本发明涉及一种含普鲁兰多糖的鲜切花花期延长剂及其制备方法,属于保鲜剂技术领域。本发明的鲜切花花期延长剂包括普鲁兰多糖、类黄腐酸、氯化钾和苯甲酸钠,各组分共同作用,与市场现有的鲜切花花期延长剂相比,具有用量少、成本低、耐寒耐热和作用持久的特点,在5~25℃范围之间,都具有良好的延长鲜切花花期的效果。本发明的鲜切花花期延长剂配方中不含生理毒性强及污染环境的银盐,高效环保、环境友好;具有抗菌防腐、补充营养物质和保持水分代谢平衡的功效;所述的制备方法科学合理,简单易行。

技术领域
本发明涉及一种含普鲁兰多糖的鲜切花花期延长剂及其制备方法,属于保鲜剂技术领域。
背景技术
鲜切花产业经过多年的发展,已成为许多国家农业的支柱产业之一,成为农业新的经济增长点。每年的情人节一般中型城市的鲜切花消费量都达到几十吨。在鲜花的生理发育过程中,影响鲜花寿命的因素主要包括温度、空气湿度、光照和乙烯等。鲜切花离开母体后,若处理不当,会枯萎,缩短观赏期。按照各种因素在不同时期的影响程度不同,可以将鲜切花保鲜处理分为5个环节,即花卉产品的采后处理、预冷处理、贮藏环境、保鲜营养液和产品包装五个步骤。其中,保鲜营养液在整个鲜切花保鲜过程中发挥着至关重要的作用,同时也是研究的主要热点。玫瑰花因其花朵颜色鲜艳、美丽芳香而深受人们的青睐,但其鲜切花瓶插寿命较短,影响了观赏品质,严重阻碍了它的商品化发展。国内外学者致力于玫瑰鲜切花保鲜方法的研究,但保鲜剂配方大多数是含硝酸银、硫代硫酸银或硫酸铝等成分。这些物质配置工艺复杂、不易保存、成本较高并且有毒,会对人体和环境造成不良的影响。至今为止,鲜切花保鲜剂的研究主要包括抑制乙烯生成、保持水分平衡、减少ABA(脱落酸)含量和增加能源物质等方面,但由于鲜切花品种繁多,且各品种间鲜切花代谢差异大,因此没有通用有效的花期延长剂。以往的保鲜方法多以蔗糖为糖源,对拮抗衰老、杀菌或抗菌方面比较重视,而对水分代谢调节、温度适应等方面鲜见报道。
中国专利文献CN 113383770 A(申请号202110820150.7)公开了一种玫瑰鲜花保鲜方法,包括以下步骤:S1、采摘:于日出后采摘半开的呈酒杯状的玫瑰鲜花;S2、预冷:将采摘的玫瑰鲜花及时送入0~4℃车间内,摊凉预冷1~2小时;S3、速冻:将上述预冷后的玫瑰装入纸箱中,并在-25~-20℃的温度下冷冻15~20h;S4、冷藏:将经过速冻的玫瑰置于0~4℃冷藏,并在玫瑰鲜花的根部包裹保鲜剂。所述保鲜剂包括以下组分:营养剂16~20份,抗氧化剂2~8份,杀菌剂3~5份,表面活性剂1~2份,植物生长调节剂8~10份,金属离子可溶性无机盐12~16份,蜂胶2~3份;蜂蜜1~2份;其中,所述营养剂包括蔗糖、葡萄糖、果糖和壳聚糖中的至少一种,所述抗氧化剂包括维生素C、水杨酸、柠檬酸、抗坏血酸、酒石酸、苹果酸、咖啡酸中的至少一种,所述杀菌剂包括高锰酸钾、亚硫酸氢钠、硼酸、阿斯匹林、抗生素、马来酸、a-琥珀酸、丁二酸、酒精、锰酸钾、8-羟基喹啉、正烷基二甲苄基氯化氨、噻苯咪唑中的任意一种,所述表面活性剂包括聚氧乙烯月桂醚、吐温-20、中性洗衣粉和黄腐酸中的任意一种,所述植物生长调节剂包括细胞激动素、赤霉素和复硝酚钠,所述金属离子可溶性无机盐为氯化钾、磷酸氢钾、碳酸钾、氯化钙、硝酸钙和氯化铝中的任意一种。
上述保鲜剂成分较为复杂,成本较高,因此,一种简单易行,操作方便,无污染,价廉物美、保鲜效果好且能进入普通家庭使用的鲜切花花期延长剂产品是迫切需要的。
