Abstract
本发明属于生物医药领域,涉及一种抗VEGF和补体双功能融合蛋白及其应用。所述融合蛋白包含VEGF结合域和补体结合域,还包含人IgG Fc结构域。本发明通过分子组合优化和分子改造提供了具有高表达、高活性和性质稳定的抗VEGF和补体双功能融合蛋白,特别是VEGFR1D2‑VEGFR2D3‑mhIgG Fc‑(GGGGS)3‑sCD59和VEGFR1D2‑mhIgG Fc‑(GGGGS)3‑sCD59。所述双功能融合蛋白可以同时高效地结合VEGF因子和补体分子,表现出更强地抗VEGF及抗补体的功效;对治疗年龄相关性黄斑变性,尤其是并发有眼底血管新生的干性年龄相关性黄斑变性,或者伴随有黄斑萎缩的湿性年龄相关性黄斑变性,具有更优的治疗效果。
Description
技术领域
本发明属于生物医药领域,涉及一种抗VEGF和补体双功能融合蛋白及其应用。
背景技术
年龄相关性黄斑变性(Age-related macular degeneration,AMD)是65岁以上患者视力损害的主要原因,全世界有1.96亿人患有AMD,预计到2040年将增至2.88亿人。AMD可分为两种类型:干性AMD(非血管性, dAMD)和新生血管性AMD(nAMD)。在干性AMD的早期阶段,存在慢性低水平炎症、视网膜下间隙的玻璃体疣沉积、黄斑部视网膜色素上皮(RPE)和光感受器的不可逆性丧失,以及随后进行性的中央视力丧失。在干性AMD的晚期,视网膜色素上皮变性变得融合,视力丧失恶化,称为地图样萎缩(Geographic atrophy,GA)。在大约10%的患者中,AMD可进展为更具攻击性的nAMD,该疾病与中心视力的快速丧失有关,其特征是异常的脉络膜血管生长进入黄斑,称为脉络膜新生血管(Choroidal neovasculation,CNV)。这些血管“渗漏”,并且有过多的瘢痕组织。
AMD的发病机制复杂,据文献报道,相关的信号通路主要包括血管新生相关通路(VEGF、PDGF、FGF等)、补体相关通路(C3、C5等)、纤维化相关通路(FGF、TGFβ等)和免疫炎症因子通路(IL-6、TNF-α等)。
血管内皮生长因子(Vascular endothelial growth factor,VEGF)能诱导已有血管的再生(血管再生)或新血管的生长(血管新生),是胚胎发育和血管修复的关键因子。VEGF 家族包括 VEGF-A、VEGF-B、VEGF-C、VEGF-D、VEGF-E 以及胎盘生长因子 1、2(PIGF-1、PIGF-2)。VEGF-A 是迄今最有效且研究最多的血管生长诱导因子。VEGF的家族成员由多个外显子编码,选择性剪接后可以产生不同的亚型,从而影响可溶性和受体结合情况。例如,VEGF-A有7个亚型,而VEGF-B 有2个亚型。VEGF的家族成员通过与VEGF受体(VEGFR)结合,激活信号转导。VEGFR为酪氨酸激酶受体,具有由7个免疫球蛋白样(immunoglobulin ,IG)结构域所组成的细胞外区域。VEGFR-1(Flt-1)结合VEGF-A、VEGF-B,以及PIGF,并可作为VEGF的诱饵受体或VEGFR-2的调节剂。VEGFR-2(KDR/Flk-1)结合所有VEGF同工型,且为VEGF诱导的血管生成通路的主要媒介。VEGFR-3(Flt-4)结合VEGF-C与VEGF-D,但不结合VEGF-A,是淋巴管生成的主要媒介。
补体系统为先天免疫系统的功能性效应子,由多种可溶性蛋白、膜结合蛋白和补体受体组成。补体活化导致一系列蛋白酶活化的级联反应,刺激细胞因子的释放与活化级联反应的放大。补体系统可通过三种不同的途径活化,即典型途径、旁路途径和凝集素途径。这3种途径都通过关键的C3转化酶与C5转化酶复合物,分别裂解补体分子C3与C5。补体活化的最终结果为细胞杀伤膜攻击复合物(membrane attack complex,MAC)的活化、刺激免疫细胞释放炎症因子和趋化因子、由过敏毒素C3a与C5a引起的过敏反应,以及病原体的吞噬作用。
