Abstract
本发明公开了一种可提高工程化细胞外泌体中特定核酸分子载量的方法及其应用,公开了工程化改造细胞获得高载量目标核酸外泌体的工程化设计原理,基于多个桥接蛋白L7Ae或其变体的串联体与含有C/D box序列的RNA核酸分子自主对接,并通过桥接蛋白L7Ae或/和其变体与CD47等外泌体膜蛋白构建的融合蛋白(即支架蛋白),将RNA核酸分子靶向装载到外泌体中。通过该方法可有效提升外泌体对特定待装载RNA核酸分子的装载量,为以外泌体作为核酸药物递送系统的药物开发提供了重要理论和技术基础。本发明所生产的工程化外泌体能携带的核酸药物种类广、功能强,具有RNA核酸药物递送的通用性。
Description
技术领域
本发明属于外泌体作为核酸药物递送载体系统的领域,可用于疫苗、再生医学、药物治疗等各个领域的核酸药物的装载。具体涉及一种将含有特定序列的核酸靶向富集到外泌体中的工程化外泌体的制备手段及其应用。
背景技术
外泌体是细胞来源的具有膜结构的细胞外囊泡,其直径大小为30-150nm,在多泡体(MVBs)中产生,并通过MVBs与细胞膜的融合分泌到胞外液中。目前,外泌体被认为是细胞特异性分泌的细胞外囊泡,主要用于细胞间通讯并参与各种生命过程。由于其具有较高的生物相容性、生物利用度和穿越血脑屏障的特性,因此被用作核酸药物的递送载体。
迄今为止,在体内和体外外泌体已被成功地用于传递小分子、短RNA和蛋白质。与合成材料相比,外泌体递送系统具有多种优势,如它们具有内源性、稳定性、生物相容性、纳米尺寸、跨越血脑屏障的能力以及成分的可设计性等。目前使用外泌体的装载策略主要包括电穿孔和转染试剂等方法,但存在装载效率低,以及影响药物活性等问题。
近期,外泌体的工程化改造得到了广泛的关注,此方法可以在一定程度上提高外泌体装载的可控性和装载效率。但是,有限的囊泡空间仍会影响到装载效率,如何提高外泌体的有效装载率并定向将药物运送到靶点是需要解决的问题。
递送系统是mRNA等核酸药物发挥功能的关键,负责将小分子药物成分完整地运送至目标靶点,并且在合适的时机和环境条件下将其释放。但是,递送载体需要经过人体免疫系统的层层保护,容易引起过敏等免疫反应。递送系统还很大程度决定了小分子药物的储藏条件和储藏时限。目前拥有此技术的公司非常稀少,同时具有专利保护壁垒,是包括mRNA疫苗行业在内的“卡脖子”问题之一。
传统的核酸药物递送载体包括合成脂质纳米颗粒和病毒载体等,目前它们分别被用于BioNTech/Pfizer和Oxford/AstraZeneca的COVID-19疫苗,以及许多蛋白质、RNA和基因疗法。但是这两种方法都有很大的局限性,包括它们能到达的身体区域,能到达的组织内细胞数量,以及避免触发有害免疫应答的能力。
外泌体是内源性脂质纳米颗粒,在大小和功能上与合成的纳米颗粒类似,作为天然内源性转运载体,具有毒性低、无免疫原性、长半衰期、渗透性好更能跨越血脑屏障(BBB)等生物屏障等优势。外泌体在介导细胞间通讯、调节免疫反应方面也发挥着关键作用。此外,研究表明,外泌体由于从其亲本细胞而来的同源表面粘附蛋白、mRNA、DNA和脂质分子而具有先天归巢能力。更重要的是,外泌体表面CD47(一种跨膜蛋白)的表达有利于避免单核巨噬细胞系统(MPS)的免疫清除,延长血液循环时间。更重要的是,与病毒载体和脂质纳米颗粒等其他递送方法相比,外泌体的一个关键优势是可以精准地递送有效载荷,并不激活先天或后天免疫系统,因此患者在第一次治疗后不会产生对运载工具的免疫力,这使得重复给药变得更加容易。
目前外泌体的装载策略主要包括电穿孔和转染试剂等方法,这些方法都存在一些问题,包括装载效率低(装载效率10-40%),会破坏外泌体结构和活性以及影响药物活性等。近期,亦有工程化改造外泌体的设计方案,在一定程度上提高外泌体装载的可控性和装载效率。但是,在外泌体有限的囊泡空间内进一步提高其装载效率仍然是亟需解决的问题。
