Abstract
The invention discloses a permeation-promoting composition containing arginine/lysine polypeptide and application thereof. The permeation promoting composition containing arginine/lysine polypeptide comprises 15-25% of arginine/lysine polypeptide solution, 30-50% of ethoxydiglycol and 30-50% of multi-element plant fermentation solution by weight percent; the multi-element plant ferment solution is obtained by fermenting lucid ganoderma, dwarf lilyturf tuber and saccharomycetes together and ultrasonically extracting the filtrate. The preparation method comprises adding arginine/lysine polypeptide solution and ethoxydiglycol into a container, heating to 30-50deg.C, and stirring; adding the multi-element plant fermentation solution, stirring, and cooling to obtain the composition. It can be used in cosmetic or personal care products. The invention can promote permeation promotion effect from multiple dimensions, increase transdermal efficiency of cosmetics, promote permeation, facilitate absorption of effective components in cosmetics, and better improve skin state.
Description
Technical Field
The invention relates to the field of cosmetics, in particular to a penetration-promoting composition capable of improving the absorptivity of cosmetics, and a preparation method and application thereof.
Background
Albert Kligman MD ph.d. dermatologists clearly propose that the functional cosmetic must possess: "whether there is a sufficient concentration to penetrate the stratum corneum to the target site within a given time", i.e., whether the cosmetic is able to penetrate the stratum corneum. Transdermal absorption of cosmetics is particularly important, and is a key point affecting whether cosmetics are effective. The more the amount of the functional component absorbed by the skin, the more remarkable the skin nourishing effect. Transdermal absorption of cosmetics on the skin mainly has several pathways: 1. dissolution and diffusion. The dissolution of cosmetic ingredients in the stratum corneum is absorbed by the skin through cutaneous arterial pathways, stratum corneum transport (including intracellular diffusion, interstitial diffusion) and deep epidermis and dermis transport, and thus into the body tissue. 2. Hydration. When the skin absorbs a lot of moisture and is sufficiently hydrated, the stratum corneum can expand to a porous state, and the barrier function is lowered, thereby facilitating penetration of substances. 3. Osmotic pressure effects. The greater the osmotic pressure outside the skin, the greater the osmotic capacity. 4. The penetration enhancer acts. Reversibly reduces the barrier function of the stratum corneum of the skin, making it easier for the cosmetic ingredients to be absorbed transdermally to reach the circulatory system. 5. The substances at the skin acupoints are absorbed. Compared with other skin, the skin impedance at the acupoint is low, the capacitance is large, the potential is high, and the transdermal absorption of substances is facilitated. Substances with low charge are easily absorbed transdermally. In cosmetic manufacture, penetration of the active ingredients of the cosmetic into the skin is generally aided by the addition of penetration enhancers.
Penetration enhancers commonly used in the cosmetic art are of a wide variety, such as moisturizers, polyols, lipids and transdermal peptides. Wherein the humectant comprises amino acids and hydrolyzed glycosaminoglycan; polyols include ethoxydiglycol, glycerol, propylene glycol, 1, 3-butanediol and pentanediol; the lipid comprises ceramide, cholesterol and lecithin; transdermal peptides include transdermal peptide TD-1, skin and cell penetrating peptide SPACE.
However, the components of the conventional penetration enhancers in actual production of real cosmetics are relatively single, one or a class of penetration enhancers are usually used, the penetration enhancing effect of the product cannot be increased from multiple dimensions, and the penetration enhancing effect is not good enough. In addition, the prior penetration enhancer is not concerned about the absorption and utilization rate of skin to effective components in cosmetics, but is required to have good transdermal effect for the cosmetics, and the absorption effect of the skin to the effective components is considered, so that the penetration enhancer and the absorption have influence on the final effect of improving the skin, and the penetration enhancer and the absorption are essential.
Therefore, research on new penetration-promoting technology is urgently needed in the field, and the transdermal and absorption effects of cosmetics are greatly improved through multiple dimensions.
Disclosure of Invention
The object of the present invention is to provide a permeation enhancing composition comprising an arginine/lysine polypeptide. The invention can promote the permeation promotion effect from multiple dimensions, and greatly promote the transdermal and absorption effects of the cosmetics.
