本发明属于基因工程技术领域,具体涉及一种与胶原具有特异结合能力VEGF活性抑制蛋白及其制备方法与应用,尤其涉及该抑制蛋白在制备用于治疗由于大量新生血管生成所引发的视网膜新生血管性疾病的药物中的应用。
背景技术
多种视网膜疾病与新生血管有关,该类疾病的共同病理改变是视网膜由于缺血、缺氧而产生新生血管,进而血管发生渗漏、增殖、牵拉,导致反复玻璃体积血、牵拉性视网膜脱离及新生血管性青光眼,病程进展迅速,如不及时治疗,最终会导致失明。
血管内皮生长因子(vascular endothelial growth factor,VEGF),是一种高度特异性的促血管内皮细胞生长因子,具有促进血管通透性增加、细胞外基质变性、血管内皮细胞迁移、增殖和血管形成等作用。VEGFR主要包括VEGFR1、VEGFR2、VEGFR3,其中VEGFR2是血管新生和有丝分裂过程主要的VEGF信号转导受体。VEGF/VEGFR2所介导的信号级联通路可以调控血管内皮细胞的增殖、迁移、存活,引起血管通透性的改变,控制血管的新生。
现有技术中,治疗与新生血管有关的视网膜疾病,主要采用内皮生长因子单克隆抗体药物和可溶性FLT-1和KDR药物。其治疗原理类似,均为与细胞表面VEGFR受体竞争性结合游离的VEGF,最终实现抑制眼底血管增生的能力。但是这两种药物在体内的半衰期短,也会被体内蛋白酶降解,为了维持药物的抑制血管的作用,需要持续、频繁的给药才能达到理想的作用效果。频繁的、高剂量的眼内注射给药,会增加眼内感染风险,同时也给患者造成生活上的不便。

本发明为解决现有技术中治疗与新生血管有关的视网膜疾病的药物均不具有在玻璃体内长效的作用机制等技术问题,提供一种与胶原具有特异结合能力VEGF活性抑制蛋白及其制备方法与应用。

将氨基酸序列SEQ ID NO:1克隆入质粒中,得到的重组质粒转化入大肠杆菌中进行诱导表达,诱导表达结束后,离心收集菌体,并将菌体破碎,再离心获得上清蛋白,上清蛋白经过亲和层析进行纯化,脱盐层析,分子筛柱纯化,获得血管内皮生长因子抑制蛋白。
优选的是,使用NED1和XhoII将氨基酸序列SEQ ID NO:1克隆入PET21b质粒中。

本发明的原理为:本发明的与胶原具有特异结合能力VEGF活性抑制蛋白序列中包含VEGFR1和VEGFR2的细胞外功能结构域,这个结构域主要负责与游离的VEGF结合降低其生物学活性,序列中N端包含也包括与胶原特异结合的氨基酸序列(MTKKTLRT),这个氨基酸序列来源于胶原酶,胶原酶和胶原是酶和底物的关系,因此能够特异性的结合。故而本发明的与胶原具有特异结合能力VEGF活性抑制蛋白就具有了和胶原结合和VEGF结合的双效功能,不但能够抑制VEGF的活性,同时也能和玻璃体内的胶原结合,结合后的蛋白能够更加稳定的滞留在玻璃体内,延长药物的半衰期和作用时间。从而减少药物使用量,并降低副作用,减轻患者的负担。

本发明的与胶原具有特异结合能力VEGF活性抑制蛋白安全,稳定,药效明确直接,毒副作用小,不易于产生耐药性,存储和运输条件较为温和。

具体实施方式
为了进一步理解本发明,下面对本发明优选实施方案进行描述,但是应当理解,这些描述只是为进一步说明本发明的特征和优点,而不是对本发明权利要求的限制。
