Abstract
本发明提供了一种泥蚶STING蛋白及其应用,该泥蚶STING蛋白的氨基酸序列如SEQ ID NO:1或SEQ ID NO:3所示;泥蚶有两个STING蛋白,根据长短把STING序列分别命名为TgSTINGL、TgSTINGS,两个STING蛋白在各个组织内普遍表达,其中在血细胞和鳃内表达量最高,泥蚶STING蛋白在受到溶藻弧菌、李斯特菌或poly(I;C)等致病因子刺激后,表达量会上升,表明泥蚶STING蛋白基因参与了抗菌免疫反应,可用于调控泥蚶抗病原菌感染,为泥蚶病害治疗防御策略提供支持,促进泥蚶养殖业的健康发展。

技术领域
本发明涉及泥蚶免疫蛋白领域,具体涉及一种泥蚶STING蛋白及其应用。
背景技术
泥蚶(Tegillarca granosa)是我国东南沿海地区重要的海水养殖贝类之一,它含有非常丰富的营养物质,因此具有极高的经济价值和广阔的市场前景。然而近年来,由于水体污染以及致病菌频繁爆发,泥蚶大量死亡,给泥蚶养殖业造成了巨大的经济损失。目前,通过分子研究辅助来预防水产养殖病害已成为现代水产技术发展的必然趋势,干扰素基因刺激因子STING(Stimulator of interferon gene,STING)是cGAS/STING信号通路的关键蛋白,能够调节宿主的先天免疫系统来抵御病原菌的入侵。该STING蛋白是重要的接头蛋白,在病原感染时能够发挥防御作用。在细菌、病毒等刺激下,环状GMP-AMP酶能够被激活,释放产生第二信使分子cGAMP并与STING结合,被活化的STING蛋白能够二聚化并从内质网转移到高尔基体,进而招募其他的信号激酶TBK1,IKK等,激活并磷酸化干扰素调节因子IRF3、NF-κB等,从而进入细胞核介导Ⅰ型干扰素和促炎症因子的转录表达。STING在哺乳动物中的研究比较系统,但是在低等生物免疫方面的阐述还比较少。泥蚶具有复杂的先天免疫系统,主要依靠血细胞进行细胞免疫、伤口修复以及抵御外来病原菌的入侵,cGAS/STING信号通路是否参与了泥蚶对病原菌的免疫调控还不得而知。
发明内容
为了解决上述问题,本发明提出一种泥蚶STING蛋白及其应用,为泥蚶病害治疗防御策略提供支持,促进泥蚶养殖业的健康发展。
为了实现上述目的,本发明的实施例在第一方面提出了一种泥蚶STING蛋白,其氨基酸序列如SEQ ID NO:1或SEQ ID NO:3所示。
根据本发明实施例的一种泥蚶STING蛋白,泥蚶有两个STING蛋白,根据长短把STING序列分别命名为TgSTINGL、TgSTINGS,两个STING基因在各个组织内普遍表达,其中在血细胞和鳃内表达量最高,泥蚶STING基因在受到溶藻弧菌、李斯特菌或poly(I;C)等致病因子刺激后,相对表达量会上升,表明泥蚶STING蛋白基因参与了抗菌免疫反应,使得其可用于调控泥蚶抗病原菌感染,为泥蚶病害治疗防御策略提供支持,促进泥蚶养殖业的健康发展。

以下实施例涉及的材料和方法:
一、细胞和质粒载体
细胞为人肾上皮细胞(HEK-293T)、宫颈癌细胞(HeLa),由实验室保存。上述细胞使用的是添加10%的胎牛血清(FBS)、10%的100μg/mL链霉素和100IU的盘尼西林的DMEM高糖培养基,链霉素/盘尼西林双抗生素DMEM培养基溶液购于Gibco公司。
基于对泥蚶通路内基因的序列分析推测,为了研究泥蚶STING目的蛋白的功能表达,需要将TgSTINGL基因克隆到表达载体上制备成携带目的基因的表达载体,载体选择带有His标签原核表达载体pET-30b(+),载体图谱信息如图1所示;为了进一步研究TgSTINGL基因在细胞内的功能性表达,后续将TgSTINGL基因克隆到真核表达载体pcDNA3.1(+)-EGFP。pcDNA3.1-EGFP真核质粒是实验室通过由含CMV启动子的质粒pcDNA3.1(+)在HindⅢ酶切位点后插入能够在哺乳动物细胞内高效表达的绿色荧光蛋白EGFP报告基因改进而成,能够与目的基因形成融合蛋白表达,载体图谱信息如图2。
二、实验所用的泥蚶购买自厦门市第八海鲜市场,将刚买的新鲜泥蚶,洗净淤泥,挑选大小相近的,放入水槽中海水充氧饲养。并于每天更换1/3的干净海水,清理状态不好的泥蚶。
海水中养殖3天以后的泥蚶,每只泥蚶注射20μL溶藻弧菌、李斯特菌(2×108CFU/mL)。在感染0、1.5、3、6、12小时后,随机收集3只血蚶的血细胞作为样本,提取总RNA。
从健康的泥蚶中随机取3只混合不同组织,分别提取血细胞、外套膜、斧足、闭壳肌、鳃、内脏团的总RNA。
三、分子相关实验
1、PCR反应所需的引物序列如表1。

2、RNA的提取
