Abstract
本发明属于脂质纳米胶束药物制剂技术领域,公开了基于抑制肿瘤COX‑2和CXCR4的共载脂质纳米胶束及制备方法。在超声条件下,取磷脂和COX‑2抑制剂溶于无水乙醇中,作为有机相;将甘氨胆酸加入到水性溶液中,并使用氢氧化钠溶液使其溶解,作为水相;将有机相溶液缓慢滴加到水相溶液中,除去乙醇后,超声得到载COX‑2抑制剂的胶束溶液;将CXCR4拮抗剂水溶液加入的胶束溶液,过滤膜,得到共载COX‑2抑制剂和CXCR4拮抗剂的脂质纳米胶束。本发明能够更高效地实现肿瘤渗透,制备方法简便、经济、有效,具有良好的包封率、载药量和稳定性。

技术领域
本发明属于脂质纳米胶束药物制剂,尤其涉及基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束及制备方法。
背景技术
恶性肿瘤是影响人类健康的典型疾病之一。肿瘤微环境(TME)作为肿瘤细胞生长、侵袭和转移所必需的土壤条件,是一个复杂的网络系统,包括细胞成分和非细胞成分,如肿瘤细胞、炎性细胞、成纤维细胞、免疫细胞及分泌的各种细胞因子等。各成分之间密切联系,促进了肿瘤的生长和发展。在多数癌症中,炎症又是肿瘤微环境的重要特征,影响着肿瘤的发生与转移。
炎症是肿瘤的重要特征之一。慢性炎症可引发并促进肿瘤的发展。长期慢性炎症能够引起人体正常细胞基因突变形成癌基因,促使细胞癌变并引发癌症。慢性炎症又激活肿瘤细胞中的关键炎症通路,促进多种炎症因子的分泌,实现对多种炎性细胞、免疫抑制细胞的招募,释放大量的肿瘤相关性炎症信号,从而产生促肿瘤炎症(Tumor-promotinginflammation),使其成为肿瘤生长的帮凶,促进肿瘤发展、转移以及实现免疫逃逸。《Nature Reviews Clinical Oncology》杂志也刊文指出,促肿瘤炎症能够降低机体固有免疫并抑制其适应性免疫应答,是肿瘤治疗失败的重要因素。因此,通过改善肿瘤炎症,降低肿瘤生长与转移,可作为肿瘤研究领域的重要科学问题。
环氧合酶-2(cyclooxygenase-2,COX-2)是一种诱导酶,可以促进促炎介质前列腺素2(PGE2)产生。COX-2/PGE2信号能够招募巨噬细胞、髓源性抑制细胞、肥大细胞等炎性细胞进入肿瘤组织,诱导分泌促肿瘤相关炎症因子和免疫抑制因子,进而促进炎症的发展。另外,COX-2还能够促进细胞增殖、抑制细胞凋亡、促进血管形成,参与肿瘤的发生与发展。有报道称其在不同类型的癌症中过度表达,包括乳腺癌、结肠癌、肺癌、黑色素瘤和前列腺癌等。因此,COX-2有望成为炎性肿瘤的治疗靶点。
CXCR4是趋化因子CXCL12的功能受体,能够通过作用于白细胞来调节炎症和免疫过程。CXCL12/CXCR4生物轴能够促进异源三聚体G蛋白的释放,进而激活细胞内各种信号转导途径和下游效应器,促进肿瘤细胞生长与转移、血管生成、上皮间充质转化、促进肿瘤炎症和免疫抑制。CXCR4作为炎性介质又能招募巨噬细胞、髓源性抑制细胞等炎症细胞进入肿瘤。研究发现,CXCR4在许多实体瘤和血液肿瘤中高表达,是肿瘤诊断和治疗的有效靶点。
脂质纳米胶束是由两亲性材料自组装而成的纳米结构,与生物膜类似,具有良好的生理环境稳定性和生物相容性,并能够有效装载难溶性药物。通过改变材料的组合,可以实现胶束对药物的最佳装载,并提高稳定性。聚合物胶束由于其体积较小,在生物体内具有良好的通透性和滞留效应,并能进行配体修饰而功能化,因此在药物传递方面具有广泛应用。
通过上述分析,现有技术存在的问题及缺陷为:现有技术CXCR4拮抗剂和COX-2抑制剂改善肿瘤相关炎症以及抑制肿瘤细胞的增殖与转移效果差。
发明内容
