Abstract

技术领域
本申请涉及一种新型的裂殖壶菌属(Schizochytrium sp.)菌株以及使用其的多不饱和脂肪酸生产方法。
背景技术
不饱和脂肪酸(unsaturated fatty acid)是在脂肪酸链中具有一个或多个双键的脂肪酸,并在包括两个或多个双键时被称为多不饱和脂肪酸(polyunsaturated fattyacid:PUFA)。其中,二十二碳六烯酸(docosahexaenoic acid:DHA)和二十碳五烯酸(eicosapentaenoic acid:EPA)是具有代表性的omega-3脂肪酸,其为大脑、眼球组织和神经系统的必需脂肪酸。此外,已知其在诸如婴儿的视力和运动神经能力等的神经系统的发育和心血管疾病的预防中发挥重要作用,并且是大脑结构脂质中最丰富的构成要素。
包括人类在内的大多数高等生物无法自行合成多不饱和脂肪酸,因此必须将其作为必需营养素摄入,多不饱和脂肪酸主要来源于金枪鱼、鲑鱼等在海洋生态环境中处于上位的深海鱼类。目前被公开的多不饱和脂肪酸的主要工业来源是从鲭鱼、秋刀鱼、金枪鱼、竹筴鱼、沙丁鱼和青鱼等青色的鱼的油中提取的鱼油,其对诸如咸水鱼类初始饲料的养鱼饲料也非常有用。然而,鱼油的品质因鱼种、季节、捕捞地点不同而异,导致难以持续供应鱼油,并且由于鱼油受重金属和有机化学物质污染的问题、鱼油特有的腥味、以及加工工艺中双键氧化导致生产量受限等问题,对多不饱和脂肪酸的替代来源的需要求正在出现。
为了解决这个问题,目前正在研究通过微生物培养的包括二十二碳六烯酸的多不饱和脂肪酸的制造方法。特别地,微细藻类可以生产脂质并且易于培养,因此能够制造具有相对一定的生化学组成的生物量。另外,由微细藻类制造的脂质不像鱼油那样具有令人不快的气味,并且具有比鱼油更简单的脂肪酸组成,因此可容易地分离特定脂肪酸。因此,能够以高含量稳定供应特定脂肪酸的新型微细藻类具有非常重要的工业价值,因此,有必要开发这样的微细藻类。
发明内容

本申请的目的在于提供一种裂殖壶菌属(Schizochytrium sp.)微细藻类,其以保藏号KCTC14344BP或KCTC14345BP保藏;以及具有二十二碳六烯酸(docosahexaenoic acid:DHA)、二十碳五烯酸(eicosapentaenoic acid:EPA)、以及棕榈酸(palmitic acid:PA)的生产能力。
本申请的另一目的在于提供一种源自所述裂殖壶菌属微细藻类的生物量以及包括其的饲料组合物。

一方面提供一种裂殖壶菌属微细藻类,其以保藏号KCTC14344BP或KCTC14345BP保藏;以及具有二十二碳六烯酸、二十碳五烯酸、以及棕榈酸的生产能力。
在本说明书中使用的术语“破囊壶菌(Thraustochytrid)”是指破囊壶菌(Thraustochytriales)目的微细藻类。此外,在本说明书中使用的术语“裂殖壶菌属(Schizochytrium sp.)”为属于破囊壶菌目的破囊壶菌(Thraustochytriaceae)科的属名之一,其与术语“裂殖壶菌属(genus Schizochytrium)”可以互换使用。另外,所述术语“微细藻类(microalgae)”是指以叶绿素进行光合作用的植物中以肉眼看不见并只能通过显微镜才能看到的在水中自由漂浮地生活的生物,其也称为浮游植物(Phytoplankton)。
在本说明书中使用的术语“二十二碳六烯酸(docosahexaenoic acid:DHA)”是具有C22H32O2的化学式的多不饱和脂肪酸中的一种,其与α-亚麻酸(α-linolenic acid:ALA)和二十碳五烯酸(eicosapentaenoic acid:EPA)一起对应于omega-3脂肪酸,通用名为鱼油酸(cervonic acid),也可缩写为22:6n-3。
在本说明书中使用的术语“二十碳五烯酸(eicosapentaenoic acid:EPA)”是具有C20H30O2的化学式的多不饱和脂肪酸中的一种,其与ALA和DHA一起对应于omega-3脂肪酸,也可缩写为20:5n-3。
在本说明书中使用的术语“棕榈酸(palmitic acid:PA)”是指具有C16H32O2的化学式的饱和脂肪酸中的一种。
所述裂殖壶菌属微细藻类是以保藏号KCTC14344BP保藏的新型的野生型裂殖壶菌属微细藻类CD01-5000或其突变株,也可以是以保藏号KCTC14345BP保藏的裂殖壶菌属微细藻类CD01-5004。
所述裂殖壶菌属微细藻类可具有SEQ ID NO:1的18S rRNA碱基序列,但不限于此。例如,所述裂殖壶菌属微细藻类可具有由与SEQ ID NO:1的碱基序列显示80%或更高、85%或更高、90%或更高、95%或更高、98%或更高、或99%或更高的序列同一性的碱基序列组成的18S rRNA,但不限于此。

