还提出了常规的基于脂质体的药物递送系统以改善DFO的生物分布。这由Lang等提出(ACS Nano,2019,13,2176-2189)。它们还在该脂质体中将DFO与HIF1α(低氧诱导因子1α)抑制剂组合以限制HIF1α的过表达,这通常用DFO观察到。脂质体内的包封使得可获得约100纳米的纳米颗粒。啮齿动物模型中的体外和体内临床前试验使得可能证明抗肿瘤作用。然而,由于纳米颗粒的尺寸,观察到肝脏和脾脏中的高度积聚。
遵循相同的推理,M.Theerasilp et al.(RSC Advances,2017,7,11158)提出将不同的铁螯合剂包封在聚合物胶束内。这些胶束由具有形成胶束核心的疏水部分和提供胶体稳定性的亲水部分的聚合物形成。这些胶束在不同细胞系中表现出抗癌活性,大小为约25nm,且基于螯合剂和基于pH表现出盐析。
从所有这些研究中可清楚地看出,目前没有溶液能够有效和特异性地靶向肿瘤,以及用于治疗肿瘤的内源性铜和/或铁的充分局部螯合,特别是通过以大约mg/l范围的有效浓度施用纳米颗粒。

根据本公开的纳米颗粒可通过用于制备纳米颗粒的胶体溶液的方法获得,所述纳米颗粒包含接枝到聚合物基质的螯合基团,仅一部分螯合基团与金属阳离子络合,另一部分未络合,所述方法包括:
(1)合成或提供作为起始材料的下式[Ch2]n-PS的纳米颗粒NP1的胶体溶液,其中:

根据本公开的纳米颗粒的药物制剂
包含根据本公开的纳米颗粒的组合物以纳米颗粒的胶体悬浮液的形式施用。它们可以如本文所述或根据本领域技术人员已知的其他方法制备,且根据治疗和待治疗的区域,通过不同的局部或全身途径施用。
因此,本公开涉及如前述部分中所述的式[Ch1]n-PS-[Ch2]m的纳米颗粒的胶态悬浮体,以及药物组合物,所述药物组合物包含这些胶态悬浮体的,合适的情况下,与一种或多种药学上可接受的赋形剂组合。
药物组合物可以特别地配制成用于静脉内注射的冻干粉末或水溶液的形式。在一个优选实施方案中,所述药物组合物包含具有治疗有效量的纳米颗粒的胶体溶液,所述纳米颗粒为如前述部分所述的式[Ch1]n-PS-[Ch2]m,特别是基于与钆螯合的聚硅氧烷的纳米颗粒,且更具体地,从如上所述的AGuIX纳米颗粒获得。
在一些实施方案中,其为每小瓶包含200mg-15g,优选250mg-1250mg纳米颗粒的冻干粉末。所述粉末还可包含其他赋形剂,特别是CaCl2。
冻干粉末可在水溶液(典型地是无菌注射用水)中重构。因此,本公开涉及药物组合物,其用于注射溶液,其包含作为活性成分的如前述部分所述的式[Ch1]n-PS-[Ch2]m的纳米颗粒,特别是基于聚硅氧烷的与钆螯合的纳米颗粒,且更具体地从如上所述的AGuIX纳米颗粒获得。

纳米颗粒的用途
由于存在游离的或与金属阳离子M1络合的螯合基团Ch1,根据本发明的纳米颗粒能够在将它们施用于有需要的受试者后捕获内源性铜和/或铁。内源性铜捕获优选意指内源性铜的局部捕获量在100ppb(0.1mg铜/升)与10000ppb(10mg铜/升)间。因此,本公开更特别地靶向用于捕获受试者中的内源性铜的方法,特别是患有癌症的受试者。

如果所述纳米颗粒静脉内或肺内施用,则所述铜和/或铁可在全身血液循环中被捕获,然后,之后,在纳米颗粒已在肿瘤中积累后,在肿瘤内被捕获,特别地通过与EPR效应相关的被动靶向。被动靶向肿瘤和累积的这种作用已特别地通过AGuIX型超细纳米颗粒清楚地证明。
因此,本公开还涉及用于治疗受试者中的肿瘤的方法,所述方法包括向所述受试者施用有效量的如前述部分所述的式[Ch1]n-PS-[Ch2]m的纳米颗粒的药物组合物,特别是基于聚硅氧烷的与钆螯合的纳米颗粒,且更特别地,所述纳米颗粒从如上所述的AGuIX纳米颗粒获得,且其特征在于,所述纳米颗粒能够部分地通过捕获内源性铜和/或铁来治疗肿瘤。
在所述方法的一个具体实施方案中,所述组合物还包含有效量的金属阳离子M2,优选钆或铋,用作放射增敏剂,且所述方法包括在施用所述组合物后,用有效剂量照射受试者以通过放疗治疗肿瘤的步骤。
