本文使用的术语“抗体”包括保留结合特异性的多聚(例如四聚)以及单域抗体以及抗体片段。本文所述的抗体可以由一种或多种多肽组成,所述多肽基本上由免疫球蛋白基因或免疫球蛋白基因片段编码。已被识别的免疫球蛋白基因包括κ、λ、α、γ、δ、ε和μ恒定区基因,以及大量免疫球蛋白可变区基因。轻链被归类为κ或λ。重链被归类为γ、μ、α、δ或ε,这进而确定了免疫球蛋白类别,分别为IgG、IgM、IgA、IgD和IgE。在一些实施方式中,抗体是IgG(例如,IgG1、IgG2、IgG3、IgG4)、IgM、IgA、IgD或IgE。
一种示例性免疫球蛋白(抗体)的结构单元包含四聚体。各四聚体包含相同的两对多肽链,每对包含一条“轻”链(约25kD)和一条“重”链(约50-70kD)。每条链的N末端确定约100至110个或更多个氨基酸构成的可变区,所述可变区主要负责抗原识别。术语可变轻链(VL)和可变重链(VH)分别指这些轻链和重链。
有许多已确定特征的抗体片段。因此,例如,胃蛋白酶消化铰链区中二硫键C端的四聚抗体,以产生(Fab)’2,其是Fab的二聚体,所述Fab本身是通过二硫键接合到VH-CH1的轻链。可在温和条件下还原F(ab')2以打断铰链区中的二硫键,从而将F(ab')2二聚体转化为Fab'单体。Fab’单体本质上是具有部分铰链区的Fab(参见Fundamental Immunology,W.E.Paul编,Raven Press,N.Y.(1993)以获得其他抗体片段的更详细描述)。虽然根据对完整抗体的消化定义了多种抗体片段,但是本领域技术人员会理解,这类片段也可利用化学方法或重组DNA方法从头合成。因此,本文使用的术语抗体还包括通过修饰完整抗体产生或使用重组DNA方法合成的抗体片段。
除了本文提供的特异性抗体序列外,也可以是抗体取代变体。取代变体可以去除至少一个氨基酸残基,并在其位置插入不同的残基。取代诱变最感兴趣的位点包括高变区,但也考虑了框架的改变。
通过选择取代产生抗体生物学特性的实质改变,所选取代在维持以下方面的效果上差异显著:(a)取代区域中多肽骨架的结构,例如β-片层或螺旋构型,(b)靶位点处分子的电荷或疏水性,或者(c)侧链体积。基于共同的侧链特性将天然残基分组如下:
(1)非极性:正亮氨酸,Met,Ala,Val,Leu,Ile;
(2)极性不带电:Cys,Ser,Thr,Asn,Gln;
(3)酸性(带负电):Asp,Glu;
(4)碱性(带正电):Lys,Arg;
(5)影响链取向的残基:Gly,Pro;和
(6)芳族:Trp,Tyr,Phe,His。
非保守性氨基酸取代可通过将一组的成员换成其他组的成员来产生。
一种可产生的取代类型是将抗体中的一个或多个可能为化学反应性的半胱氨酸换成其他残基,例如但不限于换成丙氨酸或丝氨酸。例如,可以存在非经典半胱氨酸的取代。取代可以在可变结构域的CDR或框架区或抗体的恒定区中进行。在一些实施方式中,半胱氨酸是经典的(例如参与二硫键的形成)。任何不参与维持抗体正确构象的半胱氨酸残基也可以被取代,通常用丝氨酸取代,以提高分子的氧化稳定性并防止异常交联。相反,可以将半胱氨酸键添加到抗体中以提高其稳定性,特别是当抗体是抗体片段如Fv片段时。
抗体可以包括VH-VL二聚体,包括单链抗体(作为单条多肽链存在的抗体),例如单链Fv抗体(sFv或scFv),其中可变重区和可变轻区连接在一起(直接或通过肽接头)以形成连续多肽。单链Fv抗体是共价连接的VH-VL,其可以由包括直接连接或通过编码肽的接头连接的VH-和VL-编码序列的核酸表达(例如,Huston等人Proc.Nat.Acad.Sci.USA,85:5879-5883,1988)。当VH和VL作为单条多肽链彼此连接时,VH和VL结构域非共价结合。或者,抗体可以是另一个片段。也可以产生其他片段,例如使用重组技术,作为可溶性蛋白或作为从展示方法获得的片段。抗体也可以包括双抗体和小抗体。如上所述,抗体还包括单域抗体,例如重链二聚体,例如来自骆驼的抗体。
如本文所用,“嵌合抗体”是指免疫球蛋白分子,其中(a)恒定区或其一部分被改变、取代或交换,从而抗原结合位点(可变区)连接到不同或改变的类、效应功能和/或物种的恒定区,或赋予嵌合抗体新特性的完全不同的分子,例如,酶、毒素、激素、生长因子、药物等;或(b)可变区或其一部分被具有不同或改变的抗原特异性的可变区或部分;或具有来自另一物种或来自另一抗体类或亚类的相应序列改变、取代或交换。

