Abstract
本发明属于生物医药技术领域,公开了六氯酚在制备治疗乳腺癌的药物中的新应用。本发明经研究证实化合物六氯酚对于三阴性乳腺癌小鼠有显著的抗肿瘤效果,且对小鼠无明显毒性,本发明为乳腺癌的治疗提供了一种新的可选药物,具有良好的应用前景。

技术领域
本发明涉及生物医药技术领域,涉及一种化合物的新应用,具体涉及六氯酚在制备治疗乳腺癌的药物中的新应用。
背景技术
三阴性乳腺癌(triple negative breast cancer,TNBC)是指肿瘤组织免疫组织化学检查结果为雌激素受体(ER)、孕激素受体(PR)和原癌基因Her-2均为阴性的乳腺癌。这类乳腺癌占所有乳腺癌病理类型的10.0%~20.8%,具有特殊的生物学行为和临床病理特征,预后较其他类型差。与其他类型乳腺癌相比,三阴性乳腺癌具有高度侵袭性,发病年龄更早、转移潜能更大、临床治疗结果更差、复发率更高、存活率更低,并迅速扩散到远端器官,如肺、肝、脑和中枢神经系统。三阴性乳腺癌的常规治疗方法是手术和化疗,但是疗效不佳、预后差,因此迫切需要找到新的治疗策略来提高三阴性乳腺癌患者的生存率。
六氯酚(Hexachlorophene)是一种有机氯化合物,直到20世纪70年代才被广泛用作有效的局部防腐剂。六氯酚目前主要应用为消毒杀菌防腐,例如皮肤消毒药;对革兰氏阳性菌效果优异,而对革兰氏阴性菌效力较差。
已有研究表明:β-连环蛋白信号传导的激活可以促进肿瘤发生,B淋巴瘤细胞被Epstein-Barr病毒(EBV)感染并表达其潜伏膜蛋白(LMP)-1,会导致β-连环蛋白表达增加和细胞转化,抗生素六氯酚可以下调β-连环蛋白水平,从而为感染EBV的B淋巴瘤细胞提供一种新的治疗策略。Kirste大鼠肉瘤病毒癌基因同源物(KRAS)突变是非小细胞肺癌(NSCLC)中最常见的靶向和突变分子亚型。六氯酚可以通过抑制RAF/MEK/ERK和PI3K/AKT通路,抑制KRAS-mut NSCLC细胞增殖和转移。
目前还未见将六氯酚应用于乳腺癌治疗中的报道。
发明内容
本发明的目的是针对上述问题,提供一种六氯酚的新应用,为乳腺癌的治疗提供一种新药物。
本发明为了实现其目的,采用的技术方案是:
六氯酚在制备治疗乳腺癌的药物中的新应用。
在上述的应用的技术方案中,所述乳腺癌为三阴性乳腺癌。
在上述的应用的技术方案中,所述药物的活性成分为六氯酚。
在上述的应用的技术方案中,所述药物还包括药学上可接受的载体。

本发明的有益效果是:本发明研究证实化合物六氯酚对人源三阴性乳腺癌细胞生长具有显著抑制作用,对于三阴性乳腺癌小鼠有显著的抗肿瘤效果,且对小鼠无明显毒性,本发明为乳腺癌的治疗提供了一种新的可选药物,具有良好的应用前景。