基于脂肪酸的总重量,所述裂殖壶菌属的微细藻类可生产0.5重量%至10重量%的EPA。例如,基于脂肪酸的总重量,所述裂殖壶菌属的微细藻类可生产0.5重量%至8重量%、0.5重量%至5重量%、0.5重量%至3重量%、0.8重量%至10重量%、0.8重量%至8重量%、0.8重量%至5重量%、0.8重量%至3重量%、1重量%至10重量%、1重量%至8重量%、1重量%至5重量%、或1重量%至3重量%的EPA。另外,基于脂肪酸的总重量,所述裂殖壶菌属的微细藻类可包括0.5重量%至10重量%的EPA。

所述裂殖壶菌属的微细藻类可具有类胡萝卜素的生产能力。例如,所述裂殖壶菌属的微细藻类可生产选自由β-胡萝卜素(β-carotene)、叶黄素(lutein)、虾青素(astaxanthin)、辣椒红(capsanthin)、胭脂树橙色素(annatto)、斑蝥黄质(canthaxanthin)、番茄红素(lycopene)、β-阿扑-8-胡萝卜醛(β-apo-8-carotenal)、玉米黄素(zeaxanthin)、β-阿扑-8-胡萝卜醛-酯(β-apo-8-carotenal-ester)组成的组中的任一个或多个。

在本说明书中使用的术语“生物量(biomass)”是指可以用作化学能的植物、动物、微生物等生物体,即生物能源的能源,其在生态学上也指存在于单位时间和空间内的特定生物体的重量或能量。此外,所述生物量包括但不限于细胞分泌的化合物,并且可以不仅含有细胞外物质而且可以含有细胞和/或细胞内的内容物。在本申请中,所述生物量可以为通过培养或发酵裂殖壶菌属微细藻类、其培养产物、其干燥产物、其粉碎产物、或所述微细藻类而生产的产物,或者可以是所述生物量的浓缩产物或干燥产物,但不限于此。
所述裂殖壶菌属微细藻类的“培养产物”是指通过培养所述微细藻类而生产的产物,具体可以是包括微细藻类的培养液或从所述培养液中去除微细藻类的培养滤液,但不限于此。所述裂殖壶菌属微细藻类培养产物的“干燥产物”是从所述微细藻类培养产物中去除水分后获得的,例如可以是所述微细藻类的干燥菌体形式,但不限于此。另外,所述干燥产物的“粉碎产物”是对从所述微细藻类培养产物中去除水分而获得的干燥产物进行粉碎而获得的结果的总称,例如可以是干燥菌体粉末,但不限于此。所述裂殖壶菌属微细藻类的培养产物可以通过将所述微细藻类接种在微细藻类培养基中,并按照本领域已知的培养方法制造,且所述培养产物的干燥产物和其粉碎产物也可以按照本领域已知的微细藻类或培养液的处理或干燥方法来制造。
基于脂肪酸的总重量,源自所述裂殖壶菌属微细藻类的生物量可包括35重量%至60重量%的DHA,并基于脂肪酸的总重量,可包括0.5重量%至10重量%的EPA,且基于脂肪酸的总重量,可包括10重量%至30重量%的PA。
另外,所述源自裂殖壶菌属微细藻类的生物量包括类胡萝卜素,例如可包括选自由β-胡萝卜素(β-carotene)、叶黄素(lutein)、虾青素(astaxanthin)、辣椒红(capsanthin)、胭脂树橙色素(annatto)、斑蝥黄质(canthaxanthin)、番茄红素(lycopene)、β-阿扑-8-胡萝卜醛(β-apo-8-carotenal)、玉米黄素(zeaxanthin)、β-阿扑-8-胡萝卜醛-酯(β-apo-8-carotenal-ester)组成的组中的任一个或多个。

