Abstract
本发明提供了一种复方蒜皮组合物,属于药食同源组合物技术领域。本发明提供的复方蒜皮组合物包括如下重量份的组分:蒜皮2‑4份、姜1‑2份、姜黄1‑2份、茄子1‑2份、车前子壳1‑2份、绿茶0.5‑1份、芦荟1‑2份、仙人掌1‑2份、桂皮1‑2份、金针菇0.5‑1份。本发明提出的配方均为药食同源材料,药性温和且无副作用,临床上适合大部分体质人群长期服用。本发明能够显著降低高脂饲喂小鼠体重、体脂率、血脂和肝脏脂肪,对改善高脂饮食条件下血脂健康水平以及预防肥胖具有显著效果。

技术领域
本发明属于药食同源组合物技术领域,尤其涉及一种复方蒜皮组合物及其制备方法和应用。
背景技术
肥胖不仅是全球流行性慢性疾病,且肥胖与一系列疾病之间有较强关联性,肥胖和250多种遗传变异和广泛的临床疾病有关,可直接导致血栓栓塞、糖尿病、肿瘤等疾病。肥胖的形成对人体健康造成了潜在的威胁。目前,临床上用于治疗肥胖的西药副作用很大,对患者的有些器官造成了损害,成为困惑患者的难题。因此副作用较低的防治高血脂、高血糖及肥胖症的药食同源组方成为患者的迫切需要。
蒜皮是大蒜加工的副产物,含有脂肪酸、硫醚类化合物及膳食纤维等生理活性物质,但目前蒜皮作为大蒜加工过程中产生的副产品,常被抛弃或燃烧掉,没有得到人们的重视和良好的利用,不仅造成环境的污染,同时也是对资源的极大浪费。
配伍是中药应用的基本形式之一,食物的配伍就是把中医的食物配伍理论融入进去,通过合理选择与配搭来达到保健作用。为了增强食物的效用和可食性,把蒜皮与食品进行配伍验证其具有的功效,同时提高蒜皮的利用率,是值得期待的。
发明内容
有鉴于此,本发明的目的在于提供一种蒜皮与其它食品配伍的组合物,能够有效的降低血脂水平,预防肥胖,同时提高蒜皮的利用率。
为了实现上述发明目的,本发明提供了以下技术方案:
一种复方蒜皮组合物,包括如下重量份的组分:蒜皮2-4份、姜1-2份、姜黄1-2份、茄子1-2份、车前子壳1-2份、绿茶0.5-1份、芦荟1-2份、仙人掌1-2份、桂皮1-2份、金针菇0.5-1份。
优选的,包括如下重量份的组分:蒜皮2.5-3.5份、姜1.3-1.6份、姜黄1.3-1.6份、茄子1.3-1.6份、车前子壳1.3-1.6份、绿茶0.7-0.8份、芦荟1.3-1.6份、仙人掌1.3-1.6份、桂皮1.3-1.6份、金针菇0.7-0.8份。
本发明的另一目的在于提供上述复方蒜皮组合物的制备方法,包括如下步骤:按比例称取各原料;分别粉碎;将各原料粉混合即得复方蒜皮组合物。
优选的,所述原料粉碎后过10-200目筛。
本发明的另一目的在于提供上述复方蒜皮组合物在制备维持血脂健康水平产品中的应用。
本发明的另一目的在于提供上述复方蒜皮组合物在制备抗肥胖产品中的应用。

本发明的另一目的在于提供一种含有所述复方蒜皮组合物的产品,所述产品中复方蒜皮组合物的质量百分比为1-10%。

相对于现有技术,本发明具有如下有益效果:
本发明提供了一种复方蒜皮组合物,通过特定的成分配比制备成复方蒜皮组合物,该组合物具有显著的抗肥胖作用,解决了现有技术中天然食品减肥降脂效果所需剂量大、材料昂贵等问题,且药性温和无副作用,临床上适合大部分体质人群长期食用。本发明复方蒜皮组合物为天然食品减肥降脂功能开发提供依据。本发明复方蒜皮组合物食用剂量小,解决了传统天然食品需在服用高剂量条件下才能达到降脂和抗肥胖的技术问题。本发明组合物提高了对蒜皮的利用率,有效避免了大蒜加工过程中造成的资源浪费现象。

