Abstract
本发明公开一种基于荧光的高通量筛选方法来收集蛋白质和聚合物之间相互作用的数据。我们提出了一个高通量平台,用于量化蛋白质和聚合物之间的相互作用,以简化用于靶向蛋白质递送的适当聚合物的选择,具体实验促进了基于聚六亚甲基双胍(PHMB)的聚合物递送系统的开发,该系统专为更广泛的通用递送应用而设计,以及专门为应用而设计的基于聚乙烯亚胺(PEI)的聚合物递送系统神经元靶向递送,这些系统已在体外和体内环境中证明了有效性,为推动基于蛋白质的尖端疗法的发展带来了巨大的希望,并建立在蛋白质递送领域相关研究的先前进展的基础上。
Description
技术领域
本发明涉及一种借助荧光以高通量的测定聚合物-蛋白质相互作用关系的方法,根据该方法简化筛选蛋白质的递送载体,属于高通量平台构建技术、蛋白质药物递送材料和生物技术领域等技术领域。
背景技术
目前,基因治疗的关键是如何高效地将基因递送到目标细胞内。基因递送载体包括病毒载体和非病毒载体。病毒载体的递送效率虽然高于非病毒载体,但其引发的免疫毒性及致癌性等潜在危害限制了其临床应用。近年来,高效、低毒、具生物相容性的非病毒基因载体逐步成为基因治疗载体研究的热点。常用的非病毒载体有阳离子脂质体与聚合物两类。其中,由于具有较高的转染效率,以Lipo2000等为代表的阳离子脂类化合物和、以线性聚乙烯亚胺(L-PEI)为代表的聚合物,都经过多年的发展和完善,已被广泛应用于体外基因转染实验。但这类化合物也有明显的缺点,比如它们都价格昂贵,其中相当便宜的L-PEI-25k其当前市场价都达到了8000元每克。此外,伴随着基因治疗的深入研究,与基因相关的蛋白质类药物也越来越成为新的治疗策略,然而,由于这些蛋白质的结构、电荷和分子量不同,将这些蛋白质递送到靶位点仍然是一个重大挑战。近年来,以聚合物为载体进行蛋白质递送的研究已然成为主要选择,归因于其多功能性和生物相容性。然而,由于庞大的聚合物多样性和蛋白质的复杂性,研究者难以寻找一种通用型的蛋白质递送载体。实际上,基于小分子以及蛋白质与蛋白质之间相互作用的高通量筛选(HTS)方法的研究或许可作为借鉴,但蛋白质-聚合物相互作用的研究相对有限。此外聚合物-蛋白质相互作用关系缺少统一数据库,并且传统的生化高通量筛选方法多是以特定蛋白质而构建的,涉及蛋白质的纯化、酶活性测定、蛋白抑制激动功能和蛋白抗原抗体测试等。这些实验往往具有价格昂贵、应用范围狭窄、实验条件要求高以及研究周期长等缺点。此外,这些方法无法简单往聚合物-蛋白质相互关系套用,因此急需能够高效快速获取聚合物-蛋白质相互关系的简便方法,以高通量筛选可递送特定蛋白质的聚合物载体,并能成功实现体内转化。
发明内容
本发明目的之一在于提供一种新的高通量聚合物-蛋白质相互作用关系的测定方法,实现高通量筛选蛋白质递送聚合物的筛选、高通量聚合物-蛋白质相互作用关系数据平台和数据库的建立。聚合物不同于小分子、蛋白质,如果能与蛋白质结合,往往会直接引起其自组装从而诱导蛋白聚集。以化学荧光标记的蛋白质作为标准物质,利用已知的荧光聚集淬灭和荧光聚集增强两类荧光原理,测定组装前后荧光标记蛋白的荧光强弱,就可以高效快速区分聚合物-蛋白间的相互作用。因此本发明以蛋白质组装前后荧光变化量为基准,构建起高通量聚合物-蛋白质相互作用关系测定方法。
具体而言,本发明的蛋白质递送用聚合物载体的筛选方法,其特征在于,对聚合物-蛋白自组装性、聚合物-蛋白作用强度、代表性细胞递送性进行评估,其包括以下步骤:
聚合物-蛋白自组装性评估步骤:构建标准化的聚合物-蛋白质荧光梯度,测定一系列聚合物组装前后荧光强度变化,
聚合物-蛋白作用强度评估步骤:40分钟以下的时间内快速结合聚合物-蛋白质,检测荧光变化值与其尺寸变化值,判定它们之间的相互作用程度,
