The washing products prepared in examples 1 to 9 and comparative examples 4 to 5 were scored and rated for irritation, and specific test methods were referred to the "cosmetic safety Specification" (2015 edition) acute eye irritation/corrosiveness test, and the specific procedures were as follows:
1. experimental animals: the number of healthy adult white rabbits, 33, females, 1.8-2.2kg, is divided into 11 groups of 3.
2. The experimental steps are as follows:
adapting the environment of a healthy adult white rabbit in an experimental animal house for 12 hours; both eyes of the test animals were examined (including examination with sodium fluorescein) within 24 hours before the start of the test. Animals with symptoms of eye irritation, corneal defects, and conjunctival lesions cannot be used for the test.
The lower eyelid of the rabbit's eye was gently pulled open, and 0.1mL of the composition was instilled into the conjunctival sac, and the upper and lower eyelids were passively closed for 1s to prevent loss of the test substance. The other eye was not treated as a self-control. The test subjects were instilled into the eyes of rabbits for 3 minutes, and then rinsed with a sufficient amount of water at a relatively rapid flow rate without causing damage to the eyes of animals for at least 30 seconds, and then examined and scored as shown in Table 1, and the eye irritation response grade of the products was judged as shown in Table 2.
TABLE 1
TABLE 2
The results show that the compositions of examples 1-9 of the present application are tested: the scores of cornea, iris, conjunctiva and edema are all 0, and the cornea, iris, conjunctiva and edema are nonirritating; the cleaning product of comparative example 4 appeared in the test: the integral of cornea and iris of the rabbit is equal to 0, the integral in conjunctival fluid is 0.5, and the integral is 0 within 0.5 day, which belongs to microstimulation; whereas the commercial care product of comparative example 5 appeared in the test: the integral of cornea, iris and conjunctiva of rabbit falls to 0 in 5 days, which is light-stimulative. Therefore, compared with the commercial washing and caring products, the composition provided by the application is milder to eyes and skin, has no irritation, and is very friendly to sensitive skin people.
2. Evaluation of antiallergic efficacy
The compositions of examples 1-9 and comparative examples 1-5 were tested for sensitization using the Elson-Morgan method, which comprises the following steps: 0.1mL of 2.5mmol/L CaCl was added 2 Adding into 0.5mL of hyaluronidase (600U/mL), and maintaining at 37deg.C for 20min; adding the composition0.5mL, heat preservation at 37 ℃ for 20min; adding 0.4mg/mL sodium hyaluronate 2mL, and preserving heat at 37 ℃ for 40min; adding 1mL of acetylacetone solution, heating in boiling water bath for 15min, and immediately cooling with ice water for 5 mm; finally, ehrlich reagent 1.0mL was added, and the mixture was left at room temperature for 60min to develop color, and the absorbance at 530nm was measured. The calculation formula for measuring the inhibition rate of the sample to the hyaluronidase is as follows:
hyaluronidase inhibition ratio (%) = [ (a-B) - (C-D) ]/(a-B) ×100%
Wherein A represents: absorbance of control solution (acetate buffer instead of sample solution), B represents: absorbance of control blank solution (acetate buffer instead of sample solution and enzyme solution), C represents: absorbance of the sample solution, D represents: absorbance of sample blank (acetate buffer solution instead of enzyme solution).
The number of parallel samples for each group of samples was five, and the average hyaluronidase inhibition rate for each group was calculated and recorded in table 3.
3. Capsaicin stimulation test
Capsaicin stimulation tests were performed on volunteers using the washes of examples 1-9 and comparative examples 1-5, as follows: subjects 140 were invited to age 21 to 42 years, randomized into 14 groups of 10 subjects each using the same test sample, the first group of subjects using the mask of example 1, the second group of subjects using the mask of example 2, and so on. The test area was at the nasolabial sulcus on the right side of the volunteer. Applying 0.01% capsicum alkali solution to the nasal labial sulcus of the right side of the volunteer, when the tingling sensation reaches more than a moderate degree, applying a mask to a test area, observing the redness degree of the test area, evaluating the change of the tingling sensation degree, recording the tingling sensation value of the test area after applying the mask for 60min, wherein the score range is 0-3, the score is 0, the tingling sensation value is not felt, 1 is mild tingling, 2 is moderate tingling, 3 is heavy tingling, and the average tingling sensation value of each group is calculated and recorded in table 3.
