技术领域
本发明涉及口腔护理技术领域,尤其涉及一种银耳多糖洁牙护理液及其制备工艺。
背景技术
口腔护理用品是人们日常生活中不可或缺的清洁、护理及保健用品,这一类产品种类繁多,包括洁牙护理液、牙膏、口香糖、漱口水、牙粉、口腔喷雾剂等,主要用于保持口腔和牙齿清洁、促进口气清新、护理牙龈、维护口腔良好微环境、缓解口腔不适症状等。洁牙护理液是口腔护理用品的一种,借助牙刷的机械摩擦的作用清洁牙齿表面,对牙齿及其周边进行清洁,使口腔净化清爽。
许多成人有不同程度的牙本质敏感症。牙本质敏感症是指暴露的牙本质因受温度、机械、化学或渗透压等外界刺激而产生的短暂并尖锐的牙齿酸痛症状,具有发作迅速、疼痛尖锐、时间短暂等特点,是一种常见的口腔病症,患病率较高,严重影响患者的生活质量。造成牙本质敏感的原因很多,包括能使釉质完整性受到破坏,牙本质暴露的各种牙体疾病,如磨耗、楔状缺损、牙折、龋病以及牙周萎缩致牙颈部暴露等均可发生牙齿感觉过敏症。
因此、需要开发一种洁牙护理液,针对性的去解决牙本质敏感症的问题。
发明内容
为了解决上述背景技术中提到的问题,本发明提供一种银耳多糖洁牙护理液,按照重量份计算包括如下组分原料:乙二胺四乙酸二钠0.05-0.15份、十二烷基硫酸钠0.10-1.00份、甘草酸二钾0.05-0.15份、C8-16烷基糖苷15.00-35.00份、丙二醇5.00-15.00份、薄荷脑0.05-0.25份、对羟基苯甲酸甲酯0.01-1.50份、聚甘油-10月桂酸酯1.00-3.00份、丹皮酚0.05-0.15份、柠檬酸0.10-1.00份、西吡氯铵0.05-0.15份、功能辅助成分0.5-5份、银耳多糖0.01-1.50份、糖精钠0.10-1.00份、茉莉花提取物0.05-0.25份、氨甲环酸0.05-1.00份、忍冬花提取物0.05-1.00份,纯化水补充至100份。

复合材料的制备
将碳纳米管加入浓硝酸、浓硫酸混合体系中拌均匀,加热、超声处理,冷却后过滤、洗涤、烘干,得到酸化的碳纳米管,将酸化的碳纳米管分散到无水乙醇中,加入壳聚糖,加热、超声处理,过滤、洗涤、烘干,得到碳纳米管-壳聚糖复合材料;
改性复合材料的制备
使用氯化钙溶液浸渍上述复合材料,搅拌均匀,调节pH至碱性,添加右旋泛酸钙、L-赖氨酸、烟酰胺后搅拌均匀,过滤、洗涤、烘干,得到改性复合材料,即组分A;
所述组分B的制备方法如下:
使用氯化镁溶液浸渍羟基磷灰石,搅拌均匀,调节pH至碱性,添加氯化锶搅拌均匀,过滤、洗涤、烘干,得到改性羟基磷灰石,即组分B;
所述组分C为氢化钙;
将组分A、组分B、组分C充分混合均匀,得到功能辅助成分。

取新鲜茉莉花在120℃条件下杀青5min,50℃干燥10min,研磨至过100目筛,得茉莉花粉;
按固液比1:10的质量比将茉莉花粉分散在水中,在15MPa条件下处理5min,加入茉莉花粉质量0.1%的十二烷基硫酸钠搅拌混合均匀,60℃提取10min,5000rpm离心5min、过滤,取滤液A;
向滤液A中加入无水乙醇至含醇量30%,搅拌均匀后4℃静置1h,过滤,去除沉淀,得滤液B,90℃减压蒸馏,得到茉莉花提取物。

