Description

Traditional Chinese medicine composition for treating hyperlipidemia and preparation method thereof
Technical Field
The invention relates to the field of traditional Chinese medicines, in particular to a traditional Chinese medicine composition for treating hyperlipidemia and a preparation method thereof.
Background
Hyperlipidemia, including hypercholesterolemia, hypertriglyceridemia and mixed hyperlipidemia, is an important risk factor for cerebral apoplexy, coronary heart disease, myocardial infarction and sudden cardiac death, and is mainly characterized by dizziness, headache, chest distress, chest pain, abdominal distention, obesity and the like. The occurrence and development of hyperlipidemia are related to various factors such as genetic inheritance, environmental factors (diet, nutrition, medicines and the like) and other factors (such as age, sex, season, drinking, smoking, mental stress, mood instability and the like), and research shows that the occurrence of hyperlipidemia is mostly caused by the comprehensive effect of various factors, while hyperlipidemia caused by a single factor is less, which also brings a certain difficulty to clinical treatment of hyperlipidemia.
At present, lipid-lowering western medicines are mainly used for preventing and treating clinically, but most of western medicines have great side effects and damage liver and kidney functions, such as simvastatin. The traditional Chinese patent medicines for reducing blood fat are also of various types and are full of eyes and various, but on one hand, the traditional Chinese medicine has slow blood fat reducing effect, and on the other hand, the traditional Chinese patent medicines for reducing blood fat have uncertain curative effect, so that people have worry about treating hyperlipidemia by using the traditional Chinese medicine, and meanwhile, the traditional Chinese medicine is limited in clinical use. In addition, the traditional Chinese medicine raw materials are directly used for decoction and administration, but the method has the problems of inconvenient use, difficult preservation and the like. Therefore, the novel traditional Chinese medicine for treating the hyperlipidemia, which has definite curative effect and convenient use, is very important for solving the existing technical problems.
Disclosure of Invention
The invention aims to provide a traditional Chinese medicine composition for treating hyperlipidemia and a preparation method thereof, which are used for solving the problems existing in the prior art, realizing the purpose of reducing triglyceride, total cholesterol, high-density lipoprotein cholesterol and low-density lipoprotein cholesterol in hyperlipidemia serum, and can be used for reducing hyperlipidemia.

Animal experiments were performed on the Chinese medicinal composition prepared in the above examples to verify the therapeutic effects thereof.
The formula of the high-fat feed comprises the following components: 90.1% of basic feed, 1.5% of cholesterol, 8.2% of lard and 0.2% of pig bile salt. Basal feed: 73.5% of corn, 20% of wheat bran, 5% of fish meal, 1% of cereal flour and 0.5% of salt.
120 clean-class C57 mice were selected, the quality of the mice was 15-25g, and after 1 week of the adaptive feeding of 120 mice, the mice were randomly divided into 12 groups of 10 animals each, and the model group, the positive group, the example 1 drug group-example 4 drug group, the example 5 low dose group, the example 5 medium dose group, the example 5 high dose group, the example 6 drug group, and the example 7 drug group were given high fat feeds in addition to the normal feed. The feed is fed for 14 days continuously for molding, and the feed is fed for 3 times per day, 2g each time, and normally drinks water.
After successful modeling of the hyperlipidemia of the mice, the gastric lavage is carried out once every morning and evening for 4 weeks continuously, and the dosage is given: simvastatin positive control was administered in 5mg/kg in the positive group, 1.5g/kg in the example 1 drug group-4 drug group, 5.3.0 g/kg in the example 7 drug group, and 6.0g/kg in the example 5 high dose group, 6.0g/kg in the example 5 drug group, and the normal group and the model group were administered with the same volume of physiological saline per mouse.
Mice were fasted overnight after 6 weeks, blood was collected from the orbital veins, and serum was centrifuged to determine Triglyceride (TG), total Cholesterol (TC), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C). Statistical data with SPSS software, experimental resultsThe comparison between groups is shown by t-test. The results are shown in Table 1.
TABLE 1
As can be seen from table 1 above, compared with the normal group, the traditional Chinese medicine composition prepared in examples 1-7 of the present application can reduce the content of TG, TC, HDL-C and LDL-C in serum, and especially the traditional Chinese medicine composition prepared in example 5 can significantly reduce the content of TG, TC, HDL-C and LDL-C, and the reduction effect is more remarkable with increasing dosage.
In order to further verify the therapeutic effect of the Chinese medicinal composition of the present invention, the present invention adopts the low dose group of example 5, the medium dose group of example 5, the high dose group of example 5, and the positive control group (simvastatin) for clinical experiments, and the experimental method is as follows:
1. case selection
80 patients meeting the hyperlipidemia criteria were randomly divided into 4 groups, of which 43 men, 37 women, the largest of the ages of 78 years, the smallest of 30 years, and the largest of 35-45 years. Occupation: the maximum number of cases is 15 years, the minimum number is half a year, and the average number is 6 years. Wherein, the degree of the disease among the groups is not significantly different in 25 cases of hypercholesterolemia, 35 cases of hypertriglyceridemia and 20 cases of mixed hyperlipidemia. All the patients are from the traditional Chinese medicine hospitals in Yunnan province, the ethical society of the traditional Chinese medicine hospitals in Yunnan province are informed of the consent of all the patients, and the consent of the patients is obtained.
2. Diagnostic criteria
Cholesterol (CT) >5.5mmol/L, triglyceride (TC) >1.69mmol/L, high Density Lipoprotein (HDL) < 1.03mmol/L, and low density lipoprotein (DC) >3.1mmol/L, which meet the diagnostic standards of hyperlipidemia of the teaching materials (fifth edition) of national higher medical institutions.
3. Therapeutic method
Example 5 low dose group: the traditional Chinese medicine composition prepared in the embodiment 5 of the invention is prepared into 50g of powder per bag, and 200mL of liquid is taken orally in the morning and evening each day.
Dose group in example 5: the traditional Chinese medicine composition prepared in the embodiment 5 of the invention is prepared into powder with 100g per bag, and 200mL of liquid is taken orally in the morning and evening each day.
Example 5 high dose group: the traditional Chinese medicine composition prepared in the embodiment 5 of the invention is prepared into 200g of powder per bag, and 200mL of liquid is taken orally in the morning and evening each day.
The positive control group was orally administered simvastatin once a day at night, 30mg each time.
All the patients in the above groups take the medicine for 1 month, and take the medicine for two courses of treatment, and blood lipid is checked for 1 time per month, and the statistical result is obtained after 2 months. During the treatment period, patients are instructed to take a reasonable diet, exercise properly, quit smoking and abstinence from alcohol.