在本说明书中使用的术语“生物油(bio-oil)”是指通过生物学、热化学和物理化学提取工艺从生物量中获得的油,并在本申请中制造的生物油可含有多不饱和脂肪酸,具体可含有DHA和EPA,但不限于此。
所述组合物可以是溶液、粉末或悬浮液的形式,但不限于此。所述组合物可以是例如食品组合物、饲料组合物、或饲料添加剂组合物。
在本说明书中使用的术语“饲料组合物”是指供给动物的饲料。所述饲料组合物是指提供维持动物生命或生产肉、奶等所必需的有机或无机营养素的物质。所述饲料组合物可额外包括维持动物生命或生产肉、奶等所必需的营养成分。所述饲料组合物可制造成本领域已知的各种形式的饲料,并且具体地,可以包括精饲料、粗饲料和/或特殊饲料。
在本说明书中使用的术语“饲料添加剂”包括为各种效果如补充营养素和预防体重下降、提高饲料中纤维素的消化易溶性、改善油质、预防繁殖障碍和提高受胎率、预防夏季高温应激等而添加到饲料中的物质。本申请的饲料添加剂对应于饲料管理法中的补充饲料,还可以包括碳酸氢钠、膨润土(bentonite)、氧化镁、复合矿物质等的矿物质制剂、锌、铜、钴、硒等微量矿物的矿物制剂、胡萝卜素、维生素E、维生素A、D、E、烟酸、维生素B复合体的维生素剂、蛋氨酸和裂解酸等的受保护氨基酸、脂肪酸钙盐等的受保护脂肪酸剂、活菌剂(乳酸菌剂)、酵母培养产物、真菌发酵产物等的活菌、酵母剂等。
在本说明书中使用的术语“食品组合物”包括功能性食品(functional food)、营养补充剂(nutritional supplement)、保健食品(health food)和食品添加剂(foodadditives)等的所有形式,且所述类型的食物组合物可按照本领域已知的常规方法制造成各种形式。
本申请的组合物可进一步包括谷物,例如磨碎或粉碎的小麦、燕麦、大麦、玉米和大米;植物蛋白饲料,例如以大豆和向日葵作为主要成分的饲料;动物蛋白饲料,例如血粉、肉粉、骨粉、鱼粉;糖和乳制品,例如由各种奶粉和乳清粉组成的干燥成分等,此外,还可以包括营养补充剂、消化吸收促进剂、生长促进剂等。
本申请的组合物可以单独施用或与可食用载体中的其他饲料添加剂组合施用于动物。此外,所述组合物可以作为施顶肥或直接与饲料混合,或作为与饲料分开的口服剂型而容易地施用于动物。当所述组合物与饲料分开施用时,其可以通过与本领域公知的药学上可接受的可食用载体组合而制造为即时放出或缓释性剂型。这样的可食用载体可以是固体或液体,例如玉米淀粉、乳糖、蔗糖、大豆片、花生油、橄榄油、芝麻油和丙二醇。当使用固体载体时,所述组合物可以是片剂、胶囊剂、散剂、锭剂或含糖片剂,或微分散形式的施顶肥。当使用液体载体时,所述组合物可以是明胶软质胶囊剂、糖浆剂、悬浮液、乳液剂、或溶液剂的剂型。
本申请的组合物可以含有例如防腐剂、稳定化剂、润湿剂或乳化剂、冻结保护剂、或赋形剂等。所述冻结保护剂可以为选自由甘油、海藻糖、麦芽糖糊精、脱脂奶粉和淀粉组成的组中的一个或多个。
所述防腐剂、稳定化剂或赋形剂可以以足以减少包括在所述组合物中的裂殖壶菌属微细藻类的下降(deterioration)的有效量包括在组合物中。另外,当所述组合物处于干燥状态时,所述冻结保护剂可以以足以减少包括在所述组合物中的裂殖壶菌属微细藻类的下降(deterioration)的有效量包括在组合物中。
所述组合物可以通过浇注、喷雾或混合来添加到动物饲料中来使用。
本申请的组合物可应用于包括哺乳类、鸟类、鱼类、甲壳类、头足类、爬虫类、和两栖类的多种动物的饮食,但不限于此。例如,所述哺乳类可包括猪、牛、绵羊、山羊、试验用的啮齿动物、或宠物等,所述鸟类可包括家禽类,所述家禽类可包括鸡、火鸡、鸭、鹅、野鸡或鹌鹑等,但不限于此。此外,所述鱼类可包括商业畜养的鱼类和其鱼苗类、观赏鱼等,且所述甲壳类可包括虾、藤壶等,但不限于此。另外,所述组合物也可以应用于动物性浮游生物的轮虫(rotifer)的饮食。
另一方面提供一种源自裂殖壶菌属微细藻类的生物量的制造方法,其包括培养裂殖壶菌属微细藻类,其以保藏号KCTC14344BP或KCTC14345BP保藏并具有二十二碳六烯酸、二十碳五烯酸、和棕榈酸的生产能力;以及从所述微细藻类、所述微细藻类的培养产物、所述培养产物的干燥产物、或所述干燥产物的粉碎产物中回收生物量。
所述裂殖壶菌属微细藻类、生物量、所述微细藻类的培养产物、所述培养产物的干燥产物、以及所述干燥产物的粉碎产物如上所述。
在本说明书中使用的术语“培养”是指在适当调节的环境条件下生育所述微细藻类。本申请的培养过程可以根据本领域已知的合适的培养基和培养条件进行。本领域技术人员可以根据选择的微细藻类容易地调整和使用这种培养过程。
具体地,本申请裂殖壶菌属微细藻类的培养可以在从属营养条件下进行,但不限于此。
在本说明书中使用的术语“从属营养”是依靠从体外获得的有机物作为能源或营养源的营养方法,其是与独立营养相对应的术语,并与术语“暗培养”互换使用。
所述培养裂殖壶菌属微细藻类不特别限于此,而是可以通过已知的分批培养法、连续培养法、补料分批培养法等进行。用于本申请的微细藻类的培养的培养基和其他培养条件没有特别限制,只要是用于培养常规微细藻类的培养基即可。具体地,本申请的微细藻类可以在有氧条件下在含有适当碳源、氮源、磷、无机化合物、氨基酸和/或维生素等的常规培养基中在调节温度、pH等的同时进行培养。
具体地,可以通过使用碱性化合物(例如:氢氧化钠、氢氧化钾或氨)或酸性化合物(例如:磷酸或硫酸)来调节合适的pH(例如,pH 5至9,具体是pH 6至8,最具体是pH 6.8),但不限于此。
另外,为了维持培养产物的好氧状态,可以向培养产物中注入氧气或含氧气体,或为了维持厌氧和微氧状态,可以不注入气体,或者可以注入氮气、氢气或二氧化碳气体,但不限于此。
此外,培养温度可维持在20℃至45℃,或25℃至40℃,并可培养约10小时至160小时,但不限于此。另外,在培养过程中,可通过使用诸如脂肪酸聚乙二醇酯的消泡剂来抑制起泡生成,但不限于此。
在所述培养裂殖壶菌属微细藻类中使用的培养基中包括的碳源可以为选自由葡萄糖、果糖、麦芽糖、半乳糖、甘露糖、蔗糖、阿拉伯糖、木糖和甘油组成的组中的任一个或多个,但只要是用于培养微细藻类的碳源即可,而不限于此。
在所述培养裂殖壶菌属微细藻类中使用的培养基中包括的氮源可以为i)选自由酵母提取物(yeast extract)、牛肉提取物(beef extract)、蛋白胨、和胰蛋白胨组成的组中的任一个或多个的有机氮源,或者为ii)选自由乙酸铵、硝酸铵、氯化铵、硫酸铵、硝酸钠、尿素、和谷氨酸钠(Monosodium glutamate:MSG)组成的组中的任一个或多个的无机氮源,但只要是用于培养微细藻类的氮源即可,而不限于此。
在所述培养裂殖壶菌属微细藻类中使用的培养基中,可单独包括或混合且包括磷酸二氢钾、磷酸氢二钾、及其对应的含钠盐等作为磷源,但不限于此。
从所述微细藻类、所述微细藻类的培养产物、所述培养产物的干燥产物、或所述干燥产物的粉碎产物回收生物量可通过使用本领域已知的合适方法收集所需的生物量。例如,可使用离心、过滤、阴离子交换层析、结晶化、高效液相色谱(High Performance LiquidChromatography:HPLC)等,并可进一步包括纯化工艺。
另一方面提供一种源自裂殖壶菌属微细藻类的生物油的制造方法,其包括培养裂殖壶菌属微细藻类,其以保藏号KCTC14344BP或KCTC14345BP保藏并具有二十二碳六烯酸、二十碳五烯酸、和棕榈酸的生产能力;以及从所述微细藻类、所述微细藻类的培养产物、所述培养产物的干燥产物、或所述干燥产物的粉碎产物中回收脂质。
所述裂殖壶菌属微细藻类、生物油、所述微细藻类的培养产物、所述培养产物的干燥产物、和所述干燥产物的粉碎产物、所述培养微细藻类如上所述。
从所述微细藻类、所述微细藻类的培养产物、所述培养产物的干燥产物、或所述干燥产物的粉碎产物回收脂质可通过使用本领域已知的合适方法收集所需的脂质。例如,可使用离心、过滤、阴离子交换层析、结晶化、HPLC等,并可进一步包括纯化工艺。
例如,诸如脂肪醛、脂肪醇和烃类(例如,烷烃)的脂质和脂质衍生物可以用诸如己烷的疏水性溶剂提取。也可以通过使用液化、油液化和超临界CO2提取等方法提取脂质和脂质衍生物。另外,已知的微细藻类脂质回收方法包括,例如,i)通过离心收集细胞,用蒸馏水洗涤,然后通过冻结干燥进行干燥、ii)粉碎所获得的细胞粉末,然后用正己烷提取脂质的方法(Miao,X和Wu,Q,Biosource Technology(2006)97:841-846)。
有益效果
根据一方面的新型裂殖壶菌属(Schizochytrium sp.)微细藻类在生物量中脂肪含量高,其中,诸如二十二碳六烯酸(docosahexaenoic acid:DHA)和二十碳五烯酸(eicosapentaenoic acid:EPA)的多不饱和脂肪酸的含量高,因此,所述微细藻类、从其制造的生物量或生物油可有效地用作饲料原料等。