本发明运用传统中医理论和食物的药性理论,选择本发明中复方蒜皮组合物中的组分,通过特定的成分配比,使获得的复方蒜皮组合物具有显著的抗肥胖作用,在高脂饮食条件下能够维持血脂的健康水平。
本发明研究发现,蒜皮富含多糖、多酚、果胶及膳食纤维等活性成分,是天然的农副产品,具有抗菌、保健、无副作用的特点,蒜皮中含有的大蒜素能通过抑制肥胖小鼠体质量增加、降低肥胖和改善肝脏脂肪变性而发挥显著作用。本发明以蒜皮为主要原料,促进了对蒜皮的利用,并使制备的组合物具有良好的维持血糖、血脂健康水平的效果。
姜为姜科植物姜的鲜根茎,具有镇静及抗惊厥、解热、镇痛和抗炎的作用,能够促进体内活性物质释放,与本发明复方蒜皮组合物中其他原料相互配合,具有促进吸收,降血脂的功效;姜黄保肝、利胆、抗菌、抗炎、抗肿瘤、降血脂,对消化系统有保护作用;姜及姜黄对大鼠血脂具有降血脂调节作用。
茄子为常见食材,富含多种营养元素;能降低胆固醇,还能防止高脂血症引起的血管损害,可辅助治疗高血压、高血脂、动脉硬化等病症。
车前子壳是一种传统草药,富含丰富的膳食纤维;可以降低二型糖尿病患者的血浆NEFFA、TC、TG、LDL-C、HDL-C水平,降低血尿酸水平。
绿茶具有提神清心、清热解暑、消食化痰、去腻减肥、清心除烦、解毒醒酒、生津止渴、降火明目、止痢除湿等药理作用,对多种现代疾病具有一定的药理功效;绿茶多酚能够降低小鼠的血糖水平,改善小鼠的糖尿病病情。
芦荟健胃下泄、杀菌、强心活血;芦荟多糖有较好的降糖作用,还可以效预防糖尿病心血管病的发生。
仙人掌具有解毒散结、清热去火、抗疲劳、提高免疫力的作用;仙人掌改善体重增加、肝脏和腹部脂肪重量增加,能够改善与肥胖相关的代谢参数。
桂皮含有蛋白质、脂肪、纤维素等成分,具有温肾壮阳、温中散寒、疏通经络等功效;可改善糖尿病患者体内肥胖相关指标。
金针菇的维生素和膳食纤维含量高,具有补肝益肾、宽肠胃、益智健脑、调脂降胆固醇、防疲劳等功效。金针菇多糖能改善小鼠由高脂饮食引起的高脂血症,对血脂和肝脂中TC和LDL-C等指标具有显著降低的效果。
本发明针对目前多为对单一食材或食材单体的研究现状,创造性的选择将十种食材进行配伍。本发明将十种食药同源食材配伍后研究发现,本发明复方蒜皮组合物富含黄酮、多酚、多糖类化合物等有效成分,具有调节血脂的功效,能够通过抑制脂质合成与吸收、促进脂质代谢转运及抑制脂质氧化等机制,改善机体的TC、TG、LDL-C以及HDL-C水平而调节血脂。
本发明获得的复方蒜皮组合物,将十种药食同源的食材进行配伍,各原料中的活性成分有机结合,共同作用发挥出良好的维持血脂健康水平,抗肥胖的效果。
本发明提供了上述复方蒜皮组合物的制备方法,包括如下步骤:按比例称取各原料;分别粉碎;将各原料粉混合即得复方蒜皮组合物。
作为一种可实施的方式,本发明中原料粉碎后过10-200目筛,优选过30-100目筛。
本发明提供了上述复方蒜皮组合物在制备维持血脂健康水平产品中的应用。
本发明提供了上述复方蒜皮组合物在制备抗肥胖产品中的应用。
本发明中,所述抗肥胖产品降低血脂水平。
本发明还提供了一种含有所述复方蒜皮组合物的产品,包括质量百分比为1-10%的上述复方蒜皮组合物;优选的,所述产品包括质量百分比为3-6%的上述复方蒜皮组合物。本发明复方蒜皮组合物具有显著的抗肥胖作用,在维持血脂健康水平的产品中添加剂量小,即可达到良好的降血脂的效果。
