Abstract
本公开提出了一种单一分子量环状聚乙二醇脂质,为由单一分子量环状聚乙二醇连接双烷基链形成,在本公开的另一方面公开了上述单一分子量聚乙二醇脂质合成脂质纳米粒的方法,包括:将单一分子量聚乙二醇脂质与胆固醇、磷脂和可离子化脂质溶解混合,得到有机相溶液;将核酸溶解于柠檬酸缓冲液中,得到水相溶液;将水相溶液和有机相溶液通过微流控法或闪沉法反应,得到负载核酸分子的脂质纳米粒。在本公开的另一方面,公开了一种使用上述负载核酸分子的脂质纳米粒在制备用于核酸递送的药物中的应用。

技术领域
本公开涉及高分子材料领域,具体涉及一种单一分子量聚乙二醇脂质及其应用。
背景技术
脂质体(Liposomes,Ls)以其安全无毒、易被生物降解等特点,可以作为药物载体,并能提高药物治疗指数、降低药物毒副作用、减少用药剂量。但是传统的脂质体容易被生物体内的细胞识别并摄取,降低血循环的半衰期,导致药物在到达靶器官前被清除。
聚乙二醇(Polyethylene glycol,PEG)具有亲水性、结构简单、无毒廉价、无免疫原性等特点,可以在脂质体表面引入,避免脂质体被免疫系统快速清除,防止颗粒聚集,建立聚乙二醇的空间位阻作用以增加聚乙二醇脂质的稳定性,延长脂质体的血循环作用。
在相关技术中,由于传统聚乙二醇为不同聚合度的同系混合物,导致不同批次的聚乙二醇性能不同,使得由聚乙二醇改性的脂质体的性能不稳定,在药物传递过程中效率不尽相同。
发明内容
有鉴于此,为解决相关技术中的所述以及其他方面的至少一种技术问题,本公开提出了一种单一分子量环状聚乙二醇脂质,为由单一分子量环状聚乙二醇连接双烷基链形成,其结构如下式(Ⅰ)所示:

将单一分子量环状聚乙二醇脂质与胆固醇、磷脂和可离子化脂质溶解混合,得到有机相溶液;
将核酸溶解于柠檬酸缓冲液中,得到水相溶液;
将水相溶液和有机相溶液通过微流控法或闪沉法反应,得到负载核酸分子的脂质纳米粒。
根据本公开实施例,聚乙二醇脂质在所述有机相溶液中的摩尔百分比为0.4%~50%,所述核酸与所述离子化脂质质量比为1:5~1:50。
根据本公开实施例,水相溶液的pH为3.0~5.0。
根据本公开实施例,微流控或闪沉法中,所述水相溶液和所述有机相溶液的流速比为2:1~8:1。
根据本公开实施例,上述单一分子量环状聚乙二醇脂质合成脂质纳米粒的方法,还包括将所述脂质纳米粒置于分子量为3000Da的透析袋中,于10mM的磷酸缓冲溶液中进行透析纯化。
在本公开的另一方面,公开了一种上述方法合成的单一分子量环状聚乙二醇脂质合成的负载核酸分子的脂质纳米粒在制备用于核酸递送的药物中的应用。
根据本公开实施例,通过单一分子量的聚乙二醇合成一种环状的聚乙二醇脂质。其中,单一分子量的聚乙二醇两端分别连接环状聚乙二醇和双烷基链,构成单一分子量环状聚乙二醇脂质。这种环状结构的单一分子量聚乙二醇脂质精准控制了后续合成脂质纳米粒的具体成分和结构,调控了脂质纳米粒的粒径和功能性,从而实现稳定优异的体内稳定性和生物可降解性。根据本公开实施例,单一分子量的环状的聚乙二醇脂质将其具有亲脂性的尾部插入脂质纳米粒磷脂层,使聚乙二醇锚定于脂质纳米粒表面。锚定的聚乙二醇在体循环过程中可发生解离、脱落,脂质纳米粒表面的聚乙二醇脱落后有利于细胞对脂质纳米粒的摄取。聚乙二醇锚定在脂质纳米粒中的强度由聚乙二醇化脂质的亲脂性尾部长度决定。聚乙二醇亲脂性尾部越长,锚定强度越大。
