Abstract
本发明提供一种含有改性壳聚糖可修复牙龈的脱敏牙膏及其制备工艺,将改性壳聚糖作为功效物质加入到牙膏中,在使用时改性壳聚糖能够很好的与牙组织黏附,刺激免疫因子释放,减少牙髓神经刺激,抑制牙石、菌斑的形成,结合赤藓醇使用,能够提高口腔清洁能力,减少牙石、菌斑,起到抑菌消炎、脱敏的效果,最后再通过精氨酸和β‑丙氨酸二乙酸盐营养牙齿,使牙齿再矿化,增加牙齿的耐酸性,提高抗敏作用,减少龋齿,延长牙齿脱敏时间,能够预防或辅助治疗牙周炎引起的牙龈萎缩,还具有一定的防龋齿作用。

本发明涉及口腔护理领域,特别涉及一种含有改性壳聚糖可修复牙龈的脱敏牙膏及其制备工艺。
背景技术
我国约有29.7%的成人有不同程度的牙本质敏感症。牙本质敏感症是指暴露的牙本质因受温度、机械、化学或渗透压等外界刺激而产生的短暂并尖锐的牙齿酸痛症状,具有发作迅速、疼痛尖锐、时间短暂等特点,是一种常见的口腔病症,患病率较高,严重影响患者的生活质量。造成牙本质敏感的原因很多,包括能使釉质完整性受到破坏,牙本质暴露的各种牙体疾病,如磨耗、楔状缺损、牙折、龋病以及牙周萎缩致牙颈部暴露等均可发生牙齿感觉过敏症。
临床上治疗方法众多,包括药物治疗、激光脱敏、电子脱敏、粘接修复治疗、微波治疗、冷冻治疗、紫外线、电凝等。脱敏牙膏因其使用方便、安全、经济实惠,且具有显著的疗效而被患者广泛使用,是治疗牙本质敏感症最为常用的治疗方法。
市场上的脱敏类牙膏都是从两个方面去解决牙本质过敏的问题:一是采用脱敏剂封闭牙本质小管,降低小管内液体流动性以缓解牙本质及牙髓神经末梢敏感度,二是镇静牙髓神经,采用钾盐等物质降低牙髓神经末梢的兴奋性而使疼痛减低,起到减轻牙本质敏感的作用。
中国专利CN102512336A《一种脱敏牙膏及一种脱敏的早晚组合牙膏》,公开了一种脱敏性牙膏,通过采用乙烯基甲醚/马来酸酐共聚物在牙齿表面形成生物膜,通过锶盐脱敏剂封闭牙本质小管和柠檬酸锌营养牙龈、抑制牙菌斑的功效,达到使口腔处于长时间的保护状态的目的。
但由牙周萎缩致牙颈部暴露引起的牙本质敏感,除封闭牙本质小管、降低牙髓神经末梢兴奋性外,还需解决由牙周萎缩引起的其他可导致牙本质敏感的问题,目前专门针对牙龈萎缩问题设计的脱敏牙膏较为少见,因此,需要开发一种针对性脱敏牙膏,可以针对性解决由牙龈萎缩引起牙本质敏感问题。
发明内容
针对上述问题,本发明提出一种含有改性壳聚糖可修复牙龈的脱敏牙膏及其制备工艺。
本发明一种含有改性壳聚糖可修复牙龈的脱敏牙膏,包括以下原料:按质量百分比计,4.2~5%赤藓醇和2.1~4.4%功效提取物;
所述功效提取物包括改性壳聚糖、精氨酸和β-丙氨酸二乙酸盐。
进一步的,所述改性壳聚糖、精氨酸和β-丙氨酸二乙酸盐的质量比为1:(0.62~0.75):(0.49~0.65)。
进一步的,所述改性壳聚糖的制备方法,包括以下步骤:先用10~15wt%氢氧化钙溶液浸渍β-壳聚糖30~40min,在搅拌状态下加入尿囊素泛酸钙,超声处理1~2min,在避光条件下加入多巴胺,于100~110rpm振荡5~10min,取出壳聚糖,水洗,烘干,得改性壳聚糖。
进一步的,氢氧化钙溶液、β-壳聚糖、尿囊素泛酸钙和多巴胺的比例为(20~25)mL:1g:(0.8~1.1)g:(0.4~0.6)g。
进一步的,超声处理的超声功率为180~240W。
