Abstract

Description
技术领域
本发明属于分子生物学和生物医药领域,具体地,本发明涉及一种UTP11抑制剂及其在肿瘤抑制中的用途。
背景技术
p53位于人17号染色体短臂(17p13.1),由11个外显子、10个内含子和393个氨基酸残基组成。p53是一种重要的抑癌基因,也是人类癌症中最常见的突变基因。突变型p53除了失去其抑癌功能外,通常还具有固有的、新的致癌功能,被称为“功能获得”。越来越多的证据表明,突变p53与晚期恶性肿瘤和不良预后密切相关。在对不同人类癌症细胞进行基因组测序的结果表明,在所有恶性肿瘤中50%以上会出现该基因的突变。因此,开发针对p53突变型癌症的抑制剂具有重要的临床意义。
强大的核糖体生物合成对蛋白质合成机制的运作和癌细胞的快速生长至关重要。这一过程涉及大约70种核糖体蛋白质(RPs),四种核糖体RNA(rRNA),包括28S、18S、5.8S和5S rRNA,以及超过150个辅助因子。核糖体生物发生与癌细胞的生长和增殖密切协调。致癌基因MYC通过增加rRNA和RPs的表达来促进核糖体的生物合成和整体蛋白质的合成,而肿瘤抑制因子p53抑制RNA Pol I-和iii介导的转录,导致rRNA和核糖体生物合成的减少。因此,癌细胞通常保持更活跃的核糖体生物合成,这可能是癌症的一个可靶向的脆弱靶标。
对包括基因改变、营养物质消耗或治疗药物在内的对核糖体生物合成的任何步骤的干扰都会导致癌细胞生长停滞和凋亡,这被称为核糖体应激或核仁应激,因为rRNA的合成、加工和核糖体前组装都发生在核仁中。真核生物核糖体是癌细胞存活所必需的。干扰核糖体生物合成的诱发核仁应激或核糖体应激,抑制癌症的发展,原因在于快速增殖的癌细胞需要更活跃的核糖体生物合成。
因此,本领域技术人员致力于开发能够靶向核糖体生物发生的抑制剂,以期获得更为高效的抗肿瘤药物,特别是针对p53突变型癌症有效的抑制剂。
发明内容
本发明的目的是提供一种UTP11抑制剂及其在肿瘤抑制中的用途。
在本发明的第一方面,提供了一种UTP11基因或其编码蛋白的抑制剂的用途,用于:
(1)制备预防或治疗肿瘤的药物;
(2)制备激活p53的试剂;
(3)制备通过p53依赖的机制抑制肿瘤的药物;或者
(4)制备通过p53非依赖的机制抑制肿瘤的药物。
在另一优选例中,所述的UTP11基因选自下组:
(A)编码如SEQ ID NO.2所示多肽的多核苷酸序列;
(B)如SEQ ID NO.1所示的多核苷酸序列;
(C)将SEQ ID NO.1所示的多核苷酸序列经过一个或几个核苷酸的取代、缺失或添加而形成的多核苷酸序列;
(D)序列与SEQ ID NO.:1所示的多核苷酸序列相比,同源性≥90%,较佳地≥95%,更佳地≥98%,最佳地≥99%的多核苷酸序列;
(E)与(A)-(D)任一所述的多核苷酸序列互补的多核苷酸序列。
在另一优选例中,所述的UTP11基因来源于哺乳动物(包括人)。
在另一优选例中,所述UTP11基因的抑制剂为:UTP11基因特异性的siRNA或其前体、UTP11基因特异性的microRNA或其前体、抑制UTP11基因启动子的抑制剂、或其组合。
在另一优选例中,所述UTP11基因的抑制剂为UTP11基因特异性的siRNA或其前体、或microRNA或其前体。

在本发明的第五方面,提供了一种siRNA前体(shRNA),所述siRNA前体是本发明第四方面所述的siRNA的前体。
在另一优选例中,所述siRNA前体的5’至3’端依次包括:第一序列单元、茎环序列单元和第二序列单元,所述第一序列单元和所述第二序列单元反向互补使得所述siRNA前体形成发夹结构,并且所述第一序列单元选自下组:

具体实施方式
本发明人经过广泛而深入的研究,意外发现靶向UTP11的抑制剂不仅能够抑制p53野生型肿瘤而且能够抑制p53突变型的恶性肿瘤。本发明首次公开了UTP11的缺失通过p53依赖和非p53依赖的机制在体外和体内抑制癌细胞生长。具体地,本发明揭示了UTP11在维持癌细胞生存和生长方面的关键作用,靶向UTP11会导致p53依赖性癌细胞生长停滞和p53非依赖性的癌细胞生长停滞。在此基础上,完成了本发明。
术语
UTP11基因及其编码蛋白
人类UTP11最初是通过比较蛋白质组学方法以秀丽隐杆线虫蛋白质组为模板鉴定出来的。然而,它的生物学功能几十年来一直不清楚。