The technical scheme of the invention is as follows: in a first aspect of the invention there is provided a permeation enhancing composition comprising an arginine/lysine polypeptide, the composition comprising the components:
a arginine/lysine polypeptide solution; b ethoxydiglycol; c a multi-element plant ferment solution.
Preferably, the multi-element plant fermentation solution comprises ganoderma lucidum extract, ophiopogon root extract and fermentation filtrate of the saccharomyces cerevisiae.
Preferably, the mass ratio of the ganoderma lucidum extract to the ophiopogon root extract to the fermentation filtrate of the saccharomyces cerevisiae in the multi-element plant fermentation solution is as follows: 1:2:88.
Preferably, the composition comprises: 15-25% of component a, 30-50% of component b and 30-50% of component c.
Preferably, the composition comprises: component a20% and component b40% and component c 40%.
In a second aspect of the invention, there is provided a method of preparing a permeation enhancing composition comprising an arginine/lysine polypeptide, the method comprising the steps of: A. adding arginine/lysine polypeptide solution and ethoxydiglycol into a container, heating to 30-50deg.C, and stirring at 10-30r/min for 3-8min to obtain A1 product;
B. adding the multi-element plant fermentation solution into the A1 product, stirring at a speed of 10-30r/min for 3-8min, and cooling to obtain the composition of claim 1.
In a third aspect of the invention there is provided the use of said permeation enhancing composition comprising an arginine/lysine polypeptide as a cosmetic or personal care product; or in the preparation of cosmetics or personal care products.
Preferably, the personal care product comprises cream, emulsion, essence, toning lotion, gel, facial mask, eye essence and eye mask.
In a fourth aspect of the invention, a cosmetic or personal care product is provided comprising the penetration enhancing composition comprising an arginine/lysine polypeptide and a cosmetically acceptable carrier.
In a fifth aspect of the present invention, there is provided a method of preparing the cosmetic or personal care product, the method comprising the steps of: the cosmetic or personal care product is obtained by mixing the arginine/lysine polypeptide-containing penetration-promoting composition with an emulsified or non-emulsified cosmetically acceptable carrier.
The invention has the main advantages that:
1. the arginine/lysine polypeptide solution of the invention can be synergistically enhanced in the aspect of cosmetic penetration promotion due to the unique 3D structure and the hydrophilic and lipophilic amphoteric structure of the ethoxydiglycol.
2. The multi-element plant ferment solution (ganoderma lucidum extract, dwarf lilyturf tuber extract and yeast bifidus ferment filtrate) contains multiple compound amino acid small molecules, so that the skin hydration is improved and the cosmetic permeation is promoted; the combination of the multielement plant fermentation product solution and the arginine/lysine polypeptide solution has a synergistic effect, improves the permeation effect of each time period of the test, and has very remarkable effect.
3. The invention provides a method for synergistically promoting the penetration and absorption of cosmetics by skin from three dimensions of dissolution and diffusion, hydration and penetration enhancer.
Therefore, the invention has better permeation promotion effect by improving the permeation promotion effect from multiple dimensions, is beneficial to the absorption of effective components in cosmetics while promoting permeation, and can more effectively improve the skin state.

The specific embodiment is as follows:
cosmetics can improve skin condition only by being transdermally and absorbed by skin. Three pathways of cosmetic transdermal absorption: 1. dissolution and diffusion 2, hydration 3, and penetration enhancer. The inventor tries a plurality of substances based on the transdermal absorption path, encounters great obstacle, and finally unexpectedly discovers a combination, namely, three paths of precise synergistic transdermal absorption can be realized by combining arginine/lysine polypeptide solution, ethoxydiglycol, ganoderma lucidum extract, ophiopogon root extract and fermentation filtrate of the saccharomyces cerevisiae: 1. the dissolution and diffusion effects 2, the hydration effects 3 and the penetration enhancer effects, the penetration enhancer effect is improved in a multi-dimensional way, a better penetration enhancing effect is presented, and the absorption of effective components in the cosmetics is facilitated while the penetration is enhanced. On this basis, the present invention has been completed.
