Abstract
本发明涉及免疫制剂技术领域,提供了一种RNA疫苗递送制剂及其制备方法。本发明将可电离脂质或阳离子脂质、磷脂辅助脂质、甾醇、磷脂聚乙二醇衍生物和有机溶剂混合,得到有机相;将编码目标蛋白的RNA、海藻糖和水混合,得到水相;两相混合后去除有机溶剂,得到RNA疫苗递送制剂的悬浮液形式;后续再经冻干,得到RNA疫苗递送制剂的冻干粉形式。本发明以脂质与海藻糖的混合物为载体构建脂质纳米粒(LNPs),使LNPs内部和外部均存在海藻糖,所得RNA疫苗递送制剂包封能力强,RNA的体外转染和体内表达效率高,并且在后续的冻干过程中,LNPs内外部的海藻糖皆有助于保护LNPs的完整性和稳定性,所得冻干粉稳定性更好。

技术领域
本发明涉及免疫制剂技术领域,尤其涉及一种RNA疫苗递送制剂及其制备方法。
背景技术
病毒传染性极强,对公众健康造成威胁,疫苗接种是预防传染病最有效的措施。目前,常见的疫苗主要有mRNA疫苗、灭活疫苗以及腺病毒疫苗等,其中灭活疫苗技术最为成熟,腺病毒疫苗仅需接种一次,但二者开发和生产周期均较长,至少需要10~12个月。mRNA疫苗基于体外转录技术,只需测序病毒基因组,利用病毒的基因序列而不是病毒本身,因此,mRNA疫苗具有不带有病毒成分,没有感染风险的特点。并且研发周期短,能够迅速根据变异株的突变基因更新mRNA序列,可以在几周内快速设计和生产临床规模的mRNA疫苗,是一种高效便捷的防疫手段。
脂质纳米颗粒(LNPs)是现阶段mRNA疫苗的主要递送体系,LNPs包覆mRNA使其不被RNA内切或外切酶降解,并且其类似于细胞膜的结构使其易于与受体细胞融合,促进细胞摄取及表达。Pfizer和Moderna研制的mRNA疫苗利用LNPs的多种独特的理化性质,通过可电离的氨基来增强疫苗的mRNA负载率,并促进疫苗的溶酶体逃逸;通过调控可电离脂质或阳离子脂质比例,实现了基于表面电荷及尺寸大小的组织特异性mRNA递送,而且这些特性还协同提高了疫苗的免疫原性,可以更为有效的激活免疫反应。
mRNA疫苗在常温下不稳定、容易降解,超低温的储存条件使得其储存和分配复杂化,目前以冻干粉的形式储存和运输是mRNA药物最常用的方法。然而,冷冻干燥过程中结晶和真空脱水引起的应力可能会使LNPs的结构发生改变,导致活性丧失,因此制剂中常加入冻干保护剂来保持其稳定性。
合适的冻干保护剂需要具备以下四个特性:玻璃化转变温度高、吸水性差、结晶率低以及不含还原基。在生物药品的冷冻干燥配方中,蔗糖和海藻糖是最常用的两种保护剂。既能在冷冻过程中充当低温保护剂,又能在干燥脱水过程中起到脱水保护剂的作用,而与蔗糖相比,海藻糖具有更高的玻璃化相变温度(120 ℃),因此海藻糖溶液更不容易形成冰晶,能使脱水状态下的膜脂保持液晶状态,并减少脂质纳米颗粒之间的亲和力,避免聚集,使脂质纳米颗粒更均匀地分散。此外,海藻糖具有很强的水合能力,生物膜表面结合水的存在对于维持膜的稳定性和完整性至关重要,海藻糖含有多个羟基,它能够替代水分子与磷脂头部形成氢键结合,填补失去的结合水位点,保持生物膜表面的水化状态,从而达到保护作用。
目前,海藻糖主要是作为赋形剂和冻干保护剂,在冻干之前与mRNA-LNPs混悬液混合,通过搅拌使海藻糖均匀分散在混悬液中,然后进行后续的冻干流程。但是,LNPs的内部也含有水分,在冻干过程中LNPs内部包载的mRNA也会与内部水产生的结晶发生相互作用,进而导致部分mRNA活性丧失。因此,这种方法所得的制剂稳定性也有待进一步提高。
发明内容