具体实施方式
在下文中,将通过实施例更详细地描述本发明。然而,这些实施例是为了举例说明一个或多个具体例,本发明的范围不限于这些实施例。
实施例1.破囊壶菌(Thraustochytrid)系列微细藻类的分离
为了分离破囊壶菌系列的微细藻类,从韩国忠清南道泰安和全罗北道群山地区海岸的总共50几个地区中采集了海水、土壤和沉积物形式的环境样品。所采集的环境样品运至试验室环境,并在进行7天内去除真菌和其他细菌类、微生物、原生生物等污染物的操作的同时,通过使用直接涂抹法(Direct plating)和松花粉诱饵法(Pine pollen baiting)来分离破囊壶菌系列微细藻类。在连续去除污染物和分离微细藻类的过程中,通过显微镜,形成作为破囊壶菌系列微细藻类的特征的游动孢子(Zoospore),并分离出与包括其的细胞具有相似形态的样品,从而涂抹在作为分离海洋微细藻类的培养基的改良的YEP培养基(酵母提取物(Yeast extract)1g/L、蛋白胨1g/L、MgSO4·7H2O 2g/L、海盐(Sea salt)20g/L、H3BO3 5.0mg/L、MnCl2 3.0mg/L、CuSO4 0.2mg/L、NaMo4·2H2O 0.05mg/L、CoSO4 0.05mg/L、ZnSO4·7H2O 0.7mg/L、以及琼脂(Agar)15g/L)。所获得的菌落通过传代培养数次进行纯化。将具有持续污染的菌落暴露于抗生素混合物混合溶液(硫酸链霉素0mg/L至100mg/L、氨苄青霉素0mg/L至100mg/L、苄青霉素0mg/L至100mg/L、以及硫酸卡那霉素0mg/L至100mg/L),从而控制和去除污染源。通过所述过程分离并获得50几种菌落。
纯分离的菌落通过使用改良的GYEP培养基(葡萄糖10g/L、酵母提取物1g/L、蛋白胨1g/L、MgSO4·7H2O 2g/L、海盐20g/L、H3BO3 5.0mg/L、MnCl2 3.0mg/L、CuSO4 0.2mg/L、NaMo4·2H2O 0.05mg/L、CoSO4 0.05mg/L、以及ZnSO4·7H2O 0.7mg/L)在250ml的烧瓶中在10℃至35℃且100rpm至200rpm的条件下培养约7天。其中,最终选定了一种能够在25℃或更高的温度条件下生长、生长速度优异、并且能够确保菌体量的微细藻类,并将其命名为CD01-5000。通过使用光学显微镜观察所选定的菌株的形式(图1)。
实施例2.新型的裂殖壶菌属菌株CD01-5000的鉴定
为了对在所述实施例1中分离且选定的微细藻类菌株CD01-5000进行分子生物学鉴定,分析了18S rRNA基因序列。
具体地,从纯分离的微细藻类CD01-5000的菌落中提取和分离gDNA后,通过使用下表1的引物作为用于18S rRNA部位的基因增幅用的引物来进行聚合酶链式反应(Polymerase Chain Reaction:PCR)反应。