发明内容
本发明的目的是提供一种可提高细胞外泌体中特定核酸分子载量的方法及其应用,基于L7Ae-C/D box的自主对接,通过对外泌体膜蛋白的筛选,将核酸成功装载到外泌体膜蛋白CD47与L7Ae及其变体构成的融合蛋白(即支架蛋白)上,建立了基于外泌体膜蛋白CD47-LAe(n)的高载量外泌体-核酸装载系统。同时,对CD47的蛋白结构和L7Ae结构进行了深度的分析,通过结构的优化成功提高了该装载系统的装载效率,建立了全新的基于P4支架蛋白(CD47-L7Ae(n))的高载量外泌体-核酸装载系统。
为解决上述技术问题,本发明的第一方面,采用如下技术方案:一种高载量外泌体-核酸装载系统,包括外泌体装载核酸用支架蛋白表达质粒、特定待装载核酸序列表达质粒;
外泌体装载核酸用支架蛋白表达质粒含有外泌体膜蛋白的编码序列,以及至少一个桥接蛋白L7Ae或其变体的编码序列;
特定待装载核酸序列表达质粒包括特定待装载核酸分子和与特定待装载核酸分子直接或间接连接的特殊序列,所述的特殊序列为C/D box及其同功能衍生序列(即与C/Dbox具有同等功能的C/D box衍生序列),所述的特殊序列与L7Ae或/和其变体自主对接。
特定待装载核酸分子为RNA,例如mRNA、circRNA和lncRNA。
优选地,所述的支架蛋白,即外泌体膜蛋白与多个桥接蛋白L7Ae或/和其变体串联融合表达形成外泌体膜蛋白-L7Ae(n)融合蛋白,外泌体膜蛋白-L7Ae(n)融合蛋白即所述的支架蛋白,n为L7Ae及其变体的数量;所述的外泌体装载核酸用支架蛋白表达质粒和特定待装载核酸序列表达质粒,通过共转染使特定待装载核酸分子装载到外泌体中。特殊序列为C/D box序列或其同功能衍生序列,该序列可以特异性识别并结合L7Ae蛋白及其变体。
所述的支架蛋白为外泌体膜蛋白如CD63、CD47、CD9和CD81或其变体。
优选的,所述的特定待装载核酸序列表达质粒还包括相互连接的特定核酸分子和C/D box序列。在实际药物应用中,只需使特定待装载核酸分子与C/D box序列连接,不需加入EGFP或其等效类似物(仅作荧光标记之用)。
外泌体膜蛋白与桥接蛋白L7Ae(或其变体)之间通过柔性接头连接;当不止一个所述的桥接蛋白L7Ae或其变体形成串联体时,外泌体膜蛋白与第一个桥接蛋白L7Ae(或其变体)之间由柔性接头连接,为了减少桥接蛋白间以及装载核酸间的相互干扰,第一个桥接蛋白与第二个桥接蛋白及后面的桥接蛋白之间由刚性接头连接,例如第一个桥接蛋白与第二个桥接蛋白之间由刚性接头连接,第二个桥接蛋白与第三个桥接蛋白之间由刚性接头连接,以此类推。
桥接蛋白L7Ae的变体指在野生型L7Ae(即L7Ae-WT)的羧基端去除21个氨基酸(整个ɑ螺旋)所得的L7Ae变体(即L7Ae_delC21),或者为L7Ae的可以减少空间位阻的其他变体,例如其它长度的截断体或突变体。
优选地,所述的柔性接头包括但不限于GSSS、GSSSGSSSGS或GGS,所述的刚性接头包括但不限于PWRPWRP或PWRP,也可以为其他柔性或刚性接头;一个外泌体装载核酸用支架蛋白表达质粒中,当不止一个所述的桥接蛋白L7Ae或其变体形成串联体时,柔性接头和/或刚性接头为不止一种或仅为一种序列,桥接蛋白L7Ae或其变体为不止一种或仅为一种序列。
进一步地,所述的特定待装载核酸分子为RNA核酸药物分子或RNA核酸分子。
更进一步地,所述的特定待装载核酸分子为编码疫苗抗原的核酸序列、抗肿瘤核酸药物、核酸抗体药物和其它编码各种生长因子、抗体和蛋白的核酸分子。