有鉴于此,本发明提供了一种RNA疫苗递送制剂及其制备方法。本发明提供的制备方法能够使海藻糖部分包裹进脂质纳米颗粒的内部,此部分海藻糖替代了脂质纳米粒内部部分水的空间位置,因此在冻干过程中减少了mRNA与纳米粒内部水产生的结晶的相互作用,减少了冻干过程对mRNA的破坏,从而提高冻干粉制剂的稳定性;同时,所得RNA疫苗递送制剂可以在将mRNA递送至胞内的同时也将部分海藻糖递送进胞内,而海藻糖会降低胞内由LNPs导致的细胞毒性,减少胞内氧化应激状态,从而使体外转染和体内表达效率更高。
为了实现上述发明目的,本发明提供以下技术方案:
一种RNA疫苗递送制剂的制备方法,包括以下步骤:
将脂质、磷脂辅助脂质、甾醇、磷脂聚乙二醇衍生物和有机溶剂混合,得到有机相;所述脂质为电离脂质或阳离子脂质;
将编码目标蛋白的RNA、海藻糖和水混合,得到水相;
将所述有机相加入所述水相中,去除所得混合料液中的有机溶剂,得到RNA疫苗递送制剂。
优选的,所述可电离脂质为DLin-MC3-DMA、SM-102和ALC-0315中的一种;所述阳离子脂质为DOTAP、DOTMA、DIMRIE和DOTIM中的一种;
所述磷脂辅助脂质为二硬脂酰磷脂酰胆碱;
所述甾醇为胆固醇和植物甾醇中的一种或多种;
所述磷脂聚乙二醇衍生物为1,2-二肉豆蔻酰-rac-甘油-3-甲氧基聚乙二醇。
优选的,所述植物甾醇包括β-谷甾醇、豆甾醇和麦角甾醇中的一种或多种。
优选的,所述脂质、磷脂辅助脂质、甾醇、磷脂聚乙二醇衍生物的摩尔比为(20~40):(15~30): (50~80): (7~20)。
优选的,所述水相中编码目标蛋白的RNA的浓度为5~100 μg/mL,海藻糖的浓度为1~25% w/v。
优选的,所述编码目标蛋白的RNA为编码目标蛋白的siRNA、编码目标蛋白的mRNA和编码目标蛋白的saRNA中的一种或多种。

本发明提供了一种RNA疫苗递送制剂的制备方法,包括以下步骤:将脂质、磷脂辅助脂质、甾醇、磷脂聚乙二醇衍生物和有机溶剂混合,得到有机相;所述脂质为可电离脂质或阳离子脂质;将编码目标蛋白的RNA、海藻糖和水混合,得到水相;将所述有机相加入所述水相中,去除所得混合料液中的有机溶剂,得到RNA疫苗递送制剂。传统方法直接将海藻糖和包载RNA的脂质纳米颗粒搅拌混合,此过程中海藻糖都分布在脂质纳米颗粒的外部;而本发明以脂质与海藻糖的混合物为载体构建脂质纳米颗粒(LNPs),即"coload"方法,使海藻糖负载到脂质纳米颗粒之中,同时脂质纳米颗粒的外部也存在部分海藻糖。脂质纳米颗粒内部的海藻糖能够代替水分子与包载的RNA形成氢键相互作用,其次脂质纳米颗粒外部的海藻糖也可以与脂质分子形成氢键作用,在后续的冻干的过程中,脂质纳米颗粒内外部的海藻糖皆有助于保护负载RNA的脂质纳米颗粒的完整性和稳定性。
实施例结果表明,本发明制备的脂质纳米颗粒在粒径、分散系数方面与传统方法基本无差异,但能够提高脂质纳米颗粒对mRNA的包封能力,在体内外实验中表现出更高的转染效率和药物递送的效果。冻干后的制剂与单纯将海藻糖与脂质纳米颗粒混合所制备的制剂(仅外部有海藻糖)相比,稳定性更好,转染效率更高。根据实验结果,推测海藻糖促进细胞转染的机制与海藻糖的细胞保护作用和抗氧化应激作用有关,具体原理为:RNA疫苗递送制剂可以在将mRNA递送至胞内的同时也将部分海藻糖递送进胞内,海藻糖可以提供细胞保护作用,具有更高的细胞活力,能减轻细胞的氧化应激,并且与仅在外部含有海藻糖的LNP相比,本发明制备的RNA疫苗递送制剂可以同时递送更多的海藻糖至胞内,因此海藻糖的保护作用更为显著,细胞内mRNA转染效率更高。

本发明提供了一种RNA疫苗递送制剂的制备方法,包括以下步骤:

将编码目标蛋白的RNA、海藻糖和水混合,得到水相;
将所述有机相加入所述水相中,去除所得混合料液中的有机溶剂,得到RNA疫苗递送制剂。