发明内容
针对现有技术的不足,本发明提供了一种含普鲁兰多糖的鲜切花花期延长剂,具有抗菌防腐、补充营养物质和保持水分代谢平衡的功效,还具有高效、经济和无污染的特点。
同时,本发明还提供了上述含普鲁兰多糖的鲜切花花期延长剂的制备方法,科学合理,简单易行。
普鲁兰多糖是一种由出芽短梗霉通过发酵产生的天然水溶性多糖,安全性高。其结构特征为:麦芽三糖重复单位是由葡萄糖以α-1,4糖苷键连接而成,普鲁兰多糖是麦芽三糖重复单位之间通过α-1,6糖苷键相互连接而成的高分子聚合物。普鲁兰多糖分子量1.5×104Da~1.0×107Da,其中较高分子量组分有较强的成膜性、阻气性和可塑性,较低分子量的组分可能具有刺激植物生理代谢,增强其耐逆性的功能。
黄腐酸是一类可用于提高水稻耐盐能力的重要生物刺激素,但不可再生的特性很大程度上限制了其在农业上的规模化应用。造纸行业制浆过程排放的黑液已成为重要的污染来源,但其中大量以木质素为代表的有机物质却未得到充分利用而被浪费。虽然有研究证实造纸黑液中的类黄腐酸与天然黄腐酸具有相似的特征官能团且能提高植物的耐盐性,但其应用范围仍有待拓宽。
本发明的技术方案如下:
一种含普鲁兰多糖的鲜切花花期延长剂,每升溶剂包含以下成分:
普鲁兰多糖2-8 g,类黄腐酸50-150 mg-C,氯化钾0.1-0.6g,苯甲酸钠10-50 mg。
根据本发明优选的,所述类黄腐酸的分子量≤10 kDa。
根据本发明优选的,所述含普鲁兰多糖的鲜切花花期延长剂的溶剂为水。
根据本发明优选的,所述含普鲁兰多糖的鲜切花花期延长剂每升溶剂包含以下成分:
普鲁兰多糖3-7 g,类黄腐酸60-120 mg-C,氯化钾0.1-0.6 g,苯甲酸钠10-50 mg。
根据本发明优选的,所述含普鲁兰多糖的鲜切花花期延长剂每升溶剂包含以下成分:
普鲁兰多糖4-6 g,类黄腐酸80-100 mg-C,氯化钾0.2-0.4 g,苯甲酸钠15-35 mg。
本发明中,所述类黄腐酸的含量以其中的碳(C)含量为准,即mg-C。
本发明中,所述普鲁兰多糖可购自山东弥美生物科技股份有限公司,类黄腐酸来源于山东泉林集团有限公司提供的造纸黑液,按照Yao et al (2019)的方法制备,分子量≤10 kDa,其余也均为市售产品。
上述含普鲁兰多糖的鲜切花花期延长剂的制备方法,包括以下步骤:
(1)取苯甲酸钠,加入水中,调节溶液pH至4.0~4.5;
(2)取类黄腐酸和氯化钾,依次加入至步骤(1)所得的溶液中,搅拌均匀,再加入普鲁兰多糖,溶解后搅拌均匀,得到鲜切花花期延长剂。
根据本发明优选的,步骤(1)中调节溶液pH采用磷酸二氢铵、磷酸二氢钾、磷酸二氢钠、碳酸氢钠、碳酸氢钾、碳酸氢铵、硫酸钙、硫酸铝、硫酸铝铵、柠檬酸及其盐、山梨酸、植酸、十八碳酸或硬脂酸及其盐中的一种或几种。
进一步优选的,步骤(1)中调溶液节pH的采用磷酸二氢铵、碳酸氢钠、硫酸钙、山梨酸或植酸中的一种或几种。
上述含普鲁兰多糖的鲜切花花期延长剂在延长花卉鲜切花花期中的应用。
根据本发明优选的,所述花卉为玫瑰、百合花。

上述鲜切花花期延长剂中,普鲁兰多糖可以作为一种免疫诱抗剂,充分利用植物自身对病害发生的潜在控制能力,通过调节植物防卫、代谢系统,诱导产生基础免疫反应,延迟或减轻病害的发生和发展;此外普鲁兰多糖还可以增加叶绿素含量,提高光合作用。