TRIzol法提取组织总RNA。提前准备试剂:三氯甲烷,异丙醇,75%乙醇,无RNase水。
(1)将组织在液氮中磨碎,每100mg组织加入1mL TRIzol,用匀浆仪进行匀浆处理,用移液器吸打混匀;混匀后的样品室温静置放5min,使核酸蛋白复合物完全分离。
(2)每使用1mL TRIzol加入200μL三氯甲烷,漩涡振荡器上剧烈振荡15秒,室温放置3min;4℃,10000rpm,离心15min;此时RNA主要在上层水相中,吸取550μL上层水相,把上层水相转移到新EP管中,用异丙醇混匀沉淀水相中的RNA。
(3)加入500μL异丙醇,室温放置10min;4℃,10000rpm离心10min,移去上清。
(4)用75%乙醇洗涤RNA沉淀。加入1mL 75%乙醇,8℃,7000rpm离心5min,移去上清。
(5)室温放置6min干燥RNA沉淀;加入50μL的无RNase水,枪头吹打胶状沉淀混匀,室温放置5min使RNA溶解;吸取5μL,通过凝胶电泳验证RNA质量并使用NanoDrop1000UV/可见分光光度计测定RNA浓度后,使用PrimeScript RT试剂盒将高质量RNA反转录为第一链cDNA。剩余的RNA分装完后-80℃保存。
3、RNA逆转录为cDNA
按照逆转录试剂盒说明书,RNA逆转录为cDNA分为两步反应。
基因组DNA去除反应如下表1:

4、实时荧光定量PCR
NCBI数据库和SnapGene软件设计qPCR引物,以Tg18S rDNA为内参基因,RNA逆转录得到的第一链cDNA为模板,稀释50倍之后,进行荧光定量PCR。
(1)检测目的基因的表达

室温下融解Biomarker 2×SYBR Green Fast qPCR Mix,然后置于4℃备用,待完全融解后,小心振荡混匀,离心收取底部液体。
制备预混液(内参基因同上),分别加入相应的引物和模板,待所有试剂(Mix,引物,模板),混匀,并瞬时离心。先计算实验所需Mix体积,对照内参基因引物为Tg18s的上下游。
将上述反应体系转移到专用的qPCR板内,用光学密封薄膜仔细密封。
2500rpm离心qPCR板,收集所有的反应液至孔底部,避免气泡,准备上机。

(2)统计分析方法
Real Time PCR结果以泥蚶的18s rDNA基因为内参,采用2-△△ct的方法处理实验组与对照组的数据,获得差异表达倍数;应用One-way ANOVA方法进行显著性差异分析,图中的“*”代表与对照组相比存在显著差异(P<0.05),“**”代表与对照组相比存在极显著差异(P<0.01)。
5、RACE PCR(RACE方法扩增3'端和5'端)
3'端RACE克隆:
通过基因组测序以及从NCBI数据库内比对,已经找到了STINGL和STINGS的一个序列片段,为获得两个基因的全长序列,选用RACE方法来扩增3'端和5'端。试剂盒选择:3′-Full RACE Core Set with PrimeScriptTMRTase(Takara)来获得3′端的序列。该试剂盒运用巢式PCR的原理,增加PCR扩增的灵敏度和特异性。TaKaRa LA Taq酶扩增所得的PCR产物在其3′端含有一个碱基“A”,可直接连接到pEASY-T1的T-Vector。3′RACE Adaptor引物有TaKaRa设计的dT区域,提高了逆转录的效率。5'端的扩增则采用TDT末端加尾法对片段进行锚定。
3'端RACE克隆,实验步骤:

结束反应后,取6μL PCR反应产物进行琼脂糖凝胶电泳来确认PCR产物大小。若这轮反应没有得到明显的目的产物条带,则需要进行下一轮Inner PCR反应。

48μL体系94℃反应3min,短暂离心后冰水浴2min,轻轻混匀后于37℃反应5h,70℃孵育15min以终止EDTA反应,-20℃储存。

反应结束后,将反应液跑琼脂糖凝胶电泳确认PCR扩增产物。然后将符合目的条带大小的所有条带切胶回收,并做好标记。用Nano Drop测量纯化回收产物浓度。
T载体连接目的片段。将目的片段连在T载体pEASY-T1载体或19-T Vector上,按T-Vector pMD 19(Simple)与插入片段DNA摩尔比1:5制备成5μL混合液,向混合液中加入5μL Solution I,轻轻混匀;在16℃条件下反应30min。(若连接到pEASY-T1 SimpleCloning Vector,则按照插入片段每1kb长度加入20ng切胶回收产物,再加入1μL pEASY-T1Simple Cloning Vector,无菌水补足至总体积5μL,于25℃条件下连接10min)。将连接产物加入到100μL刚融化的感受态细胞中,转化步骤同上,涂板时要涂含(1%氨苄青霉素,X-Gal IPTG终浓度为50μg/mL,2%,20%),37℃的LB固体培养基中,次日多挑取白色单菌落到96深孔板,37℃,180rpm摇床摇12h,然后选样品去送测序,比对测序结果。
