Abstract
The invention relates to antibacterial and anti-inflammatory toothpaste containing pure natural plant extracts and a preparation method thereof, belonging to the technical field of toothpaste. The antibacterial and anti-inflammatory toothpaste provided by the invention comprises the following raw materials: dendrobium officinale, centella asiatica, salvia miltiorrhiza, humifuse euphorbia herb, fibraurea stem, prinsepia utilis royle, agilawood, clove, honeysuckle flower, carboxymethyl chitosan, sodium alginate, bamboo salt, lysozyme, tetrasodium pyrophosphate, a humectant, a friction agent, a surfactant, a flavoring agent, a thickening agent, a preservative and water. The antibacterial and anti-inflammatory toothpaste provided by the invention utilizes a gel network formed by carboxymethyl chitosan and oxidized sodium alginate, and combines the extremely strong viscosity of the dendrobium candidum extract, so that the plant extract adsorbed in the gel network is firmly adsorbed on teeth, gums and oral inner films, the slow release effect of the toothpaste is improved, and the effects of inhibiting bacteria, diminishing inflammation and promoting healing of fine wounds of the oral cavity are achieved for a long time.

s1, crushing dendrobium candidum, centella asiatica, salvia miltiorrhiza, humifuse euphorbia herb, fibraurea stem, prinsepia utilis royle, agilawood, clove and honeysuckle into 180-200 meshes, mixing, adding normal saline, soaking, boiling, filtering and concentrating to obtain pure natural plant paste;
s2, adding carboxymethyl chitosan, sodium alginate, bamboo salt, lysozyme, tetrasodium pyrophosphate, a humectant, a friction agent, a surfactant, a flavoring agent, a thickening agent, a preservative and water into the pure natural plant paste in the S1, stirring to prepare an aqueous solution, sucking the aqueous solution into a paste preparation machine, stirring and grinding for 20-30 min until the paste is uniform, and synchronously carrying out vacuum defoaming to obtain the antibacterial and anti-inflammatory toothpaste containing the pure natural plant extract.

(3) The bamboo salt added in the invention contains abundant mineral components and microelements necessary for human bodies such as potassium, magnesium, zinc, calcium and the like, and has good curative effect on inflammation. 10 parts of dendrobium candidum, 10 parts of centella asiatica, 10 parts of red sage root, 5 parts of humifuse euphorbia herb, 5 parts of fibraurea stem, 5 parts of privet fruit, 5 parts of agilawood, 5 parts of clove, 5 parts of honeysuckle flower, 15 parts of carboxymethyl chitosan, 10 parts of sodium alginate, 10 parts of bamboo salt, 0.2 part of lysozyme, 0.1 part of tetrasodium pyrophosphate, 20 parts of sorbitol, 10 parts of polyethylene glycol-40010 parts of silicon dioxide, 15 parts of sodium lauryl sulfate, 1.5 parts of essence, 0.9 part of erythritol, 0.2 part of saccharin sodium, 0.1 part of xanthan gum, 0.1 part of cellulose gum, 0.2 part of sodium benzoate and 15 parts of water.

5 parts of dendrobium candidum, 5 parts of centella asiatica, 5 parts of red-rooted salvia root, 1 part of humifuse euphorbia herb, 1 part of fibraurea stem, 1 part of privet fruit, 1 part of agilawood, 1 part of clove, 1 part of honeysuckle flower, 10 parts of carboxymethyl chitosan, 5 parts of sodium alginate, 5 parts of bamboo salt, 0.1 part of lysozyme, 0.2 part of tetrasodium pyrophosphate, 40 parts of sorbitol, 40 parts of polyethylene glycol-40010 parts of silicon dioxide, 20 parts of sodium lauryl sulfate, 2.5 parts of essence, 0.9 part of erythritol, 0.2 part of saccharin sodium, 1 part of xanthan gum, 0.5 part of cellulose gum, 1 part of sodium benzoate and 30 parts of water.
The method specifically comprises the following steps:

Basically the same as in example 1, except that Dendrobium officinale was not added.
Comparative example 2
Substantially the same as in example 1, except that no root of red-rooted salvia was added.
Comparative example 3
Substantially the same as in example 1, except that fibraurea stem was not added.
Comparative example 4
Substantially the same as in example 1, except that prinsepia utilis royle was not added.
Comparative example 5
Substantially the same as in example 1, except that carboxymethyl chitosan was not added.
Comparative example 6
Substantially the same as in example 1, except that lysozyme was not added.
Test example 1
120 Balb/C mice (60 male and female mice) were selected and divided into 5 test groups (corresponding to examples 1-5) and 1 control group, and 10 male and female mice were fed with the extract obtained in examples 1-5 (concentration of 0.8g/10 mL), and fasted for 8 hours before and 4 hours after administration, but not water. Each mouse was perfused with 0.3mL/10g of the suspension at a weight of each mouse once a day, each time in the morning, in the evening, and at 4h intervals, three times total, and observed continuously for 7 days. Observing the growth state and active diet of the white mice, and identifying hematology, hematochemistry, organ tissue structure, urine routine and the like. The result shows that all mice survive healthily without any toxic or side effect; the heart, liver, spleen, kidney, lung and intestine tissue states were observed anatomically, and were indistinguishable from the control mice. The dose of mice in the test is 94 times of the clinical daily dose of adults, and the single dose of adults is 315 times, which means that the product is safe for adults.
Test example 2
With reference to the antibacterial performance test method of the disposable hygienic product hygienic standard GB 15979-2002C4 dissolution antibacterial products, the antibacterial and anti-inflammatory toothpastes prepared in examples 1-5 and comparative examples 1-6 are used as test objects, and the inhibition rates (%) of the toothpastes on staphylococcus aureus, escherichia coli, candida albicans, streptococcus sanguinis, streptococcus mutans and actinomycetes viscosus are measured, and the results are shown in Table 1:

As can be seen from table 1, the toothpaste of the invention has excellent antibacterial and antiseptic effects, and the combination of dendrobium candidum, red sage root, fibraurea stem, prinsepia utilis royle, carboxymethyl chitosan, sodium alginate, bamboo salt, lysozyme and the like synergistically has an effect on antibacterial and antiseptic, and the addition of lysozyme and carboxymethyl chitosan is beneficial to better improving oral environment.
Test example 3
110 Balb/C mice (male and female halves) are selected, the mice are randomly divided into 11 groups, 10 mice in each group are anesthetized by diethyl ether, 20 mu L of dimethylbenzene is uniformly smeared on the right ear of the mice, after 30min, 0.5mL of toothpaste prepared in examples 1-5 and comparative examples 1-6 is smeared on the right ear inflammation part of the mice for 10min for 6 times continuously, the left ear is not treated, after 4h, the mice are killed, the ears are cut off immediately, the ears on the same part are picked by an 8mm puncher, and the swelling degree and swelling rate are calculated by weighing; swelling degree = right ear weight-left ear weight, swelling rate = swelling degree/left ear weight, average, results are shown in table 2:

As can be seen from Table 2, the anti-inflammatory effects of the present invention in examples 1 to 5 were significantly different from those in comparative examples 1 to 6, indicating that the antibacterial and anti-inflammatory toothpaste of the present invention had good anti-inflammatory effects.
Test example 4
About 22g of female Balb/C mice are placed on a hot plate pain tester, 200 qualified mice are screened (the foot time after licking is less than 5s or more than 30s for discarding, the cut-off time is set for preventing foot scalding, and the cut-off time is generally 60 s), 110 female mice are taken as pain threshold indexes by taking the incubation period of the foot reaction of the mice, and the mice are divided into 11 groups, namely, the groups of examples 1-5 and comparative examples 1-6. The toothpastes prepared in examples 1 to 5 and comparative examples 1 to 6 were applied to the foot scalding portion of mice once a day for 6 times a day for 2 consecutive days, and the time for the foot addition to occur within 60 minutes and 120 minutes after the last dose was measured, and the results are shown in Table 3:

As can be seen from Table 3, after the mice are coated with the toothpastes for 1h and 2h, the time for adding the toothpastes prepared in the embodiments 1-5 of the present invention to the mice is obviously shorter than that of the toothpastes prepared in the comparative examples 1-6, which indicates that the toothpaste of the present invention can reduce the time for adding the toothpastes to the mice, and further indicates that the compatibility of the active ingredients in the ingredients of the present invention has good analgesic effect.
It is apparent that the above examples are given by way of illustration only and are not limiting of the embodiments. Other variations and modifications of the present invention will be apparent to those of ordinary skill in the art in light of the foregoing description. It is not necessary here nor is it exhaustive of all embodiments. And obvious variations or modifications thereof are contemplated as falling within the scope of the present invention.
Claims (10)
1. An antibacterial and anti-inflammatory toothpaste containing pure natural plant extracts is characterized by comprising the following raw materials in parts by weight: 5-10 parts of dendrobium candidum, 5-10 parts of centella asiatica, 5-10 parts of red sage root, 1-5 parts of humifuse euphorbia herb, 1-5 parts of caulis fibraureae recisae, 1-5 parts of prinsepia utilis, 1-5 parts of agilawood, 1-5 parts of clove, 1-5 parts of honeysuckle flower, 10-15 parts of carboxymethyl chitosan, 5-10 parts of sodium alginate, 5-10 parts of bamboo salt, 0.1-0.2 part of lysozyme, 0.1-0.2 part of tetrasodium pyrophosphate, 30-50 parts of humectant, 15-20 parts of friction agent, 1.5-2.5 parts of surfactant, 1.0-2.0 parts of flavoring agent, 0.2-1.5 parts of thickening agent, 0.2-1 part of preservative and 15-30 parts of water.
2. The antibacterial and anti-inflammatory toothpaste containing pure natural plant extracts according to claim 1, wherein the humectant is selected from one or more of sorbitol, polyethylene glycol, glycerin and propylene glycol.
3. Antibacterial and anti-inflammatory toothpaste containing purely natural plant extracts according to claim 1, characterized in that the abrasive is selected from silica and/or calcium pyrophosphate.
4. The antibacterial and anti-inflammatory toothpaste containing pure natural plant extracts according to claim 1, wherein the surfactant comprises sodium lauryl sulfate and sodium lauroyl sarcosinate.
5. The antibacterial and anti-inflammatory toothpaste containing pure natural plant extracts according to claim 1, wherein the flavoring agent comprises essence, erythritol and saccharin sodium; the mass ratio of the essence to the erythritol to the saccharin sodium is 0.5-1:0.5-1:0.1-0.3.
6. The antibacterial and anti-inflammatory toothpaste containing pure natural plant extracts according to claim 1, wherein the thickener is selected from one or more of xanthan gum, cellulose gum, carboxymethyl cellulose and hydroxyethyl cellulose.
7. The antibacterial and anti-inflammatory toothpaste containing pure natural plant extracts according to claim 1, wherein the preservative is selected from one or more of sodium benzoate, potassium sorbate and parahydroxybenzoate.
8. The method for preparing antibacterial and anti-inflammatory toothpaste containing pure natural plant extract according to any one of claims 1 to 7, characterized by comprising the following steps,
s1, crushing dendrobium candidum, centella asiatica, salvia miltiorrhiza, humifuse euphorbia herb, fibraurea stem, prinsepia utilis royle, agilawood, clove and honeysuckle into 180-200 meshes, mixing, adding normal saline, soaking, boiling, filtering and concentrating to obtain pure natural plant paste;
s2, adding carboxymethyl chitosan, sodium alginate, bamboo salt, lysozyme, tetrasodium pyrophosphate, a humectant, a friction agent, a surfactant, a flavoring agent, a thickening agent, a preservative and water into the pure natural plant paste in the S1, stirring to prepare an aqueous solution, sucking the aqueous solution into a paste preparation machine, stirring and grinding for 20-30 min until the paste is uniform, and synchronously carrying out vacuum defoaming to obtain the antibacterial and anti-inflammatory toothpaste containing the pure natural plant extract.
9. The method for preparing antibacterial and anti-inflammatory toothpaste containing pure natural plant extracts according to claim 8, wherein in S1, the mass ratio of the physiological saline to the natural plant is 7-10:1.
10. the method for preparing antibacterial and anti-inflammatory toothpaste containing pure natural plant extract according to claim 8, wherein in S1, the soaking time is 3-4 h; the boiling time is 1h-2h; the relative density after concentration is 1.05-1.10.