本发明将脂质、磷脂辅助脂质、甾醇、磷脂聚乙二醇衍生物和有机溶剂混合,得到有机相。在本发明中,所述脂质为可电离脂质或阳离子脂质;所述可电离脂质优选为DLin-MC3-DMA、SM-102和ALC-0315中的一种;所述阳离子脂质优选为DOTAP、DOTMA、DIMRIE和DOTIM中的一种;在本发明的具体实施例中,所述脂质更优选为SM-102;所述磷脂辅助脂质优选为二硬脂酰磷脂酰胆碱(DSPC);所述甾醇优选为胆固醇和植物甾醇中的一种或多种;所述植物甾醇优选包括β-谷甾醇、豆甾醇和麦角甾醇中的一种或多种;所述磷脂聚乙二醇衍生物优选为1,2-二肉豆蔻酰-rac-甘油-3-甲氧基聚乙二醇(又称为聚乙二醇-二肉豆蔻酸甘油酯,简写为PEG-DMG);所述磷脂聚乙二醇衍生物的数均分子量优选为500~3000。
在本发明中,可电离脂质或阳离子脂质是决定RNA转染效率的关键因素,LNPs经内吞作用到达溶酶体,其中的可电离脂质或阳离子脂质在低pH环境下带正电,可通过“质子海绵效应”突破溶酶体屏障,将RNA运送至胞质中,进行目标蛋白的翻译。细胞的阴离子脂质通过中和LNPs可电离脂质或阳离子脂质负载的电荷,破坏脂质载体与核酸之间的静电相互作用,帮助RNA从LNPs中释放;磷脂辅助脂质可提高可电离脂质或阳离子脂质的相变温度,支持层状脂质双层结构的形成并稳定其结构排列;甾醇在LNPs中是细胞转染中极其重要稳定因子,可以调节脂质双分子层的流动性,因为其是细胞膜的固有组成成分,具有较强的膜融合性,可以促进RNA递送;磷脂聚乙二醇衍生物是一种可以提高稳定性、防止颗粒聚集、避免被免疫系统快速清除的LNPs表面脂质。
在本发明中,所述脂质、磷脂辅助脂质、甾醇、磷脂聚乙二醇衍生物的摩尔比优选为(20~40):(15~30):(50~80):(7~20),更优选为(25~35):(20~27): (40~70): (8~15),进一步优选为35:27:70:10。
在本发明中,所述有机溶剂优选为四氢呋喃(THF)、乙醇和甲醇中的一种或多种。
本发明将编码目标蛋白的RNA、海藻糖和水混合,得到水相。在本发明中,所述编码目标蛋白的RNA优选为编码目标蛋白的siRNA、编码目标蛋白的mRNA和编码目标蛋白的saRNA中的一种或多种,更优选为编码目标蛋白的mRNA;在本发明中,所述目标蛋白优选包括病毒的SPIKE蛋白、功能蛋白的阻断剂、功能蛋白的激动剂中的一种或多种,所述病毒的SPIKE蛋白优选包括乙肝HBsAg抗原蛋白或流感病毒HA;本发明对编码目标蛋白的RNA序列及其制备方法没有特殊要求,采用本领域技术人员熟知的方法即可。
在本发明中,所述水相中编码目标蛋白的RNA的浓度优选为5~100 μg/mL,更优选为10~50 μg/mL,进一步优选为15~20 μg/mL;海藻糖的浓度优选为1~25% w/v,更优选为5~20% w/v。
得到有机相和水相后,本发明将所述有机相加入所述水相中,去除所得混合料液中的有机溶剂,得到RNA疫苗递送制剂。在本发明中,所述有机相和水相的体积比为1:(1~10);所述有机相中的脂质和所述水相中的编码目标蛋白的RNA的氮磷比(N/P)优选为(3~15):1,更优选为(4~10):1,进一步优选为6:1。本发明优选在搅拌条件下将有机相逐滴加入水相中,所述搅拌的转速优选为400 r/min。有机相加入水相中后,脂质、磷脂辅助脂质、甾醇和磷脂聚乙二醇衍生物发生自组装形成脂质纳米粒,同时将编码目标蛋白的RNA和部分海藻糖包载到脂质纳米粒内部,形成包载有RNA和部分海藻糖的脂质纳米颗粒,并且脂质纳米颗粒的外部也负载有部分海藻糖,所得脂质纳米颗粒记为海藻糖共包载的mRNA-LNPs。
有机相加入完毕后,本发明去除所得混合料液中的有机溶剂;去除所述有机溶剂的方法优选为常温旋转蒸发;去除有机溶剂后,所得料液为海藻糖共包载的mRNA-LNPs的混悬液,即为本发明的RNA疫苗递送制剂的混悬液形式。