As used in the present invention, the arginine/lysine polypeptide solution is produced by ACTIVEN, switzerland, model XEP-018 in particular, is a 23-residue-containing peptide having three disulfide bridges, which is prepared by solid phase peptide synthesis-purification of a primary linear peptide-folding: cysteine residue pairing-purification of folded peptide-arginine/lysine polypeptide powder-dilution with purified water to a concentration of 0.0024% produced an arginine/lysine polypeptide solution.
As used herein, ethoxydiglycol is produced by gattaefosse, france, and is specifically model Transcutol CG.
As used in the invention, the multi-element plant ferment solution is produced by the biological technology institute of the department of Chinese patent Hua Qi (Beijing), and the specific model is winter enzyme, and consists of ganoderma lucidum extract, ophiopogon root extract and fermentation filtrate of the saccharomyces cerevisiae. Fermenting Ganoderma, radix Ophiopogonis and yeast for 7 days by solid state fermentation and ultrasonic extraction, fermenting at 28-35 deg.C, adding purified water, ultrasonic extracting, centrifuging, filtering, and adding antiseptic.
As used herein, "composition" refers to a penetration-promoting composition that, when applied to a subject (generally referred to as a human), is capable of promoting the transdermal and absorptive effects of a cosmetic product.
Herein, "cosmetically acceptable carrier" refers to a carrier that makes the cosmetic or personal care product applicable, including various excipients. They are not per se essential active ingredients and are not particularly toxic after administration. Suitable carriers are familiar to the person skilled in the cosmetic arts. A sufficient discussion of cosmetically acceptable excipients can be found in cosmetic hygiene specifications 2015. Such carriers in such compositions may include: moisturizers, emollients, thickeners, and the like. Such as but not limited to propylene glycol, butylene glycol, water, glycerin, xanthan gum, and the like.
The term "individual" or "personal" is used herein to refer to a person who may receive the composition or method for application to the skin.
The inventors have found that synergistic effects in promoting penetration can be produced if an arginine/lysine polypeptide solution is combined with ethoxydiglycol and a multi-plant ferment solution. Generally, the arginine/lysine polypeptide solution and the ethoxydiglycol are mixed with the multielement plant fermentation solution according to the mass ratio: 1:2:2.
In one embodiment of the invention, the arginine/lysine polypeptide solution and ethoxydiglycol are heated to 30-50 ℃ and stirred at a speed of 10-30r/min for 3-8min; then adding the multi-element plant fermentation solution, stirring for 3-8min at the speed of 10-30r/min, and cooling to obtain the composition provided by the invention.
In one embodiment of the present invention, the present invention provides a composition comprising, based on total weight of the composition:
15% -25% arginine/lysine polypeptide solution
30-50% ethoxydiglycol
0.3% -0.5% of ganoderma lucidum extract
0.6% -1% of ophiopogon root extract
24% -44% of fermentation filtrate of two-split yeast
2.7% -4.5% of an alcohol solvent
The rest of preservative is p-hydroxyacetophenone.
The alcohol solvent forming the above solution may be, but is not limited to, one or two or more of the following: glycerol, 1, 2-pentanediol, butanediol, propylene glycol, and the like.
Further, the compositions provided herein are mixed with a cosmetically acceptable carrier to produce a variety of cosmetic or personal care products that can be applied to human skin, including, but not limited to, lotions, essences, emulsions, creams, facial masks, eye essences, eye masks, neck masks, and the like.
In one embodiment of the invention, the aqueous phase formed by the other cosmetically acceptable carrier is added to the oil phase formed by the other cosmetically acceptable carrier, and the composition provided by the invention is added after emulsification to obtain the cosmetic or personal care product which can be applied to human skin.
In some embodiments of the present invention, the cosmetically acceptable carrier used to form the present invention includes, but is not limited to, one, but two or more of propylene glycol, butylene glycol, water, glycerin, xanthan gum.
Unless defined otherwise herein, the meanings of scientific and technical terms used herein are the same as commonly understood and used by one of ordinary skill in the art.