为克服相关技术中存在的问题,本发明公开实施例提供了基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束及制备方法,本发明的目的就是为了针对改善肿瘤炎症、克服游离药物难溶性问题而提供一种基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束及其制备方法,可实现CXCR4拮抗剂和COX-2抑制剂特异性靶向释放,改善肿瘤相关炎症以及抑制肿瘤细胞的增殖与转移,提高炎症肿瘤的治疗效果。
所述技术方案如下:基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束的制备方法,该方法采用次序包载的方法,将具有截然不同性质的两种药物进行包载,具体包括以下步骤:
S1,在超声条件下,取磷脂和COX-2抑制剂溶于无水乙醇中,作为有机相;所述COX-2抑制剂与磷脂的质量比为1:5~1:25,磷脂浓度范围为10~30mg/ml;
S2,在超声条件下,将甘氨胆酸加入到水性溶液中,并使用氢氧化钠溶液使其溶解,作为水相;其中,甘氨胆酸与磷脂的质量比为2:1~1:2;所述水性溶液为:纯化水、磷酸盐缓冲液、生理盐水、葡萄糖溶液的一种;
S3,在温度为30~75℃条件下,将有机相溶液缓慢滴加到水相溶液中,除去乙醇后,超声得到载COX-2抑制剂的胶束溶液;
S4,在振荡涡旋条件下,将CXCR4拮抗剂水溶液加入胶束溶液,过滤膜,得到共载COX-2抑制剂和CXCR4拮抗剂的脂质纳米胶束;所述CXCR4拮抗剂与磷脂的质量比为1:10-1:25,涡旋时间3~15min。
在步骤S1中,所述磷脂选用天然磷脂或选用合成磷脂,所述天然磷脂包括大豆卵磷脂、蛋黄卵磷脂、氢化大豆磷脂中的任意一种或多种,
所述合成磷脂包括二棕榈酰磷脂酰胆碱DPPC、二油酰磷脂酰甘油DOPG、二硬脂酰磷脂酰胆碱DSPC中的任意一种或多种;
所述COX-2抑制剂为塞来昔布、帕瑞昔布、尼美舒利、美洛昔康、依托考昔中的任意一种。
在步骤S2中,所述水性溶液加入甘氨胆酸后的混合溶液的pH值范围为5.0~6.5;氢氧化钠使用浓度为0.1~1M。
在步骤S3中,有机相和水相溶液的体积比1:2~1:10,转速范围10~100rpm,除去乙醇方法通过真空旋转蒸发或常压下敞口挥发。
进一步,所述的CXCR4拮抗剂为普乐沙福AMD3100、MotixafortideBKT140、MSX-122中的任意一种。
本发明的另一目的在于提供一种基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束,所述基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束利用所述制备方法制备而成,正电性的CXCR4拮抗剂采用静电力作用吸附在共载脂质纳米胶束表面;疏水性的COX-2抑制剂通过物理包封形式装载在共载脂质纳米胶束内部;通过被动靶向进入肿瘤组织后,CXCR4拮抗剂主动靶向CXCR4高表达的肿瘤细胞并从共载脂质纳米胶束表面脱落,进而释放COX-2抑制剂抑制PGE2生成,COX-2抑制剂和CXCR4拮抗剂协同降低炎症因子的表达,抑制炎症细胞、免疫抑制细胞的招募,改善促肿瘤炎症。
结合上述的所有技术方案,本发明所具备的优点及积极效果为:本发明的制备过程中,使用氢氧化钠调节pH主要作用是使甘氨胆酸溶解,以离子形式存在。若不加氢氧化钠调节酸度,甘氨胆酸不会在PBS溶液中溶解,无法制备纳米胶束。超声有助于胶束的粒径稳定。温度过高或过低都会导致产率下降。