代表性细胞递送性评估步骤:利用Hela、3T3-L1和Neuro2a细胞系评估聚合物对蛋白质的递送能力。
经过综合评估,发明人认为上述特性的组合可以代表聚合物作为蛋白体内递送载体的性能。
在本发明优选的实施方式中,聚合物-蛋白自组装性评估步骤包括如下工序:
a,第一步,定量溶解标准的荧光标记蛋白质,以BSA-FITC为例,得到多梯度浓度蛋白溶液,分别加入96孔板,准备待测;
b,第二步,将配置好的蛋白溶液送至酶标仪中测试,得到对应的荧光测定值;
c,第三步,将测定所得浓度和相应荧光测定值构建标准曲线。随后重复以不同类型荧光标记后的蛋白质,构建相应蛋白荧光测定标准曲线。
在本发明优选的实施方式中,聚合物-蛋白作用强度评估步骤包括如下工序:
a,第一步,按照变化值排列筛选聚合物,将变化绝对值[F1-0]较大的10种聚合物溶液,及其对应蛋白分别进行粒径测定得到相应值Sj和Sd;
b,第二步,将聚合物溶液与相应蛋白混合,共孵育30分钟,随后转移送至动态光散射中测试,得到对应复合物的粒径Sm;以及进行透射电镜的扫描分析以确定两者间的相互作用程度;
c,第三步,比较粒径变化并于荧光差值进行比较,得出最终筛选匹配度最高的几类聚合物进行进一步体内实验。
在本发明优选的实施方式中,代表性细胞递送性评估步骤包括如下工序:
a,第一步,培养Hela、3T3-L1和Neuro2a细胞系;
b,第二步,聚合物与FITC-BSA反应,并分别处理上述三种细胞系,共聚焦显微镜检测递送效果;
c,第三步,使用FM4-64对细胞膜染色,探究聚合物递送蛋白质是否入胞,以判别聚合物对蛋白质是否入胞递送。
在本发明优选的实施方式中,该筛选方法还包括体内递送蛋白质能力评估步骤,
a,第一步,将Cy5-BSA与所筛选的聚合物反应,随后进行小鼠腹腔注射;
b,第二步,在注射后的50小时内,选取3个时间点将小鼠麻醉,进行小动物活体成像检测,实验结束后处死并进行组织器官的成像检测;
c,第三步,统计荧光强度,并分析荧光在不同组织器官的分布情况;
d,第四步,使用筛选的聚合物向体内递送功能性蛋白质。
作为筛选方法,与现有技术相比,本发明具有如下有益效果:
1、所采用的技术手段简单,高通量实施可行性极高;
2、所应用到的测试条件温和,相应设备常见,高通量实施难度低;
3、所得结果可重复性,可拓展性高,实验参数固定,标准意义明确与聚合物-蛋白质相互作用相关;
4、所用方法可以很好的标准化,适用范围极其广泛,不局限于蛋白或聚合物种类。
5、通过体内实验验证了本发明方法的实用价值,并在细胞水平解释了对蛋白质的递送方式。
发明人根据该高通量定量蛋白质与聚合物的相互作用,成功筛选出向体内递送功能性蛋白质的聚合物载体,本发明的另一方面是提供一种聚合物-蛋白体内递送系统,其特征在于,含有基于聚六亚甲基双胍PHMB,通过与作为递送目标的蛋白质自组装结合形成复合体,通过注射将作为递送目标的蛋白质递送至体内。
本发明的第三个方面,发明人发现的聚合物PEI在细胞和体内水平表现出在神经细胞中具备一定的转染选择性,并进行了结构修饰,验证蛋白质转染中的应用价值,具体目的是通过对其进行多种结构修饰最终获得神经元靶向、高效低毒性递送的聚合物载体。具体而言,本发明还提供一种神经靶向聚合物-蛋白体内递送系统,其特征在于,含有以式(1)、式(2)、式(3)为结构单元的聚合物,该聚合物分子量为500~50000,式(1)所示结构在聚合物中所占比例为90%以上,通过与作为递送目标的蛋白质自组装结合形成复合体,通过注射将作为递送目标的蛋白质递送至体内神经的细胞,

在优选的事实方式中,Z选自氢、甲基、苯基。
