Description
Technical Field
The application relates to the technical field of traditional Chinese medicines, in particular to a traditional Chinese medicine composition for reducing blood sugar and application thereof.
Background
Diabetes is a metabolic disease characterized by hyperglycemia due to relative insulin deficiency or insulin resistance. The diabetes patients have fat and protein metabolism disorder, and clinically can show the symptoms of diuresis, polydipsia, emaciation and the like, and the long-term hyperglycemia is easy to cause acute complications such as ketoacidosis and the like or chronic complications such as blood vessels, nerves and the like. Among them, type ii diabetes is the most common type of diabetes, and its occurrence and development are related to various factors such as insulin resistance, obesity, or impaired beta cell function.
The existing treatment of diabetes mainly controls blood sugar and reduces complications. The hypoglycemic medicine mainly comprises oral western medicines hypoglycemic medicine and injection insulin analogues, but the sustained administration is required, and the side effect of the medicine is obvious. Therefore, in recent years, the adoption of traditional Chinese medicine treatment has become an important choice for treating diabetes, but at present, a traditional Chinese medicine formula with ideal blood glucose reducing effect is still lacking.
Disclosure of Invention
In order to solve the problems, the application provides a traditional Chinese medicine composition for reducing blood sugar and application thereof, and the traditional Chinese medicine composition has remarkable blood sugar reducing effect and can effectively treat and/or prevent diabetes.

45-55 parts of tortoise plastron, 45-55 parts of medlar, 35-40 parts of aspongopus, 20-25 parts of semen astragali complanati, 5-10 parts of mulberry, 5-10 parts of ginseng, 10-20 parts of hawthorn and 15-20 parts of corn silk.
Further, the traditional Chinese medicine composition for reducing blood sugar comprises the following components in parts by weight:
50 parts of tortoise plastron, 50 parts of medlar, 37 parts of aspongopus, 22 parts of aspongopus, 20 parts of semen astragali complanati, 10 parts of mulberry, 10 parts of ginseng, 20 parts of hawthorn and 18 parts of corn silk.
In a second aspect, the application provides an application of the hypoglycemic Chinese medicinal composition in any one of the first aspect in preparing hypoglycemic products.
In a third aspect, the application provides a traditional Chinese medicine honeyed pill for reducing blood sugar, and the functional components of the traditional Chinese medicine honeyed pill for reducing blood sugar comprise the traditional Chinese medicine composition for reducing blood sugar in any one of the first aspect.
Further, the preparation method of the traditional Chinese medicine honeyed pill for reducing blood sugar comprises the following steps:
decalcification treatment is carried out on tortoise plastron, and enzymolysis is carried out to obtain tortoise plastron protein extract;
carrying out supercritical extraction on the aspongopus to obtain aspongopus extract;
extracting fructus Lycii, herba Aspongopus, semen astragali Complanati, mori fructus, ginseng radix, fructus crataegi and stigma Maydis with water to obtain mixed extract;
mixing and sieving the tortoise plastron protein extract, the aspongopus extract and the mixed extract, adding the mixture into honey, stirring, and preparing the traditional Chinese medicine honeyed pill for reducing blood sugar.
Further, the steps of decalcification treatment of tortoise plastron and enzymolysis to obtain tortoise plastron protein extract comprise the following steps:
crushing tortoise plastron, adding the crushed tortoise plastron into 0.5-1.0 mol/L ethylenediamine tetraacetic acid, and stirring for 1-3 hours to obtain a first reaction feed liquid;