更具体地,所述的特定待装载核酸分子为编码EGF的核酸序列、编码胰岛素的核酸序列或其他生长因子的核酸分子。
在一个实施例中,C/D box序列如SEQ ID NO.17所示或为其同功能衍生序列。
本发明的第二方面,是提供一种可以在外泌体中提高特定核酸分子载量的方法,包括如下步骤:
构建外泌体装载核酸用支架蛋白表达质粒和特定待装载核酸序列表达质粒;外泌体装载核酸用支架蛋白表达质粒含有外泌体膜蛋白的编码序列,以及至少一个桥接蛋白L7Ae或其变体的编码序列;特定待装载核酸序列表达质粒包括特定待装载核酸分子和与特定待装载核酸分子直接或间接连接的C/D box序列;
将外泌体装载核酸用支架蛋白表达质粒和特定待装载核酸序列表达质粒共转染至细胞中,外泌体膜蛋白与桥接蛋白L7Ae或其变体融合表达形成融合蛋白(即支架蛋白),其中C/D box与L7Ae或/和其变体自主对接,使特定待装载核酸分子通过融合蛋白装载到外泌体中,外泌体被分泌到细胞外的同时将特定核酸分子携带到细胞外;
从细胞培养上清中富集含有特定核酸分子的外泌体。
优选地,通过瞬时转染或构建稳转细胞株实现特定核酸分子装载到外泌体中;所述的特定核酸分子为RNA核酸药物或RNA核酸分子。
本发明的第三方面是提供包含前述的高载量外泌体-核酸装载系统的核酸药物或核酸分子。
优选地,提供包含前述的高载量外泌体-核酸装载系统的核酸疫苗、核酸抗体药物、抗肿瘤药物、基因治疗或肿瘤细胞治疗用核酸分子或再生医学药物。
本发明的第四方面是提供高载量外泌体-核酸装载系统在制备核酸药物中的用途。
优选地,所述的核酸药物用途为核酸疫苗、抗肿瘤、基因治疗、细胞治疗或再生医学药物领域。
基于L7Ae蛋白或其变体与含有C/D box序列的核酸药物的自主对接,并且通过L7Ae蛋白或其变体与CD47等外泌体膜蛋白的融合表达,将核酸药物靶向装载到外泌体中。
一种提高工程化细胞外泌体中特定核酸药物载量的方法,所述方法包含:
1)核酸分子,所述核酸分子含有特殊序列以达到外泌体靶向富集。
2)支架蛋白,所述支架蛋白在外泌体生物发生过程中会被组装到外泌体中。
3)桥接蛋白,所述桥接蛋白与外泌体膜蛋白融合表达,可以结合核酸分子上的特定序列,最终达到核酸药物在外泌体中的靶向富集。
4)外泌体膜蛋白和第一个桥接蛋白相融合的GSSS柔性接头以及第二个桥接蛋白及以后L7Ae变体之间的柔性接头GSSSGSSSGS或刚性接头PWRPWRP。所述刚性接头PWRPWRP可以将多个桥接蛋白之间保持更好的空间结构,减少桥接蛋白之间和所装载核酸之间的空间位阻,从而增强桥接蛋白与核酸分子装载效率和稳定性;所述柔性接头GSSSGSSSGS导致所连接的L7Ae可能摆动或折叠弯曲从而空间位阻增大,引起核酸装载效率和装载量较低。
5)支架蛋白和桥接蛋白的结构改造,所述结构通过桥接蛋白的多个串联,可以增强桥接蛋白与核酸分子上的特定序列的结合,最终提高外泌体装载核酸药物的载量。
设计含有C/D box的核酸药物序列,并构建相应的质粒。
设计外泌体膜蛋白和L7Ae或其变体的融合蛋白(即支架蛋白),例如、P1、P2、P3、P4,并构建相应的质粒。
将质粒通过转染试剂或电转的方法转染至细胞,例如HEK293T(简写为293T)、HEK293F(简写为293F)、干细胞或免疫细胞或其它真核细胞,建立稳转细胞系或仅是达到瞬转的目的。
转染上述质粒的细胞在产生外泌体时,会将含有特定序列的核酸分子通过融合蛋白表达质粒P1、P2、P3和P4等富集到外泌体中,外泌体被分泌到细胞外的同时会将这些核酸分子携带到细胞外,当携带有目标核酸分子的外泌体被受体细胞吞噬后,外泌体装载物被受体细从外泌体释放,并发挥其功能。
通过超离或分子筛的方法从细胞上清中富集外泌体。