目前,抗VEGF疗法(如贝伐单抗、雷尼单抗和阿柏西普)是唯一经批准上市的治疗nAMD的标准药物疗法,但是临床上仍有相当一部分患者对抗VEGF药物的响应不完全,表现为持续性地渗漏、出血和视力恢复欠佳。此外长期注射抗VEGF药物的nAMD患者中,有超过10%的比例转化为干性AMD。在抗VEGF药物长期使用的患者中,有超过40%的患者会伴随黄斑萎缩,与干性AMD引起的地图样萎缩较为类似。而针对干性AMD患者视力丧失的情况,Apellis公司的补体因子3抑制剂pegcetacoplan(商品名Syfovre)和Astellas公司的补体因子5抑制剂avacincaptad pegol(商品名Izervay)先后获FDA批准上市,但在临床研究中均出现nAMD发生率增加的情况,在药品说明书中也特别指出了该种风险。本申请预期通过结合抑制VEGF及抑制补体通路来进一步解决干性及湿性AMD治疗过程中出现的问题。
发明内容
本发明的目的是提供一种表达量高、活性好、纯度高的抗VEGF及补体靶点的双功能性融合蛋白,通过有效地抑制VEGF及补体途径,能够治疗多种VEGF及补体通路相关的疾病,例如湿性和干性AMD,进而解决上述临床治疗过程中出现的问题。结果发现,本发明提供的抗VEGF及补体双功能性融合蛋白既体现出更加高效的VEGF拮抗活性,同时显著提高了抑制补体溶血活性,在干性和湿性AMD动物模型中均体现出更优的药效,同时通过对分子中Fc结构域的改造显著提高了分子的稳定性。

有益效果:
本发明通过分子组合优化和分子改造提供了一种具有高表达、高活性和性质稳定的抗VEGF和补体双功能融合蛋白,特别是VEGFR1D2-VEGFR2D3-mhIgG Fc-(GGGGS)3-sCD59和VEGFR1D2-mhIgG Fc-(GGGGS)3-sCD59。
所述双功能融合蛋白采用经过人工改造的hIgG Fc后,能够显著改善采用天然IgGFc片段时制备双功能融合蛋白造成的蛋白聚集问题,一方面避免融合蛋白的活性降低,另一方面防止引起免疫原性问题,因此本发明中Fc片段的改造有利于提高双功能性融合蛋白的稳定性避免聚集,并且更加安全有效。
所述双功能融合蛋白可以同时高效地结合VEGF因子和补体分子,尤其是对于sCD59的活性有显著提升,与sCD59相比,所述双功能融合蛋白抑制补体溶血活性分别提高了4.6倍和10倍。
另外,所述双功能融合蛋白在干性和湿性AMD模型中均有效。其中干性AMD模型(实施例13中碘酸钠诱导的干性AMD小鼠模型)中,显示出显著抑制ERG波幅降低的效果。在湿性AMD模型(实施例9中激光诱导小鼠CNV模型)中,从渗漏面积和渗漏评分来看,能有效抑制CNV,且双功能蛋白的抑制活性优于单体分子。
因此,本发明提供的双功能融合蛋白预期对治疗年龄相关性黄斑变性,尤其是并发有眼底血管新生的干性年龄相关性黄斑变性,或者伴随有黄斑萎缩的湿性年龄相关性黄斑变性,具有显著的治疗效果。

除非本文另有定义,否则本文使用的科学及技术术语具有本领域普通技术人员通常理解的含义。
于某些实施例中,本发明涉及同时以血管内皮生长因子VEGF和补体C3b、C5、C8、或C9作为靶点的双功能性融合蛋白。由于VEGF及补体信号途径与许多疾病有关,包括年龄相关性黄斑变性(AMD),因此相较于补体及血管内皮生长因子(VEGF),具有双特异性抑制活性的蛋白可能会提供比分别抑制补体或血管内皮生长因子(VEGF)的蛋白更优的治疗效果。
于某些实施例中,所述双功能性融合蛋白可以是VFC,V表示VEGF结合域,F表示人IgG1 Fc结构域,C表示补体结合域,即通过在N端将一VEGF结合域与人IgG Fc结构域以及C端与一补体结合域序列融合而产生。于另一方面,所述双功能性融合蛋白可以是CFV,即通过在N端将一补体结合域与人IgG Fc结构域以及C端与一VEGF结合域序列融合而产生。于其他方面,所述双功能性融合蛋白也可以是VCF或CVF的组合形式,即蛋白序列从N端至C端按照VEGF结合域、补体结合域、Fc结构域或者补体结合域、VEGF结合域、Fc结构域的顺序融合而产生。
进一步地,所述的VEGF结合域可以是VEGFR1胞外结构域2以及VEGFR2细胞外结构域3的串联结构域;另一方面也可以是单个VEGFR1胞外结构域2或两个串联VEGFR1胞外结构域2;于其他方面也可以是衍生自雷尼单抗Fab结构域;或衍生自雷尼单抗的Scfv片段。

进一步地,于某些实施例中,所述人IgG Fc结构域与补体结合域sCD59连接后,显著提高了sCD59的体外活性。