人UTP11基因(NCBI序列号Gene ID:NM_016037)位于1p34.3染色体上。在人类细胞中,该基因编码的蛋白可能参与核糖体小亚基的生物合成,可能参与肿瘤的发生过程,但其具体功能和作用机制未知。

UTP11基因的抑制剂
如本文所用,术语“UTP11基因的抑制剂”是指抑制UTP11基因基因复制或转录的物质,或减少UTP11基因表达的物质,UTP11基因的抑制剂包括(但不限于):siRNA、microRNA、化合物、或其组合。UTP11基因的抑制剂优选siRNA、或microRNA。
如本文所用,术语“RNAi”(RNA interference,RNA干扰)是指在进化过程中高度保守的、由双链RNA(dsRNA)诱发的、高效特异性降解具有互补配对序列的RNA的现象。由于使用RNAi技术可以特异性关闭特定基因的表达,所以该技术已被广泛用于探索基因功能和传染性疾病及肿瘤的基因治疗等领域。
如本文所用,术语“siRNA”(Small interfering RNA,siRNA)是指一种小RNA分子(约21-25个核苷酸),可由Dicer(RNA酶Ⅲ家族中对双链RNA具有特异性的酶)从其前体(比如dsRNA、shRNA等)加工而成,也可由化学方法合成或由其它蛋白加工产生。siRNA是siRISC的主要成员,激发与之序列互补的目标RNA被迅速切割降解,导致目标基因的沉默,因此成为RNAi中的关键功能分子。
在本发明的一个优选地实施方式中,本发明提供了一种具有本发明siRNA序列的siRNA前体。如本文所用,术语“siRNA前体”是指可以在哺乳动物细胞中被加工产生siRNA的RNA分子,具体地说,是由Dicer或其它类似蛋白选择性加工从而产生成熟的siRNA,进而实施RNAi。
在本发明的一个优选地实施方式中,本发明提供了一种核酸构建物。如本文所用,术语“构建物”是包含本发明siRNA前体的核酸构建物。
在本发明的一个优选地实施方式中,本发明提供了一种表达盒。如本文所用,术语“表达盒”是指包含本发明siRNA前体的编码序列以及与所述编码序列操作性相连的启动子和终止信号的表达盒,所述表达盒在转录后产生本发明的siRNA前体。
在活体中产生“小干扰RNA”(siRNA)的一种办法是,将siRNA序列作为“短发夹”的一部分克隆进质粒载体中。当送入动物体内时,该发夹序列)被表达出来,形成一个带有顶端环结构的“双链RNA”(shRNA),被细胞内的Dicer蛋白所识别和加工,产生有功能的siRNA。
如本文所用,术语“shRNA”是以人miR-26b的前体作为骨架构建的一种特殊的shRNA。所述shRNA从5′端到3′端依次为:(a)5′端旁侧序列区;(b)5′端配对siRNA区域;(c)顶端环区域;(d)3′端配对siRNA区域,并且所述5′端配对siRNA区域与3′端配对siRNA区域形成双链区域;(e)3′端旁侧序列区;所述shRNA产生siRNA,且所述siRNA的核苷酸序列对应于所述3′端配对siRNA区域或5′端配对siRNA区域。
广义的shRNA是short hairpin RNA的缩写,即,“短发夹RNA”。shRNA包括两个短反向互补序列,中间由一顶端环(loop)序列分隔的,组成发夹结构,通常由细胞内源的RNA聚合酶III(RNApolymeraseIII)启动子控制转录,shRNA序列的末端连接5-6个T作为RNA聚合酶Ⅲ的转录终止子。shRNA也可以由其它RNA聚合酶的启动子转录产生。
如本文所用,术语“miRNA”(microRNA)是一类由内源基因编码的长度约20-24个核苷酸的非编码单链RNA分子,在动植物中参与对大量基因的表达调控。到目前为止,在动植物以及病毒中已经发现四千多种miRNA分子。大多数miRNA基因以单拷贝、多拷贝或基因簇(cluster)的形式存在于基因组中。每种miRNA可以调控多个靶基因,而几种miRNA也可以共同参与调节同一基因,组成复杂的调节网络。据推测,miRNA调节着人类一半以上基因的表达。miRNA存在多种形式,最原始的是pri-miRNA;pri-miRNA经过Drosha加工后,成为pre-miRNA,即miRNA前体,长度大约为50-90个核苷酸;pre-miRNA再经过Dicer酶酶切后,成为长约20-24个核苷酸的成熟miRNA。miRNA主要通过抑制翻译和加速mRNA的脱腺苷酸化抑制靶基因表达,其机制有别于siRNA介导的mRNA降解。