根据本公开实施例,单一分子量的聚乙二醇脂质相较于传统多分散聚乙二醇脂质会诱导更低的抗聚乙二醇抗体产生,有效降低了重复注射脂质纳米粒产生的加速清除效应,缓解潜在的过敏等不良反应。同时,单一分子量的聚乙二醇脂质合成脂质纳米粒可以通过控制分子量(或聚合度)精准调控不同生产批次的脂质纳米粒的重复性以及质量稳定性。
根据本公开实施例,相较于线性末端的聚乙二醇脂质,由于独特的环状末端结构,一方面,单一分子量的环状结构聚乙二醇脂质组装得到的脂质纳米粒在静脉注射后更不容易吸附血液中的蛋白质,从而更好地保护脂质纳米粒在血液循环中的稳定性,有效降低了脂质纳米粒在生物体的不良反应;另一方面,单一分子量的环状结构聚乙二醇脂质不会诱导机体产生抗聚乙二醇抗体,既保护了脂质纳米粒在体内的稳定性和有效性,又降低纳米粒诱导的炎症反应。
根据本公开实施例,脂质纳米粒中各组分相互配合,以负载药物实现核酸递送的功能。其中,可电离脂质含有正电荷基团,在纳米粒形成过程中与带负电荷的核酸相互作用,并在细胞摄取过程中促进了核酸分子的递送;中性的单一分子量环状脂质结构支撑了纳米颗粒脂质双层结构的形成并稳定其结构排列;胆固醇的膜融合性促进核酸分子的递送;脂质纳米粒表面的聚乙二醇改善了亲水性,避免了脂质纳米粒被免疫系统快速清除,在存储过程中防止颗粒聚集,增加稳定性。

环状结构的聚乙二醇在得到聚乙二醇脂质与胆固醇、蛋白质等物质连接后,相较于同样分子量的直链聚乙二醇会具有更大的表观分子量。有鉴于此,本公开中通过成醚反应制得单一分子量的等长三臂的甘醇分子,保护其中一臂的羟基,诱导剩余两臂闭合成环,脱去另一臂的保护基团从而得到单一分子量的环状聚乙二醇结构。单一分子量的聚乙二醇结构是相关生物医药领域的研究热点,具有应用转化前景,值得深入研究和重点探索。

根据本公开实施例,使用亲脂性尾部碳链长度为14碳的聚乙二醇脂质可使脂质纳米粒具有较为合适的循环时间以及最佳的递送效果。
根据本公开实施例,聚乙二醇脂质在有机相溶液中的摩尔百分比为0.4%~50%,例如可选为0.4%、0.8%、1.6%、3.2%、4.8%、10%、25%、50%等,核酸与所述离子化脂质质量比为1:5~1:50,例如可选为1:5、1:10、1:20、1:35、1:40、1:50等。
根据本公开实施例,在上述成分范围内,验证了不同配比的负载核酸分子的脂质纳米粒具有不同的性质,且负载核酸分子的脂质纳米粒的性质与各成分配比并无具体对应关系。
根据本公开实施例,水相溶液的pH为3.0~5.0,例如可选为3.0、3.5、4.0、4.5、5.0等。

根据本公开实施例,通过研究对不同环状结构的单一分子量聚乙二醇脂质纳米粒在核酸递送过程中的体内分布,代谢和诱导PEG抗体产生以及免疫原性等,验证了脂质纳米粒在核酸治疗领域有着巨大的潜力,其在COVID-19mRNA疫苗中可发挥重要作用。
需要说明的是,所描述的实施例仅仅是本公开一部分实施例,而不是全部的实施例。基于本公开中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的其他实施例,都属于本公开保护的范围。