Unless otherwise specifically indicated, the various raw materials, reagents, instruments, equipment and the like used in the present application are commercially available or may be prepared by existing methods.
In a first aspect, the application provides a traditional Chinese medicine composition for reducing blood sugar, which comprises the following components in parts by weight:
40-60 parts of tortoise plastron, 40-60 parts of medlar, 30-40 parts of aspongopus, 15-25 parts of semen astragali complanati, 5-15 parts of mulberry, 5-15 parts of ginseng, 5-20 parts of hawthorn and 5-20 parts of corn silk.
Tortoise plastron: is prepared from the tortoise shell and the abdominal shell, and has the effects of nourishing yin, suppressing yang, tonifying kidney, strengthening bone, nourishing blood, tonifying heart, and the like.
Wolfberry fruit: is a dried mature fruit of Lycium barbarum of Solanaceae, and has effects of replenishing kidney essence, moistening lung, and nourishing liver.
Aspongopus (L.) kuntze: is the body of Aspongopus of stinkbugaceae, and has effects of activating qi-flowing, relieving pain, warming kidney, and supporting yang.
Murraya paniculata: is prepared from dried leaves and twigs of Murraya koenigii and Murraya koenigii of Rutaceae, and has effects of activating qi-flowing, relieving pain, promoting blood circulation, removing blood stasis, etc.
Semen astragali Complanati: is a dry mature seed of astragalus membranaceus which is a leguminous plant, and has the effects of tonifying kidney, supporting yang, securing essence, reducing urination, nourishing liver, improving eyesight and the like.
Mulberry: is dry fruit ear of Morus alba Linne of Moraceae, and has effects of nourishing yin, replenishing blood, promoting salivation, and moistening dryness.
Ginseng: is prepared from dried root and rhizome of Panax ginseng of Araliaceae, and has effects of invigorating primordial qi, recovering pulse, relieving depletion, invigorating spleen, benefiting lung, promoting salivation, nourishing blood, tranquilizing mind, improving intelligence, etc.
Hawthorn fruit: is a dried and mature fruit of the hawthorn plant, and has the effects of invigorating stomach, promoting digestion, promoting qi circulation, removing blood stasis, resolving turbidity, reducing lipid and the like.
Corn silk: is prepared from the flower column and the column head of maize, which are the gramineous plants, and has the effects of inducing diuresis to alleviate edema, clearing liver-fire, promoting bile flow and the like.

In some embodiments, the step of decalcifying tortoise plastron and then performing enzymolysis to obtain the tortoise plastron protein extract comprises the following steps:

In some specific embodiments, the weight of each hypoglycemic Chinese medicinal honeyed pill is 5-8 g, preferably 6g.
The preparation method of the traditional Chinese medicine honeyed pill for reducing blood sugar provided by the application is simple to operate, does not need extra special equipment, and is suitable for industrial production. Meanwhile, the preparation method of the traditional Chinese medicine honeyed pill for reducing blood sugar is realized based on the traditional Chinese medicine composition for reducing blood sugar in any one of the first aspect, so that the traditional Chinese medicine honeyed pill for reducing blood sugar has at least all the beneficial effects brought by the technical scheme of the embodiment and is not repeated here. The operation steps involved in the preparation method of the traditional Chinese medicine honeyed pill for reducing blood sugar provided by the embodiment of the application can be carried out in a conventional manner in the art without special limitation or explanation, and are not repeated here.

The example provides a traditional Chinese medicine composition for reducing blood sugar, which comprises the following components in parts by weight (1 g in each 1 part by weight): 40 parts of tortoise plastron, 40 parts of medlar, 30 parts of aspongopus, 15 parts of semen astragali complanati, 5 parts of mulberry, 5 parts of ginseng, 5 parts of hawthorn and 5 parts of corn silk.
The preparation method of the traditional Chinese medicine composition for reducing blood sugar comprises the following steps:
grinding 40g of tortoise plastron, sieving with a 50-100 mesh sieve, adding 100mL of ethylenediamine tetraacetic acid with the concentration of 0.7mol/L, stirring for 2 hours, adding excessive sodium hydroxide with the concentration of 1mol/L, continuously stirring for 7 hours, centrifuging, filtering, and washing to obtain a tortoise plastron solid after decalcification treatment; adding the decalcified tortoise plastron solid into 200mL of water, steaming and boiling for 2 hours, cooling to room temperature, and adding trypsin with the enzyme amount of 2.5 ten thousand U to carry out enzymolysis for 7 hours to obtain the tortoise plastron protein extract;
grinding 30g of aspongopus, sieving with a 80-mesh sieve, adding into a supercritical device, performing supercritical extraction, placing the extracted product in an environment of 5 ℃ for 2 hours at constant temperature, centrifuging at 6000r/min for 20 minutes to remove bottom sediment, and vacuumizing the supernatant in a vacuum box at 45 ℃ for 12 hours to obtain aspongopus extract; wherein, the working parameters of the supercritical extraction comprise: the solvent is supercritical carbon dioxide fluid, the pressure is 45Mpa, the temperature is 50 ℃, the time is 1.5 hours, and CO 2 The flow rate of the fluid is 15kg/h;
pulverizing 40g of medlar, 30g of Murraya paniculata, 15g of semen astragali Complanati, 5g of mulberry, 5g of ginseng, 5g of hawthorn and 5g of corn silk, adding water with the weight being 7 times of the total weight of the traditional Chinese medicine raw materials in a water extraction mode for extraction, and concentrating under reduced pressure and drying to constant weight to obtain the mixed extract; wherein the extracted working parameters include: the temperature is 105 ℃, the time is 2 hours, and the times are 1 time;
mixing the tortoise plastron protein extract, the aspongopus extract and the mixed extract, and sieving the mixture with a 50-100-mesh sieve to obtain the traditional Chinese medicine composition for reducing blood sugar.