本发明提供聚六亚甲基双胍PHMB以及上述聚合物作为蛋白质递送用聚合物载体的应用。
本发明中的可靶向性递送蛋白的载体,是一类水溶性聚合物通过适当化学修饰,赋予其独特功能。其与蛋白可以组装成纳米颗粒,其递送方式简单,只需物理共混后直接实用。此外,本发明提供的载体已经被验证能够在难递送的神经元细胞器模型中实现蛋白递送。此外,通过体内实验验证,我们验证了相较于已有文献报道的材料BARD及商业材料transex相比,本发明提供的载体,体内递送效率更好,应用价值更广。
本发明通过以下技术路线来实现:以支链branched PEI为例,大致的合成过程如下,应该注意的是以下的示例仅仅是为了更明确的解说,本发明的合成方法不受其限制,第一步是将支链PEI、溶解在甲醇溶剂中;待其充分溶解后,随后将3-烯丙基酰胺苯硼酸(BA),并连通氮气,充氮;随后缓慢搅拌12-24h;随后旋转蒸发除去混合液中溶剂,最终得到无色油状物。
第二步是将油状物溶解于去离子水中,随后将转移至透析袋中。去离子水透析24小时,随后转移冻干,得到最终产物PEI-BA。
本发明提供了一种基于已有聚合物进行理性化改造的方法。其中PEG修饰、胍基修饰、苯硼酸修饰均有相关关于递送改造的报道,然而,将其与特定的阳离子聚合物,如b-PEI结合是否能够达到改进递送效果的作用未见报道。且以往报道改造后的递送载体筛选往往从Hela、HEK293细胞等开始,针对难转染的神经元细胞的载体改造极少。因此,通过巧妙设计了如上所述结构及相应体外及体内实验探索,本发明最终优选出了能够用于向神经元细胞递送蛋白质且同时具备递送核酸的多功能载体。
此外,针对体内和体外实验条件的不同,本发明通过多组实验将递送载体的具体配方进行详细探索和优化,提供了更具可行性的优良载体。
本发明提供的新型载体与现有技术相比较,具有如下显著优点:
1,本发明提供的新型载体在细胞水平上,其蛋白递送效果相较能够超越或者至少相当于传统商业产品效果;
2,本发明提供的新型载体其合成过程相当简单,可快速大规模生产;
3,本发明提供的新型载体其所用原料易得,价格低廉,制备成本较传统递送材料具备较大优势
4,本发明提供的新型载体体内水平上,其蛋白递送效果能够超越大多数传统商业产品效果,且无明显副作用。

实施例
下面通过实施例对本发明进行详细描述,但并不意味着对本发明任何不利限制。本文已经详细地描述了本发明,其中也公开了其具体实施例方式,对本领域的技术人员而言,在不脱离本发明精神和范围的情况下针对本发明具体实施方式进行各种变化和改进将是显而易见的。
实施例1以BSA-FITC荧光标记蛋白为例,详细叙述蛋白质递送用聚合物载体的筛选方法的步骤
1,荧光蛋白质和聚合物的准备
以BSA-FITC荧光蛋白质为示例蛋白质(商业化试剂),随机选取了10中聚合物:
Dextran sulfate sodium(简称DSS),Pluronic F68(简称F-68),Poly-hydroxyethylmethacrylate(简称PHEMA),Poly(DL-Lactide)(简称PDLLA),Ureaformaldehyde(简称UF),Linear polyethylenimine(简称lPEI),and Branchedpolyethylenimine(简称bPEI)以及4ARM-PEG-SH,Triton X-114和Tween 20。
取适量上述聚合物溶解于PBS缓冲液混匀,置电磁搅拌器上(速度适当以不起泡沫为宜)5~10min,随后将所有待测聚合物均统一浓度为0.4mg/mL。分别加入96孔板,每种聚合物至少重复12次。
2,聚合物@荧光蛋白的荧光变化测定
a)荧光蛋白与聚合物的混合过程