进一步的,还包括:0.1~0.3%苯甲酸钠、0.1~0.35%糖精、28~35%山梨醇液、12~18%甘油、1~3%聚乙二醇、0.4~0.7%羧甲基纤维素钠、0.5~0.8%黄原胶、18~31%二氧化硅、0.15~0.25%十二烷基硫酸钠、0.2~0.8%椰油酰胺丙基甜菜碱、1.2~1.5%香精和0.01~0.03%薄荷烷甲酰乙胺。

①将苯甲酸钠、糖精、薄荷烷甲酰乙胺、聚乙二醇、功效提取物和赤藓醇混合,搅拌均匀,得混合物1;
②用甘油溶解羧甲基纤维素钠、黄原胶,得混合物2,在低速搅拌下将混合物2加入到山梨醇液中,搅拌均匀后加入混合物1,继续搅拌均匀,加入二氧化硅,高速搅拌35~45min,得混合物3;
③向混合物3中加入十二烷基硫酸钠、椰油酰胺丙基甜菜碱,低速搅拌10~18min,加入香精搅拌均匀,静置5~10h,得膏体;
④研磨膏体1~2遍后,在真空度≤-0.092Mpa的条件下,搅拌膏体12~15min进行脱气处理,脱气结束后灌膏包装。
进一步的,所述低速搅拌的速度为60~80r/min;所述高速搅拌的速度为3000~6000r/min。
进一步的,所述脱敏牙膏的pH为7.50~7.94,稠度为17~19mm,粘度为260~280Pa.s,泡沫量为50~60mm。
与现有技术相比,本发明的有益效果是:
本申请将改性壳聚糖作为功效物质加入到牙膏中,在使用时改性壳聚糖能够很好的与牙组织黏附,刺激免疫因子释放,减少牙髓神经刺激,抑制牙石、菌斑的形成,结合赤藓醇使用,能够提高口腔清洁能力,减少牙石、菌斑,起到抑菌消炎、脱敏的效果,最后再通过精氨酸和β-丙氨酸二乙酸盐营养牙齿,使牙齿再矿化,增加牙齿的耐酸性,提高抗敏作用,减少龋齿,延长牙齿脱敏时间,能够预防或辅助治疗牙周炎引起的牙龈萎缩,还具有一定的防龋齿作用。
本发明利用多巴胺和尿囊素泛酸钙经过特殊处理改性壳聚糖,加速牙周支持组织重建,减轻牙槽骨吸收情况,抗敏坚固牙齿,缓解牙本质过敏性酸痛,增强牙膏的脱敏效果,此外,壳聚糖还具有良好的稀释保湿性,和山梨醇、甘油复配使用能够增强保湿作用,使牙膏具有良好的使用性。

为了更好理解本发明技术内容,下面提供具体实施例,对本发明做进一步的说明。
本发明使用的山梨醇液为75%(v/v)的山梨醇液。
实施例1
一种含有改性壳聚糖可修复牙龈的脱敏牙膏,包括以下原料:按质量百分比计,4.8%赤藓醇、3.6%功效提取物、0.1%苯甲酸钠、0.15%糖精、34%山梨醇液、16%甘油、2%聚乙二醇、0.6%羧甲基纤维素钠、0.6%黄原胶、28%二氧化硅、0.18%十二烷基硫酸钠、0.6%椰油酰胺丙基甜菜碱、1.3%香精、0.02%薄荷烷甲酰乙胺、余量水;
功效提取物的改性壳聚糖、精氨酸和β-丙氨酸二乙酸盐的质量百分比分别为1.6%,1.16%,0.84%;
改性壳聚糖的制备方法,包括以下步骤:先用230mL 10wt%氢氧化钙溶液浸渍10gβ-壳聚糖35min,在搅拌状态下加入10g尿囊素泛酸钙,超声处理1min,超声功率为200W,在避光条件下加入5g多巴胺,于110rpm振荡8min,取出壳聚糖,水洗,烘干,得改性壳聚糖。

功效提取物的改性壳聚糖、精氨酸和β-丙氨酸二乙酸盐的质量百分比分别为1.83%,1.38%,1.19%;
改性壳聚糖的制备方法,包括以下步骤:先用250mL 12wt%氢氧化钙溶液浸渍10gβ-壳聚糖40min,在搅拌状态下加入11g尿囊素泛酸钙,超声处理2min,超声功率为180W,在避光条件下加入6g多巴胺,于110rpm振荡5min,取出壳聚糖,水洗,烘干,得改性壳聚糖。