具体实施方式
下面结合实施例对本发明作进一步说明,但并不因此而限制本发明。
下述实施例中的实验方法,如无特别说明,均为常规方法。
主要试剂、材料来源:
CCK8试剂:购自GLPBIO公司(美国);
六氯酚:CAS号:70-30-4,购自GLPBIO公司(美国);
TDP43抗体:Proteintech公司,美国;
GRP78抗体:Affinity公司,美国。
实施例1
1.细胞培养
向人源性三阴性乳腺癌细胞MDA-MB-231细胞和小鼠三阴性乳腺癌细胞4T1细胞中加入含有10%血清和1%青霉素-链霉素双抗(10000U/mL)的RPMI-1640培养基,5%CO237℃培养箱内培养,每两天传代一次,当细胞的融合度达到80%-90%左右,即可进行试验。
2.CCK8实验
以每孔1×104个细胞的密度,在37℃、5%CO2存在下,将细胞接种在96孔培养板上24小时,以使细胞粘附在板上。之后,分别将细胞用新鲜培养基和含六氯酚的新鲜培养基取代上述培养基。其中,六氯酚使用DMSO溶解,对照组加入等量DMSO。培养24小时和48小时后,弃去原培养基,每孔加入100μl CCK8试剂和不完全培养基预混液,37℃孵育30分钟上机检测450nm处的光密度值,评估细胞增殖能力。
分别在24小时和48小时使用CCK8检测三阴性乳腺癌细胞增殖能力。结果如图1所示,给药组与对照组相比,细胞增殖能力明显受到抑制。
3.构建BALb/c小鼠乳腺癌肿瘤4T1细胞株移植瘤模型
将处于对数期的4T1细胞,经过胰酶消化、离心后,用PBS重悬,将细胞悬液的细胞密度调整为107个/mL,置于冰盒中待用。先将雌性小鼠的腹部进行脱毛处理后,用1mL的注射器吸取100μL的细胞悬液在小鼠右下腹第四对乳房脂肪垫处皮下注射。完成接种后的小鼠继续饲养14天后,从中选择出移植瘤体积为100mm3以上的小鼠用于实验。
4.抗肿瘤效应
从上述的小鼠中,挑选出肿瘤体积大于100mm3的乳腺癌肿瘤4T1细胞株移植瘤模型的BALB/C小鼠15只,随机分成3组,每组5只:①对照组(单纯注射药物溶剂);②六氯酚2.5mg/kg组;③六氯酚5mg/kg组。六氯酚药物使用丙二醇溶解,采取腹腔注射给药的方式。
每2d给药一次,总共给药13次,每2d测一次肿瘤体积和小鼠体重,将测得的肿瘤最长径(A)和最短径(B)代入肿瘤体积计算公式中进行计算得出各组肿瘤体积(V)。随后以肿瘤体积为纵坐标,时间为横坐标,绘制肿瘤生长曲线图。将小鼠处死,取出肿瘤,并称重记录,以肿瘤重量为纵坐标,分组为横坐标,对各组肿瘤的最终重量进行比较。肿瘤的体积计算公式如下:
小鼠处死后取出瘤体计算体积,结果如图2所示,对照组小鼠瘤体体积均值为1453.9mm3,2.5mg/kg组小鼠瘤体体积均值为932.6mm3,5mg/kg组小鼠瘤体体积均值为765.7mm3。给药组小鼠瘤体体积明显小于对照组,且5mg/kg组小鼠瘤体体积明显小于2.5mg/kg。
取出瘤体后对小鼠瘤体重量进行测量,结果如图3所示,对照组小鼠瘤体重量均值为1.424g,2.5mg/kg组小鼠瘤体重量均值为0.8898g,5mg/kg组小鼠瘤体重量均值为0.5998g。给药组小鼠瘤体重量明显低于对照组,且5mg/kg组小鼠瘤体重量明显低于2.5mg/kg。
从第一次给药起,每2天测量小鼠瘤体大小,绘制小鼠瘤体体积曲线,结果如图4所示。给药组瘤体生长速度明显较对照组缓慢,且5mg/kg组小鼠瘤体生长速度小于2.5mg/kg。
从第一次给药起,每2天记录一次小鼠的体重,给药前各组小鼠的体重相当,经过13次给药后对照组和给药组体重变化趋势较为一致(图5),均未观察到体重变化差异,说明药物不会影响小鼠的体重增长,未观察到明显毒性。
5.六氯酚抗肿瘤的作用机制研究
TDP-43是一种DNA和RNA结合的调节因子,其表达受核定位信号和转移信号等调控并受机体生理状态影响。TDP-43在正常生理状态下行使着多项重要功能,在多种疾病中参与RNA代谢和转录调节。GRP78是一种关键的细胞伴侣蛋白,有助于调节其他细胞蛋白的折叠。GRP78可以作为内质网应激的标志物。
我们在小鼠三阴性乳腺癌细胞中验证了六氯酚对TDP43的作用。以每孔1×106个细胞的密度,在37℃、5%CO2存在下,将细胞接种在6孔培养板上24小时,使细胞密度达80%。之后,分别将细胞用新鲜培养基和含六氯酚(3uM)的新鲜培养基取代上述培养基,其中,六氯酚使用DMSO溶解,对照组加入等量DMSO。培养24小时后,提取蛋白。使用蛋白免疫印迹检测蛋白中TDP43的表达。结果如图6所示,在使用六氯酚处理24小时后,TDP43的表达显著性降低。六氯酚处理组TDP43蛋白表达同对照组相比,平均降低52%。
提取小鼠瘤体中的蛋白,使用蛋白免疫印迹检测瘤体蛋白中TDP43的表达。结果如图7所示,在六氯酚处理组TDP43的表达同对照组相比显著性降低。2.5mg/kg组TDP43的表达量同对照组相比降低23.2%,5mg/kg组TDP43的表达量同对照组相比降低47.2%。以上结果证明,六氯酚对三阴性乳腺癌肿瘤的抑制作用可能是通过抑制TDP43的表达。
随后使用蛋白免疫印迹检测瘤体蛋白中GRP78的表达。结果如图8所示,在六氯酚处理组GRP78的表达同对照组相比显著性降低。2.5mg/kg组GRP78的表达量同对照组相比降低24.9%,5mg/kg组GRP78的表达量同对照组相比降低52.7%。GRP78可以作为内质网应激的标志物。
以上结果证明,六氯酚对三阴性乳腺癌肿瘤的抑制作用可能是通过抑制TDP43表达从而抑制内质网应激。

1.六氯酚在制备治疗乳腺癌的药物中的新应用。
2.根据权利要求1所述的应用,其特征在于:所述乳腺癌为三阴性乳腺癌。
3.根据权利要求1所述的应用,其特征在于:所述药物的活性成分为六氯酚。
4.根据权利要求3所述的应用,其特征在于:所述药物还包括药学上可接受的载体。
5.根据权利要求1所述的应用,其特征在于:六氯酚通过抑制TDP43蛋白的表达和内质网应激,从而抑制乳腺癌的发展。
6.一种用于治疗乳腺癌的药物组合物,其特征在于:含有作为活性成分的六氯酚以及药学上可接受的载体。
7.根据权利要求6所述的药物组合物,其特征在于:所述乳腺癌为三阴性乳腺癌。
8.根据权利要求6所述的药物组合物,其特征在于:所述药物组合物含有治疗有效量的六氯酚。
9.根据权利要求6所述的药物组合物,其特征在于:六氯酚通过抑制TDP43蛋白的表达和内质网应激抑制乳腺癌的发展。