上述鲜切花花期延长剂中,类黄腐酸可以通过提高渗透性调节物质含量和保护酶活性以缓解干旱胁迫,促进鲜切花对养分的吸收利用,还可以降低叶片中碳水化合物含量,维持鲜切花较高的氮代谢过程,保持较高的净光合效率。
上述鲜切花花期延长剂中,苯甲酸钠具有防腐抑菌的作用,pH为4.0~4.5时,能更好地发挥苯甲酸钠的防腐抑菌作用,又不会对鲜切花产生酸胁迫伤害。
上述鲜切花花期延长剂中,氯化钾能增加花瓣细胞的渗透浓度,促进水分平衡,延缓衰老过程。
有益效果:
1、本发明的鲜切花花期延长剂对各种花卉,特别是玫瑰花和百合花,均可起到延长花期的作用;普鲁兰多糖、类黄腐酸、氯化钾和苯甲酸钠共同作用,与市场现有的鲜切花花期延长剂相比,具有用量少、成本低、耐寒耐热和作用持久的特点,在5~25℃范围之间,都具有良好的延长鲜切花花期的效果。
2、本发明的鲜切花花期延长剂配方中不含生理毒性强及污染环境的银盐,高效环保、环境友好;具有抗菌防腐、补充营养物质和保持水分代谢平衡的功效。
3、本发明鲜切花花期延长剂的制备方法科学合理,简单易行,制得的鲜切花花期延长剂效果好。
具体实施方式
下面结合实施例对本发明做进一步的说明,但是本发明的保护范围并不仅限于此。
实施例中,普鲁兰多糖可购自山东弥美生物科技股份有限公司,粘度值≥60mm2/s;类黄腐酸来源于山东泉林集团有限公司提供的造纸黑液,按照Yao et al,MolecularComposition of Size-Fractionated Fulvic Acid-Like Substances Extracted fromSpent Cooking Liquor and Its Relationship with Biological Activity,(2019)的方法制备,分子量≤10 kDa;其余原料均为市售产品。
实施例1
一种含普鲁兰多糖的鲜切花花期延长剂,每升溶剂包括如下成分:
普鲁兰多糖4 g,类黄腐酸80 mg-C,氯化钾0.2 g,苯甲酸钠20 mg;溶剂为水。
上述鲜切花花期延长剂的制备方法,步骤如下:
1)先称取苯甲酸钠20 mg加入1000 mL水中,使用磷酸二氢铵调节溶液pH至4.0;
2)然后称取类黄腐酸(以其中C含量达到80 mg为准)、氯化钾0.2 g加入步骤1)得到的溶液中,搅拌均匀;再加入普鲁兰多糖4 g,完全溶解搅拌均匀后即得鲜切花花期延长剂。
实施例2
一种含普鲁兰多糖的鲜切花花期延长剂,每升溶剂包括如下成分:
普鲁兰多糖5 g,类黄腐酸100 mg-C,氯化钾0.3g,苯甲酸钠35 mg;溶剂为水。
上述鲜切花花期延长剂的制备方法,步骤如下:
1)先称取苯甲酸钠35 mg加入1000 mL水中,使用磷酸二氢铵调节溶液pH至4.0;
2)然后称取类黄腐酸(以其中C含量达到100 mg为准)、氯化钾0.3 g加入步骤1)得到的溶液中,搅拌均匀;再加入普鲁兰多糖5 g,完全溶解搅拌均匀后即得鲜切花花期延长剂。
实施例3
一种含普鲁兰多糖的鲜切花花期延长剂,每升溶剂包括如下成分:
普鲁兰多糖3 g,类黄腐酸60 mg-C,氯化钾0.6 g,苯甲酸钠50 mg;溶剂为水。
上述鲜切花花期延长剂的制备方法,步骤如下:
1)先称取苯甲酸钠50 mg加入1000 mL水中,使用碳酸氢钠调节溶液pH至4.1;
2)然后称取类黄腐酸(以其中C含量达到60 mg为准)、氯化钾0.6 g加入步骤1)得到的溶液中,搅拌均匀;再加入普鲁兰多糖3 g,完全溶解搅拌均匀后即得鲜切花花期延长剂。
实施例4
一种含普鲁兰多糖的鲜切花花期延长剂,每升溶剂包括如下成分:
普鲁兰多糖6 g,类黄腐酸120 mg-C,氯化钾0.3 g,苯甲酸钠15 mg;溶剂为水。
上述鲜切花花期延长剂的制备方法,步骤如下:
1)先称取苯甲酸钠15 mg加入1000 mL水中,使用硫酸钙调节溶液pH至4.2;
2)然后称取类黄腐酸(以其中C含量达到120 mg为准)、氯化钾0.3 g加入步骤1)得到的溶液中,搅拌均匀;再加入普鲁兰多糖6 g,完全溶解搅拌均匀后即得鲜切花花期延长剂。
实施例5
一种含普鲁兰多糖的鲜切花花期延长剂,每升溶剂包括如下成分:
普鲁兰多糖2 g;类黄腐酸50 mg-C,氯化钾0.3g,苯甲酸钠25 mg;溶剂为水。
上述鲜切花花期延长剂的制备方法,步骤如下:
1)先称取苯甲酸钠25 mg加入1000 mL水中,使用山梨酸调节溶液pH至4.3;
2)然后称取类黄腐酸(以其中C含量达到50 mg为准)、氯化钾0.3 g加入步骤1)得到的溶液中,搅拌均匀;再加入普鲁兰多糖2 g,完全溶解搅拌均匀后即得鲜切花花期延长剂。
实施例6
一种含普鲁兰多糖的鲜切花花期延长剂,每升溶剂包括如下成分:
普鲁兰多糖8 g,类黄腐酸150 mg-C,氯化钾0.3g,苯甲酸钠35 mg;溶剂为水。
上述鲜切花花期延长剂的制备方法,步骤如下:
1)先称取苯甲酸钠35 mg加入1000 mL水中,使用植酸调节溶液pH至4.4;
2)然后称取类黄腐酸(以其中C含量达到150 mg为准)、氯化钾0.3 g加入步骤1)得到的溶液中,搅拌均匀;再加入普鲁兰多糖8 g,完全溶解搅拌均匀后即得鲜切花花期延长剂。
对比例1
一种含普鲁兰多糖的鲜切花花期延长剂,每升溶剂包括如下成分:
普鲁兰多糖5 g,氯化钾0.3 g,苯甲酸钠35 mg;溶剂为水。
上述鲜切花花期延长剂的制备方法,步骤如下:
1)先称取苯甲酸钠35 mg加入1000 mL水中,使用磷酸二氢铵调节溶液pH至4.0;
2)然后称取氯化钾0.3 g加入步骤1)得到的溶液中,搅拌均匀;再加入普鲁兰多糖5 g,完全溶解搅拌均匀后即得鲜切花花期延长剂。
对比例2
一种含麦芽糖的鲜切花花期延长剂,每升溶剂包括如下成分:
麦芽糖5 g,类黄腐酸100 mg-C,氯化钾0.3 g,苯甲酸钠35 mg;溶剂为水。
上述鲜切花花期延长剂的制备方法,步骤如下:
1)先称取苯甲酸钠35 mg加入1000 mL水中,使用磷酸二氢铵调节溶液pH至4.0;
2)然后称取类黄腐酸(以其中C含量达到100 mg为准)、氯化钾0.3 g加入步骤1)得到的溶液中,搅拌均匀;再加入麦芽糖5 g,完全溶解搅拌均匀后即得鲜切花花期延长剂。
实验例1
实验对象:香槟玫瑰
形态指标:主要根据花瓣的颜色与膨胀度评分,评分标准如下:
100 分——花色纯正,无色变,鲜艳,膨胀度好(一般指刚采时的状态);
90 分——有轻微的色变,但鲜艳,花色轻微黯淡但纯正;
80分——有轻微色变且花色略黯淡,膨胀度稍差;
70分——色变较明显,花色黯淡或膨胀度较差,出现蓝变,花朵已失去观赏价值但叶色尚好,饱满直立;
60 分——花瓣色变明显,有坏死斑块或斑点,花瓣失水明显,叶片中度萎蔫。
每组固定取6枝鲜切花进行评分,取平均值。