“表位”或“抗原决定簇”是指抗体与之结合的抗原上的部位。表位既可以由连续氨基酸形成,也可以由蛋白质三级折叠并列的非连续氨基酸形成。由连续氨基酸形成的表位通常在暴露于变性溶剂时保留,而由三级折叠形成的表位通常在用变性溶剂处理时丢失。表位通常包含呈现独特空间构象的至少3个、更通常至少5个或8-10个氨基酸。确定表位空间构型的方法包括,例如,x射线晶体学和二维核磁共振。见例如分子生物学方法中的表位映射方案,第66卷,Glenn e.Morris编(1996)。
本文使用的术语“价态”是指抗体对于一种抗原的不同结合位点的数量。单价抗体包含对抗原的一个结合位点。多价抗体包含多个结合位点。
短语“特异性(或选择性地)结合”抗原或靶,或“特异性(或选择性地)与……免疫反应”,当指蛋白质或肽时,指抗体结合到感兴趣的抗原或靶的结合反应。在本公开的上下文中,抗体以比其对其他抗原(例如CD20作为一个例子)的亲和力大至少100倍的KD结合人CD94病毒。
术语“相同”或“相同性”百分数,在两个或多个多肽序列的上下文中,指在比较窗口或指定区域上进行比较和比对以获得最大对应时,两个或多个相同的序列或子序列,在指定区域具有相同(例如,至少70%、至少75%、至少80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%或更高)种类的特定氨基酸残基百分比。用于确定氨基酸序列相同性百分数的比对可以使用公开可用的计算机软件如BLAST-2.0进行。BLAST和BLAST 2.0算法在Altschul等人,Nuc.Acids Res.25:3389-3402(1977)和Altschul等人,J.Mol.Biol.215:403-410(1990)中描述。因此,BLAST 2.0可以与默认参数一起使用,以确定序列相同性百分数。

具体实施方式
发明人已经确定,自然杀伤细胞靶向并可以杀死高度活化的T细胞。虽然这在许多情况下是有益的(例如,防止过度活跃的免疫反应),但在一些实施方式中,效果不理想。例如,在一些实施方式中,期望用T细胞靶向个体中的癌细胞或其他不需要的物质。在这些方面,耗竭个体中的自然杀伤细胞以允许靶向癌细胞抗原的活化T细胞(例如CD8+效应T细胞)靶向并杀死个体中的癌症细胞可能是有利的。因此,在一些实施方式中,给予患有癌症的个体抗CD94抗体,从而耗竭自然杀伤细胞,从而通过T细胞增强对个体中癌症细胞的靶向,否则这些细胞会被自然杀伤细胞去除。该方法可以进一步包括给予一种或多种检查点抑制剂。因此,可以诱导对癌症增强的免疫反应。在一些实施方式中,给予优先靶向自然杀伤细胞的抗体或抗-CD94抗体可以伴随或跟随或后随有给予抗原,所述抗原活化针对一种或多种癌抗原的T细胞。
本发明人还发现,可以通过(i)给予个体足以瞬时耗竭自然杀伤细胞的量的NK特异性靶蛋白特异性抗体(例如,抗CD94特异性抗体),以及(ii)将非自身细胞或组织移植到个体以减少个体中对非自身细胞或组织的NK细胞介导的免疫反应。非自身细胞或组织的表达最初可以在给予抗体(例如抗CD94抗体)之前或之后发生。示范性非自身细胞或组织可以包括但不限于嵌合抗原受体(CAR)T细胞(包括但不限于修饰为表达非自身CAR的自体或同种异体T细胞)或表达异源T细胞受体的T细胞(包含但不限于自体或同种异体T细胞)。