UTP11基因编码蛋白的抑制剂
如本文所用,术语“UTP11基因编码蛋白的抑制剂”是指抑制UTP11基因编码蛋白活性的物质。
在一个优选地实施方式中,UTP11基因编码蛋白的抑制剂选自下组:特异性抗UTP11基因编码蛋白的抗体、特异性结合UTP11基因编码蛋白的结合蛋白、抑制UTP11基因编码蛋白活性的化合物等。
药物组合物
本发明提供了一种药物组合物,包括药学上可接受的载体和有效量的以下活性成分:UTP11基因或其编码蛋白的抑制剂。
如本文所用,术语“有效量”或“有效剂量”是指可对人和/或动物产生功能或活性的且可被人和/或动物所接受的量。
如本文所用,“药学上可接受的”的成分是适用于人和/或哺乳动物而无过度不良副反应(如毒性、刺激和变态反应)的,即具有合理的效益/风险比的物质。术语“药学上可接受的载体”指用于治疗剂给药的载体,包括各种赋形剂和稀释剂。
本发明的药物组合物含有安全有效量的本发明的活性成分以及药学上可接受的载体。这类载体包括(但并不限于):盐水、缓冲液、葡萄糖、水、甘油、乙醇、及其组合。通常药物制剂应与给药方式相匹配,本发明的药物组合物的剂型为注射剂、口服制剂(片剂、胶囊、口服液)、透皮剂、缓释剂。例如用生理盐水或含有葡萄糖和其他辅剂的水溶液通过常规方法进行制备。所述的药物组合物宜在无菌条件下制造。
本发明所述的活性成分的有效量可随给药的模式和待治疗的疾病的严重程度等而变化。优选的有效量的选择可以由本领域普通技术人员根据各种因素来确定(例如通过临床试验)。所述的因素包括但不限于:所述的活性成分的药代动力学参数例如生物利用率、代谢、半衰期等;患者所要治疗的疾病的严重程度、患者的体重、患者的免疫状况、给药的途径等。通常,当本发明的活性成分每天以约0.00001mg-50mg/kg动物体重(较佳的0.0001mg-10mg/kg动物体重)的剂量给予,能得到令人满意的效果。例如,由治疗状况的迫切要求,可每天给予若干次分开的剂量,或将剂量按比例地减少。
本发明所述的药学上可接受的载体包括(但不限于):水、盐水、脂质体、脂质、蛋白、蛋白-抗体缀合物、肽类物质、纤维素、纳米凝胶、或其组合。载体的选择应与给药方式相匹配,这些都是本领域的普通技术人员所熟知的。
应用
本发明提供了一种治疗肿瘤细的方法,包括步骤:给肿瘤患者施用治疗有效量的UTP11基因或其编码蛋白的抑制剂。
本发明还提供了一种体外非治疗性的抑制肿瘤细胞的方法,包括步骤:在UTP11基因或其编码蛋白的抑制剂存在的条件下,培养肿瘤细胞,从而抑制肿瘤细胞。
在另一优选例中,所述肿瘤为p53野生型肿瘤,或者所述肿瘤细胞为p53野生型肿瘤细胞。
在另一优选例中,所述肿瘤为p53突变型肿瘤,或者所述肿瘤细胞为p53突变型肿瘤细胞。

p53野生型肿瘤和p53突变型肿瘤
p53蛋白是一种转录因子,通常分为三个功能区:氨基末端结构域、DNA结合域和羧基末端结构域。野生型p53(wt p53)通过调控多个靶基因的转录,在许多重要的生物学过程中发挥关键作用。
然而,突变的p53不仅失去了wt p53的肿瘤抑制功能,而且还获得了促进恶性肿瘤进展的新功能。p53的主要突变类型包括错义突变、截短突变、框架内突变和剪接突变。错义突变导致单一氨基酸替换,在肿瘤发生过程中显示出功能增强的活性,例如促进肿瘤细胞侵袭和迁移的p53 R175H和R273H突变。截短突变会导致蛋白质截断,这也可以促进肿瘤的发展。例如,p53外显子6截短突变体R196*和R213*促进肿瘤细胞的增殖和转移。框架内突变由核苷酸的缺失或插入引起。剪接突变是由发生在剪接部位的突变引起的。因此,本发明中术语“p53突变型肿瘤(肿瘤细胞)”指携带p53突变的肿瘤(肿瘤细胞),p53突变包括错义突变、截短突变、框架内突变和剪接突变。这些突变通常会导致更具侵袭性和耐药性的肿瘤。
本发明的主要优点在于:
(1)首次发现UTP11在维持癌细胞生存和生长方面的关键作用,靶向UTP11会导致p53依赖的癌细胞生长停滞和非p53依赖的癌细胞生长停滞。
(2)验证了UTP11基因及其编码蛋白可以作为新型的肿瘤治疗靶点。
(3)获得了高效抑制UTP11基因的siRNA。