实验过程:温度控制在15~20℃,取实施例1-6及对比例1-2所述的鲜切花花期延长剂。采切的是同一批次的花色、大小、生理状况基本一致且生长健壮的玫瑰花。花枝用消毒剪刀斜剪,以增大吸水面积。保留花枝长度约为30cm,留2~3片叶;然后插入装有不同鲜切花花期延长剂的三角瓶中,每瓶插2枝,每组重复处理3次。用脱脂棉塞紧瓶口,置于室内散射光下,在施用时间第0,4,8天,分别按照上述形态指标进行评价。表1为采用实施例1-6及对比例1-2鲜切花花期延长剂的香槟玫瑰的形态指标比较。

实验例2
实验对象:蓝色妖姬玫瑰
形态指标:主要根据花瓣的颜色与膨胀度评分,评分标准如下:
100 分——花色纯正,无色变,鲜艳,膨胀度好(一般指刚采时的状态);
90 分——有轻微的色变,但鲜艳,花色轻微黯淡但纯正;
80分——有轻微色变且花色略黯淡,膨胀度稍差;
70分——色变较明显,花色黯淡或膨胀度较差,出现蓝变,花朵已失去观赏价值但叶色尚好,饱满直立;

形态指标:主要根据花瓣的颜色与膨胀度评分,评分标准如下:
100 分——花色纯正,无色变,鲜艳,膨胀度好(一般指刚采时的状态);
90 分——有轻微的色变,但鲜艳,花色轻微黯淡但纯正;
80分——有轻微色变且花色略黯淡,膨胀度稍差;

结果分析:
结合上述实施例及对比例的实验数据,可以看出普鲁兰多糖、类黄腐酸、氯化钾、苯甲酸钠全部存在于鲜切花花期延长剂中,且按适当比例配置才能达到最佳的延长花期的效果。缺少普鲁兰多糖、类黄腐酸中的任何一种,延长花期的效果均不理想。虽然普鲁兰多糖和麦芽糖都能同时提供麦芽糖单元,但是普鲁兰多糖被麦芽糖替换,延长花期效果也不理想。在5~25℃范围之间,该鲜切花花期延长剂对其它鲜切花也可达到很好的延长花期效果,显示出适应温度范围广、花卉品种多的特点。
Claims (9)
1.一种含普鲁兰多糖的鲜切花花期延长剂,其特征在于,每升溶剂包含以下成分:
普鲁兰多糖2-8 g,类黄腐酸50-150 mg-C,氯化钾0.1-0.6g,苯甲酸钠10-50 mg;
所述类黄腐酸来源于造纸黑液,其在鲜切花花期延长剂的含量以其中的碳含量为准,即mg-C,所述类黄腐酸的分子量≤10 kDa。
2.如权利要求1所述的鲜切花花期延长剂,其特征在于,所述含普鲁兰多糖的鲜切花花期延长剂的溶剂为水。
3.如权利要求1所述的鲜切花花期延长剂,其特征在于,所述含普鲁兰多糖的鲜切花花期延长剂每升溶剂包含以下成分:
普鲁兰多糖3-7 g,类黄腐酸60-120 mg-C,氯化钾0.1-0.6 g,苯甲酸钠10-50 mg。
4.如权利要求1所述的鲜切花花期延长剂,其特征在于,所述含普鲁兰多糖的鲜切花花期延长剂每升溶剂包含以下成分:
普鲁兰多糖4-6 g,类黄腐酸80-100 mg-C,氯化钾0.2-0.4 g,苯甲酸钠15-35 mg。
5.权利要求1所述的含普鲁兰多糖的鲜切花花期延长剂的制备方法,其特征在于,包括以下步骤:
(1)取苯甲酸钠,加入水中,调节溶液pH至4.0~4.5;
(2)取类黄腐酸和氯化钾,依次加入至步骤(1)所得的溶液中,搅拌均匀,再加入普鲁兰多糖,溶解后搅拌均匀,得到鲜切花花期延长剂。
6.如权利要求5所述的制备方法,其特征在于,步骤(1)中调节溶液pH采用磷酸二氢铵、磷酸二氢钾、磷酸二氢钠、碳酸氢钠、碳酸氢钾、碳酸氢铵、硫酸钙、硫酸铝、硫酸铝铵、柠檬酸及其盐、山梨酸、植酸、十八碳酸或硬脂酸及其盐中的一种或几种。
7.权利要求1所述的含普鲁兰多糖的鲜切花花期延长剂的应用,其特征在于,应用于延长花卉鲜切花花期。
8.如权利要求7所述的应用,其特征在于,所述花卉为玫瑰、百合花。
9.如权利要求7所述的应用,其特征在于,所述延长花卉鲜切花花期的应用温度为5~25℃。
