技术领域
本发明涉及犊牛养殖技术领域,具体涉及一种防治犊牛转场运输应激组合物及其制备方法。
背景技术
应激(stress)是机体受到外界或内部非特异的有害因子的刺激所表现的防御反应。常见应激类型包括运输应激、热应激、天气应激、断水断料应激、饲料变更应激等。对于育肥牛养殖来讲,运输应激是危害最大的应激。
运输应激是养牛生产过程中常见的一种应激类型,我国地缘辽阔,不同地区饲养的牛种类不同,一般犊牛出生后至半岁龄是一个保犊期,之后发往全国各地进行育肥,运输距离短则数百公里,长的能达3000km以上,即使全程高速运输也需要1~2d时间才能到达目的地,期间牛一般不饮水,不喂料,全依靠机体自身抵抗力来对抗运输带来的身体反应。在温度和湿度相对适宜的天气下,运输应激造成的危害一般不大,只要犊牛不存在基础性疾病,一般都能安全到达新场。但若是遇到高温、高湿、高寒等天气,则运输应激往往较大,有些牛甚至在运输途中就出现死亡,有些牛在到达新场后短时间内无法适应,最终也会死亡,给养殖户带来非常大的经济损失。
运输应激对牛有着诸多的危害,这些危害一般表现在疾病的发生和对后期生长发育的影响上,应激反应越大,对牛的生产性能影响就越大。
首先,运输应激虽然是暂时性的应激,但能导致机体免疫力出现暂时性下降,从而使体内条件致病原乘机繁殖。长途运输导致机体免疫力下降后,这些受抑制的病原体被解除,乘机大量繁殖,寄生部位也从上呼吸道向下迁移,到达支气管、肺,最终使病牛出现支气管肺炎病。还有些大肠杆菌、沙门氏菌、魏氏梭菌等病原主要分布在肠腔中,使得动物表现出消化道症状。
其次,过长时间的应激会使瘤胃微生态平衡被打破。一方面运输过程对中枢神经影响较大,进一步抑制了瘤胃的正常蠕动和消化。另一方面牛到达新场后必然会出现饲料变更,瘤胃菌群需要重新适应新的日粮结构,而调整过程中会导致牛出现消化道症状,比如腹泻、消化不良等。
再次,运输后的牛因应激影响,通常在10~15d内生长是基本停滞的,从而对生长发育造成影响。这种影响一方面和犊牛运输前后减料饲喂有较大关系,另一方面运输应激造成瘤胃菌群紊乱后,短时间内无法恢复,从而造成饲料的消化吸收利用率下降。再者,牛变更新环境后需要对场地环境、饲养员、兽医、饲料等有个熟悉过程,需要机体重新适应。
目前常见的防治运输应激的措施为采用镇静安定作用的化学药物来缓冲运输应激反应,如中国专利文献CN201110409655.0提供了一种适用于缓解商业动物特别是水牛公牛长途运输产生应激反应的混合药物处理方法,其特征包括:在运输动物运输前的第2小时给被运输的动物施用盐酸氯丙嗪1mg/kg体重+酒精度50°(V/V)的白酒1g/kg体重或盐酸氯丙嗪1mg/kg体重+酒精度50°(V/V)的白酒2g/kg体重或盐酸氯丙嗪1mg/kg体重+酒精度50°(V/V)的白酒3g/kg体重;但该方法使用化学药物易造成药物残留,还易造成牛胃、肾、肝等内脏的损害,不利于牛的生长。
发明内容
为了解决现有技术存在的不足,本发明的目的在于提供一种防治犊牛转场运输应激组合物及其制备方法,所得组合物能够提升犊牛的肝肾功能,调节矿物质元素的代谢,帮助牛体纠正平衡失调,提高犊牛机体免疫机能,降低炎症反应,促进应激犊牛瘤胃机能恢复,减少犊牛消化疾病的发病率,增加采食量,从而预和防治疗犊牛运输应激综合征,并使犊牛在运达后快速恢复正常生长。
