An acid-resistant and bile-salt-resistant probiotic preparation is prepared from crystal ball probiotics and capsule shells, wherein the crystal ball probiotics are filled in the capsule shells, the capsule shells are transparent capsule shells, and the crystal ball probiotics are probiotic dripping pills.
The capsule shell is selected from enteric gelatin capsule or enteric plant capsule.
The probiotics dripping pill has the following color: red, orange, yellow, blue.
The probiotics dripping pill formula is prepared from the following raw and auxiliary materials in percentage by weight: probiotics: 10-15%, carrier oil: 75-89% of emulsifying agent: 1-10%;
the emulsifier is MCT oil.

The probiotic dripping pill is a plain pill or a coated pill.

adding 2.5-3.5% of MCT oil by weight of beeswax into the mixture, melting at 85-90deg.C, adding into the rest MCT oil, adding probiotic powder, stirring, and dripping at 30-40deg.C.

adding 2.5-3.5% of MCT oil by weight of the beeswax into the formula, melting the materials at 85-90 ℃, adding the rest MCT oil after the melting materials are uniform, adding the probiotic powder by the formula, stirring uniformly, transferring to a pill dropping machine, dropping at 30-40 ℃ to obtain pellet probiotic pills, dissolving enteric coating materials with water according to the proportion of 6% of coating weight gain, fully stirring uniformly, transferring the pellet into coating equipment, and coating under the conditions of average air inlet temperature of 60 ℃, average tablet bed temperature of 35-40 ℃, atomizing pressure of 2bar, average rotating speed of 10-20rpm and average coating liquid flow rate of 2-5g/min to obtain the pellet.
The grain size of the crystal ball probiotics is 0.5-3mm.

Dissolving enteric coating material with water according to the weight increasing proportion of 6%, stirring thoroughly, transferring the pill into coating equipment, and coating at average air inlet temperature of 60deg.C, average tablet bed temperature of 35deg.C, atomizing pressure of 2bar, average rotation speed of 10rpm, and average coating liquid flow rate of 2-5 g/min.

Example 3 coated pellet of pellet probiotics
The pellet composition and the preparation were the same as in example 2, except that the red coating was applied.
Example 4 coated pellet of pellet probiotics
The pellet composition and the preparation were the same as in example 2, except that orange coating was applied.
Example 5 coated pellet of pellet probiotics
The pellet composition and the preparation were the same as in example 2, except that a yellow coating was applied.
Example 6 coated pellet of pellet probiotics
The pellet composition and the preparation were the same as in example 2, except that a blue coating was applied.

and (3) detecting the crystal ball probiotics: taking 5g of crystal ball probiotics, dissolving the probiotics in 60mmol/L sodium citrate solution, and detecting the number of viable bacteria by referring to GB4789.35-2016 food safety national standard food microbiology test lactobacillus test;

Simulating gastric juice digestion
The pH value of human gastric juice is usually about 3.0, the gastric juice is about 2.0 in the fasting state, and the digestion time is usually 2-3 hours. 1g of crystal ball probiotic capsules are taken and placed in simulated gastric fluid (Chinese pharmacopoeia 2015 edition) for 60 minutes:

As can be seen from table 2, the pellet probiotic capsules prepared by the present invention can resist digestion of gastric juice for 60min without disintegration, so as to smoothly enter the intestinal tract, thereby improving the intestinal health.
Simulation of bile and intestinal fluid digestion
The food stays in the small intestine longer than in the stomach for about 3-8 hours. After passing through the stomach, food enters the duodenum, bile and pancreatic juice are injected into the duodenum, the mass fraction of the bile salt in the small intestine is about 0.3%, and the survival condition of probiotics in simulated bile salt and artificial intestinal juice in the pellet probiotic capsules is tested:

As can be seen from table 3, the pellet probiotic capsules prepared by the method can be dissolved in intestinal tracts only by about 3 hours, and the probiotics in the pellet probiotic capsules still have higher survival rate after being digested by the bile salt solution for 3.0 hours, so that the intestinal tract environment can be effectively improved.
The survival rate bar graph of probiotics in the bile salt solution digested 3.0h crystal ball probiotics is shown in figure 1.

the probiotics dripping pill is prepared from the following raw and auxiliary materials in percentage by weight: probiotics: 10-15%, carrier oil: 75-89% of emulsifying agent: 1-10%;
the emulsifier is medium chain fatty acid triglyceride;
the probiotics are formed by mixing one or more of lactobacillus acidophilus, lactobacillus rhamnosus, lactobacillus casei, lactobacillus paracasei, lactobacillus plantarum, lactobacillus grignard, lactobacillus reuteri, lactobacillus bulgaricus, lactobacillus johnsonii, bifidobacterium bifidum, bifidobacterium longum, bifidobacterium breve, bifidobacterium adolescentis, bifidobacterium infantis, bacillus coagulans, propionibacterium freudenreichii, enterococcus faecalis, lactococcus lactis, streptococcus thermophilus and kluyveromyces marxianus;
the carrier oil is beeswax;
the probiotics dripping pill is a plain pill or a coated pill;
the specific preparation method of the element pill of the probiotics dripping pill comprises the following steps:
adding 2.5-3.5% of medium chain fatty acid triglyceride by weight of Cera flava into the mixture, dissolving at 85-90deg.C, adding into the rest medium chain fatty acid triglyceride, adding probiotic powder, stirring, and dripping at 30-40deg.C;
the specific preparation method of the coated pill of the probiotics dripping pill comprises the following steps:
adding 2.5-3.5% of medium chain fatty acid triglyceride by weight of beeswax into the formulation, melting at 85-90 ℃, adding the rest medium chain fatty acid triglyceride after melting uniformly, adding the formulation of probiotic powder, stirring uniformly, transferring to a dripping pill machine, dripping at 30-40 ℃ to obtain a probiotic dripping pill, dissolving enteric coating material with water according to the proportion of coating weight gain of 6%, fully stirring uniformly, transferring the dripping pill into coating equipment, and coating under the conditions of average air inlet temperature of 60 ℃, average tablet bed temperature of 35-40 ℃, atomization pressure of 2bar, average rotating speed of 10-20rpm and average coating liquid flow rate of 2-5g/min to obtain the probiotic dripping pill;
the particle size of the probiotics dripping pill is 0.5-3mm.
2. An acid and bile salt resistant probiotic formulation according to claim 1, characterized in that the capsule shell is selected from an enteric gelatin capsule or an enteric plant capsule.