下面结合具体实施例,进一步详陈本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。下列实施例中未注明具体条件的实验方法,通常按照常规条件如Sambrook等人,分子克隆:实验室手册(New York:Cold Spring Harbor LaboratoryPress,1989)中所述的条件,或按照制造厂商所建议的条件。除非另外说明,否则份数和百分比按重量计。
实施例
(I)材料与方法
人类乳腺癌和结直肠癌标本
采用来自中国医科大学第一附属医院的6对乳腺癌及和10对其邻近正常组织分别进行免疫印迹和RT-qPCR分析。取中国医科大学第一附属医院乳腺癌组织9 1石蜡包埋切片进行免疫组化分析。本研究获中国医科大学第一附属医院人体研究伦理委员会批准。此外,利用南昌大学第一附属医院获得的10对结直肠癌及其邻近正常组织和150份结直肠癌石蜡包埋切片进行免疫组化分析。本研究获南昌大学第一附属医院人体研究伦理委员会批准。
免疫组织化学
石蜡包埋的乳腺或结直肠癌组织切片在65℃脱蜡1-2小时,然后置于二甲苯和分级系列酒精中。随后,用柠檬酸钠-EDTA抗原修复液(catNo:P0086,碧云天)用于抗原揭膜。冷却后,与一抗在室温孵育1-2小时。然后用辣根过氧化物酶(HRP)偶联二抗(catNo:GK500705,GeneTech)在室温中孵育30-60分钟,与3'-二氨基联苯胺(No:GK500705GK500705,Gentech)孵育5分钟。随后,用苏木精复染载玻片,用分级系列乙醇脱水,用盖玻片和培养基进行封片。染色密度测量使用徕卡CCD相机DFC420连接徕卡DM IRE2显微镜(徕卡微系统成像解决方案有限公司)。用染色强度(0=阴性,1=弱,2=中,3=强)与阳性染色率(0=阴性,1≤10%,2=10-50%,3≥50%)相乘计算IHC评分。免疫组化评分≤4时暗示UTP11为低表达,免疫组化评分为>4时暗示UTP11为高表达。
生物信息学分析
为了探索UTP家族基因与p53信号通路相关的潜在生物学机制,我们提取了40个UTP和587个p53信号基因之间的已知蛋白-蛋白相互作用关系。简单地说,基于所有已知的蛋白质-蛋白质相互作用,我们使用Pathway Commons数据库(V版本12)(35)创建了一个非冗余的人类交互组,其中包括30918个基因和1787402个基因-基因连接。接下来,我们使用子网络提取工具GenRev(36)研究了UTP相关rRNA加工与587个p53信号基因之间的联系。总共有627个与UTP和p53信号相关的基因首先通过GenRev映射到PathCommons互作组。然后,该算法将映射的基因连接起来,形成一个尽可能含有多个输入基因的完全连接的子网络。
统计学
体外实验均进行生物学三次重复。p值通过GraphPad Prism 5.0进行测试或方差分析获得。采用Kaplan-Meier法分析两组患者生存期的显著差异。P<0.05认为差异有统计学意义。采用多变量Cox比例风险模型计算95%置信区间的风险比。星号表示有统计学意义:*p<0.05;**p<0.01;***P<0.001。定量数据以平均值±标准差表示。
细胞培养和瞬时转染
将人癌细胞系CAL-51、MCF-7、HCT116 p53+/+、HCT116 p53-/-和RKO(购自ATCC)培养在添加含有10%胎牛血清、青霉素(100U/ml)和链霉素(0.1mg/ml)的Dulbecco改良Eagle培养基中,孵育在含5% CO2的37℃加湿培养箱中培养。
按照S说明书(中国上海易森),质粒和RNAsiRNA使用Hieff反式脂质体转染试剂被瞬时转染到细胞中,并按照图例中所示在板上过夜。转染后36-72小时收集细胞用于后续实验。环己酰亚胺(CHX)和蛋白酶体抑制剂MG132购自Med Chem Express(中国)。
siRNA的设计和筛选
针对UTP11基因设计siRNA并进行敲减效率有效性的检测。
siRNA敲减效率筛选:
将设计的siRNA进行合成后,通过转染细胞后提取细胞RNA,进行反转录反应后,通过qPCR检测UTP11基因的敲减效率。