4. Testing of skin moisture loss (TEWL)
Volunteers using the washes of examples 1-9 and comparative examples 1-5 were tested for skin moisture loss (TEWL) as follows: subjects 420 were invited to age 21 to 42 years, randomized into 14 groups of 30 subjects each using the same test sample, the first group of subjects using the mask of example 1, the second group of subjects using the mask of example 2, and so on. The preparation is used once in the morning and evening every day for 4 weeks. Require sitting still and remaining relaxed in the test environment before each acquisition, test TEWL values with a Vapometer percutaneous moisture loss meter for 30 min; the Δtewl calculation formula is:
△TEWL=(TEWL 1 -TEWL 2 )/TEWL 1 ×100%
in TEWL (TeWL) 1 : values before the product is not used; TEWL (Tewl) 2 : the values after 4 weeks of use of the product;
the larger Δtewl indicates that the larger the percutaneous water loss reduction range is, the better the repair effect is. The average Δtewl value for each group was calculated and recorded in table 3.
5. Antibacterial property test
Antibacterial properties of the cleaning products prepared in examples 1 to 9 and comparative examples 1 to 4 were tested by a suspension quantification method, and test strains were staphylococcus aureus ATCC6538 and staphylococcus epidermidis ATCC12228, respectively. Preparation of the concentration was about 1X 10 6 Fresh test bacterial liquid of CFU/mL. The culture medium is commercial pancreatic protein soybean agar dry powder culture medium. The culture medium is prepared and then sterilized by high-pressure steam and cooled to 40-45 ℃ for standby. The specific test steps are as follows:
1. 5.0mL of each of the test samples of examples 1-9 and comparative examples 1-5 was aspirated and placed in a sterile test tube, and the temperature was kept constant at 20℃for 5 minutes;
2. sucking 0.1mL of test bacterial liquid, adding into a test tube containing 5.0mL of sample, rapidly mixing, timing immediately, and selecting action time (such as 30min, 1h, 1d, etc.) according to antibacterial components and product characteristics;
3. after the test bacteria and the sample react for a set time, 0.5mL of mixed solution of the test bacteria and the sample is taken and added into a sterilized 0.03mol/L Phosphate Buffer Solution (PBS) containing 4.5mL, and the mixture is uniformly mixed;
4. after 10 minutes of standing, 1mL of sample liquid is sucked (or diluted appropriately, 2-3 dilutions of the sample liquid are taken) and placed in a sterile plate, pancreatic protein soybean agar culture medium cooled to 40-45 ℃ is poured, the plate is rotated to be fully and uniformly mixed, the plate is turned over after agar is solidified, and viable bacteria colony counting is carried out after 2-3 days of culture in a biochemical incubator at 30-35 ℃;
5. replacing the test sample with sterilized PBS, and performing the above steps to obtain a control sample;
6. experiments were repeated 3 times and their average value was calculated;
7. survival (%) = A1/a2×100%, A1 is the number of colonies of the control sample, and A2 is the number of colonies of the test sample.
The results of the test for the survival rate of staphylococcus epidermidis and the survival rate of staphylococcus aureus are shown in Table 3.
TABLE 3 Table 3
By combining examples 1-3, comparative examples 1-5 and tables 1-3, it can be seen that the composition prepared from the waterweed extract, the privet fruit oil, the water temple laver polysaccharide and the conditioner has the effects of no stimulation and no tears, has strong antiallergic activity, can establish a stable skin protection barrier on the surface of sensitive skin, can avoid invasion of harmful miscellaneous bacteria (staphylococcus aureus), can provide a promoting effect for growth and survival of beneficial bacteria (staphylococcus epidermidis), can regulate the distribution of skin bacteria, and can remarkably improve the capability of sensitive skin against interference of harmful factors in external environment. The lack of either component in the composition can negatively impact the antiallergic activity or/and barrier repair effects of the skin.
By combining examples 1, 4-7 and tables 1-3, it can be seen that by adding a plant complex comprising rice extract and velvet flower extract and controlling the mass ratio of the two, the antiallergic activity of the composition, the regulation of the skin surface flora and the protection of the skin, and the stability of sensitive skin can be further improved without affecting the non-irritating and non-lacrimating properties of the composition.
In combination with examples 1, 8-9 and tables 1-3, it can be seen that the interaction between the two components of rice extract and velvet flower extract is important for further enhancing the antiallergic activity of sensitive skin and cannot be simply replaced.
The present embodiment is only for explanation of the present application and is not to be construed as limiting the present application, and modifications to the present embodiment, which may not creatively contribute to the present application as required by those skilled in the art after reading the present specification, are all protected by patent laws within the scope of claims of the present application.