首先,进行预实验以摸索合适的聚合物、蛋白质浓度及配比,按照聚合物:BSA-FTIC为1:1、1:0.5、1:0.25、1:0.125的比例计算出所需BSA-FITC质量的用分析天平准确称取所需的粉末。边搅拌边将称取的BSA-FITC溶液溶解,保证完全溶解。随后将配置好的溶液直接加入96孔板,并做好标记。结合期间应保持蛋白溶液于25℃左右,轻微振荡混匀,静置避光孵育30min。最后,根据优化的配比及浓度进行荧光蛋白质与聚合物混合。
b)混合物荧光测定
在充分混匀30min后,将96孔板转移至bio-red酶标仪,随后进行荧光读数,得到荧光数值,并进行数据分析。
3,验证聚合物@荧光蛋白的组装
根据上述测得的荧光变化数值,可将其分类成增强、不变以及降低三类,并对其进行相应的粒径测定,包括动态光散射以及透射电镜。
第一步,按照变化值排列筛选聚合物,将变化绝对值[F1-0]较大的以及未发生显著变化的聚合物溶液,及其对应蛋白分别进行粒径测定得到相应值Sj和Sd。
第二步,将聚合物溶液与相应蛋白混合,共孵育30分钟,随后转移送至动态光散射中心以及电镜中心测试,得到对应复合物的粒径Sm。
第三步,计算[Sm-Sj]和[Sm-Sd]并比较其与[F1-F0]的关系,随后根据检测结果判别蛋白质与聚合物相互作用能否形成纳米颗粒,作为下一步蛋白递送载体筛选(图1)。
4,高通量检测荧光蛋白质和聚合物的相互作用
a)荧光蛋白质准备
为尽可能增加检测蛋白质的多样性,所测试荧光蛋白质的等电点(pI)范围从3.4到10.8,分子量范围更是从12.4~240kD,共计13种蛋白质,荧光素标记的蛋白质委托公司合成。
b)聚合物的准备
测试用聚合物的纳入标准涵盖了溶解性、分子结构、分子量等商业可用的聚合物种类,其中FDA批准的聚合物种类占比约40%。
c)混合物进行荧光测定
在充分混匀30min后,将96孔板转移至bio-red酶标仪,随后进行荧光读数,得到荧光数值。
实施例2细胞水平蛋白递送能力测试
利用与实施例1相同的方法,共计测试13种荧光蛋白质分别于101种聚合物反应后的荧光变化情况,结果如图2所示(图2中仅部分列出序号1~18的聚合物的名称,19~101聚合物名称未赘述)。选择排序靠前的聚合物进行细胞水平的蛋白质递送验证实验如下:
1,细胞培养
Hela、3T3-L1和Neuro2a细胞生长条件为含10%胎牛血清、1%双抗的DMEM培养基,37℃、5%CO2培养箱,将生长状态良好的细胞传代至含盖玻片(多聚赖氨酸处理)的24孔板中,接种密度~2.5×105/孔,培养箱孵育12~16小时以使细胞充分贴壁。
2,蛋白递送
预先准备聚合物和蛋白质复合物,并使用空白DMEM将复合物稀释至10μg/mL的工作浓度,使用37℃预热的PBS缓冲液洗涤细胞2次,将上述稀释后的复合物加入24孔板中(1mL/孔),37℃转染12小时。
3,效果观察
转染结束前10min,除去培养基,使用FM4-64染色1~10min,PBS洗涤后转染效果通过共聚焦显微镜直接镜检的方式进行验证,即将培养/转染12小时的细胞取出,载玻片滴加含DAPI的封片剂、封片后镜检,根据细胞绿色荧光判别材料在神经细胞中转染蛋白质的能力(参见附图3)。
实施例3进行蛋白质体内递送效果比较
根据实施例2的结果,选择细胞水平递送效果理想的聚合物进行体内实验,需要说明的是,以下的实验选取聚六亚甲基双胍PHMB作为聚合物载体,该载体在筛选中细胞水平的递送效率靠前(在图2中的,图2c是不同化合物的筛选结果,左边的序号代表不同的聚合物,其中65号是PHMB,综合排名位于前五),具体步骤如下:
1、蛋白质体内递送
