技术领域
本发明涉及中药技术领域,具体涉及一种提高睾丸精子数量、质量和改善碎片成熟的生物组合物制剂及其制备方法。
背景技术
目前,在世界范围内男性生育质量普遍降低,据统计约有15%~20%的育龄夫妇存在生育问题,并呈逐年上升趋势,其中男性造成的不育占30%~40%。男性不育症病因比较复杂,受环境污染、心理压力及烟酒嗜好等多种因素影响,其发病机制尚未完全明了,一般认为可能与内分泌因素、生精相关基因缺失或突变、微生物感染及微量元素缺乏等因素有关。精液质量与男性不育症有着十分密切的关系,也是男性不育症诊断及疗效观察的重要客观依据,精液质量主要由精子数量、质量和改善碎片成熟决定。其治疗药物多以性激素、抗炎、锌制剂及抗氧化剂等为主,但临床应用提高精子质量效果不明显,受孕率低,疗效欠佳。辅助生殖技术(Assisted Reproductive Technology,ART)虽然为男性不育症的患者提供了生育的可能,但ART技术的成功率仍较低,妊娠率仅为4%~40%,且子代出生缺陷的风险高于自然妊娠。因此研发安全有效的治疗少弱精子症的药物具有重大的社会价值和研究意义。
发明内容
为了克服现有技术的不足,本发明的目的之一在于提供一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂,该生物组合物制剂能够实现精子数量明显增加,精子质量明显提高。
本发明的目的之二在于提供一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂的制造方法,该方法步骤简单,能够工业化生产提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的中药。

君药
补骨脂:味苦、辛,性温,归肾、脾经,具有补肾助阳,固精缩尿,暖脾止泻,用于治疗肾虚阳痿、腰膝冷痛的功效。
锁阳:味甘,性温,归肝、肾、具有补肾阳,益精血,阳痿滑精的功效。
韭菜子:味辛、甘,性温,归肝、肾经,具有补肝肾,暖腰膝,助阳,固精的功效。
沙苑子:味甘,性温,归肝、肾经,具有补肾固精,阳痿遗精的功效。
冬虫夏草:味甘,性温,归肾、肺经,补肾益精,具有兴阳起痿之功,用于治肾阳不足,精血亏虚之阳痿遗精,腰膝酸痛的功效。
分心木:苦、涩,平,归脾、肾经,具有涩精缩尿,止血止带,止泻痢的功效,用于遗精滑泄,尿频遗尿,崩漏,带下,泄泻,痢疾的治疗。3~9
金樱子:味酸而涩,善于收敛固涩,又入肾、膀胱、大肠经,能够敛肾气、固精关、止遗滑(遗精滑精)、缩小便、止泻,对于因肾气不足、下元不固而导致的神疲乏力、腰膝酸软、遗精滑精、尿频遗尿、久泻,患者可以通过适量饮用金樱子泡酒,来帮助改善症状。
以上七味药皆为君药,利用其固摄精气和精液,防其滑精,从而起固精的作用;同时又利用其补肾的功能,温补肾气以气化水液,使排尿正常,避免尿频、遗尿,从而起缩尿的作用,往往用于因为肾阳虚不固而引起的遗精、滑精、遗尿、尿频的症状。
佐助药,即配合君、臣药以加强治疗作用,或直接治疗次要症状的药物臣药
紫河车:味甘、咸,性温,归肺、心、肾经,具有补精助阳,养血益气的功效。
鹿角霜:味咸、涩,性温。归肝、肾经。具有温肾助阳,收敛止血的功效,用于脾肾阳虚,白带过多,遗尿尿频,崩漏下血,疮疡不敛的治疗。
蛤蚧:味咸,性平,归肺、肾经,具有补肺益肾,纳气定喘,助阳益精的功效。
以上三味药皆为臣药,补益人体肾脏精气,用于改善肾阳虚,以助君药起治疗不固而引起的遗精、滑精、遗尿、尿频的症状。
佐药
阳起石:味咸,性温,归肾经,具有温肾壮阳,强阳起痿,治男子阳痿不举,女子宫冷不孕,崩中漏下,以及腰膝冷痛等症的功效。
仙茅:味辛,性热,归肾、肝经,具有温肾壮阳,祛寒除湿的功效,主治阳痿精寒,遗尿尿频,寒湿痹痛,筋骨痿软,腹痛冷泻等的功效。
狗肾:味甘、咸,性温,归肾经,具有壮阳补精的功效的功效。
肉桂:药性辛、大热,归肾、脾、心、肝经,具有补火助阳、引火归元、散寒止痛、温通经脉的作用,用于治疗阳痿宫冷、腰膝冷痛、肾虚作喘、虚阳上浮、眩晕目赤、心腹冷痛、虚寒吐泻、寒疝腹痛、痛经经闭。
姜黄:味辛、苦,性温,归肝、脾经,具有活血行气,通经止痛的功效,主治气滞血瘀痛证,风湿痹痛。
火麻仁:味甘,性平,归肝、脾经,具有活血行气,通经止痛的功效,主治气滞血瘀痛证,风湿痹痛。
山药:味甘,性平,归脾、肺、肾经,具有益气养阴,补脾肺肾,固精止带的功效。实践还发现补阳药主要有增强精子活力的作用,而滋阴药则可明显增加精子数量。
以上七味药皆为佐药,利用阳起石、仙茅、狗肾、肉桂壮阳的功效,达到提高肾阳、强阳健体,起增强精子活力的作用,且能提升性能力、增强性欲的目的,并且还能起直接治疗腰膝冷痛、精索静脉曲张等次要症状的作用;辅以利用姜黄、火麻仁,利用其活血行气,通经止痛的功效,用于改善气滞血瘀,起治疗精索静脉曲张的作用;山药具有益气养阴的功效,达到使人体气血滋生,改善气阴两虚的症状,可明显增加精子数量。
本发明的目的之二采用如下技术方案实现:
一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂的制备方法,包括以下步骤:
S1、按照重量份数计算,将补骨脂6~10份、冬虫夏草3~6份、锁阳5~10份、蛤蚧3~6份、山药15~30份、沙苑子9~15份、姜黄3~9份、火麻仁10~15份、肉桂1~5份、鹿角霜9~15份、仙茅3~10份、阳起石9~30份、紫河车1.5~3份、分心木3~9份、金樱子6~12份、韭菜子5~15份、狗肾2.5~5份十七味药加适量水煎煮2次,每次1~2小时,合并煎液,滤过,得到所述提高卵巢卵子数量、质量和储备功能的生物组合物制剂的煎液;
S2、将所述提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂的煎液中加入所述药学上可接受的辅料,制备成所述提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂。
本发明的目的之三是提供一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂、或者所述的一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂的制备方法制备的生物组合物制剂在制备用于提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的药物中的应用。
相比现有技术,本发明的有益效果在于:

实验例
1实验动物分组及给药
1.1少弱精子症小鼠模型建立
体重18~22g之间的雄性成年ICR小鼠清洁级饲养间适应性喂养1周后,按体重梯度抽样法将其平均分至对照组和模型组,模型组20只,对照组10只,每天早上称取小鼠体重,模型组腹腔注射环磷酰胺(Cyclophosphamide,CTX)60mg/kg,连续给5天,即造成少弱精子症模型。
1.2实验分组
造模成功小鼠按体重均匀分为4组,记为组1~4,每组5只,组1不作处理,组2~4分别给予实施例1~3提供提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂进行灌胃,每组5只。组2~4分别于每天上午固定时间点连续给药4周,对照组和模型组小鼠均给予相同容量的生理盐水,以每0.1ml/10g的给药容积灌胃给药;其余实验条件相同。
2精子参数测定
2.1实验方法:
精子数目的计数
将盛有2mL生理盐水的小平皿置于恒温水浴箱内预热至37℃,连续给药4周后将照组和模型组小鼠右侧附睾尾称重并在小平皿剪碎,水浴5分钟,得到的精子原浆用生理盐水稀释5倍,在400倍光镜下计数血细胞计数板上4大格精子的数量并计算。精子数目的计数结果如表1所示。

精子活力的测定
半定量法评估精子活力,对照组和模型组小鼠每个样本取10μl稀释5倍后的精子混悬液,400倍光镜下观察大约100个精子的游动情况,计算游动精子的比例。
精子分级标准参照《世界卫生组织人类精液及精子-宫颈粘液相互作用实验室检验手册》。精子标准可以被分为4级:1级:精子活动性良好,运动时呈快速直线运动;2级:精子运动性一般,运动时做直线或非直线运动;3级:精子运动能力差,呈徘徊状或转圈运动,几乎无前进性;4级:精子没有运动能力。精子活力计算依照以下公式:精子活力(%)=(1级+2级+3级)/(1级+2级+3级+4级)*100%。精子活力的测定的结果如表2所示。

