Abstract
本发明提供了一种腹主动脉瘤动物模型的制备方法与应用。本发明首先提供一种腹主动脉瘤动物模型的制备方法,该方法包括:施予动物有效量的血管紧张素II‑1型受体自身抗体,以制备得到腹主动脉瘤动物模型。本发明还提供了所述的方法制备得到的腹主动脉瘤动物模型在筛选和/或制备用于防治腹主动脉瘤的药物中的应用。

技术领域
本发明是关于一种动物模型的制备技术,具体地说,是关于一种腹主动脉瘤的制备方法与相关应用。
背景技术
腹主动脉瘤(abdominal aortic aneurysm,AAA)是一种异常凶险且死亡率极高的大血管疾病,特征为腹主动脉管壁呈现局限性、永久性扩张。当血管直径超过3cm,或者超过临近血管直径的50%,即可诊断为腹主动脉瘤。AAA男性患病率为1.9%-18.5%,女性为0%-4.2%。随着直径增大,动脉瘤破裂风险增加。腹主动脉瘤破裂前无症状,一旦破裂,致死率高达90%。目前手术是AAA患者的主要治疗方式,包括腔内修复术和开放手术等,尚无明确适应证为AAA的药物上市。因此,深入探究 AAA的病因和病理机制,为临床提供有效的药物治疗靶点和防治手段具有十分重要的意义。
目前,AAA研究中常用的造模方式包括血管紧张素II(angiotensin II, AngII)皮下埋泵、CaPO4血管周围涂抹、弹力蛋白酶血管内灌注等。肾素-血管紧张素系统(rennin-angiotensin-aldosterone system,RAS)的异常激活在AAA发病过程中占据重要地位。作为RAS的主要效应分子,AngII在AAA的形成、进展及破裂中起关键作用。使用ApoE-/-小鼠进行AngII皮下埋泵诱导AAA是最经典、应用最为广泛的AAA造模方式。研究数据显示,AngII诱导的AAA模型重复性较高,具有与人类AAA众多相似的病理特征:血管壁炎性细胞浸润、细胞外基质降解、平滑肌细胞丢失、弹力板断裂、主动脉扩张甚至瘤体破裂,且常有动脉粥样硬化和血栓等伴随发生。机制上,AngII通过作用于血管紧张素II-1型受体(angiotensin IItype 1 receptor,AT1R),使其及下游信号过度活化,引起血管损伤,从而诱发小鼠发生AAA。临床样本研究结果显示,与正常主动脉相比,AAA组织中AT1R表达水平升高。敲除小鼠全身的AT1R可完全阻断AAA的形成,且AT1R拮抗剂(ARB类药物)治疗效果优于抑制AngII生成的血管紧张素转化酶抑制剂(angiotensin converting enzyme inhibitor,ACEI)。提示可能存在其他激活AT1R的物质参与AAA的发生。
然而,AAA模型的成功率及重复性仍有待进一步提高。
发明内容
本发明的一个目的在于提供一种腹主动脉瘤动物模型的制备方法。
本发明的另一目的在于提供腹主动脉瘤动物模型的相关应用。
本案发明人在研究中发现,血管紧张素II-1型受体自身抗体(autoantibodyagainst angiotensin II type 1 receptor,AT1-AA)与腹主动脉瘤(abdominal aorticaneurysm,AAA)的发生相关。AT1-AA为二十世纪初发现的在子痫前期患者血清中存在的一种针对 AT1R胞外第二环(the second extracellular loop of AT1R,AT1R-ECL2)的自身抗体。现有技术已知AT1-AA通过激活AT1R,触发下游 IP3/Ca2+和 DAG/PKC信号通路,引起胞内Ca2+升高,从而发挥类AngII的受体激动剂样效应。近十几年的研究发现,AT1-AA分布于多种血管疾病患者体内,且能够促进疾病的发生和发展,如高血压、急性冠脉综合征、外周动脉疾病等。但是现有技术中并没有关于AT1-AA与腹主动脉瘤关系的研究报道。本案发明人通过对小鼠进行AAA造模的方式,验证AT1-AA与AAA发生之间的关系。并且,为了弥补单一模型的不足,本发明同时进行AngII皮下埋泵和CaPO4血管周涂抹两种模型进行研究,研究结果显示,AT1-AA可能通过增加管壁炎症和基质金属蛋白酶MMP表达加重AAA。进而,本发明提出了一种新的制备腹主动脉瘤动物模型的方法与相关应用。

