本发明属于药品、保健品、食品或日化用品领域,涉及左、右旋丹参素及其药学上可接受的盐以及其酯在抗衰老、祛斑及美白中的应用,具体涉及右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯在单独或者作为有效成分在制备抗衰老及皮肤美白、祛斑和治疗色素沉着性疾病以及食品添加剂中的用途。
背景技术
丹参素(danshensu,DSS)又名D(+)-β-(3,4-二羟基苯基)乳酸或(R)-3-(3,4-二羟基苯基)-2-羟基丙酸,是丹参水溶性活性成分之一,相对分子质量为198.17,分子式为C9H10O5。天然植物来源的丹参素均为右旋异构体。文献中,有关丹参素的合成、衍生物以及生物活性报道较多,如丹参素具有心脑血管疾病防治(心肌保护、脑保护、抗动脉粥样硬化、血栓形成)、抗肿瘤、抗炎、抗病毒、抗肝脏纤维化、脏器损伤保护等(西北药学杂志,2011,26(04):310-312;CN112516124A,山西医药杂志,2006(02):108-110+186;中西医结合肝病杂志,1999(02):30-32.;中西医结合肝病杂志,1996(03):29-30+32-56.)。右旋丹参素异丙酯具有血管扩张、抗炎、抗氧化、抑制自由基所致的大鼠心律失常、保护心肌细胞、降脂、抗缺氧等生物活性(中国药理学通报,2023(08):1541-1547;中草药,2013,44(01):59-64.;中国中药杂志,2011,36(08):1094-1096.;心脏杂志,2011,23(01):27-30+34.中草药,2009,40(01):82-86.)。对于手性化合物,尤其是对映异构体间的生物活性往往有较大的不确定性。因此,随着潜在右旋丹参素系列药物的不断开发,其左旋异构体(L(-)-β-(3,4-二羟基苯基)乳酸或(S)-3-(3,4-二羟基苯基)-2-羟基丙酸或左旋丹参素)及其酯类衍生物也逐渐备受关注。然而除左旋丹参素的化学与生物合成报道外,相关生物活性鲜有报道(CN104744242),尤其左旋丹参素及其药学上可接受的盐以及其酯的毒性以及相应对映异构体间的药效学比较至今未有报道。
随着人们对抗衰老、美白机制与产品的不断研究,市面涌现出大量的防治药品、保健品与护肤品,然而由于多数产品针对性过于单一,在抗衰老与美白功能上的改善能力仍显不足,有些甚至对身体或皮肤带来伤害。而人们对延缓衰老与“白里透红”的向往,始终是抗衰老与美白产品的发展方向。本专利针对以上不足,结合血管保护(促进血管新生、抗炎、抗氧化等)与美白功能,公开一类具有抗衰老与美白效果的结构通式(I)化合物(左旋丹参素及其药学上可接受的盐以及其酯以及右旋丹参素及其药学上可接受的盐以及其酯)。结构通式(I)化合物通过养血、血管新生平衡与美白的协同作用达到抗衰老与美白的效果,使人们在身体健康、延年益寿的基础上展现出由里及表的“容光焕发”与“白里透红”,进一步提高人们健康生活质量。
发明内容
针对现有技术的缺陷或不足,本发明提供了一种涉及左、右旋丹参素及其药学上可接受的盐以及其酯在抗衰老及美白中的应用,具体涉及右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯在单独或者作为有效成分在制备抗衰老及皮肤美白、祛斑和治疗色素沉着性疾病以及食品添加剂中的用途。
左、右旋丹参素及其药学上可接受的盐以及其酯类衍生物(结构通式I)的生物活性。
结构通式(I)中:
*碳为手性碳,可为R型或S型。
R选自氢、烷基、环烷基、杂烷基、杂环基、芳基、芳烷基、杂芳基或杂芳烷基。
