Abstract
本发明公开了一种具有降尿酸功能的固体饮料,茶叶固体饮料按质量百分比包括以下原料:茶叶提取物粉末10%‑15%、米草提取物粉末5%‑10%、天然草药提取物粉末5%‑10%、柠檬酸15%‑25%、碳酸氢钠15%‑25%、羟丙基淀粉15%‑30%、乳糖3%‑5%、聚乙二醇3%‑5%和微晶纤维素5%‑15%。本发明将米草和天然草药提取物以及茶叶的提取物制作成泡腾片,既补充了茶汤普遍缺少的营养成分,又极大地提高了米草生物利用度,同时泡腾片的形式有利于冷水或温水冲泡,不局限于滚烫的热水才能冲泡出馥郁茶香的饮品,刷新了冲泡流程所需条件。本发明在保有茶叶饮品馨香味道的基础上,增添了具有降尿酸功能的米草提取物,特别适用于痛风和高尿酸血症患者饮用,天然草药的添加有益于提高免疫力、抗炎抗氧化。

技术领域
本发明涉及一种固体饮料,特别涉及一种具有降尿酸功能的固体饮料及其制备方法,属于固体饮料技术领域。
背景技术
茶叶是我国传统的营养健康食物资源,纯茶按照发酵程度的不同分为六类。根据多酚氧化酶的作用导致发酵程度自浅而深,可以将茶叶分为:绿茶、白茶、黄茶、乌龙茶、红茶、黑茶。我国是世界上最早种植茶树和饮茶的国家,茶可以滋润爽口,解酒消食,除烦去腻,还能使人心境平和,提神益思。但纯茶饮品主要是因其特殊的茶香而深受人们的喜爱,功能单一,营养价值不高,因此丰富茶叶饮品的营养价值对茶饮市场具有重要意义。
同时随着人类饮食结构的变化,高尿酸血症和痛风患病人群呈现逐年递增的趋势。在高尿酸血症及痛风的众多诱因中,遗传因素、年龄构成、性别差异等客观因素对痛风均有不同程度的影响。从生活饮食习惯来说,喜欢海鲜和肉类等高嘌呤荤食、饮酒、吸烟、肥胖和作息不规律容易提升高尿酸血症及痛风的患病风险。高尿酸血症及痛风与高血压、动脉粥样硬化、慢性肾脏疾病以及其它代谢紊乱症的发生具有内在联系。痛风发作时以反复出现小关节炎为特点,后期常累及大关节,形成痛风石导致关节畸形,其症状为多为关节的红、肿、热、痛,病情反复,迁延难愈,目前常用高尿酸血症临床治疗药物(如别嘌呤醇)均存在较大毒副作用,对胃肠道、肝脏、肾脏等存在一定程度损伤,引起人们对传统类高尿酸血症治疗药物安全性的关注。
发明内容
本发明的目的在于提供一种具有降尿酸功能的固体饮料及其制备方法,以解决上述背景技术中提出的问题。
为实现上述目的,本发明提供如下技术方案:一种具有降尿酸功能的固体饮料,所述茶叶固体饮料按质量百分比包括以下原料:茶叶提取物粉末 10%-15%、米草提取物粉末5%-10%、天然草药提取物粉末5%-10%、柠檬酸 15%-25%、碳酸氢钠15%-25%、羟丙基淀粉15%-30%、乳糖3%-5%、聚乙二醇 3%-5%和微晶纤维素5%-15%。
作为本发明的一种优选技术方案,一种具有降尿酸功能的固体饮料的制备方法,包括以下步骤:
S1:将茶叶、米草和天然草药分别提取得到茶叶提取物粉末、米草提取物粉末和天然草药提取物粉末按比例混合;
S2:将步骤S1中得到的提取物混合粉末与泡腾剂及辅料按比例混合,加入80%乙醇混合均匀;
S3:将步骤S2中得到的混合物在40±3℃恒温干燥箱中干燥30min后,干法制粒压片,得到具有降尿酸功能的茶叶固体饮料。
作为本发明的一种优选技术方案,所述步骤S1中,茶叶提取投茶量5g、茶水比1∶30浸提、超声时间10min、提取两次、合并两次提取滤液、浓缩烘干备用;米草和天然草药分别称取切断,6倍量70%乙醇加热回流提取3次,每次2h,过滤后合并滤液,减压回收乙醇,滤渣水回流提取3次,每次2h,过滤后合并滤液,减压浓缩烘干。