1)提取外泌体中的总RNA,并通过qPCR的方法检测核酸分子在外泌体中是否被富集。
2)将富集的外泌体加入到受体细胞的培养上清中,例如293T、293F、干细胞或免疫细胞或其它真核细胞,检测mRNA的表达情况,或miRNA等核酸的调控情况。
与现有技术相比,本发明的优点在于:可以提高在工程化细胞外泌体中靶向装载核酸药物的载量;可以装载不同序列的核酸药物;可以在不同的细胞系中实现该方法。

具体实施方式
下面结合实施例及附图对本发明作进一步详细的描述,但不用来限制本发明的范围。
本发明中使用的相关核酸由生物工程(上海)有限公司合成。P1、P2、P3和P4序列由生物工程(上海)有限公司合成。转染试剂:Polyethylenimine购买自sigma;lipofectamine3000购买自ThermoFisher。Luciferase Reporter Gene Assay Kit萤火虫萤光素酶报告基因检测试剂盒购买自翌圣生物科技(上海)股份有限公司。RNA提取试剂盒MiniBESTUniversal RNA Extraction Kit购买自TaKaRa,HiScript III 1st Strand cDNASynthesis Kit反转录试剂盒购买自诺唯赞。Premix Ex TaqTM II qPCR试剂盒购买自TaKaRa。
以下实施例的实验原理是基于桥接蛋白L7Ae蛋白与C/D box的自主对接,本发明将外泌体的膜蛋白与桥接蛋白L7Ae连接,同时将需装载的核酸分子与C/D box连接,通过桥接蛋白L7Ae与C/D box的自主对接,可以将需装载的核酸分子装载到外泌体上。具体的实验操作方法是将含有桥接蛋白L7Ae的外泌体装载核酸用支架蛋白表达质粒和含有C/D box的特定待装载核酸序列表达质粒进行细胞的共转染,转染验证成功后,将含有装载核酸分子的外泌体的上清进行富集。再将富集的外泌体的上清加入到新的受体细胞中进行细胞培养(其中外泌体中所装载核酸分子为预设的携带有C/D box或同功能衍生序列的mRNA、circRNA和/或lncRNA)。将受体细胞回收进行验证和定量,如果装载成功,那么受体细胞就会吸收上清中的外泌体并表达出外泌体装载核酸药物。
实施例1:外泌体核酸装载用支架蛋白的筛选
(1)利用GFP荧光进行筛选
构建含有L7Ae的支架蛋白质粒和C/D box-GFP表达质粒。质粒构建好后,准备24孔板的293T细胞,将含有L7Ae的支架蛋白质粒和C/D box-GFP质粒进行共转染,除去上清后更换为不含外泌体血清的培养基继续进行培养,培养条件为37℃,5%CO2,24h进行显微镜观察,观察到绿色荧光,如图1中的1A所示,证明质粒共转染成功。富集培养了34h的培养基上清,300×g,5min离心,将上清移至新的1.5mL管中,2000×g,5min再进行一次离心,将上清加入到新的24孔板的293T受体细胞中,48h后用显微镜观察荧光。CD47-L7Ae支架蛋白质粒与C/D box-GFP质粒的共转染组观察到了微弱的绿色荧光,如图1中的1B所示,293T空白对照组和C/D box-GFP质粒单独转染组无荧光。同时,还在其它支架蛋白的共转染组观察到了微弱的绿色荧光。该实验结果证明,CD47-L7Ae可作为支架蛋白用于外泌体的装载,但该实验中GFP的荧光比较微弱,只能观察到少许细胞有发光,故增加上清中的外泌体含量,从而提高受体细胞的荧光强度。
无法准确测定荧光强度是该实验存在的一个问题,故在此实验基础上进行质粒改进,将C/D box-GFP质粒中GFP部分变换为Luciferase报告基因。Luciferase报告基因系统是以荧光素(luciferin)为底物来检测萤火虫荧光素酶活性的实验,荧光素底物会在荧光素酶作用下发光(波长540-600nm),且光强能反映荧光素酶的表达量。