制备例1环PEG32
合成路线如下式所示:CPEG32-LPEG16-OH:
取如上图所述的8mmol的式1所示的化合物和2mmol的式2所示的化合物混合于250mL的圆底烧瓶中,加入100mL的四氢呋喃充分搅拌使得溶解均匀。再将12mmol的NaH分批次加入体系中,并加热至70℃回流反应12h。过柱纯化得到上图反应式中式3所示的白色固体。
将2mmol的式3所示的化合物加入至250mL圆底烧瓶中,再加入50mL的甲醇充分溶解,然后取2mmol的式4所示的化合物加入至圆底烧瓶中充分搅拌,于20℃的室温条件下反应6h。最终过柱纯化得式5所示的三臂甘醇,其为无色粘稠物。
合成闭环分子的前体(即为下图式9所示化合物)。其合成途径如下:
取1.5mmol的式5所示的化合物加入至250mL圆底烧瓶中,再加入无水50mL的二氯甲烷充分溶解均匀,将1.5mmol的式6所示的化合物、3.0mmol的式7所示的化合物、0.15mmol的式8所示的化合物加入至圆底烧瓶中,并使用惰性气体氮气进行保护,室温反应12h。过柱纯化得式9所示的化合物为淡黄色粘稠物。
再取0.5mmol的式9所示的化合物加入至250mL圆底烧瓶中,加入100mL的四氢呋喃充分搅拌溶解均匀,置于0℃的冰浴环境中充分冷却,将0.5mmol的式10所示的化合物溶解于四氢呋喃中,逐滴加到反应体系中,于20℃的室温条件下反应12h。过柱纯化得到式11所示的化合物为淡黄色固体。
制备得到单一分子量精准环状聚乙二醇,其合成途径如下:
取0.2mmol式11所示的化合物加入至250mL的圆底烧瓶中,加入1000m四氢呋喃充分溶解均匀,置于0℃的冰浴环境中充分冷却,取0.5mmol的NaH加入到反应体系中,于20℃的室温条件下反应72h。过柱纯化得式12所示的化合物为淡黄色粘稠物。
取0.1mmol的式12所示的化合物加入至250mL的圆底烧瓶中,再加入10mL的甲醇充分溶解均匀,取0.1mmol的NaOH水溶液10mL加到反应体系中,进行回流反应12h。过柱纯化得产物14为白色固体。图1示出了本发明实施例中分子量为n=8时的单一分子量精准环状聚乙二醇的核磁共振氢谱图和质谱图,其中a是该n=8时的聚乙二醇的核磁共振氢谱图,b是该n=8时的聚乙二醇的质谱图,通过核磁氢谱图和质谱图的表征能够明确该式14所示的化合物的具体结构。根据与实施例1相似的方式,本公开还合成了n=16对应的单一分子量环状聚乙二醇结构。
实施例1环PEG32-DMA
S1:将如下式所示的化合物1和化合物2溶于5mL甲苯,共沸除水,加入无水四氢呋喃溶液(etrahydrofuran,THF)10mL进行溶解,然后加入氢化钠,在氮气氛围中室温反应12个小时。
S2:反应结束后,过滤除去反应体系内的不溶物,将油泵旋除体系中的溶剂至1mL,再沉降到8mL乙醚与石油醚混合液(1:1)中,离心得到淡黄色粘稠物即为环PEG32-DMA(聚乙二醇脂质)。
对实施例1中的白色固体进行核磁和质谱表征。
图2是本公开中环PEG32-DMG的核磁氢谱图。
如图2所示,核磁氢谱中特征峰a~h验证了实施例1中合成了上述环状聚乙二醇结构。
图3是本公开中环PEG32-DMG的电喷雾质谱图。