药学上可接受的盐包括:Na+、K+、Mg2+、Zn2+、Ca2+、Cu2+、Fe2+等。
本发明的理论基础是血管生成不足、炎症、氧自由基损害是衰老与黑色素生成的临床病理表现,是造成衰老与黑色素生成的重要原因。改善血管新生、抗炎、抗氧自由基以及抑制黑色素生成可有效抗衰老与美白。
发明人采用斑马鱼的两种模型说明通式(I)化合物具有促进血管新生的作用。一是通过强效VEGF受体抑制剂(PTK787)诱导斑马鱼节间血管生成不足模型,模拟疾病血管生成不足的病理状态,研究了通式(I)化合物促进血管新生的功效,结果表明:部分左旋丹参素及其药学上可接受的盐以及其酯以及右旋丹参素及其药学上可接受的盐以及其酯与阳性药丹红注射液比较,可明显促进PTK787抑制的斑马鱼节间血管的生长而没有明显毒副作用。二是通过斑马鱼肠下静脉血管新生实验证明:部分左旋丹参素及其药学上可接受的盐以及其酯以及右旋丹参素及其药学上可接受的盐以及其酯类衍生物具有促血管新生的作用。
另一方面,本发明还采用斑马鱼与小鼠的抗炎模型分别研究左旋丹参素及其药学上可接受的盐以及其酯以及右旋丹参素及其药学上可接受的盐以及其酯类衍生物的抗炎活性。证明部分左旋丹参素及其药学上可接受的盐以及其酯以及右旋丹参素及其药学上可接受的盐以及其酯类衍生物具有显著的抗炎活性。
另一方面,本发明还采用细胞模型分别研究部分右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯类衍生物的抗氧自由基活性。证明部分右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯类衍生物具有显著的抗氧自由基活性。
另一方面,本发明还采用斑马鱼模型研究部分右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯类衍生物的抗黑色素生成活性。证明部分右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯类衍生物具有显著的抑制黑色素生成活性。
基于上述研究结果完成了本发明。本发明的研究表明部分右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯类衍生物至少通过抗炎、抗氧化、促血管新生活性以及抑制黑色素实现抗衰老和美白,可单独或者作为有效成分在制备抗衰老及皮肤美白、祛斑和治疗色素沉着性疾病药物、保健品、日化用品(如化妆品、洁肤用品等)以及食品添加剂中的用途。
本发明中,右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯类衍生物可以和药品、保健品、食品或日化用品上允许的任意一种辅料制成组合物,也可以和其他与右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯类衍生物不发生拮抗作用的治疗物组成复方制剂。这些制剂可以是药学上允许的任意一种类型,包括但不限于慕斯、喷剂、霜、乳剂、冻干粉剂以及液体制剂等。部分右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯类衍生物的用量为1-2000μM,可以根据使用方式、使用者年龄、体重、体表面积,可以一次或者多次使用。
本发明的突出优点是对比右旋丹参素及其药学上可接受的盐以及其酯与左旋丹参素及其药学上可接受的盐以及其酯类衍生物的促血管生成、抗炎、抗氧自由基、抑制黑色素生成等活性,并进一步肯定部分右旋丹参素及其药学上可接受的盐以及其酯以及左旋丹参素及其药学上可接受的盐以及其酯的抗衰老与美白活性,其效果好,毒性低,具有广泛的应用前景。