如图1所示,本发明的与胶原具有特异结合能力VEGF活性抑制蛋白,包括3个VEGFR的细胞外功能结构域和一个胶原酶结构域(C),1个结构域(D1)来自于VEGFR1蛋白,2个(D2和D3)来自于VEGFR2蛋白。具体的,本发明的与胶原具有特异结合能力VEGF活性抑制蛋白包括以下氨基酸序列:MTKKTLRTGGGGPEIIHMTEGRELVIPCRVTSPNITVTLKKFPLDTLIPDGKRIIWDSRKGFIISNATYKEIGLLTCEATVNGHLYKTNYLTHRGGGGSHGIELSVGEKLVLNCTARTELNVGIDFNWEYPSSKHQHKKLVNRDLKTQSGSEMKKFLSTLTIDGVTRSDQGLYTCAASSGLMTKKNSTFVRVHGGGGTVGERVRIPAKYLGYPPPEIKWYKNGIPLESNHTIKAGHVLTIMEVSERDTGNYTVILTNPIS,记作SEQ ID NO:1。
受体是一种蛋白质,多居于靶器官的质膜上或核内/胞液中,与之结合的相应的信息分子叫配体。不同的配体只能与其相应的受体结合,启动细胞内的信息传递体系,导致细胞功能的改变,VEGF即为配体,VEGFR1和VEGFR2即为受体,属于一种跨膜蛋白,包括胞内和胞外部分,胞外部分负责与游离的配体VEGF结合从而激活细胞内的信号传导通路,实现新血管的生成。本发明的与胶原具有特异结合能力VEGF活性抑制蛋白,使用基因重组表达技术,在大肠杆菌中表达了含有VEGFR1和VEGFR2部分胞外序列的新的蛋白质,这种VEGFR拮抗蛋白能够和VEGF配体结合,但是不会激发细胞内信号传导,因此阻碍了VEGF介导的新生血管,从而降低了由于新生血管而导致的多种视网膜疾病的发生几率。序列中N端包含也包括与胶原特异结合的氨基酸序列(MTKKTLRT),这个氨基酸序列来源于胶原酶,胶原酶和胶原是酶和底物的关系,因此能够特异性的结合。
本发明的与胶原具有特异结合能力VEGF活性抑制蛋白的制备方法为:
将氨基酸序列SEQ ID NO:1克隆入质粒中,得到的重组质粒转化入大肠杆菌中进行诱导表达(融合表达),诱导表达结束后,离心收集菌体,并将菌体破碎,再离心获得上清蛋白,上清蛋白经过亲和层析进行纯化,脱盐层析,分子筛柱纯化,获得血管内皮生长因子抑制蛋白。
上述技术方案中,优选使用NED1和XhoII将氨基酸序列SEQ ID NO:1克隆入PET21b质粒中。
上述技术方案中,诱导表达的温度优选为37℃;诱导表达优选采用的是0.2mM的IPTG。
上述技术方案中,离心收集菌体的离心转速优选为4000rpm;菌体破碎采用的设备优选为超声波破碎仪;离心获得上清蛋白的离心转速优选为13000rpm。
上述技术方案中,亲和层析优选采用Ni-NTA亲和层析柱,使用Wash Buffer洗涤杂蛋白,然后使用Elution Buffer洗脱蛋白;优选过G25脱盐柱进行脱盐层析;优选分子筛柱纯化使用Superdex75分子筛柱子对上清蛋白进行纯化,流速为0.5ml/min。
大肠杆菌表达系统以其细胞繁殖快速、产量高、IPTG诱导表达相对简便等优点成为生产重组蛋白的最常用的系统。对于表达不同的蛋白,需要采用不同的载体。目前已知的大肠杆菌的表达载体可分为非融合型表达载体和融合型表达载体两种。融合表达是将目的蛋白或多肽与另一个蛋白质或多肽片段的DNA序列融合并在菌体内表达。融合型表达的载体包括分泌表达载体、带纯化标签的表达载体、表面呈现表达载体、带伴侣的表达载体。大肠杆菌表达系统优点在于遗传背景清楚、繁殖快、成本低、表达量高、表达产物容易纯化、稳定性好、抗污染能力强以及适用范围广等。
本发明的与胶原具有特异结合能力VEGF活性抑制蛋白能够在制备用于治疗视网膜新生血管性疾病的药物中的应用。
上述技术方案中,与胶原具有特异结合能力VEGF活性抑制蛋白的浓度为>100pM。