取新鲜忍冬花在120℃条件下杀青5min,50℃干燥10min,研磨至过100目筛,得忍冬花粉;
按固液比1:10的质量比将忍冬花粉分散在水中,在15MPa条件下处理5min,加入忍冬花粉质量0.1%的十二烷基硫酸钠搅拌混合均匀,60℃提取10min,5000rpm离心5min、过滤,取滤液C;
向滤液C中加入无水乙醇至含醇量30%,搅拌均匀后4℃静置1h,过滤,去除沉淀,得滤液D,90℃减压蒸馏,得到忍冬花提取物。
本发明还提供一种银耳多糖洁牙护理液制备工艺,按照质量份计算,将乙二胺四乙酸二钠、十二烷基硫酸钠、甘草酸二钾,纯化水按照配比在85℃乳化、保温20min,降温至60℃,按配比添加C8-16烷基糖苷、丙二醇、薄荷脑、对羟基苯甲酸甲酯、聚甘油-10月桂酸酯、丹皮酚、柠檬酸搅拌均匀,降温至50℃,按配比添加西吡氯铵、功能辅助成分搅拌均匀,降温至45℃,按配比添加银耳多糖、糖精钠、茉莉花提取物搅拌均匀,接着按配比加入氨甲环酸、忍冬花提取物搅拌均匀,灌装,得到银耳多糖洁牙护理液。
有益效果:
1、本发明的洁牙护理液中加入了功能辅助成分,能够有效的降低牙龈指数和牙本质敏感程度,在动物实验中可知,采用实施例1的配方的洁牙护理液产生了很好的效果,使用两周后的牙龈指数降低至0,牙本质敏感程度降低至0.1,在人体临床试验中也产生了很好的效果,有94%的患者,牙本质敏感程度降低至0-0.5。
2、本发明的功能辅助成分包括组分A,在碱性条件下将右旋泛酸钙、L-赖氨酸、烟酰胺填充在碳纳米管-壳聚糖多孔复合材料的内部空隙中,组分A能够很好的与牙组织黏附,减少牙髓神经刺激,抑制牙石、菌斑的形成,结合西吡氯铵、氨甲环酸的使用,能够明显的提高口腔内部清洁能力,降低牙石、菌斑的产生,起到抑菌消炎的效果,再通过右旋泛酸钙、L-赖氨酸、烟酰胺营养牙齿,使牙齿再矿化,增加牙齿的耐酸性,提高抗敏作用,能够预防或辅助治疗牙周炎引起的牙龈萎缩。
3、本发明的功能辅助成分包括组分B,在碱性条件下将氯化锶填充在羟基磷灰石的内部空隙中,组分B能够很好的与牙组织黏附,通过吸附内源性锶盐、钙磷成为牙本质矿物质阻塞牙本质小管,用于降低治疗牙本质敏感,促进牙本质小管再修复。
4、本发明的功能辅助成分包括组分C,组分C为氢化钙,氢化钙在口腔中遇水会分解,产生微量的氢气和钙离子,氢气可以调节碳纳米管-壳聚糖多孔复合材料和羟基磷灰石空隙中功能物质的释放速度,使功能物质可以在口腔中长时间的保持有效浓度,产生了协同增效的配合作用,使得牙齿长时间处于保护状态。
5、本发明的牙膏中使用了银耳多糖、茉莉花提取物、忍冬花提取物气味清洗,香味宜人,具有很好的使用体验。
具体实施方式
制备例1
功能辅助成分的制备,所述功能辅助成分包括组分A、组分B和组分C,所述组分A的制备方法如下:
复合材料的制备
将50g碳纳米管分批(每批10g)加入浓硝酸、浓硫酸(体积比3:1,总体积400ml)混合体系中拌均匀,加热至70℃、超声处理2h,冷却后过滤、蒸馏水洗涤、60℃烘干,得到酸化的碳纳米管,将30g酸化的碳纳米管分散到1000ml无水乙醇中,加入50g壳聚糖,加热至50℃、超声处理1h,过滤、无水乙醇洗涤、60℃烘干,得到碳纳米管-壳聚糖复合材料;
改性复合材料的制备
使用500ml质量分数为10%氯化钙溶液浸渍上述50g复合材料,1000rpm搅拌30min,搅拌状态下滴加氨水调节pH至9,添加10g右旋泛酸钙、5gL-赖氨酸、2g烟酰胺后搅拌2h,过滤、无水乙醇洗涤、50℃烘干,得到改性复合材料,即组分A;
所述组分B的制备方法如下:
使用500ml质量分数为5%氯化镁溶液浸渍50g羟基磷灰石,搅拌状态下滴加氨水调节pH至9,添加10g氯化锶搅拌2h,过滤、无水乙醇洗涤、60℃烘干,得到改性羟基磷灰石,即组分B;
所述组分C为氢化钙;
将20g组分A、10g组分B、1g组分C充分混合均匀,得到功能辅助成分。
制备例2
茉莉花提取物的制备方法如下:
取新鲜茉莉花在120℃条件下杀青5min,50℃干燥10min,研磨至过100目筛,得茉莉花粉;
按固液比1:10的质量比将50g茉莉花粉分散在500ml水中,在15MPa条件下处理5min,加入茉莉花粉质量0.1%的十二烷基硫酸钠搅拌混合均匀,60℃提取10min,5000rpm离心5min、过滤,取滤液A;
向滤液A中加入无水乙醇至含醇量30%,搅拌均匀后4℃静置1h,过滤,去除沉淀,得滤液B,90℃减压蒸馏,得到茉莉花提取物。
制备例3
忍冬花提取物的制备方法如下:
取新鲜忍冬花在120℃条件下杀青5min,50℃干燥10min,研磨至过100目筛,得忍冬花粉;
按固液比1:10的质量比将50g忍冬花粉分散在水中,在15MPa条件下处理5min,加入忍冬花粉质量0.1%的十二烷基硫酸钠搅拌混合均匀,60℃提取10min,5000rpm离心5min、过滤,取滤液C;
向滤液C中加入无水乙醇至含醇量30%,搅拌均匀后4℃静置1h,过滤,去除沉淀,得滤液D,90℃减压蒸馏,得到忍冬花提取物。
实施例1
一种银耳多糖洁牙护理液制备工艺,按照质量份计算,将乙二胺四乙酸二钠0.1份、十二烷基硫酸钠0.5份、甘草酸二钾0.1份,适量纯化水(补足100份)在85℃乳化、保温20min,降温至60℃,添加C8-16烷基糖苷20份、丙二醇10份、薄荷脑0.1份、对羟基苯甲酸甲酯1份、聚甘油-10月桂酸酯2份、丹皮酚0.1份、柠檬酸0.5份搅拌均匀,降温至50℃,按配比添加西吡氯铵0.1份、功能辅助成分3份搅拌均匀,降温至45℃,按配比添加银耳多糖1份、糖精钠0.5份、茉莉花提取物0.1份搅拌均匀,接着按配比加入氨甲环酸0.5份、忍冬花提取物0.5份搅拌均匀,灌装,得到含银耳多糖的洁牙护理液。