The present example provides a hypoglycemic Chinese medicinal composition, which differs from example 5 only in that: extracting tortoise plastron by adopting a water extraction mode, wherein the specific process is as follows: grinding tortoise plastron, sieving with a 50-100 mesh sieve, adding water with the weight being 7 times of that of the tortoise plastron, extracting, concentrating under reduced pressure, and drying to constant weight to obtain the aspongopus extract; wherein the extracted working parameters include: the temperature is 105 ℃, the time is 2 hours, and the times are 1 time; the rest steps and parameters are the same.
Test case
This example demonstrates the efficacy of the hypoglycemic Chinese medicinal compositions provided in examples 1 to 5 and comparative examples 1 to 4.
The testing method and the grouping are as follows: 120 were used: SPF-class C57BL/6J male mice are randomly divided into 12 groups, namely a blank group (1 group; not molded), a model group (1 group; molded, not administered), a positive control group (1 group; molded, administered with commercial rosiglitazone sodium tablet at 5 mg/kg/day, and administered by intragastric administration), and a hypoglycemic Chinese medicinal composition group (9 group; molded, numbered 1 st to 11 hypoglycemic Chinese medicinal composition groups; 1 st to 5 th hypoglycemic Chinese medicinal composition groups are sequentially administered with hypoglycemic Chinese medicinal compositions provided in examples 1 to 5 respectively, and 6 th to 9 th hypoglycemic Chinese medicinal compositions are sequentially administered with hypoglycemic Chinese medicinal compositions provided in comparative examples 1 to 4 respectively, and the administration amounts are 50 mg/kg/day, and administered by intragastric administration). The positive control group and the hypoglycemic Chinese medicinal composition group are administered by gastric lavage for 1 time a day for 30 days continuously, and the blank blood sugar value and body weight of the mice are measured.
The molding method comprises the following steps: the mice are fasted for 24 hours (free drinking water), the molding is carried out by injecting the tetraoxypyrimidine into the abdominal cavity for 130mg/kg, and after the free diet for 7 days, the fasting blood glucose value is measured, and the blood glucose value is more than 10mmol/L, thus the molding is carried out successfully.
The test results are shown in Table 1.

Application example
In the embodiment, the hypoglycemic Chinese medicinal composition provided in the embodiment 5 is prepared into honeyed pills and then applied to treating diabetics.

Diabetes patient information: men, 41 years old, were diagnosed with type ii diabetes. 40-60 parts of tortoise plastron, 40-60 parts of medlar, 30-40 parts of aspongopus, 15-25 parts of semen astragali complanati, 5-15 parts of mulberry, 5-15 parts of ginseng, 5-20 parts of hawthorn and 5-20 parts of corn silk.
2. The hypoglycemic traditional Chinese medicine composition according to claim 1, wherein the hypoglycemic traditional Chinese medicine composition comprises the following components in parts by weight:
45-55 parts of tortoise plastron, 45-55 parts of medlar, 35-40 parts of aspongopus, 20-25 parts of semen astragali complanati, 5-10 parts of mulberry, 5-10 parts of ginseng, 10-20 parts of hawthorn and 15-20 parts of corn silk.
3. The hypoglycemic traditional Chinese medicine composition according to claim 1, wherein the hypoglycemic traditional Chinese medicine composition comprises the following components in parts by weight:
50 parts of tortoise plastron, 50 parts of medlar, 37 parts of aspongopus, 22 parts of aspongopus, 20 parts of semen astragali complanati, 10 parts of mulberry, 10 parts of ginseng, 20 parts of hawthorn and 18 parts of corn silk.