以下实施例所用药物均为市售产品。
实施例1:左、右旋丹参素及其酯对斑马鱼的毒性作用研究
1.实验方法:
挑选发育良好的3dpf AB系斑马鱼,斑马鱼在无菌24孔板中培养,对待测化合物(R-DSS、R-DSS-Na、S-DSS、S-DSS-Na、R-IDHP、S-IDHP)进行耐受性评价,给药浓度分别是50、100、250、500、1000、1500、2000、2500、5000、8000μmol/L(溶剂为水);各孔溶液体系终体积为2mL,待加药完毕后,将15条幼鱼加入至相对应的孔中,加盖并标记;
加药完毕后将培养板放置于光照培养箱中,每隔4h观察一次,记录死亡数量并移除死亡幼鱼,统计于给药24h后终止,实验重复三次。LC50在1000μmol/L以上属于低毒物质。
2.实验结果:
由图1可知,R-DSS:LC50>5000μmol/L;R-DSS-Na:LC50>5000μmol/L;S-DSS:LC50>5000μmol/L;S-DSS-Na:LC50>5000μmol/L;R-IDHP:LC50=3040μmol/L;S-IDHP:LC50=3129μmol/L。由此说明,R-DSS及其钠盐,S-DSS及其钠盐,R-IDHP与S-IDHP属于低毒性化合物,在1000μmol/L以下对斑马鱼模式动物无毒性。
实施例2:左、右旋丹参素及其酯对PTK787诱导斑马鱼节间血管生成不足模型的血管影响研究
1.原理:
血管新生是指在已有血管结构的基础上长出新血管的生物学过程,是一个涉及多种细胞、基质及细胞因子的复杂过程。其中血管内皮细胞是循环血液与血管平滑肌的机械屏障,在缺血性疾病的治疗中,促进内皮细胞的增殖,重建缺血区微循环,促进血管新生是重要的治疗手段之一。斑马鱼在发育起始阶段,存活不依赖血液循环,不会像哺乳动物那样由于循环缺失导致胚胎死亡。因此,可用瓦他拉尼(PTK787)诱导斑马鱼血管缺失,进而筛选血管再生促进剂。
2.实验方法:
2.1促进节间血管生成活性评价:
正式实验前进行成鱼交配产卵,在收集受精卵6h后,按照30μL/mL的比例加入苯基硫脲(PTU,1mg/mL),以抑制黑色素生长,保证鱼体透明,避免造成统计结果误差。在模型稳定建立后,在显微镜下挑选发育良好的24hpf Tg(fli1-EGFP)系斑马鱼胚胎,转移至5mL离心管中,加入1mg/mL脱膜剂(链蛋白酶E)进行脱膜,并用新鲜养鱼水清洗三次。使用无菌24孔板,实验设置正常对照组、模型组、阳性组(丹红注射液)以及不同浓度(25、50、100μmol/L)的R-DSS、S-DSS、R-IDHP、S-IDHP、R,S-IDHP、R,R-DBZ(10μmol/L)、S,R-DBZ(10μmol/L)给药组。本实验采用造模药物与治疗药物共同处理,即除空白组外,其余所有组都要加入0.175μg/mL的PTK787,最终各孔溶液体系终体积为2mL。将10条幼鱼移入相对应的孔中,加盖并标记,转移至光照培养箱中,培养箱温度控制在28.5℃±0.5℃。在给药处理过程中,每隔4h观察一次,移除因偶然误差死亡的幼鱼,避免造成水体污染影响统计结果。待其发育24h后使用荧光显微镜对每条鱼进行观察,采集图像。为了让统计结果更加准确,幼鱼需保证在两眼重合状态下采集图像,使用三卡因对其麻醉。使用IPP 5.1进行节间血管总长度的测量,实验重复三次。
2.2促进肠下静脉血管生成活性评价:
实验采用3dpf的发育良好的Tg(fli1-EGFP)转基因品系斑马鱼,在收集受精卵的6h后按照30μL/mL的比例加入PTU(1mg/mL),以抑制黑色素生长,保证鱼体透明,避免造成统计结果误差。使用无菌24孔板,实验设置正常对照组、不同浓度(25、50、100μmol/L)的R-DSS、S-DSS、R-IDHP、S-IDHP、R,S-IDHP、R,R-DBZ(10μmol/L)、S,R-DBZ(10μmol/L)给药组。空白对照组加入DMSO,其体积与最高浓度相同,最终各孔溶液体系终体积为2mL。将15条幼鱼移入相对应的孔中,加盖并标记,转移至光照培养箱中。待其发育24h后使用荧光显微镜对每条鱼进行观察,采集腹部肠下静脉的图像。由于肠下静脉为网篮形状,呈45°角,因此需使用三卡因进行麻醉和甲基纤维素进行固定。最后使用IPP 5.1对图像中的斑马鱼进行肠下静脉总长度进行测量,实验重复三次。
3.实验结果:
3.1促进节间血管生成活性评价结果
图2结果表明:R-DSS、S-DSS、R-IDHP、S-IDHP、R,S-IDHP、R,R-DBZ、S,R-DBZ均有显著的促进血管新生活性。R-DSS、S-DSS与其相应钠盐活性无显著性差异。
3.2促进肠下静脉血管生成活性评价结果
图3结果表明:R-DSS、S-DSS、R-IDHP、S-IDHP、R,S-IDHP、R,R-DBZ、S,R-DBZ均有显著的促进血管新生活性。R-DSS、S-DSS与其相应钠盐活性无显著性差异。
实施例3:左、右旋丹参素及其酯在细胞层面的抗氧化作用研究1.材料与方法
1.1实验细胞
该实施例中使用的细胞为人肝细胞WRL68,购于上海富衡生物科技有限公司。WRL68细胞使用DMEM培养液,培养液中含有10% FBS和1%青霉素、链霉素混合物,于37℃5% CO2条件下培养。
1.2实验药品与仪器
S-DSS、R-DSS、S-IDHP和R-IDHP,DMEM培养基和胰蛋白酶-EDTA(0.25%)及PBS购自美国HyClone公司;培养细胞用的胎牛血清FBS购自澳洲Noverse公司;棕榈酸(palmiticacid,PA)、油酸(oleic acid,OA)、牛血清白蛋白(BSA)购自美国Sigma公司,TG、TC测定试剂盒购自南京建成生物公司。二氧化碳培养箱(Thermofisher,BB150);倒置显微镜(Olympus,CKX53);流式细胞仪;酶标仪(Thermo Fisher,MK3)
1.3实验分组
正常培养的细胞为对照组(Control);PAOA诱导的细胞为实验组(PAOA);PAOA诱导细胞脂肪化后,分别加入不同药物为干预组(S-DSS、R-DSS、S-IDHP和R-IDHP)。
1.4实验方法
1.4.1 CCK-8法检测PAOA及各种化合物对细胞活力的影响
各细胞以5 000个每孔的密度接种至96孔板培养24h,分别加入不同浓度的PAOA(PA、OA以摩尔比1:2配制)或不同浓度的S-DSS、R-DSS、S-IDHP和R-IDHP(由PBS配制),每组6个复孔,培养24h后每孔加入100μL的CCK8工作液,培养箱内孵育1h,设定酶标仪于450nm处测定各孔的吸光度值并计算细胞存活率。
1.4.2细胞内ROS水平检测
收集经处理的细胞,加入ROS工作液,37℃避光孵育30min后,离心弃去染色液并加入PBS洗涤细胞,于Ex/Em=488nm/525nm下使用流式细胞仪检测。
1.4.3数据统计分析
使用GraphPad prism 9.3.1软件进行数据处理及绘图,计量资料采用均数±标准误表示,组间差异比较采用单因素方差分析。
2.结果
2.1化合物浓度筛选
