背景技术
烟草特有亚硝胺(Tobacco-Specific Nitrosamines,TSNAs)是烟草特有的N-亚硝基类化合物,从分子水平上看,其是由烟草生物碱和亚硝基反应生成的复合物,在烟叶和烟气中都有存在,目前已证实的有8种,其中最主要的4种为N-亚硝基降烟碱(NNN),4-(甲基亚硝胺)-1-(3-吡啶基)-1-丁酮(NNK),N-亚硝基假木贼碱(NAB)和N-亚硝基新烟碱(NAT)。有研究表明,NNN和NNK具有一定的致癌性和致突变性。NNK又被确定为影响卷烟烟气危害性最重要的有害成分指标之一。因此,减少烟草和烟草产品中的TSNAs含量对减少其有害影响至关重要。
烟叶中存在丰富的氨基化合物,包括氨基酸、蛋白质、生物碱等,其中烟碱是烟草中最重要的生物碱,占总生物碱含量的94%以上,另外比较重要但含量较少的是降烟碱、新烟草碱和假木贼碱。目前普遍认为NNK来源于烟碱,NNN来源于烟碱和降烟碱,NAT来源于新烟草碱,NAB来源于假木贼碱。因此,减少烟草和烟草产品中的烟碱含量对减少TSNAs有一定帮助。
在各类烟草制品中,雪茄烟的总TSNAs含量排在首位。雪茄烟TSNAs含量比混合型卷烟和烤烟型卷烟高1-2个数量级(史宏志等,2002;史宏志等,2012;冷红琼等,2011),其中雪茄烟总TSNAs含量可高达50g/g以上(Hilldrup et al.,2019)。从目前的研究可知,TSNAs的形成主要有两条途径,一是烟草中的生物碱直接发生亚硝化反应形成,二是通过亚硝酸盐或氮氧化物与生物碱的反应形成的,而其中的亚硝酸盐是烟叶中的硝酸盐被微生物还原得到的,所以普遍认为微生物在烟叶中TSNAs的形成中起到了非常重要的调节作用。近年来烟草行业利用微生物在降低晾晒烟和烤烟中TSNAs前体物方面做了一些探索性的研究和应用,但关于雪茄烟降TSNAs技术的研究几乎处于空白。国产雪茄烟均采用高温高湿发酵工艺对雪茄烟叶原料进行堆积发酵,为微生物菌制剂的施用提供了可能性,然而,雪茄特有的堆积发酵过程中会伴有剧烈的升温,目前有关研究报道中所筛选的基于烟叶调制和卷烟加工环节应用的降TSNAs的菌株可能存在不适用性,因此,在雪茄发酵过程中亟需筛选高温耐受性强的菌株,以期发挥最大的功效。
发明内容
为实现解决上述问题,本发明的第一个目的是提供一种弯曲芽孢杆菌(Bacillusflexus)Ni3,所述菌株为弯曲芽孢杆菌(Bacillus flexus)Ni3,可以高效降解烟草中的烟碱和TSNAs。
本发明的第二个目的是提供所述的弯曲芽孢杆菌(Bacillus flexus)在降解烟草特有亚硝胺方面的应用。
本发明的第三个目的是提供一种微生物菌制剂。
本发明的第四个目的是提供微生物菌制剂在降解烟草中烟草特有亚硝胺方面的应用。
本发明的第五个目的是提供弯曲芽孢杆菌(Bacillus flexus)在降解烟碱方面的应用。
为了实现上述目的,本发明所采用的技术方案是:
一种弯曲芽孢杆菌(Bacillus flexus)Ni3,所述菌株为弯曲芽孢杆菌(Bacillusflexus)Ni3,保藏于中国典型培养物保藏中心,保藏编号为CCTCC NO:M 20221334。
弯曲芽孢杆菌能分泌多种酶,如:果胶酶、蛋白酶等,是微生物环境改良的优选菌剂。本发明从雪茄烟中分离筛选到一株耐高温的弯曲芽孢杆菌(Bacillus flexus)Ni3,其既可以高效降解烟碱,又可以降解烟草特有亚硝胺,为烟碱和TSNAs的降解提供一种新的微生物资源。
上述弯曲芽孢杆菌(Bacillus flexus)在降解烟草特有亚硝胺方面的应用。
该弯曲芽孢杆菌筛选分离自雪茄烟叶表面,比一般的外源细菌更容易在雪茄烟叶上生长,可以高效降解烟草特有亚硝胺的前体物质——烟碱,也可以高效降解烟草特有亚硝胺,在提高烟草安全性、保护烟草消费者健康方面有重要意义。

图7是本发明实施例2中弯曲芽孢杆菌(Bacillus flexus)Ni3对雪茄烟叶CX-84的TSNAs各组分的降解率。