甘氨胆酸酸性很强,pH值很低,如果不加氢氧化钠调pH值的话,根本就无法溶解,制备也就没法继续往下进行。pH值必须要在合适的范围。在本发明中,甘氨胆酸既是作为形成脂质纳米胶束的载体材料,还可与CXCR4拮抗剂通过静电结合,得到“药物-甘氨胆酸”的成盐形式,保证药物稳定地吸附在胶束纳米颗粒的表面。
本发明合成脂质纳米胶束,改善药物的溶解度,降低药物的毒副作用。纳米胶束因为其具有合适的粒径,可以通过渗透增强和滞留(EPR)效应在肿瘤部位蓄积,实现药物的靶向释放,具有良好体内安全性。设计并合成负电性的纳米胶束,具有良好血液相容性,通过静电作用实现与药物的物理连接,通过受体结合实现对肿瘤组织的主动靶向。
本发明合成负电性的脂质纳米胶束,COX-2抑制剂为疏水性药物,通过物理包封形式包载于脂质纳米胶束内部;CXCR4拮抗剂呈正电性,与负电性脂质纳米胶束通过静电吸附连接,在体内具有良好的生物安全性,通过EPR效应到达肿瘤部位,通过静电结合的CXCR4拮抗剂可以主动靶向CXCR4高表达的肿瘤细胞,由于受体与配体的结合作用力大于CXCR4拮抗剂与负电性胶束的静电吸引力,从而实现AMD在与肿瘤细胞上CXCR4靶向结合后,从胶束上响应性脱落。本发明的共载脂质纳米胶束可充分发挥COX-2抑制剂与CXCR4拮抗剂的协同治疗作用,能够同时改善肿瘤炎症,抑制细胞增殖和转移,提高癌症治疗效果。
与现有技术相比,本发明制备的共载脂质纳米胶束的平均粒径可控制在10~50nm左右,能够更高效地实现肿瘤渗透,制备方法简便、经济、有效,具有良好的包封率、载药量和稳定性。

为使本发明的上述目的、特征和优点能够更加明显易懂,下面结合附图对本发明的具体实施方式做详细的说明。在下面的描述中阐述了很多具体细节以便于充分理解本发明。但是本发明能够以很多不同于在此描述的其他方式来实施,本领域技术人员可以在不违背本发明内涵的情况下做类似改进,因此本发明不受下面公开的具体实施的限制。
本发明实施例提供的基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束及制备方法创新点在于:本发明提出了通过抑制肿瘤COX-2和CXCR4改善促肿瘤炎症的新的治疗策略;采用次序包载的方法将具有截然不同性质的两种药物进行包载;将正电性药物在纳米颗粒表面上与载体生成“药物-甘氨胆酸”盐形,保证了药物在胶束上稳定性;所制备脂质纳米胶束粒径在10~50nm范围内,能够增加在肿瘤部位的分布,表现出优异的抗肿瘤效果。该技术路线简便、经济、有效,脂质纳米胶束具有良好的包封率、载药量和稳定性,具有较高的转化潜力。
实施例1,本发明实施例提供的基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束,作为一种能够改善肿瘤炎症的共载脂质纳米胶束制剂,其具有合适粒径、且能够包载两种不同性质的药物。
本发明创新的限制了两种药物的类型,同时作用于COX-2和CXCR4。
实施例2,如图1所示,本发明实施例还提出了一种基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束的制备方法,可以改善肿瘤炎症,包括以下步骤:
S1,在超声条件下,取磷脂和COX-2抑制剂溶于无水乙醇中,作为有机相;所述COX-2抑制剂与磷脂的质量比为1:5~1:25,磷脂浓度范围为10~30mg/ml;
S2,在超声条件下,将甘氨胆酸加入到水性溶液中,并使用氢氧化钠溶液使其溶解,作为水相;其中,甘氨胆酸与磷脂的质量比为2:1~1:2;所述水性溶液为:纯化水、磷酸盐缓冲液、生理盐水、葡萄糖溶液的一种;
S3,在温度为30~75℃条件下,将有机相溶液缓慢滴加到水相溶液中,除去乙醇后,超声得到载COX-2抑制剂的胶束溶液;