PCR反应通过使用含有taq聚合酶(taq polymerase)的反应溶液来重覆进行以下循环38次:在95℃下变性5分钟,然后在95℃下变性30秒,在55℃下退火30秒,在72℃下聚合1分30秒。然后在72℃下进行聚合反应5分钟。将通过PCR过程增幅的反应液在1%琼脂糖凝胶上进行电泳,从而确认增幅出大小为约1600bp至2000bp的DNA片段,并进行碱基序列测序分析。作为分析的结果,确保了大小为约1800bp的碱基序列(SEQ ID NO:1),该序列通过NCBI BLAST检索显示出具有与属于破囊壶菌系列微细藻类的裂壶藻(Schizochytriumlimacinum)菌株(NCBI登录号.:HM042913.2)的18S rRNA基因碱基序列99.3%的同源性,并确认了显示出具有与裂殖壶菌属(Schizochytrium sp.)菌株SH104(NCBI登录号.:KX379459.1)的18S rRNA基因碱基序列99.3%的同源性。由此确认所分离的微细藻类CD01-5000为新型裂殖壶菌属菌株,并将其命名为裂殖壶菌属(Schizochytriumsp.)CD01-5000,并在2020年10月26日保藏于布达佩斯条约下的国际保藏机构的韩国生命工学研究院韩国典型菌种保藏中心(Korean Collection for Type Cultures:KCTC),且获得保藏号KCTC14344BP。
实施例3.突变微细藻类菌株的开发
实施例3-1.根据伽马线照射的死亡率测量
为了从所述实施例2中确认的新型微细藻类CD01-5000开发人工突变菌株,通过测量根据伽马线线量的死亡率,并选择伽马线照射条件。
具体地,新型微细藻类CD01-5000在包括3%的葡萄糖的改良的GYEP培养基中培养20小时或更长,从而达到指数生长期(Exponential phase)。将所培养的细胞培养液样品以4,000rpm离心15分钟以收获菌体,并将悬浮于PBS中以成为109细胞/mL的微细藻类培养液样品置于50mL锥形管中,从而用于伽马线照射试验中。伽马线照射试验在韩国原子能研究院高级放射线研究所进行,并向微细藻类培养液样品照射2000、2500、3000、3500、4000、4500、5000、5500、6000、7000、或8000Gy的伽马线。对经伽马线照射的微细藻类培养液样品进行离心并去除上清液后,接种于包括1%的葡萄糖的GCBS培养基(葡萄糖10g/L、)中,玉米浆(Corn Steep liquor)5g/L、牛肉提取物(Beef extract)5g/L、MgSO4·7H2O 5g/L、海盐15g/L、柠檬酸20.08mg/L、FeSO4·7H2O 5.3mg/L、ZnSO4·7H2O 0.5mg/L、MnCl2 1.0mg/L、CuSO4 0.1mg/L、NaMo4·2H2O 0.1mg/L、CoSO4 0.1mg/L、ZnSO4·7H2O 0.5mg/L、生物素1.0mg/L、盐酸硫胺素(Thiamine hydrochloride)1.0mg/L、CAPA 1.0mg/L、以及维生素B120.1mg/L),并在30℃下培养约48小时。此后,将培养液接种并涂抹于包括2%(20g/L)琼脂(agar)的GYEP培养基上,在30℃下培养48小时后,计数生育菌落数,从而通过下计算式1测量每伽马线量的死亡率。