(2)利用Luciferase报告基因进行筛选
将C/D box-GFP质粒的序列中GFP部分变换为Luciferase报告基因后,进行质粒构建,质粒构建后再次进行支架蛋白的筛选实验。准备6孔板的293T细胞,进行质粒共转染,除去上清后更换为不含外泌体血清的培养基继续进行培养,培养条件为37℃,5%CO2,富集培养了34h的培养基上清,300×g,5min离心,将上清移至新的1.5mL管中,2000×g,5min再进行一次离心,将上清加入到新的24孔板的293T受体细胞中,48h回收细胞进行荧光素酶活性检测。荧光素酶活性检测结果显示CD47-L7Ae支架蛋白质粒与C/D box-Luc质粒的共转染组高于293T空白对照组和C/D box-Luc质粒单独转染组,如图2所示。该实验结果证明,CD47-L7Ae可作为支架蛋白用于外泌体的装载。
该实验结果虽然显示CD47-L7Ae可作为支架蛋白用于外泌体的装载,但其装载效率并不是很高,为了提高其装载效率,我们对CD47-L7Ae结构进行了深入的分析和改造。
实施例2:支架蛋白的结构优化及验证
(1)对支架蛋白和桥接蛋白L7Ae结构进行改造
为了提升外泌体的核酸药物装载效率,本发明对外泌体膜蛋白CD47和桥接蛋白L7Ae的结构进行了深入的分析和改造,针对两种蛋白的连接方式设计了全新的GSSS柔性linker。同时,通过对桥接蛋白L7Ae的结构进行改造,实现了多个桥接蛋白构成的串联体。设计了P1、P2、P3和P4四个质粒,质粒包括CD47外泌体膜蛋白、GSSS柔性linker和桥接蛋白L7Ae。为了更好的将两个蛋白连接,本发明设计了PWRPWRP的刚性linker序列,并应用到了P4质粒中。四个质粒的图谱为图6-9所示,以下为四个质粒分别表达的支架蛋白的氨基酸序列。
P1支架蛋白氨基酸序列(CD47-GSSS-1×L7Ae-WT序列,其中斜体部分为GSSS柔性linker序列,大写的字母均为氨基酸缩写,SEQ ID NO.1)
P2支架蛋白氨基酸序列(CD47-GSSS-1×L7Ae-del-C21序列,其中斜体部分为GSSS柔性linker序列,大写的均为氨基酸缩写,SEQ ID NO.2)
P3支架蛋白序列(CD47-GSSSGSSSGS×3-3×L7Ae-WT序列,其中斜体部分为GSSSGSSSGS柔性linker序列,大写的均为氨基酸缩写,SEQ ID NO.3)
P4支架蛋白序列(CD47-GSSS-2×L7Ae_delC21-1×L7Ae-WT序列,其中斜体部分为GSSS柔性linker序列和PWRPWRP刚性linker序列,大写的均为氨基酸缩写,其中前两个桥接蛋白为L7Ae_delC21,第三个桥接蛋白为L7Ae-WT,CD47与第一个桥接蛋白通过GSSS连接,第一个桥接蛋白与第二个桥接蛋白之间通过PWRPWRP连接,第二个桥接蛋白与第三个桥接蛋白之间通过PWRPWRP连接,对应于SEQ ID NO.4)
构建P1-P4质粒的方法为:将分别编码上述P1-P4支架蛋白的核苷酸序列插入目标载体pCDNA3(+),其中编码上述P1-P4支架蛋白的核苷酸序列的5’端加上核酸序列AAGCTT(酶切位点),而3’端加上核酸序列GAATTC(酶切位点)。
(2)利用Luciferase报告基因进行验证及筛选
