发明内容
本发明为用户提供了测定来自食品生产、食品加工或食品服务场所的环境样品中李斯特菌的存在的手段,其中在当天或同一工作班次,例如在约12小时或更短或约6至10小时内获得结果。用于测定来自环境样品的李斯特菌的常规方法需要16至24小时或更长的孵育时间,并且因此结果不可在当天得到或至少不可在同一个工作班次得到。如本文所述,当天结果为食品工业提供了优势,例如,通过允许在食品产品离开设施之前检测李斯特菌。

孵育
在收集环境样品时,样品经受孵育以增加样品中可用于检测的细胞的数量(并且从而增加靶核酸的数量)。样品中活细胞与死细胞的相对比例将转化(在一些情况下显著地)为更大比例的活细胞。这在食品安全测试中是有利的,在食品安全测试中,环境表面上活细胞的存在是特别关注的。

用于等温检测和扩增核酸(例如,RNA)的方法和试剂描述于国际专利申请公开号WO 2009/012246中,将所述专利申请其通过引用整体并入本文。简言之,扩增核酸靶序列的方法依赖于产生切口(nicking)和延伸反应,以在比传统扩增反应(例如像链置换扩增反应)更短的时间范围内扩增更短的序列。本发明的实施方案包括,例如,在等温条件下仅使用两种模板(以扩增靶序列)、一种或两种切口酶和一种聚合酶的反应。在示例性实施方案中,所述聚合酶和切口酶是嗜热的,并且反应温度显著低于杂交靶区域的解链温度。所述切口酶仅在双链双链体中的一条链中产生切口,使得不需要像常规链置换扩增的情况那样掺入经修饰的核苷酸。本发明的方法不需要初始热变性步骤。由于反应简单,因此在示例性实施方案中,反应非常容易进行,不需要特殊的设备,诸如热循环仪,并且可以在仅约2.5至约10分钟内从基因组DNA扩增20-30聚体产物108至约1000倍。所述方法能够在同时逆转录步骤中扩增RNA。

美国专利号7,662,594中描述了用于等温检测和扩增核酸的其他方法,将所述专利申请其通过引用整体并入本文。这些方法描述了核酸(例如,RNA)的解旋酶依赖性扩增。本发明的实施方案包括,例如,向李斯特菌RNA中添加包含逆转录酶、解旋酶(例如,热稳定UVrD解旋酶)、任选地单链结合蛋白、寡核苷酸引物和一种或多种聚合酶的混合物;以及逆转录所述RNA以形成cDNA,并且通过解旋酶依赖性扩增来扩增所述cDNA。
在一些实施方案中,通过自动读取器读取测定结果。在其他实施方案中,通过酶检测方法或凝胶电泳检测测定结果。

实施例1
用于从环境样品中检测李斯特菌的示例性方法(诸如图1中所示)如下:
1)用环境海绵或拭子擦抹表面以收集环境样品。
2)将海绵放置在含有2-25ml LESS Plus富集肉汤或非选择性肉汤的Whirl-袋中。
3)将样品在35℃下孵育约4-6小时。(在富集肉汤中留出1ml样品用于培养确认)

环境样品制备:
在制造设施中使用环境拭子(Letheen拭子)和海绵(EZ海绵)擦抹环境表面(池排放管(sink drain)、地漏)(使用拭子擦抹1X1英寸表面以及使用海绵擦抹4X4英寸表面),使得拭子和海绵被天然微生物群落污染。用低水平的单核细胞增多性李斯特菌(2-7CFU)接种受污染的拭子/海绵,并且在冰箱中保持3天以模拟用于李斯特菌细胞的推荐冷应激程序。

ANSR李斯特菌程序:
然后通过等温ANSR李斯特菌测定对样品进行检测。简言之,将样品在37℃下裂解10min,然后在80℃下进行热灭活步骤20min。添加一等分试样(50μL)的裂解物以再水合LRN试剂,用于在ANSR读取器中检测。在表1中,阳性ANSR李斯特菌结果标注为总检出样品比总测试样品(#阳性/#测试),表明环境样品中存在或不存在李斯特菌。
培养确认程序:
继续将剩余的拭子和海绵样品与肉汤在35℃下孵育总共24h,然后划线到MOX板上,用于观察和计数典型的李斯特菌菌落。在表1中,阳性ANSR培养结果标注为总检出样品比总测试样品(#阳性/#测试),表明环境样品中存在或不存在李斯特菌。
结果和结论:
表1示出了研究结果。
在存在背景微生物群落的情况下,使用拭子和海绵进行6小时富集(通过离心步骤浓缩或不浓缩)的李斯特菌当天测定有效检测出低水平(2-7CFU)的李斯特菌。所述测定显示出与需要样品富集24小时的培养方法可比较的结果。因此,出人意料地发现,此实施例2的李斯特菌当天测定可以用于在样品收集的当天或同一工作班次内检测来自环境表面(包括低浓度环境)的李斯特菌。

