本发明公开了一种基因修饰多潜能干细胞衍生的增强型NK细胞的方法及其应用,提供了一种经修饰的细胞或其群体,细胞或其群体包含(a)CD16的胞外结构域中的F158V和S197P;(b)源自CD16的全部或部分胞外结构域;(c)天然CD16跨膜结构域;(d)天然刺激性结构域为2B4、DAP10中至少一种的胞内结构域;(e)天然信号传导结构域CD3ζ胞内结构域,表现出成熟的分化表型和更强的免疫杀伤能力,具有更显著的CD16表达。还提供了所述细胞或其群体的制备方法和其用途,通过该方法得到的FR1‑iNK细胞中成熟相关膜蛋白CD94和杀伤功能相关蛋白NKp44/NKG2A的表达量相比CTRL‑iNK和FR2‑iNK有显著上升。

技术领域
本发明涉及免疫细胞产物领域,具体涉及一种基因修饰多潜能干细胞衍生的增强型NK细胞的方法及其应用。
背景技术
现有技术采用在外源表达高亲和力非剪切型的hnCD16或其变体(嵌合抗原受体hnCD16,CAR-hnCD16)的方式来增强免疫细胞抗体依赖细胞介导的毒作用(antibody-dependent cell-mediated cytotoxicity,ADCC)以杀伤肿瘤细胞。目前,在免疫细胞治疗中已有的基于CD16工程改造的iPSC来源的NK细胞治疗产品与技术主要是围绕“高亲和力不可切割的CD16(hnCD16)或其变体”的iPSC工程化改造。
关于这一技术路线,已有技术罗列了多种免疫细胞(T,NK,巨噬细胞等)膜信号蛋白的跨膜域,刺激域和信号传导域作为hnCD16变体CAR-hnCD16的结构组合来源,然而这样随机组合的CAR-hnCD16是否能功能性的发挥ADCC作用现有技术并没有验证。更重要的是针对不同的免疫细胞不同的CAR-hnCD16结构组合产生ADCC作用强弱千差万别,将极大影响到其作为细胞治疗产品对肿瘤杀伤的能力。另外,针对自然杀伤细胞特定的CAR结构,特别是高效的胞内域,信号刺激域,也是影响非野生型CD16蛋白介导ADCC杀伤肿瘤效力高低的关键。如中国专利CN113166232A公开了一种使用增强的iPSC衍生效应细胞的免疫疗法,该专利仅验证了外源表达hnCD16a蛋白(胞外为携带点突变的高亲合和非剪切型CD16a,跨膜区与胞内区为野生型CD16a序列)在iPSC分化的NK细胞中的作用,并没有证实并优选适合NK细胞的CAR-hnCD16结构组合。
发明内容
本发明提供一种基因修饰多潜能干细胞衍生的增强型NK细胞的方法及其应用,目的是解决背景技术中存在的上述问题。
本发明提供的技术解决方案如下:
第一方面,本发明提供一种经修饰的细胞或其群体,其中:
(i)所述细胞包括:(a)诱导性多能细胞(iPSC)、(b)造血干细胞或造血祖细胞;或(b)通过分化(a)的所述细胞获得的衍生细胞;并且
(ii)表达至少一种编码以下的内源基因:
(1)Fc受体FcγRIIIa(CD16a);
(2)CD94、CD56和/或CD45;
(3)NKp46、NKG2D和/或FasL;

在一个实施例中,所述细胞为衍生的NK细胞,所述NK细胞来源于诱导多能干细胞或造血干(祖)细胞。
在一个实施例中,所述细胞与由诱导多能干细胞分化的天然对应细胞相比具有包括以下的以下特性中的至少一个特性:
(i)激活免疫细胞;
(ii)提高肿瘤细胞抗原的识别和杀伤力;
(iii)增强免疫细胞的ADCC作用;
(iv)细胞毒作用增加;
(v)增加杀伤的肿瘤细胞的种类。
在第二方面,本发明提供一种产生上述的经修饰的细胞或其群体的方法,该方法包括:
(i)设计和生产转染iPSC的病毒载体;
(ii)进行慢病毒包装;
(iii)通过慢病毒转染表达hnCD16-FR和GFP蛋白的人诱导多能干细胞hiPSCs生成hnCD16-FR-iPSCs,然后进行单细胞分选和克隆选择,获得hnCD16-FR-iPSCs阳性克隆株。
在一个实施例中,所述诱导多能干细胞生成hnCD16-FR-iPSCs,包括:
(i)将诱导多能干细胞首先分化为造血干(祖)细胞,然后再将所述造血祖细胞分化为NK细胞,当CD34+细胞出现时,将所述NK细胞转移到NK细胞分化培养基中;
(ii)将所述NK细胞在所述NK细胞分化培养基中放置28-35天,每周更换培养基,直到测定其发育为CD45+CD56+细胞。
在一个实施例中,所述培养基含有DMEM/Ham F12、l-谷氨酰胺、青霉素/链霉素、β-硫代乙醇、亚硒酸钠、乙醇胺、抗坏血酸、白细胞介素-3、干细胞因子、白细胞介素-15、fms样酪氨酸激酶3配体和白细胞介素-7。
在一个实施例中,所述进行慢病毒包装,包括:
(i)准备HEK 293T细胞;
(ii)慢病毒包装系统转染;
(iii)慢病毒液收集及浓缩。
在一个实施例中,所述设计和生产转染iPSC的病毒载体,包括:
将包含一个嵌合的5'长末端重复序列(LTR),包装信号,旋转响应元件(RRE),土拨鼠肝炎病毒转录后调控元件(WPRE)和3'自我失活(SIN)的LTR的CAR-hnCD16慢病毒转移质粒置于延伸因子1α(elongation factor 1alpha)启动子(EF1α)的控制下,通过IRES与EGFP共同表达。
在第三方面,本发明提供一种上述的经修饰的细胞或其群体在癌症免疫治疗中的应用。
与现有技术相比,本发明的有益效果是:
1.本发明提供了一种经修饰的细胞或其群体,通过设计不同胞内刺激域结构的CAR-hnCD16,从而优选出在iPSC来源的NK细胞中介导ADCC作用最佳的CAR-hnCD16结构,以提高携带工程化改造CD16的iPSC来源NK细胞治疗产品对肿瘤杀伤的效力;
2.本发明提供的两个工程化表达hnCD16FR的iNK细胞(FR1和FR2)相比CTRL有更显著的CD16表达,特别是FR1-iNK细胞中成熟相关膜蛋白CD94和杀伤功能相关蛋白NKp44/NKG2A的表达量相比CTRL-iNK和FR2-iNK有显著上升;
3.本发明同时提供一种上述NK细胞的制备方法,通过该方法得到的NK细胞成熟相关膜蛋白如CD94以及杀伤功能型蛋白如NKp46、NKG2D、FasL等在iPSC分化制备的终末细胞中都有明显的表达,证明得到具有成熟的分化表型和免疫杀伤能力的NK细胞。

为使本申请实施例的目的、技术方案和优点更加清楚,下面将结合本申请实施例中的附图,对本申请实施例中的技术方案进行清楚、完整地描述。显然,下面所描述的实施例是本申请的一部分实施例,而不是全部的实施例。通常在此处附图中描述和示出的本申请实施例的组件可以以各种不同的配置来布置和设计。
因此,以下结合附图提供的本申请实施例的详细描述旨在仅仅表示本申请的选定实施例,并非限制本申请要求保护的范围。基于本申请中的实施例,本领域普通技术人员在没有作出创造性劳动的前提下所获得的其他所有实施例,都属于本申请保护的范围。
自然杀伤(NK)细胞是先天免疫系统的成员之一,具有裂解靶细胞并为免疫调节提供早期细胞因子的两种关键作用。近年来,研究者们在自然杀伤(NK)细胞对肿瘤和病毒的免疫效力作用机制研究方面取得了长足的进步。作为具有细胞毒性的先天性淋巴细胞,自然杀伤细胞能产生炎性细胞因子和趋化因子,从而通过裂解转化或感染目标细胞来限制肿瘤的生长和病毒的感染。研究表明供体自身NK细胞可以抑制异体基因造血干细胞移植后白血病治疗的复发,更重要的是同种异体NK细胞的输注已被证明能诱导和/或维持急性髓系白血病(AML)的缓解。更重要的是相对由MHC分子识别激活的T细胞,NK细胞表达广泛识别癌细胞表面压力诱导蛋白的受体。因此NK细胞在癌症免疫治疗方面显现出巨大的潜力,通过自身的特异性受体识别多种肿瘤抗原和细胞激活无MHC I类分子的限制。另外与T细胞相反,同种异体NK细胞具有低免疫原性的特点,可在HLA亚型不匹配的异体间使用,也不会发生移植物抗宿主病,这个特点结合NK细胞的广谱抗癌活性,使NK细胞疗法成为一种新兴的免疫细胞肿瘤治疗选择。
在人体免疫细胞中,NK细胞分布于骨髓、外周血、肝、脾、肺和淋巴结,然而含量较低,占循环淋巴细胞的8-20%,表型上表现为CD56阳性和CD3阴性。相对的,T细胞在胸导管淋巴液中可占90%,淋巴节中约占75%,末梢血中可占60%~80%。通过人外周血提取PBMC扩增得到的NK细胞数量有限,且不同供者来源产品批次间稳定性较差。2006年日本科学家山中伸弥在世界著名学术期刊《细胞》上率先发表了关于成功制备诱导性多能干细胞(induced pluripotent stem cells,简称iPS细胞)的技术方法,并在2012年凭借此技术获得了诺贝尔生理医学奖。具体来说,iPSC技术利用特定的转录因子,可以将终末分化的体细胞(如外周血细胞、尿液细胞、皮肤细胞等)逆转为具有亚全能性的干细胞,即诱导性多能干细胞,这一逆转的过程即为细胞的重编程(Cell reprogramming)。由于iPS细胞具有与胚胎干细胞类似的亚全能性,可以分化为人体多种组织和器官细胞,如神经元、胶质细胞、心肌细胞、肝细胞、胰岛细胞、免疫细胞等,实现了对这些难以获取、难以扩增的细胞的大量制备,为相关疾病的机理机制研究提供了全新平台。因此由iPS细胞分化制备成熟的NK细胞为NK细胞治疗临床转化提供了可能的实现途径。
CD16蛋白和抗体IgG的Fc段结合,通过ADCC作用,激活免疫细胞,使其杀伤抗体识别抗原的肿瘤细胞,因此CD16可以搭配不同的抗体杀伤肿瘤细胞。目前,CD16已鉴别为两种异构体:Fc受体FcγRIIIa(CD16a;NM_000569.6)和FcγRIIIb(CD16b;NM_000570.4)。CD16a是由NK细胞表达的跨膜蛋白,为抗体IgG Fc段的受体,结合被抗体识别的靶细胞,激活NK细胞的ADCC作用。CD16b仅由人类嗜中性粒细胞表达,不介导NK细胞对肿瘤的ADCC杀伤作用。在成熟的自然杀伤细胞中CD16的表达水平很低,而且蛋白结合抗体后会被剪切而失效,因此自然条件下野生型CD16的ADCC作用有限。在iPSC分化制备的成熟NK细胞中CD16a的表达很低,一般CD16阳性iNK细胞有10%左右。另外,野生型CD16a对IgG Fc段的亲和力较低,并且胞外结构域的特定区域会被蛋白酶ADAM17剪切而脱落,从而调控NK等细胞上的表面密度。因此野生型CD16在结合靶细胞表面抗体后会被剪切而脱落,降低其介导的肿瘤杀伤作用。
hnCD16是由野生型CD16引入胞外F158V突变使其具有高亲和力,以及胞外S197P突变,不被ADAM17蛋白酶切割而脱落。hnCD16的变体主要是hnCD16的胞外区和非源自CD16并且源自相同或不同多肽的跨膜结构域、信号传导结构域和刺激性结构域的融合蛋白。多项细胞和动物实验已验证hnCD16-iPSC-NK细胞搭配相应抗体使用能大大提升NK细胞对多种肿瘤的杀伤效力。