如本文所用,术语[Fab]指抗体上与抗原结合的区域。其由轻链的可变及恒定结构域(即前述LC)以及重链抗体的可变结构域以及第一恒定结构域(即前述HC)所组成。
如本文所用,术语[Scfv]指由重链(VH)以及轻链(VL)的可变区组成的单链片段,其通过一柔性连接肽连接在一起,其可以功能形式容易地在大肠杆菌中表达,可进行蛋白质工程改造以改善Scfv的特性,例如增加亲和力及改变特异性。
于本发明中,上述双功能性融合蛋白由Expi293细胞瞬时表达,并通过重组protein G亲和层析法或固定化金属离子亲和层析法从转染的细胞培养上清液中纯化。经过再一步的分子筛纯化过程获得了纯度大于90%的产物,且所有融合蛋白都被正确地形成及表达。
于某些实施例中,双功能性融合蛋白与VEGF的结合能力通过ELISA试验得以验证,双功能性融合蛋白表现出与VEGF165的强结合能力。
本发明提供的双功能融合蛋白可用于治疗血管内皮生长因子(VEGF)及补体相关疾病,以及用于制备治疗血管内皮生长因子(VEGF)及补体相关疾病的药物组合物;相关疾病包括眼部疾病、炎性疾病、自身免疫性疾病或肿瘤。
相关眼部疾病包括年龄相关性黄斑变性、糖尿病性视网膜病变、糖尿病性视网膜水肿、糖尿病性黄斑水肿、视网膜后纤维化、视网膜中央闭塞、视网膜静脉阻塞、缺血性视网膜病变、高血压性视网膜病、眼色素层炎(例如,前、中、后或全葡萄膜炎)、白塞病、Bietti结晶样角膜视网膜营养不良、睑缘炎、开角型青光眼、新生血管性青光眼、角膜新生血管、脉络膜新生血管形成(CNV)、视网膜下新生血管形成、角膜发炎及角膜移植并发症。其中年龄相关性黄斑变性包含湿性年龄相关性黄斑变性或干性年龄相关性黄斑变性,尤其是并发有眼底血管新生的干性年龄相关性黄斑变性,或者伴随有黄斑萎缩的湿性年龄相关性黄斑变性。
相关炎性疾病包括类风湿性关节炎、银屑病或强直性脊柱炎等。
相关肿瘤疾病包括大肠癌、非小细胞肺癌、神经胶质母细胞瘤、转移性肾癌、乳腺癌、结直肠癌、肺癌、胃癌或视网膜细胞瘤等。
以下结合具体实施例对本发明作进一步地解释说明。
实施例1双功能性融合蛋白结构设计
本实施例中,构建了19种双功能性融合蛋白及12种单体蛋白,其结构分别如下表1所示。其中:
VBD表示VEGF结合域蛋白;CBD表示补体结合域蛋白;BFP表示双功能性融合蛋白;
在VBD中,VEGFR1D2表示VEGF受体1胞外结构域2(SEQ ID NO.3),两侧含天然VEGF受体1胞外结构域2的N端侧翼22个氨基酸和C端侧翼16个氨基酸;VEGFR2D3表示VEGF受体2胞外结构域3(SEQ ID NO.4),C端含天然VEGF受体2胞外结构域3的C端侧翼7个氨基酸;VEGFR1D2D2表示两个VEGF受体1胞外结构域2的串联结构域;Rani HC表示Ranibizumab重链结构域;Rani LC表示Ranibizumab轻链结构域;Rani VH表示Ranibizumab重链可变结构域;Rani VL表示Ranibizumab轻链可变结构域;
在CBD中,Eculi-Scfv1和Eculi-Scfv2表示Eculizumab重链可变区VH和轻链可变区VL通过(GGGGS)3连接肽连接形成的Scfv片段,Eculi-Scfv1表示VH-(GGGGS)3-VL,Eculi-Scfv2表示VL-(GGGGS)3- VH;Pexe表示Pexelizumab;FHL表示因子H样蛋白1;FI表示因子I;sCD59表示可溶性CD59蛋白(SEQ ID NO.5),为天然人全长CD59蛋白成熟体,不含N端25个信号肽氨基酸序列及C端GPI-锚定序列;
hIgG1 Fc表示天然人IgG1 Fc结构域,包含天然人IgG1 Hinge区末端10个氨基酸,CH2结构域和CH3结构域,即表1中的hIgG1 Fc; mhIgG Fc表示经过改造的人IgG Fc结构域(SEQ ID NO.6),即表1中的mhIgG Fc。
P2A表示porcine teschovirus-1 2A肽;His tag表示6-10个histidine标签肽。His标签肽便于单体蛋白纯化,其对蛋白空间结构影响较小,通常情况下可不进行去除。

实施例2双功能性融合蛋白的表达及纯化