检测不同浓度化合物干预下的细胞存活率,由图4(A、B、C)可知,S-DSS、R-DSS、S-IDHP在100μmol·L-1及以下浓度时细胞活力无明显降低,结合前期研究发现R-IDHP在100μmol·L-1对细胞无明显毒性,因此后续实验选用100μmol·L-1的浓度作为干预脂肪化细胞的最适浓度。
2.2细胞内ROS含量
通过流式细胞术检测人肝细胞WRL68内ROS的含量(图4D),结果发现,PAOA诱导后ROS荧光强度显著增强,而经S-DSS、R-DSS、S-IDHP和R-IDHP干预后,ROS水平显著降低,说明四种化合物可抑制脂肪化细胞内ROS的产生,具有显著的抗氧化性。
实施例4:左、右旋丹参素及其酯在小鼠耳肿胀模型上的抗炎作用研究
1.材料与方法
1.1实验动物
昆明系小鼠,SPF级,体质量(20±2)g,雄性,购于河南斯克贝斯生物科技股份有限公司,许可证号:SCXK(豫)2020-0005。实验前在本实验室适应性饲养3d,室温20~24℃。
1.2实验药品与仪器
二甲苯溶液(批号:20230414,国药集团化学试剂有限公司);吲哚美辛(批号:I26251LL31,天津希恩思生化科技有限公司);电子分析天平(XS105DU,梅特勒-托利多科技有限公司),打孔器、手术剪、移液枪、灌胃针。
1.3实验方法
1.3.1实验动物的选择与分组
KM雄性小鼠140只,(20±2)g,适应性喂养3天,随机分成14组,每组10只。实验药物分低、中、高剂量组进行试验。模型组灌胃等体积的生理盐水作空白对照,吲哚美辛作阳性对照药物。实验分组及给药剂量见表1。

1.3.2化合物对小鼠耳肿胀的影响
1.3.2.1给予抗炎药物
分组后的实验动物适应性喂养3天后,每日灌胃给药1次,每次灌胃体积为10mL/kg即0.1mL/10g,连续给药4天。模型组灌胃等体积的生理盐水作空白对照。
1.3.2.2建立小鼠耳廓肿胀急性炎症模型
末次给药(第4天)30min后,在小鼠右耳正反两表面用移液枪各滴加20μL二甲苯,均匀涂抹。左耳不做任何处理,作平行对照。
1.3.2.3取耳片计算肿胀度
造模1h后,将小鼠脱颈椎处死,沿耳廓基部剪下两耳。对齐后用直径6毫米的打孔器在左右耳的同一部位打下圆形耳片。
用电子天平称重,以左右两耳片重量之差衡量肿胀程度,以肿胀率和抑制率衡量抗炎作用强度。
肿胀度(mg)=右耳片重(肿胀耳)-左耳片重(对照耳)。
肿胀率(%)=肿胀度/左耳片重(对照耳)×100%
肿胀抑制率(%)=(模型组平均肿胀度-给药组平均肿胀度)/模型组平均肿胀度×100%。
1.3.3统计方法
各组实验数据结果以(x±s)表示,采用SPSS20.0软件对数据进行方差分析,组间比较采用t检验,以P<0.05为差异有统计学意义。
2.结果
表2实验组和对照组小鼠耳廓肿胀度
由表2可以得出,在小鼠耳廓肿胀抗炎实验中,R-DSS、S-DSS、R-IDHP、S-IDHP与空白对照比均有显著的抗炎活性。
实施例5:左、右旋丹参素及其酯在斑马鱼模型上的抗炎作用研究
采用硫酸铜诱导的炎症模型评价化合物的抗炎活性。
1.原理:
炎症是免疫系统对组织损伤和感染的一种反应,主要特点就是白细胞(粒细胞、巨噬细胞)会聚集在感染组织周围。斑马鱼的免疫系统非常类似于哺乳动物。创伤发生时,中性粒细胞、巨噬细胞对创伤性炎症几乎同时作出应答,中性粒细胞迁移速度快,先募集到损伤部位,随后巨噬细胞才到达。数小时后,炎症开始消退,巨噬细胞和中性粒细胞离开损伤部位。硫酸铜损伤斑马鱼神经丘(斑马鱼体表侧线器的末稍器官),造成神经丘细胞死亡,斑鱼的中性粒细胞发生免疫应答,聚集在神经丘周围,吞噬死亡的细胞。