以筛选得到的PHMB为例,因其带正电荷,我们提出其能向体内的白色脂肪组织(WAT)递送蛋白质,向小鼠腹腔注射PHMB/Cy5-BSA混合物,空白对照和阴性对照Cy5-BSA,以及筛选出的另一种聚合物bPEI。
2、活体动物成像
借助PerkinElmer IVIS Spectrum系统,在小鼠完成腹腔注射的5h、12h、30h分别进行麻醉后的活体荧光成像,统计荧光强度。
3、组织荧光分布
为进一步探究聚合物递送时是否存在组织选择性,小鼠麻醉处死后分离各组织器官进行荧光成像,观测组织靶向性(参见图4)。
实施例4PHMB作为递送载体的药效实验
以筛选到的PHMB为例,尝试递送功能性蛋白Irisin以探究其能否发挥显著的促进作用。
饮食诱导的肥胖(DIO)小鼠及维持
订购肥胖小鼠,并用维持性饲料(含60%脂肪)适应饲喂2周,腹腔注射PHMB/Irisin,对照组分别为PBS、Irisin和PHMB,注射期间记录体重变化,实验末期进行糖耐量测试,实验结束后分离血清及组织器官,进行生理生化以及组织染色等以判别聚合物递送功能性蛋白质的效果(参见图5)。
实施例5聚合物的结构修饰及细胞水平递送能力的探究
以支链PEI600为骨架,合成制备PEI600-BA的制备过程,及细胞效果验证如下:
第一步,制备PEI600-BA。
首先将支链PEI600约0.2g、溶解在甲醇溶剂中;待其充分溶解后,随后将3-烯丙基酰胺苯硼酸(BA)0.15g,并连通氮气,充氮;水浴45℃,随后缓慢搅拌12-24h;随后旋转蒸发除去混合液中溶剂,最终得到无色油状物。
随后,将油状物溶解于去离子水中,随后将转移至透析袋中。去离子水透析24小时,随后转移冻干,得到最终产物PEI600-BA。
第二步,验证PEI600-BA蛋白转染效果。
首先,将生长状态良好的小鼠脑神经瘤细胞(Neuro2a,简称N2a)传代至24孔板中(约2.5×105/孔),孔内预先铺入多聚赖氨酸处理的盖玻片,置入培养箱继续培养24小时以充分贴壁。
其次,称取等质量的PEI600-BA和异硫氰酸荧光素标记的牛血清白蛋白(FITC-BSA),使用HEPES缓冲液溶解至终浓度为100mg/mL,室温静置30min以使两者形成复合物。
然后,向上述复合物中加入空白DMEM培养基至终浓度为20μg/mL,取出细胞并使用PBS(或空白DMEM)洗涤细胞2次,每孔添加1mL含复合物的空白DMEM,置于培养箱孵育12小时。
最后,蛋白转染效果通过共聚焦显微镜直接镜检的方式进行验证,即将培养/转染12小时的细胞取出,用PBS洗涤细胞5分钟/次(共3次),载玻片滴加含DAPI的封片剂、封片后镜检,根据细胞绿色荧光判别材料在神经细胞中转染蛋白质的能力。
以上的操作关键信息和结果参见附图6。
实施例6以支链PEI0.6k为骨架,合成制备PEI0.6k-BAX及细胞效果验证如下:
第一步,制备PEI0.6k-BAX。
首先将支链PEI0.6k约0.2g、溶解在甲醇溶剂中;待其充分溶解后,随后将4-(溴甲基)苯硼酸(BAX)0.18g,并连通氮气,充氮;水浴35℃,随后缓慢搅拌12-24h;随后旋转蒸发除去混合液中溶剂,最终得到无色油状物。
随后,将油状物溶解于去离子水中,随后将转移至透析袋中。去离子水透析24小时,随后转移冻干,得到最终产物PEI0.6k-BAX。
第二步,同实施例5的第二步蛋白质的递送效果探究。
实施例7体内眼部注射递送FITC-BSA的效率探究
将上述不同分子量PEI及修饰后聚合物分别进行体内递送实验探索。具体步骤如下:
第一步,制备聚合物和FITC-BSA复合物,使用生理盐水稀释复合物至10μg/μL以备体内注射。