Claims (27)
1.一种检测环境样品中李斯特菌(Listeria)的方法,所述方法包括以下步骤:
使用收集装置收集包含细胞的环境样品;
将所述样品转移到含有约50ml或更少富集培养基的孵育容器中;
将所述孵育容器中的细胞孵育约12小时或更短;
裂解所述细胞以从所述细胞中释放核酸;以及
进行靶向核酸序列的检测测定,用于扩增和检测从所述细胞中释放的李斯特菌核酸。
2.根据权利要求1所述的方法,所述方法进一步包括在裂解所述细胞之前浓缩所述细胞。
3.根据权利要求1所述的方法,其中所述孵育容器含有约1ml至约50ml富集培养基。
4.根据权利要求1所述的方法,其中所述富集培养基对李斯特菌的生长是选择性的。
5.根据权利要求1所述的方法,其中所述富集培养基对李斯特菌的生长是非选择性的。
6.根据权利要求1所述的方法,所述方法包括将所述孵育容器中的细胞孵育约4至约12小时。
7.根据权利要求1所述的方法,其中所述收集装置是拭子、棉签或海绵。
8.根据权利要求6所述的方法,其中所述收集装置是海绵。
9.根据权利要求1所述的方法,其中从收集所述样品到检测所扩增的核酸的时间是12小时或更短。
10.根据权利要求8所述的方法,其中从收集所述样品到检测所扩增的核酸的时间是8小时或更短。
11.根据权利要求1所述的方法,其中所述检测测定是热稳定解旋酶依赖性等温扩增(tHDA)测定。
12.根据权利要求1所述的方法,其中所述检测测定是等温切口酶扩增反应(NEAR)测定。
13.根据权利要求2所述的方法,其中所孵育的细胞的浓缩包括离心或过滤。
14.根据权利要求1所述的方法,其中所述环境样品是从包含约2至约50个李斯特菌细胞的低浓度环境中收集的,并且其中所述检测测定检测所述样品中的李斯特菌。
15.根据权利要求13所述的方法,其中所述低浓度环境包含约5至约50个李斯特菌细胞。
16.根据权利要求1所述的方法,其中在孵育之前,所述环境样品包含约2至约50个李斯特菌细胞,并且其中所述检测测定检测所述样品中的李斯特菌。
17.根据权利要求15所述的方法,其中在孵育之前,所述环境样品包含约5至约50个李斯特菌细胞。
18.根据权利要求1所述的方法,其中所述检测测定靶向的核酸序列来自从所述细胞中释放的RNA。
19.根据权利要求1所述的方法,其中所述方法具有足够的灵敏度来检测具有少至7个李斯特菌细胞的环境样品中李斯特菌的存在。
20.根据权利要求1所述的方法,其中所述方法具有足够的灵敏度来检测具有少至3个李斯特菌细胞的环境样品中李斯特菌的存在。
21.根据权利要求3所述的方法,其中所述收集装置是海绵,并且所述孵育容器含有约5ml至约50ml富集培养基。
22.根据权利要求3所述的方法,其中所述收集装置是拭子,并且所述孵育容器含有约1ml至约10ml富集培养基。
23.一种用于检测环境样品中李斯特菌的存在的试剂盒,所述试剂盒包括:
用于收集包含细胞的环境样品的收集装置;
含有约50ml或更少富集培养基的孵育容器;以及
用于扩增和检测所述样品中的李斯特菌核酸的测定;
其中所述试剂盒能够在从收集所述样品到检测所扩增的核酸的12小时或更短的时间内检测来自低浓度环境的李斯特菌。
24.根据权利要求22所述的试剂盒,其中所述孵育容器含有约1ml至约50ml富集培养基。
25.根据权利要求22所述的试剂盒,其中所述富集培养基是LESS Plus富集培养基。
26.根据权利要求22所述的试剂盒,其中所述收集装置是拭子、棉签或海绵。
27.根据权利要求22所述的试剂盒,其中所述测定是热稳定解旋酶依赖性等温扩增(tHDA)测定或等温切口酶扩增反应(NEAR)测定。