结果,当以6000Gy或更高的线量照射伽马线时,所有微细藻类死亡导致无法确保菌落,并选择显示出99.99%的死亡率的5500Gy的伽马线线量条件。
实施例3-2.突变微细藻类菌株的分离
以与所述实施例3-1中所记载的方法相同的方法,对新型微细藻类菌株CD01-5000以5000Gy的线量照射伽马线后,将微细藻类培养液样品分别接种并涂抹在GYEP固体培养基和添加了丁醇和异烟肼(isoniazid)的GYEP固体培养基上,并在30℃下进行培养。挑选在每个固体平板上生育约4周的微细藻类菌落,并在相同的培养基和培养条件下进行传代分离。经传代分离的每个菌落被判断为彼此不同的一个突变菌株,并且额外挑选能够持续生长的菌落和菌株。
实施例3-3.挑选优异的突变微细藻类菌株
对在所述实施例3-2中挑选的突变菌株在烧瓶规模中进行培养且评估,并通过粗脂肪和脂肪酸分析挑选出优异的突变菌株。
具体地,为了将在所述实施例2中确认的新型的野生型微细藻类CD01-5000和在实施例3-2中挑选的突变微细藻类在烧瓶规模中进行培养,在500mL烧瓶中,将最终体积(Working volume)定为50mL,并在包括3%(30g/L)葡萄糖的GYEP培养基中,在30℃和180rpm条件下培养24小时,从而获得可以分析的一定量的菌体。此后,通过用50mL锥形管对培养液进行离心,然后去除上清液,并获得菌体,用pH 7.5的磷酸缓冲盐溶液(phosphatebuffered saline:PBS)洗涤3次,然后在60℃的干燥箱中干燥过夜,从而获得每个干燥菌体。
向所获得的每个干燥菌体2g加入8.3M的盐酸溶液,在80℃下将微细藻类菌体的细胞壁水解后,加入30mL的乙醚和20mL的石油醚(Petroleum ether),并混合30秒后,离心过程重覆3次或更多次。回收经分离出的溶剂层,转移至预先称重的圆底烧瓶中,然后通过氮气吹扫去除溶剂,并在干燥器(Desicator)中干燥至恒量。干燥后,通过从烧瓶的重量中减去空烧瓶的重量来测量经干燥的油的重量。另外,将所获得的经干燥的油用甲醇性0.5NNaOH和14%三氟硼烷甲醇(BF3)进行预处理后,通过气相色谱法测量油中所含的棕榈酸(PA)、二十碳五烯酸(EPA)、二十二碳六烯酸(DHA)的含量,其被计算并表示为对于总油即粗脂肪重量的百分比。