质粒(如图6-9所示)构建好后,将培养于6孔板内的293T细胞分别进行P1、P2、P3和P4膜蛋白-L7Ae融合支架蛋白质粒与C/D box-Luc质粒的共转染,然后更换为不含外泌体血清的培养基继续进行培养,培养条件为37℃,5%CO2,富集培养了34h的培养基上清,300×g,5min离心,将上清移至新的1.5mL管中,2000×g,5min离心一次,将上清加入到新的24孔板的293T受体细胞中,培养48h后回收细胞进行荧光素酶活性检测。荧光素酶活性检测结果显示新构建的P1和P4质粒与C/D box-Luc质粒的共转染组明显提高了外泌体的装载效率,如图3所示。特别是P4组与对照组相比,装载效率提升了约3倍,其中CD47-L7Ae(即CD47直接与一个野生型L7Ae形成融合蛋白所对应的质粒)及C/D box-Luc组为对照组,293T组为空白对照组。该实验结果证明,新构建的P4可作为支架蛋白(即外泌体膜蛋白CD47-L7Ae(n)融合蛋白,这里n为3)用于外泌体的装载。
实施例3:P4富集了外泌体中连接C/D box的核酸药物的mRNA
在细胞蛋白水平筛选到较强的支架蛋白P4之后,本发明通过qPCR检测了P4富集外泌体内核酸药物的能力。本实施例所涉及到的工程化外泌体的制备及其应用主要包括以下步骤:
(1)核酸药物的质粒设计及核酸药物的蛋白表达:
本发明设计了两个核酸药物质粒,一个是胰岛素的氨基酸序列连接EGFP蛋白,在终止密码子之后加入C/D box序列(I01-EGFP-C/D box,p458);另一个是IL-2的信号肽连接Flag-EGF以及EGFP,终止密码子之后为C/D box序列(E01-EGFP-C/D box,p459)。
I01-EGFP-C/D box的序列(其中粗体为I01序列,I01序列为胰岛素氨基酸序列,斜体部分是EGFP(增强绿色荧光蛋白)氨基酸序列,大写的均为氨基酸缩写。其中,SEQ IDNO.5为胰岛素与EGFP融合蛋白的氨基酸序列。在构建质粒时,胰岛素-EGFP融合蛋白CDS序列后紧接有C/D box序列,SEQ ID NO.17)
在构建相应的质粒时,将编码胰岛素的核酸序列、编码EGFP蛋白的核酸序列、终止密码子、C/D box序列依序连接,编码EGFP蛋白的核酸序列之后是终止密码子,在终止密码子之后为C/D box序列,C/D box序列为5’gggcgtgatccgaaaggtgaccc3’(SEQ ID NO.17)。
E01-EGFP-C/D box的序列(其中大写加下划线为IL2信号肽序列,粗体加斜体序列为Flag序列,粗体为E01序列,E01为EGF(表皮细胞生长因子)的序列,斜体不加粗部分是EGFP序列,大写的均为氨基酸缩写,其中SEQ ID NO.6是EGF与EGFP融合蛋白的氨基酸序列。在构建质粒时,EGF-EGFP融合蛋白CDS序列后紧接有C/D box序列,SEQ IDNO.17)
在构建相应的质粒时,将编码IL2信号肽的核酸序列、编码Flag的核酸序列、编码EGF的核酸序列、编码EGFP蛋白的核酸序列、终止密码子和C/D box序列依序连接,编码EGFP蛋白的核酸序列之后是终止密码子,在终止密码子之后为C/D box序列,C/D box序列为5’gggcgtgatccgaaaggtgaccc3’(SEQ ID NO.17)。
其中,CD47的氨基酸序列如SEQ ID NO.18所示,L7Ae-del-C21的氨基酸序列如SEQID NO.19所示,L7Ae-WT的氨基酸序列如SEQ ID NO.20所示,EGFP的氨基酸序列如SEQ IDNO.21所示。
本发明首先在293T细胞内验证了两个质粒的表达,如图4所示。在用Lipo3000试剂转染293T细胞24h后,我们在荧光显微镜下观察到了EGFP的表达。实验结果表明该核酸药物序列可以正常表达相应的目的蛋白。
(2)RT-qPCR验证P4介导的外泌体中目标mRNA载量
