Abstract
The invention discloses a traditional Chinese medicine composition for improving the semen quality of a male duck, and a preparation method and application thereof, wherein the active ingredients of the traditional Chinese medicine composition are prepared from the following components: angelica sinensis, dangshen, aconite, cinnamon, prepared rehmannia root, semen cuscutae, shizandra berry, dwarf lilyturf tuber, largehead atractylodes rhizome and liquorice. The composition provided by the invention has the effects of improving the sexual desire of the male ducks, promoting the secretion of the male ducks semen, improving the quality of the male ducks semen and enhancing the reproductive performance, can obviously improve the quality of the male ducks semen in the production and breeding work of the breeding ducks, improves the use value of the breeding ducks, and improves the breeding benefit and the breeding efficiency.

s1: weighing the components according to the formula, and crushing;
s2: soaking cortex Cinnamomi powder, distilling, and collecting distillate;
s3: dissolving radix Aconiti lateralis powder with hydrochloric acid, filtering, extracting the filtrate with organic solvent, collecting acid water layer, adjusting pH of acid water layer to 9-10, extracting with organic solvent, and collecting alkali water layer;
s4: soaking the powder of other components except cortex Cinnamomi and radix Aconiti lateralis in acidic alcohol solution, reflux extracting, and filtering to obtain filtrate and residue; recovering ethanol from the obtained filtrate, and regulating pH to 6.5-7.5; decocting the residue with water, and filtering to obtain filtrate;
s5: mixing the distillate obtained in step S2, the alkali water layer obtained in step S3, the filtrate obtained in step S4 after the pH adjustment, and the filtrate obtained after decoction of the residues, adjusting the pH to 6.5-7.5, concentrating, adsorbing the obtained concentrate with silicon dioxide, and drying to obtain the final product.

The beneficial effects are that: compared with the prior art, the invention has the following advantages:
(1) In the preparation process, the alkaloid components in the aconite are extracted by adopting stepwise extraction and adopting dilute hydrochloric acid ethanol solution, and other effective components are extracted by water, so that the extraction yield of the effective components is improved; the silica adsorption drying process is adopted, so that the drying time is shortened, the drying temperature is reduced, and the product flowability is increased.
(2) The composition provided by the invention has the effects of warming yang and warming kidney, tonifying kidney and securing essence, and tonifying qi and nourishing blood, can obviously improve the sexual desire of the male ducks, promote the semen secretion of the male ducks, improve the semen quality of the male ducks, enhance the reproductive performance, promote the seed value of the male ducks, fully exert the genetic advantages of the excellent male ducks, improve the breeding benefit and the breeding efficiency, and has obvious economic benefit and social benefit.

Detailed Description
The experimental methods described in the following examples are all conventional methods unless otherwise specified; the reagents and materials, unless otherwise specified, are commercially available.
Example 1
The formula of the components comprises: according to the weight portions, 20g of angelica, 40g of dangshen, 20g of aconite, 20g of cinnamon, 20g of prepared rehmannia root, 20g of semen cuscutae, 20g of shizandra berry, 20g of dwarf lilyturf tuber, 20g of bighead atractylodes rhizome and 10g of liquorice are respectively weighed, screened and cleaned.
The preparation process comprises the following steps:
(1) Weighing the raw materials according to the parts by weight; pulverizing the above materials into coarse powder of about 2 mm;
(2) Soaking cortex Cinnamomi powder in water at a mass ratio of 1:6 for 2 hr, extracting by steam distillation for 8 hr, and collecting distillate;
(3) Dissolving radix Aconiti lateralis powder in hydrochloric acid of 0.1mol/L at a mass ratio of 1:5, and filtering. The obtained filtrate was treated with the same volume of CHCl 3 Extracting, collecting acid water layer, adjusting pH to 10 with ammonia water, and adding CHCl with the same volume 3 Extracting, and collecting an alkali water layer for later use;
(4) Adding 0.05mol/L dilute hydrochloric acid 50% ethanol solution into the powder of other components, soaking for 60 minutes, carrying out reflux extraction at 77-79 ℃ for 2 hours, wherein the mass ratio of the solvent to the raw materials is 6:1, filtering to obtain filtrate and dregs, extracting the filtrate to recover ethanol, decocting the obtained dregs in water, keeping micro-boiling for 2 hours, filtering, combining the filtrates, and regulating the pH value to 7.0 by using sodium hydroxide;