如图3所示,电喷雾质谱图中仅有一个特征峰,证明本公开成功合成聚合度为32的单一分子量聚乙二醇。
实施例2环PEG16-DMG
S1:在氩气氛围中,将如下式所示的化合物1,化合物2和4-二甲氨基吡啶(4-Dimethylaminopyridine,DMAP)混合,加入5mL甲苯进行甲苯共沸除水,重复2次,然后加入然后3mL二氯甲烷(Dichloromethane,DCM)溶解。
S2:将二环己基碳二亚胺(Dicyclohexylcarbodiimide,DCC)溶解于1mL DCM中,加入步骤S1的反应体系中。在氩气的保护下,用锡箔纸保护,室温反应24h。
S3:反应结束后,抽滤除去滤渣,得黄色滤液;将有机相浓缩至1mL左右,逐滴沉淀于石油醚/乙醚(v/v=1:1)中,离心,得淡黄色粘稠产物即为环PEG16-DMG(聚乙二醇脂质)。
对实施例2中的白色固体进行核磁和质谱表征。
图4是本公开中环PEG16-DMA的核磁氢谱图。
如图4所示,核磁氢谱中特征峰a~d验证了实施例1中合成了上述环状聚乙二醇结构。
图5是本公开中环PEG16-DMA的电喷雾质谱图。
如图5所示,电喷雾质谱图中仅有一个特征峰,证明本公开成功合成聚合度为16的单一分子量聚乙二醇。
实施例3负载虫荧光素酶mRNA的脂质纳米粒(微流控法)
S1:将环状PEG32-DMA溶解在含有胆固醇、磷脂和可离子化脂质的乙醇溶液中作为有机相(其中,可离子化脂质:磷脂:胆固醇:环状PEG32-DMA的百分摩尔比为46.3%,9.4%,43.9%,0.4%);
S2:将一定量的虫荧光素酶mRNA溶解于pH=4.0的柠檬酸缓冲液中作为水相;
S3:将水相和有机相以3:1的流速比通入微流控设备中,制备脂质纳米粒;
S4:将步骤S3中制备得到的脂质纳米粒置于分子量为3000Da的透析袋中,并放置于10mM的磷酸盐缓冲液中透析2h,得到负载虫荧光素酶mRNA的脂质纳米粒。
在本公开中,基于同样方法,改变了环状PEG32-DMA的含量合成了一系列负载虫荧光素酶mRNA脂质纳米粒,其中,PEG32-DMA的含量分别为0.8%、1.6%、3.2%、4.8%。
实施例4负载CRISPR mRNA和sgRNA的脂质纳米粒
S1:将环状PEG32-DMA溶解在含有胆固醇、磷脂和可离子化脂质的乙醇溶液中作为有机相(其中,可离子化脂质:环状PEG32-DMA:胆固醇:磷脂的百分摩尔比为46.3%:1.6%:42.7%:9.4%);
S2:将一定量的CRISPR mRNA和sgRNA溶解于pH=4.0的柠檬酸缓冲液中作为水相;
S3:将水相和有机相以3:1的流速比通入微流控设备中,制备脂质纳米粒;
S4:将步骤S3中制备得到的脂质纳米粒置于分子量为3000Da的透析袋中,并放置于10mM的磷酸盐缓冲液中透析2h,得到负载CRISPR mRNA和sgRNA的脂质纳米粒。
实施例5负载虫荧光素酶mRNA的脂质纳米粒(闪沉法)
S1:将环状PEG32-DMA溶解在含有胆固醇、磷脂和可离子化脂质的乙醇溶液中作为有机相(其中,可离子化脂质:PEG-脂质:胆固醇:磷脂的百分摩尔比为46.3%:1.6%:42.7%:9.4%);
S2:将一定量的虫荧光素酶mRNA溶解于pH=4.0的柠檬酸缓冲液中作为水相;