6、PCR产物纯化
(1)预处理:取一个新的硅胶膜吸附柱子装在收集管中,吸取100μL的平衡液于柱子中。13000rpm离心1min,倒掉收集管中废液,将吸附柱子重新放回收集管。
(2)称一个空1.5mL离心管重量。用干净刀片将所需回收的DNA条带在紫外灯下切下,将切下的含有DNA条带的凝胶条带放入1.5mL离心管,再次称重,两次重量相减。
(3)加3倍体积溶胶液DD。56℃,水浴放置10min(或直至胶完全溶解)。中间上下混匀加速溶解。
(4)所得溶液加入到吸附柱EC中,室温放置2min。12,000rpm离心30秒,倒掉收集管中的废液。
(5)加入600μL漂洗液WB(已加入无水乙醇),12,000rpm离心30秒,弃掉废液。重复用漂洗液再清洗一次。将吸附柱EC放回空收集管中,12,000rpm离心2min,除去漂洗液。
(6)取出吸附柱EC,放入一个干净的1.5mL离心管中,在吸附膜的中间部位加50μL洗脱缓冲液EB(事先在65℃水浴中加热),室温放置2min后,12,000rpm离心1min。取5μL洗脱液用Nanodrop测量胶回收浓度。
7、质粒提取
在过夜培养的平板上挑取单克隆菌落到LB培养基中(含有相应的抗生素),在37℃摇床中摇12小时后提取质粒。(P1放置于4度冰箱)
(1)取约5mL菌液12000rpm离心1min,去上清。
(2)取250μL的P1重悬菌体沉淀,然后再加250μL的P2温和地上下转动试管数次,至菌液变得透明使得菌体得到充分地裂解(过程小于5min),再向菌液中加入350μL的P3,立即温和地上下转动试管数次,充分混匀试剂(可以看到有白色絮状物沉淀)。12000rpm常温离心10min。
(3)向吸附柱CP3加入500μL平衡液BL来平衡吸附柱,12000rpm离心1min后弃废液。将第二步离心得到的上清转移到平衡好的吸附柱CP3中,12000rpm离心1min后弃废液。
(4)向CP3中加入600μL的漂洗液PW,12000rpm离心1min后弃废液。此步骤重复两次清洗。
(5)重新将CP3用12000rpm离心2min以去除残留的漂洗液,然后打开CP3的盖子室温放置数分钟使乙醇挥发干净。
(6)向CP3中滴加50-100μL洗脱液,室温静置3min。12000rpm离心2min将洗脱液收集于灭菌后的1.5mL离心管中。测定浓度后置于-20℃冰箱。
8、重组表达载体构建
(1)感受态细胞DH5α和BL21(DE3)制备。
①提前配置无抗性的LB固体培养基平板和TB缓冲液(预冷)。对实验室保存的DH5α和BL21(DE3)菌株进行平板划线(无抗性)并置于37℃的培养箱中生长14小时。
②挑取DH5α和BL21(DE3)单克隆菌于50mL锥形瓶(内含10mL液体TB培养基)中,37℃摇床摇至OD600为0.5,按1:100转接到200mL TB培养基中,18℃度摇至OD600为1-3后,4℃,3000rpm,5min离心收菌。
③加入TB缓冲液(预冷),4℃,3000rpm,5min离心,去上清,清洗菌体一遍。再加入TB缓冲液(预冷)和DMSO,冰上操作,保证重悬后DMSO终浓度为7%,OD600读值为5。
④置冰上,每管分装100μL的感受态细胞于1.5mL离心管中(预冷)。迅速放入液氮中1min左右,转放入-80℃冰箱保存。
(2)质粒载体酶切回收
选取了两个原核表达载体pET-30b(+)、pGEX-4T-1,对选择的2个不同的载体选取BamH I、xho I两个酶切位点进行双酶切,并对酶切完的质粒DNA片段进行纯化回收。
双酶切反应体系:

反应条件:37℃培养箱,反应2小时,加入10×的Loading Buffer终止反应。反应后酶切产物核酸电泳,胶回收。
(3)TEDA法克隆连接

(4)转入感受态细胞
将连接产物20μL加入到100μL刚融化的感受态细胞DH5α中,立即放冰上冰浴30min,42℃水浴,热激60-90s,立即放冰上10min。加1mL不含抗性的LB液体培养基,37℃,180rpm孵育活化1h。3,000rpm室温离心1min,去除部分上清,留100μL吹打混匀。均匀涂至含终浓度为1%氨苄青霉素或卡那霉素的平板,37℃培养箱倒置过夜培养14个小时。