The units in the percentages mentioned by weight in the present invention are well known to the person skilled in the art. Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art. In addition, any methods and materials similar or equivalent to those described herein can be used in the methods of the present invention. The preferred methods and materials described herein are presented for illustrative purposes only.
Example 1. The penetration-promoting composition containing arginine/lysine polypeptide comprises 15% of arginine/lysine polypeptide, 35% of ethoxydiglycol and 50% of a multi-element plant fermentation solution; the commodity name of the multi-element plant ferment solution is winter enzyme.
The arginine/lysine polypeptide used in the examples of the present invention was identified by the trade name XEP-018 and the ethoxydiglycol used was identified by the trade name Transcutol CG.
A method for preparing a permeation enhancing composition comprising an arginine/lysine polypeptide, comprising the steps of,
A. adding arginine/lysine polypeptide and ethoxydiglycol into a container, heating to 30deg.C, and stirring at 30r/min for 3min to obtain product A1;
B. adding the multi-element plant fermentation product solution into the A1 product, stirring for 3min at the speed of 30r/min, and cooling to obtain the composition.
Example 2. The permeation promoting composition containing arginine/lysine polypeptide comprises 25% of arginine/lysine polypeptide, 45% of ethoxydiglycol and 30% of a multi-element plant fermentation solution in percentage by weight.
A method for preparing a permeation enhancing composition comprising an arginine/lysine polypeptide, comprising the steps of,
A. adding arginine/lysine polypeptide and ethoxydiglycol into a container, heating to 50 ℃, and stirring at a speed of 10r/min for 8min to obtain an A1 product;
B. adding the multi-element plant fermentation product solution into the A1 product, stirring at the speed of 10r/min for 8min, and cooling to obtain the composition.
Example 3. The permeation promoting composition containing arginine/lysine polypeptide comprises, by weight, 20% of arginine/lysine polypeptide, 40% of ethoxydiglycol, and 40% of a multi-element plant fermentation solution.
A method for preparing a permeation enhancing composition comprising an arginine/lysine polypeptide, comprising the steps of,
A. adding arginine/lysine polypeptide and ethoxydiglycol into a container, heating to 40 ℃, and stirring at 15r/min for 5min to obtain A1 product;
B. adding the multi-element plant fermentation product solution into the A1 product, stirring for 5min at the speed of 15r/min, and cooling to obtain the composition.
Application example 1. 1g of nicotinamide, 3g of glycerol, 5g of butanediol and 0.2g of carbomer are added into 82g of deionized water, heated to 60 ℃, homogenized for 3 minutes, cooled to 40 ℃,1.5g of acetyl hexapeptide-81 g of the composition provided by the invention and a proper amount of preservative and essence are added, and uniformly stirred; adding proper amount of arginine water solution, and homogenizing for 2 min.
Application example 2. Preparation of emulsion, 2g of polydimethylsiloxane, 2g of isononyl isononanoate, 2g of octyl polymethylsiloxane and 2g of glyceryl stearate citrate are heated to be dissolved (oil phase), 6g of glycerin, 6g of butanediol, 1g of nicotinamide, 0.3g of carbomer and 0.02g of EDTA-2NA are added, 68g of deionized water is added to be heated to 60 degrees (water phase), the oil phase is slowly added to the water phase while being homogenized, the homogenization is carried out for 5 minutes, the temperature is reduced to 40 degrees, 2.5g of the purslane extract, a proper amount of arginine water solution, preservative and essence are added to the composition provided by the invention, and the composition is stirred uniformly.
Application example 3. Preparation of essence, 1g of squalane, 2g of polydimethylsiloxane, 3g of isononyl isononanoate, 1.5g of cetostearyl-oleyl oleate and sorbitan-oleyl oleate complex are heated to be dissolved (oil phase), 2g of glycerin, 4g of butanediol, 1g of nicotinamide, 0.2g of carbomer, 0.02g of EDTA-2NA, 65g of deionized water are added to be heated to 60 degrees (water phase), the oil phase is slowly added to the water phase while being homogenized, the mixture is homogenized for 3 minutes, the temperature is reduced to 40 degrees, 8g of the composition provided by the invention, 2g of acetyl hexapeptide-8, 2g of palmitoyl tripeptide-5 and a proper amount of arginine water solution are added, and the mixture is stirred uniformly.
