技术领域
本发明涉及桃红四物汤颗粒制备技术领域,具体为一种桃红四物汤颗粒及其制备方法。
背景技术
桃红四物汤以祛瘀为核心,辅以养血、行气。方中以强劲的破血之品桃仁、红花为主,力主活血化瘀;以甘温之熟地、当归滋阴补肝、养血调经;芍药养血和营,以增补血之力;川芎活血行气、调畅气血,以助活血之功。全方配伍得当,使瘀血祛、新血生、气机畅,化瘀生新是该方的显著特点。
桃红四物汤为调经要方之一,是《玉机微义》转引的《医垒元戎》中的一个方子,也称加味四物汤,桃红四物汤这一方名始于见《医宗金鉴》。该方由四物汤加味桃仁、红花而成,功效为养血活血。现代研究表明,桃红四物汤具有扩张血管、抗炎、抗疲劳、抗休克、调节免疫功能、降脂、补充微量元素、抗过敏等作用。
长期以来,由于汤剂因其易吸收、起效快、疗效好的特点,一直是中医临床的主要用药形式,历代医家在使用桃红四物汤时,也大多采用汤剂,但汤剂存在服用剂量大,携带和运输不便,药效不稳定,不易保存等缺点,改制成颗粒剂,不仅减少了患者的服药剂量,增加了药品的稳定性,且有利于携带和运输。
随着现代研究手段的提升,对其药理、药效及化学成分的研究已十分迅速;然而传统工艺步骤繁杂、耗费时间长;需大量使用易燃的有机溶剂,较长时间的加热易造成热敏性成分的降解且残留在提取物中的有机溶剂很难较彻底的去除,本项目旨在研究对桃红四物汤有效成分提取完全,开发出产品纯度高、质量好的桃红四物汤颗粒。
发明内容
(一)解决的技术问题
针对现有技术的不足,本发明提供了一种桃红四物汤颗粒及其制备方法,具备制作的桃红四物汤颗粒的药材中有效成分提取完全,制作的桃红四物汤颗的纯度高、质量好,并且在整个制作的过程中对药材本身的选择和处理、设备的选择和整个工艺流程设计均有助于对高质量桃红四物汤颗粒的制作等优点,解决了传统工艺步骤繁杂、耗费时间长;需大量使用易燃的有机溶剂,较长时间的加热易造成热敏性成分的降解且残留在提取物中的有机溶剂很难较彻底的去除的问题。

为实现上述制作的桃红四物汤颗粒的药材中有效成分提取完全,制作的桃红四物汤颗的纯度高、质量好,并且在整个制作的过程中对药材本身的选择和处理、设备的选择和整个工艺流程设计均有助于对高质量桃红四物汤颗粒的制作目的,本发明提供如下技术方案:
一种桃红四物汤颗粒,包括以下原料:
燀山桃仁、酒红花、酒地黄、酒当归、川芎和酒白芍。
优选的,所述酒红花在制作时,每100kg红花饮片,用黄酒10kg,闷润至黄酒被吸尽;酒地黄在制作时,每100kg的黄饮片,用黄酒12kg,闷润至透;酒当归在制作时,每100kg当归饮片,用黄酒10kg,闷润至透;酒白芍在制作时,每100kg白芍饮片,用黄酒15kg,闷润至黄酒被吸尽。