将编码上述表1所示融合蛋白或单体蛋白的核苷酸序列构分别建入PTT5载体中(其中,包含BFP6、BFP10编码基因的重组载体质粒图谱如图1-2所示,BFP6的编码基因如SEQID NO.7所示,BFP10的编码基因如SEQ ID NO.8所示,构建时在融合蛋白N段添加信号肽序列:MGVKVLFALICIAVAEA)。按照转染试剂说明书的方法,采用PEI MAX转染试剂(购自翌圣生物)将上述构建好的质粒载体转入Expi293细胞中。细胞培养5天后收集上清,留1ml细胞上清用ELISA方法进行表达量检测,剩余上清进行蛋白纯化。
针对含hIgG1 Fc或mhIgG Fc的蛋白,采用重组protein G亲和层析介质(重组protein G亲和介质购自金斯瑞)进行蛋白的初步分离和纯化。取3-5ml重组protein G填料装柱,用0.02M PBS 缓冲液(pH7.4)平衡,然后将细胞培养液上清缓慢上样,使蛋白与层析介质充分结合;使用PBS缓冲液清洗未发生结合的非特异性杂蛋白,最后用0.1M pH3.0甘氨酸缓冲液进行洗脱,洗脱液用pH9.0 Tris缓冲液调节pH值至中性。参考填料使用说明书,采用Superdex 200 prep grade凝胶过滤填料(购自Cytiva)进行精细纯化。纯化蛋白经Millipore Amicon Ultra-15 30kD超滤浓缩管浓缩,用GE PD-10 脱盐柱换液至20mM PBS缓冲液中(20mM sodium phosphate, 150mMNaCl, pH7.4)。
针对含His tag的蛋白,采用固定化金属离子亲和层析(IMAC)进行纯化。取镍NTA琼脂糖凝胶FF预装柱,用0.02M PBS 缓冲液(pH7.4,含10mM咪唑)平衡,直至流出液pH为7.4。对样品进行离心处理,加入咪唑使样品中咪唑终浓度为10mM,缓慢上样使蛋白与层析柱充分结合。使用平衡液冲洗未结合的非特异性杂蛋白,用0.02M PBS 缓冲液(pH7.4,含200mM咪唑)进行洗脱。经0.22μm滤膜过滤获得纯化的表1中的双功能融合蛋白和单体蛋白,无菌分装,超低温保存。
实施例3 双功能性融合蛋白表达量的测定
用ELISA方法测定质粒瞬转细胞培养上清中的蛋白表达量。用碳酸盐缓冲液稀释VEGF165(购自R&D systems)至200ng/mL,每孔100μL,2-8℃包被过夜。次日,洗板后加入1%BSA封闭。用1%BSA稀释样品,以VBD1作为参考标准品配制标准曲线和质控样品。封闭结束后洗板,每孔分别加入100μL标准曲线、质控样品和稀释后的待测样品(实施例2制备的细胞上清液),室温孵育1小时。洗板,加入1:4000稀释的HRP偶联羊抗人IgG Fc抗体(购自Sigma),室温孵育1小时。洗板,加入TMB显色,10分钟后终止反应,用酶标仪读取450nm OD值。用软件绘制标准曲线,计算待测样品浓度,根据分子量大小进行归一化处理。结果如图3所示,结果表明双功能性融合蛋白分子BFP5、BFP6、BFP7、BFP10、BFP18和BFP19均具有较高的瞬转表达水平。
实施例4 纯化的双功能性融合蛋白电泳纯度分析
为比较经改造的人IgG Fc片段对融合蛋白稳定性的影响,对经过一步proteinG亲和纯化的BFP6、BFP10、BFP18和BFP19(实施例2制备)进行SDS-PAGE电泳分析,不稳定的融合蛋白具有更高的聚集倾向。将样品稀释至1mg/ml,取1μL稀释后样品,加入2.5μL 4×LDSSample Buffer(金斯瑞M00676)和6.5μL超纯水,混匀后95℃处理5min,离心。取SDS-PAGE预制胶(金斯瑞M00652),撕掉胶板底部胶带,安装在微型垂直电泳槽中,电泳槽内倒入Tris-MOPS-SDS Running Buffer。平稳地将梳子拔出,每孔加入10μL处理后样品,盖好电泳槽盖,设置恒压140V,电泳40分钟。
结果如图4A所示,泳道1为BFP18,泳道2为BFP6,可以看出约140kd处的条带为主条带,也即实施例2制备的双功能融合蛋白,泳道2除约140 kd处的条带外,并无其他杂带,而泳道1在约140 kd处的条带上方还存在多个分子量更大的杂带,从主条带上方相邻条带的分子量来看(主条带上方相邻条带的分子量约为主条带的2倍)推测杂带是由于BFP18形成了聚集体造成的。同样地,泳道3、4也出现了上述情况,即BFP10所示在的泳道4中除约115kd处的条带外,并无其他杂带,而泳道3的BFP19在约115 kd处的条带上方还存在多个分子量更大的杂带。