结果,如表3所示,新型的裂殖壶菌属菌株CD01-5000显示出非常高的50%或更高的DHA含量,并显示出比其他突变株更高水平的棕榈酸和EPA含量。另外,在试验的突变株中,相比于野生型菌株显示出更高的DHA含量且相比于其他突变株显示出更高水平的棕榈酸和EPA含量的‘突变株3’被挑选为优异的突变株,其命名为裂殖壶菌属CD01-5004,并在2020年10月26日保藏于布达佩斯条约下的国际保藏机构的韩国生命工学研究院韩国典型菌种保藏中心(Korean Collection for Type Cultures:KCTC),并获得保藏号KCTC14345BP。
实施例4.确认野生型裂殖壶菌属菌株CD01-5000和突变裂殖壶菌属菌株CD01-5004的培养特性
确认新型的野生型裂殖壶菌属菌株CD01-5000和其突变裂殖壶菌属菌株CD01-5004的额外培养特性。
实施例4-1.生物量和脂肪酸含量分析
将CD01-5000菌株、CD01-5004菌株、以及作为对照组的裂殖壶菌属ATCC20888菌株在5L发酵罐中培养至总体积达到3L。在包括酵母提取物5g/L和蛋白胨10g/L作为氮源的GYEP培养基中在20℃至35℃、100rpm至500rpm、以及0.5vvm至1.5vvm的条件下进行培养,按照与所述实施例3-3中所记载的相同的方法从培养液中获得干燥的菌体,并分析细胞中粗脂肪和脂肪酸的含量。

结果,如表4所示,确认出,CD01-5000和CD01-5004菌株在显示出与作为对照组所培养的裂殖壶菌属ATCC20888菌株相似的生物量生长性的同时,可以在细胞内以高含量生产和积累EPA和DHA。
实施例4-2.蛋白质和氨基酸含量分析
针对显示出最高的生物量含量和EPA含量的CD01-5004菌株进行额外的培养试验,从而进行细胞内蛋白质和氨基酸含量分析。
具体地,除了使用包括酵母提取物10g/L和氯化铵10g/L作为氮源的GYEP培养基之外,在与所述实施例4-1所记载的相同的培养条件下培养共63小时。培养终结后,以与所述实施例3-3中所记载的相同的方法,从培养液中获得干燥的菌体。
为了蛋白质含量分析,使用元素分析仪对0.5g至1.0g的每个干燥的菌体定量且分析样品中存在的氮含量。将每个样品中存在的氮的重量比率(TN%)乘以6.25,从而计算并表示样品中的粗蛋白含量。
为了分析氨基酸含量,对0.5g至1g的每个干燥的菌体进行酸水解,然后针对其进行液相色谱。各氨基酸的组成比率(%)计算为个别氨基酸相对于样品中总氨基酸浓度的百分比。

结果,如表5所示,确认出,总菌体生产量为60.5g/L,且干燥的菌体内的粗蛋白含量为74.38%。确认出,细胞内氨基酸由23.31%的谷氨酸和20.51%的苯丙氨酸作为主要成分组成,其次是由组氨酸、丙氨酸、缬氨酸、脯氨酸、蛋氨酸、天冬氨酸、甘氨酸、亮氨酸、异亮氨酸、和丝氨酸成分构成。
实施例5.确认野生型裂殖壶菌属菌株CD01-5000和突变裂殖壶菌属菌株CD01-5004的生长速度
测量并比较新型的野生型裂殖壶菌属菌株CD01-5000和其突变裂殖壶菌属菌株CD01-5004的生长速度。
具体地,CD01-5000和CD01-5004菌株分别在500mL烧瓶中接种于包括3%葡萄糖的GYEP培养基中,使最终体积(Working volume)为50mL,从而在摇床培养箱(Shakingincubator)中在30℃和180rpm的条件下进行培养。在培养时间内,通过使用分光光度计(UV/Visible Spectrophotometer)在波长为680nm的条件下测量吸光度作为光密度(Optical Density:OD)值,从而测量各步骤的细胞生长程度。
结果,如图2所示,确认出,与野生株CD01-5000相比,突变株CD01-5004显示出同等或更高水平的糖消耗速度和细胞生长,并且比野生株快约5小时达到指数生长期(Exponential phase)。
实施例6.确认野生型裂殖壶菌属菌株CD01-5000和突变裂殖壶菌属菌株CD01-5004的生长特性
确认了新型的野生型裂殖壶菌属菌株CD01-5000和其突变裂殖壶菌属菌株CD01-5004在培养时是否可以在宽pH范围内生长。