具体实施方式
下面结合具体实施方式对本发明作进一步描述,但本发明的保护范围并不仅限于此;若无特殊说明,实施例中所用的各类试剂、仪器等均为市售商品。
下述实施例及试验例中涉及到的部分生物材料、实验试剂、实验设备等情况简要介绍如下:
培养基:
烟碱富集培养基:K2HPO4 3H2O 13.3g/L,KH2PO4 4g/L,MgSO4 7H2O 0.2g/L,0.5mL/L微量元素溶液。培养基灭菌后,烟碱用0.22μm滤膜过滤除菌后按1.5g/L的含量加入。
微量元素溶液:CaCl2·2H2O 0.05g/L,CuCl2·2H2O 0.05g/L,MnSO4·H2O 0.008g/L,FeSO4·7H2O 0.004g/L,ZnSO4·7H2O 0.1g/L,Na2MoO4·2H2O 0.1g/L,用少量0.1mol/LHCl溶解。
烟碱分离培养基:富集培养基中加入15.0g/L琼脂粉。
LB培养基:氯化钠10g/L,蛋白胨10g/L,酵母粉5g/L。
NNN培养基:氯化钠10g/L,蛋白胨10g/L,酵母粉5g/L,pH7.2~7.4,121℃灭菌30min,培养基灭菌后,NNN用0.22μm滤膜过滤除菌后按10mg/L的最终含量加入。
NNK培养基:氯化钠10g/L,蛋白胨10g/L,酵母粉5g/L,pH7.2~7.4,121℃灭菌30min,培养基灭菌后,NNK用0.22μm滤膜过滤除菌后按10mg/L的最终含量加入。
NAT培养基:氯化钠10g/L,蛋白胨10g/L,酵母粉5g/L,pH7.2~7.4,121℃灭菌30min,培养基灭菌后,NAT用0.22μm滤膜过滤除菌后按10mg/L的最终含量加入。
NAB培养基:氯化钠10g/L,蛋白胨10g/L,酵母粉5g/L,pH7.2~7.4,121℃灭菌30min,培养基灭菌后,NAB用0.22μm滤膜过滤除菌后按10mg/L的最终含量加入。
实施例1一种弯曲芽孢杆菌(Bacillus flexus)
本实施例的弯曲芽孢杆菌(Bacillus flexus)分离于雪茄烟表面,以烟碱为唯一碳源和能源进行初筛和复筛,再使用TSNAs进行降解效果评价,分离一株能高效降解烟碱和TSNAs的细菌Ni3,通过形态学、MALDI-TOF MS和分子生物学鉴定,鉴定为弯曲芽孢杆菌(Bacillus flexus)。所述弯曲芽孢杆菌(Bacillus flexus)Ni3,保藏编号为CCTCC NO:M20221334;保藏日期:2022年8月26日;保藏单位:中国典型培养物保藏中心,保藏地址:湖北省武汉市武昌区八一路299号武汉大学。具体分离、筛选、评价和鉴定的具体实施操作如下:
1、降解烟碱的菌株分离、筛选
1.1、样品筛选
根据国内外主要品牌和规格的手工雪茄烟产品和雪茄烟叶生产情况,考虑不同国别、不同烟支与烟叶样品质量、不同品种与不同年份烟叶的差异,选择和搜集代表性手工雪茄烟和雪茄烟叶样品,并对国内代表性产地待初次发酵烟叶样品进行采集,冷藏备用;所筛样品名称、类型及来源如下表1。

1.2、初筛
将国内外不同品种、地区的雪茄烟叶剪碎后加入无菌水,在室温下振荡提取3h,得到提取悬浊液,离心取上清液60℃水浴加热30min,定向保留耐热菌株。取加热后上清液加入以烟碱为唯一碳氮源的富集培养基中,富集培养2d。取离心后菌悬液用稀释平板的方法分离能在烟碱分离培养基上生长的菌株。将筛选出的菌株用LB培养基平板进行划线纯化,获得长势旺盛的单菌落。
1.3、复筛
挑取分离保存的菌株接种于液体LB中,37℃培养12h做种子液。将种子液以3%(V/V)的接种量接种至初始浓度为1.5g/L的烟碱发酵培养基中,37℃培养24h,以不接种菌的作为对照。菌液离心取上清液,在72h时使用紫外分光光度法检测上清液中烟碱含量,其原理是高锰酸钾与烟碱能在NaOH溶液中反应形成水溶性绿色产物,绿色产物在610nm处有最大吸收值。测试完成计算烟碱降解率:
烟碱降解率=(原培养液中烟碱含量-发酵液中烟碱含量)/原培养液中烟碱含量×100%