一种银耳多糖洁牙护理液制备工艺,按照质量份计算,将乙二胺四乙酸二钠0.15份、十二烷基硫酸钠1.00份、甘草酸二钾0.15份,适量纯化水(补足100份)在85℃乳化、保温20min,降温至60℃,添加C8-16烷基糖苷35份、丙二醇15份、薄荷脑0.25份、对羟基苯甲酸甲酯1.5份、聚甘油-10月桂酸酯3份、丹皮酚0.15份、柠檬酸1.00份搅拌均匀,降温至50℃,按配比添加西吡氯铵0.015份、功能辅助成分3份搅拌均匀,降温至45℃,按配比添加银耳多糖1.5份、糖精钠1.00份、茉莉花提取物0.25份搅拌均匀,接着按配比加入氨甲环酸1.00份、忍冬花提取物1.00份搅拌均匀,灌装,得到银耳多糖洁牙护理液。

一、牙周炎治疗效果试验
牙周炎动物模型:6-8周龄的雄性C57BL/6J小鼠先适应性喂养一周,第8天进行牙周炎造模。配制7%质量体积的水合氯醛,按0.5mL/100g的剂量腹腔注射麻醉小鼠。置小鼠于固定器,悬吊下颌,暴露口腔,显露上颌后牙,显微外科镊将5-0的慕斯线结扎于小鼠双侧上颌第二磨牙颈部,舌侧打结,饮用15%高糖水,使其形成牙周炎,造模2周后,经牙周炎临床指标检测,筛选出牙龈指数为1.8的小鼠70只,去除丝线后,随机分成10组,其中7组为实验组、2组为对比组,1组为空白组;
实验方法:实验组1-7小鼠适配实施例1-7制备的洁牙护理液(例如试验组1对应实施例1的洁牙护理液,试验组2对应实施例2的洁牙护理液),每日上、下午牙龈局部涂抹洁牙护理液,涂抹厚度为0.1-0.2mm,给药时间为2min,连续给药3周。对比组1、2小鼠适配对比例1、2的牙膏,按照同法涂抹洁牙护理液,空白组按照同法涂抹蒸馏水,在涂抹洁牙护理液前、涂抹洁牙护理液后1周和2周,检测牙龈指数和牙本质敏感程度,结果见表1。
牙龈指数:采用GI分析牙龈颜色和质的改变,评价标准:0=无炎症;1=轻度炎症,探诊无出血,牙龈色泽和质地略有变化;2=中度炎症,探诊出血,牙龈光亮,红肿、增生;3=重度炎症,牙龈有自发性出血,明显发红、增生;
牙本质敏感程度:采用Schiff冷空气敏感指数评估牙本质敏感度程度:0=刺激时不引起疼痛;1=可诱发轻度疼痛或不适;2=可诱发明显疼痛,停止刺激后不引发疼痛;3=可诱发剧烈疼痛,停止刺激后仍疼痛。