一种含有改性壳聚糖可修复牙龈的脱敏牙膏,包括以下原料:按质量百分比计,6.5%赤藓醇、1.6%功效提取物、0.1%苯甲酸钠、0.15%糖精、34%山梨醇液、16%甘油、2%聚乙二醇、0.6%羧甲基纤维素钠、0.6%黄原胶、28%二氧化硅、0.18%十二烷基硫酸钠、0.6%椰油酰胺丙基甜菜碱、1.3%香精、0.02%薄荷烷甲酰乙胺、余量水;
功效提取物的改性壳聚糖、精氨酸和β-丙氨酸二乙酸盐的质量百分比分别为0.715%,0.515%,0.37%;
改性壳聚糖的制备方法,包括以下步骤:先用230mL 10wt%氢氧化钙溶液浸渍10gβ-壳聚糖35min,在搅拌状态下加入10g尿囊素泛酸钙,超声处理1min,超声功率为200W,在避光条件下加入5g多巴胺,于110rpm振荡8min,取出壳聚糖,水洗,烘干,得改性壳聚糖。
实施例1-5和对比例1-2一种含有改性壳聚糖可修复牙龈的脱敏牙膏的制备工艺,包括以下步骤:
①用水将苯甲酸钠、糖精、薄荷烷甲酰乙胺、聚乙二醇、功效提取物和赤藓醇混合,搅拌均匀,得混合物1;
②用甘油溶解羧甲基纤维素钠、黄原胶,得混合物2,在低速搅拌下将混合物2加入到山梨醇液中,低速搅拌的速度为60r/min,搅拌均匀后加入混合物1,继续搅拌均匀,加入二氧化硅,高速搅拌45min,高速搅拌的速度为6000r/min,得混合物3;
③向混合物3中加入十二烷基硫酸钠、椰油酰胺丙基甜菜碱,低速搅拌10~18min,低速搅拌的速度为80r/min,加入香精搅拌均匀,静置10h,得膏体;
④研磨膏体2遍后,在真空度≤-0.092Mpa的条件下,搅拌膏体15min进行脱气处理,脱气结束后灌膏包装。

安全性试验
1.急性经口毒性试验
参照《化妆品安全技术规范》(2015版)急性经口毒性试验进行;
实验动物:选用体重150±10g的成年、健康大鼠80只,雌雄各半,雌性大鼠为未孕和未曾产仔的,试验前在需实验动物房环境中适应5d时间,实验动物房的环境为20~26℃,相对湿度40~70%,选用标准配合饲料喂养,饮水自由。
受试物:实施例1~5和对比例1~3的牙膏,将受试物用水配制成溶液;
试验方法:试验前,实验动物禁食过夜,不限制饮水,称量动物体重,随机分成8组,然后对各组动物用管饲法,在24h内分2次进行染毒,染毒总剂量为5000mg/kg,染毒后继续禁食4h,染毒后定时观察大鼠中毒表现及死亡情况,每天进行一次仔细的检查,记录是否有被毛和皮肤、眼睛和粘膜,呼吸、循环、自主神经和中枢神经系统、肢体活动和行为等的改变,观察10d,观察期内每周称重一次,观察期结束称重一次,处死后进行尸检。
试验结果,在观察期内,未发现大鼠被毛和皮肤、眼睛和粘膜的改变,同时观察到大鼠生命活动规律正常,试验前后体重也无异常变化,处死解剖未发现病理改变。
试验结果表明,本发明的牙膏对大鼠无经口毒性。
2.口腔黏膜刺激试验
参照YY/T0127.13-2018《口腔医疗器械生物学评价第13部分:口腔黏膜刺激试验》进行试验;
实验动物:选用体重70±5g、成年、健康的金黄色地鼠24只,雌雄各半,雌性金黄色地鼠应无孕;
受试物:实施例1~5和对比例1~3的牙膏;
试验方法:在试验前,麻醉动物,使用消毒剂清洁动物口腔内外组织,将左侧口腔颊囊设为试验区,右侧口腔颊囊为对照区;每日早上8点,晚上10点使用受试物涂抹试验区,每次涂抹剂量为0.5mg/cm2,停留3min后清洗口腔,重复7d,对照区按同法给予蒸馏水,每次涂抹前后检查颊囊,末次接触后24h观察颊囊,并无痛处死动物,取口腔颊囊进行观察。试验期间观察记录动物有无异常表现。