将初筛得到的可以利用烟碱生长的菌株发酵,进一步复筛获得一株可以高效降解烟碱的菌株Ni3,复筛得到高效降解烟碱菌株的烟碱降解率如表2。X5同批次筛选出来的菌株。

2、降解烟草特有亚硝胺(TSNAs)菌株的评价
取上述降解烟碱能力强的菌株Ni3接种至LB培养基上,37℃培养12h得到发酵种子,然后将发酵种子接种至液体LB培养基中,37℃培养12h作为种子液,将种子液以3%(V/V)的接种量分别接种至5mL NNN培养基(NNN 10mg/L)、5mL NNK培养基(NNK10 mg/L)、5mLNAT培养基(NAT 10mg/L)、5mL NAB培养基(NAB 10mg/L)中进行培养,37℃培养72h,以不接种菌的作为对照。发酵后菌液离心取上清液,测试NNN、NNK、NAT、NAB的含量,计算NNN、NNK、NAT、NAB的降解率,比较弯曲芽孢杆菌Ni3对TSNAs各组分的降解能力。
TSNAs的检测方法:HPLC条件:流动相A:水溶液,流动相B:甲醇溶液,流速为0.8mL/min,柱温40℃,进样量为10μL,检测波长235nm,色谱柱Poroshell EC-C18(4.6*250mm4um)。下表3为梯度洗脱的条件。

通过HPLC检测分析,并计算出对照样品和发酵后样品中NNN、NNK、NAT、NAB含量,计算出NNN、NNK、NAT、NAB降解率如图1。从图中可以看出Ni3号菌对TSNAs的关键组分10mg/L的NNN的降解率为23.31%;对TSNAs的关键组分10mg/L的NNK的降解率为64.10%;对TSNAs的关键组分10mg/L的NAT的降解率为26.42%;对TSNAs的关键组分10mg/L的NNK的降解率为27.42%,与对照菌株相比,具有较明显优势。
3、弯曲芽孢杆菌Ni3的鉴定
3.1、形态学鉴定
将上述筛选获得的Ni3接种到LB固体培养基平板上,培养后观察菌落形态。结果如图2、图3所示,弯曲芽孢杆菌Ni3菌体呈现棒状,培养基上菌落形状不规则,淡黄色,边缘呈现透明锯齿状。
3.2、MALDI-TOF MS鉴定
制备弯曲芽孢杆菌Ni3的单菌落,使用“提取法”获得菌株悬液,吸取1μL样本至靶板点位,晾干后滴加1μL基质溶液,使用全自动微生物质谱检测系统(Autof ms2000,安图生物科技有限公司公司)进行菌株鉴定。Ni3经Autof ms2000鉴定为弯曲芽孢杆菌,质谱指纹图谱及匹配棒状图如图4所示。
3.3、分子生物学鉴定
将弯曲芽孢杆菌Ni3菌株接种于液体LB培养基震荡培养12小时,离心收集菌体,用16S rDNA通用引物正向引物27F(5’-AGAGTTTGATCCTGGCTCAG-3’)和反向引物1492R(5’-GGTTACCTTGTTACGACTT-3’),对其16S rDNA基因进行PCR扩增,PCR条件为:95℃,5min;95℃,30s,55℃,30s,72℃,90s,30循环;95℃,40s,55℃,45s,72℃,1.5min,30循环;72℃,5min,10℃,5min。PCR产物长度为1458bp,其核苷酸序列表如序列表SEQ ID NO:1所示。
菌株Ni3的16S rDNA基因片段经PCR扩增、纯化回收,由北京擎科生物科技有限公司完成DNA测序工作。将测得的序列用NCBI的Blastn程序进行序列相似性分析,从Genbank数据库和核糖体数据库选取相近种属的代表性菌株的16S rDNA基因序列,通过MEGA7.0软件进行系统发育分析,用邻接法(Neighbor Joining)构建基于16S rDNA基因序列的系统发育树,用于检验自举支持率(Bootstrap)的重复抽样次数为1000次。测序所得Ni3的16SrDNA基因序列有1458个碱基,同源性分析结果显示菌株Ni3的16S rDNA基因序列与弯曲芽孢杆菌(Bacillus flexus)高度相似,且通过MEGA 7.0软件构建系统发育树,系统进化树如图5所示,显示菌株Ni3属于弯曲芽孢杆菌。结合形态学特点和16S rDNA基因序列分析,该菌株被鉴定为弯曲芽孢杆菌(Bacillus flexus),命名为弯曲芽孢杆菌Ni3,已于2022年8月26日保藏于中国典型培养物保藏中心(CCTCC),保藏编号为CCTCC NO:M 20221334。