本发明取用天然草药包括蒲公英、荞麦、桑叶、丹参、金银花。蒲公英是常见的药食两用植物,富含多糖、黄酮、酚酸等多种生物活性物质,中医和现代医学研究表明其具有保肝沥胆,利尿,抗炎,抗菌和辅助保护化学性肝损伤作用;荞麦被誉为“五谷之王”,其营养成分丰富,富含蛋白质、脂肪、膳食纤维、维生素、矿物元素等,相比其它的大宗粮食作物具有独特的营养价值。荞麦淀粉颗粒比其它富含淀粉的农作物小,但其黏度大于谷类淀粉,且持水能力较强、释放葡萄糖速度慢。荞麦中脂肪的含量与其它谷物食品相当,但其中的不饱和脂肪酸所占比例较高,多为油酸和亚油酸。荞麦中不仅富含如维生素B、维生素E、芦丁等,还含有钾、镁、铁、铜、锌、硒和锰等矿物质元素;桑叶异名铁扇子,为桑科桑属植物桑的干燥叶。桑叶中含有17种氨基酸成分,并且含有丰富的d-葡萄糖苷酶抑制剂1-脱氧野尻霉素(DNJ),这是一种降糖成分,约占桑叶干物质的9%,是具有特殊功能的黄酮类化合物,其中人体自身不能合成的必需氨基酸约占3.28%,非必需氨基酸占约 1.84%。其所含的碳水化合物、维生素以及钙、锌、铁等微量元素、矿物质等多种人体必需的微量元素,营养价值极高;丹参为唇形科植物丹参的根及根茎,是一味临床常用中药,主要药效成分为脂溶性的丹参酮类以及水溶性的酚酸类化合物,是其发挥药理作用的重要物质基础。丹参中共有201种化合物,包括亲脂性二枯类化合物,水溶性酚酸和其他成分,这些化合物表现出多种药理活性,例如抑菌消炎、抗氧化、抗肿瘤、肝脏保护、抗动脉粥样硬化和抗糖尿病等作用;金银花是忍冬科植物忍冬的干燥花蕾,其性甘、寒,有清热解毒,凉血止血的功效,主要用于治疗温病发热、热毒血痢、痈肿疔疮、喉痹等病症,研究表明,金银花中主要含有挥发油类、黄酮类、酚酸类、三萜类等成分,具有较好的抗氧化和抑菌效果。
实施例1
该茶饮制备工艺中,原料配方按重量百分比计为茶叶提取物粉末15%、米草提取物粉末10%、天然药草提取物粉末10%、柠檬酸15%、碳酸氢钠15%、羟丙基淀粉15%、乳糖5%、聚乙二醇3%、微晶纤维素12%。
(1)将茶叶提取投茶量适当、茶水比1∶30(g/mL)浸提、超声时间10min、提取两次、合并两次提取滤液浓缩烘干备用,米草和天然草药分别称取适当切断,6倍量70%乙醇加热回流提取3次,每次2h,过滤后合并滤液,减压回收乙醇,滤渣水回流提取3次,每次2h,过滤后合并滤液,减压浓缩烘干;取米草和天然草药提取物粉末与茶叶提取物粉末均匀混合;
(2)将步骤(1)中得到的提取物混合粉末与柠檬酸(聚乙二醇分别)、碳酸氢钠(聚乙二醇包裹)、乳糖、聚乙二醇、微晶纤维素按比例混合,加入 80%乙醇混合均匀;

本发明为一种具有降尿酸功能的茶叶固体饮料,对其降尿酸效果进行测试。该测试基于具有降尿酸功能的茶叶固体饮料动物实验。
1、实验材料与仪器
动物:20-25克雄性昆明种小白鼠,清洁级二级40只,由南京医科大学动物饲养中心提供;造模剂:次黄嘌呤;样品:具有降尿酸功能的茶叶固体饮料粉末;空白对照组:生理盐水;阳性对照:苯溴马隆;溶剂:羧甲基纤维素钠-CMC-Na;贝克曼LX20全自动生化分析仪。
2、试剂配制:
(1)样品配制:准确称取样品具有降尿酸功能的茶叶固体饮料粉末(实施例1)1g,用0.8%CMC-Na溶液配制成4.5mg/ml剂量试液;
