Abstract

Description
技术领域
本发明涉及生物医药技术领域,尤其涉及一种从多能干细胞通过诱导分化制备巨噬细胞的方法。
背景技术
巨噬细胞是一种存在于所有组织中的白细胞,它们既参与先天免疫(吞噬能力)又参与自适应免疫(细胞极化),而且参与个体发育,体内平衡和组织修复,具有显著的表型和功能多样性。
目前巨噬细胞研究主要使用外周血单核细胞衍生的巨噬细胞,不仅需要大量来自献血者的血液,并且相对难以进行基因编辑。利用诱导性多能干细胞(iPSCs)诱导产生的巨噬细胞(iMac)可以解决巨噬细胞的获取问题,并可利用先前构建的基因型特异性的iPSCs来衍生出核型正常和基因型稳定的巨噬细胞。这种方法为研究巨噬细胞特定的功能、免疫应答机制、活化和极化分子及巨噬细胞相关疾病或肿瘤治疗提供了良好的工具。
发明内容
本发明的目的在于提供一种制备周期短,纯度较高(>95%)的成熟的诱导的巨噬细胞,本发明通过包括以下步骤的方法实现了这一目的。
在一些实施方式中,本发明的方法包括:
(I)使多能干细胞分化为拟胚体,其中使多能干细胞分化为拟胚体的步骤包括:
I-1:在增补有干细胞因子(SCF)、骨形态发生蛋白4(BMP-4)和ROCK抑制剂的第一培养基中孵育所述多能干细胞;
I-2:在增补有干细胞因子(SCF)、骨形态发生蛋白4(BMP-4)、碱性成纤维细胞生长因子(bFGF)、血管内皮生长因子(VEGF)、胰岛素样生长因子1(IGF-1)、和血小板生成素(TPO)的第二培养基中孵育从步骤I-1获得的细胞;
(II)使所述拟胚体与一种或多种选自以下的细胞因子接触,从而得到所述巨噬细胞:胎肝激酶配体(Flt3L)、粒细胞-巨噬细胞集落刺激因子(GM-CSF)、巨噬细胞集落刺激因子(M-CSF)、血管内皮生长因子(VEGF)、干细胞因子(SCF)、胰岛素样生长因子1(IGF-1)、碱性成纤维细胞生长因子(bFGF)和白介素3(IL-3),其中使所述拟胚体与多种细胞因子接触的步骤包括:
II-1:在增补有bFGF、SCF、VEGF-A、IGF-1和Flt3L的第三培养基中孵育所述拟胚体;
II-2:在增补有SCF、VEGF-A、IGF-1、IL-3、M-CSF、GM-CSF和Flt3L的第四培养基中孵育从步骤II-1获得的细胞;
II-3:在增补有SCF、IL-3、M-CSF、GM-CSF和Flt3L的第五培养基中孵育从步骤II-2获得的细胞;
II-4:在增补有IL-3、M-CSF、GM-CSF和Flt3L的第六培养基中孵育从步骤II-3获得的细胞,从而得到所述巨噬细胞。
在一些实施方式中,所述多能干细胞是诱导的多能干细胞。
在一些实施方式中,所述ROCK抑制剂选自Y-27632、法舒地尔或H-1152,优选地,所述ROCK抑制剂是Y-27632,还优选地,所述ROCK抑制剂的含量选自5~15µM。
在一些实施方式中,所述第一培养基和所述第二培养基相同,并且/或者是含血清蛋白的无饲养层完全培养基。
在一些实施方式中,所述第三培养基、所述第四培养基、所述第五培养基和所述第六培养基相同,并且/或者是无血清培养基。
本发明还提供了一种用于使多能干细胞分化为巨噬细胞的试剂盒,所述试剂盒包括用于诱导多能干细胞分化为巨噬细胞的多种细胞因子,所述多种细胞因子包括胎肝激酶配体(Flt3L)、粒细胞-巨噬细胞集落刺激因子(GM-CSF)、巨噬细胞集落刺激因子(M-CSF)、血管内皮生长因子(VEGF)、干细胞因子(SCF)、胰岛素样生长因子1(IGF-1)、碱性成纤维细胞生长因子(bFGF)、白介素3(IL-3)、骨形态发生蛋白4(BMP-4)、ROCK抑制剂和血小板生成素(TPO),或者由胎肝激酶配体(Flt3L)、粒细胞-巨噬细胞集落刺激因子(GM-CSF)、巨噬细胞集落刺激因子(M-CSF)、血管内皮生长因子(VEGF)、干细胞因子(SCF)、胰岛素样生长因子1(IGF-1)、碱性成纤维细胞生长因子(bFGF)、白介素3(IL-3)、骨形态发生蛋白4(BMP-4)、ROCK抑制剂和血小板生成素(TPO)构成。