在本发明中,还包括所述混悬液进行冷冻干燥,得到RNA疫苗递送制剂的冻干粉;所述冷冻干燥优选包括:将混悬液在-80 ℃下冷冻24 h后,在冷冻干燥系统中冻干24 h。冷冻干燥后,得到海藻糖共包载的mRNA-LNPs的固体产物,即为本发明的RNA疫苗递送制剂的冻干粉形式;所述冻干粉形式的RNA疫苗递送制剂容易储存和运输,在使用时,优选用生理盐水或PBS缓冲液重新分散成混悬液形式。
本发明还提供了上述方案所述制备方法制备得到的RNA疫苗递送制剂。本发明制备的RNA疫苗递送制剂对RNA的包封能力强,稳定性好,并且RNA的体外转染和体内表达效率更高。
下面将结合本发明中的实施例,对本发明中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
下列实施例中,以编码荧光素酶的mRNA作为模型RNA。
实施例1
本实施例制备的样品共为5组:其中非冻干3组:不含海藻糖的mRNA-LNPs混悬液组(记为LNP组),海藻糖与mRNA-LNPs的混悬液组(记为Tre-mixed组),海藻糖共包载的mRNA-LNPs混悬液组(记为Tre-coload);冻干2组:分别将Tre-mixed组和Tre-coload组的悬浮液冻干,记为FD-mixed组和FD-coload组。
1、样品制备:采用溶剂挥发法合成LNPs,具体步骤如下:
海藻糖共包载的mRNA-LNPs混悬液的制备:将可电离脂质(SM-102)、磷脂辅助脂质(DSPC)、胆固醇以及聚乙二醇-二肉豆蔻酸甘油酯(PEG-DMG,分子量为2509)以35:27:70:10的摩尔比溶解在THF中,得到有机相。将编码荧光素酶的mRNA和海藻糖溶于水中,得到水相;水相中编码荧光素酶的mRNA的浓度为16.7 μg/mL,海藻糖的浓度为20% w/v。按照SM-102和mRNA的氮磷比(N/P)= 6: 1,有机相与水相体积比为1: 3的比例,在400 r/min的搅拌转速下,将有机相逐滴加入水相中;然后通过常温旋转蒸发将有机试剂去除,即得海藻糖共包载的mRNA-LNPs混悬液(Tre-coload)。
mRNA-LNPs混悬液的制备:其他条件和Tre-coload混悬液的制备方法相同,仅在水相中不加入海藻糖,得到mRNA-LNPs混悬液(LNP)。
海藻糖与mRNA-LNPs的混悬液的制备:在mRNA-LNPs混悬液制备完成后,加入20%w/v的海藻糖,搅拌混合均匀,得到海藻糖与mRNA-LNPs的混悬液(Tre-mixed)。
FD-coload组样品的制备:将海藻糖共包载的mRNA-LNPs混悬液在-80 ℃冷冻24 h后,在冷冻干燥系统中冻干24 h,得到冻干粉(FD-coload)。
FD-mixed组样品的制备:将海藻糖与mRNA-LNPs的混悬液在-80 ℃冷冻24 h后,在冷冻干燥系统中冻干24 h,得到冻干粉(FD-mixed)。
2、表征
通过马尔文粒径测试仪测定各组的粒径、PDI和zeta电位,结果见图1和表1;
对各组样品的mRNA包封率进行测试,结果见表1;包封率的测试方法如下:
(1)建立标准曲线:通过Ribogreen检测试剂盒测定mRNA的包封率。首先制备线性浓度梯度的mRNA标样,Ribogreen检测液按照1:2000的体积比稀释,然后按照mRNA标样:Ribogreen检测液=1:1的体积比混合,避光孵育5 min,随后用荧光分光光度计测定标样中荧光强度,激发光500 nm,发射光525 nm,根据标样浓度梯度和读取的荧光值,绘制标准曲线,得到浓度计算公式。
(2)游离mRNA浓度测定:待检测样品用1×TE缓冲液稀释至适宜浓度(30 ng/mLmRNA),待测样品与Ribogreen检测液等体积混合,用荧光分光光度计测定荧光强度,把样品荧光值代入浓度计算公式,得到样品中游离mRNA的浓度(记为C游离)。