(5)次日挑取白色单菌落至5mL抗性LB培养液中,37℃,180rpm振荡16h。将白色的单菌落摇起来,选几个样品送去测序公司测序。
9、蛋白诱导表达
将构建好的重组质粒进行浓度测定,然后取1-2μL表达质粒转入感受态细胞BL21(DE3)表达,轻轻吹打混匀。冰上放30min,42℃水浴锅热激90s,冰上再放10min后加入1mL的LB培养基,混匀后37℃摇床活化1h。离心后留100μL重悬均匀涂在分别在含有卡那霉素抗性的平板上过夜培养,诱导表达重组蛋白pET30b-TgSTINGL。
第二天,平板划线菌落于5mL的LB培养基中,37℃摇床摇至OD600为0.5时加入IPTG诱导剂进行诱导表达。设计正交试验,分别在18℃,25℃,37℃的条件下对目的蛋白进行诱导,保证IPTG终浓度为0.1mM。离心收集菌体,用Lysis buffer重悬起来后,35%功率,超声3s停4s,直到裂解液变清亮。4℃,10000rpm,15min离心。
10、蛋白分离与纯化
裂解离心后的上清,用0.45μM的滤膜过滤后,加入300μL平衡过的Ni-NTA resin(预平衡:5mL的Lysis buffer重悬清洗两遍)于4℃缓慢摇晃结合1个半小时。上柱前,用Buffer A柱平衡,流穿2个柱体积后上柱;将结合后液体混匀后流穿层析柱,再用含25mM咪唑的Buffer A清洗约6个柱体积,除去非特异性结合的杂蛋白;加入1mL Elution buffer B(250mM咪唑)用4℃缓慢结合半个小时。
收集最后一步洗脱液,高浓度咪唑250mM咪唑洗脱目的蛋白。
BCA蛋白定量法测定蛋白浓度:
(1)标准液稀释。首先用PBS配制2mg/mL的牛血清蛋白标准液。均匀混匀后,在96孔酶标板里按梯度稀释成1mg/mL、0.5mg/mL、0.25mg/mL、0mg/mL的蛋白标准液。
(2)BCA试剂配制。按试剂盒里的A液和B液比例为1:50取样,充分混匀在2mL的EP管内。
(3)在干净的酶标板孔里,加入10μL按梯度配置的蛋白标准液、待测的蛋白溶液,每孔再加入混匀后的100μL BCA溶液,将两种溶液轻轻摇晃混匀后,放入37℃培养箱静置30min。
(4)酶标仪读取OD562时读值,以吸光值为Y,牛血清蛋白含量为X,绘制标准曲线。根据标准曲线,测定蛋白浓度。
SDS-PAGE电泳鉴定蛋白的纯度:
(1)制备样品。取纯化好的蛋白液30μL于1.5mL离心管中,加入10μL的4×SDS上样缓冲液(加入还原剂DTT),充分混匀后,放入100℃的沸水中煮10min,加热煮沸使蛋白样品充分变性。放入冰上短暂冷却,高速离心后,取上样缓冲液上层20μL上样。
(2)电泳。配置12%的蛋白胶,凝固后加入1×电泳缓冲液并上样,每孔20μL,电泳条件为:在80V恒压下,观察到蛋白marker分层后,将电压增加到120V,继续电泳,直至分离胶最小的marker条带跑至绿色底端后即可。
(3)染色脱色。取下蛋白胶,放入考马斯亮蓝染色剂中,常温下轻轻摇晃染色2h。清水洗掉染色剂后加入脱色液,摇晃至蛋白胶背景干净即可。
蛋白免疫印迹Western blot:
(1)转膜。将制备好的样品先SDS-PAGE电泳(80V,60min;120V,60min)跑完,取下SDS-PAGE胶放在转膜的棉片上,将蛋白胶多余部分切掉,剪一块与蛋白胶大小的PVDF膜,用甲醇浸泡活化2min后小心附在蛋白胶上。压至无气泡后,夹紧放进转膜装置。加入转膜液(提前4℃预冷1h),低温(周围放冰)状态下,100V,60min下进行转膜。
(2)免疫反应。转膜结束后,用镊子将PVDF膜小心取出。用5%的脱脂牛奶封闭2小时,2小时后TBST清洗1遍。再用1:3000的1%的脱脂牛奶稀释anti-GFP抗体并过夜敷。第二天早上,TBST清洗3遍,每5min一次。HRP标记的羊抗鼠IgG按1:5000的比例稀释在1%的脱脂牛奶,孵育1个小时。TBST清洗5遍,每10min一次。
(3)显影。在暗匣内平铺一张保鲜膜并固定,然后将胶片小心放置在显影暗匣内,ECL显色液按A液:B液=1:1充分混匀后,滴在膜上,盖上保鲜膜并吸走多余的发光液。暗匣子内反应2min后,在暗室无光的条件下将胶片剪开附上,迅速合上暗匣暗反应5min。反应后在显影液里摇晃显影1-2min,在红光下能够看到条带后,迅速放入到定影液里定影。
四、细胞实验
1、293T细胞复苏:从液氮罐中取出HEK-293T细胞冻存管,迅速放入37℃水浴锅中,并不停摇晃使其快速解冻。完全解冻后,2000rpm,离心2min。在超净台里,吸掉上清,并加入1mL新鲜的DMEM培养基重悬细胞,将细胞重悬液接种到含有7mL DMEM完全培养基的10cm细胞培养板中,轻轻晃匀后置于37℃,5% CO2培养箱培养,6个小时后更换一次培养液后继续培养。