根据本发明的具体实施方案,本发明的腹主动脉瘤动物模型的制备方法中,所述血管紧张素II-1型受体自身抗体是与血管紧张素II皮下埋泵和/或CaPO4血管周围涂抹方式联合应用于制备腹主动脉瘤动物模型。
根据本发明的具体实施方案,本发明的腹主动脉瘤动物模型的制备方法中,所述动物为小鼠。
根据本发明的具体实施方案,本发明的腹主动脉瘤动物模型的制备方法包括:
ApoE-/-小鼠,使用血管紧张素II-1型受体自身抗体进行尾静脉注射,4周后进行血管紧张素II皮下埋泵,血管紧张素II-1型受体自身抗体仍持续注射,再经4周后制备得到腹主动脉瘤动物模型。
根据本发明的具体实施方案,本发明的腹主动脉瘤动物模型的制备方法包括:
C57BL6/J小鼠,使用血管紧张素II-1型受体自身抗体进行尾静脉注射,4周后进行手术,分离腹主动脉后,以CaCl2联合PBS,对腹主动脉管壁外周进行湿敷,手术结束后14天制备得到腹主动脉瘤动物模型。
根据本发明的具体实施方案,本发明的腹主动脉瘤动物模型的制备方法中,所述腹主动脉瘤动物模型具有增加的腹主动脉的最大直径和/或弹力板断裂程度。

另一方面,本发明还提供了所述的方法制备得到的腹主动脉瘤动物模型在筛选和/或制备用于防治腹主动脉瘤的药物中的应用。
在本发明的一些具体实施方案中,本发明的方法制备得到的腹主动脉瘤动物模型,具有更高的腹主动脉瘤发生率。并且,动物体重在模型组与对照组间没有明显差异。血压监测结果显示,AT1-AA不会引起 Ang II输注的小鼠血压的进一步升高。生存情况无统计学差异。甘油三酯、总胆固醇、高/低密度脂蛋白含量均无差异。
在本发明的一些具体实施方案中,AT1-AA与AngII联合使用,增加了小鼠腹主动脉的最大直径、弹力板断裂程度。
在本发明的一些具体实施方案中,AT1-AA增加了AngII介导的血管炎症及基质金属蛋白酶水平,具体表现在:AT1-AA+AngII联合应用造模,GO分析发现炎症反应和细胞外基质相关基因水平上调; AT1-AA与Ang II联合使用,小鼠腹主动脉血管壁的炎症水平升高;AT1-AA与AngII联合使用,小鼠腹主动脉血管壁的MMP表达增加。
在本发明的一些具体实施方案中,AT1-AA增加了CaPO4诱导腹主动脉瘤血管最大直径、弹力板断裂程度。
综上所述,本发明提供了一种制备腹主动脉瘤动物模型的方法与相关应用,本发明的方法制备得到的AAA模型发生率高,且模型具有与人类AAA众多相似的病理特征。并且,本发明制备得到的模型具有增加的管壁炎症和基质金属蛋白酶MMP表达,加重了AAA,本发明的模型可应用于AAA病因和病理机制研究,筛选防治AAA的药物,对于为临床提供有效的药物治疗靶点和防治手段具有十分重要的意义。

具体实施方式
为了对本发明的技术特征、目的和有益效果有更加清楚的理解,现对本发明的技术方案进行以下详细说明,但不能理解为对本发明的可实施范围的限定。下文各实验中所用各原始试剂材料均可商购获得,未注明具体条件的实验方法为所属领域熟知的常规方法和常规条件,或按照仪器制造商所建议的条件。
除非另外专门定义,本文使用的所有技术和科学术语都与相关领域普通技术人员的通常理解具有相同的含义。
实施例中所用具体实验方法:
1、AngII皮下埋泵
(1)Alzet渗透性微泵(2004 model)浸泡于无菌生理盐水,37℃浸泡48小时。血管紧张素II(1000 ng/kg/min输注速率,持续28天)用生理盐水溶解,注入渗透性微泵。
(2)小鼠1%戊巴比妥钠腹腔注射麻醉后,酒精消毒右侧耳后,剪开约1厘米皮肤,用止血钳分离背部皮下组织,在右侧后背形成一个长约3厘米的皮下孔腔。将渗透性微泵从切口后处送入背部空腔中,头端朝向尾侧,缝合切口。
2、 CaPO4诱导腹主动脉瘤造模