为了进一步确定上述BFP18和BFP19产生的杂带是否为聚合体,对BFP18和BFP19经过还原后再次进行SDS-PAGE,结果如图4B所示,经过还原后的泳道2和泳道4中均只有一条带,可以确定泳道3中的杂带是由于BFP18形成了聚集体造成的,泳道7中的杂带是由于BFP19形成了聚集体造成的。
BFP6和BFP18,以及BFP10和BFP19结构上的区别仅在于BFP6/ BFP10采用的是经过改造的IgG Fc片段,BFP18/ BFP19采用的是天然IgG Fc片段,上述SDS-PAGE结果表明,含经改造的人IgG Fc片段的融合蛋白单体纯度明显高于含天然Fc片段的融合蛋白,聚集体显著降低。通常认为聚集体会导致融合蛋白的活性降低,以及引起免疫原性,并且不稳定的融合蛋白具有更高的聚集倾向,因此本发明中Fc片段的改造有利于提高双功能性融合蛋白的稳定性避免聚集,并且更加安全有效。
由于BFP18和BFP19纯化产品存在杂带,将对BFP18和BFP19蛋白进行进一步的分子筛精纯后,再进行后续关于Fc片段改造对双功能性融合蛋白活性影响的分析实验。
实施例5 双功能性融合蛋白与VEGF165结合活性的测定
以VBD为参考,考察双功能性融合蛋白分子与VEGF165的结合活性是否会受到影响,测试方法与实施例3描述的方法相似,不同之处在于所考察的样品为经过实施例2纯化后的蛋白样品,各蛋白样品从50nM开始,经3倍比梯度稀释至0.00085nM。最后用四参数回归模式进行曲线拟合,计算各样品结合活性EC50。
如图5及下表2所示,结果表明双功能性融合蛋白分子BFP6 和BFP10与VEGF165的结合能力未受影响,Fc片段改造对融合分子与VEGF165的结合无明显影响;而BFP5和BFP7与VEGF165的结合能力受到补体结合域的影响,结合能力显著下降(评价标准为EC50比值在80%~120%范围内,认为不受影响)。

实施例6 双功能性融合蛋白抑制HUVEC细胞增殖活性
以VBD为参考品,考察双功能性融合蛋白分子抑制VEGF依赖性HUVEC细胞增殖的生物学活性是否会受到影响。在96孔细胞培养板中每孔接种6×103个HUVEC细胞(购自Promcell),贴壁过夜。次日,用DMEM+2%FBS(购自Gibco)培养基作为稀释液稀释纯化后的蛋白样品,各蛋白样品(实施例2制备的纯化蛋白)从50nM开始,经1.5倍比梯度稀释至1.3nM。用DMEM+2%FBS培养基稀释液稀释VEGF165至400ng/mL。将各蛋白梯度稀释样品与VEGF165按1:1比例混合。吸去细胞孔中原培养液,每孔加入100μL前述混合液,注意不要使用边缘孔。将处理后的细胞置于37℃,5%CO2培养箱中培养,96h后取出细胞板,每孔加入10μL CCK-8显色液(购自Dojindo);放置于37℃,5%CO2培养箱中孵育3h,用酶标仪读取450nm OD值。最后用四参数回归模式进行曲线拟合,计算各样品抑制HUVEC细胞增殖的生物学活性IC50。
如图6及表3所示,结果表明,双功能性融合蛋白BFP5、BFP6、BFP7 和BFP10抑制HUVEC细胞增殖的生物学活性未受影响(评价标注为IC50比值在70%~130%范围内,认为不受影响)。

实施例7 双功能性融合蛋白抑制VEGF-KDR报告基因细胞活性
双功能蛋白对VEGFA诱导的报告基因细胞的转录激活具有抑制作用。将VEGF-KDR报告基因细胞培养于白壁透明底的PDL coating 96孔细胞培养板中培养过夜,次日,加入VEGFA蛋白(终浓度10 ng/mL)以及梯度稀释的蛋白样品(实施例2制备的纯化蛋白,最高终浓度从30 nM开始,3倍梯度稀释9个浓度),将96孔板放入37度细胞培养箱继续孵育6小时后加入100 μL/孔 Bright-Glo™ 萤光素酶检测试剂,放置3到5分钟,放入酶标仪中选取化学发光模式读取数值。计算每个浓度梯度的信号抑制率,绘制浓度-抑制率曲线,计算各样品IC50。