试验结果,在试验期间没有观察到动物的异常表现,试验前后颊囊无充血、肿胀、溃疡等情况,试验颊囊黏膜和对照黏膜无明显差异。
试验证明,本发明的牙膏对金黄色地鼠无口腔黏膜刺激性。
使用效果
1.牙周炎治疗效果试验
牙周炎动物模型:选择8周龄,体重210±10g的大鼠,用3.0丝线于大鼠实验牙牙颈部进行结扎,饮用10%高糖水,使其形成牙周炎,造模4周后,经牙周炎临床指标检测,筛选出符合指标的大鼠45只,去除丝线后,随机分成9组,其中8组为实验组、1组为模型对照组;
受试物:实施例1~5和对比例1~3制备所得牙膏;
实验方法:实验组大鼠每日上、下午牙龈局部涂抹给药,涂抹厚度为0.1~0.2mm,给药时间为3min,连续给药4周。对照模型组按照同法给予蒸馏水,在给药前、给药2周和4周,检测菌斑指数(PLI)、牙石指数(CI)、牙龈指数(GI)和牙本质敏感程度,结果见表1。
菌斑指数(plaque index,PLI):采用Silness和Loe所提出的标准进行评分,0=龈缘区无菌斑,1=龈缘区的牙面有薄的菌斑,仅用探针才能刮及;2=在龈缘或邻面可见中等量菌斑;3=龈沟内或龈缘区及邻面有大量软垢;
牙石指数(calculus Index,CI):采用CI评估牙石的沉积情况:0=龈上、龈下无牙石;1=龈上牙石覆盖牙面不超过1/3;2=龈上牙石覆盖牙面1/3-1/2或有散在龈下牙石;3=龈上牙石超过牙面2/3或有连续而厚的龈下牙石;
牙龈指数(gingival index,GI):采用GI分析牙龈颜色和质的改变,评价标准:0=无炎症;1=轻度炎症,探诊无出血,牙龈色泽和质地略有变化;2=中度炎症,探诊出血,牙龈光亮,红肿、增生;3=重度炎症,牙龈有自发性出血,明显发红、增生;
牙本质敏感程度:采用Schiff冷空气敏感指数评估牙本质敏感度程度:0=刺激时不引起疼痛;1=可诱发轻度疼痛或不适;2=可诱发明显疼痛,停止刺激后不引发疼痛;3=可诱发剧烈疼痛,停止刺激后仍疼痛。

实验结果表明,与对照模型组相比,本发明制备的脱敏牙膏可有效去除牙石菌斑,减轻牙周炎症状,还可通过改善牙周炎症状来减缓牙本质敏感程度,说明本发明的牙膏可针对性解决由牙龈萎缩引起的牙本质敏感问题。
与实施例1-3相比,对比例1功效提取物的成分与本发明不同,牙龈指数较高,牙本质敏感程度的改善不明显;对比例2赤藓醇与功效提取物的用量比例不同,各项指数的改善程度均低于实施例;对比例3的脱敏牙膏未使用赤藓醇,牙石、菌斑和牙龈指数的变化不明显,牙本质敏感程度的改善程度低。
2.防龋齿效果试验:
受试物:实施例1~5和对比例1制备所得牙膏;
试验前准备:选取表面光洁的牛切牙70颗,将每颗牛切牙切割成4mm×4mm的牙釉质块,抛光后用去离子水冲洗后,随机分成7组,其中6组为实验组,1组为空白对照组,分别测量牙釉质表面显微硬度(Surface Microhardness,SMH)后,进行试验。
试验方法:取实验组的牛切牙进行酸蚀处理,将酸蚀后的牙釉质放入牙膏浆中,牙膏浆要完全没过牙釉质,浸泡3分钟后,冲洗干净,放入人工唾液中浸泡10~12小时,再用牙膏浆浸泡3分钟,清洗干净后放入人工唾液浸泡过夜,再放入牙膏浆中浸泡3分钟,冲洗干净,放入人工唾液中浸泡10~12小时,再用牙膏浆浸泡3分钟,清洗干净后放入人工唾液浸泡过夜,循环处理;空白对照组不使用牙膏浆浸泡,其余操作同实验组。分别在酸蚀处理开始第0,2,6,8,10天测量SMH,结果见表2。