为了实现上述目的,本发明采用如下技术方案:
一种防治犊牛转场运输应激组合物,由以下组分制备而成:中药提取物20~30份、改性松针粉18~26份、虾油8~16份、酵母细胞壁8~12份、茶多酚4~8份、番茄红素4~8份、乳清粉10~16份、蔗糖3~6份、微量元素3~6份。
优选的,所述中药提取物的制备方法,包括如下步骤:取中药原料,干燥、粉碎,得中药粉,用无水乙醇提取两次,过滤,合并两次提取液,蒸馏除去乙醇,得中药提取物。
优选的,以重量份数计,中药原料为6~10份乌梅、6~10份当归、4~8份金银花、4~8份半夏、4~8份柴胡、1~5份厚朴、1~3份生姜、2~4份甘草;干燥至中药原料含水量为8~10wt%,粉碎至80~100目。
优选的,一次提取中,中药粉与无水乙醇的重量比为1:2~3,在20~35℃下提取3~4h;二次提取中,中药粉与无水乙醇的重量比为1:3~5,在40~55℃下提取1~2h。
优选的,所述改性松针粉的制备方法,包括如下步骤:
(1)取马尾松松针,粉碎为松针粉,装入到蒸汽爆破罐中进行蒸汽爆破,得到预处理松针粉;
(2)将预处理松针粉与碳酸氢钠混合均匀,然后分散到二乙醇胺中,搅拌,然后加入棕榈酸,搅拌反应,反应结束后抽滤、洗涤、烘干,得到改性松针粉。
优选的,步骤(1)中,松针粉粒径<2mm;蒸汽爆破过程为向蒸汽爆破罐中通入温度为130~150℃的水蒸汽至压力为1.7~2.5Mpa,保压120~180s瞬间打开阀门,泄压至常压后取出。
优选的,步骤(2)中,预处理松针粉、碳酸氢钠、二乙醇胺、棕榈酸的重量比为10:0.8~1:20~30:4~6。
优选的,步骤(2)中,搅拌反应条件为70~80℃、300~500r/min下搅拌反应60~90min。
优选的,所述微量元素为氯化钠、碳酸锂、硫酸锌、氯化钾、氧化镁按重量比1:1:1~2:1~3:1混合而得。
优选的,所述组合物的灌服量为1~3g/kg。
本发明还要求保护一种防治犊牛转场运输应激组合物的制备方法,包括如下步骤:按重量份称取各原料,搅拌混合均匀,50~60℃下干燥至恒重,粉碎为1~5mm颗粒,得到所述防治犊牛转场运输应激组合物。
乌梅:味酸,性平,归肝、脾、肺、大肠经,具有敛肺止咳,涩肠止泻,止血,生津的功效,主治久咳不止,久泻久痢,尿血便血,崩漏,虚热烦渴,疮痈肿毒等证。
当归:甘、辛、温,归肝、心、脾经,具有补气养血、活血止痛、润燥通便的功能;主治血虚劳伤、血瘀疼痛;当归作为补益药具有促进新陈代谢、活血散瘀、旺盛血液循环的功能。
金银花:性甘,微苦,清香,辛,寒。归肺,胃,心,大肠经。重在清下焦及血分之热毒,主要用于治疗痢疾等。
半夏:辛散温燥,入脾、胃、肺经,善祛脾胃痰湿,内服能燥湿化痰、降逆止呕、消痞散结。
柴胡:性味苦平,入肝胆经;解热退热平稳可靠,镇静、镇痛、抗菌消炎,降压醒脑。
厚朴:性味苦、辛,温,苦燥泄降,辛散温通,入脾、胃、大肠经,既除胃肠之湿滞、食积,又理胃肠之气滞,有行气化湿、温中止痛、降逆平喘的功效。
生姜:性辛、微温,归肺脾胃经。具有发汗解表,温中止呕,温肺止咳,解药毒等功效。
甘草:甘、平,归肺、心、脾经,具有补脾益气、祛痰止咳、和中缓急、解毒、调和诸药、缓解药物毒性、烈性的功能;主治脾胃虚弱、咳喘、中毒。
与现有技术相比,本发明具有如下有益效果:
(1)本发明提供了一种防治犊牛转场运输应激组合物及其制备方法,所得组合物能够提升犊牛的肝肾功能,调节矿物质元素的代谢,帮助牛体纠正平衡失调,提高犊牛机体免疫机能,降低炎症反应,促进应激犊牛瘤胃机能恢复,减少犊牛消化疾病的发病率,增加采食量,从而预和防治疗犊牛运输应激综合征,并使犊牛在运达后快速恢复正常生长;中药提取物和改性松针粉能够防治犊牛由于运输应激导致的饮水、采食量下降,调节犊牛在运输过程中的内分泌和机体代谢,大幅提高抗应激能力,同时还具有镇静、抗狂躁的作用;虾油具有较多不饱和脂肪酸活性物质,能够调节机体代谢,增加细胞生物活性,快速补充机体能量水平,提高犊牛对低压缺氧的耐受力,起到抗疲劳作用;酵母细胞壁可延长食糜在消化道内的滞留时间、降低胃肠道消化道pH,促进肠道内有益菌增殖,改变肠道微生态环境,从而有效抑制犊牛体内有害菌生长,改善犊牛肠道微生态菌群平衡环境,预防腹泻,刺激免疫系统,强化特异性细胞免疫反应,提高犊牛机体免疫力;氯化钠、碳酸锂、硫酸锌、氯化钾、氧化镁作为电解质,能够调节犊牛在运输过程中由于应激导致的电解质紊乱,并防治代谢性中毒;乳清粉和蔗糖起到诱食作用,能够有效掩盖中药、松针的苦涩,有利于犊牛采食,使组合物发挥出应有的效果。
(2)本发明提供了一种中药提取物,组方合理,配伍得当,扶正祛邪,标本兼治,具有安神定惊、滋阴养血、补肝肾的作用,能够有效降低应激牛的体温、减缓呼吸频率,提高犊牛对运输应激的耐受力,与茶多酚、番茄红素配伍组合,能综合增强机体免疫功能,提高机体抗应激能力,还具有理气健脾、清热生津、调顺胃肠之气的作用,能够增加犊牛采食量,保证其消化和吸收功能,有利于短时间内恢复体质;该组方绿色环保、安全无毒副作用、无药物残留,不产生耐药性,不污染环境、不引发药源性疾病,且原料来源广泛、成本低廉。
(3)本发明提供了一种改性松针粉,首先将松针粉碎获得更大表面积,然后利用蒸汽爆破破坏松针细胞壁,打破松针粉内部纤维素结晶结构,使纤维素大分子中共价键断裂产生小分子低聚糖,同时还使得松针粉末变得疏松多孔,预处理后松针粉的有效成分能够更好被犊牛吸收利用,同时还能够改善犊牛肠道微生态环境;而后经二乙醇胺处理,能够在松针粉表面产生大量羟基活性位点并引入氮原子,棕榈酸则能将羧基官能团和长链烷烃引入松针粉表面,改性处理能够有效增加其与中药提取物中多糖、黄酮等有效成分的吸附性和亲和性,起到增强药效的作用;另外,松针粉自身具有较佳的抑菌活性,还能够提高机体抗氧化活性、抵抗疾病能力,其与中药组合物能够协同缓解犊牛运输应激反应,保障犊牛健康,增加犊牛转场运输的存活率,保证犊牛在转运后能够快速恢复至正常状态。

如无特殊说明外,本发明中的化学试剂和材料均通过市场途径购买或通过市场途径购买的原料合成。
马尾松采自长春博瑞科技股份有限公司园区树林;
酵母细胞壁由长春博瑞科技股份有限公司提供;
虾油购自汕头市明德食品添加剂有限公司。
下面通过具体实施例,来对本发明作进一步的说明。
实施例1
一种防治犊牛转场运输应激组合物的制备方法,包括如下步骤:
(1)取80g乌梅、80g当归、60g金银花、60g半夏、60g柴胡、30g厚朴、20g生姜、30g甘草,干燥至中药原料含水量为10wt%、粉碎至100目,得中药粉,将10g中药粉加入到25g无水乙醇中,在30℃下提取3.5h,过滤,将药渣加入到4倍药渣重量的无水乙醇中,在50℃下提取1.5h,过滤,合并两次提取液,蒸馏除去乙醇,得中药提取物;