分离暴露腹主动脉后对照组以0.5 mol/L NaCl 溶液纱布管周湿敷 10 分钟,然后以PBS缓冲液湿敷5分钟,其余实验组以0.5 mol/L CaCl2溶液纱布管周湿敷10分钟后PBS 缓冲液湿敷5分钟。
3、免疫组化
(1)水化:PBS洗 3 min,3次。
(2) 3% H2O2甲醇封闭内源性过氧化物酶 15 min,避光。
(3) PBS洗 3 min,3次。
(4)滴加 10%羊血清封闭 37℃ 1 hr。
(5) 滴加抗小鼠 CD4,CD45,CD68抗体 37℃ 1 hr,然后 4℃过夜。
(6) PBS洗 3 min/次,3次。
(7) 滴加二抗 37℃ 1 hr。PBS洗 3 min/次,3次。
(8)DAB显色 3~10 min。PBS中止。
(9)脱水,透明,封片,镜检。
4、免疫荧光
(1)将组织片用PBS洗三遍。
(2)4%的多聚甲醛室温固定15 min。
(3)滴加10%羊血清封闭37℃ 1 hr。
(4)滴加MMP2和MMP9抗体 37℃1 hr,然后4℃过夜。
(5)PBS洗3 min/次,3次。
(6)加入荧光二抗避光孵育37℃ 1 hr。PBS洗3 min/次,3次。
(7)用含DAPI的封片剂封片。
5、实时荧光定量PCR
5.1 血管组织中RNA提取
取小钢珠用固相RNAse清除剂浸泡过夜,次日置于烘箱内烘干。血管组织置于2 mlEP管中,放入钢珠,加入500 μl Trizol,使用匀浆机匀浆1-2个循环后,取出EP管,将钢珠用磁石沿管壁吸出,EP管冰上静置2-3分钟。
(1)向上述每个EP管中加入氯仿(0.2 ml 氯仿/1 ml Trizol),剧烈晃动15秒,冰上静置10分钟。
(2)4 ℃,12000 RPM,离心15分钟。混合物分为有机相、中间相、上层水相,RNA保留在上层水相中。
(3)将<80%的水相转移到新的2 ml EP管中,加入等体积的无水乙醇,涡旋。
(4)将<700 μl体积的步骤3)中的混合物加入RNA mini柱上,室温,10000g,离心30-60秒,弃去下层流出液。
(5)重复步骤4),再次向mini柱中加入剩下混合物,10000g,离心30-60秒,弃去流出液,至所有混合物加完。
(6)加入500 μl RNA洗涤缓冲液I,室温,10000g,离心60秒,弃去下层流出液。
(7)加入500 μl RNA洗涤缓冲液II,室温,10000g,离心60秒,弃去下层流出液。
(8)重复步骤7)。
(9)RNA mini柱放入倒空的收集管中,13000g,离心2分钟,高速旋转完全干燥RNAmini柱。
(10)总RNA洗脱,将RNA mini柱转移到新的1.5 ml 无菌EP管中,用20-30 μl DEPC水洗脱RNA,确保DEPC水直接加入mini柱的基质中,室温放置2分钟,13000 RPM,离心1分钟。离心下来的水吸出再次加入mini柱基质中,再次离心。洗脱时DEPC水应多浸润一会儿再离心。若RNA预产量>20 g,则可进行二次洗脱。
(11)RNA浓度测定。
5.2 RT-qPCR反转录成cDNA
(1)取无菌1.5 ml EP管,设计逆转录总质量为2 μg,依据样品浓度计算加样体积,每管中加入1 μl的Random Hexamer Primer,再用无菌水补至12 μl。
(2)65 ℃金属浴5分钟。
(3)取出EP管立即冰上静置2分钟。
接着每管依次加入以下试剂:
5×Reaction buffer 4 μl
Ribolock Rnase Inhibitor 1 μl
10 mM dNTP Mix 2 μl
RevertAid M-MuLV RT 1 μl
总体积至20 μl。
(4)轻轻混匀,并瞬离,室温静置10分钟。
(5)42 ℃水浴锅内孵育1小时。
(6)70 ℃金属浴内5分钟,终止反应。
(7)冰上静置2-3分钟,直接扩增或者-80 ℃保存备用。
5.3 RT-qPCR扩增流程
(1)配制反应体系:

计算:目的基因的表达量=2-ct(表示实验组目的基因较对照组的变化倍数)。ct=(ct(目的基因)-ct(管家基因))实验组-(ct(目的基因)-ct(管家基因))对照组。