本发明中,所述含有所述复方蒜皮组合物的产品以复方蒜皮组合物为有效成分之一或唯一有效成分,可维持高脂饮食条件下血脂的健康水平,具有预防肥胖的效果。
作为一种可实施的方式,本发明维持血脂健康水平的产品可制备为不同剂型,所述剂型包括颗粒剂、片剂、胶囊剂、口服液、散剂、饮料、茶剂、冲剂、酒剂、饼干类或糕点类中的任意一种。
如无特殊说明,本发明涉及的试剂、耗材等均可从商业途径获得,如未注明具体使用条件,通常按照常规条件,或按照公司所推荐的条件进行。

实施例5
本实施例对比了不同组合物的减重降脂功效
1、实验动物分组及处理
(1)SPF级4周龄雄性C57BL/6J小鼠30只,体重12-13g,购自长春市亿思实验动物技术有限责任公司。小鼠购进后给予标准饲料和自由饮水适应性饲养一周;
小鼠称重后随机分为5组(n=6):正常饮食组(NCD)、高脂饮食组(HFD)、复方蒜皮组合物低剂量组(HFD+L-1%)、复方蒜皮组合物中剂量组(HFD+M-3%)、复方蒜皮组合物高剂量组(HFD+H-5%);
其中,NCD给予普通饲料,HFD组给予60%脂肪供能高脂饲料,HFD+L-1%、HFD+L-3%、HFD+L-5%组分别给予添加了质量百分比为1%、3%、5%实施例1中组合物的60%脂肪供能高脂饲料,实验期间每周定期监测小鼠的体重和饮食摄入变化。
其中,普通饲料购于江苏省协同医药生物工程有限责任公司,组成为:18.8%粗蛋白、5.2%粗脂肪、3%粗纤维、5.4%粗灰分、1.1%钙、0.7%总磷;
60%脂肪供能高脂饲料购于江苏省协同医药生物工程有限责任公司,组成为:22.9%粗蛋白、34.1%粗脂肪、3%粗纤维、4.4%粗灰分、1.1%钙、0.7%总磷;
(2)试验12周后,各组小鼠禁食12h(自由饮水),称量最终体重,眼球采血,分离血清后,测定血清指标。解剖时收集肝脏、心脏、肾脏、附睾脂肪和肾周脂肪组织,滤纸吸干表面水分后称重,保留到0.001g,计算脏器指数、体重变化、体重增加量、摄食量及能量摄入量。
1.2血清指标检测
(1)采集小鼠的新鲜血液,常温下全血静置30min。
(2)4℃,5000r/min,10min离心,吸取上清即为小鼠血清。
(3)使用酶标仪测定血清的甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)、谷丙转氨酶(ALT)、谷草转氨酶(AST)水平。
1.3肝脏及附睾脂肪结构检测
(1)将肝脏组织及附睾脂肪组织取出后,取部分组织固定于10%福尔马林溶液中避光保存。
(2)组织切成4-5mm3的小块,流水过夜冲洗,用乙醇从低浓度到高浓度梯度脱水后二甲苯透明,软蜡浸泡后硬蜡包埋,待石蜡凝固后取下组织块进行修块,38℃摊片时应尽量避免褶皱,烘片机上40℃烘片3h以上。
(3)烘干后按照H&E染色程序染色,中性树脂封片后制成组织切片,在光学显微镜下进行观察组织形态并对视野进行拍照。
(4)使用ImageJ软件分析附睾脂肪细胞平均面积。
1.4统计学分析
实验结果以平均值±标准误差形式表示。试验数据采用GraphPadPrismprogram8.0统计学软件进行分析和制图,使用ImageJ软件对实验图片结果进行定量分析,实验数据表示为Mean±SD,用单因素方差分析(One-way ANOVA)检验进行显著性比较,认为P<0.05时具有统计学意义,P<0.001表示具有极显著差异。
1.5测定结果