根据初筛结果,针对UTP11基因表现出敲减效果的10个siRNA进行复筛,根据复筛结果,将敲减效果最好的05#、08#siRNA分别命名为siUTP11-1(SEQ IDNO.7)、siUTP11-2(SEQ ID NO.10)进行后续实验。
质粒和抗体
编码HA-MDM2,p53,His-Ub的质粒为已公开质粒。标记的pENTER-UTPUTP11质粒购自维吉尼生物科学公司(中国)。将人UTP11亚克隆到带有Flag标签的pcDNA3.1载体中。
本实施例所用抗体均购买自相应的商业化公司,例如:
抗UTP11抗体:Cat NO:46701,圣克鲁斯生物技术公司;
抗p53抗体:Cat NO:sc-126,DO-1,圣克鲁斯生物技术公司;
抗GAPDH抗体:Cat NO:60004-1-Ig,Proteintech公司;
抗p21抗体:细胞信号技术公司;
二抗为酶联亲和山羊抗兔IgG(Cat NO:SA00001-2,武汉三鹰)和抗小鼠IgG(CatNO:SA00001-1,武汉三鹰)。
蛋白质显影用ECL化学发光试剂(Yeasen公司)。
RNA-测序
收集转染了siNC或siUTP11 48小时后的CAL-51细胞,按照说明书(日本,Takara公司)使用RNAiso Plus分离总RNA,由OEbiotech(中国)提供RNA测序服务。
逆转录和实时定量PCR
使用RNAiso Plus(日本,Takara公司)分离总RNA。以0.2-0.5微克RNA为模板,用Hiscript III qRT SuperMix(中国,诺唯赞公司)RNA合成互补DNA(cDNA)。根据说明书(中国,诺唯赞公司)使用SYBR qPCR Master Mix进行定量PCR(qPCR)。用GAPDH作为内部参照,通过比较Ct法计算RNA的mRNA的相对表达量。
免疫印迹
在冰-冷的裂解缓冲液[50mm Tris/HCl(pH 7.5),0.5%NP-40,1毫摩尔EDTA,150毫摩尔NaCl,1毫摩尔二硫苏糖醇(DTT),0.2毫摩尔苯甲基磺酰氟(PMSF),10微摩尔pepstatatin A,1微克/毫升leupeptin和10%蛋白酶抑制剂鸡尾酒]中提取蛋白质。使用等量的透明细胞裂解物(20-80微克)用于免疫印迹(IB)分析。
免疫沉淀反应
CAL-51细胞转染对照或UTP11 siRNA 48小时,MG132处理4-6小时后收获。免疫沉淀(IP)使用抗体进行。简单地说,500-1000微克的蛋白质与指示抗体在4℃孵育5小时。然后加入蛋白A或g珠(美国,圣克鲁斯生物技术有限公司),混合物在4℃孵育2小时。用裂解缓冲液洗涤6-8次。用免疫吸附法检测蛋白质相互作用。
免疫荧光染色
转染siRNA和质粒的HCT116 p53+/+和HEK293T细胞在-20℃预冷的甲醇中固定过夜。用磷酸盐缓冲液(PBS)冲洗固定细胞,用8%牛血清白蛋白(BSA)在PBS中封闭1小时,然后与含2%牛血清白蛋白的一抗,在4℃孵育过夜。之后,用PBS清洗细胞,并与相应的荧光二抗和DAPI孵育。用倒置荧光显微镜(德国,徕卡)获取图像。

将稳定表达shNC或shUTP11的HCT116 p53-/-细胞转染编码p53、HA-MDM2或His-Ub的质粒,收样前用MG132处理4-6小时。转染48小时后,细胞被收集并分裂成两个等分体,一个用于免疫吸附,另一个用于泛素化试验。简言之,细胞碎片溶解在缓冲液[8摩尔尿素,0.1摩尔Na2HPO4/NaH2PO4(pH值8.0),10毫摩尔Tris-HCl(pH值8.0),10毫摩尔β巯基乙醇,咪唑和5毫摩尔咪唑],在室温下与Ni-NTA珠子共孵育4小时为了捕获His-tagged蛋白质/复合物。用缓冲液I洗Ni-NTA珠子和洗了两次,然后用缓冲液II[8摩尔尿素,0.1摩尔Na2HPO4/NaH2PO4(pH值6.3),10毫摩尔Tris-HCl(pH值6.3),10毫摩尔β巯基乙醇]洗两次。洗脱捕获的蛋白并与指示抗体吸附用于免疫印迹分析。
RNA干扰和稳定细胞系的产生
针对UTP11、RPL5和RPL11的siRNA由GenePharma(中国,上海)合成和纯化。
siRNA序列如下:
siUTP11-1:5'-GAAGCTAAGAAAATCGAAA-3'(SEQ ID NO.7);
siUTP11-2:5'-GGATGGAGTACATATTATT-3'(SEQ ID NO.10);
