Abstract
The invention relates to a method for preparing a food, a health-care food or a medicine with the weight-losing effect by taking fresh potentilla anserina as a raw material and adopting a freeze-drying method to dry the fresh potentilla anserina. The freeze-dried potentilla anserina powder is characterized by obviously inhibiting the activities of alpha-glucosidase and lipase. Preliminary test feeding shows that the dry potentilla anserina has better weight-losing effect.
Description
Technical Field
The invention relates to a dry potentilla anserina with weight-losing effect, which is prepared from fresh potentilla anserina by adopting a sun-drying, drying or freeze-drying method. Belongs to the technical field of medicines.
The background technology is as follows:
obesity has been identified by the World Health Organization (WHO) as a disease that can cause a range of health problems, such as increased risk of various chronic diseases such as hypertension, diabetes, dyslipidemia, coronary heart disease, myocardial infarction, stroke, and partial tumors, as well as social and psychological problems. Obesity and related chronic diseases have become a major public health problem in China and the world.
The radix Potentillae Anserinae is root tuber of Potentilla anserina Potentilla anserina L, removing fibrous root, cleaning, removing sediment, and sun drying. The potentilla anserina is also called ginseng fruit and cactus fruit, is also a conventional medicinal material in Tibetan and belongs to a plant for both medicine and food. Sweet and neutral in nature and taste, and enters spleen meridian; has effects in invigorating spleen, invigorating stomach, promoting salivation, quenching thirst, invigorating qi, replenishing blood, and promoting diuresis. The potentilla anserina contains flavonoid, steroid, terpene, phenolic acid coumarin and other components. The flavonoid component contained in the plant can be classified into flavonoid, flavonols, flavan-3-ols and anthocyanidins. The aglycone is mainly quercetin (quercetin), kaempferol (kaempferol), luteolin (luteolin), myricetin (myricetin) and isorhamnetin (isohamnetin). Phenolic compounds are largely divided into cinnamic acids and benzoic acids. The water extract and the ethanol extract of the potentilla anserina are important active components of the potentilla anserina.
The potentilla anserina has better hypoxia tolerance, fatigue resistance and stress resistance; the water extract and the ethanol extract can obviously improve the immune function; the potentilla anserina saponin has anti-HBV effect. The daily application experience shows that the potentilla anserina extract has better weight-losing effect, and particularly has obvious effect of reducing abdominal fat and waistline. The daily observation shows that the silverweed cinquefoil root obtained by directly adopting the freeze-drying method has more obvious weight-losing effect than the silverweed cinquefoil root obtained by sun-drying, and the superiority of the freeze-dried silverweed cinquefoil root in the aspect of weight losing is also proved by comparing the inhibition rate of the freeze-dried silverweed cinquefoil root and the sun-dried silverweed cinquefoil root on alpha-glucosidase and pancreatic lipase.
The potentilla anserina is distributed in Shaanxi, gansu, ningxia, tibet, qinghai and inner Mongolia etc., but according to the description of Xinhua Ben Cao gang Chart, the potentilla anserina only grows on Qinghai-Tibet plateau, and the seed starts to grow with root tuber. The potentilla anserina used in the invention is produced by the Qinghai province, the Jersey county.
The invention comprises the following steps:
aiming at the problem that the freeze-dried potentilla anserina and the sun-dried potentilla anserina have great difference in weight loss, the invention is obtained by the following technical scheme.
1. Cutting fresh root tuber of Potentilla anserina Potentilla anserina L of Potentilla of Rosaceae, removing fibrous root, cleaning, and slicing into 2-4mm thick slices;
2. placing the fresh potentilla anserina root slabs in a drying oven of a freeze dryer, cooling to-30 ℃ to-40 ℃ and maintaining for 2-4 hours; simultaneously, cooling a condenser of the freeze dryer to-60 ℃ to-70 ℃;
3. and (3) opening vacuum pumping, slowly raising the temperature of the drying box, controlling the temperature to be raised to 0 ℃ for 12-16 hours, continuously raising the temperature to 35-40 ℃, maintaining the temperature for 6-8 hours, controlling the water content of the potentilla anserina slices to be below 5%, and crushing the potentilla anserina slices into coarse powder.
The invention has the following advantages
The freeze-dried potentilla anserina coarse powder has more obvious effect than the potentilla anserina obtained by the traditional sun-drying method when being used as the raw material of food, health-care food or medicine with the weight-losing effect.
Detailed Description
The present invention will be further illustrated with reference to the following examples, but the present invention is not limited to the following examples.
Example 1 the lyophilized water extract of potentilla anserina contained more active ingredient than the sun-dried water extract of potentilla anserina.
The potentilla anserina contains saponins, flavonoids and multi-classified components. More and more researches show that the flavonoid components and the polyphenol components have the effects of inhibiting alpha-glucosidase and pancreatic lipase, reducing sugar and fat absorption and playing a role in losing weight. For this purpose. Comparing the total flavone content and total polyphenol content of the water extracted extracts of freeze-dried potentilla anserina and sun-dried potentilla anserina, the specific method is as follows:
respectively decocting 100g of radix Potentillae Anserinae with water for 2 hr and 8 times of water for 1 hr respectively, mixing decoctions, filtering, concentrating the filtrate, and fixing volume to 200 ml. The content of total flavonoids in the extracting solution is measured according to the method under the pagodatree flower item of the Chinese pharmacopoeia of 2020 edition, and the result is shown in table 1; the total phenolic acid content of the extract was determined according to the method for determining total phenolic acid in foods, and the results are shown in Table 1.