如图7及表4所示,结果表明BFP6和BFP10对VEGF-KDR报告基因细胞转录激活显示出更高的抑制活性,而通常情况下双功能融合蛋白与VBD相比增加了补体结构域会增大空间位阻,降低活性。

实施例8 双功能性融合蛋白抑制补体溶血活性
利用绵羊红细胞与相应抗体(溶血素)结合成的复合物,可激活血清中的补体,形成MAC复合体(C5b-9),导致红细胞溶解。CBD6(sCD59)与补体C9结合,阻止MAC的形成,从而抑制溶血。
首先将2%绵羊红细胞与等量稀释后的(1:2000)溶血素混匀,置37℃孵育30分钟;用GVB++缓冲液(0 .1%明胶,5mM Veronal,145mM NaCl,0 .025%NaN3,pH 7.3)稀释不同比例正常人血清,然后将活化的绵羊红细胞与不同比例的正常人血清在37℃培养30分钟后,确定导致绵羊红细胞发生90%裂解的正常人血清稀释度。通过将可裂解90%绵羊红细胞的正常人血清稀释液分别与0-20μM的CBD6蛋白、BFP6、BFP10、BFP18和BFP19蛋白在37℃下混合1小时,然后将上述混合物与绵羊红细胞在37℃下培养30分钟后测定绵羊红细胞的溶血程度。通过四参数回归模式进行曲线拟合,计算IC50。
如图8和表5所示,结果表明,与CBD6相比,双功能性融合蛋白BFP6和BFP10抑制补体溶血活性分别提高了4.6倍和10倍,未经Fc改造的双功能融合蛋白BFP18和BFP19体现出类似的增益效果。

实施例9双功能性融合蛋白抑制激光诱导小鼠CNV模型
采购约2月龄SPF级C57BL/6J雄性小鼠,在实验室饲养3-5天。
造模前,使用舒泰(25 - 50 mg/kg, i.p.)和盐酸赛拉嗪(5 mg/kg, i.p.)注射液麻醉动物,检眼镜检查眼底有无异常,挑选正常动物入组。
双眼造模,使用YAG激光光凝仪(VITRA,Quantel Medical)在距离视盘 1-1.5PD处围绕视乳头光凝3点(波长532 nm,250 mw,50 μm,100 ms),激光斑位置避开视网膜大血管和注射点。
给药,造模后第3天,通过玻璃体腔(IVT)分别注射2μl PBS、0.15mM 各供试品蛋白(实施例2制备的纯化BFP6、BFP10、VBD1、VBD2、CBD6)。
给药后第10天,进行荧光素钠眼底血管造影(FFA)检查,评价脉络膜新生血管(CNV)评分和渗漏面积。评分标准如下:
I级 | 无强荧光。 |

各组FFA代表性图片如图9所示,渗漏评分和渗漏面积统计如表6所示,结果表明,相对于PBS组,各供试品蛋白均能有效抑制CNV,且双功能蛋白的抑制活性优于单体分子。

实施例10 携带目标蛋白基因的AAV8病毒制备
采用三质粒共转染VPC2.0细胞的方法包装AAV病毒,其中的三质粒分别是辅助包装质粒、AAV8 rep-cap质粒、以及各转基因质粒GOI。
将实施例2中分别含VBD1、VBD2、BFP6、BFP10、CBD6编码基因核酸序列的PTT5载体进行EcoRI/HindIII双酶切,电泳回收目的基因片段。将目的基因片段同源重组到EcoRI/HindIII双酶切的pAAV-MCS载体中,大肠杆菌转化,挑取克隆测序验证获得分别将VBD1、VBD2、BFP6、BFP10、CBD6编码基因克隆至pAAV-MCS载体的pAAV-VBD1、pAAV-VBD2、pAAV-BFP6、pAAV-BFP10和pAAV-CBD6载体,作为GOI质粒。
GOI质粒、AAV8 rep-cap质粒和pHelper辅助质粒需经过大量抽提,浓度大于1μg/μL,A260/280 在1.8-2.0 间用以包装病毒。无血清悬浮培养VPC2.0细胞至细胞密度为1E+6cells/ml。将抽提的三质粒按摩尔比1:1:1的比例混合,再把质粒DNA按总质量与PEIpro转染试剂按1:2的质量比例进行混匀,室温孵育20分钟后缓慢加到细胞悬液(细胞与三质粒的比例为1mL:1μg)中混匀。放37℃,8% CO2摇床培养3天,收集细胞悬液。