在某些情况下,将CDR转移到人框架会导致人源化抗体的特异性丧失。在这些情况下,可以将回复突变引入抗体人类部分的框架区。制造回复突变的方法描述于例如,Co等人,PNAS USA 88;2269-2273(1991)和WO 90/07861中。
在一些实施方式中,抗体是抗体片段,例如Fab、F(ab’)2、Fv或scFv。抗体片段可以使用本领域已知的任何手段产生,包括化学消化(例如木瓜蛋白酶或胃蛋白酶)和重组方法。分离和制备重组核酸的方法是本领域技术人员已知的(参见Sambrook等人,MolecularCloning.A Laboratory Manual(2d ed.1989);Ausubel等人,Current Protocols inMolecular Biology(1995))。抗体可在多种宿主细胞中表达,包括大肠杆菌、其他细菌宿主、酵母、和多种高级真核细胞如COS、CHO和HeLa细胞系和骨髓瘤细胞系。
竞争性结合测定可用于鉴定与本文所述的抗体竞争与例如CD94的特异性结合的抗体。本领域已知的多种竞争性结合测定中的任何一种都可以用于测量两种抗体对同一抗原的竞争。简单说,测试不同抗体抑制另一种抗体结合的能力。例如,抗体可以通过使用夹心ELISA测定法由与其结合的表位来区分。这是通过使用捕获抗体来涂覆孔表面进行的。然后将亚饱和浓度的标记的抗原添加到捕获表面。这种蛋白质将通过一种特定的抗体:表位相互作用与抗体结合。洗涤后,将已共价连接到可检测部分(例如HRP,其中标记的抗体被定义为检测抗体)的第二抗体加入ELISA。如果该抗体识别与捕获抗体相同的表位,则其将不能与靶蛋白结合,因为该特定表位将不可再结合。然而,如果该第二抗体识别靶蛋白上的不同表位,则其将能够结合,并且这种结合可以通过使用相关底物量化活性水平(由此获得抗体结合)来检测。背景通过使用单个抗体作为捕获和检测抗体来确定,而最大信号可以通过用抗原特异性抗体捕获和用针对抗原上标签的抗体检测来建立。通过使用背景和最大信号作为参照,可以成对方式评估抗体以确定表位特异性。
使用任何上述测定,如果第二抗体与抗原的结合在第一抗体存在下减少至少30%,通常至少约40%、50%、60%或75%,并且通常至少约90%,则第一抗体被认为竞争性抑制第二抗体的结合。
在一些实施方式中,抗体(例如,抗CD94抗体)与毒素(例如,细胞毒素)连接,使得抗体与NK细胞(例如,表达CD94的细胞)结合并杀死它们。示例性毒素可以包括但不限于鹅膏蕈碱、蒽环类药物、奥瑞他汀、巴卡丁类药物、加利车霉素、喜树碱类药物、西马多丁、秋水仙素(colchicines)、秋水仙胺(colcimids)、考布他汀(combretastatin)、隐藻素、圆皮海绵内酯、多卡霉素类药物、棘霉素类、刺五加素类、埃坡霉素类、雌莫司汀类、来红毒素类、美登木素生物碱类、纺锤菌素类、嘌呤霉素类、二聚吡咯并苯并二氮杂类、根瘤菌素类、紫杉烷类、微管蛋白裂解素类和长春花生物碱类。毒素可以通过接头,例如本领域已知的接头与抗体连接。参见例如欧洲专利申请EP3165237A1。在一些实施方式中,毒素通过抗体中(例如在Fc区)天然存在或工程改造的半胱氨酸残基与抗体连接。
在一些实施方式中,由于延长其血液半衰期的部分或氨基酸序列的修饰或融合,抗体可以具有增加的半衰期。在一些实施方式中,抗体是PEG化的或包含至少一个结合位点,用于结合血清蛋白(如血清白蛋白)或其他部分(尤其是至少一个氨基酸序列),这增加了抗体的半衰期。例子包括但不限于:血清白蛋白(如人血清白蛋白)、血清免疫球蛋白如IgG,或转铁蛋白;Fc部分(例如人Fc)或其合适的部分或片段;或一种或多种可与血清蛋白结合的小蛋白或肽,例如但不限于WO 91/01743、WO 01/45746、WO 02/076489、WO 08/062820、WO 09/127691和WO 11/095545中描述的蛋白质和肽。