“患者”或“受试者”优选是指哺乳动物或人,包括例如患有肿瘤的受试者。

纳米颗粒的“有效量”是指如上所述的纳米颗粒的量,当施用于患者时,足以位于肿瘤中并能够通过捕获内源性铜和/或铁来治疗肿瘤,适当情况下与放射增敏作用和放疗治疗组合。
该量基于诸如受试者的年龄、性别和体重的因素来确定和调节。
如上所述的纳米颗粒可以以瘤内、皮下、肌内、静脉内、皮内、腹膜内、口服、舌下、直肠、阴道、鼻内,通过吸入或通过透皮给药来施用。优选肿瘤内和/或静脉内施用。
在施用纳米颗粒作为放射增敏剂后用于治疗肿瘤的照射方法是本领域技术人员熟知的且已在以下出版物中具体描述:WO2018/224684,WO2019/008040和C.Verry et al.,Science Advances,2020,6,eaay5279;and C.Verry et al.,“NANO-RAD,a phase I studyprotocol”,BMJ Open,2019,9,e023591。
放疗期间的总照射剂量将根据癌症类型、阶段和待治疗的受试者进行调整。对于治疗剂量,实体瘤的典型总剂量约为20-120Gy。可以考虑其他因素,诸如通过化疗的治疗,并发症和/或放疗在手术干预前或后进行的情形。总剂量通常分成几部分。根据本公开的方法中的放疗步骤可以例如包括,每天2-6Gy的几次,例如每周5天,且特别地连续2-8周,总剂量可在20-40Gy,例如30Gy。

具体实施方式
材料和方法
CuPRiXx产品通过将由Nh TherApuix(France)提供的起始产品引入到强酸性介质中而获得,所述介质获自来自CarlRoth的37%超纯氢氯酸。
采用Vivaflow手册中所述的条件,用蠕动泵和Sartorius Stedim Biotech(France)的Vivaflow/>盒进行过滤步骤。
用Malvern Instruments(USA)的Zetasizer Nano-S(633nm He-Ne激光)进行流体动力学直径的测量和等电点的滴定。为测量等电点,将该装置与Malvern Instruments(USA)的MPT-2自动滴定器偶联。
用具有DAD检测器的Agilent 1200进行HPLC-UV。使用的反相柱是Jupiter的C4,5μm,150x4.6mm。通过UV检测器在295nm的波长下进行检测。相A(H2O/ACN/TFA:98.9/1/0.1)和相B(H2O/ACN/TFA:10/89.9/0.1)的梯度如下:95/5,5分钟,然后10分钟内线性梯度,使得可达到10/90的比例,保持15分钟。在这15分钟结束时,A的含量在1分钟内恢复到95%,然后在95/5下,保持7分钟稳定水平(plateau)。洗脱剂相的组成中使用的产物都是经认证的HPLC级。
在Institut des Sciences Analytiques,UMR 5280,Pole Isotopes &Organique,5 rue de la Doua,69100 Villeurbanne进行元素分析。
用Perkin-Elmer(USA)的Nexion 2000进行HPLC-ICP/MS。使用具有以下组成的洗脱相以等度模式测量介质中的游离元素:95%A和5%B。相A和B的组成与HPL-UV方法相同。使用的反相柱是Jupiter的C4,5μm,150x4.6mm。洗脱剂相的组成中使用的产物都是经认证的HPLC级。
按照“初级干燥”程序,通过Christ(Germany)的Alpha 2-4LSC冻干机冻干颗粒。
在补充有10%胎牛血清(Dutscher)和1%青霉素-链霉素(GibcoTM,Thermofischer)的F12-K培养基(GibcoTM,Thermofischer)中培养A549细胞(ECACC86012804)。对于每个实验,将细胞在PBS 1X(GibcoTM,Thermofischer)中漂洗两次,在培养箱中,在37℃,5%CO2下用胰蛋白酶-EDTA孵育5分钟,然后置于完全培养基中。