(2)50mg/ml次黄嘌呤的配制:称取1g次黄嘌呤,溶于20ml 0.8%CMC-Na 溶液中;
(3)0.5mg/ml苯溴马隆配制:称取20mg苯溴马隆,溶于40ml 0.8%CMC-Na 溶液中,使之浓度为0.5mg/ml。
3、模型动物试验组别设计:
(1)空白对照组(生理盐水)10只动物;
(2)模型组(CMC-Na)10只动物;
(3)样品组(4.5mg/ml)10只动物;
(4)阳性对照组(0.5mg/ml)10只动物。
4、实验方法:
对各组受试动物每天给药一次(每天给药前称体重),连续给药7天,各动物组实验方法:对1-4组动物分别进行胃内给药(i.g.),其中(1)组i.g. 生理盐水0.1ml/10g;(2)组i.g.0.8%CMC-Na 0.1ml/10g;(3)组i.g.样品 0.1ml/10g;(4)组i.g.溴苯马隆0.1ml/10g。
第8天最后一次给药30min后,除生理盐水组,其他各组腹腔(i.p.) 注射已配制的50mg/ml次黄嘌呤,按剂量1000mg/kg体重给药,在给药后 30-40min,立刻摘除眼球采血,置离心管中,在室温下,待血清渗出,取出离心(15000rpm,4min),吸取上层血清置带盖的样品管中,根据所得血清体积,全部用生理盐水配制到所需体积,在贝克曼LX20全自动生化分析仪上测定血尿酸(UA)、尿素氮(BUN)、肌酐(CR)等指标。所有数据经Spss11.5软件包处理,采用单因素方差分析,用均数±标准差(x±s)表示。
5、实验结果
实验鼠血尿酸(UA)、尿素氮(BUN)、肌酐(CR)测定结果如下:样品组(茶叶泡腾片)及阳性对照组(溴苯马隆)都有不同程度的对高尿酸血症小鼠血尿酸(UA)、尿素氮(BUN)、肌酐(CR)水平降低的影响,而且在本实验设计范围内,样品组降低血尿酸水平与模型组对比差异显著,即明显降低了高尿酸血症小鼠的血尿酸水平,说明样品对高尿酸血症小鼠有降低血尿酸水平的作用。
注:*P<0.05,表示差异显著,**P<0.01,表示差异非常显著,***P<0.001,表示差异极其显著。

Claims (3)
1.一种具有降尿酸功能的固体饮料,其特征在于,所述茶叶固体饮料按质量百分比包括以下原料:茶叶提取物粉末10%-15%、米草提取物粉末5%-10%、天然草药提取物粉末5%-10%、柠檬酸15%-25%、碳酸氢钠15%-25%、羟丙基淀粉15%-30%、乳糖3%-5%、聚乙二醇3%-5%和微晶纤维素5%-15%。
2.一种具有降尿酸功能的固体饮料的制备方法,其特征在于,包括以下步骤:
S1:将茶叶、米草和天然草药分别提取得到茶叶提取物粉末、米草提取物粉末和天然草药提取物粉末按比例混合;
S2:将步骤S1中得到的提取物混合粉末与泡腾剂及辅料按比例混合,加入80%乙醇混合均匀;
S3:将步骤S2中得到的混合物在40±3℃恒温干燥箱中干燥30min后,干法制粒压片,得到具有降尿酸功能的茶叶固体饮料。
3.根据权利要求1所述的一种具有降尿酸功能的固体饮料的制备方法,其特征在于:所述步骤S1中,茶叶提取投茶量5 g、茶水比1∶30 浸提、超声时间10 min、提取两次、合并两次提取滤液、浓缩烘干备用;米草和天然草药分别称取切断,6倍量70%乙醇加热回流提取3次,每次2h,过滤后合并滤液,减压回收乙醇,滤渣水回流提取3次,每次2h,过滤后合并滤液,减压浓缩烘干。