(2)取马尾松松针,粉碎为粒径<2mm的松针粉,装入到蒸汽爆破罐中,向蒸汽爆破罐中通入温度为140℃的水蒸汽至压力为2.0Mpa,保压180s瞬间打开阀门,泄压至常压后取出,得到预处理松针粉;
(3)将100g预处理松针粉与9g碳酸氢钠混合均匀,然后分散到250g二乙醇胺中,搅拌,然后加入50g棕榈酸,75℃、400r/min下搅拌反应80min,反应结束后抽滤、洗涤、烘干,得到改性松针粉;
(4)取10g氯化钠、10g碳酸锂、10g硫酸锌、10g氯化钾、10g氧化镁混合均匀得到微量元素;
(5)按重量份称取300g中药提取物、260g改性松针粉、160g虾油、120g酵母细胞壁、80g茶多酚、80g番茄红素、160g乳清粉、60g蔗糖、60g微量元素,搅拌混合均匀,55℃下干燥至恒重,粉碎为1~5mm颗粒,得到防治犊牛转场运输应激组合物。
实施例2
一种防治犊牛转场运输应激组合物的制备方法,包括如下步骤:
(1)取80g乌梅、80g当归、60g金银花、60g半夏、60g柴胡、30g厚朴、20g生姜、30g甘草,干燥至中药原料含水量为10wt%、粉碎至100目,得中药粉,将10g中药粉加入到25g无水乙醇中,在30℃下提取3.5h,过滤,将药渣加入到4倍药渣重量的无水乙醇中,在50℃下提取1.5h,过滤,合并两次提取液,蒸馏除去乙醇,得中药提取物;

实施例3
一种防治犊牛转场运输应激组合物的制备方法,包括如下步骤:
(1)取80g乌梅、80g当归、60g金银花、60g半夏、60g柴胡、30g厚朴、20g生姜、30g甘草,干燥至中药原料含水量为10wt%、粉碎至100目,得中药粉,将10g中药粉加入到25g无水乙醇中,在30℃下提取3.5h,过滤,将药渣加入到4倍药渣重量的无水乙醇中,在50℃下提取1.5h,过滤,合并两次提取液,蒸馏除去乙醇,得中药提取物;

对比例1
一种防治犊牛转场运输应激组合物的制备方法,包括如下步骤:
(1)取马尾松松针,粉碎为粒径<2mm的松针粉,装入到蒸汽爆破罐中,向蒸汽爆破罐中通入温度为140℃的水蒸汽至压力为2.0Mpa,保压180s瞬间打开阀门,泄压至常压后取出,得到预处理松针粉;
(2)将100g预处理松针粉与9g碳酸氢钠混合均匀,然后分散到250g二乙醇胺中,搅拌,然后加入50g棕榈酸,75℃、400r/min下搅拌反应80min,反应结束后抽滤、洗涤、烘干,得到改性松针粉;
(3)取10g氯化钠、10g碳酸锂、10g硫酸锌、10g氯化钾、10g氧化镁混合均匀得到微量元素;
(4)按重量份称取200g改性松针粉、120g虾油、100g酵母细胞壁、60g茶多酚、60g番茄红素、130g乳清粉、50g蔗糖、40g微量元素,搅拌混合均匀,55℃下干燥至恒重,粉碎为1~5mm颗粒,得到防治犊牛转场运输应激组合物。
对比例2
一种防治犊牛转场运输应激组合物的制备方法,包括如下步骤:
(1)取80g乌梅、80g当归、60g金银花、60g半夏、60g柴胡、30g厚朴、20g生姜、30g甘草,干燥至中药原料含水量为10wt%、粉碎至100目,得中药粉,将10g中药粉加入到25g无水乙醇中,在30℃下提取3.5h,过滤,将药渣加入到4倍药渣重量的无水乙醇中,在50℃下提取1.5h,过滤,合并两次提取液,蒸馏除去乙醇,得中药提取物;