Application example 4. Preparation of eye essence, 1g of PEG-40 stearate, 1.5g of glycerol stearate and PEG-100 stearate complex, 2g of polydimethylsiloxane, 1g of isononyl isononanoate are heated to be dissolved (oil phase), 2g of glycerol, 1g of butanediol, 1g of nicotinamide, 0.3g of carbomer and 0.02g of EDTA-2NA are added, 60g of deionized water is added to be heated to 60 degrees (water phase), an oil phase is slowly added to the water phase while being homogenized, the mixture is homogenized for 3 minutes, the temperature is reduced to 40 degrees, 10g of composition provided by the invention, 3g of palmitoyl tripeptide-5, 3g of spilanthol extract are added, and a proper amount of arginine water solution, preservative and essence are added and stirred uniformly.
Application example 5. The preparation of the facial mask comprises the steps of adding 2g of trehalose, 4g of glycerol, 2g of butanediol and 0.08g of xanthan gum into 85g of deionized water, heating to 60 ℃, homogenizing for 3 minutes, cooling to 40 ℃, adding 1.5g of the composition provided by the invention, 1g of the purslane extract, a proper amount of preservative and essence, and uniformly stirring.

From the examples, the present invention has excellent ability to promote percutaneous absorption of ascorbyl glucoside (AA 2G), and the permeation promoting effect is better than 1% azone. The penetration promoting effect of the composition is better than that of a control group 1+a control group 2+a control group 3. The invention can promote the absorption of effective components in the cosmetics and has the effect of adding the cosmetics due to the increase of the skin hydration capability at the same time of promoting the permeation.

Claims (10)
1. An arginine/lysine polypeptide-containing permeation enhancing composition, comprising the following components: a arginine/lysine polypeptide solution; b ethoxydiglycol; c a multi-element plant ferment solution.
2. The arginine/lysine polypeptide-containing permeation promoting composition according to claim 1, wherein the multi-component plant fermentation solution comprises ganoderma lucidum extract, ophiopogon root extract, and fermentation filtrate of saccharomyces cerevisiae.
3. The arginine/lysine polypeptide-containing permeation promoting composition according to claim 1, wherein the mass ratio of the ganoderma lucidum extract to the ophiopogon root extract to the fermentation filtrate of the saccharomyces cerevisiae in the multi-plant fermentation solution is: 1:2:88.
4. The arginine/lysine polypeptide-containing permeation promoting composition according to claim 1, wherein the composition comprises 15% to 25% of component a and 30% to 50% of component b and 30% to 50% of component c.
5. The arginine/lysine-containing polypeptide permeation promoting composition according to claim 1, wherein the composition comprises 20% of component a and 40% of component b and 40% of component c by mass: 1:2:2.
6. A method for preparing a permeation enhancing composition comprising an arginine/lysine polypeptide according to any one of claims 1 to 5, wherein the method comprises the steps of:
A. adding arginine/lysine polypeptide solution and ethoxydiglycol into a container, heating to 30-50deg.C, and stirring at 10-30r/min for 3-8min to obtain A1 product;
B. adding the multi-element plant fermentation product solution into the A1 product, stirring at the speed of 10-30r/min for 3-8min, and cooling to obtain the final product.
7. Use of the arginine/lysine-containing polypeptide-based penetration enhancing composition according to any one of claims 1 to 5 in cosmetics or personal care products.
8. The use according to claim 7, wherein the personal wash comprises creams, lotions, essences, lotions, gels, facial masks, eye essences, eye patches.
9. A cosmetic comprising the arginine/lysine polypeptide-containing penetration-promoting composition of any one of claims 1 to 5 and a cosmetically acceptable carrier.
10. A method of preparing the cosmetic according to claim 9, comprising the steps of: mixing with penetration promoting agent of the arginine/lysine-containing polypeptide using emulsifying or non-emulsifying cosmetically acceptable carrier to obtain the final product.