(3)总mRNA浓度测定:待测样品用2%Triton-TE缓冲液稀释,以破坏LNPs结构释放所有的mRNA,然后与Ribogreen检测液等体积混合,用荧光分光光度计测定荧光强度,把样品荧光值代入浓度计算公式,得到样品中总mRNA的浓度(记为C总)。
(4)包封率计算:包封率% = 1-(C游离/C总)×100%。

表1中的结果显示,各组所得脂质纳米颗粒的粒径均在150 nm左右,电位在35 mV左右,包封率均大于95%,并且Tre-coload组和Tre-mixed组相比,包封率更高。
通过透射电子显微镜(TEM)评估LNP、Tre-coload组、FD-coload样品颗粒的大小和分布,结果见图2。图2中的结果显示,三组样品外观呈球形,大小相对均一,分布均匀。
实施例2:海藻糖包封率以及细胞内海藻糖含量的测定
采用海藻糖含量检测试剂盒-酶法(货号:NMKD0233,分光法 50T/48S)检测Tre-coload组样品包载在LNPs之中的海藻糖的量。具体步骤如下:将样品经超滤离心(5000rpm,120 min)获取样品中游离的海藻糖(C游离)。游离海藻糖直接加入试剂避光反应30 min,其余样品经2%Triton-100稀释以破坏LNPs结构释放所有的海藻糖,用于计算C总,然后在紫外分光光度计510 nm处测定吸光值,按照下式计算海藻糖包封率:海藻糖包封率=1-(C游离/C总)×100%;
采用以上方法测得Tre-coloaded组样品中海藻糖的包封率(%)为:86.37±1.481。
细胞内海藻糖含量测定:取对数生长期的HEK293T细胞种植于6cm培养皿,每皿接种3×106个细胞,48 h后分别加入Tre-mixed组和Tre-coload组样品,其mRNA为2.5 μg每皿,4 h后弃培养基,细胞经RIPA裂解液裂解后,超声破碎10min,释放所有的海藻糖,然后在紫外分光光度计510 nm处测定吸光值。
按照下式计算细胞内海藻糖含量:细胞内海藻糖含量(%)=胞内海藻糖/总海藻糖×100%
采用以上方法测得细胞内海藻糖的含量占加入总量的比值如下:Tre-mixed 组为2.79±0.21(%),Tre-coload组为3.94±0.19(%) (P < 0.05, t test),具体测试结果如图3所示。以上结果说明,Tre-coload组样品可以促进海藻糖进入细胞内。
实施例3:mRNA体外转染效率测试
为了验证体外转录的mRNA产物的蛋白表达情况,本实施例使用编码荧光素的mRNA转染HEK293T细胞。本实施例共设置5组实验:LNP组,Tre-mixed组,Tre-coload组,FD-mixed组和FD-coload组。取对数生长期的HEK293T细胞种植于96孔板,每组5个复孔,每孔接种2×104个细胞,48 h后加入不同的制剂(均为新鲜制备的液体制剂),其mRNA为100 ng每孔剂量,4 h后更换为完全培养基,继续培养24 h后使用luciferase相对荧光强度检测试剂盒检测制剂的转染效率。
结果如图4~图6所示,其中图4为LNP组和Tre-coload组样品的mRNA体外转染效率对比图,图5为Tre-mixed组和Tre-coload组样品的mRNA体外转染效率对比图,图6为LNP组、Tre-mixed组、Tre-coload组、FD-mixed组和FD-coload组样品的mRNA体外转染效率对比图。图4~图6中的结果表明,Tre-coload组的mRNA体外转染效率明显高于LNP组及Tre-mixedLNP,差异有统计学意义(P < 0.05,单因素方差分析)。以上结果表明,Tre-coload组表达的抗原蛋白量高,免疫反应将更加明显,在实际应用中,同样剂量的疫苗,本发明制备的Tre-coload样品中所含的mRNA能够在接种后高效地进入宿主细胞并合成更多的抗原蛋白(目标蛋白)。