(2)取10g氯化钠、10g碳酸锂、10g硫酸锌、10g氯化钾、10g氧化镁混合均匀得到微量元素;
(3)按重量份称取250g中药提取物、120g虾油、100g酵母细胞壁、60g茶多酚、60g番茄红素、130g乳清粉、50g蔗糖、40g微量元素,搅拌混合均匀,55℃下干燥至恒重,粉碎为1~5mm颗粒,得到防治犊牛转场运输应激组合物。
对比例3
一种防治犊牛转场运输应激组合物的制备方法,包括如下步骤:
(1)取80g乌梅、80g当归、60g金银花、60g半夏、60g柴胡、30g厚朴、20g生姜、30g甘草,干燥至中药原料含水量为10wt%、粉碎至100目,得中药粉,将10g中药粉加入到25g无水乙醇中,在30℃下提取3.5h,过滤,将药渣加入到4倍药渣重量的无水乙醇中,在50℃下提取1.5h,过滤,合并两次提取液,蒸馏除去乙醇,得中药提取物;
(2)取马尾松松针,粉碎为粒径<2mm的松针粉,装入到蒸汽爆破罐中,向蒸汽爆破罐中通入温度为140℃的水蒸汽至压力为2.0Mpa,保压180s瞬间打开阀门,泄压至常压后取出,得到预处理松针粉;
(3)取10g氯化钠、10g碳酸锂、10g硫酸锌、10g氯化钾、10g氧化镁混合均匀得到微量元素;
(4)按重量份称取250g中药提取物、200g预处理松针粉、120g虾油、100g酵母细胞壁、60g茶多酚、60g番茄红素、130g乳清粉、50g蔗糖、40g微量元素,搅拌混合均匀,55℃下干燥至恒重,粉碎为1~5mm颗粒,得到防治犊牛转场运输应激组合物。
2022年10月5日在甘肃张掖市肉牛交易市场上随机挑选91头经临床检查健康、体重相近(约300kg)的8月龄西门塔尔杂交肉牛,装入货车运送至山西省大同市伊磊牧场,里程大约1600km,总运输时间20h左右。将犊牛随机分为7组,在运输前2天和运达后5天的早上8:00,前6组分别对犊牛灌服实施例1~3、对比例1~3制备所得组合物,第7组(空白组)灌服蒸馏水,剂量600g/头,试验期间,7组牛的基础日粮完全相同,自由饮水,居住相对湿度、光照均相同。
分别于运前2d、运达时、运达后7d、30d的上午9:00对犊牛进行称重,具体数据见图1。可知,7组犊牛在运达时体重均发生了显著降低,而在运达后30d体重又有了恢复,其中实施例1~3组犊牛的变化最为平缓,其次为对比例3组,而对比例1组、空白组的体重下降最为剧烈,且在运达后30d体重也未完全恢复至运输前水平。
从犊牛运达至运达后10d,连续观察记录犊牛腹泻与死亡数量,记录每头犊牛平均腹泻次数、发病率与病死率(试验过程中及时治疗病牛),具体数据见表1。

运达后7d,将直插式精密pH计插进试验牛直肠进行pH值检测。同时,用无菌玻璃棒采集试验牛直肠中适量的肠道内容物,称取0.5g后置于10mL无菌EP管中,用无菌生理盐水倍比稀释(稀释度为1×10-6),最后吸取上层稀释液0.1mL分别均匀涂布于分离培养基中,培养后计数。每克肠道微生物菌落数=lg[菌落平均数×稀释倍数×10mL/(0.1×0.5)],具体数据见表2。

分别在运前3d、运后10h、运达时、运达后5d的上午9:00对各组犊牛进行采血,每次采血5mL,进行激素和免疫指标测定,结果见图2、图3、表3。
