技术领域
本发明涉及药物制备技术领域,具体涉及新型抗微生物肽Perceptide-TJ-2(简称P2)及其在制备广谱抗病毒药物中的用途。
背景技术
在人类传染病中有70%以上是由病毒引起的,长期严重危害人类健康,尤其是近几年在全球爆发的新型冠状病毒,造成了数百万人的死亡,即使痊愈康复,也会遗留多种后遗症,如易疲劳、呼吸急促、肌肉无力、关节疼痛和记忆力衰退等症状。尽管现代医药科技的发展已经使许多难治性疾病得到了有效控制,但病毒性疾病缺乏特效药的困境仍是摆在医学科学家面前的难题。
对于通过特异性抑制病毒而不是其宿主细胞起作用的抗病毒药物,存在耐药性的可能性,通常是由抗病毒药物分子靶点的突变而引起,使抗病毒药物治疗效果降低或完全无效,因此产生抗病毒耐药性的病毒治疗难度更大。综合以上情况,使得抗病毒药物的临床应用以及新型药物的开发面临着巨大的挑战。
抗微生物肽(AMP)是一类广泛存在于自然界生物体中的小肽类物质,通常少于50个氨基酸,强阳离子,两亲性,是机体先天性免疫系统的重要组成部分,在抑制多种病原微生物中起重要作用。与干扰病原微生物代谢过程的抗生素不同,抗微生物肽通常是利用自身带正电荷的阳离子特性,可识别和结合带阴离子电荷的病原微生物,使抗微生物肽分子的疏水端插入包膜的磷脂中,进而改变膜的通透性,同时形成跨膜电位,打破酸碱平衡,影响渗透压平衡,诱导胞内内容物外漏而发挥抗病毒作用。由于病原微生物一般不会改变自己的阴离子性质,膜结构变化较小,因此具有较强的耐药性和广谱性,在抗病毒药物存在局限性的情况下,抗微生物肽有望解决长期困扰临床病毒耐药性的问题,具有巨大的医学应用前景。
抗微生物肽虽然是机体固有免疫的产物,具有一定抗病毒活性,但目前大多数是从生物体内提取,不仅存在生物活性较低,稳定性差,化学合成成本较高,纯化困难等缺点,在实际使用中,还会受到高浓度盐离子屏蔽、毒性大、结构稳定性差等各种因素限制,使其活性效能下降。目前,病毒性疾病的治疗方式主要有两种:疫苗注射和药物治疗。疫苗研制周期长,一般需要2年以上甚至更长的时间。常规的抗病毒药物一般为化学成分,首先,其毒副作用较大,长期积累还会对多种器官,尤其是排毒器官造成很大负担。其次,由于化学药物主要作用于病毒生命周期关键酶和蛋白质等易发生突变的靶点,因此面临不同程度的耐药性,大大限制了此类药物的应用和发展。
发明内容
为获得一种新型稳定性好、耐药性低且具有广谱性的抗病毒活性多肽,本发明提供了新型抗微生物肽Perceptide-TJ-2(P2)及其在制备广谱抗病毒药物中的用途。本发明提供的新型抗微生物肽Perceptide-TJ-2对汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒等多种传染性病原体具有广谱抑制作用,不仅有效降低细胞毒性和耐药性,还显著提高抗病毒活性,为开发新一代广谱抗病毒药物提供依据和基础,为保障人类健康提供全新的技术手段。

进一步地,所述抗微生物肽Perceptide-TJ-2能够抑制汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒的增殖,具有广谱抗病毒作用。
进一步地,所述抗微生物肽Perceptide-TJ-2对汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒的半数有效抑制浓度分别为:HTNV EC50=2.076μM,CHIKV EC50=0.367μM,HSV-1EC50=0.928μM,DENV-2EC50=0.989μM。
进一步地,所述抗微生物肽Perceptide-TJ-2对汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒的mRNA均具抑制作用。
进一步地,从0.078μM低浓度到50μM高浓度呈梯度依赖性,浓度越高,Perceptide-TJ-2对汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒mRNA的抑制作用越显著。
进一步地,所述抗微生物肽Perceptide-TJ-2对正常细胞没有影响。
进一步地,所述抗微生物肽Perceptide-TJ-2能够抑制Vero、Huh7和A549中的汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒的增殖。
与现有技术相比,本发明的有益效果在于:
1、本发明提供的新型抗微生物肽Perceptide-TJ-2对汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒等多种传染性病原体的具有广谱抑制活性,不仅有效降低细胞毒性和耐药性,还显著提高抗病毒活性,最终达到构建广谱性多肽治疗药物研究平台的目的。

下面对本发明的具体实施方式进行详细描述,但应当理解本发明的保护范围并不受具体实施方式的限制。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动的前提下所获得的所有其他实施例,都属于本发明保护的范围。本发明各实施例中所述实验方法,如无特殊说明,均为常规方法,下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实施例1:一种新型抗微生物肽Perceptide-TJ-2(简称P2)及其构建方法和用途。
一、新型抗微生物肽Perceptide-TJ-2的设计、合成及细胞毒性评价。
1、全新抗微生物肽Perceptide-TJ-2基于人工智能方法构建深度学习网络模型,设计并合成:利用已知数据库中抗菌肽序列及最小抑菌浓度数据集完成训练和性能评估,建立抗菌活性预测模型和抗微生物肽生成模型。人工智能模型由Python语言编写,基于PyTorch深度学习框架开发。抗微生物肽生成模型是一个生成对抗网络,其生成器和判别器均使用二维卷积神经网络。随后,对完成训练的生成器输入随机数组生成特征矩阵,根据与氨基酸特征编码的相似性转换为氨基酸字符,得到一系列全新抗微生物短肽序列。最后,将生成序列输入至抗菌活性预测模型中进行打分并排序,结合生物信息学软件计算其各类理化性质(分子量、电荷数、等电点、疏水氨基酸占比等),保证人工设计多肽在各项性质中的分布多样性和功能有效性,挑选并合成全新多肽Perceptide-TJ-2,多肽Perceptide-TJ-2的氨基酸序列如SEQ ID NO.1所示,序列性质轮状图和预测结构如图1所示。

2、抗微生物肽Perceptide-TJ-2细胞毒性评价
在96孔培养板中接种处于对数生长期的三种易感细胞,每孔2×104个。24h后加入从100到0.2μmol·L-1的2倍浓度梯度稀释的多肽P2,同时设PBS对照组和空白对照组。置于37℃、5% CO2条件下孵育48h后,每孔加入10%CCK8试剂,培养箱放置1h,使用酶标仪在450nm处测定吸光度值,GraphPad Prism软件计算细胞活力。CC 50表示将细胞活力降低50%对应的多肽浓度。
结果如图2所示:三种病毒易感细胞Vero、Huh7和A549的CC 50分别为49.91μM、56.49μM和65.92μM,即多肽P2的细胞毒性均≥50μM,毒性较低,对细胞影响小,可用于体外抗病毒实验。
二、抗微生物肽Perceptide-TJ-2广谱抗病毒活性评价
1、体外抗病毒活性实验细胞感染和检测时间点
汉滩病毒(HTNV)在A549细胞中感染5天后检测;基孔肯雅病毒(CHIKV)在Vero-E6细胞中感染24小时后检测;1型单纯疱疹病毒(HSV-1)在Vero-E6细胞中感染24小时后检测;2型登革病毒(DENV-2)在Huh 7细胞中感染48小时后检测。
2、抗微生物肽Perceptide-TJ-2在蛋白水平抗病毒活性评价
将易感细胞传代至96孔板细胞培养板,过夜培养,待细胞密度达到70%时,取MOI=1的病毒与12.5-0.078μM 2倍浓度稀释的多肽Perceptide-TJ-2预混10min后感染细胞,同时作病毒对照和正常细胞对照,5% CO2培养箱中感染1h后,弃病毒液,PBS洗1次,加入细胞培养液,置于37℃,5% CO2培养箱中培养至检测时间,用4%多聚甲醛固定15min,PBS洗1次后加入0.2%Triton X-100的缓冲液渗透10min,PBS洗1次后加入5% BSA室温封闭1h,再加入病毒特异性一抗,4℃孵育过夜,PBS洗3次,加入相应种属的荧光二抗,37℃孵育1h,洗涤后用DAPI染色细胞核5min,荧光显微镜观察,并用ImageJ软件计算荧光值。
实验结果如图3和图4所示,相对于病毒对照,多肽Perceptide-TJ-2对四种病毒均具有明显的抑制作用,从0.078μM低浓度到12.5μM高浓度呈梯度依赖性;其半数有效抑制浓度(EC50)分别为:HTNV EC50=2.076μM,CHIKV EC50=0.367μM,HSV-1EC50=0.928μM,DENV-2EC50=0.989μM。
3、抗微生物肽Perceptide-TJ-2在mRNA水平抗病毒活性评价