第二步,将小鼠(C57BL6/J,6~8周龄)按体重进行腹腔注射三溴乙醇麻醉剂(0.4mL/20g)完全麻醉。同时安装好微量注射装置(共两套),一套用于从眼球吸出2μL液体,另一套安装于微量注射泵上,将注射参数设置为注射速度0.5μL/s,总量2μL。
第三步,在体式显微镜下,用小动脉血管夹夹住小鼠眼睑边缘,暴露结膜,向玻璃体内注射1-3μL上述复合物(于视网膜锯齿缘后1-2mm处),玻璃电极针尖倾斜插入眼球深度1-2mm,避免损伤晶状体。
第四步,将深度麻醉的小鼠用4%的PFA磷酸缓冲液进行心脏灌流,剪刀剪取小鼠整个头部置于4% PFA磷酸缓冲液中固定12-16小时。尔后,剥取小鼠眼球,置于30%蔗糖PBS溶液中脱水24小时直至眼球沉入底部,取出眼球,根据需要用O.C.T包埋后放于-80℃冰箱保存。
第五步,取出已包埋眼球于-20℃冰冻切片机中复温,然后进行冰冻切片,切片机设置20μm(视网膜)厚度,并立即贴片至粘附性载玻片上,37℃烤片半小时后于-80℃冰箱保存。体内转染效率通过荧光显微镜镜检评估,即将切片置于PBS缓冲液洗涤5min/次共3次,向载玻片上滴加含DAPI防荧光淬灭封片剂盖玻片封片后于显微镜镜检观察。
其结果显示PEI1.8k-BAX能够有效递送蛋白质,效果明显由于商业递送试剂,且无明显副作用。
以上结果参见附图7。
实施例8以筛选获得的1.8k的BAX为例,探究体内递送功能性重组蛋白质UbV.E4B后的促进视神经损伤后轴突再生能力
第一步,首先,视神经损伤造模采用小鼠眶内视神经夹伤手术实现,具体步骤:无菌手术室内小鼠麻醉,小心剪开结膜暴露视神经,在视盘后方1-2mm处用尖部0.1mm宽的镊子对视神经夹持5秒(注意:整个过程要尽量避开眼底层血管,防止出血以引起炎症反应),术中使用生理盐水润湿眼球,术后在眼睛上涂抹红霉素眼膏,并将其放置在加热毯上,待清醒后再放回鼠笼。
其次,将PEI1.8k-BAX和重组UbV.E4B蛋白等比混合至终浓度4μg/μL,进行小鼠眼玻璃体注射(方法参考同上)。在视神经损伤24小时后,将PEI1.8k-BAX和重组UbV.E4B蛋白复合物注射至视神经损伤小鼠的眼玻璃体内。
然后,注射2周后,小鼠注射1-2μL Alexa偶联的霍乱毒素β亚单位(CTB-555,1mg/mL)以顺行示踪再生的RGCs轴突。2~3天后,对小鼠进行麻醉灌流(方法参考同上)。
最后,视神经取材、脱水(同上),轴突再生评估通过视神经透明成像技术来实现,具体方法:配制透明化试剂,将视神经置于上述试剂中室温透明5min,后置于含DAPI的封片剂中封片、荧光显微镜下观察,并统计量化再生轴突数量以及长度以探究促再生能力。
综上所述,本发明的筛选方法,用于量化蛋白质和聚合物之间的相互作用,以简化用于靶向蛋白质递送的适当聚合物的选择。我们的方法导致从那些市售聚合物中发现了候选聚合物,它们可以提供我们感兴趣的蛋白质递送载体。此后,深入的体外实验和合理的设计促进了基于聚六亚甲基双胍(PHMB)的聚合物递送系统的开发,该系统专为更广泛的通用递送应用而设计,以及专门为应用而设计的基于聚乙烯亚胺(PEI)的聚合物递送系统神经元靶向递送。我们的体内概念验证研究提供了令人信服的证据,证明神经元特异性递送系统有效刺激视神经再生。总之,我们创新的高通量筛选方法显着加速了蛋白质递送系统候选聚合物的发现。这些系统已在体外和体内环境中证明了有效性,为推动基于蛋白质的尖端疗法的发展带来了巨大的希望,并建立在蛋白质递送领域相关研究的先前进展的基础上。

Claims (10)
1.一种蛋白质递送用聚合物载体的筛选方法,其特征在于,对聚合物-蛋白自组装性、聚合物-蛋白作用强度、代表性细胞递送性进行评估,其包括以下步骤:
聚合物-蛋白自组装性评估步骤:构建标准化的聚合物-蛋白质荧光梯度,测定一系列聚合物组装前后荧光强度变化,