将细胞悬液10,000 g 离心10min,所得离心上清转移到一个新离心管中,所得细胞沉淀用少量PBS溶液重悬后反复冻融法裂解细胞;冻融后细胞再10,000 g 离心10min,收集离心所得上清。将两次离心收集的上清混合在一起,用 0.45μm 滤器过滤除杂质。加入1/2 体积的 1M NaCl,10% PEG8000 溶液,混合均匀,4度过夜。12,000 rpm 离心 2h,弃上清,病毒沉淀用适量的 PBS 溶液溶解,待完全溶解后用 0.22μm 滤器过滤除菌。加入Benzonase 核酸酶消化去除残留的质粒DNA(终浓度为50 U/ml)。合上管盖,颠倒几次以充分混合。在37℃孵育30 分钟;用 0.45 μm 针头滤器过滤,取滤出液,即为浓缩的AAV病毒。
向病毒浓缩液中添加固体 CsCl 直到密度为1.41 g/ml(折射率为1.372);将样品加入到超速离心管中,用预先配好的 1.41 g/ml CsCl 溶液将离心管剩余空间填满;在175,000 g 下离心24 小时,以形成密度梯度。按顺序分步收集不同密度的样品,取样进行滴度测定。收集富集有AAV 颗粒的组分。
重复上述过程一次。将病毒装入100 kDa的透析袋,4度透析脱盐过夜,透析缓冲液成分为含0.001% Pluronic F68的PBS,pH7.2。收获的透析后样品即为纯化的AAV病毒,可用于体内药效验证。
本发明使用的AAV病毒均采用上述方法进行制备,包装不同GOI质粒,即可制备携带不同目的基因的AAV,通过上述方法获得携带VBD1的AAV8-VBD1病毒、携带VBD2的AAV8-VBD2病毒、携带BFP6的AAV8-BFP6病毒、携带BFP10的AAV8-BFP10病毒、携带CBD6的AAV8-CBD6病毒。
实施例11 携带目标基因的AAV8病毒体外抑制MAC沉积
在补体激活途径的末端,C5b可与C6稳定结合为C5b6,后者自发与C7结合成C5b67,该复合物中的C7初步插入靶细胞膜脂质双分子层,继而C8与插入膜上的C5b67高亲和力结合,形成稳定的、深插入细胞膜的C5b678,该复合物可与12~18个C9分子结合为C5b6789n,即攻膜复合体MAC。CD59是一种MAC抑制蛋白,可以阻止C9聚集到C5b678复合体上,从而阻止MAC的形成。
将2E5/孔 hepa-1c1c7细胞种在含有聚D-赖氨酸涂层载片的6孔板中,孵育过夜。分别用AAV8-BFP6、AAV8-BFP10和AAV8-CBD6感染细胞,MOI为2E5 vg/cell,同时设置未感染作为对照组。72h后,用PBS清洗细胞2次,用GVB++ 缓冲液配制10%正常人血清,加入细胞孔中37°C孵育5min。吸出正常人血清,立即用预冷的PBS清洗细胞3次,然后用4%多聚甲醛固定15min。固定结束用PBS清洗3次,加入mouse Anti-C5b-9 + C5b-8抗体[aE11] (Abcam,ab66768)孵育2.5h。清洗后加入Goat Anti-Mouse IgG H&L (Cy3®) pre-adsorbed(Abcam,ab97035)在暗盒中孵育1.5h。清洗后加入DAPI染色1min,再清洗2次取出载片,加入一滴Fluoromount-G荧光封片剂,倒置于载玻片上,于荧光显微镜下观察MAC染色结果,各组条件下MAC染色的代表性图片如图10所示。将各组图片通过ImageJ软件处理,扣除背景后计算总荧光强度,统计结果如图11所示,结果显示相对于AAV8-CBD6,AAV8-BFP10和AAV8-BFP6显示了更高的抑制MAC沉积活性。
实施例12 携带目标基因的AAV8病毒体内转基因表达
6-8周C57BL/6小鼠,在实验室饲养3~5天。筛选双眼无异常的动物进行视网膜下腔注射,分别给予AAV8-VBD1、AAV8-VBD2、AAV8-BFP6和AAV8-BFP10,给药剂量为3E8vg/眼。视网膜下腔注射给药结束后连续2天给予涂抹氧氟沙星眼膏,每天2次。给药后1周、2周和4周,分别将动物安乐死,取眼球,加200μl/眼的PBS(含有蛋白酶抑制剂)匀浆,离心取上清。用实施例3的方法检测转基因蛋白表达。如图12所示,结果表明,各AAV分子均能实现转基因蛋白的稳定表达。
实施例13 携带目标基因的AAV8病毒抑制碘酸钠诱导的干性AMD小鼠模型
取6-8周C57BL/6J小鼠,在实验室饲养3-5天。分组视网膜下注射PBS、AAV8-CBD6、AAV8-BFP6和AAV8-BFP10,给药剂量为3E8 vg/眼,给药体积为1μl,双眼注射。21天后,通过小鼠尾静脉注射40mg/kg碘酸钠进行造模。造模后第3天和第7天进行光学相干断层扫描(OCT)和视网膜膜电位(ERG)检测,测量视网膜外核层厚度变化和ERG波幅变化。