实施例4:mRNA体内表达实验
有效的体内递送是mRNA疫苗实现治疗效果的关键性要素,为了进一步观察Tre-coload组和LNP组mRNA体内表达的差异,本实施例通过小动物成像技术进行验证。SPF级雄性BALB/C小鼠(9周龄)适应性饲养1周,进行腿部脱毛,在双侧后腿分别肌肉注射含5 μgmRNA的制剂。注射后24 h和48 h,以150 mg/kg的剂量腹腔注射D-Luciferin(15 mg/mL),随后将小鼠放入含4%异氟烷的通风麻醉室中麻醉,15 min后,经IVIS成像系统获取生物发光图像,使用Living Image软件对图像进行定量分析。不同的颜色代表了不同的发光,由蓝色到红色的渐变分别对应发光值由低到高的变化,发光值间接反映荧光素酶蛋白的表达量。
结果如图7~图8所示,图7为生物发光图像,图8为生物发光图像的定量分析结果;图7~图8中的结果显示,注射后48 h,Tre-coload组mRNA表达水平明显高于LNP组,差异有统计学意义(p < 0.05, One-Way ANOVA analyses)。
实施例5:细胞活力测定
本实施例实验组采用的制剂为LNP、Tre-coload和Tre-mixed组的制剂,并采用PBS作为对照组。具体实验步骤如下:
取对数生长期的HEK293T细胞种植于96孔板,每组5个复孔,每孔接种2×104个细胞,48 h后加入不同的制剂,其mRNA为100 ng每孔剂量,8 h后更换为完全培养基,继续培养24 h后,加入10 μL CCK8室温孵育4h,在酶标仪490 nm波长下测定各孔OD(opticaldensity)值。并按照下式计算细胞活力:细胞活力=(实验组吸光度平均值-空白组吸光度平均值)/(对照组吸光度平均值-空白组吸光度值)×100 %。
测试结果如图9所示。图9中的结果表明,含海藻糖的样品具有更高的细胞活力,说明海藻糖可以发挥细胞保护作用,降低细胞因外部应激引起的损伤,从而增加细胞活力。
实施例6:细胞氧化应激水平测定
本实施例实验组采用的制剂为LNP、Tre-coload和Tre-mixed组的制剂,并采用PBS作为对照组。具体实验步骤如下:
取对数生长期的HEK293T细胞种植于6 cm培养皿,每皿接种3×106个细胞,48 h后加入不同的制剂,其mRNA为2.5 μg每皿,8 h后采用丙二醛(MDA)(Elabscience,E-BC-K028-M)、超氧化物歧化酶(SOD)(Elabscience,E-BC-K019-S)、谷胱甘肽(GSH)(Elabscience,E-BC-K030-M)试剂盒检测MDA、SOD、GSH表达。
测试结果如图10所示。图10中的结果表明,海藻糖具有抗氧化的作用,与LNP组以及Tre-mixed组相比,Tre-coload组可以明显降低胞内的MDA含量,同时提高胞内的SOD及GSH等抗氧化物质的含量,能够减轻细胞受氧化应激的影响,有助于维持RNA的完整性,提高转染效率。
实施例7:mRNA完整性检测
1.样品准备:检测LNP、FD-mixed、FD-coload样品中的RNA完整性,以游离mRNA为对照,其中游离mRNA(RNA浓度0.2 mg/mL)直接与RNA 上样缓冲液等体积混合,LNP、FD-mixed、FD-coload样品(RNA浓度均为0.05 mg/mL)经Triton-100裂解后,与RNA 上样缓冲液等体积混合,95 ℃孵育1 min。
2.制胶:1×TAE 40 mL,琼脂糖0.8 g,配制2%琼脂糖凝胶,摇匀后于微波炉中中火加热,至琼脂糖完全融化,加入溴化乙锭(EB)染色液,使其浓度为0.5 μg/mL。制胶板插入梳子,待胶稍冷(约60 ℃)倒入制胶板中,厚度3-5 mm。室温静置30-45 min至凝胶完全凝结。