S3:将水相和有机相以3:1的体积比将有机相快速注入到水相中,制备脂质纳米粒;
S4:将步骤S3中制备得到的脂质纳米粒置于分子量为3000Da的透析袋中,并放置于10mM的磷酸盐缓冲液中透析2h,得到负载虫荧光素酶mRNA的脂质纳米粒。
应用例1:基于不同环状结构的聚乙二醇脂质的脂质纳米粒包载核酸分子静脉注射后在小鼠体内的转染情况。
按照实施例1、2的方法制备不同环状结构的单一分子量聚乙二醇脂质(环PEG16-DMA、环PEG16-DMG、环PEG32-DMA、环PEG32-DMG),并按照实施例3的步骤制备包载虫荧光素mRNA的脂质纳米粒。通过静脉将包载虫荧光素mRNA的脂质纳米粒注射注射入实验小鼠体内,其中,虫荧光素酶mRNA脂质纳米粒的给药剂量为0.1mg kg-1。分别于静脉注射后的6h和24进行检测,检测前15min向小鼠腹腔注射虫荧光素底物150mg。然后取出小鼠心、肝、脾、肺和肾,通过生物发光成像观测虫荧光素酶mRNA在小鼠体内和脏器的分布和表达情况。
图6是本公开应用例1中小鼠注射包载虫荧光素mRNA的脂质纳米粒6小时后的生物发光成像图。
如图6所示,本公开制备得到的不同环状结构的单一分子量聚乙二醇脂质在小鼠体内均有明显分布。其中,由环PEG32-DMA和环PEG32-DMG制备得到的包载虫荧光素mRNA的脂质纳米粒的表达情况最佳。
图7是本公开应用例1中小鼠注射包载虫荧光素mRNA的脂质纳米粒6、12小时后的生物发光成像图。
如图7所示,本公开制备得到的不同环状结构的单一分子量聚乙二醇脂质主要在小鼠的肝脏和肾脏中有所分布,在心脏中少量分布,其中,环PEG32-DMA和环PEG32-DMG的表达效果最好。同时,其含量与时间并无显著关系。
应用例2:基于不同环状结构的精准PEG脂质制备的脂质纳米粒包载eGFP mRNA静脉注射后在小鼠肝脏细胞群中的表达情况
将基于上述实施例的方法制备得到的不同聚乙二醇脂质和线型聚乙二醇2000脂质含有绿色荧光蛋白mRNA(eGFPmRNA)的脂质纳米粒(其中聚乙二醇脂质的种类包括:线型PEG2000-DMA、环状PEG16-DMA、环状PEG32-DMA、线型PEG2000-DMG、环状PEG16-DMG、环状PEG32-DMG))通过静脉注射到实验鼠体内,其中,绿色荧光蛋白mRNA的给药剂量为0.15mg kg-1。于12h取出小鼠肝脏,采用脏组织解离试剂盒(美天旎公司)制备肝脏单细胞悬液,进一步对不同细胞表面的标记进行染色,通过流式细胞仪分析肝脏中肝细胞、肝内皮细胞和肝枯否细胞中eGFP mRNA的表达情况。
图8~10是本公开中包含eGFP mRNA的环状聚乙二醇脂质纳米粒在小鼠肝实质(肝细胞)细胞、肝内皮细胞、肝巨噬细胞中的表达情况柱状图。
如图8~10所示,在小鼠的肝细胞内,环状单一分子量的聚乙二醇脂质纳米粒的表达情况显著优于传统的线型PEG2000脂质纳米粒。同时,在相同分子量相同结构、成分的基础上,PEG-DMA脂质纳米粒具有更好的表达效果。
应用例3:
选用6周龄的雌性Balb/c小鼠,以150μg/鼠的剂量通过静脉注射各组脂质纳米粒后,分别于4h和24h通过眼眶收集小鼠血液。