外源表达不可切割的hnCD16一定程度上增强了ADCC作用以及双特异性、三特异性或多特异性接合体的接合。但是其在NK胞内的传导信号通路仍为野生型CD16的胞内信号域。因此为了进一步增强CD16介导的ADCC杀伤作用,特别是其和多特异性抗体结合针对多种靶点的肿瘤细胞杀伤力的增强,将hnCD16改造为hnCD16-CAR(hnCD16-chimera antigenreceptor),替换原生CD16的跨膜域和/或胞内域,产生嵌合hnCD16包含非野生型跨膜域、非野生型刺激域和/或非野生型信号传导域。
针对不同来源的不同类型免疫细胞,例如PBMC或iPSC来源的T细胞或NK细胞,不同CAR结构的hnCD16变体激活免疫细胞ADCC作用的效果相差甚大,最终将导致CAR-hnCD16-免疫细胞产品对肿瘤细胞的消除治疗效果千差万别。本发明优选适合iPSC来源NK细胞的CAR-hnCD16结构组合,设计了不同来源的免疫细胞膜信号蛋白,组合为胞外、跨膜和胞内刺激域结构的hnCD16变体结构。我们提出并证明了“信号肽-hnCD16胞外区-跨膜结构域-信号传导结构域-刺激性结构域”的CAR-hnCD16结构,介导了更好的肿瘤杀伤效力。因此,本发明相比已有技术,优选出效果更好的在iPSC来源的NK细胞中高效介导ADCC作用杀伤肿瘤细胞的CAR-hnCD16结构和序列,为工程改造的CD16在iPSC来源NK细胞治疗产品的应用提供了重要的支持。
贯穿本说明书提及“一个实施例”、“一实施例”、“特定实施例”、“相关实施例”、“某一实施例”、“另外实施例”或“另一实施例”或其组合意指结合实施例描述的特定特征、结构或特性包含于本发明的至少一个实施例中。因此,贯穿本说明书中的各处出现的前述短语不一定全部指代同一实施例。此外,特定特征、结构或特性可在一个或多个实施例中以任何合适方式组合。
术语“体内”一般是指在生物体内部进行的活动。
如本文所用,术语“分化”是未特化(“未专门化”)或弱特化细胞借以获得特化细胞(例如血细胞或肌肉细胞)的特征的过程。分化细胞或分化诱导的细胞是已处在细胞谱系内的更特化(“专门化”)位置的细胞。术语“专门化”在应用于分化过程时是指一种细胞,所述细胞在分化路径中已经行进到在正常情况下其将继续分化成特定细胞类型或细胞类型的亚群并且在正常情况下其不能分化成不同细胞类型或恢复为更弱分化细胞类型的时刻。如本文所用,术语“多能”是指细胞形成身体或胞体(即,胚胎本身)的所有谱系的能力。例如,胚胎干细胞是一种类型的多能干细胞,其能够从三种胚层:外胚层、中胚层和内胚层中的每一种形成细胞。多能性是一种连续的发育效能,范围为不能产生完整生物体的不完全或部分多能细胞(例如外胚层干细胞或EpiSC)到能够产生完整生物体的更原始、更多能细胞(例如胚胎干细胞)。
如本文所用,术语“诱导性多能干细胞”或iPSC意指由分化的成体、新生儿或胎儿细胞产生的干细胞,所述干细胞已经诱导或改变,即重编程为能够分化成所有三种胚层或真皮层:中胚层、内胚层和外胚层的组织的细胞。所产生的iPSC并非指如其在自然界中被发现的那样的细胞。
如本文所用,术语“多潜能干细胞”是指具有分化成一种或多种胚层(外胚层、中胚层和内胚层)、但非全部三种胚层的细胞的发育潜能的细胞。因此,多潜能细胞也可以称为“部分分化的细胞”。多潜能细胞为所属领域中熟知的,并且多潜能细胞的实例包含成体干细胞,例如造血干细胞和神经干细胞。“多潜能”指示细胞可以形成既定谱系内的许多类型的细胞,而非其它谱系的细胞。例如,多潜能造血细胞可形成许多不同类型的血细胞(红细胞、白细胞、血小板等),但其不能形成神经元。因此,术语“多潜能性”是指一种细胞状态,其发育潜能的程度低于分化全能和多能。
可部分地通过评定细胞的多能性特征确定多能性。多能性特性包含但不限于:(i)多能干细胞形态;(ii)无限自我更新的潜能;(iii)多能干细胞标记物的表达,所述标记物包含但不限于SSEA1(仅小鼠)、SSEA3/4、SSEA5、TRA1-60/81、TRA1-85、TRA2-54、GCTM-2、TG343、TG30、CD9、CD29、CD133/凸素(prominin)、CD140a、CD56、CD73、CD90、CD105、OCT4、NANOG、SOX2、CD30和/或CD50;(iv)分化成所有三种体细胞谱系(外胚层、中胚层和内胚层)的能力;(v)由三种体细胞谱系组成的畸胎瘤形成;以及(vi)由来自三种体细胞谱系的细胞组成的类胚胎体的形成。
术语“培养”或“细胞培养”是指细胞在体外环境中维持、生长和/或分化。“细胞培养基”、“培养基”(在各种情况下为单数“培养基(medium)”)、“补充剂”和“培养基补充剂”是指培养细胞培养物的营养组合物。
术语“造血干细胞和祖细胞”、“造血干细胞”、“造血祖细胞”或“造血前驱细胞”是指定型为造血谱系,但能够进一步造血分化的细胞,且包含多潜能造血干细胞(血胚细胞)、骨髓祖细胞、巨核细胞祖细胞、红细胞祖细胞和淋巴祖细胞。造血干细胞和祖细胞(HSC)是多潜能干细胞,其产生所有血细胞类型,包含骨髓(单核细胞和巨噬细胞、嗜中性粒细胞、嗜碱粒细胞、嗜酸性粒细胞、红细胞、巨核细胞/血小板、树突状细胞)和淋巴谱系(T细胞、B细胞、NK细胞)。如本文所用,术语“永久性造血干细胞”是指能够产生成熟骨髓细胞类型和淋巴细胞类型两者,包含T细胞、NK细胞和B细胞的CD34+造血细胞。造血细胞还包含原始造血细胞的多种亚群,其产生原始红细胞、巨核细胞和巨噬细胞。