实施例2弯曲芽孢杆菌Ni3在降解烟草特有亚硝胺方面的应用
本实施例的弯曲芽孢杆菌Ni3在降解烟草中烟碱和烟草特有亚硝胺方面的应用,将所述降解菌弯曲芽孢杆菌Ni3的菌液喷洒在雪茄烟叶的表面,经过一段时间发酵后,经检测相比于对照组,喷洒弯曲芽孢杆菌Ni3的雪茄烟叶中TSNAs显著降低。具体实施操作如下:
弯曲芽孢杆菌Ni3接种至LB平板上活化培养12h,将活化后的弯曲芽孢杆菌Ni3接种到液体LB培养基中在37℃,180rpm的摇床中发酵12h制备发酵液。将发酵液在12000rpm,离心5min,除去上清液,收集菌体。将收集的菌体进行重悬,使用分光光度计测定菌体浓度,将菌体浓度稀释为108cfu/mL,即为弯曲芽孢杆菌Ni3的发酵菌剂。称取一定量的两种高TSNAs含量的雪茄烟叶,使用无菌剪刀剪碎至三角瓶中作为雪茄烟叶发酵培养基,以保持雪茄烟叶共含水量为60%的量,施加一定量的菌剂,均匀喷洒在烟叶上,在37℃、RH75%发酵72h。
发酵结束后,取雪茄烟叶,60℃烘干3h后,磨碎,过60目筛,取样,通过YQ/T 29—2013《烟草及烟草制品烟草特有N-亚硝胺的测定高效液相色谱-串联质谱联用法》检测发酵后雪茄烟叶中NNN、NNK、NAT、NAB含量,计算得出各成份降解率,分析弯曲芽孢杆菌Ni3对不同种类的高TSNAs含量的雪茄烟叶TSNAs的降解能力。
通过高效液相色谱-串联质谱联用法的检测分析,弯曲芽孢杆菌Ni3对不同种类的的雪茄烟叶中TSNAs各组分的作用效果如下表4。

注:CX-81为楚雪81号雪茄烟品种;CX-84为楚雪84号雪茄烟品种。CK代表对照组。
由上表可知,弯曲芽孢杆菌Ni3可使发酵后的两种高TSNAs雪茄烟叶CX-81、CX-84中TSNAs明显降低,总TSNAs含量降解率分别达到了52%和35%,对两种雪茄烟叶的TSNAs各成分降解率如图6、图7所示。
实施例3一种微生物菌制剂
本实施例的微生物菌制剂,包括微生物和辅料,所述微生物为弯曲芽孢杆菌(Bacillus flexus)Ni3,保藏编号为CCTCC NO:M 20221334。辅料可以是本领域中常用的辅料。
综上所述,本发明所述的菌株Ni3既可高效降低烟碱的含量,又对TSNAs的各组份NNN、NNK、NAT、NAB有较高的降解能力,并且在雪茄烟的实际应用中也表现出了较好的降解性能。该菌在雪茄烟叶及其产品的降害方面,具有极大的应用潜力,可为后续菌剂发酵工艺优化及雪茄发酵改良奠定基础。

1.一种弯曲芽孢杆菌(Bacillus flexus)Ni3,其特征在于:所述菌株为弯曲芽孢杆菌(Bacillus flexus)Ni3,保藏于中国典型培养物保藏中心,保藏编号为CCTCC NO:M20221334。
2.一种如权利要求1所述的弯曲芽孢杆菌(Bacillus flexus)在降解烟草特有亚硝胺方面的应用。
3.根据权利要求2所述的弯曲芽孢杆菌(Bacillus flexus)在降解烟草特有亚硝胺方面的应用,其特征在于:所述的烟草为雪茄烟叶。
4.根据权利要求2所述的弯曲芽孢杆菌(Bacillus flexus)在降解烟草特有亚硝胺方面的应用,其特征在于:所述的烟草特有亚硝胺为N-亚硝基去甲基烟碱、4-(甲基亚硝胺)-1-(3-吡啶基)-1-丁酮、N-亚硝基假木贼碱、N-亚硝基新烟碱。
5.一种微生物菌制剂,其特征在于:包含如权利要求1所述的弯曲芽孢杆菌(Bacillusflexus)Ni3。
6.一种如权利要求5所述的微生物菌制剂在降解烟草中烟草特有亚硝胺方面的应用。
7.一种如权利要求1所述的弯曲芽孢杆菌(Bacillus flexus)在降解烟碱方面的应用。