siRPL5:5'-GGAGGAGATGTATAAGAAA-3'(SEQ ID NO.19);
siRPL11:5'-GGAACTTCGCATCCGCAAA-3'(SEQ ID NO.20)。
使用Hieff反式脂质体转染试剂,按照说明书的要求将siRNA导入细胞。转染IB或RT-qPCR 48-72小时后收集细胞。将获得的UTP11的shRNA序列亚克隆到pLKO.1质粒中。
shRNA质粒和包装质粒psPAX2和pMD2.G一起转染进HEK293T细胞。转染48小时后收集慢病毒颗粒,然后用于CAL-51、HCT116 p53+/+和HCT116 p53-/-细胞的感染。用1微克/毫升嘌呤霉素培养稳定细胞。
细胞活力测定
根据生产商的说明书(Dojindo),用细胞计数Kit-8(CCK-8)法评估细胞活力。转染后6~12小时,在96孔培养板中每孔接种2~3.5×103细胞,每孔重复3次。在不同时间点以最终浓度为10%的CCK-8添加到每个孔中,使用Microplate Reader在450纳米处测量样品的吸光度。
菌落形成试验
转染后6~18小时,将1×103细胞置于6厘米板上,培养14天。每3天更换一次培养基,直到菌落可见为止。用甲醇固定菌落,用0.2%结晶紫溶液在室温下下染色30分钟。用ImageJ软件对菌落计数进行量化。
细胞周期分析
转染siRNA的细胞用70%乙醇固定过夜,用250微升缓冲液(50微克/毫升RNase A,0.1% Triton X-100in PBS)在37℃下处理30分钟。然后用250微升PBS缓冲液[50微克/毫升碘化丙啶(PI)(诺唯赞公司),0.1% Triton X-100(上海生工有限公司)]在黑暗中染色30分钟。最后,用流式细胞仪(CytoFLEX S,Beckman Coulter,Indianapolis,USA)分析细胞周期。
Transwell迁移试验
Transwell腔室插入24孔板中。在200微升无血清培养液中加入5~10×104细胞。下腔填充800微升20% FBS培养基。37℃培养36-48小时后,刮洗上表面细胞,甲醇固定下表面细胞,0.2%结晶紫染色。在光学显微镜下,在至少三个随机选择的区域内计数迁移细胞,并用ImageJ软件进行量化。
小鼠异种移植研究
4周龄雌性BALB/c裸鼠购于复旦大学上海癌症中心并在实验动物科学中心饲养。将稳定表达shNC或shUTP11的CAL-51细胞[6×106细胞悬浮在DMEM与50%Matrigel(BDBiosciences)]注射到小鼠右侧。为了验证UTP11缺陷介导的肿瘤抑制作用是否依赖于p53,我们使用5×106HCT116 p53+/+和HCT116 p53-/-稳定表达shNC或shUTP11的细胞进行了另外一组实验。用电子数字卡尺监测肿瘤的长度和宽度,肿瘤体积计算公式为:肿瘤体积(mm3)=(长×宽)×0.52。最后采用安乐死法处死小鼠,采集肿瘤进行分析。动物实验方案符合伦理准则,并经复旦大学上海癌症中心动物福利委员会批准。
RNA免疫沉淀反应
收集转染空载体或Flag-UTP11的细胞,并悬浮在RIP缓冲液(10毫摩尔Tris,150毫摩尔NaCl,1毫摩尔Na2EDTA.2H2O,3.5毫摩尔SDS,1毫摩尔DTT,1%NP-40,pH 7.4)。细胞裂解物用抗flag磁珠(Cat No:B26101,bimake,上海,中国)在4℃下共孵育过夜。用RIP缓冲液冲洗6次,然后进行RNA纯化和RT-qPCR分析。
(II)结果
UTP11在癌症中作为一种潜在的癌蛋白
本发明评估了三种UTP在乳腺癌中的预后意义,发现只有UTP11与不良预后显著相关,p值为1.8E-09。此外,与正常组织相比,在不同类型的癌组织中UTP11的mRNA和蛋白水平都被发现上调。在各种癌症中,较高水平的UTP11与较差的预后相关。这些结果初步表明,UTP11可能在癌症中起致癌蛋白的作用。
进一步地研究,证实了UTP11过表达与乳腺癌和结直肠癌不良预后相关。
首先,通过免疫印迹和RT-qPCR分析UTP11在乳腺癌和配对正常组织中的表达。
与正常组织相比,乳腺癌组织中UTP11的蛋白和mRNA水平均上调(图1、图2)。
此外,91例乳腺癌标本的UTP11免疫组化(IHC)染色显示,较高水平的UTP11与较高的肿瘤/节点/转移(TNM)分期和较差的患者总生存期显著相关(图3)。