室温静置2h后,在温度为4℃下,2500rpm的转速离心20min。将分离的血清收集至1.5mL离心管中,于-20℃储存备用。
采用流式细胞术检测环状PEG16-DMA、环状PEG32-DMA和线性多分散PEG2000-DMA所制备的脂质纳米粒在血液树突状细胞、中性粒细胞、单核细胞、B细胞、T淋巴细胞和自然杀伤性细胞中的摄取情况。
图11~16是本公开中环状不同脂质纳米粒在血液树突状细胞、中性粒细胞、单核细胞、血液B细胞、血液T淋巴细胞和自然杀伤性细胞中的摄取情况柱状图。其中,1表示PEG2000-DMA脂质纳米粒,2表示环状PEG16-DMA脂质纳米粒,3表示环状PEG32-DMA脂质纳米粒。
如图11~16所示,在血液树突状细胞、中性粒细胞、B细胞、T淋巴细胞和自然杀伤性细胞中环状单一分子量聚乙二醇脂质纳米粒的摄取表达情况均优于传统PEG2000-DMA脂质纳米粒。同时,在结构、成分的基础上,环状PEG16-DMA脂质纳米粒在树突状细胞中具有更好的表达效果。
应用例4:基于不同环状结构的精准PEG脂质制备的脂质纳米粒抵抗蛋白吸附的研究
血清离心处理:将小鼠血清用20000g的离心速度离心1h以除去蛋白的聚集体,然后用Bradford法测定其中的总蛋白浓度为66.7g/L.
0.7M的蔗糖溶液配置:3.5mL的2M的蔗糖溶液混合6.5mL的PBS buffer溶液(其中包含:4.3mM Na2HPO4,1.4mM KH2PO4,137mM NaCl,2.7mM KCl)。
脂质纳米粒与血清共孵育:取1mL不同环状结构的聚乙二醇脂质纳米粒(MeO-PEG2000-DMA脂质纳米粒、环状PEG16-DMA脂质纳米粒、环状PEG32-DMA脂质纳米粒)。向其中加入2.7mL的经过离心之后的血清,在37℃恒温摇床中孵育30min。然后将脂质纳米粒与血清混合液以与蔗糖溶液体积比1:1的比例,缓慢加入到蔗糖溶液上方,在转速为20000g,,温度为4℃的条件下下离心30min,将离心后的脂质纳米粒用PBS重新分散后加入到蔗糖溶液上方再次离心,此过程重复三次。
取经过离心后的脂质纳米粒加入30μL的PBS,吸取5μL用于BCA蛋白定量,使用酶标仪测量其在562nm的紫外吸收强度。
图17是本公开中不同环状结构的聚乙二醇脂质纳米粒的蛋白吸附归一化结果柱状图。其中,以传统商业的MeO-PEG2000-DMA制备的脂质纳米粒为标准,将各组吸附蛋白质的含量进行归一化处理。
如图17所示,传统的MeO-PEG2000-DMA脂质纳米粒对蛋白质的吸附能力最强,环状PEG16-DMA脂质纳米粒次之,环状PEG32-DMA脂质纳米粒吸附的蛋白质含量最少。
应用例5:基于不同环状结构的精准PEG脂质制备的脂质纳米粒在4T1细胞中的递送mRNA的能力
制备Cy5荧光染料标记的负载mRNA分子的MeO-PEG2000-DMA脂质纳米粒、MeO-PEG2000-DMG脂质纳米粒、环状PEG16-DMA脂质纳米粒、环状PEG32-DMA脂质纳米粒、环状PEG16-DMG脂质纳米粒和环状PEG32-DMG脂质纳米粒。
取对数生长期的4T1细胞接种于玻璃板底的共聚焦孔板中(1×105cells/well),于细胞培养箱中孵育24h至细胞贴壁完全。去除旧的培养基并用新制PBS溶液润洗两遍。每孔中加入含不同组别的脂质纳米粒的新鲜培养基,并将孔板置于37℃分别孵育6h。
