Evaluation of bacteriostatic Effect
Helicobacter in the mouth pylorus is distributed in plaque, dorsum lingualis, saliva and gingival crevicular fluid, and is one of the factors causing periodontitis, halitosis, recurrent oral ulcer, chronic pharyngitis. Helicobacter pylori in the mouth and stomach has homology. The swallowing action of the oral cavity can send dental plaque and helicobacter pylori in saliva into intestines and stomach from esophagus, so that gastritis is caused; helicobacter pylori in the stomach may also enter the mouth by the route of oesophageal reflux.
Streptococcus mutans is gram-positive coccus, one of the largest-proportion Streptococcus in the natural flora of the oral cavity, and one of the main components of dental plaque. Caries is a bacterial infectious disease that occurs in dental hard tissues, with the highest incidence among oral diseases. Streptococcus mutans is the primary pathogen of caries.
Evaluation was performed with reference to WS/T650-2019, antibacterial and bacteriostatic Effect evaluation method.
The antibacterial rate is more than or equal to 50-90 percent, and the antibacterial effect is judged. The antibacterial rate is more than or equal to 90 percent, and the antibacterial effect is higher.
The nicotine buccal products of examples 1 to 5 containing the antibacterial and antioxidant composition and the nicotine buccal product of comparative example 1 were evaluated for antibacterial effect, and the evaluation results are shown in table 3 below:
table 3 results of evaluation of antibacterial effect of Nicotine buccal products containing antibacterial and antioxidant compositions of examples 1 to 5 and Nicotine buccal product of comparative example 1
As can be seen from the evaluation results of Table 3, the nicotine buccal product (example 1-example 5) containing the antibacterial and antioxidant composition prepared by the component proportion and the preparation method of the application has a strong antibacterial effect, while the nicotine buccal product (comparative example 1) prepared by the lack of the antibacterial and antioxidant composition has an antibacterial effect, but the effect is not obvious. The content of the antibacterial and antioxidant composition is not in the range of the application, and the prepared nicotine buccal product (comparative examples 2-4) containing the antibacterial and antioxidant composition has antibacterial effect, but the effect is not obvious, but the antibacterial effect is stronger than that of the nicotine buccal product (comparative example 1) prepared by lacking the antibacterial and antioxidant composition component. The nicotine buccal product (comparative examples 5-7) containing the antibacterial and antioxidant composition prepared by lacking one or more components in the antibacterial and antioxidant composition has antibacterial effect, but the effect is not obvious, wherein the nicotine buccal product (comparative example 5) containing the antibacterial and antioxidant composition prepared by lacking a single component in the antibacterial and antioxidant composition has better antibacterial effect than the nicotine buccal product (comparative examples 6-7) containing the antibacterial and antioxidant composition prepared by lacking multiple components in the antibacterial and antioxidant composition. Therefore, the nicotine buccal product containing the antibacterial and antioxidant composition realizes the antibacterial effect through the synergistic effect of hericium erinaceus polysaccharide, procyanidine, resveratrol, ellagic acid, rosmarinic acid and hyperoside in the antibacterial and antioxidant composition.
Evaluation of antioxidant efficacy
The DPPH scavenging measurement method is applied to quantitative measurement of antioxidant capacity of biological samples, phenols and other substances, and the measurement principle is that the DPPH (1, 1-diphenyl-2-trinitrophenylhydrazine) free radical has single electron, has strong absorption at 517nm, and the alcohol solution has purple characteristic. When the free radical scavenger exists, the DPPH single electron is paired with the free radical scavenger, so that the absorption of the free radical scavenger gradually disappears, the color development reaction of the alcohol solution starts to weaken, the color development degree is quantitatively related to the number of the electrons accepted by the alcohol solution, and therefore, the rapid quantitative analysis can be performed by a spectrophotometer.
DPPH inhibition rate detection method:
preparation of sample dilution 1g of the nicotine buccal product containing the antibacterial and antioxidant composition of examples 1 to 5 and the nicotine buccal product of comparative example 1 were taken and diluted 100 times with ultrapure water to prepare sample dilutions.
(1) 2ml of sample diluent and 0.5ml of DPPH solution with the concentration of 0.5mmol/ml are added into the same test tube with a plug successively, the mixture is shaken uniformly, the mixture is placed for 20min at 37 ℃ in the dark, and the absorbance Ai is measured at 514nm by taking methanol as a blank.
(2) 0.5ml of a DPPH solution of 0.5mmol/ml was mixed with 2ml of methanol, shaken well, left to stand in the dark at 37℃for 20min, and the absorbance Ao was measured at 514nm with methanol as a blank.
(3) 2ml of the sample dilution was mixed with 0.5ml of methanol, shaken well, left to stand in the dark at 37℃for 20min, and the absorbance Aj was measured at 514nm with methanol as a blank.
Calculation of inhibition of DPPH by sample: inhibition ratio (%) = [ 1- (Ai-Aj)/Ao ] x 100
Wherein Ai is absorbance of DPPH solution after adding antioxidant; ao is the absorbance of the DPPH solution without antioxidant; aj is the absorbance of the sample at the measurement wavelength. Aj is introduced in the formula to eliminate the interference of the sample diluent itself on the assay. The greater the inhibition, the higher the antioxidant activity.
The DPPH inhibition rate test results of the nicotine buccal products of examples 1 to 5 of the present application containing the antibacterial and antioxidant composition and the nicotine buccal product of comparative example 1 are shown in FIG. 1. As can be seen from fig. 1, the nicotine buccal preparation (examples 1 to 5) containing the antibacterial and antioxidant composition of the present application has a better antioxidant effect than the nicotine buccal preparation (comparative example 1). Therefore, the application has remarkable effect in the experiment through the antioxidation synergistic effect of the hericium erinaceus polysaccharide, the procyanidine, the resveratrol, the ellagic acid, the rosmarinic acid and the hyperin.