在一些实施方式中,所述试剂盒包括6个子试剂盒,其中第一子试剂盒包括干细胞因子(SCF)、骨形态发生蛋白4(BMP-4)和ROCK抑制剂,或由其构成;第二子试剂盒包括干细胞因子(SCF)、骨形态发生蛋白4(BMP-4)、碱性成纤维细胞生长因子(bFGF)、血管内皮生长因子(VEGF)、胰岛素样生长因子1(IGF-1)、和血小板生成素(TPO),或由其构成;第三子试剂盒包括bFGF、SCF、VEGF-A、IGF-1和Flt3L,或由其构成;第四子试剂盒包括SCF、VEGF-A、IGF-1、IL-3、M-CSF、GM-CSF和Flt3L,或由其构成;第五子试剂盒包括SCF、IL-3、M-CSF、GM-CSF和Flt3L,或由其构成;第六子试剂盒包括IL-3、M-CSF、GM-CSF和Flt3L,或由其构成。
在一些实施方式中,所述试剂盒中的ROCK抑制剂选自Y-27632、法舒地尔或H-1152。

“多能干细胞(PSC)”是指具有分化为构成一种或多种组织或器官,或优选地,三种胚层中的任何一种:内胚层(例如,内部胃内膜、胃肠道、肺)、中胚层(例如,肌肉、骨骼、血液、泌尿生殖器)或外胚层(例如,表皮组织和神经系统)的所有细胞的潜力的干细胞。“胚胎干(ES)细胞”是未分化的多能细胞,其在早期阶段从胚胎获得,例如在胚泡阶段的内细胞团,或通过人工方法(例如核移植)产生和可以在胚胎或成人中产生任何分化的细胞类型,包括生殖细胞(例如精子和卵)。
“诱导的多能干细胞”是通过重新编程因子的组合的表达来重新编程体细胞而生成的细胞。使用胎儿、出生后、新生儿、幼年或成年体细胞能够生成iPSC。能够用于将体细胞重新编程为多能干细胞的因子包括,例如,Oct4(有时称为Oct3/4)、Sox2、c-Myc、Klf4、Nanog和Lin28。可以通过表达至少两种重新编程因子、至少三种重新编程因子、至少四种重新编程因子、至少五种重新编程因子、至少六种重新编程因子或至少七种重新编程因子来重新编程体细胞,从而将体细胞重新编程为多能干细胞。
术语“体细胞”是指生殖细胞(例如卵、精子等)以外的任何细胞,其不直接将它的DNA转移至下一代。通常,体细胞具有有限的多能性或没有多能性。本文使用的体细胞可以是天然存在的或遗传修饰的。
“分化”是较不特化的细胞变成更特化的细胞类型的过程。
在本发明的第一方面,提供了一种使多能干细胞分化为巨噬细胞的方法,所述方法包括:(I)使多能干细胞分化为拟胚体,(II)使所述拟胚体与一种或多种特定的细胞因子接触,从而得到所述巨噬细胞。
当在本发明中使用时,“巨噬细胞”和“诱导的巨噬细胞”可以交替使用。
术语“拟胚体”(Embryoid bodies,EBs)是指当允许多能干细胞在悬浮培养物或聚集体中以非特异性方式分化时出现的包含未分化、分化和部分分化的细胞的异质簇。
拟胚体生成法是目前最常用的使iPSCs诱导分化为巨噬细胞的方法。EBs生成法是使未分化的多能干细胞在悬液中生长,形成一种被称为EBs的结构。EBs可以分化为内胚层、中胚层、外胚层的三胚层结构。一旦形成,EBs就可以使用特定的酶分离,获得一个细胞群体,该细胞群体利用特定诱导条件可以分化为三胚层中所有类型的细胞。常见的EBs生产方法主要有静态悬浮培养、旋转悬浮培养、悬挂滴片法、微孔和微模型芯片法等(参见例如Carpenedo RL等. Rotary suspension culture enhances the efficiency, yield, andhomogeneity of embryoid body differentiation(旋转悬浮培养提高了拟胚体分化的效率、产量和均匀性). Stem Cells, 2007, 25(9): 2224-2234;Wu HW等. 