结果如图13显示,随着时间延长,碘酸钠引起小鼠视网膜功能改变,体现为ERG暗适应b波振幅降低,与对照PBS组相比,AAV8-BFP6和AAV8-BFP10显示出显著抑制ERG波幅降低的效果;且与AAV8-CBD6相比,AAV8-BFP6和AAV8-BFP10对干性AMD小鼠模型的改善效果具有明显优势。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本专利构思的前提下,上述各实施方式还可以做出若干变形、组合和改进,这些都属于本专利的保护范围。因此,本专利的保护范围应以权利要求为准。
Claims (10)
1.一种双功能性融合蛋白,其特征在于,所述双功能性融合蛋白包含VEGF结合域、经过改造的人IgG Fc结构域和补体结合域;
所述VEGF结合域包含VEGF受体1的胞外结构域2,或VEGF受体1的胞外结构域2和VEGF受体2的胞外结构域3;
所述经过改造的人IgG Fc结构域,所采用的点突变为L234A/L235A/H435A;
所述补体结合域包含可溶性CD59结构域结构sCD59;
所述VEGF结合域与人IgG Fc结构域,补体结合域与人IgG Fc结构域,VEGF结合域与补体结合域之间直接串联或通过连接肽连接。
2.如权利要求1所述的一种双功能性融合蛋白,其特征在于,所述VEGF结合域包含VEGF受体1的胞外结构域2,所述补体结合域包含可溶性CD59结构域结构sCD59,经过改造的人IgG Fc结构域和sCD59通过连接肽(GGGGS)3连接,所述双功能性融合蛋白结构为:VEGFR1D2-mhIgG Fc-(GGGGS)3-sCD59,氨基酸序列如SEQ ID NO.2所示;或者,
所述VEGF结合域包含VEGF受体1的胞外结构域2和VEGF受体2的胞外结构域3,所述补体结合域包含可溶性CD59结构域结构sCD59,经过改造的人IgG Fc结构域和sCD59通过连接肽(GGGGS)3连接,所述双功能性融合蛋白结构为:VEGFR1D2-VEGFR2D3-mhIgG Fc-(GGGGS)3-sCD59,氨基酸序列如SEQ ID NO.1所示。
3.权利要求1或2所述双功能性融合蛋白的编码基因。
4.一种表达权利要求1或2所述双功能性融合蛋白的表达盒或重组载体,其特征在于,所述表达盒从5’-3’端包括如下式I所述结构:
E1-E2-E3-E4 (式I)
其中:E1为启动子;E2为信号肽;E3为编码权利要求1或2所述双功能性融合蛋白的核苷酸序列;E4为Poly A序列。
5.一种包含权利要求3所述编码基因或权利要求4所述表达盒或重组载体的重组宿主细胞。
6.一种包含权利要求3所述编码基因或权利要求4所述表达盒的AAV病毒,其特征在于,将所述基因或表达盒克隆至腺相关病毒AAV骨架中两个末端重复序列ITR之间,构建目的基因质粒GOI,应用于AAV包装载体系统,所述包装载体系统包括:目的基因质粒GOI,携带AAVrep、cap基因的载体以及辅助载体,上述载体通过三质粒瞬时转染生产细胞包装成为AAV病毒。
7.一种药物组合物,其特征在于,包含权利要求1或2所述的双功能性融合蛋白、权利要求3所述编码基因、权利要求5所述的重组宿主细胞、或权利要求6所述AAV病毒。
8.权利要求1-2所述双功能性融合蛋白,或权利要求3所述编码基因,或权利要求4所述表达盒,或权利要求5所述的重组宿主细胞,或权利要求6所述AAV病毒,或权利要求7所述组合物的应用。
9.如权利要求8所述的应用,其特征在于,是在抑制VEGF和/或补体途径中的应用,或是在制备治疗血管内皮生长因子和/或补体相关疾病的制剂或配方或药物组合物中的应用。
10.如权利要求9所述的应用,其特征在于,所述疾病包括眼部疾病、炎性疾病、自身免疫性疾病或肿瘤;
所述眼部疾病包括年龄相关性黄斑变性、糖尿病视网膜病变、视网膜静脉阻塞、病理性近视性脉络膜新生血管;
所述年龄相关性黄斑变性,是湿性年龄相关性黄斑变性,或者干性年龄相关性黄斑变性,或者并发有眼底血管新生的干性年龄相关性黄斑变性,或者伴随有黄斑萎缩的湿性年龄相关性黄斑变性。