2、细胞传代
将生长至90%密度的细胞进行传代,吸除掉培养基液体,加入1mL灭菌的1×PBS(37度水浴锅预热)到细胞培养板,PBS清洗细胞2遍,然后再弃去该溶液。再加入1mL浓度为0.25%的胰酶,室温消化大约1-2min,待细胞完全消化下来时,加入1mL DMEM培养基,用移液枪轻轻吹打数次,将壁上的细胞吹打下来。将细胞重悬液移至1.5mL EP管中,2000rpm,离心2min,在超净台里,吸去上清,加入1mL新鲜的DMEM培养基重悬细胞,接种到96cm板,置于37℃,5% CO2培养箱过夜培养。
3、细胞转染
将脂质体PEI稀释成1mg/mL,按体积比质粒:PEI为(1:3)将质粒和PEI分别用100μL的opti-DMEM无血清培养基稀释成A液和B液,充分混匀,静置5min;将充分混匀的B液加入到A液中,再次充分混匀,静置20min后,加入到过夜培养的96cm盘的293T细胞中,轻轻晃动;六个小时后将培养液换成DMEM完全培养基。37℃,5% CO2培养箱过夜培养。
4、免疫共聚焦
①显微镜下观察到细胞共定位,将1×PBS提前放在37℃水浴锅中预热半个小时。
②细胞固定。将细胞培养小皿从培养箱中取出,贴壁吸除掉培养液,并缓慢加入PBS轻轻摇晃清洗(2-3次)。吸除掉PBS后加入1mL 4%的多聚甲醛固定细胞,静置30min。
③弃掉后每5min加入1mL PBS清洗3次,摇床上轻轻摇晃。
④通透。吸除掉后加入1mL的免疫染色通透液0.1% Triton X-100,静置15min室温透膜。弃掉后PBS清洗,步骤同上③。
⑤DAPI染色。PBS清洗完,加入100μL DAPI染色剂没过盖玻片,静置避光染色15min。弃掉后PBS清洗,步骤同上③。
⑥清洗完后在载玻片中央滴一滴抗荧光淬灭封片液,然后将盖玻片小心夹到载玻片封片液上,轻轻从一侧缓慢放,避免过程中产生气泡,用吸水纸吸掉一侧多余的液体,待晾干后用指甲油轻轻封住,激光共聚焦扫描显微镜拍照。
5、双荧光素酶报告基因
转染HeLa细胞:
提前复苏HeLa细胞,细胞密度生长到80%时,传代到实验用的96孔板,用1mg/mL的PEI进行转染操作。需要提前准备好报告基因质粒。
对照组:①HeLa细胞:正常培养后不处理
②HeLa细胞:转染NF-κB luciferase质粒
实验组:③HeLa细胞:转染NF-κB luciferase+STINGL质粒
质粒+STINGL+Renilla每孔转染质粒约100-200ng,根据质粒浓度配制质粒稀释液和PEI稀释液,每个实验组做5个重复,充分混匀后,室温静置30min,加到每孔中,37℃,5%的CO2中培养24h。
试剂配制:
把Luciferase Reaction Substrate按1:5(1Vial Substrate+5buffer)的比例充分溶解,避光保存。同时,按1:50比例将Luciferase Reaction Substrate II和LuciferaseReaction Substrate II混合,避光保存。用5×Cell lysis buffer同RNAse free water稀释成1×的混合备用。
手动荧光检测:
细胞去掉培养基,用1×PBS润洗两次后加入20μL Cell lysis buffer,室温裂解10min后取细胞到1.5mL离心管中,4℃,12000rpm离心10min,取20μL裂解液到干净的离心管中备用。
取100μL的Luciferase Reaction reagent(已平衡至室温)加入到裂解液上清的离心管中,移液枪轻轻吹打3次,混匀于发光检测仪中检测,记录发光值firely Luciferase(RLU1中)。
取100μL的Luciferase Reaction reagentII(已平衡至室温)加入上述上清的离心管中,移液枪轻轻吹打3次,立刻记录检测发光值(RLU2中)。最后记录RLU1/RLU2比值。
下面参考具体实施例,对本发明进行描述,需要说明的是,这些实施例仅仅是描述性的,而不以任何方式限制本发明。
实施例1转录组学分析
文库构建:从泥蚶的血细胞中提取总RNA,富集mRNA,逆转录SMARTerTMPCR cDNASynthesis Kit对泥蚶mRNA进行全长cDNA的合成,PCR大量扩增合成全长cDNA,纯化PCR产物,对全长cDNA进行末端修复,修复损坏的或者不完整的末端,基于单分子测序(Sequencing By Synthesis,SMRT)技术,把修复的末端和SMRT哑铃型接头连接。然后进行核酸外切酶消化,获得测序文库。