将由多能干细胞产生的EBs在无血清状态下在含有特定的细胞因子的培养基中进行培养,即可将EBs诱导分化为巨噬细胞。在本发明中使用的用于将EBs诱导分化为巨噬细胞的细胞因子选自:胎肝激酶配体(Flt3L)、粒细胞-巨噬细胞集落刺激因子(GM-CSF)、巨噬细胞集落刺激因子(M-CSF)、血管内皮生长因子(VEGF)、干细胞因子(SCF)、胰岛素样生长因子1(IGF-1)、碱性成纤维细胞生长因子(bFGF)和白介素3(IL-3)。可以使用的细胞因子的浓度为约0.1ng/mL至约500ng/mL,通常为5ng/mL至200ng/mL,更通常为10ng/mL至100ng/mL,例如10、20、30、40、50、60、70、80、90、100ng/mL及其间任意值。
在某些方面,多能干细胞是iPSC。在某些方面,PSC是人iPSC。在某些方面,人iPSC是健康人的多种组织细胞来源的iPSC、患者来源的iPSC、或者经过基因修饰的iPSC,例如病毒重编程的或附加体重编程的iPSC。
在一些实施方式中,通过包括以下步骤的方法将多能干细胞分化为拟胚体:I-1:将多能干细胞置于增补有干细胞因子SCF(例如0.1ng/mL至500ng/mL,例如10ng/mL至100ng/mL,例如50 ng/mL)、骨形态发生蛋白4(例如0.1ng/mL至300ng/mL,例如10ng/mL至50ng/mL,例如20 ng/mL)和ROCK抑制剂(例如0.1µM至50µM,例如1µM至30µM,例如10µM)的第一培养基中,例如在适当的条件下孵育一定的时间,例如1~5天,或2~3天,I-2:将从步骤I-1中获得的细胞置于增补有干细胞因子(例如0.1ng/mL至500ng/mL,例如10ng/mL至100ng/mL,例如50 ng/mL)、骨形态发生蛋白4BMP-4(例如0.1ng/mL至300ng/mL,例如10ng/mL至50ng/mL,例如20 ng/mL)、碱性成纤维细胞生长因子bFGF(例如0.1ng/mL至300ng/mL,例如10ng/mL至50ng/mL,例如20 ng/mL)、血管内皮生长因子VEGF(例如0.1ng/mL至300ng/mL,例如10ng/mL至50ng/mL,例如20 ng/mL)、胰岛素样生长因子1IGF-1(例如0.1ng/mL至300ng/mL,例如10ng/mL至50ng/mL,例如20 ng/mL)、和血小板生成素TPO(例如0.1ng/mL至500ng/mL,例如10ng/mL至100ng/mL,例如50 ng/mL)的第二培养基中,例如在适当的条件下孵育一定的时间,例如5~10天,或6~9天,或7天。
在一些实施方式中,第一培养基和第二培养基都是含血清蛋白的无饲养层完全培养基,例如mTeSR1培养基、STEMdiff™ APEL™2 Medium(STEMCELL Technologies),或StemPro™-34 SFM (Thermo)。在一些实施方式中,第一培养基和第二培养基相同。在一些实施方式中,第一培养基和第二培养基不同。
术语“ROCK”为GenBank登录号:NP_005397.1和NP_004841.2中所示蛋白,其具有丝氨酸/苏氨酸激酶活性,能够调节细胞分裂、平滑肌收缩、肌动蛋白应激纤维的形成以及c-fos血清响应元件的激活。
术语“ROCK抑制剂”是指通过抑制ROCK磷酸化水平(通过蛋白质印迹法分析检测)来抑制ROCK活性的任何分子。

在一些实施方式中,通过使从步骤I-2获得的拟胚体在四个阶段中分别与多种细胞因子、优选至少4种细胞因子接触,获得需要的巨噬细胞。在优选的实施方式中,在所有阶段中均使用Flt3L。Flt3L被报道能够促进造血干细胞和祖细胞在体外的扩增, 促进髓系前体细胞的存活。