在测试前至少1小时,将产物CuPRiX20和CuPRiX30以等于10mM的游离DOTA浓度(分别为30mM和16.7mM的钆)下,置于无菌蒸馏水中并储存在4℃。
对于迁移和侵袭实验,将A549细胞接种(50000个细胞/孔)在ImageLock 96孔板(Essen BioScience)中,并在37℃和5%CO2下孵育16小时直至达到90-100%汇合。使用WoundMakerTM(Essen BioScience)在每个孔的细胞单层中产生伤口。然后,用PBS 1X漂洗各孔两次。对于侵袭,向各孔中加入50μl Matrigel(Corning)-预先在含有或不含有CuPRiX20的F12-K培养基中稀释-终浓度为1mg/ml。将板在37℃下孵育30分钟,以实现Matrigel的聚合。最后,将100μl含有增加浓度的CuPRiX的培养基加入到每个孔(含有或不含有Matrigel)中。将板置于Incucyte(10x物镜)中,在含有CO2的培养箱中通过Zoom Incucyte软件(EssenBioScience)每2小时自动拍摄伤口填充的图像。通过软件分析数据并将结果表示为伤口的汇合百分比。
对于趋化性迁移实验,将A549细胞胰蛋白酶化、离心,然后重悬于耗尽培养基(depleted medium)(不含FCS的F12-K)中。随后将细胞接种(1000个细胞/孔,40μl/孔)在 Clearview insert(Essen BioScience)中。在相应的孔中,加入含有或不含有CuPRiX20(500μM游离螯合物)的20μl耗尽培养基。最后,将200μl含有10%FCS且含有或不含有CuPRiX20(500μM游离螯合物)的F-12K培养基加入趋化室的下部隔室中。每小时拍摄每个插入物的图像。与接种在膜上的细胞的初始汇合相比,从上层储库到下层储库的趋化迁移被量化为膜下的细胞汇合。使用IncuCyte ZOOM 2015A显微镜软件自动进行计算。
对于照射后的迁移实验,将A549细胞以20000个细胞/孔在孔培养板(Essen BioScience)中培养,在37℃,5%CO2下过夜。将细胞用含有或不含有CuPRiX30(500μM游离螯合物,相当于800μM钆)的不含FCS的F-12K处理24小时,然后以8Gy(X-Rad 320辐照器,250kV,15mA)照射。照射后,使用96孔WoundMakerTM(Essen BioScience)制备伤口。将细胞用PBS 1X漂洗两次以除去漂浮的细胞,并将100μl含有CuPRiX30(0和500μM游离螯合物,相当于约0和800μM钆)的培养基加入到每个相应的孔中。
为评价克隆源性存活,将细胞以40000个细胞/cm2,即1百万个细胞接种在25cm2烧瓶(Dutscher)中,并在37℃,5%CO2下孵育过夜。用含有或不含有CuPRiX30(500μM游离螯合物,相当于800μM钆)的不含FCS的培养基处理细胞24小时。然后,以不同剂量(0、2、3、4、6和8Gy)照射细胞。照射后,用PBS 1X洗涤细胞,胰蛋白酶化并计数。然后,将细胞重新接种在25cm2烧瓶中,并在固定和染色前生长六次分裂(7天)。用96%乙醇溶液(VWR)固定集落30分钟,然后用Giemsa溶液(Sigma-Aldrich)稀释至1/20染色30分钟。随后冲洗烧瓶,然后使用ColcountTM集落计数器(Oxford Optronix)数字计数含有64个或更多个细胞的集落。使用该方程式的线性二次模型测定克隆源性存活:其中SF是存活量,α和β分别表示致死和亚致死损伤的概率。
细胞系4T1是来源于BALB/c小鼠品系乳腺的乳腺癌细胞系。在补充有10%FCS和1%青霉素-链霉素的RPMI培养基(GibcoTM,Thermofischer)中培养细胞。