皮质醇(Cor):动物在应激原的刺激下血液中皮质醇含量会发生明显变化,皮质醇被认为是评价动物应激程度的客观指标。
促肾上腺皮质激素(ACTH):应激状态下,下丘脑垂体-肾上腺皮质轴(HPA)的功能增强,皮质醇(Cor)和促肾上腺皮质激素(ACTH)水平也不同程度的增加,进而对淋巴细胞、中性粒细胞、巨噬细胞等几乎所有免疫细胞呈现抑制作用,对免疫细胞增殖、细胞因子和抗体的生成进行调控,从而影响动物的免疫功能。
组合物对犊牛的皮质醇(Cor)、促肾上腺皮质激素(ACTH)的影响见图2、图3。如图所示,对比例1、对比例2、空白组犊牛的Cor、ACTH在运后10h和运达时均极显著升高(P<0.01),运达后慢慢恢复正常,实施例组犊牛的Cor、ACTH在运后有升高趋势但是不显著,运达后也慢慢恢复正常。
组合物对犊牛免疫球蛋白的影响见表3,由表3可知,实施例1~3组的免疫球蛋白含量在运达时均有降低趋势但差异不显著,且在运达后慢慢恢复正常;对比例1、空白组的犊牛免疫球蛋白含量在运输后发生了显著降低,且在运达后5d也未完全恢复至正常水平。

Claims (4)
1.一种防治犊牛转场运输应激组合物,其特征在于,由以下组分制成:中药提取物20~30份、改性松针粉18~26份、虾油8~16份、酵母细胞壁8~12份、茶多酚4~8份、番茄红素4~8份、乳清粉10~16份、蔗糖3~6份、微量元素3~6份;
所述改性松针粉的制备方法,包括如下步骤:
(1)取马尾松松针,粉碎为松针粉,装入到蒸汽爆破罐中进行蒸汽爆破,得到预处理松针粉;
(2)将预处理松针粉与碳酸氢钠混合均匀,然后分散到二乙醇胺中,搅拌,然后加入棕榈酸,搅拌反应,反应结束后抽滤、洗涤、烘干,得到改性松针粉;
步骤(1)中,松针粉粒径<2mm;蒸汽爆破过程为向蒸汽爆破罐中通入温度为130~150℃的水蒸汽至压力为1.7~2.5Mpa,保压120~180s瞬间打开阀门,泄压至常压后取出;
步骤(2)中,预处理松针粉、碳酸氢钠、二乙醇胺、棕榈酸的重量比为10:0.8~1:20~30:4~6;
步骤(2)中,搅拌反应条件为70~80℃、300~500r/min下搅拌反应60~90min;
所述中药提取物的制备方法,包括如下步骤:取中药原料,干燥、粉碎,得中药粉,用无水乙醇提取两次,过滤,合并两次提取液,蒸馏除去乙醇,得中药提取物;
以重量份数计,中药原料为6~10份乌梅、6~10份当归、4~8份金银花、4~8份半夏、4~8份柴胡、1~5份厚朴、1~3份生姜、2~4份甘草;干燥至中药原料含水量为8~10wt%,粉碎至80~100目。
2.根据权利要求1所述的组合物,其特征在于,一次提取中,中药粉与无水乙醇的重量比为1:2~3,在20~35℃下提取3~4h;二次提取中,中药粉与无水乙醇的重量比为1:3~5,在40~55℃下提取1~2h。
3.根据权利要求1所述的组合物,其特征在于,所述微量元素为氯化钠、碳酸锂、硫酸锌、氯化钾、氧化镁按重量比1:1:1~2:1~3:1混合而得。
4.一种如权利要求1~3任一项所述防治犊牛转场运输应激组合物的制备方法,其特征在于,包括如下步骤:按重量份称取各原料,搅拌混合均匀,50~60℃下干燥至恒重,粉碎为1~5mm颗粒,得到所述防治犊牛转场运输应激组合物。