本发明还提供含有上述与胶原具有特异结合能力VEGF活性抑制蛋白的治疗视网膜新生血管性疾病的药物。
上述技术方案中,与胶原具有特异结合能力VEGF活性抑制蛋白的浓度为>100pM。
在本发明中所使用的术语,一般具有本领域普通技术人员通常理解的含义,除非另有说明。为了使本领域的技术人员更好地理解本发明的技术方案,下面将结合实施例对本发明作进一步的详细介绍。
在以下实施例中,未详细描述的各种过程和方法是本领域中公知的常规方法。下述实施例中所用的材料、试剂、装置、仪器、设备等,如无特殊说明,均可从商业途径获得。
以下结合实施例进一步说明本发明。
实施例1
(1)使用NED1和XhoII将氨基酸序列SEQ ID NO:1克隆入PET21b质粒中,然后将重组质粒转化入BL21大肠杆菌中。
(2)诱导剂IPTG的使用量为0.1mM、0.15mM、0.2mM、0.3mM,诱导温度为16℃、20℃、25℃、37℃,使用上面的诱导条件对与胶原具有特异结合能力VEGF活性抑制蛋白进行诱导表达条件摸索。
(3)诱导表达结束后,4000rpm离心收集菌体,然后使用超声波破碎仪对细菌进行破碎处理,然后13000rpm离心获得上清蛋白。
(4)上清蛋白过Ni-NTA亲和层析柱子,使用Wash Buffer洗涤杂蛋白,然后使用Elution Buffer洗脱蛋白,获得的蛋白在过G25脱盐柱进行脱盐处理。
(5)使用Superdex75分子筛柱子对蛋白进行精细纯化,流速为0.5ml/min,得到与胶原具有特异结合能力VEGF活性抑制蛋白。
使用SDS-PAGE电泳对纯化过程中各组分和蛋白进行检测。其中,IPTG浓度分别为0mM、0.1mM、0.15mM、0.2mM、0.3mM,温度25℃制备与胶原具有特异结合能力VEGF活性抑制蛋白,纯化结果如图2所示,从图2可以看出,当IPTG浓度为0.2mM时,制备的与胶原具有特异结合能力VEGF活性抑制蛋白效果最好。再以IPTG浓度为0.2mM,温度分别为16℃、20℃、25℃、37℃制备与胶原具有特异结合能力VEGF活性抑制蛋白,结果如图2所示,从图2可以看出,当温度为37℃时,制备的与胶原具有特异结合能力VEGF活性抑制蛋白效果最好。
使用SDS-PAGE电泳对纯化过程中各组分和蛋白进行检测。以IPTG浓度为0.2mM,温度为37℃制备的与胶原具有特异结合能力VEGF活性抑制蛋白,亲和层析纯化后蛋白的电泳图如图3所示,经过Superdex75分子筛进一步纯化后去除了多余的杂蛋白,如图4所示。表明经过亲和层析和分子筛层析后的蛋白可用于后续的实验使用。
获得的与胶原具有特异结合能力VEGF活性抑制蛋白使用紫外分光光度计进行定量分析,使用0.22μm的滤头进行无菌处理以备后续活性检测。使用人血管内皮细胞(HUVEC)作为研究细胞,细胞以5×104/ml密度种植于24孔板中,次日更换维持培养基DMEM(0.05%血清),培养基中加入1ng/ml的VEGF,然后再分别加入25pM、75pM、100pM、1200pM的VEGFR抑制蛋白,对贴壁的细胞进行划线,24h后对划痕进行显微镜拍照,计算各组细胞的迁移率(细胞迁移率(%)=(D0-Dt)*100%/D0,其中D0是0天时细胞划痕距离,Dt是时间点时细胞划痕距离),结果如图5所示,从图5可以看出,当与胶原具有特异结合能力VEGF活性抑制蛋白浓度大于100pM时能够有效抑制HUVEC的细胞迁移。