在6孔板中接种易感细胞,病毒感染和多肽Perceptide-TJ-2处理方法同2中。以病毒各自检测时间点提取样本总RNA,参照TaKaRa说明书将mRNA反转录为cDNA,qRT-PCR SYBRgreen法检测病毒基因,GAPDH为内参对照。病毒检测基因及引物序列见表1。根据各样品的CT值,运用2-ΔΔCT法计算各样品相对于对照组的mRNA水平。

结果如图5所示,相对于病毒对照组,多肽Perceptide-TJ-2(P2)对四种病毒mRNA均具有明显的抑制作用,从0.078μM低浓度到50μM高浓度呈梯度依赖性,浓度越高,抑制作用越显著。
4、抗微生物肽Perceptide-TJ-2对病毒增殖抑制活性的评价
在25cm3培养瓶中铺易感细胞,病毒感染和多肽P2处理方法同2。以病毒各自检测时间点胰酶消化细胞后,加入10% DMEM培养基,3000rpm离心15分钟,转移至1.5ml离心管,PBS缓冲液,悬浮细胞后,1000rpm离心10分钟,弃上清,加入电镜固定液,4℃放置24小时后送电镜中心制备超薄切片后进行电镜观察。
结果如图6所示:在没有加入多肽P2的病毒对照中,细胞核或细胞质中可观察到HTNV、CHIKV、HSV-1和DENV-2四种病毒颗粒;加入多肽P2后,细胞中的病毒(HTNV、CHIKV、HSV-1和DENV-2)增殖被抑制,在细胞核或细胞质中没有观察到病毒颗粒;正常细胞加入多肽P2后,细胞形态完整,毒性较小。
本发明基于人工智能方法,融合已知抗菌肽库不同层级信息,构建深度神经网络模型,在数据集完成训练和性能评估,在明确优势序列后,设计并合成全新抗微生物肽序列。合成的多肽经过体外实验评估显示新型多肽P2不仅有效降低细胞毒性和耐药性,还能够显著提高广谱抗病毒活性,为研制新型广谱抗病毒药物,构建多肽治疗药物研究平台提供重要依据。
尽管已描述了本发明的优选实施例,但本领域内的技术人员一旦得知了基本创造性概念,则可对这些实施例作出另外的变更和修改。所以,所附权利要求意欲解释为包括优选实施例以及落入本发明范围的所有变更和修改。
显然,本领域的技术人员可以对本发明进行各种改动和变型而不脱离本发明的精神和范围。这样,倘若本发明的这些修改和变型属于本发明权利要求及其等同技术的范围之内,则本发明也意图包含这些改动和变型在内。
Claims (9)
1.新型抗微生物肽Perceptide-TJ-2,其特征在于,所述新型抗微生物肽Perceptide-TJ-2的氨基酸序列如SEQ ID NO.1所示;
SEQ ID NO.1:GWKDFKKTIKKLLRGASRLLKF。
2.权利要求1所述的新型抗微生物肽Perceptide-TJ-2在制备广谱抗病毒药物中的用途。
3.根据权利要求2所述的用途,其特征在于,所述病毒包括汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒。
4.根据权利要求3所述的用途,其特征在于,所述抗微生物肽Perceptide-TJ-2能够抑制汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒的增殖,具有广谱抗病毒作用。
5.根据权利要求4所述的用途,其特征在于,所述抗微生物肽Perceptide-TJ-2对汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒的半数有效抑制浓度分别为:HTNVEC50=2.076μM,CHIKV EC50=0.367μM,HSV-1EC50=0.928μM,DENV-2EC50=0.989μM。
6.根据权利要求5所述的用途,其特征在于,所述抗微生物肽Perceptide-TJ-2对汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒mRNA均具有抑制作用。
7.根据权利要求6所述的用途,其特征在于,从0.078μM低浓度到50μM高浓度呈梯度依赖性,浓度越高,Perceptide-TJ-2对汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒mRNA的抑制效果越显著。
8.根据权利要求5所述的用途,其特征在于,所述抗微生物肽Perceptide-TJ-2对正常细胞没有影响。
9.根据权利要求5所述的用途,其特征在于,所述抗微生物肽Perceptide-TJ-2能够抑制Vero、Huh7和A549中的汉滩病毒、基孔肯雅病毒、1型单纯疱疹病毒和2型登革病毒的增殖。