表2防龋齿效果
由实验数据可知,使用本发明的脱敏牙膏在酸蚀作用下,可明显降低牙釉质硬度损失率,还可提高牙釉质表面显微硬度,表明本发明的牙膏可通过促进牙釉质再矿化,减轻酸蚀效果,说明本发明的脱敏牙膏还可以防治龋齿。与实施例1相比,对比例1功效提取物的成分与本发明不同,酸蚀效果明显,防治龋齿效果差。
以上所述仅为本发明的较佳实施例而已,并不用以限制本发明,凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (7)
1.一种含有改性壳聚糖可修复牙龈的脱敏牙膏,其特征在于,包括以下原料,按质量百分比计,4.2~5%赤藓醇和2.1~4.4%功效提取物;
所述功效提取物包括改性壳聚糖、精氨酸和β-丙氨酸二乙酸盐;所述改性壳聚糖、精氨酸和β-丙氨酸二乙酸盐的质量比为1:(0.62~0.75):(0.49~0.65);
所述改性壳聚糖的制备方法,包括以下步骤:先用10~15wt%氢氧化钙溶液浸渍β-壳聚糖30~40min,在搅拌状态下加入尿囊素泛酸钙,超声处理1~2min,在避光条件下加入多巴胺,于100~110rpm振荡5~10min,取出壳聚糖,水洗,烘干,得改性壳聚糖;氢氧化钙溶液、β-壳聚糖、尿囊素泛酸钙和多巴胺的比例为(20~25)mL:1g:(0.8~1.1)g:(0.4~0.6)g。
2.如权利要求1所述的一种含有改性壳聚糖可修复牙龈的脱敏牙膏,其特征在于,所述超声处理的超声功率为180~240W。
3.如权利要求1所述的一种含有改性壳聚糖可修复牙龈的脱敏牙膏,其特征在于,还包括:0.1~0.3%苯甲酸钠、0.1~0.35%糖精、28~35%山梨醇液、12~18%甘油、1~3%聚乙二醇、0.4~0.7%羧甲基纤维素钠、0.5~0.8%黄原胶、18~31%二氧化硅、0.15~0.25%十二烷基硫酸钠、0.2~0.8%椰油酰胺丙基甜菜碱、1.2~1.5%香精和0.01~0.03%薄荷烷甲酰乙胺。
4.如权利要求3所述的一种含有改性壳聚糖可修复牙龈的脱敏牙膏,其特征在于,所述山梨醇液为70~75%v/v的山梨醇液。
5.如权利要求3所述的一种含有改性壳聚糖可修复牙龈的脱敏牙膏的制备工艺,其特征在于,包括以下步骤:
①将苯甲酸钠、糖精、薄荷烷甲酰乙胺、聚乙二醇、功效提取物和赤藓醇混合,搅拌均匀,得混合物1;
②用甘油溶解羧甲基纤维素钠、黄原胶,得混合物2,在低速搅拌下将混合物2加入到山梨醇液中,搅拌均匀后加入混合物1,继续搅拌均匀,加入二氧化硅,高速搅拌35~45min,得混合物3;
③向混合物3中加入十二烷基硫酸钠、椰油酰胺丙基甜菜碱,低速搅拌10~18min,加入香精搅拌均匀,静置5~10h,得膏体;
④研磨膏体1~2遍后,在真空度≤-0.092Mpa的条件下,搅拌膏体12~15min进行脱气处理,脱气结束后灌膏包装。
6.如权利要求5所述的一种含有改性壳聚糖可修复牙龈的脱敏牙膏的制备工艺,其特征在于,所述低速搅拌的速度为60~80r/min;所述高速搅拌的速度为3000~6000r/min。
7.如权利要求1~4任一项所述的一种含有改性壳聚糖可修复牙龈的脱敏牙膏,其特征在于,所述脱敏牙膏的p H为7.50~7.94,稠度为17~19mm,粘度为260~280Pa.s,泡沫量为50~60mm。