该实验将样品分为以下6组:P458组、P459组、P458+P1组、P458+P4组、P459+P1组和P459+P4组。将构建好的支架蛋白质粒(P1和P4)分别与携带有C/D box的待装载目标核酸质粒(P458和P459)通过PEI共转染至悬浮培养的293F细胞中,转染前细胞密度为1×106/mL,每组准备两瓶T75细胞进行实验。细胞转染实验以c组为例:先配置A液:用移液器将10μgp458+10μg p1(共约20μg)质粒加入到1.5mL的无菌离心管中,再用移液器吸取375μL的Opti-MEM加到离心管中,用移液器轻柔吹打混匀,室温静置5min。配置A液的同时准备B液,用移液器将37.5μL的PEI(终浓度:1μg/μL)加入到375μL的Opti-MEM中,轻柔吹打混匀,室温静置5min。用1mL移液器将B液加到A液中,轻柔混匀,室温静置20min。转染5h后更换为新鲜的完全培养基继续进行细胞培养,培养条件为37℃,5%CO2。为了提高上清中外泌体的含量,将细胞继续培养约50h。细胞培养50h完成后,将细胞悬液转移到50mL离心管中,3,000×g离心30min。将上清转移到恩泽康泰SEC柱子上,利用SEC柱分离纯化上清中的外泌体。
需要对富集到的外泌体中的mRNA进行qPCR的定量分析,首先使用RNA试剂盒提取外泌体中的mRNA,然后利用反转录试剂盒将mRNA反转录成cDNA,最后通过荧光定量PCR实验对mRNA进行相对定量,具体的操作流程如下:
a)去除基因组DNA

qPCR结果显示C/D box及编码支架蛋白P4质粒的共转染能够明显增强外泌体内携带C/D box的目标蛋白mRNA的装载量与细胞蛋白水平的筛选结果相同,如图5所示,其中P458及P459组为对照组,293F组为空白对照组。
本发明通过对细胞进行定向的工程化改造,达到提高其工程化细胞外泌体对特定核酸分子装载量的效果。本发明公开了工程化改造细胞获得高载量目标核酸外泌体的工程化设计原理,基于多个L7Ae蛋白或/和其变体的串联体与含有C/D box序列的待装载核酸自主对接,并通过桥接蛋白L7Ae或/和其变体与CD47等外泌体膜蛋白构建的融合蛋白,将核酸药物靶向装载到外泌体中。通过该方法可有效提升外泌体对特定待装载核酸的装载量,为以外泌体作为核酸药物递送系统的核酸疫苗、抗肿瘤药物、核酸抗体药物、细胞治疗和再生医学等方向的药物开发提供了重要理论和技术基础。本发明所生产的工程化外泌体能携带的核酸药物种类广、功能强,具有核酸药物递送的通用性。
上述说明并非对本发明的限制,本发明也并不限于上述举例。本技术领域的普通技术人员在本发明的实质范围内,作出的变化、改型、添加或替换,也应属于本发明的保护范围,本发明的保护范围以权利要求书为准。
Claims (15)
1.一种高载量外泌体-核酸装载系统,其特征在于,包括外泌体装载核酸用支架蛋白表达质粒、特定待装载核酸序列表达质粒;
外泌体装载核酸用支架蛋白表达质粒含有外泌体膜蛋白的编码序列,以及至少一个桥接蛋白L7Ae或/和其变体的编码序列;
特定待装载核酸序列表达质粒包括特定待装载核酸分子和与特定待装载核酸分子直接或间接连接的特殊序列,其中特殊序列为C/D box序列或具有同等功能的C/D box衍生序列,所述的特殊序列与L7Ae或其变体自主对接。
2.根据权利要求1所述的一种高载量外泌体-核酸装载系统,其特征在于,所述的支架蛋白为:由外泌体膜蛋白与多个桥接蛋白L7Ae或/和其变体串联融合表达形成的外泌体膜蛋白-L7Ae(n)融合蛋白,n为L7Ae或/和其变体的数量;所述的外泌体装载核酸用支架蛋白表达质粒和特定待装载核酸序列表达质粒,通过共转染使特定待装载核酸分子装载到细胞外泌体中。
3.根据权利要求1所述的一种高载量外泌体-核酸装载系统,其特征在于,所述的支架蛋白由CD47或其它外泌体膜蛋白(即包括但不限于CD47)与在其C末端串联的L7Ae或/和其变体组成。