精子畸形率的测定
精子原浆采用0.05%龙胆紫染色,记录光镜十个视野里精子的形态变化,观察500个精子,其中颈部膨大、纤细、屈折、不全、双体等、尾部中段畸形(膨大、纤细、弯曲、屈折、不全、双尾)、尾部主段畸形,然后分类计数,有的精子尾部发育未完成,为未成熟精子,可计为畸形精子,精子畸形率的测定的结果如表3所示。

精子存活率测定
精子原浆采用0.05%龙胆紫染色,记录光镜十个视野里精子的形态变化,观察500个精子,,记录精子的着色状况。精子头部着紫色时为死亡精子,精子头部不着色或着浅紫色时为活精子,计数后计算活精子百分率,精子存活率的测定的结果如表4示。

2.2实验结果:
如表1所示,模型组组2~4的小鼠精子数量与对照组相比显著下降,模型组组2~4在给予实施例1~3提供提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂治疗后,小鼠精子精子数量与模型组相比显著提高,精子畸形率显著下降,恢复到对照组的水平,示本发明提供的提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂对于少弱精子症小鼠的精子的数量有很好的保护作用。
如表2所示,模型组组2~4在给予实施例1~3提供提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂治疗后,小鼠精子精子活力与模型组相比显著提高,精子畸形率显著下降,恢复到对照组的水平;如表3所示,模型组组2~4在给予实施例1~3提供提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂治疗后,小鼠精子精子畸形率与模型组相比显著提高,精子畸形率显著下降,恢复到对照组的水平;如表4示,模型组组2~4在给予实施例1~3提供提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂治疗后,小鼠精子精子存活率与模型组相比显著提高,精子畸形率显著下降,恢复到对照组的水平,提示本发明提供的提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂对于少弱精子症小鼠的精子质量有很好的保护作用。
上述实施方式仅为本发明的优选实施方式,不能以此来限定本发明保护的范围,本领域的技术人员在本发明的基础上所做的任何非实质性的变化及替换均属于本发明所要求保护的范围。
Claims (6)
1.一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂,其特征在于,它是由如下重量配比的原料药制备而成的制剂:补骨脂6~10份、冬虫夏草3~6份、锁阳5~10份、蛤蚧3~6份、山药15~30份、沙苑子9~15份、姜黄3~9份、火麻仁10~15份、肉桂1~5份、鹿角霜9~15份、仙茅3~10份、阳起石9~30份、紫河车1.5~3份、分心木3~9份、金樱子6~12份、韭菜子5~15份、狗肾2.5~5份。
2.如权利要求1所述的一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂,其特征在于,它是由如下重量配比的原料药制备而成的制剂:补骨脂8份、冬虫夏草5份、锁阳8份、蛤蚧5份、山药20份、沙苑子12份、姜黄5份、火麻仁12份、肉桂3份、鹿角霜10份、仙茅8份、阳起石15份、紫河车2份、分心木5份、金樱子8份、韭菜子10份、狗肾3份。
3.如权利要求1所述的一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂,其特征在于,所述生物组合物制剂为片剂、胶囊剂、口服液、水针剂或粉针剂中的一种。
4.如权利要求1所述的一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂,其特征在于,它还包括药学上可接受的辅料,所述辅料包括但不限于填充剂、崩解剂、润滑剂、助悬剂、赋形剂、粘合剂、甜味剂、矫味剂、防腐剂中的一种或多种的组合。
5.如权利要求1所述的一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂的制备方法,其特征在于,包括以下步骤:
S1、按照重量份数计算,将补骨脂6~10份、冬虫夏草3~6份、锁阳5~10份、蛤蚧3~6份、山药15~30份、沙苑子9~15份、姜黄3~9份、火麻仁10~15份、肉桂1~5份、鹿角霜9~15份、仙茅3~10份、阳起石9~30份、紫河车1.5~3份、分心木3~9份、金樱子6~12份、韭菜子5~15份、狗肾2.5~5份九味药加适量水煎煮2次,每次1~2小时,合并煎液,滤过,得到所述提高卵巢卵子数量、质量和储备功能的生物组合物制剂的煎液;
S2、将所述提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂的煎液中加入所述药学上可接受的辅料,制备成所述提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂。
6.权利要求1~5中任一项所述的一种提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的生物组合物制剂在制备用于提高睾丸精子数量、质量和改善碎片成熟及提高男性功能和生育能力、优生优育的药物中的应用。