孵育完成后,用移液枪吸去含上述脂质纳米粒的培养基并用PBS清洗细胞两次。向共聚焦培养皿中加入200μL细胞核染色液,室温染色10min。染色结束后,加入500μL PBS清洗细胞。吸去PBS后加入300μLPBS并将培养皿置于共聚焦显微镜下观察脂质纳米粒中mRNA在细胞中的分布情况。
图18是本公开中以不同环状脂质纳米粒递送的mRNA在细胞中的分布情况图。其中,红色荧光表示mRNA的分布,绿色荧光表示溶酶体的分布
脂质纳米粒在递送核酸的过程中会吸附在细胞膜表面,通过细胞的内吞作用,脂质纳米粒进入到细胞内形成内体,内体成熟后会和溶酶体相融合,溶酶体内的酸性环境和水解酶可以降解脂质纳米粒和脂质纳米粒内部的mRNA。因此,mRNA降解前的内体逃逸被认为是核酸药物疗法成功的关键步骤。mRNA逃逸进入细胞质,和细胞核糖体相结合进行翻译步骤,产生相应的蛋白,病毒蛋白刺激机体产生免疫反应,进而形成免疫记忆。
如图18所示,环状结构的单一分子量聚乙二醇脂质纳米粒递送的mRNA在细胞质的分布情况优于传统MeO-PEG2000聚乙二醇脂质。同时,被溶酶体捕捉的环状结构的单一分子量聚乙二醇脂质纳米粒递送的mRNA所产生的荧光效应明显低于传统MeO-PEG2000聚乙二醇脂质负载的mRNA所产生的荧光效应。因此,本公开中合成的环状结构的单一分子量聚乙二醇脂质纳米粒在核酸递送过程中具有优异的传递性能和药物效力。

Claims (10)
1.一种单一分子量环状聚乙二醇脂质,为由单一分子量环状聚乙二醇连接双烷基链形成,其结构如下式(Ⅰ)所示:
其中,k=0-500,n=1-500,R为R1为C8-C20烷基链。
2.根据权利要求1所述的单一分子量环状聚乙二醇脂质,其中,所述单一分子量环状聚乙二醇脂质的结构如下式A或B所示:
其中,k=0-500,n=1-500,m=8-20。
3.根据权利要求1或2所述的单一分子量环状聚乙二醇脂质,其中,k=8~50,n=4~25,且k=2n-1。
4.根据权利要求1所述的单一分子量环状聚乙二醇脂质,其中,所述单一分子量环状聚乙二醇脂质的结构如下式所示:
5.一种使用如权利要求1~4任一项所述的单一分子量环状聚乙二醇脂质合成脂质纳米粒的方法,包括:
将单一分子量环状聚乙二醇脂质与胆固醇、磷脂和可离子化脂质溶解混合,得到有机相溶液;
将核酸溶解于柠檬酸缓冲液中,得到水相溶液;
将所述水相溶液和所述有机相溶液通过微流控法或闪沉法反应,得到负载核酸分子的脂质纳米粒。
6.根据权利要求5所述的方法,其中,所述聚乙二醇脂质在所述有机相溶液中的摩尔百分比为0.4%~50%,所述核酸与所述离子化脂质质量比为1:5~1:50。
7.根据权利要求5所述的方法,其中,所述水相溶液的pH为3.0~5.0。
8.根据权利要求5所述的方法,其中,所述微流控或闪沉法中,所述水相溶液和所述有机相溶液的流速比为2:1~8:1。
9.根据权利要求5所述的方法,还包括将所述脂质纳米粒置于分子量为3000Da的透析袋中,于10mM的磷酸缓冲溶液中进行透析纯化。
10.一种使用如权利要求5~9任一项所述方法得到的负载核酸分子的脂质纳米粒在制备用于核酸递送的药物中的应用。