上呼吸消化道(UAT)癌细胞系SQ20B衍生自复发性喉癌。在使用流式细胞术进行两个连续的细胞分选(Hoechst流出和CD44标记)步骤后,收集目标亚群SQ20B/CSC(干细胞样癌细胞)。该群体具有低的EGFR表达和间质表型,与迁移和侵袭特性的获得相关。这些细胞SQ20B-CSC,在补充有5%FCS,1%PS,0.04mg/ml氢化可的松和20ng/ml表皮生长因子(EGF)的DMEM:F12培养基(3∶1,v∶v)中培养。
体内研究方案(2021021714569264)被Université Claude Bernard Lyon1的伦理委员会接受。该研究在三阴性乳腺癌小鼠模型(细胞系4T1)上进行。使用7周龄雌性BALB/c小鼠(Janvier Labs)。在异氟烷麻醉下将4T1细胞(5×105,基质胶50∶50)皮下注射到第4个乳房垫中。选择该位置是为了在放疗治疗期间保留重要组织。移植后10天,将小鼠随机分成2组:NaCl(对照)组(n=4)和CuPRiX组(n=6)。小鼠接受3次静脉内注射,每次50μl,间隔48h,每次注射NaCl 0.9%(对照)或CuPRiX(200mg/kg)。称重小鼠,评价其一般状况,每周测量肿瘤3次。最后一次注射后5天将小鼠安乐死,并在靶器官(肺和肝)中定量转移。
根据Pulaski等(2000)的方案定量转移。为此,在异氟烷麻醉下通过颈脱位使小鼠安乐死后,取出肺和肝。用HBSS洗涤器官,机械切割,然后用与用于肺的DNA酶I(Roche)结合的IV型胶原酶(2mg/ml)和用于肝脏的I型胶原酶(2mg/ml)、BSA(1mg/ml)和透明质酸酶(2mg/ml)混合物进行酶解。在37℃下在旋转轮上进行2小时(肺)和40分钟(肝)的酶解。消化的器官随后通过70μm尼龙过滤器过滤。将它们在1500G离心5分钟并用HBSS洗涤两次。将细胞沉淀重悬于含有60μM的6-硫代鸟嘌呤的RPMI培养基中,用于克隆生长研究。将悬浮液放回培养物中,在10cm培养皿中稀释至1/6用于肺,原悬浮液用于肝。8天后,将细胞用96%乙醇固定并用Giemsa(在蒸馏水中稀释至1/20)染色。随后使用ColcountTM菌落计数器(OxfordOptronix)对克隆进行数字计数。
实施例1:介质的酸化和Gd3+离子的盐析
为获得能够络合铜离子同时保持其作为放射增敏剂特性的纳米颗粒,将产物置于酸性介质中,目的是质子化DOTA基团并因此释放一部分初始络合的Gd3+离子。
首先,通过将10g产物溶于50ml超纯水中制备200g/l 溶液。将溶液在环境温度下搅拌1小时。同时,通过向50ml超纯水中加入10ml37%盐酸(37%盐酸,超纯,2.5L塑料包装,CarlRoth)制备2M盐酸溶液。
搅拌1小时后,将50ml 2M的盐酸溶液加入到50ml 中。然后,测量pH,其小于0.5。所得溶液呈橙棕色。将装置置于预热至50℃的烘箱中4小时。每小时取出样品以通过HPLC-ICP/MS观察钆离子的盐析(图1)。观察到在保留时间Tr=2.3min时介质中游离Gd3+的峰随反应时间增加。
实施例2:CuPRiX20:反应4h
4h后,用超纯水将溶液稀释10倍。然后,测量pH,如果需要,用1M氢氧化钠调节至1±0.2,以便不破坏过滤膜。通过蠕动泵和Sartorius Vivaflow 200-5kDa盒纯化由此获得的500ml溶液,以将颗粒与释放的Gd3+分离,从而防止这些离子重新络合。
将500ml的初始体积浓缩至50ml并重复该操作。总共重复4次稀释/浓缩操作,最终体积为50ml。纯化后,将50ml溶液分装在小瓶中,每个小瓶含有2ml溶液。将小瓶置于-80℃以冷冻溶液,然后冻干以获得棕色粉末形式的最终产物。
获得产物后,从该批次中取出一个小瓶以表征所述产物。通过加入超纯水制备新产物的1ml溶液(100g/l)。在溶液中1小时后,使用我们的DLS装置测量直径,表明直径为4.4nm±1.2nm。进行HPLC-UV/Vis色谱图,且指示保留时间为11min±0.1min,与原始颗粒相同。还测量了CuPRIX20的等电点,且其等于6.29,优于的等电点(等于7.15)。由于钆具有磁性,因此测量CuPRiX20的弛豫常数r1,且等于18.9mM-1.s-1/钆原子。