6、Western blot技术检测蛋白表达水平
6.1 缓冲液配制:
丙烯酰胺贮备液:称取29 g丙烯酰胺及1 g甲叉双丙烯酰胺,加入50 ml双蒸水中溶解,定容至100 ml,置入棕色瓶中4 ℃保存。可水浴加热至37 ℃助溶。
1.5 mol/L Tris-HCl (pH8.8):称取Tris 18.16 g,充分溶解在80 ml双蒸水中,用HCl调节pH至8.8,用双蒸水定容至100 ml,4 ℃保存。
1.0 mol/L Tris-HCl (pH6.8):称取Tris 12.12 g,充分溶解在80 ml双蒸中,用HCl调节pH至6.8,用双蒸水定容至100 ml,4 ℃保存。
10% SDS溶液:称取SDS 10 g,充分溶解在100 ml双蒸水中,室温保存。使用前如有结晶,用37 ℃水浴溶解。
10% 过硫酸胺溶液:称取0.1 g过硫酸胺,加入1 ml双蒸水中,4 ℃保存。
TEMED:原液。
电泳缓冲液(5×):称取37.75 g Tirs;235 g 甘氨酸;12.5 g SDS,充分溶解在2000 ml双蒸水中,定容至2500 ml,室温保存。使用时双蒸水稀释成1×工作液。
电转缓冲液(10×):称取75.7 g Tirs;360.8 g 甘氨酸,加入2000 ml双蒸水中,充分溶解后定容至2500 ml,室温保存。使用时,按照双蒸水:甲醇:电转液=7:2:1的比例配制成1×的电转缓冲液。
洗涤缓冲液(Tris Buffered Solution Tween,TBST):称取24.23 g Tris;80.06g NaCl,加入900 ml双蒸水中,用HCl调节pH值至7.6,充分溶解后定容至1000 ml,制备成10×TBS溶液,室温保存。取100 ml 10×TBS溶液,加入900 ml双蒸水,再加入1 ml Tween-20,充分混匀,制备成TBST,室温存放。
6.2 实验步骤
1) 样品制备。本部分研究所用Western blot样品为组织匀浆样品。组织匀浆液制备:将待检血管组织从-80 ℃冰箱中取出,稍微解冻,置于2 ml EP管中。按照20 μl/mg加入组织蛋白裂解液(已预先加入蛋白酶抑制剂和蛋白磷酸酶抑制剂)。在每个EP管中放入两颗小钢珠,EP管盖上做好标记后,安装在组织匀浆仪上进行匀浆。匀浆后,将小钢珠用磁铁吸出,将EP管中样品进行离心,4 ℃,12000 RPM离心15分钟。收集上清至新的1.5 ml EP管中,做好标记。处理好的蛋白样品保存于-80 ℃冰箱,尽量减少反复冻融。样品标化。为了便于分析统计,需要在电泳前对样品进行标化处理。首先通过BCA试剂盒对待测样品的蛋白浓度进行测定,然后计算使各组蛋白上样量和上样体积保持一致,一般达到蛋白总上样量15-20μg,上样体积在10-20 μl。混合加入5×蛋白上样缓冲液,99 ℃煮沸10分钟使蛋白质变性,离心使样品聚集后备用。
2) 凝胶配制。根据被检测蛋白分子量大小选择需要配制凝胶的浓度,本部分实验使用10%的分离胶,选用1.5 mm玻璃板。按照表1的配方配制凝胶。先配制10%分离胶,将配好混匀的凝胶慢慢灌入搭建好的模具中,上层加异丙醇压胶。约20分钟后,待分离胶凝固,倾斜模具吸出异丙醇,用滤纸小心吸去残余异丙醇,而后配制和灌注5%的积层胶,灌胶后慢慢插入10孔梳子,防止梳子与凝胶之间产生气泡。待胶凝固后,将其组装入电泳装置,短板向里,槽内加注新配制电泳液后轻轻拔去梳子,即可用于蛋白上样和电泳。

3) 上样及电泳。将待测样品按照设计好的顺序加入样品孔中,第一孔和第10孔加入预染Marker 3 μl,再用1×蛋白上样缓冲液补齐至和样品孔上样体积一致。若有多余孔道加入等体积1×蛋白上样缓冲液。正确连接正负极,开始电泳时,设定参数恒压为80 V,开始电泳,预电泳30分钟。当样品开始进入分离胶后,将电压调到120 V,继续电泳,当溴酚蓝抵达分离胶底部时停止电泳。