此外,91例患者总生存期的单因素和多因素分析表明,UTP11是乳腺癌的预后因素(图4)。
UTP11在结直肠癌中的临床相关性也得到了评估。
与乳腺癌的结果一致,通过免疫组化染色,结直肠癌中的UTP11水平高于邻近正常组织(图5和图6)。
此外,在150名结直肠癌患者的队列中,UTP11水平较高与TNM分期较高和预后较差显著相关(图7)。
此外,单因素和多因素分析显示,UTP11是结直肠癌的预后因素(图8)。
这些结果证明了UTP11的临床意义,为癌症治疗利用该位点提供了基础。
UTP11的消融诱导p53的激活
针对UTP11基因,设计并筛选出了敲减效果最好的siRNA(siUTP11-1、siUTP11-2)进行后续实验。siRNA复筛结果如图9所示。
通过对CAL-51乳腺癌细胞进行了RNA-测序(RNA-seq)分析,发现敲低UTP11可能会增加p53的转录活性,多个p53靶基因的上调证明了这一点(图10)。
为了进一步证实这一结果,进行了RT-qPCR和免疫印迹(IB)分析,发现在CAL-51和MCF-7乳腺癌细胞系中,用两个独立的siRNA敲低UTP11显著增加了p53及其靶基因p21、BTG2和MDM2的表达(图11-图14)。
两株等基因结直肠癌细胞系HCT116 p53+/+和HCT116 p53-/-也被用来阐明UTP11在p53调控中的可能作用。UTP11敲除可诱导HCT116 p53+/+细胞中p53靶基因的表达(图15和图16),但对HCT116 p53-/-细胞中p53靶基因的表达影响甚微(图17和图18)。
此外,我们还发现UTP11缺失可能在蛋白水平上调节p53的表达,因为UTP11敲除对RT-qPCR检测的p53 mRNA水平几乎没有影响。这些结果表明UTP11缺乏激活p53途径,再次表明该蛋白在癌症中的致癌作用。
UTP11缺乏抑制乳腺癌细胞生长和进展
由于UTP11在癌症中高度表达,其缺失触发核仁应激和p53激活,在此确定靶向UTP11是否抑制了野生型p53细胞的生长和进展。
使用两个独立的siRNA敲除UTP11显著抑制了CAL-51和MCF-7乳腺癌细胞的活力(图19和图20)和克隆形成能力(图21和图22)。
此外,在这两种乳腺癌细胞系,消融UTP11诱导G1细胞周期停滞(图23和图24),这与UTP11敲除后p21上调有关(图11-图16)。transwell试验显示UTP11消融显著抑制了乳腺癌细胞的侵袭(图25和26)。
与这些基于细胞的结果一致,我们发现靶向UTP11抑制了乳腺癌细胞在体内的生长。将稳定表达对照细胞或UTP11 shRNA的CAL-51细胞通过皮下注射于裸鼠侧部。UTP11缺乏显著降低了移植瘤的生长速度(图27),但并不显著影响小鼠体重(图28)。
此外,肿瘤的重量和大小也随着UTP11的损耗而减少(图29和30)。这些结果表明,靶向UTP11在体外和体内均能抑制肿瘤生长,提示UTP11可能是癌症治疗的潜在靶点。
UTP11缺陷介导的肿瘤抑制可不依赖于p53
接下来,我们研究了肿瘤抑制效应是否特异针对p53野生型的癌细胞。为了验证这一想法,我们使用了等基因结直肠癌细胞系HCT116 p53+/+和HCT116 p53-/-。
与乳腺癌中的结果一致,细胞活力(图31)、集落形成(图33)和transwell试验(图37)实验均显示UTP11的敲低显著抑制了HCT116 p53+/+细胞的生长和侵袭。
然而令人意外的是,UTP11敲除也减少了HCT116 p53-/-细胞的生长和侵袭(图32,图34和图38)。
这些结果表明,其他独立于p53的机制可能有助于UTP11的功能。值得注意的是,细胞周期的调节似乎需要p53的活性,因为UTP11的耗尽选择性地在HCT116p53+/+细胞中诱导G1的阻滞(图35),而在HCT116 p53-/-细胞中则没有(图36)。
这可能是由于p53依赖的p21和其他细胞周期基因的上调(图11-图18)。此外,我们发现UTP11缺失不仅能够有效抑制HCT116 p53+/+细胞源性异种移植瘤的生长(图39-图42),而且同样能够抑制HCT116 p53-/-细胞源性肿瘤的生长(图43-图46)。
综上所述,这些结果表明靶向UTP11可以通过p53依赖和非依赖机制抑制癌症的发展。