实施例3:通过铕螯合和荧光测定游离DOTAGA的数目
存在于CuPRiX20中的游离螯合物的量可通过铕的螯合以及随后的发光研究来确定。这是因为铕的发光主要集中在590(5D0→7F1)和615nm(5D0→7F2)。这种发光在水分子的存在下猝灭。该测定的原理是加入增加量的铕:随着铕被螯合时,发光增加,然后当所有螯合位点都充满时,发光达到稳定水平,如图2所示。
为进行测定,将CuPRiX20置于pH 5的乙酸盐缓冲液中;加入溶解在乙酸盐缓冲液中的氯化铕盐。然后,绘制测定曲线,在396nm处进行激发并在590nm处读取发射。该测定使得可计算出每mg CuPRiX20的螯合物的量为0.16μmol。假定初始产物中钆的含量为零或至多可忽略不计,则钆的初始量等于DOTA的量。通过元素分析测量,起始产物中的初始钆含量为每mg />为0.81μmol。
因此,产物CuPRiX20的20%的DOTA基团是游离的。
实施例4:铜螯合
因此,游离DOTA的存在表明CuPRiX20作为螯合剂在螯合疗法中的潜在应用。通过铜的螯合,随后使用HPLC-UV/Vis研究吸光度来测定CuPRiX20的络合物形成潜力。DOTA@Cu在295nm处的吸光度远大于溶液中DOTA@Gd络合物,DOTA和铜离子的吸光度。
CuPRiX20的吸光度将随着游离DOTA基团络合可用的铜离子而增加,直到其达到稳定水平,在该稳定水平添加额外的铜将不会导致吸光度的增加。为进行该实验,用增加量的氯化铜溶液和恒定量的CuPRiX20制备一系列样品。所有样品的体积均等。该实验使得可计算每mg CuPRiX20可络合铜的量为0.18μmol。在起始产物上进行的相同实验表明CuPRiX20的螯合潜力大幅增加(图3)。
实施例5:CuPRiX30:反应5h
游离DOTA的含量可基于AGuIX在酸性介质中的反应时间来改变。5h后,用超纯水将溶液稀释10倍。然后,测量pH,如果需要,用1M氢氧化钠调节至1±0.2,以免破坏过滤膜。通过蠕动泵和Sartorius Vivaflow200-5kDa盒纯化由此获得的500ml溶液,以将颗粒与释放的Gd3+分离,从而防止这些离子重新络合。将500ml的初始体积浓缩至50并重复该操作。
总共重复4次稀释/浓缩操作,且最终体积为50ml。纯化后,将50ml溶液分装在小瓶中,每个小瓶含有2ml溶液。将小瓶置于-80℃以冷冻溶液,然后冻干以获得棕色粉末形式的最终产物CuPRiX30。获得产物后,从该批次中取出一个小瓶以表征产物。
通过加入超纯水制备100g/l新产物的1ml溶液。在溶液中1小时后,使用我们的DLS装置测量直径,表明直径为5.7nm±1nn。进行HPLC-UV/Vis色谱图,且显示保留时间为10.8min±0.1min,与原始颗粒相同。
实施例6:CuPRiX30:与CuPRiX20相比改善络合物形成潜力
因此,游离DOTA的存在表明CuPRiX30作为可能的螯合剂在螯合疗法中的潜在应用。通过铜的螯合,随后使用HPLC-UV/Vis研究吸光度来确定CuPRiX20的络合潜力。DOTA@Cu在295nm处的吸光度远大于DOTA@Gd络合物,溶液中DOTA和铜离子的吸光度。随着游离DOTA基团络合可用的铜离子,产物的吸光度将增加,直到其达到稳定水平,在该水平添加额外的铜将不会导致吸光度增加。

实施例7:CuPRiX20对A549细胞运动性的影响的分析
细胞迁移是多步骤过程,是许多生物过程和疾病过程(包括肿瘤转移)的基本组成部分。在体外,伤口测试是基于细胞苔上伤口的形成和细胞运动性的研究,即细胞响应密度变化在表面上移动的能力,在该过程中闭合伤口。它是细胞在固体2D基质上的运动性的直接量度。