如本文所用,术语“外源”旨在意指参考分子或参考活性引入宿主细胞中,或对于宿主细胞是非原生的。可以例如通过将编码核酸引入宿主遗传物质中,例如通过整合到宿主染色体中或作为非染色体遗传物质(例如质粒)来引入所述分子。因此,所述术语在关于编码核酸的表达使用时是指编码核酸以可表达形式引入细胞中。术语“内源”是指存在于宿主细胞中的参考分子或活性。类似地,所述术语在关于编码核酸的表达使用时是指细胞内所含而非外源引入的编码核酸的表达。
如本文所用,术语“多核苷酸”是指具有任何长度的核苷酸的聚合物形式,脱氧核糖核苷酸或核糖核苷酸或其类似物。多核苷酸序列由四个核苷酸碱基构成:腺嘌呤(A);胞嘧啶(C);鸟嘌呤(G);胸腺嘧啶(T);和尿嘧啶(U)(当多核苷酸是RNA时,尿嘧啶代替胸腺嘧啶)。多核苷酸可以包含基因或基因片段(例如,探针、引物、EST或SAGE标签)、外显子、内含子、信使RNA(mRNA)、转移RNA、核糖体RNA、核糖核酸酶、cDNA、重组多核苷酸、分支链多核苷酸、质粒、载体、任何序列的分离DNA、任何序列的分离RNA、核酸探针和引物。多核苷酸还指双链和单链分子。
如本文所用,术语“肽”、“多肽”和“蛋白质”可互换使用,并且是指具有通过肽键共价连接的氨基酸残基的分子。多肽必须含有至少两种氨基酸,且多肽的氨基酸的最大数目不受限制。如本文所用,所述术语是指短链(在所属领域中通常也称为例如肽、寡肽和低聚物)和较长链(在所属领域中通常称为多肽或蛋白质)。“多肽”包含例如生物活性片段、大体上同源的多肽、寡肽、同源二聚体、异二聚体、多肽的变体、经修饰多肽、衍生物、类似物、融合蛋白等。多肽包含天然多肽、重组多肽、合成多肽或其组合。
术语“Fc受体”(缩写为FcR)是基于其识别的抗体类型而分类。例如,结合最常见类别的抗体IgG的受体称为Fc-γ受体(FcγR),结合IgA的受体称为Fc-α受体(FcαR)并且结合IgE的受体称为Fc-ε受体(FcεR)。FcR的类别也通过表达其的细胞(巨噬细胞、粒细胞、自然杀伤细胞、T细胞和B细胞)和每种受体的信号传导特性来区分。Fc-γ受体(FcγR)包含若干成员:FcγRI(CD64)、FcγRIIA(CD32)、FcγRIIB(CD32)、FcγRIIIA(CD16a)、FcγRIIIB(CD16b),它们的抗体亲和力因其分子结构不同而不同。
术语“长末端重复序列(long terminal repeated,LTR)”是反转录病毒的基因组的两端各有一个长末端重复序列(5'—LTR和3'—LTR)。不编码蛋白质,但含有启动子,增强子等调控元件,病毒基因组内的LTR可转移到细胞原癌基因邻近处,使这些原癌基因在LTR强启动子和增强子的作用下被激活,将正常细胞转化为癌细胞。
术语“慢病毒”是目前细胞与基因疗法研究,尤其是CAR-T细胞治疗,所使用的主要基因导入平台。慢病毒是单链RNA病毒,一般以人类免疫缺陷病毒1型(HIV-1)为基础开发的居多,可用于绝大多数哺乳动物细胞基因导入,包括使用逆转录病毒难以实现的造血干细胞及神经细胞等在内的非分裂细胞。使用重组慢病毒导入的目的基因也可以整合到宿主细胞的基因组中,可实现长期稳定的基因表达。慢病毒载体(Lentiviral vectors,LVs)是在人免疫缺陷病毒(HIV-1病毒)基础上改造而成的病毒载体系统,它能高效的将目的基因(或RNAi)导入动物和人的原代细胞或细胞系,对分裂细胞和非分裂细胞均具有感染能力。慢病毒载体基因组是单股正链RNA,其基因组进入细胞后,在细胞浆中被其自身携带的反转录酶反转为DNA,形成DNA整合前复合体,进入细胞核后,DNA整合到细胞基因组中。整合后的DNA转录成mRNA,回到细胞浆中,表达目的基因或片段。慢病毒载体介导的基因表达或RNAi干扰作用持续且稳定,其目的基因整合到宿主细胞基因组中,并随细胞基因组的分裂而分裂。另外,慢病毒载体能有效感染并整合到非分裂细胞中。以上特性使慢病毒载体与其它病毒载体相比(如不整合的腺病毒载体、整合率低的腺相关病毒载体、只整合分裂细胞的传统逆转录病毒载体),有鲜明的特色。大量文献研究表明,慢病毒载体介导的目的基因长期表达的组织或细胞包括脑、肝脏、肌肉、视网膜、造血干细胞、骨髓间充质干细胞、巨噬细胞等。
慢病毒表达载体删除了HIV病毒绝大多数基因,仅保留了HIV病毒的LTR序列,包装信号,Rev应答元件等。因此免疫原性低,在注射部位无细胞免疫反应,体液免疫反应也较低,不影响病毒载体的第2次注射。
最早使用的外源表达载体含有慢病毒载体所有的顺式作用元件(包装信号、反转录和整合元件),外源插入片段以及相关调控元件(启动子和插入片段)。具体来说,载体N端含有复制引发结合位点,剪切供体位点,包装信号,RRE序列和剪切受体位点;异源启动子和插入片段在中间;C端含有PolyA加尾信号和3’端LTR。为了增加安全性,降低重组产生RCR的可能性以及增加表达效率和丰富调控表达方式,研究人员对外源表达载体进行了一系列改造:
1),构建SIN载体:由于逆转录病毒的3’端LTR的U3区在反转录和病毒DNA复制时时会被当作模板最终生成5’端LTR所对应的U3区,因此如果要去除病毒自身的转录起始元件使得外源片段发生不依赖于Tat蛋白的转录,必须将5’端LTR的U3区替换成异源启动子,同时去除3’端LTR的U3区的133-400bp含TATA盒子和Sp1,NF-kB等转录结合位点以及其它增强子的对应序列。改造后的不含这段序列的U3区会被复制转移形成转录产物中的5’端LTR的新U3区,所以转录产物中不再含有病毒自身转录起始元件,因此也就不再依赖于Tat蛋白因子转录,使得可以在其它质粒载体中去除Tat编码基因。同时,在整合入宿主细胞基因组后,还可以减少对整合位点附近基因的转录干扰,比如避免激活下游基因(在早期逆转录病毒载体中常见,并且是运用逆转录病毒载体进行基因治疗时诱发肿瘤发生的主要因素)。
2),土拨鼠肝炎病毒转录后调控元件(WPRE,woodchuck hepatitis virusposttranscriptional regulatory element)和cPPT(central polypurine tract)序列可以极大增加外源片段表达效率。
包装因子表达载体表达除包膜蛋白外的所有包装和侵染必须的反式作用因子。为避免将此类序列包装入假病毒颗粒(慢病毒载体),因此包装因子表达载体上缺乏顺式包装信号和LTR序列,然而仍然保留旋转响应元件(RRE,Rev response element)序列和剪切供体位点。使用其它启动子,比如CMV或者RSV启动子,以及胰岛素基因的polyA加尾信号序列替代LTR的对应序列,可以使得转录产物更加稳定,表达效率更高。
术语“启动子”是RNA聚合酶识别、结合和开始转录的一段DNA序列,它含有RNA聚合酶特异性结合和转录起始所需的保守序列,多数位于结构基因转录起始点的上游,启动子本身不被转录。但有一些启动子(如tRNA启动子)位于转录起始点的下游,这些DNA序列可以被转录。启动子的特性最初是通过能增加或降低基因转录速率的突变而鉴定的。启动子一般位于转录起始位点的上游。
术语“启动子(EF1a)或EF1α启动子”是一种来源于延伸因子1α(elongationfactor 1alpha,EF1A)基因的强哺乳动物表达启动子,可在多种细胞中稳定驱动其下游基因的组成型表达,也可以用于干细胞、原代细胞、造血细胞等。
术语“内部核糖体进入位点序列(Internal ribosome entry site,IRES)”为一段非翻译RNA,能招募核糖体对mRNA进行翻译。将IRES与外源cDNA融合,发现IRES能独立地起始翻译。一般真核mRNA的翻译都需要5‘帽子来介导核糖体结合,但真核生物和病毒中还存在一些例外情况,例如一些基因5'端具有一段较短的RNA序列(约150-250BP),这类RNA序列能折叠成类似于起始tRNA的结构,从而介导核糖体与RNA结合,起始蛋白质翻译。
术语“增强绿色荧光蛋白(Enhanced Green Fluorescent Protein)”,EGFP是GFP突变系。应用较多的是GFP的突变体:增强型绿色荧光蛋白(EGFP),发射出的荧光强度比GFP大6倍以上,因此,比GFP更适合作为一种报告基因来研究基因表达、调控、细胞分化及蛋白质在生物体内定位和转运等。
术语“HEK293T细胞”,简称293T细胞是由293细胞通过基因技术派生出的细胞系,是经过腺病毒E1A基因的转染,能表达SV40大T抗原,含有SV40复制起始点与启动子区。许多真核表达载体如pcDNA3.1中含有SV40病毒的复制起始位点,可以在表达SV40病毒T抗原的细胞系中复制,从而提高外源基因的表达水平。因此293T细胞广泛应用于病毒包装。用磷酸钙转染效率可高达50%。蛋白表达水平高,转染后2-3天用碱性磷酸酶分析可较容易地检测到表达的蛋白。瞬时转染293T细胞是过表达蛋白并获得细胞内及细胞外(分泌的或膜)蛋白的便捷方式。293细胞是人肾上皮细胞系,有多种衍生株,比如HEK293,293T/17等,来源都是人胚胎肾细胞,其极少表达细胞外配体所需的内生受体,且比较容易转染,是一个很常用的表达研究外源基因的细胞株。本发明采用的293T细胞编号为“ATCC,CRL-3216”的细胞株。
术语“ddH2O”是经过两次蒸馏过的超纯水。
术语“Opti-MEM培养基”是一种可用于各种悬浮和贴壁的哺乳动物细胞,包括Sp2、AE-1、CHO、BHK-21、HEK和原代成纤维细胞的培养基。
术语“ExFectTM Transfection Reagent”是一种基于生物可降解的高分子树枝状聚合物的转染试剂,适用于多种细胞的DNA转染。由于所使用的活化树枝状聚合物具有均一的大小和确定的形状,因此可实现高重复性的转染结果。ExFectTM Transfection Reagent携带DNA进入细胞后,可迅速释放DNA,并被快速降解,极大的降低了对细胞的毒性。转染试剂-DNA复合物可以直接加入完全培养基中,血清和抗生素不影响其转染效果,转染前后都不需要更换培养基,因此操作十分简便。
术语“Package Plasmid Mix”是人类免疫缺陷病毒衍生的慢病毒包装质粒psPAX2和含VSV-G的pMD2.G质粒的混合物。这些质粒提供所有必需的,包装假型慢病毒表达所必要的结构蛋白,监管蛋白和复制蛋白。于慢病毒表达构建体(例如Cellecta PRSI shRNA表达构建体)共转染293T产生细胞系,(如ATCC,目录号CRL-11268),生产假型包装慢病毒。用VSV-G包膜蛋白假型化的慢病毒颗粒能够感染多种哺乳动物或非哺乳动物,分裂的细胞或不分裂的细胞。