聚合物-蛋白作用强度评估步骤:40分钟以下的时间内快速结合聚合物-蛋白质,检测荧光变化值与其尺寸变化值,判定它们之间的相互作用程度,
代表性细胞递送性评估步骤:利用Hela、3T3-L1和Neuro2a细胞系评估聚合物对蛋白质的递送能力。
2.根据权利要求1所述的蛋白质递送用聚合物载体的筛选方法,其特征在于,聚合物-蛋白自组装性评估步骤包括如下工序:
a,第一步,定量溶解标准的荧光标记蛋白质,以BSA-FITC为例,得到多梯度浓度蛋白溶液,分别加入96孔板,准备待测;
b,第二步,将配置好的蛋白溶液送至酶标仪中测试,得到对应的荧光测定值;
c,第三步,将测定所得浓度和相应荧光测定值构建标准曲线。随后重复以不同类型荧光标记后的蛋白质,构建相应蛋白荧光测定标准曲线。
3.根据权利要求1所述的蛋白质递送用聚合物载体的筛选方法,其特征在于,聚合物-蛋白作用强度评估步骤包括如下工序:
a,第一步,按照变化值排列筛选聚合物,将变化绝对值[F1-0]较大的10种聚合物溶液,及其对应蛋白分别进行粒径测定得到相应值Sj和Sd;
b,第二步,将聚合物溶液与相应蛋白混合,共孵育30分钟,随后转移送至动态光散射中测试,得到对应复合物的粒径Sm;以及进行透射电镜的扫描分析以确定两者间的相互作用程度;
c,第三步,比较粒径变化并于荧光差值进行比较,得出最终筛选匹配度最高的几类聚合物进行进一步体内实验。
4.根据权利要求1所述的蛋白质递送用聚合物载体的筛选方法,其特征在于,代表性细胞递送性评估步骤包括如下工序:
a,第一步,培养Hela、3T3-L1和Neuro2a细胞系;
b,第二步,聚合物与FITC-BSA反应,并分别处理上述三种细胞系,共聚焦显微镜检测递送效果;
c,第三步,使用FM4-64对细胞膜染色,探究聚合物递送蛋白质是否入胞,以判别聚合物对蛋白质是否入胞递送。
5.根据权利要求1所述的蛋白质递送用聚合物载体的筛选方法,其特征在于,该筛选方法还包括体内递送蛋白质能力评估步骤,
a,第一步,将Cy5-BSA与所筛选的聚合物反应,随后进行小鼠腹腔注射;
b,第二步,在注射后的50小时内,选取3个时间点将小鼠麻醉,进行小动物活体成像检测,实验结束后处死并进行组织器官的成像检测;
c,第三步,统计荧光强度,并分析荧光在不同组织器官的分布情况;
d,第四步,使用筛选的聚合物向体内递送功能性蛋白质。
6.一种聚合物-蛋白体内递送系统,其特征在于,含有基于聚六亚甲基双胍PHMB,通过与作为递送目标的蛋白质自组装结合形成复合体,通过注射将作为递送目标的蛋白质递送至体内。
7.一种神经靶向聚合物-蛋白体内递送系统,其特征在于,含有以式(1)、式(2)、式(3)为结构单元的聚合物,该聚合物分子量为500~50000,式(1)所示结构在聚合物中所占比例为90%以上,通过与作为递送目标的蛋白质自组装结合形成复合体,通过注射将作为递送目标的蛋白质递送至体内神经的细胞,
R表示以下基团中的一种,
其中Z表示为氢、C1~C5的烷基、C6~C10的芳基、C1~C5的烷基硫醚基。
8.根据权利要求7所述的神经靶向聚合物-蛋白体内递送系统,其特征在于,
该聚合物分子量为5000~20000,Z选自氢、甲基、苯基。
9.聚六亚甲基双胍PHMB作为蛋白质递送用聚合物载体的应用。
10.以式(1)、式(2)、式(3)为结构单元的聚合物作为蛋白质递送用聚合物载体的应用,所述聚合物分子量为500~50000,式(1)所示结构在聚合物中所占比例为90%以上,
R表示以下基团中的一种,
其中Z表示为氢、C1~C5的烷基、C6~C10的芳基、C1~C5的烷基硫醚基,优选的是该聚合物分子量为5000~20000,Z选自氢、甲基、苯基。