S4,在振荡涡旋条件下,将CXCR4拮抗剂水溶液加入胶束溶液,过滤膜,得到共载COX-2抑制剂和CXCR4拮抗剂的脂质纳米胶束;所述CXCR4拮抗剂与磷脂的质量比为1:10-1:25,涡旋时间3~15min。
在步骤S4中,过450nm、200nm滤膜,得到共载COX-2抑制剂和CXCR4拮抗剂的脂质纳米胶束。
可以理解,步骤S1-步骤S3为制备脂质纳米胶束的过程,在形成胶束的同时包载COX-2抑制剂药物;
传统上包载两种药物往往采用同时包载,这就要求两种药物的性质相同或相近(如都是疏水性的)。而本发明中采用次序包载的方法,可以实现具有截然不同性质的两种药物的包载。
在步骤S1中,COX-2抑制剂与磷脂的质量比为1:5~1:25,磷脂浓度范围在10~30mg/ml;
本发明的制备方法步骤S2中,所述水性溶液加入甘氨胆酸后的混合溶液的pH值范围为5.0~6.5;
在本发明中,氢氧化钠可使用0.1~1M浓度范围,以滴定后溶液pH值到合适范围为终点,其用量并不固定;
有机相和水相溶液的体积比1:2~1:10,转速范围10~100rpm,温度为30~75℃,除去乙醇方法可通过真空旋转蒸发或常压下敞口挥发。
可以理解,步骤S4技术作用为:CXCR4拮抗剂为正电性物质,通过静电力作用吸附在脂质纳米胶束的表面。
由于磷脂为中性,甘氨胆酸具有较强的荷电性(加入氢氧化钠后成盐),因此CXCR4拮抗剂是吸附在甘氨胆酸的表面,二者生成“药物-甘氨胆酸”盐形。
其中,CXCR4拮抗剂与磷脂(或甘氨胆酸)的比例选择中,如果药物太多,就超出了胶束所带负电荷的静电吸附能力,造成多余的药物浪费无法负载。
本发明所述的共载脂质纳米胶束,其材料由磷脂、甘氨胆酸组成。其中,所述磷脂,可选用天然磷脂,如大豆卵磷脂、蛋黄卵磷脂、氢化大豆磷脂中的任意一种或多种,也可选用合成磷脂,如二棕榈酰磷脂酰胆碱(DPPC)、二油酰磷脂酰甘油(DOPG)、二硬脂酰磷脂酰胆碱(DSPC)中的任意一种或多种。
可以理解,磷脂是制备脂质纳米胶束的必需材料。磷脂是一大类的统称,有具体不同的品种。任何一种磷脂材料都是可以制备得到该脂质纳米胶束。
本发明所述的共载脂质纳米胶束,能够同时包载两种不同性质的药物。两种药物分别为CXCR4拮抗剂和COX-2抑制剂。
本发明中共载脂质纳米胶束所包载的CXCR4拮抗剂和COX-2抑制剂中,CXCR4拮抗剂为正电性物质,通过静电力作用吸附在脂质纳米胶束的表面;COX-2抑制剂为疏水性物质,通过物理包封形式装载在脂质纳米胶束的内部。
在本发明所述的COX-2抑制剂,可以为塞来昔布、帕瑞昔布、尼美舒利、美洛昔康、依托考昔等中的任意一种;所述的CXCR4拮抗剂,可以为普乐沙福(AMD3100)、Motixafortide(BKT140)、MSX-122等中的任意一种。
可以理解,两种药物包载的方式不同。采用何种方式将药物包载在胶束内,是要根据药物性质来选择的。CXCR4拮抗剂因为是为正电性的,所以采用静电力作用(正负电吸引)载药,实际上药物是通过生成“药物-甘氨胆酸”盐形,稳定吸附在脂质纳米胶束表面的;而COX-2抑制剂由于是疏水性药物,则通过物理包封形式装载在脂质纳米胶束的内部。
如果在制备方法中,均同时采用物理包封形式包载两药,则CXCR4拮抗剂包封率低(对比例1);如果均采用静电吸附方式,则COX-2抑制剂因为不是正电性就会包载不上。
在本发明实施例中,脂质纳米胶束装载COX-2抑制剂和CXCR4拮抗剂,通过被动靶向进入肿瘤组织后,CXCR4拮抗剂可主动靶向CXCR4高表达的肿瘤细胞并从胶束表面脱落,进而释放COX-2抑制剂抑制PGE2生成,二者协同可降低炎症因子的表达,抑制炎症细胞、免疫抑制细胞的招募,改善促肿瘤炎症。