2.实验方法:实验采用3dpf的发育良好的Tg(Lyz:EGFP)转基因品系斑马鱼,在收集受精卵的6h后按照30μL/mL的比例加入PTU(1mg/mL)。使用无菌24孔板,实验设置正常对照组、模型组、阳性组(白藜芦醇(res)和吲哚美辛(Indomethacin))及不同浓度(25、50、100、150μmol/L)的R-DSS、S-DSS、R-IDHP、S-IDHP、R,S-IDHP给药组,每组3个重复孔,每孔10条斑马鱼。其中正常对照组与模型组不加药,阳性组及待测化合物给药组用各剂量化合物与斑马鱼共孵育3h后,除对照组外,其余各组用20μmol/L硫酸铜处理1h制备炎症模型。随后使用荧光显微镜对每条鱼进行观察,采集图像。为了让统计结果更加准确,幼鱼需保证在两眼重合状态下采集图像,使用三卡因对其麻醉。使用软件IPP 5.1计算躯干部荧光炎症粒细胞迁移个数,实验重复三次。
3.实验结果:
由图5可知,R-DSS、S-DSS、R-IDHP、S-IDHP、R,S-IDHP均有显著的抗炎活性。在较高浓度(100μmol以上)下,R-DSS、S-DSS、R-IDHP、S-IDHP、R,S-IDHP均较阳性药白藜芦醇与吲哚美辛抗炎活性好。
实施例6:左、右旋丹参素及其酯在斑马鱼模型上抑制黑色素生成活性研究
本实验按照《化妆品黑色素抑制测试-斑马鱼胚胎测试方法》(T/SHRH 036—2021)进行。
1.原理:
正常人的皮肤颜色主要由黑色素决定,通常说的色斑、色沉也与黑色素有关。目前认可的黑色素形成途径主要为酪氨酸在酶催化下经过多巴氧化等过程,最终形成黑色素。由于斑马鱼拥有与人类相似的皮肤结构,黑色素的形成机理也与人类高度相似。而且斑马鱼的幼鱼身体透明,表皮的黑色素易于观察,因此可以用来观察产品的美白淡斑效果。
2.实验方法:
实验采用AB系斑马鱼,在显微镜下挑选发育良好的24hpf AB系斑马鱼胚胎,转移至5mL离心管中,加入1mg/mL脱膜剂(链蛋白酶E)进行脱膜,并用新鲜养鱼水清洗三次;使用无菌24孔板,实验设置正常对照组、阳性组(苯硫脲、曲酸)以及不同浓度(50、200、2000μmol/L)的R-DSS、S-DSS、R-IDHP、S-IDHP、R,S-IDHP给药组;除正常对照组外,其余各组加入不同浓度的化合物共孵育24h,每孔10条斑马鱼,每组3个重复孔,最终各孔溶液体系终体积为2mL。加完药后加盖并标记,转移至光照培养箱中,培养箱温度控制在28.5℃±0.5℃,在给药处理过程中,每隔4h观察一次,移除因偶然误差死亡的幼鱼,避免造成水体污染影响统计结果。待其发育至48h后使用荧光显微镜对每条鱼进行观察,采集图像。使用IPP 5.1进行斑马鱼头部色素的测量统计,实验重复三次。
3.实验结果:
结果表明:R-DSS、S-DSS、R-IDHP、S-IDHP、R,S-IDHP均有显著的抑制黑色素生成活性,且效果显著优于阳性药曲酸。
Claims (3)
1.左、右旋丹参素、其药学上可接受的盐以及其酯在制备抗衰老、祛斑和美白药物、保健品、食品或日化用品中的应用,所述丹参素结构式如式(I)所示。
式(I)中:
*碳为手性碳,可为R型或S型;
所述左、右旋丹参素结构式如(I)所示,且R选自氢;
所述左、右丹参素酯的结构式如(I)所示,且R选自烷基、环烷基、杂烷基、杂环基、芳基、芳烷基、杂芳基或杂芳烷基。
2.根据权利要求1所述的应用,其特征在于,丹参素药学上可接受的盐包括:Na+、K+、Mg2 +、Zn2+、Ca2+、Cu2+或Fe2+盐。
3.根据权利要求1-2所述的应用,其特征在于,结构通式(I)化合物的有效浓度为1-2000μM。