在一些实施方式中,在第一阶段(步骤II-1)中使用bFGF、SCF、VEGF-A、IGF-1和Flt3L。在一些实施方式中,在第二至四阶段(步骤II-2~II-4)中使用的细胞因子包括IL-3、M-CSF、GM-CSF和Flt3L。在一些实施方式中,在第二阶段(步骤II-2)中使用的细胞因子还包括SCF、VEGF-A、IGF-1。在一些实施方式中,在第三阶段(步骤II-3)中使用的细胞因子还包括SCF。在一些实施方式中,在第四阶段(步骤II-4)中使用的细胞因子仅包含IL-3、M-CSF、GM-CSF和Flt3L。

本发明的方法使得能够仅仅使用8种细胞因子,就可以从拟胚体分化得到巨噬细胞,而且通过不同种类和浓度的细胞因子的组合,使得能够快速(32天)诱导出成熟的巨噬细胞,缩短生产周期。
巨噬细胞具有不同的表型,M1型和M2型巨噬细胞在免疫反应和炎症过程中有着不同的作用,而M0型巨噬细胞为未激活的、不成熟的状态。M0型巨噬细胞具有吞噬和清除细胞碎片的功能,同时也是炎症反应的起始点。M1型巨噬细胞主要参与炎症反应,产生大量的炎症因子(如IL-1β、IL-6、TNF-α等),从而引起炎症反应,吞噬和杀灭病原体。M2型巨噬细胞主要参与修复和再生过程,产生一些抗炎因子(如IL-10和TGF-β等),从而减轻炎症反应,促进组织修复和再生。在需要的情况下,还可以通过添加适当的细胞因子诱导巨噬细胞向M1和M2极化。例如使用干扰素IFN-γ和LPS脂多糖可以极化获得M1型巨噬细胞;使用Th2细胞因子如IL-4、IL-13等可以极化获得M2型巨噬细胞。
术语“极化”在本文中用于表示巨噬细胞的表型特征和功能特征。表型能够通过由巨噬细胞表达的表面标志物来定义。
巨噬细胞表达CD11b、CD14、CD18、CD45和CD64标志物,可以通过检测这些标志物的表达来定量细胞培养物中巨噬细胞的存在。M1型巨噬细胞表达CD80和CD86,而M2型巨噬细胞表达CD163和CD206,成熟/激活的巨噬细胞表达CD195和HLA-DR。巨噬细胞的另一种免疫调节功能是刺激后释放亲炎或抗炎细胞因子,这使得其释放的各种细胞因子也能成为验证靶点,亲炎细胞因子(称为M1相关) 包括IL-6、IL-8、TNF-α、CXCL8、CXCL10、CCL2和CCL4等,抗炎因子(称为M2相关) 包括IL-1RA、IL-10、VEGF和CCL22等,均可作为验证模型。
通过本发明的方法获得的巨噬细胞表达巨噬细胞表面标志物CD14、CD11b、CD86、CD163,并且还具有吞噬、杀伤和释放活性氧的功能,可用于疾病建模、药物筛选或细胞疗法,例如单独使用或与免疫检查点抑制剂/化疗剂联用、与CAR-T/T细胞或CAR-NK/NK细胞联用。
在本发明的另一方面,提供了一种用于使多能干细胞分化为巨噬细胞的试剂盒,所述试剂盒包括用于诱导多能干细胞分化为巨噬细胞的多种细胞因子,所述多种细胞因子包括胎肝激酶配体(Flt3L)、粒细胞-巨噬细胞集落刺激因子(GM-CSF)、巨噬细胞集落刺激因子(M-CSF)、血管内皮生长因子(VEGF)、干细胞因子(SCF)、胰岛素样生长因子1(IGF-1)、碱性成纤维细胞生长因子(bFGF)、白介素3(IL-3)、骨形态发生蛋白4(BMP-4)、ROCK抑制剂和血小板生成素(TPO),或者由胎肝激酶配体(Flt3L)、粒细胞-巨噬细胞集落刺激因子(GM-CSF)、巨噬细胞集落刺激因子(M-CSF)、血管内皮生长因子(VEGF)、干细胞因子(SCF)、胰岛素样生长因子1(IGF-1)、碱性成纤维细胞生长因子(bFGF)、白介素3(IL-3)、骨形态发生蛋白4(BMP-4)、ROCK抑制剂和血小板生成素(TPO)构成。

实施例
以下实施例中使用的试剂如下:
mTeSR1培养基(Stem Cell Technologies,85850);X-VIVO 15培养基(Lonza);Corning® Matrigel基质胶(Corning Cat. No. 354277);iPS无酶细胞消化液ReLeSR(StemCell Technologies,05872);人源细胞因子:BMP4、SCF、bFGF、VEGF-A、IL-3、Flt3L、MCSF、IGF-1、GM-CSF、IL-3、IL-15、TPO均购买于金斯瑞;荧光乳胶珠购买于SigmaL2778。
以下实施例中使用的诱导分化培养基包括:
(1)EB诱导培养液1:mTeSR1培养基,含有SCF (50 ng/ml)、BMP4 (20 ng/ml)、Y27632 (10µM)。
(2)EB诱导培养液2:mTeSR1培养基,含有SCF (50 ng/ml)、 bFGF (20 ng/mL)、VEGF-A (20 ng/ml)、BMP4 (20 ng/ml)、IGF-1(20 ng/ml)、TPO (50 ng/mL)。
(3)iMac细胞诱导培养液1:X-VIVO 15培养基,含有bFGF (20 ng/ml)、SCF(50 ng/ml)、VEGF-A (20 ng/ml)、IGF-1(20 ng/ml)、Flt3L(20 ng/ml)。
(4)iMac细胞诱导培养液2:X-VIVO 15培养基,含有SCF(50 ng/ml)、VEGF-A (20ng/ml)、IGF-1(20 ng/ml)、IL-3(20 ng/ml)、M-CSF (50 ng/ml) 和GM-CSF (50 ng/ml)、Flt3L (50 ng/ml)。
(5)iMac细胞诱导培养液3:X-VIVO 15培养基,SCF(50 ng/ml)、IL-3(20 ng/ml)、M-CSF (50 ng/ml) 和GM-CSF (50 ng/ml)、Flt3L (50 ng/ml)。
(6)iMac细胞诱导培养液4:X-VIVO 15培养基,含有IL-3(20 ng/ml)、M-CSF(100ng/ml)、GM-CSF (100 ng/ml)、Flt3L (50 ng/ml)。
以下实施例中使用的细胞培养条件为:在含有5%CO2的37℃恒温培养箱中培养。
实施例1从iPSC生成iMac细胞
1. 从iPSC生成拟胚体
(1)将iPSC(赛贝生物,CA4025106)接种到经Corning® Matrigel包被过的6孔板中;
(2)将iPSC在大约70%的汇合度的情况下用1mLReLeSR解离,并用70μm的滤器过滤,以去除任何结块从而制备得到单细胞悬液。在室温下300xg离心5分钟,遗弃上清液。加入1mL EB诱导培养液1,重悬细胞。吸取10 μL细胞悬液,使用Countstar细胞分析仪计算细胞密度,使用EB诱导培养液1稀释细胞悬液至50个细胞/ul的细胞密度,将细胞悬液加于圆底96孔板中(每孔100μl培养液)。室温,1500 rpm/min,离心5min,然后将培养皿放入含有5%CO2的37℃培养箱中培养3天。
(3)在诱导分化的第4天,可以观察到球型或椭球型的拟胚体开始形成(图1A),将培养基更换为EB诱导培养液2并继续诱导培养7天得到球型或椭球型的拟胚体。在诱导分化的第7天采用流式细胞术检测发现拟胚体表达干细胞标志物SSEA4(图1B)。
2. 从拟胚体生成iMac细胞
(1)诱导分化的第11天,按每孔4-6个EB的密度,将从步骤1中获得的EB转移到24孔板中,每个孔添加0.5ml iMac细胞诱导培养液1。培养7天,期间每3-4天换液一次。
(2)诱导分化的第18天,培养基换成iMac细胞诱导培养液2,培养4天。
(3)诱导分化的第22天,培养基换成iMac细胞诱导液3,培养3天。