测序及质量控制:测过滤序列。通过过滤掉片段小于50bp、序列准确性小于0.9的酶聚合序列(Polymerase reads)后,将剩余序列从接头处打断并过滤掉接头序列后得到聚合酶序列子序列(subreads),再过滤长度小于50bp的subread,剩余的得到我们要的高质量的过滤后的原始数据序列(clean data)。
TransDecoder软件预测转录组编码区序列和氨基酸序列。使用SMRTLink软件中的IsoSeq进行序列长度聚类分析。利用MISA软件做SSR分析。通过软件GraphPad Prism 5,进行显著性分析。
本次转录组测序共用1个cell,获得过滤后的clean data 23.36Gb,其中共得到CCS read325,351条,全长非嵌合序列206,330条,对高质量全长非嵌合序列聚类得到一致序列76,416个,校正得到高质量一致序列共70,030个,对高质量一致序列进行去冗余得到41,570转录本序列。对转录本进行功能注释,其中共有27,921序列得到注释。比对NR数据库27,324个序列中,泥蚶与牡蛎氨基酸序列一致性最高,其次是霸王莲花青螺,说明泥蚶与海洋贝类的基因同源性最高。
通过对转录组数据分析以及转录组基因注释信息的查找,在泥蚶全长转录组中找到了STING信号通路的其他关键基因cGAS、TBK1、IKK、NF-κB,表明了在泥蚶体内cGAS-STING信号通路是完整的,STING蛋白参与天然免疫防御过程,在抗细菌、病毒感染、抗病原微生物的免疫过程中,发挥胞质DNA和免疫防御的信号转导的关键作用。
根据长短把STING序列分别命名为TgSTINGL、TgSTINGS。通过转录组数据分析,得到了TgSTINGS的部分序列,关于两段STING蛋白序列在贝类等无脊椎动物中行使的免疫功能相关研究阐述较少,STING序列在泥蚶的免疫反应中的作用也有待进一步研究。为了比较长短两段STING序列在泥蚶体内结构和功能、在组织分布中的差异性,在感染病原菌刺激下作出的免疫应答反应在抗病原菌感染作用的机制,通过RACE法扩增出了TgSTINGS全长cDNA,再通过NCBI数据库对TgSTINGS结构域进行分析。
泥蚶TgSTINGS的cDNA全长为1,466bp,5′端UTR为81bp,3′端UTR为86bp。其中CDS编码区有1,299bp,编码的蛋白质分子含有432个氨基酸,理论分子质量约为49.3kDa。通过结构域分析,发现TgSTING的ORF区含有两个结构域。STING(位置在aa 30-206)和TIR_2(位置在aa 238-367)的结构域,分别是属于STING-like超家族和TIR_2超家族。泥蚶TgSTINGS的氨基酸序列如SEQ ID NO:1所示;核苷酸序列如SEQ ID NO:2所示。
泥蚶的TgSTINGL的cDNA全长为2,365bp,5′端UTR为22bp,3′端UTR为279bp,以及多聚A(poly A)尾巴。其中开放阅读框ORF为2,064bp,编码的蛋白分子含有687个氨基酸,理论分子质量约为79.1kDa。包含一个N-末端跨膜区和一个胞质C-末端球状结构域,跨膜结构域与面向细胞质的球状结构域相互作用,形成一个完整的区域交换二聚体。通过对泥蚶序列结构域分析(https://www.ncbi.nlm.nih.gov/Structure/cdd/wrpsb.cgi),发现TgSTINGL的ORF含有两个保守的TIR,TIR-2结构域(位置在aa 8-109,aa 356-470),TIR结构域是Toll样受体家族和胞内的衔接蛋白所共有的高度保守的结构域,属于TIR-2家族。此外还有两个保守的STING-C结构域(位置分别在aa 151-318、aa 495-687),属于sting-c家族。泥蚶TgSTINGL的氨基酸序列如SEQ ID NO:3所示;核苷酸序列如SEQ ID NO:4所示。
实施例2构建重组质粒
通过对转录组数据结果分析以及NCBI数据库比对,能够确定泥蚶体内有两个STING蛋白序列。把TgSTINGL的序列构建到原核表达载体pET-30b(+)上,构建重组质粒pET30b-TgSTINGL。通过对实验室保存的原核载体质粒pET-30b(+)进行双酶切,选择BamHI、xho I为双酶切位点,回收切胶产物,如图3。