对与胶原具有特异结合能力VEGF活性抑制蛋白与胶原的特异结合能力测定:将鼠尾胶原在盖玻片上进行粘附,在无菌通风橱下干燥成膜,使用100μg/ml的与胶原特异结合VEGFR抑制蛋白和对照蛋白VEGFR抑制蛋白(不带胶原特异结合接头)与胶原膜进行抚育(4℃下12h),然后使用His-Tag抗体进行抚育(4℃下2h),然后使用Fitc标记的二抗进行抚育(4℃下2h)使用荧光成像仪进行拍摄照片,结果如图6和图7所示,结果显示含有胶原结合结构域的C-VEGFR与胶原膜的结合能力最强,而VEGFR只有少量的蛋白粘附在胶原膜上,这说明实施例1制备的含有胶原结合结构域的C-VEGFR能够特异性的与胶原进行结合。
显然,上述实施方式仅仅是为清楚地说明所作的举例,而并非对实施例的限定。对于所属领域的普通技术人员来说,在上述说明的基础上还可以做出其它不同形式的变化或变动。这里无需也无法对所有实施例予以穷举。而由此所引申出的显而易见的变化或变动仍处于本发明创造的保护范围之中。
Claims (10)
1.与胶原具有特异结合能力VEGF活性抑制蛋白,其特征在于,包括以下氨基酸序列:SEQ ID NO:1。
2.权利要求1所述的与胶原具有特异结合能力VEGF活性抑制蛋白的制备方法,其特征在于:
将氨基酸序列SEQ ID NO:1克隆入质粒中,得到的重组质粒转化入大肠杆菌中进行诱导表达,诱导表达结束后,离心收集菌体,并将菌体破碎,再离心获得上清蛋白,上清蛋白经过亲和层析进行纯化,脱盐层析,分子筛柱纯化,获得与胶原具有特异结合能力VEGF活性抑制蛋白。
3.根据权利要求2所述的与胶原具有特异结合能力VEGF活性抑制蛋白的制备方法,其特征在于,
使用NED1和XhoII将氨基酸序列SEQ ID NO:1克隆入PET21b质粒中。
4.根据权利要求2所述的与胶原具有特异结合能力VEGF活性抑制蛋白的制备方法,其特征在于,
所述诱导表达的温度为37℃。
所述诱导表达采用的诱导剂为IPTG,浓度为0.2mM。
5.根据权利要求2所述的与胶原具有特异结合能力VEGF活性抑制蛋白的制备方法,其特征在于,
所述离心收集菌体的离心转速为4000rpm;
所述菌体破碎采用的设备为超声波破碎仪;
所述离心获得上清蛋白的离心转速为13000rpm。
6.根据权利要求2所述的与胶原具有特异结合能力VEGF活性抑制蛋白的制备方法,其特征在于,
亲和层析采用Ni-NTA亲和层析柱,使用Wash Buffer洗涤杂蛋白,然后使用ElutionBuffer洗脱蛋白;
过G25脱盐柱进行脱盐层析;
分子筛柱纯化使用Superdex75分子筛柱子对蛋白进行纯化,流速为0.5ml/min。
7.权利要求1所述的与胶原具有特异结合能力VEGF活性抑制蛋白或者权利要求2-6任何一项所述的与胶原具有特异结合能力VEGF活性抑制蛋白的制备方法制备的与胶原具有特异结合能力VEGF活性抑制蛋白在制备用于治疗视网膜新生血管性疾病的药物中的应用。
8.根据权利要求7所述的与胶原具有特异结合能力VEGF活性抑制蛋白或者权利要求2-6任何一项与胶原具有特异结合能力VEGF活性抑制蛋白的制备方法制备的与胶原具有特异结合能力VEGF活性抑制蛋白在制备用于治疗视网膜新生血管性疾病的药物中的应用,其特征在于,所述与胶原具有特异结合能力VEGF活性抑制蛋白的浓度为>100pM。
9.含有权利要求1所述的与胶原具有特异结合能力VEGF活性抑制蛋白或者权利要求2-6任何一项与胶原具有特异结合能力VEGF活性抑制蛋白的制备方法制备的与胶原具有特异结合能力VEGF活性抑制蛋白的治疗视网膜新生血管性疾病的药物。
10.权利要求9所述的治疗视网膜新生血管性疾病的药物,其特征在于,所述与胶原具有特异结合能力VEGF活性抑制蛋白的浓度为>100pM。
