA、p-SCN-Bn-DOTA、DOTA-NHS、p-SCN-Bn-DTPA和p-SCN-Bn-NOTA中的一种或几种。
5.权利要求2~4任一项所述正电子分子探针的制备方法,其特征在于,包括以下步骤:
(1)将双功能螯合剂的溶液和双聚体多肽的溶液混合后,进行螯合反应,得到标记前体;
(2)将含标记前体的溶液与正电子显像核素溶液混合,进行螯合反应后,分离纯化,得到所述正电子分子探针。
6.根据权利要求5所述的制备方法,其特征在于,所述双功能螯合剂的溶液和双聚体多肽的溶液的溶剂独立地为乙酸铵溶液、水、乙醇、磷酸盐缓冲液或二甲亚砜。
7.根据权利要求5所述的制备方法,其特征在于,所述含标记前体的溶液中还包括AlCl3。
8.根据权利要求7所述的制备方法,其特征在于,所述标记前体和AlCl3的摩尔比为1:1-3。