术语“诱导多能干细胞(induced pluripotent stem cells,iPSCs)”是通过人工诱导非多能性细胞表达某种特定基因得到的。iPSCs和天然多功能干细胞在很多方面具有相似性,比如某种干细胞基因和蛋白的表达,染色质甲基化模式,倍增之间,胚体形成,畸胎瘤形成,不同嵌合体的形成,以及分化潜能。
术语“人诱导多能干细胞(human induced pluripotent stem cells,hiPSCs)”是由人体细胞通过非整合的方法诱导获得,适合临床级的干细胞研究和应用,具有高度近似人ES细胞的克隆形态和基因表达,长期维持正常核型,并在体内外具有三胚层分化潜能。
根据上文详细说明,可以对实施例作出这些和其他改变。总体上,在以下权利要求书中,所使用的术语不应解读为将权利要求书限制为说明书和权利要求书中披露的具体实施例,而应解读为包括所有可能的实施例连同这些权利要求所享有的等效权利的全部范围。因此,权利要求书不受本披露的限制。
实施例1
慢病毒转染构建CAR-hnCD16a-iPSC的实验
1)转染iPSC的病毒载体设计和生产:CAR-hnCD16慢病毒转移质粒包含一个嵌合的5'长末端重复序列(LTR),包装信号,旋转响应元件(RRE),土拨鼠肝炎病毒转录后调控元件(WPRE)和3'自我失活(SIN)的LTR。将CAR-hnCD16慢病毒转移质粒置于EF1α启动子的控制下,并通过IRES与EGFP共同表达。该病毒通过共转染转移质粒、包装质粒和包膜质粒在293T细胞(ATCC,CRL-3216)中产生。
图1为本发明实施例中CAR-hnCD16慢病毒转移质粒结构示意图,本实施例采用的CAR-hnCD16慢病毒转移质粒整体大小为10536bp。
2)慢病毒包装流程:
A.HEK 293T细胞准备:将3~5×106个HEK 293T细胞传代接种至100mm细胞培养皿中,置于37℃,5% CO2的培养箱中,培养16h~24h。传代过程中需要将细胞充分消化为单细胞悬液,以获得较好的包装效果。
B.慢病毒包装系统转染:将慢病毒包装试剂盒中的包装质粒混合物(PackagePlasmid Mix)及慢病毒表达质粒使用ddH2O稀释为终浓度1.0μg/μL的质粒溶液;取1支1.5mL离心管(标记为A管),分别加入300μL Opti-MEM培养基及40μL ExFectTMTransfection Reagent,混匀后室温静置5min;取1支1.5mL离心管(标记为B管),加入2.5μL终浓度为1.0μg/μL的慢病毒表达质粒质粒溶液及7.5μL Package Plasmid Mix,充分混匀;将A管中溶液加入B管中,充分混匀,室温静置15~30min;将B管中的混合溶液逐滴均匀加入接种HEK 293T细胞的培养皿中,轻微水平震荡培养皿以混匀,将培养皿置于37℃,5% CO2培养6h,将培养基更换为37℃水浴预热的新鲜完全培养基。
C.病毒收集及浓缩:转染48h后,收集含有慢病毒的上清液,向培养皿中补充10~15mL新鲜的完全培养基;继续培养24h,进行第二次病毒上清液收集;将两次收集的病毒上清液混合,0.45μm滤器过滤,过滤后的病毒液可以进行浓缩或直接感染目的细胞;将病毒浓缩液分装,保存于-80℃,即取即用。
3)构建多种hnCD16-CAR-iPSC的方法:
通过转导带有表达hnCD16-FR和GFP的慢病毒粒子的hiPSCs生成hnCD16-FR-iPSCs,然后进行单细胞分选和克隆选择,获得hnCD16-FR-iPSCs阳性克隆株。
参阅图2,hnCD16-FR-iPSCs包括CAR-hnCD16-FR1和CAR-hnCD16-FR2。两个CAR-hnCD16核苷酸序列参见序列表。
参阅图3,通过流式实验检测建立的iPSC阳性克隆细胞表达外源导入基因的情况,Unstained为未加入抗体的iPSC对照组;CTRL为病毒转染GFP的iPSC对照组;FR1和FR2为转染CAR-hnCD16-FR1和CAR-hnCD16-FR2的阳性克隆细胞实验组。第一排为GFP抗体流式检测结果,第二排为CD16抗体流式检测实验结果。通过图3说明FR1和FR2两个克隆株都正常表达膜定位的CD16a蛋白。
构建均为一型跨膜蛋白的组合体(FR1)和采用二型跨膜蛋白NKG2D的跨膜区组合体(FR3)结构的CAR(嵌合抗原受体),通过慢病毒感染永生化细胞系YT-NK细胞,从而比较他们的细胞定位情况。这两种结构的胞外区均为高亲合非切割的CD16(hnCD16),跨膜区分别用CD16的跨膜区序列(FR1),或者NKG2D的跨膜区序列(FR3),并通过IRES(internalribosome entry site,胞内核糖体插入位点)与绿色荧光蛋白(GFP)共同表达,用含有FR1和FR3结构的慢病毒分别感染HEK293细胞,然后用CD16的抗体进行免疫荧光染色,并用激光共聚焦显微镜观察。从免疫荧光的结果可见,均为一型跨膜蛋白的组合体FR1,CD16的荧光信号明显定位于细胞膜,细胞内也有部分信号;而二型跨膜蛋白NKG2D的跨膜区和一型跨膜蛋白组合体FR3,CD16没有形成细胞膜特有的膜结构信号,主要定位于细胞内部(细胞质)。
FR1和FR3结构请参阅图4。
参阅图5,CD16和NKG2D跨膜区结构的细胞定位分析,HEK293细胞分别感染FR1和FR3结构的慢病毒,图中兰色为细胞核DNA,绿色为GFP,红色为CD16,标尺为10μm。