在一个方面,本文所述的一种或多种抗体被配制为药物组合物,例如,以治疗有效量使用适合治疗癌症或用于减少或预防对移植到个体中的细胞或组织的免疫反应的给药方案。所述组合物可配制成适用于多种药物递送系统。
药物组合物可包括药学上可接受的运载体。药学上可接受的运载体部分地由所给予的特定组合物以及用于给予组合物的特定方法确定。因此,有多种合适的药物组合物制剂(参见例如,《雷明顿药物科学》(Remington’s Pharmaceutical Sciences);第17版,1989)。

在本方法和组合物的背景下,给予患者的剂量应足以随着时间的推移在对象中产生有益的反应。任何患者的最佳剂量水平将取决于多种因素,包括所使用的特定调节剂的效力、患者的年龄、体重、生理活动和饮食,以及与其他药物的可能组合。剂量大小也可由伴随具体对象的特定化合物或载体给药的任何不良副作用的存在、性质、和程度来决定。
在确定要给予的活性成分(例如抗体)的有效量时,医生可以评估活性成分的循环血浆水平及其毒性。通常,对于典型的对象,抗体的剂量当量为约1ng/kg至10mg/kg。给药可以通过单次给药或分次给药来完成。
组合物可以定期(例如每天)给药一段时间(例如2、3、4、5、6天或1-3周或更长时间)。可以使用本领域已知的任何途径,包括例如通过注射(例如静脉内、腹膜内、皮下、肌肉内或皮内)、吸入或透皮应用,将组合物直接给予哺乳动物对象以耗竭自然杀伤细胞。

在一些实施方式中,引入个体的细胞包含由异源核酸表达的异源蛋白。将核酸引入细胞的合适方法的非限制性实例包括电穿孔(例如,核转染)、病毒或噬菌体感染、转染、偶联、原生质体融合、脂质转染、磷酸钙沉淀、聚乙烯亚胺(PEI)介导的转染、DEAE-葡聚糖介导的转导、脂质体介导的转基因、粒子枪技术,磷酸钙沉淀、直接显微注射、纳米粒子介导的核酸递送等。在一些实施方式中,编码蛋白质的多核苷酸通过载体递送至细胞。例如,在一些实施方式中,载体是病毒载体。示例性病毒载体可以包括但不限于腺病毒载体、腺相关病毒(AAV)载体和慢病毒载体。
目前,CAR T细胞产品是由自体T细胞制成的。这些被批准用于临床的CAR T细胞必须以定制为基础产生。由于生产过程昂贵且漫长,这种根据个别案例的自体T细胞生产平台仍然是大规模临床应用的重要限制因素。还有生产失败的内在风险。个体化、定制的自体CAR T细胞生产过程也限制了其在不同肿瘤类型上的广泛应用。因此,需要通用同种异体T细胞来制备通用CAR T细胞,该细胞可以作为大规模临床应用的“现成”即用治疗剂。NK细胞的瞬时耗竭可以实现通用CAR T细胞的移植。如果同种异体T细胞已经工程改造(例如,通过CRISPR)以消除其HLA I类蛋白的表达——通过使b2微球蛋白基因失活或通过使HLA I类重链基因如HLA-A、HLA-B、HLA-C或其他多态性HLA I类基因失活——则耗竭宿主中的NK细胞是特别有用的,因为NK细胞优先杀死HLA I类阴性细胞。在一些实施方式中,NK细胞的耗竭通过删除能够杀死高度活化的自体T细胞的NK细胞来增强患者中自体CAR T细胞的扩增。
在一些实施方式中,在给予抗NK细胞抗体(例如,抗CD94抗体)以及随后耗竭成熟NK细胞后,新产生的NK细胞可以变得对转移的同种异体细胞或组织,例如HLA I类阴性细胞或组织具有耐受性。
任选地,引入动物体内的细胞(例如CAR T-细胞或其他T细胞)可以被修饰,例如但不限于通过CRISPR,以使这些细胞中的CD94(KLRD1)基因失效,从而避免在活化的CAR T细胞已经获得CD94表达的情况下,抗-CD94抗体对引入的细胞的任何潜在毒性作用。