S1、净选
将准备的药材进行挑选整理,去除药材(含饮片)中的杂质和非药用部位;
S2、清洗
将净选后的药材分别进行清洗,直至药材表面无泥沙和杂质,将清洗后的药材沥干水分;
S3、润药
将洗净的白芍放入到蒸锅中,设定软化温度80℃,将白芍蒸软即得,将软化好的药材做好状态标识;
S4、炮制
将洗净后的山桃仁放入到煎煮容器中进行煎煮,将煎煮后的山桃仁捞出,放入冷水中稍浸,取出后除去种皮;
S5、切制
分别对处理后的酒地黄、酒当归、川芎和酒白芍进行切片操作;
S6、干燥
将待干燥物料装盘,推入热风循环烘箱中进行干燥,将干燥好的药材做好状态标识;
S7、炒制
将待炒制的药材置于炒药机内进行炒制,将炒制好的药材盛装好,做好状态标识;
S8、提取
将酒地黄、酒当归、酒白芍、川芎、酒红花和燀山桃仁投入6000L提取罐内进行煎煮,将煎煮后的提取液进行过滤;
S9、浓缩
将滤液按边进料边浓缩的方式抽入单效外循环浓缩器内,浓缩成浸膏,将浓缩后的浸膏进行过滤;
S10、喷雾制粒
将沸腾干燥机预热,分两锅次进行制粒,将1锅次麦芽糊精置于料斗内,进行预热,使麦芽糊精温度达到制粒要求温度,设定进风温度、出风温度及设定物料温度,控制颗粒水分≤8.0%;
S11、整粒
操作人员将干颗粒置入三元旋振筛内进行筛分,将颗粒通过14目、65目筛网进行筛分,将合格品置入洁净容器中或混合机料仓内备用;
S12、总混
将整粒后的颗粒加入自动提升料斗混合机内进行混合,设定转速10rpm,混合20分钟;
S13、内包装
将待包装的颗粒置自动充填包装机进料斗内,设置前横封辊温度、后横封辊温度、前纵封辊温度、后纵封辊温度和打字加热温度,温度达到稳定值后,将合格颗粒由真空上料机抽入自动充填包装机斗内,进行分装。
优选的,所述步骤S4中,在对山桃仁进行炮制时,将山桃仁放入到10倍量的沸水中烫约6-8分钟。
优选的,所述步骤S4中,将烫后的山桃仁放入到温度为10-15℃的冷水中,当冷水没过上层山桃仁后将其取出。
优选的,所述步骤S5中,在对药材进行切片时,将酒地黄和酒当归以及酒白芍切成1~2mm的薄片,将川芎切成2-4mm的厚片。
优选的,所述步骤S6中,在对药材进行干燥时,酒地黄和川芎以及冷浸后的山桃仁干燥时的温度应设置在60℃,酒当归和酒白芍干燥时的温度应设置在50-60℃。
优选的,所述步骤S7中,酒红花在炒制时,炒制温度为80~95℃,将酒红花炒干时,取出放凉,筛去药屑;酒地黄在炒制时,炒制温度为105~120℃,炒至微焦,取出放凉;酒当归在炒制时,炒制温度为105~120℃,将酒当归炒干,取出放凉;酒白芍在炒制时,炒制温度95~105℃,炒至表面呈微黄色,取出放凉,筛去药屑。
优选的,所述步骤S8中,在对药材提取时,第一次加10倍量饮用水,煎煮25-35分钟,提取液150目筛过滤,第一次提取的药渣进行第二次提取,加入8倍量饮用水,煎煮25-35分钟;提取液150目筛过滤,合并两次滤液。
优选的,所述步骤S9中,在对提取液进行浓缩时,单效外循环浓缩器中控制真空度-0.06Mpa~-0.1Mpa、温度<90℃,浓缩至相对密度约为1.05~1.15(60℃)停止。

在本发明中,使用自动化控制提取浓缩装置,通过电磁阀的开启,依托压缩空气提供的动力,自动控制蒸汽、压缩空气、真空、水系统的阀门开关状态,实现生产过程中物料自动化处理,最大限度的减少人员混淆差错对物料的影响,提高生产效率效能。
该产品质量控制过程中,采用指纹图谱与多指标含量(芍药苷、苦杏仁苷、羟基红花黄色素A)等检测性指标进行产品开发及质量控制。

燀山桃仁、酒红花、酒地黄、酒当归、川芎和酒白芍。
2.根据权利要求1所述的一种桃红四物汤颗粒,其特征在于,所述酒红花在制作时,每100kg红花饮片,用黄酒10kg,闷润至黄酒被吸尽;酒地黄在制作时,每100kg的黄饮片,用黄酒12kg,闷润至透;酒当归在制作时,每100kg当归饮片,用黄酒10kg,闷润至透;酒白芍在制作时,每100kg白芍饮片,用黄酒15kg,闷润至黄酒被吸尽。

S10、喷雾制粒
将沸腾干燥机预热,分两锅次进行制粒,将1锅次麦芽糊精置于料斗内,进行预热,使麦芽糊精温度达到制粒要求温度,设定进风温度、出风温度及设定物料温度,控制颗粒水分≤8.0%;

6.根据权利要求1所述的一种桃红四物汤颗粒的制备方法,其特征在于,所述步骤S5中,在对药材进行切片时,将酒地黄和酒当归以及酒白芍切成1~2mm的薄片,将川芎切成2-4mm的厚片。
7.根据权利要求1所述的一种桃红四物汤颗粒的制备方法,其特征在于,所述步骤S6中,在对药材进行干燥时,酒地黄和川芎以及冷浸后的山桃仁干燥时的温度应设置在60℃,酒当归和酒白芍干燥时的温度应设置在50-60℃。