1.5.1按照上述处理方法对不同组小鼠喂养12周后,各组小鼠的体重如图1所示。如图1-A所示,随着喂养时间增加,各组小鼠体重呈不同程度增加,第4周时,各组小鼠体重开始出现统计学差别(P<0.05)。在12周实验进程中,NCD组小鼠体重增长趋势较为平缓,HFD组小鼠体重速度较快,试验周期末HFD组小鼠体重显著高于NCD组(P<0.05),证明高脂饮食成功诱导小鼠营养性肥胖。
添加实施例1中组合物的三个剂量组体重增长趋势较高脂饮食组相比均有所减缓(图1-B),中和高剂量组在第4周开始体重与HFD组相比具有数据统计学差异(P<0.05)。主体呈现为,NCD组体重最低,HFD组体重最高,蒜皮配伍的剂量组介于二者之间,且剂量越高越趋近于NCD组体重。结果表明:在高脂饮食喂养第12周时,各组体重均与正常组有显著性差异,但高剂量组体重显著低于模型组,趋近于NCD体重,说明本发明复方蒜皮组合物能降低高脂模型饮食的肥胖小鼠体重。
1.5.2从图2-A可知,NCD组、HFD组及复方蒜皮组合物剂量组之间食物摄入量无显著性差异(P>0.05),这说明复方蒜皮组合物可以在不影响小鼠进食量的前提下发挥有效缓解高脂饮食诱导的小鼠体质量增加。
从图2-B可以看出,经过计算高脂饮食喂养组与复方蒜皮组合物剂量组各组小鼠能量摄入基本为相似水平且无统计学差异(P>0.05)。说明复方蒜皮组合物饮食可缓解体重增长,但不影响小鼠正常能量摄入。
1.5.3各组织脏器指数如图3所示。由图3-A、B可知,与NCD小鼠相比,HFD组小鼠与NCD组小鼠的附睾脂肪、肾周脂肪指数相比呈显著增加,说明高脂饲料可导致小鼠出现脂肪堆积、肥大等问题,而饲喂含复方蒜皮组合物高脂饲料小鼠的附睾脂肪、肾周脂肪指数明显下降,其中高剂量组的肾周脂肪指数与模型组相比存在极显著差异。复方蒜皮组合物干预后小鼠附睾脂肪及肾周脂肪重量呈下降趋势,说明复方蒜皮组合物降低体重作用有一部分是通过减少脂肪量完成的。
除心脏之外,各组肝脏、肾脏的脏器指数均无显著差异,说明复方蒜皮组合物在缓解小鼠肥胖时,对其他脏器损害小,无明显副作用。可见,以蒜皮配伍干预有利于肥胖小鼠内部脂肪代谢功能的恢复,减轻小鼠脂肪堆积情况,且未对其他器官造成明显损伤。
1.5.4从图4-A、B可知,HFD组小鼠血清中TG和TC含量显著高于NCD组(P<0.001),而复方蒜皮组合物处理各组小鼠后,血清TG和TC含量与HFD组相比呈现显著下降趋势(P<0.05),其中HFD+H组差异极显著(P<0.001),并趋近于正常组。
从图4-C可知,HFD组HDL-C含量显著低于NCD组(P<0.01),其中HFD+H组与HFD组相比HDL-C含量均有显著升高且与正常组达到相似水平(P<0.01)。从图4-D可以看出,NCD组LDL-C含量显著低于HFD组(P<0.001),复方蒜皮组合物处理后,中、高剂量组的LDL-C水平有下降趋势(P<0.05)。这说明高脂肥胖小鼠血脂代谢紊乱情况得到纠正,血脂代谢动态平衡向着更有利、健康的方向移动。
1.5.5由图5-A可知,与NCD组相比,HFD组的AST水平明显提高(P<0.001),而复方蒜皮组合物低中高三个剂量组均有明显的降低(P<0.001)。从图5-B可以看出,与HFD组相比,虽然低中两个剂量组作用不显著,但HFD组作用显著降低(P<0.001)。这说明复方蒜皮组合物可以显著缓解高脂对肝脏造成的损伤。