通过上述实施例可知,本发明技术路线简便,所选用载体材料磷脂和甘氨胆酸具有良好安全性和生物相容性,制备得到的脂质纳米胶束粒径均一、稳定性良好,具有较高的工艺转化潜力。肿瘤炎症是几乎所有实体肿瘤所共性的病理特征,因此,抑制肿瘤COX-2和CXCR4改善促肿瘤炎症的治疗具有普适性。由于我国恶性肿瘤患者患病基数大,本发明如获转化,将具有极高的市场价值。
本发明所提出的联合治疗策略具有创新性。目前针对恶性肿瘤的临床推荐指南中,尚无针对改善促肿瘤炎症的临床治疗方案,本发明通过抑制肿瘤COX-2和CXCR4改善促肿瘤炎症,改变了传统针对肿瘤细胞的治疗理念,可拓宽临床肿瘤治疗策略,为临床肿瘤治疗提供新思路。其次,在本发明的具体技术方案中,胶束中的甘氨胆酸与CXCR4拮抗剂通过静电作用结合后,得到“药物-甘氨胆酸”的成盐形式,保证了药物能够稳定地吸附在胶束纳米颗粒的表面。将药物在纳米颗粒上与载体成盐的设计,具有创新性。
利用纳米胶束将截然不同性质药物的进行共包载,是本发明解决的重要技术难题。现有技术更多是针对同种性质的药物同时包载,而本发明的技术方案实现了同时对疏水性药物和亲水正电性药物的包载。在将正电性药物包载到胶束表面时,生成“药物-甘氨胆酸”盐形,保证了药物在胶束上稳定性。本发明技术方案集两种药物包载手段于一体,从而为两种类型药物充分发挥联用协同效果提供了保证。
实施例3,本发明实施例提供一种基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束,所述共载纳米胶束为共载塞来昔布/普乐沙福脂质纳米胶束,由以下组分制备而成:蛋黄卵磷脂120mg、甘氨胆酸70mg、塞来昔布10mg、普乐沙福10mg。
上述共载脂质纳米胶束的制备方法,包括以下步骤:首先将蛋黄卵磷脂、塞来昔布溶解于2mL无水乙醇中,将甘氨胆酸溶解在8ml PBS溶液中,并用氢氧化钠溶液,调节pH值至5.5左右。在转速20rpm、温度60℃条件下,将乙醇混合溶液逐滴注入到甘氨胆酸的PBS溶液中,敞口挥发1h除去乙醇,得到负电性的载塞来昔布纳米胶束;然后将普乐沙福与上述负电性纳米胶束混合并涡旋15min,过450nm、200nm滤膜各3次,得到共载塞来昔布/普乐沙福脂质纳米胶束。
所制备的塞来昔布/普乐沙福共载脂质纳米胶束,其平均粒径为26.77nm(图2),zeta电位为-11.4mV(图3),透射电镜(TEM)照片显示胶束纳米颗粒呈规整的圆球形(图4)。使用10%曲拉通破乳并用HPLC测定药物含量,共载脂质纳米胶束中塞来昔布的浓度为0.93mg/ml,包封率78.9%,载药量3.9%;普乐沙福的浓度为0.72mg/ml,包封率61.0%,载药量3.2%,见制备的共载脂质纳米胶束的包封率载药量数据表1;

将纳米胶束分别用培养基(RPMI-1640)和PBS(pH=7.4)稀释10倍,过除菌膜后分别分装于7个离心管中,监测14天粒径变化。脂质纳米胶束在RPMI-1640和PBS(pH=7.4)中,14天内的粒径大小变化可以忽略不计,初步稳定性良好(图5)。
采用透析袋法测定药物在胶束溶液中的体外释放情况。游离药物表现出明显的突释现象,4h后释放量超过70%(如图7),相比之下,本发明胶束释放明显较慢,这有利于药物的持续释放(图6)。
考察纳米胶束载体的细胞溶血性能。不同浓度下纳米载体的溶血率均低于5%,说明载体不会引起红细胞破裂,具有良好的生物安全性(图8)。
考察共载脂质纳米胶束的小鼠体内抗肿瘤效果。与单载胶束组和两种游离药混合组相比,共载制剂组肿瘤体积最小,表现出良好的抑瘤效果(图9)。