本实施例的目的是显示CuPRiX20降低细胞运动性的能力。在含有8nM的CuSO4·5H2O的F12-K培养基(Gibco)中培养A549细胞。在培养72小时后,在有或没有CuPRiX20(500μM游离螯合物)的情况下,将细胞胰蛋白酶化,然后以每孔40000个细胞接种在孔培养板(Essen Bioscience)中。将板在37℃,5%CO2下放置过夜。然后,使用/>(Essen Bioscience)制作伤口。将细胞用PBS冲洗两次以除去漂浮的细胞,然后用100μl含有增加浓度的CuPRiX20(0、50、100、200、300、400、500和1000μM游离螯合物,相当于约0、150、300、600、900、1200、1500和3000μM钆)的培养基处理。
在含有CO2的培养箱中通过Zoom Incucyte软件(Essen Bioscience)每2小时自动拍摄伤口填充的图像,持续72小时。通过软件分析数据,结果表示为伤口的汇合百分比。获得的结果使得可证明,由于使用不同浓度的CuPRiX20处理,A549细胞的细胞运动性减慢(图4、图5和图6)。
实施例8:CuPRiX20和CuPRiX30对A549细胞运动性的影响的比较
本实施例的目的是显示CuPRiX20和CuPRiX30降低细胞运动性的能力,并比较它们的效果。将A549细胞以40000个细胞/孔在孔培养板(Essen Bioscience)中培养,在37℃,5%CO2下过夜。使用96-孔/> WoundMaker(Essen Bioscience)进行伤口测试。将细胞用PBS冲洗两次以除去漂浮的细胞,然后用100μl含有CuPRiX20(0、125、250和500μM游离螯合物,相当于约0、375、750和1500μM钆)或CuPRiX30(0、125、250和500μM游离螯合物,相当于约0、200、400和800μM钆)的培养基处理。
在含有CO2的培养箱中通过Zoom Incucyte软件(Essen Bioscience)每2h自动拍摄伤口填充的图像。通过软件分析数据,结果表示为伤口的汇合百分比。
获得的结果可显示,在游离螯合物的恒定浓度下,两种类型的CuPRiX的效果是等同的。
实施例9:CuPRiX20对A549细胞侵袭的影响的分析
细胞侵袭是癌症的特征之一。它与细胞迁移有关并在转移的发展中起关键作用。肿瘤细胞形成转移的能力主要取决于细胞改变和重组其细胞形态和降解细胞外基质(ECM)的能力。体外侵袭试验基于伤口实验方法,但包括添加模拟ECM的凝胶基质。添加3D基质意味着细胞必须降解该基质以便移动。
本实施例的目的是显示CuPRiX20降低细胞侵袭的能力,即细胞分解细胞外基质和移动的能力。将A549细胞以40000个细胞/孔在孔培养板中培养,在37℃,5%CO2下过夜。然后,使用/> WoundMaker(Essen Bioscience)制作伤口,然后用PBS冲洗细胞两次。向各孔中加入50μl的Matrigel(Corning)——预先在含有或不含有CuPRiX20的F12-K培养基中稀释——终浓度为1mg/ml。将板在37℃下孵育30分钟,以实现Matrigel的聚合。最后,加入100μl含有或不含有CuPRiX20(0和500μM游离螯合物)的培养基。在含有CO2的培养箱中通过Zoom Incucyte软件(Essen Bioscience)每2小时自动拍摄伤口填充的图像,持续72小时。通过Zoom Incucyte软件(Essen Bioscience)分析数据,结果表示为伤口的汇合百分比。获得的结果使得:由于用CuPRiX20处理,可以证明A549细胞的细胞侵袭减缓(图7)。
实施例10:CuPRiX20对A549细胞的趋化性迁移的影响