另外,从培养CD01-5000和CD01-5004菌株时培养液呈深红色来看,可以看出其为生产类胡萝卜素系列的抗氧化色素,例如,β-胡萝卜素(β-carotene)、叶黄素(lutein)、虾青素(astaxanthin)、辣椒红(capsanthin)、胭脂树橙色素(annatto)、斑蝥黄质(canthaxanthin)、番茄红素(lycopene)、β-阿扑-8-胡萝卜醛(β-apo-8-carotenal)、玉米黄素(zeaxanthin)、β-阿扑-8-胡萝卜醛-酯(β-apo-8-carotenal-ester)等的菌株。

Claims (14)
1.一种裂殖壶菌属微细藻类,其
以保藏号KCTC14344BP或KCTC14345BP保藏;以及
具有二十二碳六烯酸、二十碳五烯酸、以及棕榈酸的生产能力。
2.根据权利要求1所述的裂殖壶菌属微细藻类,其中,基于脂肪酸的总重量,所述微细藻类生产35重量%至60重量%的二十二碳六烯酸。
3.根据权利要求1所述的裂殖壶菌属微细藻类,其中,基于脂肪酸的总重量,所述微细藻类生产0.5重量%至10重量%的二十碳五烯酸。
4.根据权利要求1所述的裂殖壶菌属微细藻类,其中,基于脂肪酸的总重量,所述微细藻类生产10重量%至30重量%的棕榈酸。
5.根据权利要求1所述的裂殖壶菌属微细藻类,其中,所述微细藻类具有类胡萝卜素的生产能力。
6.根据权利要求5所述的裂殖壶菌属微细藻类,其中,所述类胡萝卜素为选自由β-胡萝卜素、叶黄素、虾青素、辣椒红、胭脂树橙色素、斑蝥黄质、番茄红素、β-阿扑-8-胡萝卜醛、玉米黄素、β-阿扑-8-胡萝卜醛-酯组成的组中的任一个或多个。
7.一种源自裂殖壶菌属微细藻类的生物量,其包括权利要求1的裂殖壶菌属微细藻类、所述微细藻类的培养产物、所述培养产物的干燥产物、或所述干燥产物的粉碎产物。
8.一种饲料组合物,其包括权利要求7的源自裂殖壶菌属微细藻类的生物量或所述生物量的浓缩产物或干燥产物。
9.一种源自裂殖壶菌属微细藻类的生物量的制造方法,包括:
培养裂殖壶菌属微细藻类,其以保藏号KCTC14344BP或KCTC14345BP保藏并具有二十二碳六烯酸、二十碳五烯酸、和棕榈酸的生产能力;以及
从所述微细藻类、所述微细藻类的培养产物、所述培养产物的干燥产物、或所述干燥产物的粉碎产物中回收生物量。
10.根据权利要求9所述的源自裂殖壶菌属微细藻类的生物量的制造方法,其中,所述培养在从属营养条件下进行。
11.根据权利要求9所述的源自裂殖壶菌属微细藻类的生物量的制造方法,其中,所述培养通过使用包括碳源和氮源的培养基来进行。
12.根据权利要求11所述的源自裂殖壶菌属微细藻类的生物量的制造方法,其中,所述碳源为选自由葡萄糖、果糖、麦芽糖、半乳糖、甘露糖、蔗糖、阿拉伯糖、木糖和甘油组成的组中的任一个或多个。
13.根据权利要求11所述的源自裂殖壶菌属微细藻类的生物量的制造方法,其中,所述氮源为i)选自由酵母提取物、牛肉提取物、蛋白胨、和胰蛋白胨组成的组中的任一个或多个的有机氮源,或者为ii)选自由乙酸铵、硝酸铵、氯化铵、硫酸铵、硝酸钠、尿素、和谷氨酸钠组成的组中的任一个或多个的无机氮源。
14.一种源自裂殖壶菌属微细藻类的生物油的制造方法,包括:
培养裂殖壶菌属微细藻类,其以保藏号KCTC14344BP或KCTC14345BP保藏并具有二十二碳六烯酸、二十碳五烯酸、和棕榈酸的生产能力;以及
从所述微细藻类、所述微细藻类的培养产物、所述培养产物的干燥产物、或所述干燥产物的粉碎产物中回收脂质。