讨论
癌细胞维持高度活跃的核糖体生物发生,以实现其快速生长和增殖。越来越多的证据表明,该过程的靶向该组分可能是癌症治疗的一种潜在策略。
本发明的结果表明,UTP11在乳腺癌和结直肠癌中普遍过表达,较高水平的UTP11与较差的预后相关。本发明还证实了UTP11缺失通过p53依赖和非依赖的机制在体外和体内抑制癌症生长。
综上所述,本发明揭示了UTP11在调节p53依赖和非依赖癌细胞生存和生长中的致癌作用。
UTP11也可能在非癌症疾病中发挥作用。有报道称UTP11在海马体神经元中表达,UTP11的下调可能与阿尔茨海默病有关,但其分子基础尚不清楚。我们的研究为UTP11在神经退行性疾病中的致病作用提供了一种潜在机制,因为核仁改变和p53诱导的细胞死亡与这些疾病的病因学相关。因此,本发明的研究揭示了UTP11通过调节核糖体生物合成和p53活性在癌症和非癌症疾病中的新机制。
在大约50%的人类癌症中,由于基因缺失、截断或错义突变,TP53基因失活。野生型p53的失活通常会导致更具侵袭性和耐药性的肿瘤。在本发明中,意外发现在没有p53的情况下,消融UTP11也能抑制癌细胞的生长(图32,图34,图38,图43-图46)。因此,UTP11作为治疗肿瘤的靶点可以用于治疗p53突变的恶性或耐药肿瘤。
总之,本发明确定了UTP11是一种致癌蛋白,在乳腺癌和结直肠癌中过度表达并与不良预后相关。这些发现表明UTP11是癌症治疗的靶点,不仅能够用于p53野生型肿瘤的治疗,而且能够用于p53突变型肿瘤的治疗。

Claims (10)
1.一种UTP11基因或其编码蛋白的抑制剂的用途,其特征在于,用于:
(1)制备预防或治疗肿瘤的药物;
(2)制备激活p53的试剂;
(3)制备通过p53依赖的机制抑制肿瘤的药物;或者
(4)制备通过p53非依赖的机制抑制肿瘤的药物。
2.如权利要求1所述的用途,其特征在于,所述的UTP11基因选自下组:
(A)编码如SEQ ID NO.2所示多肽的多核苷酸序列;
(B)如SEQ ID NO.1所示的多核苷酸序列;
(C)将SEQ ID NO.1所示的多核苷酸序列经过一个或几个核苷酸的取代、缺失或添加而形成的多核苷酸序列;
(D)序列与SEQ ID NO.:1所示的多核苷酸序列相比,同源性≥90%,较佳地≥95%,更佳地≥98%,最佳地≥99%的多核苷酸序列;
(E)与(A)-(D)任一所述的多核苷酸序列互补的多核苷酸序列。
3.如权利要求1所述的用途,其特征在于,所述UTP11基因的抑制剂为:UTP11基因特异性的siRNA或其前体、UTP11基因特异性的microRNA或其前体、抑制UTP11基因启动子的抑制剂、或其组合。
4.如权利要求1所述的用途,其特征在于,所述UTP11基因编码蛋白的抑制剂选自下组:UTP11基因编码蛋白的抗体、UTP11基因编码蛋白的结合蛋白。
5.如权利要求1所述的用途,其特征在于,所述肿瘤为p53野生型肿瘤,或者所述肿瘤为p53突变型肿瘤。
6.一种药物组合物,其特征在于,包括药学上可接受的载体和有效量的活性成分,其中所述的活性成分为UTP11基因或其编码蛋白的抑制剂。
7.一种体外非治疗性抑制肿瘤细胞的方法,其特征在于,包括步骤:在UTP11基因或其编码蛋白的抑制剂存在的条件下,培养肿瘤细胞,从而抑制抑制肿瘤细胞。
8.一种靶向UTP11基因的siRNA,其特征在于,所述siRNA选自下组:
siUTP11-1:5'-GAAGCTAAGAAAATCGAAA-3',和
siUTP11-2:5'-GGATGGAGTACATATTATT-3'。
9.一种siRNA前体(shRNA),其特征在于,所述siRNA前体是权利要求8所述的siRNA的前体;优选地,所述siRNA前体的5’至3’端依次包括:第一序列单元、茎环序列单元和第二序列单元,所述第一序列单元和所述第二序列单元互补使得所述siRNA前体形成发夹结构,并且所述第一序列单元选自下组:
5'-GAAGCTAAGAAAATCGAAA-3',和
5'-GGATGGAGTACATATTATT-3'。
10.一种敲减细胞中UTP11基因表达水平的方法,其特征在于,包括步骤:在权利要求8所述的siRNA存在的条件下培养细胞,从而实现敲减所述细胞中UTP11基因的表达水平。