4.根据权利要求2所述的一种高载量外泌体-核酸装载系统,其特征在于,所述的特定待装载核酸序列表达质粒还包括相互连接的特定待装载核酸分子和C/D box序列或其同功能衍生序列。
5.根据权利要求1所述的一种高载量外泌体-核酸装载系统,其特征在于,外泌体膜蛋白与桥接蛋白L7Ae或/和其变体之间通过柔性或/和刚性接头连接;当不止一个所述的桥接蛋白L7Ae或/和其变体形成串联体时,相邻的桥接蛋白L7Ae和其变体之间由刚性接头连接。
6.根据权利要求1所述的一种高载量外泌体-核酸装载系统,其特征在于,桥接蛋白L7Ae的变体指在野生型L7Ae的羧基端去除21个氨基酸所得的L7Ae变体,或者为任何一个减少L7Ae-RNA相结合空间位阻的其它L7Ae变体。
7.根据权利要求5所述的一种高载量外泌体-核酸装载系统,其特征在于,所述的柔性接头包括但不限于GSSS、GSSSGSSSGS或GGS,所述的刚性接头包括但不限于PWRPWRP或PWRP;一个外泌体装载核酸用支架蛋白表达质粒中,当不止一个所述的桥接蛋白L7Ae或/和其变体形成串联体时,柔性接头和/或刚性接头为不止一种或仅为一种序列,桥接蛋白L7Ae或其变体为不止一种或仅为一种序列。
8.根据权利要求2所述的一种高载量外泌体-核酸装载系统,其特征在于,所述的特定待装载核酸分子为核酸序列;支架蛋白的氨基酸序列如SEQ ID NO.4所示。
9.根据权利要求1所述的一种高载量外泌体-核酸装载系统,其特征在于,所述的特定待装载核酸分子为编码疫苗抗原的核酸序列、抗肿瘤核酸药物、核酸抗体药物或其它编码各种生长因子、抗体和蛋白的核酸分子。
10.根据权利要求1所述的一种高载量外泌体-核酸装载系统,其特征在于,所述的特定待装载核酸分子为编码EGF的核酸序列、编码胰岛素的核酸序列或其他生长因子的核酸分子。
11.根据权利要求2所述的一种高载量外泌体-核酸装载系统,其特征在于,C/D box序列如SEQ ID NO. 17所示或为其同功能衍生序列。
12.一种可提高细胞外泌体中特定核酸分子载量的方法,其特征在于,包括如下步骤:
构建细胞外泌体装载核酸用支架蛋白表达质粒和特定待装载核酸序列表达质粒;外泌体装载核酸用支架蛋白表达质粒含有外泌体膜蛋白的编码序列,以及至少一个桥接蛋白L7Ae或/和其变体的编码序列;特定待装载核酸序列表达质粒包括特定待装载核酸分子和与特定待装载核酸分子直接或间接连接的C/D box序列;
将外泌体装载核酸用支架蛋白表达质粒和特定待装载核酸序列表达质粒共转染至外泌体供体细胞中,外泌体供体细胞为HEK293T、HEK293F、干细胞或免疫细胞,外泌体膜蛋白与桥接蛋白L7Ae或/和其变体融合表达形成融合蛋白,该融合蛋白即所述的支架蛋白,其中C/D box或其同功能衍生序列与L7Ae和其变体自主对接,使特定待装载核酸分子通过融合蛋白装载到外泌体中,外泌体被分泌到细胞外的同时将特定核酸序列分子携带到细胞外;从细胞上清中富集含有特定核酸分子的外泌体。
13.根据权利要求12所述的方法,其特征在于,通过瞬时转染或构建稳定转染的细胞株实现特定待装载核酸分子装载到细胞外泌体中;所述的特定待装载核酸分子为任何携带有C/D box 或其同功能衍生序列的RNA核酸药物或RNA核酸分子。
14.包含权利要求1-11任意一项所述的高载量外泌体-核酸装载系统的核酸药物或核酸分子,其特征在于,所述的核酸药物或核酸分子为核酸疫苗、抗肿瘤核酸药物、核酸抗体药物、基因治疗或肿瘤细胞治疗用核酸分子和再生医学药物。
15.权利要求1-11任意一项所示的高载量外泌体-核酸装载系统在制备核酸药物中的用途,其特征在于,所述的核酸药物用途为核酸疫苗、抗肿瘤、基因治疗、细胞治疗或再生医学药物领域。