同时,用NKG2D的跨膜区hnCD16-CAR(FR3)结构FR3的慢病毒感染YT-NK细胞,进行流式细胞分析,以不加入CD16抗体为阴性对照,结果见图6。如果不用0.05%曲拉通打破细胞膜,加入CD16抗体,21.8%的细胞阳性,扣除对照,只有约16%的CD16阳性细胞;如果用曲拉通打破细胞膜,则有66.9%的细胞CD16阳性,两者的差值为45.1%。说明较多细胞的细胞质内存在翻译的CD16,但不能递呈或定位于细胞膜上。
编码蛋白定位于细胞膜是淋巴细胞的CAR识别和杀伤肿瘤细胞的基础,如果不能形成稳定地在细胞膜表达的结构,根据细胞生物学和CAR作用原理,则无法行使肿瘤细胞特异性识别和杀伤的功能。因此选择2B4蛋白的一型跨膜区和其他一型跨膜蛋白结构域组合CAR-hnCD16a,可以有效避免细胞内外信号混乱的情况,从而高效行驶其功能,提高淋巴细胞(如NK细胞)杀灭肿瘤细胞的能力。
实施例2
hnCD16-CAR-iPSC分化为iNK的实验方法
第一步将hiPSCs首先分化为造血祖细胞,然后分化为NK细胞,当CD34+细胞出现时,细胞群被转移到NK细胞分化培养基中,将造血祖细胞转移到NK细胞分化培养基中,该培养基含有DMEM/Ham F12(Thermo Fisher Scientific,Waltham,MA,11965092,11765054)、2mM l-谷氨酰胺(Thermo Fisher Scientific,Waltham,MA,25030081)、1%青霉素/链霉素(Thermo Fisher Scientific,Waltham,MA,15140122)、25μMβ-硫代乙醇(Thermo FisherScientific,Waltham,MA,21985023),5ng/mL亚硒酸钠(Merck Millipore,Burlington,MA,S5261),50μM乙醇胺(MP Biomedicals,ICN19384590),20mg/mL抗坏血酸(MerckMillipore,Burlington,MA,A4544),白细胞介素-3;干细胞因子(SCF),白细胞介素-15,fms样酪氨酸激酶3配体(FLT3L)和白细胞介素-7。细胞在这种条件下放置28-35天,每周更换培养基,直到流式细胞术测定其发育为CD45+CD56+细胞。如图7所示分化终点的细胞明显表达NK细胞成熟markerCD56及造血细胞marker CD45(蓝色峰)(所有抗体均来源于biolegend)。
参阅图8,通过分析一系列NK细胞相关Marker表达情况,证明1.NK细胞成熟相关膜蛋白如CD94以及杀伤功能型蛋白如NKp46、NKG2D、FasL等在iPSC分化制备的终末细胞中都有明显的表达,说明通过我们的分化方法得到具有成熟的分化表型和免疫杀伤能力的NK细胞。2.两个工程化表达hnCD16FR的iNK细胞(FR1和FR2)相比CTRL有更显著的CD16表达,特别是FR1-iNK细胞中成熟相关膜蛋白CD94和杀伤功能相关蛋白NKp44/NKG2A的表达量相比CTRL-iNK和FR2-iNK有显著上升。
实施例3
1.hnCD16-CAR-iNK细胞杀伤实验:
1)实验方法与流程:将分化得到的iNK效应细胞CTRL、FR1、FR2,搭配癌细胞对应高表达靶点的抗体,与表达luciferase的靶细胞Raji或A549以不同效靶比置于37℃,5% CO2共同孵育培养24h后,加入生物发光底物并通过酶标仪上机检测,使用以荧光素酶为基础的标准生物发光法评价对靶细胞的杀伤毒性。杀伤肿瘤细胞实验结果如下:
1)搭配anti-CD20 mab(Obinutuzumab)杀伤Raji细胞,结果见图9。
2)搭配anti-EGFR mab(Nimotuzumab)杀伤A549细胞,结果见图10。
通过图9、图10表明工程化表达FR结构的NK细胞通过联合治疗型单克隆抗体可以显著的增强对多种来源肿瘤细胞的杀伤效果,不表达CD16的CTRL细胞对肿瘤细胞的杀伤功能在加入抗体后没有显示出较大的差异,提示其ADCC作用并没有被明显激活,FR1对比FR2可以发现即使在较低的效靶比时也能造成更显著的杀伤增强效果,体现了CAR-hnCD16a结构的高效肿瘤杀伤效率。
通过上述实施例,可以得到经基因修饰的NK细胞或其群体,其中,NK细胞或其群体来源于诱导多能干细胞或造血祖细胞:
(i)所述NK细胞包括:(a)诱导性多能细胞(iPSC)、(b)造血干细胞或造血祖细胞;或(b)通过分化(a)的所述细胞获得的衍生细胞;并且
(ii)表达至少一种编码以下的内源基因:
(1)Fc受体FcγRIIIa(CD16a);
(2)CD94、CD56和/或CD45;
(3)NKp46、NKG2D和/或FasL;
(iii)包含至少一种或多种外源核酸表达构建体,其包含编码以下的核酸序列:
(1)嵌合抗原受体CAR;
(2)CD16的胞外结构域中的F158V和S197P;
(3)源自CD16的全部或部分胞外结构域;
(4)天然CD16跨膜结构域;
(5)天然刺激性结构域为2B4、DAP10中至少一种的胞内结构域;
(6)天然信号传导结构域CD3ζ胞内结构域。