从表1中可知,本发明制备的洁牙护理液可以有效的降低牙龈指数,其中效果最好的是实施例1制备的洁牙护理液,一周后牙龈指数降低至0.8,两周后牙龈指数降至0,牙龈指数的降低带来了牙本质敏感程度的降低,相对比对比例和对照组,使用了本发明实施例1-7的洁牙护理液,牙本质敏感程度均产生了明显得降低,效果最好的是实施例1制备的洁牙护理液,一周后牙本质敏感程度降低至1.0,两周后牙龈指数降至0.1。
二、实施例1制备的洁牙护理液临床效果检测
选取2022年10月-12月就诊于某口腔医院的患者50名,牙龈指数在2.0,全身健康状况良好、无重要的全身系统性疾病,年龄在18周岁-60周岁。每人每天早晚各1次,每次2克牙膏,刷牙3分钟。持续使用2周。结果见表2。

从表2中可知,本发明制备的洁牙护理液临床效果非常好,所有的患者均产生了不同程度的改善,94%的患者牙本质敏感程度降低至0-0.5。

复合材料的制备
将碳纳米管加入浓硝酸、浓硫酸混合体系中拌均匀,加热、超声处理,冷却后过滤、洗涤、烘干,得到酸化的碳纳米管,将酸化的碳纳米管分散到无水乙醇中,加入壳聚糖,加热、超声处理,过滤、洗涤、烘干,得到碳纳米管-壳聚糖复合材料;
改性复合材料的制备
使用氯化钙溶液浸渍上述复合材料,搅拌均匀,调节pH至碱性,添加右旋泛酸钙、L-赖氨酸、烟酰胺后搅拌均匀,过滤、洗涤、烘干,得到改性复合材料,即组分A;
所述组分B的制备方法如下:

4.根据权利要求1所述的一种银耳多糖洁牙护理液,其特征在于,所述茉莉花提取物的制备方法如下:
取新鲜茉莉花在120℃条件下杀青5min,50℃干燥10min,研磨至过100目筛,得茉莉花粉;
按固液比1:10的质量比将茉莉花粉分散在水中,在15MPa条件下处理5min,加入茉莉花粉质量0.1%的十二烷基硫酸钠搅拌混合均匀,60℃提取10min,5000rpm离心5min、过滤,取滤液A;
向滤液A中加入无水乙醇至含醇量30%,搅拌均匀后4℃静置1h,过滤,去除沉淀,得滤液B,90℃减压蒸馏,得到茉莉花提取物。
5.根据权利要求1所述的一种银耳多糖洁牙护理液,其特征在于,所述忍冬花提取物的制备方法如下:
取新鲜忍冬花在120℃条件下杀青5min,50℃干燥10min,研磨至过100目筛,得忍冬花粉;
按固液比1:10的质量比将忍冬花粉分散在水中,在15MPa条件下处理5min,加入忍冬花粉质量0.1%的十二烷基硫酸钠搅拌混合均匀,60℃提取10min,5000rpm离心5min、过滤,取滤液C;
向滤液C中加入无水乙醇至含醇量30%,搅拌均匀后4℃静置1h,过滤,去除沉淀,得滤液D,90℃减压蒸馏,得到忍冬花提取物。
6.一种银耳多糖洁牙护理液制备工艺,其特征在于,按照质量份计算,将乙二胺四乙酸二钠、十二烷基硫酸钠、甘草酸二钾,纯化水按照配比在85℃乳化、保温20min,降温至60℃,按配比添加C8-16烷基糖苷、丙二醇、薄荷脑、对羟基苯甲酸甲酯、聚甘油-10月桂酸酯、丹皮酚、柠檬酸搅拌均匀,降温至50℃,按配比添加西吡氯铵、功能辅助成分搅拌均匀,降温至45℃,按配比添加银耳多糖、糖精钠、茉莉花提取物搅拌均匀,接着按配比加入氨甲环酸、忍冬花提取物搅拌均匀,灌装,得到银耳多糖洁牙护理液。