4) 蛋白质印迹转移电泳。准备滤纸和PVDF膜,大小与胶一致,将PVDF膜提前置于无水甲醇中约30秒钟,使其充分活化。然后将滤纸、纤维垫、膜和凝胶置于电转液中浸泡5分钟。制作转膜三明治,由下往上(负极(黑板)到正极(白板))按下列顺序依次安放:黑板—纤维垫一滤纸一凝胶一PVDF膜一滤纸一纤维垫一白板。电转槽电极板中放入电转液,盖好盖子(注意电极),将转膜装置放入冰水槽中保持低温,接通电源,设置转膜条件为恒定电流400 mA,转膜1小时。
5) 封闭。电转结束后,取出膜,将其放入脱脂奶粉配制的封闭液中,室温摇床孵育1小时。
6) 一抗孵育。封闭结束后,将膜用TBST洗2次,每次10分钟,以充分洗去奶粉。按照待测蛋白分子量裁膜,而后放入预先铺有疏水性封口膜的平皿中,滴加配好的抗体,抗体按照抗体说明书比例用TBST配制。平皿盖好盖子,轻轻平放入4 ℃冰箱,孵育过夜。
7) 二抗孵育。次日取出膜,回收一抗。膜用TBST洗3遍,每次10分钟。之后放入二抗中孵育。二抗是种属特异性的抗体,依据一抗的来源选择孵育的二抗种类。按照说明书建议比例用TBST稀释,室温摇床晃动孵育1小时。
8) 显影。二抗孵育结束后,用TBST将膜洗3遍,每次10分钟。取化学发光液A液、B液1:1混匀,配成发光液工作液。将膜稍沥干,均匀滴加发光液,使用Bio-Rad的凝胶成像系统对目的条带进行成像。因二抗上标记有HRP,经二抗孵育后的膜在滴加发光液后可被催化发光,从而显示出待检测蛋白的条带。
9) 结果分析。蛋白的表达量用显色条带的灰度值来反应。分析统计时,样品条带灰度值需用内参条带灰度值做标化后才有对比意义。
实施例1、AT1-AA加重AngII介导的腹主动脉瘤的发生。
将8周雄性ApoE-/-小鼠随机分为4组:对照组,AT1-AA(20 μg/g)组,AngII(1000ng/kg/min)组,AngII+AT1-AA联合组。具体流程为,使用AT1-AA或生理盐水进行小鼠尾静脉注射,4周后进行AngII皮下埋泵,AT1-AA和生理盐水仍持续注射,每周一次,再经4周后取材。
AT1-AA获得方式见参考文献(《Wei M, Zhao C, Zhang S, Wang L, Liu H, MaX. Preparation and Biological Activity of the Monoclonal Antibody against theSecond Extracellular Loop of the Angiotensin II Type 1 Receptor.》J ImmunolRes. 2016;2016:1858252.)所述方法。
结果参见图1。结果显示,对照组、AT1-AA组全部小鼠都没有发生动脉瘤(0/15);与AngII埋泵组相比,AngII+AT1-AA联合组腹主动脉瘤发生率显著增加(47% vs.81%);另外,在AngII组,动脉瘤的破裂率为16%(3/19),2(10%)只小鼠发生胸主动脉瘤;在AT1-AA+AngII联合组,有29%(6/21)小鼠死于动脉瘤破裂,5(24%)只小鼠发生胸主动脉瘤。
小鼠的体重在各组间没有明显差异(图2A)。血压监测结果显示,AT1-AA不会引起Ang II输注的小鼠血压的进一步升高(图2B)。生存情况无统计学差异(图2C)。甘油三酯、总胆固醇、高/低密度脂蛋白含量均无差异(图2D)。
将AngII埋泵4周后的小鼠用异氟烷气体麻醉,腹部进行脱毛,用Veov 770TM图像系统采集腹部超声图像,测量并统计血管的最大直径。取材大体图见(图3A),腹主动脉瘤发生率统计图见(图3B)。超声统计结果显示,与对照组相比,AT1-AA组血管的直径没有明显改变,AngII输注的小鼠血管直径明显增加,而AT1-AA+AngII联合组进一步增加(图3C)。