Note that: the total flavone content refers to the amount of rutin equivalent per 1g of dry potentilla anserina (without calculating moisture); total polyphenols refer to the amount of comparable gallic acid per 1g of dry potentilla anserina (without accounting for moisture)
As can be seen from table 1, the content of total flavonoids and total phenolic acids in lyophilized potentilla anserina is significantly higher than in sun-dried potentilla anserina, which may be related to both aspects; on the one hand, the freeze drying can freeze water in the cells of the fresh potentilla anserina, increase the volume, lead the cells to swell and split, and is beneficial to the extraction of the components; on the other hand, it may be associated with instability of the total flavonoids and total phenolic acids, and freeze-drying cannot destroy the components.
Example 2 the inhibition of the aqueous lyophilized potentilla anserina extract to alpha-glucosidase and pancreatic lipase was higher than that of the aqueous sun-dried potentilla anserina extract
Inhibiting the activity of alpha-glucosidase and pancreatic lipase, reducing sugar and fat absorption is an important target for weight loss. The polyphenols and flavonoids have good inhibitory effect on two enzymes. For this reason, the inhibition of two enzymes by two water extracts of potentilla anserina were compared. The specific method comprises the following steps:
decocting radix Potentillae Anserinae with water for 2 times, mixing the extractive solutions, concentrating to obtain concentrate corresponding to 0.5g radix Potentillae Anserinae/ml, and determining the inhibition ratio of the two concentrates to alpha-glucosidase and pancreatic lipase. The method for measuring the inhibition rate of alpha-glucosidase adopts p-nitrophenyl-alpha-glucopyranoside (PNPG) as a substrate, and the alpha-glucosidase can be decomposed into glucose and p-nitrophenol (PNP) to measure PNP; the method for detecting the inhibition rate of pancreatic lipase adopts a method for detecting the PNP content of a product by using 4-nitrobutyrate (PNPB) substrate and decomposing the substrate into p-nitrophenol (PNP) by porcine pancreatic lipase. The results are shown in Table 2.

From table 2 it can be seen that the inhibition rate of the lyophilized potentilla anserina against both enzymes is significantly higher than that of the sun-dried potentilla anserina.
Example 3 lyophilized aqueous extract of Potentilla anserina significantly reduced weight gain in obese rats
The fat rats are modeled by high-fat feed, and the two potentilla anserina water extracts are administrated by stomach irrigation for 30 days, so that the weight increase of the rats can be obviously reduced by freeze-dried potentilla anserina compared with sun-dried potentilla anserina. The results are shown in Table 3.

Example 4 human trial feeding shows that the freeze-dried potentilla anserina has better weight-losing effect
The potentilla anserina belongs to a Chinese medicine for both medicine and food. When the human body tries to eat, the dosage of the tea is 25-50 g each time according to the specification of Sichuan province Chinese herbal medicine standard, and the tea is replaced by tea. The observation time is 60 days, and the weight-losing effect is judged according to waistline before and after taking. The criteria for the reference test diets were: volunteers and center obese persons (waistline men more than or equal to 90cm and women more than or equal to 85 cm). Greater than the results are shown in Table 4.

From the observation results, the waistline of the freeze-dried potentilla anserina and the sun-dried potentilla anserina can be reduced, and the freeze-dried potentilla anserina has more obvious effect than the sun-dried potentilla anserina.
Claims (3)
1. A method for preparing lyophilized radix Potentillae Anserinae powder with weight reducing effect is provided. The method is characterized by comprising the following three steps:
(1) Digging fresh radix Potentillae Anserinae tuberous root, removing fibrous root, cleaning silt, and cutting into thick slices (2-4 mm);
(2) Placing the fresh potentilla anserina slices in a drying oven of a freeze dryer, cooling to-30 ℃ to-40 ℃ and maintaining for 2-4 hours; the condenser is cooled to-60 to-70 ℃. Vacuum pumping is started, the drying box is slowly heated, the temperature is raised to 0 ℃ and controlled to be 12-16 hours, the temperature is continuously raised to 35-40 ℃ and maintained for 6-8 hours, and the product is obtained. The water content of the dry potentilla anserina tablet is controlled below 5%.
2. (3) Pulverizing lyophilized radix Potentillae Anserinae tablet into powder.
3. According to the requirement of claim 1, the prepared freeze-dried potentilla anserina powder can be directly taken or substituted for tea, and has better weight-losing effect. Therefore, can be used as raw materials for preparing weight-losing foods, health-care foods or medicines.