1.5.6复方蒜皮组合物对高脂小鼠肝脏脂质沉积的影响如图6所示,NCD组小鼠肝脏切面染色均匀,可见肝细胞以中央静脉为中心向周围放射状排列,肝小叶结构清晰,肝脏细胞结构完整且排布整齐,细胞核清晰无异变,肝脏组织中几乎没有脂肪滴形成。而HFD组小鼠肝脏脂肪变性明显,肝脏组织被大量脂滴充斥,镜下可见肝细胞肿大变圆,肝脏细胞排列紊乱且边界不清。
复方蒜皮组合物处理后小鼠肝脏组织内虽也观察到有脂肪滴形成,但脂肪滴数量和大小与HFD组相比有明显降低,中、高剂量组小鼠肝脏受损程度有显著改善,细胞排列有序且整体水平趋于NCD组小鼠,肝脏组织中脂质积累的现象也明显减少(图6-C、D)。综上所述复方蒜皮组合物可能通过抑制脂肪生成来改善高脂饮食引起的肝脏脂质代谢异常,对肝脏脂肪积累有明显改善效果。
1.5.7复方蒜皮组合物对高脂小鼠肝脏脂质水平的影响如图7所示,HFD组小鼠肝脏组织中TG和TC含量显著高于NCD组(P<0.001),复方蒜皮组合物处理各组小鼠肝脏中TG和TC含量均呈下降趋势,且具有显著性差异(P<0.001)。
1.5.8复方蒜皮组合物对高脂小鼠脂肪积累的影响如图8所示,NCD组小鼠的脂肪细胞面积小而紧密,HFD组小鼠的脂肪细胞面积明显更大而且呈现扩散状态,而复方蒜皮组合物处理可改善小鼠脂肪组织中脂滴过量积累以及脂肪肥大的现象。
1.5.9复方蒜皮组合物对高脂小鼠附睾脂肪细胞大小的影响如图9所示,HFD组小鼠脂肪细胞平均面积显著高于NCD组(P<0.001),复方蒜皮组合物干预各组平均面积与HFD组相比呈下降趋势且均具有显著差异(P<0.001)。结合对脂肪细胞切片观察和对脂肪细胞平均面积分析的结果证明,复方蒜皮组合物治疗可有效的减少小鼠附睾脂肪脂滴大小。
综上所述,将蒜皮与姜、姜黄、茄子、车前子壳、绿茶、芦荟、仙人掌、桂皮、金针菇复配可以调节肥胖小鼠脂代谢的不良变化。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (10)
1.一种复方蒜皮组合物,其特征在于,包括如下重量份的组分:蒜皮2-4份、姜1-2份、姜黄1-2份、茄子1-2份、车前子壳1-2份、绿茶0.5-1份、芦荟1-2份、仙人掌1-2份、桂皮1-2份、金针菇0.5-1份。
2.根据权利要求1所述复方蒜皮组合物,其特征在于,包括如下重量份的组分:蒜皮2.5-3.5份、姜1.3-1.6份、姜黄1.3-1.6份、茄子1.3-1.6份、车前子壳1.3-1.6份、绿茶0.7-0.8份、芦荟1.3-1.6份、仙人掌1.3-1.6份、桂皮1.3-1.6份、金针菇0.7-0.8份。
3.权利要求1或2所述复方蒜皮组合物的制备方法,其特征在于,包括如下步骤:按比例称取各原料,分别粉碎,将各原料粉混合即得复方蒜皮组合物。
4.根据权利要求3所述复方蒜皮组合物的制备方法,其特征在于,所述原料粉碎后过10-200目筛。
5.权利要求1或2所述复方蒜皮组合物在制备维持血脂健康水平产品中的应用。
6.权利要求1或2所述复方蒜皮组合物在制备抗肥胖产品中的应用。
7.根据权利要求6所述的应用,其特征在于,所述抗肥胖产品降低血脂水平。
8.一种含有权利要求1或2所述复方蒜皮组合物的产品,其特征在于,所述产品中复方蒜皮组合物的质量百分比为1-10%。
9.根据权利要求8所述的产品,其特征在于,所述复方蒜皮组合物为所述产品的有效成分之一或唯一有效成分。
10.根据权利要求8或9所述的产品,其特征在于,所述产品为颗粒剂、片剂、胶囊剂、口服液、散剂、饮料、茶剂、冲剂、酒剂、饼干类或糕点类中的任意一种。