接下来,使用血管组织切片对弹力板断裂情况进行评估。评分依据:弹力板的断裂分为1-4级,第1级是弹力板的断裂少于25%,第2级是25~50%的断裂,第3级是50~75%的断裂,第4级是超过75%的断裂。结果显示,对照组的血管壁弹力板结构完整,染色清晰;与对照组相比,AT1-AA组血管壁弹力板的断裂程度没有明显改变,Ang II输注的小鼠血管弹力板断裂明显增加,而AT1-AA+Ang II联合组断裂情况进一步加重(图3D)。
使用小鼠的主动脉血管组织进行取材,样品送至华大基因进行RNA-seq分析,结果显示,与生理盐水组相比,AT1-AA+AngII组共有283个基因表达上调。将上调的283个基因进行GO分析,发现炎症反应和细胞外基质相关基因水平上调(图4)。
利用免疫组化技术,分别通过检测CD4、CD45、CD68分子的表达和分布情况,来确定T淋巴细胞、白细胞、巨噬细胞在血管壁浸润的情况。结果显示,与对照组相比,AngII组的血管壁炎症有所增加,而AT1-AA+Ang II联合组炎症浸润情况明显加重(图5A)。血浆中致炎因子MCP-1和IL-6在动脉瘤血管炎症中起着关键的作用。对这两种关键致炎因子的检测结果显示,AT1-AA+AngII联合组相较于AngII输注组血浆炎症水平也明显增加(图5B)。
利用WB、qRT-PCR及免疫荧光技术,检测了血管组织中MMPs表达的情况。结果显示,与对照组相比, AngII输注组血管壁中MMP9和MMP2的表达水平有所增加,而AT1-AA+Ang II联合组进一步显著增加(图6)。
实施例2、AT1-AA增加CaPO4诱导腹主动脉瘤血管最大直径、弹力板断裂程度
以C57BL6/J小鼠腹主动脉管周浸润CaPO4构建AAA模型,进一步验证AT1-AA在AAA发生中的作用。将8周雄性C57BL6/J小鼠随机分成以下4组:对照组,AT1-AA(20μg/g)组,CaPO4组,CaPO4+AT1-AA组。AT1-AA(20μg/g)或生理盐水尾静脉注射4周(每周一次)后进行手术,分离腹主动脉后,以生理盐水或CaCl2联合PBS,对腹主动脉管壁外周进行湿敷,手术结束后14天取材。
结果显示,与CaPO4组相比,CaPO4+AT1-AA组小鼠腹主动脉直径显著增加(图7A),弹力板断裂程度进一步加重(图7B)。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (9)
1.一种腹主动脉瘤动物模型的制备方法,该方法包括:
施予动物有效量的血管紧张素II-1型受体自身抗体,以制备得到腹主动脉瘤动物模型;
所述动物为小鼠。
2.根据权利要求1所述的方法,其中,所述血管紧张素II-1型受体自身抗体是以尾静脉注射方式施予动物。
3. 根据权利要求1所述的方法,其中,所述血管紧张素II-1型受体自身抗体是以20 μg/g的剂量施予动物。
4.根据权利要求1所述的方法,其中,所述血管紧张素II-1型受体自身抗体是与血管紧张素II皮下包埋缓释泵和/或CaPO4血管周围涂抹方式联合应用于制备腹主动脉瘤动物模型。
5. 根据权利要求1-4任一项所述的方法,该方法包括:
ApoE-/-小鼠,使用血管紧张素II-1型受体自身抗体进行尾静脉注射,4周后进行血管紧张素II皮下埋泵,血管紧张素II-1型受体自身抗体仍持续注射,再经4周后制备得到腹主动脉瘤动物模型;或者
C57BL6/J小鼠,使用血管紧张素II-1型受体自身抗体进行尾静脉注射,4周后进行手术,分离腹主动脉后,以CaCl2联合PBS,对腹主动脉管壁外周进行湿敷,手术结束后14天制备得到腹主动脉瘤动物模型。
6.根据权利要求1所述的方法,其中,所述腹主动脉瘤动物模型具有增加的腹主动脉的最大直径和/或弹力板断裂程度。
7.根据权利要求1所述的方法,其中,所述腹主动脉瘤动物模型具有增强的管壁炎症和/或高表达的基质金属蛋白酶MMPs。
8.根据权利要求7所述的方法,其中,所述腹主动脉瘤动物模型血浆中致炎因子MCP-1和IL-6表达水平上调,和/或血管壁中MMP9和/或MMP2的表达水平上调。
9.权利要求1-8任一项所述的方法制备得到的腹主动脉瘤动物模型在筛选和/或制备用于防治腹主动脉瘤的药物中的应用。