对于作为免疫治疗剂的临床用途,例如,在免疫肿瘤学应用的背景下,产生经基因修饰的增强型NK细胞,通过联合治疗型单克隆抗体可以显著的增强对多种来源肿瘤细胞的杀伤效果,不表达CD16的CTRL细胞对肿瘤细胞的杀伤功能在加入抗体后没有显示出较大的差异,提示其ADCC作用并没有被明显激活,FR1对比FR2可以发现即使在较低的效靶比时也能造成更显著的杀伤增强效果,体现了CAR-hnCD16a结构的高效肿瘤杀伤效率。
本发明设计了嵌合抗原受体hnCD16(CAR-hnCD16)的不同结构组合,以胞外区高亲和非剪切型hnCD16与不同跨膜区域(2B4和DAP10)和胞内共刺激区域(CD3ζ)设计了两种结构的CAR-hnCD16。通过对比CN 113166232 A专利提出的CD16变型体中关键以NKG2D跨膜域的CAR-CD16组合,发现其表达的CAR蛋白在膜定位能力上显著低于本发明提供的CAR-hnCD16组合。其原因可能是CD16作为一型跨膜蛋白,和其信号肽(hnCD16胞外区)组合的跨膜结构域/信号传导结构域/刺激性结构域需要来同样来源于一型跨膜蛋白,才能够使得变体CAR-hnCD16结构传导信号不发生混乱,介导更好的肿瘤杀伤效力。将一型跨膜蛋白胞外信号域(CD16胞外域)和二型跨膜蛋白跨膜域(NKG2D跨膜域)组合在一起,产生的嵌合抗原受体组合蛋白质可能会发生信号的混乱。设计的两个CAR-hnCD16结构和序列(CAR-hnCD16-FR1和CAR-hnCD16-FR2)证明其中两种(FR1,FR2)在iPSC来源的NK细胞搭配人源抗体使用有明显增强杀伤肿瘤细胞的作用(显著高于野生型hnCD16的作用),其中FR1的杀伤效果更优。
以上所述,仅为本申请的最优具体实施方式,但本申请的保护范围并不局限于此,任何在本申请揭露的技术范围内的变化或替换,都应涵盖在本申请的保护范围之内。因此,本申请的保护范围应以所述权利要求的保护范围为准。
Claims (10)
1.一种经修饰的细胞或其群体,其特征在于,其中:
(i)所述细胞包括:(a)诱导性多能细胞(iPSC)、(b)造血干细胞或造血祖细胞;或(b)通过分化(a)的所述细胞获得的衍生细胞;并且
(ii)表达至少一种编码以下的内源基因:
(1)Fc受体FcγRIIIa(CD16a);
(2)CD94、CD56和/或CD45;
(3)NKp46、NKG2D和/或FasL;
(iii)包含至少一种或多种外源核酸表达构建体,其包含编码以下的核酸序列:
(1)嵌合抗原受体CAR;
(2)非剪切型的hnCD16或其工程化改造变体CAR-hnCD16。
2.根据权利要求1所述的细胞或其群体,其特征在于,所述hnCD16或其工程化改造变体CAR-hnCD16包括以下中的至少一种:
(a)CD16的胞外结构域中的F158V和S197P;
(b)源自CD16的全部或部分胞外结构域;
(c)天然CD16跨膜结构域;
(d)天然刺激性结构域为2B4、DAP10中至少一种的胞内结构域;
(e)天然信号传导结构域CD3ζ胞内结构域。
3.根据权利要求1所述的细胞或其群体,其特征在于:
所述细胞为衍生的NK细胞,所述NK细胞来源于诱导多能干细胞、造血干细胞或造血祖细胞。
4.根据权利要求3所述的细胞或其群体,其特征在于,所述细胞与由诱导多能干细胞分化的天然对应细胞相比具有包括以下的以下特性中的至少一个特性:
(i)激活免疫细胞;
(ii)提高肿瘤细胞抗原的识别和杀伤力;
(iii)增强免疫细胞的ADCC作用;
(iv)细胞毒作用增加;
(v)增加杀伤的肿瘤细胞的种类。
5.一种产生如权利要求1-4任一所述的细胞或其群体的方法,其特征在于,该方法包括:
(i)设计和生产转染iPSC的病毒载体;
(ii)进行慢病毒包装;
(iii)通过慢病毒转染表达hnCD16-FR和GFP蛋白的人诱导多能干细胞hiPSCs生成hnCD16-FR-iPSCs,然后进行单细胞分选和克隆选择,获得hnCD16-FR-iPSCs阳性克隆株。
6.根据权利要求5所述的细胞或其群体的方法,所述诱导多能干细胞生成hnCD16-FR-iPSCs,其特征在于,包括:
(i)将诱导多能干细胞首先分化为造血干细胞或造血祖细胞,然后再将所述造血祖细胞分化为NK细胞,当CD34+细胞出现时,将所述NK细胞转移到NK细胞分化培养基中;
(ii)将所述NK细胞在所述NK细胞分化培养基中放置28-35天,每周更换培养基,直到测定其发育为CD45+CD56+细胞。
7.根据权利要求6所述的细胞或其群体的方法,其特征在于:
所述培养基含有DMEM/Ham F12、l-谷氨酰胺、青霉素/链霉素、β-硫代乙醇、亚硒酸钠、乙醇胺、抗坏血酸、白细胞介素-3、干细胞因子、白细胞介素-15、fms样酪氨酸激酶3配体和白细胞介素-7。
8.根据权利要求5所述的细胞或其群体的方法,所述进行慢病毒包装,其特征在于,包括:
(i)准备HEK 293T细胞;
(ii)慢病毒包装系统转染;
(iii)慢病毒液收集及浓缩。
9.根据权利要求5所述的细胞或其群体的方法,所述设计和生产转染iPSC的病毒载体,其特征在于,包括:
将包含一个嵌合的5'长末端重复序列(LTR),包装信号,旋转响应元件(RRE),土拨鼠肝炎病毒转录后调控元件(WPRE)和3'自我失活(SIN)的LTR的CAR-hnCD16慢病毒转移质粒置于构成启动子(ef1a)的控制下,通过IRES与EGFP共同表达。
10.根据权利要求1-4任一所述的经修饰的细胞或其群体在肿瘤免疫治疗中的应用。