(4)诱导分化的第25天,培养基换成iMac细胞诱导液4,培养7天,期间每3-4天换液一次。在诱导分化的第32天,有悬浮细胞在培养液中出现,收集这部分细胞,通过光学显微镜可观察到细胞呈圆形或椭圆形,并有短小的突起(图1C),流式细胞术检测发现大部分细胞表达髓系细胞标志物CD14(图1D)。
(5)诱导分化33天后,开始收集iMac,吸取细胞培养上清,500 rcf离心5分钟,遗弃上清液,加入含有MCSF(100 ng/ml)的X-VIVO 15培养基并转移至6孔板中培养5-7天,以诱导iMac成熟。在诱导分化的第38天光学显微镜观察到细胞表面有许多突起和伸展,流式细胞术检测发现绝大多数细胞(95%)表达髓系细胞标志物CD14,表明已经分化形成具有巨噬细胞形态和表面标志物的巨噬细胞(图1E和图1F)。
实施例2 iMac细胞的鉴定
1.表型鉴定
采用流式细胞术分析从实施例1获得的成熟的iMac细胞表面 CD11b,CD14,CD163,CD86的表达,结果如图2所示。结果表明经过诱导分化产生的细胞具有成熟巨噬细胞的表型。
2. 功能鉴定
(1)吞噬功能分析
肿瘤靶细胞:将使用荧光标记的靶细胞(如人胃癌细胞MKN45细胞),与用其他荧光标记的iMac细胞共培养24小时,iMac细胞数8000,MKN45细胞数8000,离心去上清,用荧光计检测剩余的活靶细胞的荧光。
磁珠:将5000个iMac细胞铺入48孔板,在含有100ng/mL M-CSF的X-VIVO15培养24h。乳胶磁珠以1:1000的比例稀释到200µL X-VIVO15培养基中,37℃预热20min后,添加到iMac细胞中,37℃孵育2 ~8 h。吸掉培养基,使用预冷的 PBS 终止吞噬,并清洗 3 次,以去除多余磁珠;收集iMac细胞进行流式分析。
如图3A所示iMac能够吞噬磁珠,表明iMac具有吞噬能力。
图3B示出DIR荧光染料标记的肿瘤细胞;图3C示出CFSE荧光染料标记的iMac细胞;图3D示出存在同时被两种荧光标记的细胞群落,表明巨噬细胞能够吞噬靶细胞。
(3)检测活性氧(ROS)
将5000个iMac细胞铺入48孔板,在含有100ng/mL M-CSF的X-VIVO15中培养24 h。将DCFH-DA(2',7'-二氯二氢荧光素二乙酸酯)以1:1000的比例稀释到200 µL X-VIVO15培养基中。去除细胞培养液,将稀释好的DCFH-DA添加到iMac细胞中。37℃细胞培养箱内孵育20分钟。用预冷的 PBS洗涤细胞三次,以充分去除未进入细胞内的DCFH-DA。收集iMac细胞进行流式分析。
如图4所示iMac的ROS水平高,表明iMac具有释放ROS能力。
(4)杀伤功能检测
将效应细胞iMac和对应的肿瘤靶细胞(如人胃癌细胞MKN45细胞)每孔肿瘤细胞细胞数8000,iMac细胞数8000、16000、40000、80000,即效靶比(T:E)1:1、1:2、1:5、1:10的比例铺96孔板中,使用含有10%胎牛血清的X-VIVO15培养基,每孔体积为200 µL。共培养48小时后,配制浓度为30ng/mL的荧光素钾盐溶液,遗弃细胞培养液,加入100 µL/孔荧光素钾盐溶液,立即放入生物化学仪内检测,对应荧光值反应该孔内肿瘤细胞存活情况。
如图5所示在肿瘤细胞的死亡率随着靶效比(T:E)的升高而增加,表明iMac细胞具有杀伤肿瘤细胞的能力。
3. 纯度分析
收集三批次诱导分化获得的iMac, 使用预冷的 PBS清洗2次,将anti-humanCD14和anti-humanCD11b流式抗体以5:100的比例稀释到200µL预冷的 PBS中,添加到iMac细胞中,4℃孵育40 分钟后。使用预冷的 PBS 终止染色,并清洗 3 次,以去除多余抗体;收集iMac细胞进行流式分析。
如图6所示三批次诱导分化获得的iMac(1、2、3)中成熟的iMac即CD14和CD11b均高表达的iMac比例分别为95.41%、98.08%和99.03%,表明本发明的诱导分化体系可稳定且高纯度获得成熟的iMac。