3.电泳:倒入少量电泳缓冲液在凝胶顶部,小心拔出梳子,将凝胶放入电泳槽中,向电泳槽中加入缓冲液,刚好没过凝胶1 mm。每孔上样200 ng RNA。关上电泳槽盖,接好电源,电压80 V,使RNA从负极向阳极移动,15min后关闭电源。
4.成像:小心取出凝胶,放入0.5 μg/mL的EB液中,浸染30 min,于凝胶成像仪观察并拍摄,结果如图11所示。图11中的结果表明,FD-coloaded样品经冻干复溶之后,RNA完整性并没有遭到破坏。
实施例8:冻干稳定性检测
将FD-mixed组和FD-coload组样品放入4 ℃环境中密封保存,存放一定时间后,取出复溶,并以新鲜制备的LNP组、Tre-mixed组以及Tre-coload组的液体制剂为对照,按实施例4的方法测定细胞转染效率,结果如图12~图15所示,其中图12为FD-mixed组和FD-coload组样品冻干后立即复溶后的细胞转染效率测试结果;图13为FD-mixed组和FD-coload组样品冻干后4 ℃存放3天复溶后的细胞转染效率测试结果;图14为FD-mixed组和FD-coload组样品冻干后4 ℃存放7天复溶后的细胞转染效率测试结果;图15为FD-mixed组和FD-coload组样品冻干后4 ℃存放28天复溶后的细胞转染效率测试结果。根据图12~图15中的结果可以看出,FD-coload组具有更高的冻干稳定性。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (10)
1.一种RNA疫苗递送制剂的制备方法,其特征在于,包括以下步骤:
将脂质、磷脂辅助脂质、甾醇、磷脂聚乙二醇衍生物和有机溶剂混合,得到有机相;所述脂质为可电离脂质或阳离子脂质;
将编码目标蛋白的RNA、海藻糖和水混合,得到水相;
将所述有机相加入所述水相中,去除所得混合料液中的有机溶剂,得到RNA疫苗递送制剂。
2.根据权利要求1所述的制备方法,其特征在于,所述可电离脂质为DLin-MC3-DMA、SM-102和ALC-0315中的一种;所述阳离子脂质为DOTAP、DOTMA、DIMRIE和DOTIM中的一种;
所述磷脂辅助脂质为二硬脂酰磷脂酰胆碱;
所述甾醇为胆固醇和植物甾醇中的一种或多种;
所述磷脂聚乙二醇衍生物为1,2-二肉豆蔻酰-rac-甘油-3-甲氧基聚乙二醇。
3.根据权利要求2所述的制备方法,其特征在于,所述植物甾醇包括β-谷甾醇、豆甾醇和麦角甾醇中的一种或多种。
4.根据权利要求1所述的制备方法,其特征在于,所述脂质、磷脂辅助脂质、甾醇、磷脂聚乙二醇衍生物的摩尔比为(20~40):(15~30): (50~80): (7~20)。
5.根据权利要求1所述的制备方法,其特征在于,所述水相中编码目标蛋白的RNA的浓度为5~100 μg/mL,海藻糖的浓度为1~25% w/v。
6.根据权利要求1所述的制备方法,其特征在于,所述编码目标蛋白的RNA为编码目标蛋白的siRNA、编码目标蛋白的mRNA和编码目标蛋白的saRNA中的一种或多种。
7.根据权利要求1所述的制备方法,其特征在于,所述有机相和水相的体积比为1:(1~10);所述有机相中的脂质和所述水相中的编码目标蛋白的RNA的氮磷比为(3~15):1。
8.根据权利要求1所述的制备方法,其特征在于,所述有机溶剂为四氢呋喃、乙醇和甲醇中的一种或多种。
9.根据权利要求1所述的制备方法,其特征在于,去除所述混合料液中的有机溶剂后,所得RNA疫苗递送制剂为悬浮液;还包括将所述悬浮液进行冷冻干燥,得到RNA疫苗递送制剂的冻干粉。
10.权利要求1~9任意一项所述制备方法制备得到的RNA疫苗递送制剂。