实施例4,与实施例3相比,绝大部分都相同,除了本实施例中塞来昔布替换为尼美舒利,普乐沙福替换为Motixafortide。
经检测,共载脂质纳米胶束平均粒径32.14nm。尼美舒利的浓度为0.90 mg/ml,包封率68.4%,载药量3.4%;Motixafortide的浓度为0.66 mg/ml,包封率50.6%,载药量2.5%。
实施例5,与实施例3相比,绝大部分都相同,除了本实施例中甘氨胆酸与磷脂的比例调整为1:1.1,即:蛋黄卵磷脂90mg、甘氨胆酸100mg。
经检测,所制备的共载脂质纳米胶束粒径有所降低,平均粒径18.33nm。塞来昔布浓度为0.96mg/ml,包封率82.7%,载药量4.1%;普乐沙福浓度为0.68 mg/ml,包封率58.4%,载药量2.9%。
实施例6,与实施例3相比,绝大部分都相同,除了本实施例中蛋黄卵磷脂替换为二棕榈酰磷脂酰胆碱,溶解甘氨胆酸使用5%葡萄糖水性溶液。
经检测,共载脂质纳米胶束平均粒径22.14nm。塞来昔布浓度为1.06mg/ml,包封率81.2%,载药量4.0%;普乐沙福浓度为0.81mg/ml,包封率62.2%,载药量3.0%。
实施例7,与实施例3相比,绝大部分都相同,除了本实施例中塞来昔布替换为美洛昔康,调整其与磷脂的质量比为1:20,且温度控制在30℃、通过旋蒸抽真空除去乙醇。
经检测,共载脂质纳米胶束平均粒径29.33nm。美洛昔康的浓度为0.61mg/ml,包封率87.6%,载药量2.6%;普乐沙福浓度为0.75mg/ml,包封率65.1%,载药量3.2%。
对比例1,与实施例3相比,绝大部分都相同,除了将两种药物同时溶解于无水乙醇中作为有机相。
经测定,该方法制备的共载脂质纳米胶束普乐沙福的包封率低于10%,此外zeta电位为18.96mV,接近于未载药的空白纳米胶束的zeta电位,表面仅少量普乐沙福吸附在胶束表面。可见,COX-2抑制剂和CXCR4拮抗剂的加入顺序对本发明中的共载脂质纳米胶束的表面性质有重要影响。
对比例2,与实施例3相比,绝大部分都相同,除了本实施例中塞来昔布与磷脂的质量比为1:4。即加入磷脂120mg、塞来昔布30mg。
该方法得到的脂质纳米胶束有明显沉淀。可见,较大量的药物超出了脂质纳米胶束的包载容量,说明COX-2抑制剂的剂量是影响共载脂质纳米胶束的重要因素。
对比例3,与实施例3相比,绝大部分都相同,除了本实施例中pH未在5.0-6.5之间。
当pH=4.0时,无法得到稳定的脂质纳米胶束;pH>6.5时,脂质纳米胶束表面电荷不稳定,影响普乐沙福的静电吸附作用。可见,合适的PH范围影响共载脂质纳米胶束的稳定性和CXCR4的静电吸附。
对比例4,与实施例3相比,绝大部分都相同,除了本实施例中甘氨胆酸与磷脂的质量比为4:1,不在2:1~1:2的优化范围内,即加入蛋黄磷脂40mg,甘氨胆酸160mg。
该方法所制备的脂质纳米胶束粒径分布不均匀,呈现双峰现象,PDI>0.3。可见,甘氨胆酸与磷脂的质量比也是制备得多粒径均匀脂质纳米胶束的重要影响条件。
通过上述实施例可以获知,对比例2是磷脂与药物的比例、对比例3是pH值范围的影响、对比例4是磷脂与甘氨胆酸的比例。如果不在本发明实施例3-7提供的参数范围内,就会制备失败。
在上述实施例中,对各个实施例的描述都各有侧重,某个实施例中没有详述或记载的部分,可以参见其它实施例的相关描述。
为进一步说明本发明实施例相关效果,进行如下实验。
纳米胶束因为其具有合适的粒径,可以通过渗透增强和滞留(EPR)效应在肿瘤部位蓄积,实现药物的靶向释放,具有良好体内安全性。如图10小动物成像药物体内分布的数据图,可知脂质胶束能够增加药物在肿瘤部位的蓄积分布。