泥蚶血细胞中含有完整的STING基因序列,可以通过TRIzol裂解血细胞,提取总RNA并逆转录成cDNA。利用SnapGene软件和NCBI数据库(https://www.ncbi.nlm.nih.go)设计含有相同酶切位点的同源序列引物,然后通过PCR扩增获取TgSTINGL基因的编码CDS区域。通过琼脂糖核酸凝胶电泳检验PCR产物的大小,可以从图4中看到符合TgSTINGL基因大小的条带2,064bp,符合预期的实验结果。
过夜培养并筛选出能够通过卡那霉素抗性筛选的菌落,在提取质粒后进行双酶切验证,并通过PCR鉴定电泳结果。如图5,通过核酸电泳能够得到两条分别为5382bp和2064bp的条带。把双酶切后条带位置相符合的重组质粒测序,比对测序结果准确。表明克隆序列准确无误,将构建成功的重组质粒命名为pET30b-TgSTINGL。
实施例3融合蛋白pET30b-TgSTINGL诱导表达与纯化
将构建成功的pET30b-TgSTINGL重组质粒转化入BL21(DE3)感受态细胞,将重组蛋白TgSTINGL小量诱导表达,设置诱导表达pET-30b(+)质粒空载体为空白对照。设置温度和IPTG浓度的正交实验,在不同的温度以及不同的诱导表达条件下,通过SDS-PAGE电泳检测TgSTINGL蛋白的表达。与实验对照比较,STING蛋白在破碎完菌体的上清和沉淀中有均少量表达。同时发现在18℃,IPTG终浓度为0.1mM,16个小时的诱导条件下,TgSTINGL蛋白的表达量最多。
由于TgSTINGL蛋白在超声破碎菌体离心后的上清和沉淀中均存在少量表达,SDS-PAGE电泳无法直接确定表达量少的蛋白是目的条带的表达。为了进一步验证在marker约75kDa条带泳道的蛋白是TgSTINGL蛋白的特异性表达,采用蛋白免疫印迹的方法对该泳道的蛋白进行鉴定。设置空载体pET30b为对照,利用抗体结合特异性,Western Blot结果确定pET30b-TgSTINGL能够特异性表达,对照组没有条带,TgSTINGL目的蛋白条带大小约为75kDa。与SDS-PAGE结果一致(如图6所示)。
利用优化的诱导表达条件,对TgSTINGL蛋白进行大量的诱导表达。并通过Ni-NTA柱亲和层析,对带有His标签的融合蛋白进行纯化,低浓度咪唑梯度洗掉杂蛋白,最终在250mM高浓度的咪唑时,洗脱TgSTINGL蛋白。收集各组分,通过SDS-PAGE电泳检测,结果图7所示,显示纯化的蛋白大小约为75kDa,结果与预期一致。
BCA蛋白定量法对纯化的蛋白进行浓度检测,结果显示纯化的TgSTINGL蛋白的浓度约为0.7mg/mL。可以用于后续对蛋白功能实验使用。
实施例4基因TgSTINGL在细胞内的定位
通过瞬时转染293T细胞,观察融合蛋白在细胞内的分布,进一步分析TgSTINGL在细胞内的功能。先构建过表达重组质粒pcDNA3.1-EGFP-TgSTINGL。
将构建成功的含有目的基因和GFP荧光信号融合蛋白的重组质粒瞬时转染293T细胞,使用荧光共聚焦显微镜观察,并以pcDNA3.1-EGFP空载体为对照。结果如图8所示,对照空载体pcDNA3.1-EGFP转染的293T细胞,整个细胞结构都具有清晰地绿色荧光,而融合蛋白TgSTINGL表达后的293T细胞,只有细胞质内可见清晰地绿色荧光,细胞核内几乎没有绿色荧光。结果表明,STING蛋白主要定位在293T细胞的细胞质中。这与哺乳动物STING蛋白参与的信号通路定位一致,哺乳动物、软体动物中与泥蚶进化同源性最高的牡蛎,体内STING蛋白未受外界刺激被激活的前提下,主要定位在细胞质中的内质网。在感知到外界异常信号时,从内质网转移到高尔基体,进一步说明了STING蛋白在293T细胞中存在功能性表达。
实施例5TgSTINGL组织表达分析
通过qPCR荧光定量技术来分析两条序列表达量在泥蚶不同组织的分布差异,首先分别从健康的泥蚶中提取血细胞、外套膜、斧足、闭壳肌、鳃、内脏团的RNA并逆转录成单链cDNA,并以cDNA为模板,18S rDNA为内参基因,对其中的因素进行显著性分析。
结果如图9所示,在泥蚶的所有组织中均检测到TgSTINGL基因在mRNA水平上表达。其中在血细胞的表达量最高,鳃、内脏团次之,在斧足中的表达量最低。设置表达量最低的斧足为参考对照,血细胞的相对表达量远高于斧足,约是对照组的25倍(p<0.01)。泥蚶的所有组织中均检测到TgSTINGS基因普遍表达,TgSTINGS在鳃中的表达量最高,斧足和外套膜次之,内脏团中的表达量最低,鳃跟斧足中的相对表达量具有显著性的差异(p<0.01)。
实施例6不同病原菌处理下的TgSTING表达水平的变化
1、溶藻弧菌刺激下TgSTINGL和TgSTINGS表达水平的变化