The preparation process is the same as in example 1.
Example 3
The formula of the components comprises: according to the weight portions, 30g of angelica, 60g of dangshen, 30g of aconite, 30g of cinnamon, 30g of prepared rehmannia root, 30g of semen cuscutae, 30g of shizandra berry, 30g of dwarf lilyturf tuber, 30g of bighead atractylodes rhizome and 15g of liquorice are respectively weighed, screened and cleaned.
The preparation process is the same as in example 1.
Example 4: clinical verification
1. Test drug example 1 the water-soluble Chinese herbal compound extracted.
2. The test animals are provided by a certain duck farm in Nanjing, and the breeding mode is single cage breeding. 120 breeding ducks with healthy age and close weight are selected at 210 days.

The male ducks on the 1 st day of the test drink water freely, no medicine is added, and the administration is carried out by drinking water according to requirements for 14 consecutive days from the 2 nd day, and then the administration is stopped and the free drinking water is carried out for 3 days. Semen and venous blood of the test ducks are manually collected on days 1, 15 and 18 in the test period, and relevant indexes of each group of test ducks are measured. The measurement index is shown in Table 2.

The test results showed that the traditional Chinese medicine composition of example 1 can improve the libido (figure 2) and semen quality of the male ducks to some extent in the administration stage and a period of time after stopping administration compared with the control group. As can be seen from table 3, the semen of each group of the duckling is slightly alkaline, the pH value of the duckling semen in the administration stage and the dosing-stopping stage is slightly higher than that in the non-administration stage, but the difference is not obvious, if the pH value is lower, the motility of sperms can be influenced, and the traditional Chinese medicine composition has no adverse effect on the pH value of the semen; the traditional Chinese medicine composition in the embodiment 1 can obviously increase the semen quantity of the drake, and compared with the drug administration stage without drug administration, the average semen quantity of the drake in the drug administration stage and the drug withdrawal stage is increased by 0.551mL and 0.494mL; compared with the non-administration period, the sperm motility and the activation rate of the male ducks can be respectively increased by 20.88 percent and 25.02 percent, and the effect is still obvious in the stopping period; compared with the non-administration phase, the sperm density of the male ducks in the administration phase and the stopping phase is obviously improved, and the sperm density is respectively increased by 16.27 hundred million/mL and 15.80 hundred million/mL; the sperm malformation rate in the administration stage and the stopping stage is obviously reduced by 14.47 percent and 14.41 percent respectively, and the malformation rate in the stopping stage is still kept low.
The reproductive hormones in hypothalamus-pituitary gland-gonadal axis are closely related to the regulation of the reproductive function of the male ducks, testosterone (T), luteinizing Hormone (LH), follicle Stimulating Hormone (FSH) and the like are directly involved in spermatogenesis and maintenance, and as shown in table 4, the serum testosterone, luteinizing hormone and follicle stimulating hormone content of the male ducks are significantly increased in the administration phase compared with the non-administration phase, and the higher hormone level is still significantly maintained in the withdrawal phase, thereby promoting the normal sufficient production of sperm of the male ducks and affecting sperm quality.
The results show that: the invention discloses a traditional Chinese medicine composition for improving the semen quality of a male duck, which has the advantages of quick effect and obviously improved semen quality after the male duck is used, thereby playing a role in improving the reproductive performance of the male duck. Because the product is a pure traditional Chinese medicine, the use is safe and reliable, and the product can be widely applied to livestock and poultry cultivation.

s3: dissolving radix Aconiti lateralis powder with hydrochloric acid, filtering, extracting the filtrate with organic solvent, collecting acid water layer, adjusting pH of acid water layer to 9-10, extracting with organic solvent, and collecting alkali water layer;
s4: soaking the powder of other components except cortex Cinnamomi and radix Aconiti lateralis in acidic alcohol solution, reflux extracting, and filtering to obtain filtrate and residue; recovering ethanol from the obtained filtrate, and regulating pH to 6.5-7.5; decocting the residue with water, and filtering to obtain filtrate;
s5: mixing the distillate obtained in step S2, the alkali water layer obtained in step S3, the filtrate obtained in step S4 after the pH adjustment, and the filtrate obtained after decoction of the residues, adjusting the pH to 6.5-7.5, concentrating, adsorbing the obtained concentrate with silicon dioxide, and drying to obtain the final product.