本发明合成负电性的脂质纳米胶束,COX-2抑制剂为疏水性药物,通过物理包封形式包载于脂质纳米胶束内部;CXCR4拮抗剂呈正电性,与负电性脂质纳米胶束通过静电吸附连接,在体内具有良好的生物安全性,通过EPR效应到达肿瘤部位,通过静电结合的CXCR4拮抗剂可以主动靶向CXCR4高表达的肿瘤细胞,由于受体与配体的结合作用力大于CXCR4拮抗剂与负电性胶束的静电吸引力,从而实现AMD在与肿瘤细胞上CXCR4靶向结合后,从胶束上响应性脱落。本发明的共载脂质纳米胶束可充分发挥COX-2抑制剂与CXCR4拮抗剂的协同治疗作用,能够同时改善肿瘤炎症,抑制细胞增殖和转移,提高癌症治疗效果。
COX-2抑制剂与CXCR4拮抗剂两种药物包载方式是根据药物性质决定的,实验能证明本制备方法中两种药物的包封率和载药量,表明药物已经包载成功了。CXCR4拮抗剂与肿瘤细胞CXCR4的主动靶向能力,最终的效益是提高治疗效果如图9所示。
以上所述,仅为本发明较优的具体的实施方式,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,都应涵盖在本发明的保护范围之内。
Claims (2)
1.一种基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束的制备方法,其特征在于,该方法采用次序包载的方法,将具有截然不同性质的两种药物进行包载,具体包括以下步骤:
S1,在超声条件下,取磷脂和COX-2抑制剂溶于无水乙醇中,作为有机相;所述COX-2抑制剂与磷脂的质量比为1:5~1:25,磷脂浓度范围为10~30mg/ml;
S2,在超声条件下,将甘氨胆酸加入到水性溶液中,并使用氢氧化钠溶液使其溶解,作为水相;其中,甘氨胆酸与磷脂的质量比为2:1~1:2;所述水性溶液为:纯化水、磷酸盐缓冲液、生理盐水、葡萄糖溶液的一种;
S3,在温度为30~75℃条件下,将有机相溶液缓慢滴加到水相溶液中,除去乙醇后,超声得到载COX-2抑制剂的胶束溶液;
S4,在振荡涡旋条件下,将CXCR4拮抗剂水溶液加入胶束溶液,过滤膜,得到共载COX-2抑制剂和CXCR4拮抗剂的脂质纳米胶束;所述CXCR4拮抗剂与磷脂的质量比为1:10-1:25,涡旋时间3~15min;
所述磷脂选用天然磷脂或选用合成磷脂,所述天然磷脂包括大豆卵磷脂、蛋黄卵磷脂、氢化大豆磷脂中的任意一种或多种,
所述合成磷脂包括二棕榈酰磷脂酰胆碱DPPC、二油酰磷脂酰甘油DOPG、二硬脂酰磷脂酰胆碱DSPC中的任意一种或多种;
所述COX-2抑制剂为塞来昔布、尼美舒利、美洛昔康中的任意一种;
在步骤S2中,所述水性溶液加入甘氨胆酸后的混合溶液的pH值范围为5.0~6.5;氢氧化钠使用浓度为0.1~1M;
在步骤S3中,有机相和水相溶液的体积比1:2~1:10,转速范围10~100rpm,除去乙醇方法通过真空旋转蒸发或常压下敞口挥发;
所述的CXCR4拮抗剂为普乐沙福、Motixafortide中的任意一种。
2.一种基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束,其特征在于,所述基于抑制肿瘤COX-2和CXCR4的共载脂质纳米胶束利用权利要求1所述制备方法制备而成,正电性的CXCR4拮抗剂采用静电力作用吸附在共载脂质纳米胶束表面;疏水性的COX-2抑制剂通过物理包封形式装载在共载脂质纳米胶束内部;通过被动靶向进入肿瘤组织后,CXCR4拮抗剂主动靶向CXCR4高表达的肿瘤细胞并从共载脂质纳米胶束表面脱落,进而释放COX-2抑制剂抑制PGE2生成,COX-2抑制剂和CXCR4拮抗剂协同降低炎症因子的表达,抑制炎症细胞、免疫抑制细胞的招募,改善促肿瘤炎症。