趋化性迁移是细胞响应刺激的定向运动。该实验包括将培养插入物置于细胞培养板孔内。将细胞接种到插入物中,插入物含有确定孔径的膜,并含有无血清培养基以使细胞饥饿。将化学引诱剂介质置于下面的孔中。由于这种化学梯度,能够迁移的细胞被化学引诱剂介质吸引并穿过孔。本实施例的目的是使用趋化性测试显示CuPRiX20降低细胞迁移率的能力。为此,使用IncuCyte ZOOM趋化性模块。将A549细胞(1000个细胞/孔)再悬浮于含有0%FCS的F-12K培养基中,并接种于具有8μm孔的96孔细胞迁移Incucyte Clearview板的上部隔室中(40μl/孔)。在适当的孔中,加入20μl不含FCS的F12-K培养基,其含有或不含有CuPRiX20(500μM最终游离螯合物)。最后,将200μl含有10%FCS且含有或不含有CuPRiX20(500μM游离螯合物)的F-12K培养基加入趋化室的下部隔室中。每小时拍摄每个插入物的图像。从上层储库到下层储库的趋化迁移量化为与接种在膜上的细胞的初始汇合相比的膜下的细胞汇合。使用IncuCyte ZOOM 2015A显微镜软件自动进行计算。
所获得的结果可证明CuPRiX20对细胞的趋化性迁移大大降低(图8)。
实施例11:CuPRiX30和光子照射的组合对A549细胞运动性的作用
本实施例的目的是显示CuPRiX30在光子照射后降低细胞迁移率的能力。将A549细胞以20000个细胞/孔在孔培养板中培养,在37℃,5%CO2下过夜。将细胞与含有或不含有CuPRiX30(500μM游离螯合物,相当于800μM钆)的不含FCS的F-12K一起孵育24小时。随后以8Gy(X-Rad320辐照器,250kV)照射细胞,然后形成伤口。将细胞用PBS冲洗两次以除去漂浮的细胞,并将100μl含有CuPRiX30(0和500μM游离螯合物,相当于约0和800μM钆)的培养基加入到每个相应的孔中。
在含有CO2的培养箱中通过Zoom Incucyte软件每2小时自动拍摄伤口填充的图像。通过软件分析数据,结果表示为伤口的汇合百分比。所得的结果表明,与通过单独照射或单独使用CuPRiX30治疗相比,CuPRiX30/照射组合在限制运动性方面具有优越的功效(累加效应)(图9)。
实施例12:CuPRiX30和光子照射的组合对A549细胞存活的影响

实施例13:CuPRiX30和光子照射的组合对A549、SQ20B-CD44+和4T1细胞的存活的影响

实施例14:CuPRiX30在转移性乳腺癌小鼠模型中对肿瘤生长和转移形成的功效
本实施例的目的是显示CuPRiX30减缓肿瘤生长和减少转移瘤形成的能力。为此,给10只8周龄雌性BALB/c小鼠皮下注射50000个4T1细胞(三阴性乳腺癌细胞,50∶50PBS∶基质胶)。十天后(D10),当肿瘤平均达到100mm3时,给小鼠静脉注射50μl的CuPRiX30(200mg/kg,n=6)或NaCl(0.9%,n=4)。第一次注射后48小时(D12)和96小时(D14)再注射两次。肿瘤接种后19天,即最后一次注射后5天,在异氟烷麻醉下通过颈脱位使小鼠安乐死,并除去可能具有转移的器官(肺和肝)。
为量化转移,肺和肝被机械破碎,然后在过滤前进行酶解。随后在含有60μM的6-硫代鸟嘌呤(4T1细胞的选择剂)的培养基中培养纯的(肝)或稀释的(肺)细胞悬浮液,并在含有CO2的培养箱中在37℃下孵育。8天后,固定细胞并染色,自动计数集落;结果列于表1。

所获得的结果显示,在用CuPRiX30处理小鼠后,肿瘤生长减慢,且肺部转移数量减少。对照组中的4只小鼠有肺部转移,而由CuPRiX治疗的6只中的2只没有肺部转移。在两种情况下均未观察到肝转移(图12)。
实施例15:CuPRiX30和放疗的组合对转移性乳腺癌小鼠模型中的肿瘤生长和存活的功效
本实施例的目的是显示CuPRiX30增加放疗对肿瘤生长和存活的功效的能力。为此,给42只8周龄雌性BALB/c小鼠皮下注射50000个4T1细胞(三阴性乳腺癌细胞,50∶50PBS∶基质胶)。肿瘤移植后10天,将小鼠随机分成4组以接受治疗:NaCl组(对照,n=12),CuPRiX组(n=10),NaCl+放疗组(RT,n=9)和CuPRiX+RT组(n=11)。