本发明的技术方案不限于上述具体实施例的限制,凡是根据本发明的技术方案做出的技术变形,均落入本发明的保护范围之内。
Claims (8)
1.一种使多能干细胞分化为巨噬细胞的方法,所述方法包括:
(I)使多能干细胞分化为拟胚体,其中使多能干细胞分化为拟胚体的步骤包括:
I-1:在增补有干细胞因子、骨形态发生蛋白4和ROCK抑制剂的第一培养基中孵育所述多能干细胞;
I-2:在增补有干细胞因子、骨形态发生蛋白4、碱性成纤维细胞生长因子、血管内皮生长因子、胰岛素样生长因子1、和血小板生成素的第二培养基中孵育从步骤I-1获得的细胞;
(II)使所述拟胚体与一种或多种选自以下的细胞因子接触,从而得到所述巨噬细胞:胎肝激酶配体、粒细胞-巨噬细胞集落刺激因子、巨噬细胞集落刺激因子、血管内皮生长因子、干细胞因子、胰岛素样生长因子1、碱性成纤维细胞生长因子和白介素3,其中使所述拟胚体与多种细胞因子接触的步骤包括:
II-1:在增补有碱性成纤维细胞生长因子、干细胞因子、血管内皮生长因子-A、胰岛素样生长因子1和胎肝激酶配体的第三培养基中孵育所述拟胚体;
II-2:在增补有干细胞因子、血管内皮生长因子-A、胰岛素样生长因子1、白介素3、巨噬细胞集落刺激因子、粒细胞-巨噬细胞集落刺激因子和胎肝激酶配体的第四培养基中孵育从步骤II-1获得的细胞;
II-3:在增补有干细胞因子、白介素3、巨噬细胞集落刺激因子、粒细胞-巨噬细胞集落刺激因子和胎肝激酶配体的第五培养基中孵育从步骤II-2获得的细胞;
II-4:在增补有白介素3、巨噬细胞集落刺激因子、粒细胞-巨噬细胞集落刺激因子和胎肝激酶配体的第六培养基中孵育从步骤II-3获得的细胞,从而得到所述巨噬细胞。
2.根据权利要求1所述的方法,其中所述多能干细胞是诱导的多能干细胞。
3.根据权利要求1所述的方法,其中所述ROCK抑制剂选自Y-27632、法舒地尔或H-1152。

5.根据权利要求1所述的方法,其中所述第三培养基、所述第四培养基、所述第五培养基和所述第六培养基相同,并且/或者是无血清培养基。
6. 一种用于使多能干细胞分化为巨噬细胞的试剂盒,所述试剂盒包括用于诱导多能干细胞分化为巨噬细胞的多种细胞因子,所述多种细胞因子包括胎肝激酶配体、粒细胞-巨噬细胞集落刺激因子、巨噬细胞集落刺激因子、血管内皮生长因子、干细胞因子、胰岛素样生长因子1、碱性成纤维细胞生长因子、白介素3、骨形态发生蛋白4、ROCK抑制剂和血小板生成素,或者由胎肝激酶配体、粒细胞-巨噬细胞集落刺激因子、巨噬细胞集落刺激因子、血管内皮生长因子、干细胞因子、胰岛素样生长因子1、碱性成纤维细胞生长因子、白介素3、骨形态发生蛋白4、ROCK抑制剂和血小板生成素构成。
7.根据权利要求6所述的试剂盒,其中所述试剂盒包括6个子试剂盒,其中
第一子试剂盒包括干细胞因子、骨形态发生蛋白4和ROCK抑制剂,或由其构成;
第二子试剂盒包括干细胞因子、骨形态发生蛋白4、碱性成纤维细胞生长因子、血管内皮生长因子、胰岛素样生长因子1、和血小板生成素,或由其构成;
第三子试剂盒包括碱性成纤维细胞生长因子、干细胞因子、血管内皮生长因子-A、胰岛素样生长因子1和胎肝激酶配体,或由其构成;
第四子试剂盒包括干细胞因子、血管内皮生长因子-A、胰岛素样生长因子1白介素3、巨噬细胞集落刺激因子、粒细胞-巨噬细胞集落刺激因子和胎肝激酶配体,或由其构成;
第五子试剂盒包括干细胞因子、白介素3、巨噬细胞集落刺激因子、粒细胞-巨噬细胞集落刺激因子和胎肝激酶配体,或由其构成;
第六子试剂盒包括白介素3、巨噬细胞集落刺激因子、粒细胞-巨噬细胞集落刺激因子和胎肝激酶配体,或由其构成。
8.根据权利要求6所述的试剂盒,其中所述ROCK抑制剂选自Y-27632、法舒地尔或H-1152。