选取同生长期健康的泥蚶,将溶藻弧菌注射进泥蚶体内,并以0.85%的生理盐水为对照组。在病原菌刺激后的0h、1.5h、3h、6h、12h取泥蚶血液,收集血细胞提取总RNA,并逆转录成cDNA,利用qPCR技术分析转录水平的变化。
如图10所示,在注射溶藻弧菌后,TgSTINGL相对表达量呈上升趋势,在3小时TgSTINGL的转录水平达到最高,约为对照组的7.4倍(p<0.01)。随后降到正常水平,TgSTINGL能够对溶藻弧菌的刺激做出免疫响应,说明了血细胞是参与贝类免疫的重要组成部分。
与TgSTINGL不同的是,同对照组相比,TgSTINGS相对表达量在3小时内呈下降趋势,在3小时mRNA水平降到最低,随后开始逐渐上调。
2、李斯特菌刺激下TgSTINGL和TgSTINGS表达水平的变化
注射李斯特菌后,如图11所示,与对照组相比,TgSTINGL和TgSTINGS在mRNA表达水平上均发生了明显的变化。TgSTINGL的相对表达量在1.5小时内逐渐上调,在1.5小时相对表达量达到最高,约为对照组的5.5倍。随着时间水平推移,逐渐降低。TgSTINGS在第1.5个小时的相对表达量最高,约为对照组的7.3倍,存在显著性的差异,随后至12小时,逐渐恢复到正常水平。
3、poly(I;C)刺激下TgSTINGL和TgSTINGS表达水平的变化
如图12所示,在poly(I;C)处理下,TgSTINGL基因的相对表达量3个小时内逐渐上升,在3个小时上升达到最高,与对照组相比,差异性显著(p<0.01),约为对照组的6倍。随后降低到初始水平。与TgSTINGL基因相比较不同的是,TgSTINGS基因相对表达量在1.5小时内略有上调,逐渐上升在3小时达到最高水平,约为对照组的5倍,在第3小时到12小时又下降回到正常水平。表明poly(I;C)刺激后,TgSTINGL参与了泥蚶天然免疫防御反应。
实施例7下游通路NF-κB的调控
双荧光素酶报告基因实验是可以快速灵敏检测转录因子和目的基因启动子表达增强和抑制的一种检测方法。为了确定STING感染激活了NF-κB信号传导途径,利用双荧光素酶报告系统检测NF-κB的活化机制。将荧光素酶报告质粒NF-κB luc和pcDNA3.1-EGFP-TgSTINGL共转染HeLa细胞。
如图13所示,与未转染STING基因的HeLa细胞相比,单转染NF-κB luc报告基因质粒能够引起部分上调,共同转染的实验组荧光素酶活性明显增强。在STING和NF-κB luc报告基因质粒共同转染的实验组能够引起显著性上调(p<0.01),大约是对照组的40倍。表明STING在激活状态下,能够激活NF-κB信号传导机制,诱导促炎症因子的表达。
综上,根据本发明的实施例,获得STING的两段基因编码区序列,并根据序列长短命名为TgSTINGL、TgSTINGS。STINGL蛋白定位在细胞质中。在病原致病菌的刺激下,分别注射溶藻弧菌、李斯特菌以及poly(I;C)之后0h、1.5h、3h、6h和12h作用下,TgSTING两条序列的免疫应答响应时间是存在明显差异,在mRNA水平上存在变化。通过双荧光素酶报告基因实验,检测STING被激活后对下游基因的调控。结果表明STING被激活后能够依赖NF-κB通路,产生促炎症因子等。本申请初步探索了STING蛋白在先天免疫防御过程中作用的分子机制,阐明了泥蚶在病原菌感染过程中的信号传递机制,丰富了泥蚶等双壳贝类先天免疫理论知识,为泥蚶病害治疗防御策略提供支持,促进泥蚶养殖业的健康发展。

1.一种泥蚶STING蛋白,其氨基酸序列如SEQ ID NO:1或SEQ ID NO:3所示。
2.如权利要求1所述的泥蚶STING蛋白,其特征在于,用于编码氨基酸序列如SEQ IDNO:1所示的泥蚶STING蛋白的核苷酸序列如SEQ ID NO:2所示;用于编码氨基酸序列如SEQID NO:3所示的泥蚶STING蛋白的核苷酸序列如SEQ ID NO:4所示。
3.如权利要求1或2所述的泥蚶STING蛋白在调控泥蚶抗病原菌感染中的应用。
4.如权利要求1或2所述的泥蚶STING蛋白在抗病原菌泥蚶品种的培育中的应用。
5.如权利要求3或4所述的应用,其特征在于,所述应用为通过提高所述泥蚶STING蛋白的表达水平,提高泥蚶抗病原菌感染能力。
6.如权利要求3或4所述的应用,其特征在于,所述病原菌包括溶藻弧菌或李斯特菌。
7.如权利要求5所述的应用,其特征在于,将所述泥蚶STING蛋白的编码基因构建至载体上得到表达质粒,将所述表达质粒转移至哺乳动物细胞中进行表达。
8.如权利要求7所述的应用,其特征在于,所述载体为pET-30b(+)或pcDNA3.1(+)-EGFP。