实施例
实施例1
CD94是NK细胞表面的一种膜蛋白,在活化的NK细胞中上调。为了产生针对CD94的抗体,我们在哺乳动物细胞中表达作为Fc-融合蛋白的CD94的外结构域,并使用该蛋白与经过充分验证的合成Fab噬菌体文库进行噬菌体展示选择。产生并验证了两种重组单克隆抗体(JLS002-rAB22和JLS002-rAB23)。它们的CDR序列测定如下:
这两种抗体在大肠杆菌和哺乳动物细胞中都具有高亲和力和良好的表达和生产能力。产生了两种抗体的IgG和Fab(片段抗原结合结构域)。它们与Ba/F3细胞上表达的CD94和CD94-NKG2C复合物表现出良好的结合,表明它们作为治疗抗体和研究试剂组分的潜力。
1.Fc融合蛋白的表达和纯化
CD94是II型膜蛋白。我们产生了一种生物素化Fc融合蛋白,其CD94的胞外结构域融合到人IgG1-Fc的C端(下文称为“Fc-CD94”)。为了产生足够的与我们的噬菌体展示选择兼容的蛋白质,我们对载体进行了优化,以提高蛋白质表达和纯化的效率。在我们的实验中,在Expi293细胞中瞬时转染后,从30ml哺乳动物细胞培养物中纯化出100-130μg具有约90%生物素化的Fc-CD94蛋白。
2.选择
我们对Fc-CD94融合蛋白进行了直接选择。在图1中显示了每一轮选择的富集度。
在第3轮和第4轮选择中,共产生了54个命中。通过测序证实了两个具有独特序列的Fab噬菌体。表达并纯化了这两个Fab。通过多点ELISA测定结合亲和力。结合曲线如图2A-B所示,这表明这两种Fab具有良好的亲和力。
3.IgG的亲和力
使用octet测定法来测量IgG对两种结合剂JLS002-rAB22和JLS002-rAB23的亲和力。见图3。使用OctetRED384(ForteBio)仪器来测量IgG对两种结合剂JLS002-rAB22和JLS002-rAB23的亲和力。将Fc-CD94融合蛋白固定在涂有链霉亲和素的生物层干涉生物传感器上。用含有10μM的生物素的PBST缓冲液(PBS+0.5Tween)封闭后,加载测试的IgG 15分钟,然后换到不含IgG的PBST缓冲液中约30分钟。记录动力学结合曲线。
用编码人CD94的逆转录病毒载体或用人CD94、人NKG2C和人DAP12稳定转导小鼠Ba/F3细胞,如Lanier,L.L.,B.C.Corliss,J.Wu,和J.H.Phillips.1998Association ofDAP12 with activating CD94/NKG2C NK cell receptors.Immunity 8:693-701.PMID:9655483中所述。将Ba/F3转染子与磷酸盐缓冲盐水(未染色)或rab22 JIS002 rab 22或JSI002 rab 23抗体一起孵育,洗涤,然后使用标准方法用PE-偶联的抗人IgG1染色。结果如图4A-B所示。
实施例2
NK细胞耗竭可提高α-PD-1L检查点阻断治疗RMA肿瘤生长的效力。在第0天对C57BL/6小鼠皮下给予1x 10e6 RMA淋巴瘤细胞。如图5A所示,实验设计包括在第0天和第7天用耗竭性抗-NK1.1单克隆抗体(克隆PK136)处理小鼠,该单克隆抗体结合小鼠自然杀伤细胞上的NK细胞特异性蛋白。在第3天、第4天、第5天、第7天和第10天,将α-PD-1L抗体(克隆10F.9G2)或对照大鼠IgG2b同种型匹配抗体给予小鼠。在指定的时间点腹膜内注射在盐水中的α-NK1.1、α-PD-1L或IgG2b同种型匹配的对照抗体,剂量为150μg、250μg、和250μg。在各种处理下,小鼠肿瘤体积随时间的测量。图5B提供了所得数据的图表,显示在检查点阻断α-PD-1L抗体处理的小鼠中,与对照相比,用抗-NK细胞抗体耗竭NK细胞显著减少了肿瘤体积。斯氏t检验的显著性为*P<0.05。

轻链可变区,其分别包含SEQ ID NO:10、SEQ ID NO:11和SEQ ID NO:12的轻链CDR1、CDR2和CDR3。

21.如权利要求20所述的方法,其中所述嵌合抗体是人源化抗体。

26.一种减少或防止自然杀伤细胞排斥个体中同种异体细胞或组织移植的方法,该方法包括:
在所述移植期间、之前或之后给予所述个体足以瞬时耗竭自然杀伤细胞的量的针对所述个体的抗自然杀伤细胞特异性抗体。

重链可变区,其分别包含SEQ ID NO:7、SEQ ID NO:8和SEQ ID NO:9的重链CDR1、CDR2和CDR3;和
轻链可变区,其分别包含SEQ ID NO:10、SEQ ID NO:11和SEQ ID NO:12的轻链CDR1、CDR2和CDR3。