共给小鼠注射3次的50μl CuPRiX(200mg/ml)或NaCl(0.9%),间隔48小时,并以5×2Gy的量(每天2Gy,共5天)与放疗组合(图13)。在异氟烷麻醉下,在施用治疗后1小时进行照射。
为监测肿瘤进展和建立Kaplan-Meier存活曲线,每周对小鼠称重并测量肿瘤6次。在治疗计划结束时,连续2天达到以下极限点之一的小鼠被安乐死:体重减轻15%;肿瘤体积为≥±1000mm3;持续性溃疡;观察到痛苦征象(虚脱、毛皮粗糙、弓背)。
如下计算肿瘤体积的演变:(时间t的肿瘤体积)/(参考时间的肿瘤体积),其中参考时间对应于治疗的第一天,即D10。所得结果显示,与单独用RT或单独用CuPRiX治疗后相比,用CuPRiX+RT组合治疗后肿瘤生长减少更多。与单独的RT相比,CuPRiX和RT的组合也可延长动物的存活时间(图14)。
工业实用性
本技术方案可以特别地应用于医药领域,特别地用于治疗肿瘤。
本公开不限于上述仅作为示例给出的实例,且其涵盖本领域技术人员在所寻求的保护的上下文中可以设想的所有变体。
Claims (10)
1.下式的纳米颗粒:
[Ch1]n-PS-[Ch2]m,其中:
-PS是有机或无机聚合物基质,
-Ch1是未络合的或与金属阳离子M1络合的螯合基团,
-M1不存在,或选自金属阳离子,所述金属阳离子与Ch1形成络合物的常数小于铜,特别地至少小十倍;例如,M1选自锌或碱土金属,特别是钙或镁,
-Ch2是螯合基团,其与螯合基团Ch1相同或不同,且与金属阳离子M2络合,所述M2具有大于40,优选大于50的高原子序数Z,
其特征在于,
(i)所述螯合剂Ch1和Ch2接枝到聚合物基质,
(ii)n/(n+m)比为10%-100%,并且
(iii)所述纳米颗粒的平均流体动力学直径为1-50nm,优选2-20nm,更优选2-8nm。
2.如权利要求1所述的纳米颗粒,其特征在于,至少50%的所述Ch1与锌、钙或镁络合。
3.如前述权利要求中任一项所述的纳米颗粒,其特征在于,所述螯合基团Ch1和适当情况下的Ch2选自大环试剂,优选选自1,4,7-三氮杂环壬烷三乙酸(NOTA)、1,4,7,10-四氮杂环十二烷-1,4,7,10-四乙酸(DOTA)、1,4,7-三氮杂环壬烷-1-戊二-4,7-二乙酸(NODAGA)和1,4,7,10-四氮杂环十二烷、1-(戊二酸)-4,7,10-三乙酸(DOTAGA)、2,2’,2”,2”’-(1,4,7,10-四氮杂环十二烷-1,4,7,10-四基)四乙酰胺(DOTAM)和1,4,8,11-四氮杂环十四烷(Cyclam)、1,4,7,10-四氮杂环十二烷(Cyclen)、去铁胺(DFO)。
4.如权利要求1-3中任一项所述的纳米颗粒,其特征在于,所述金属阳离子M2选自放射增敏剂和/或磁共振成像造影剂,特别是钆或铋。
5.如权利要求1-4中任一项所述的纳米颗粒,其特征在于,
(i)PS是聚硅氧烷基质,
(ii)Ch1和Ch2是下式(I)的DOTAGA螯合基团
且通过Si-C键合接枝到聚硅氧烷基质,
(iii)M1不存在,且M2是钆阳离子Gd3+,
(iv)n+m为5-50,优选为10-30,并且
(iv)所述平均流体动力学直径为2-8nm。
6.如权利要求1-5中任一项所述的纳米颗粒的胶体溶液。
7.药物组合物,其包含如权利要求1-5中任一项所述的纳米颗粒的胶体溶液,以及一种或多种药学上可接受的赋形剂。
8.如权利要求7所述的药物组合物,其特征在于,所述药物组合物是用于在受试者中静脉内、肿瘤内或肺内施用的可注射组合物,特别地包含有效量的螯合基团Ch1,用于在体内捕获肿瘤中的铜,所述游离螯合剂例如在所述组合物中的浓度为至少10mM。
9.如权利要求7和8之一所述的药物组合物,其用于在受试者中治疗癌症,特别地用于体内捕获肿瘤中的铜和/或铁。
10.如权利要求9所述的用于其用途的药物组合物,其特征在于,所述组合物包含有效量的金属阳离子M2,优选钆,用作放射增敏剂,且在施用所述组合物后通过放疗来治疗所述受试者。
