Abstract
本发明公开了一种鸡枞菌多糖及其制备方法和应用。本发明将鸡枞菌脱脂,用提取剂,醇沉透析后得到的鸡枞菌多糖。提取剂为水、酸溶液或碱溶液,分别得到水提多糖、鸡枞菌酸提多糖和鸡枞菌碱提多糖,所述鸡枞菌多糖升高肠道有益菌的丰度,降低肠道有害菌的丰度,改善肠道菌群失调,减少稀便、腹泻、便血、结肠长度减少、肿胀和/或出血的情况,降低了结肠炎症细胞浸润、紧密连接蛋白表达水平和促炎症细胞因子表达水平,增加了结肠肠黏膜粘蛋白和短链脂肪酸的表达水平,对结肠炎症以及损伤等起到缓解作用,维持肠道稳态。

技术领域
本发明涉及生物医药技术领域,具体地,涉及一种鸡枞菌多糖及其制备方法和应用。
背景技术
炎症性肠病(Inflammatory bowel disease,IBD)是一种慢性特发性肠道炎症性疾病。根据病变部位及严重程度,IBD一般归类为溃疡性结肠炎(Ulcerative colitis,UC)、克罗恩病(Crohn disease,CD)以及未分型IBD。IBD患者临床表现常为腹痛、腹泻、直肠出血、粘液脓血便、体重下降、贫血等,其中以腹泻为主要症状。肠道菌群的异常变化或生态失调会增强肠道炎症反应,从而导致IBD的发展。因此,通过改善肠道微生物群的结构来治疗IBD已成为潜在的辅助疗法之一。
食用菌多糖对IBD中的肠道菌群失调具有一定的改善作用。鸡枞菌是我国传统的食药用真菌之一,鸡枞菌中有许多生物活性成分,其中多糖是鸡枞菌最重要的生物活性成分之一。目前鸡枞菌多糖(Termitomyces Polysaccharides,TPS)的提取方法主要包括热水提取、碱提取、酸提取、超声波辅助提取和酶辅助提取等。
尽管TPS的提取方式很多,但是不同的鸡枞菌种类、提取方式和纯化过程,所获得的TP S结构以及性质并不相同,同时也最终影响其生理活性。TPS的分子量主要分布在103~105Da。不同种类的TPS的单糖组成不尽相同。T.eurhizus和T.microcarpus的TPS单糖成分中仅含有葡萄糖(Glc)。T.striatus的TPS单糖组成主要为Glc、半乳糖(Gal)、甘露糖(Man)和岩藻糖(Fuc)。T.robustus的TPS单糖主要含有Fuc、Glc。T.albuminosus的TPS单糖成分主要为木糖(Xyl)、Fuc、Man、Gal、Glc、阿拉伯糖(Ara)、核糖(Rib)、鼠李糖(Rh a)。T.clypeatus和T.heimii中TPS主要含有Glc、Gal、Man和Fuc。
用蜗牛酶提取鸡枞菌(T.albuminosus)多糖(EIPS),在降低肝脂质水平和预防氧化应激方面,EIPS的效果明显优于水提和酸提的多糖。用酸提取法获得的鸡枞菌(T.albuminosus)多糖(ADES),ADES能降低高脂血症小鼠血清和肝脏脂质水平,降低血清酶活性,提高抗氧化酶活性和p-AMPKα的表达。碱提取得到的鸡枞菌(T.heimii)多糖能有效地抑制二甲基肼诱导的瑞士白化大鼠结肠癌的增生和杯状细胞的萌发。
发明内容
本发明的目的是克服现有技术的上述不足,提供一种鸡枞菌多糖及其制备方法和应用。
本发明的第一个目的是提供一种鸡枞菌多糖的制备方法。
本发明的第二个目的是提供所述制备方法制备得到的鸡枞菌多糖。
本发明的第三个目的是提供所述鸡枞菌多糖在制备治疗肠道炎症和/或肠道菌群失调的药物中的应用。
为了实现上述目的,本发明是通过以下方案予以实现的:
一种鸡枞菌多糖的制备方法,包括以下步骤:
S1:将鸡枞菌脱脂,得到脱脂后的鸡枞菌;
S2:混合步骤S1脱脂后的鸡枞菌和提取剂,其比例为1g:14.8~15.2mL,搅拌混匀,固液分离,得粗提液;
S3:对步骤S2的粗提液进行醇沉,固液分离,得粗提沉淀;
S4:用8000~12000Da透析袋透析步骤S3的粗提沉淀,得鸡枞菌多糖。
优选地,步骤S1用索氏提取法对鸡枞菌进行脱脂。
更优选地,将装有鸡枞菌粉末的滤纸包放入抽提筒中,注入无水乙醚,使鸡枞菌粉末包完全浸没在乙醚中;在64.9~65.1℃恒温水浴中进行抽提,至用滤纸点滴检查抽取筒内的乙醚无油迹为止;取出装有鸡枞菌粉末的滤纸包,待乙醚挥发后,将装有鸡枞菌粉末的滤纸包置于烘干,得脱脂鸡枞菌。
更优选地,所述无水乙醚为一次虹吸量2倍的无水乙醚,所述恒温水浴温度为65℃,抽提过程中,乙醚回流3次/h以上。
优选地,步骤S2所述提取剂为水、酸溶液或碱溶液,得到鸡枞菌水提多糖、鸡枞菌酸提多糖和鸡枞菌碱提多糖。
更优选地,所述酸溶液为HCl溶液,所述碱溶液为NaOH溶液。
更优选地,所述HCl溶液的浓度为0.9~1.1M,所述NaOH溶液为含有0.009~0.011MNaBH4的0.9~1.1M NaOH溶液。
更优选地,所述HCl溶液的浓度为1M,所述NaOH溶液为含有0.01M NaBH4的1M NaOH溶液。
优选地,步骤S2所述搅拌混匀时间为2.8~3.2h。

具体实施方式
下面结合说明书附图及具体实施例对本发明作出进一步地详细阐述,所述实施例只用于解释本发明,并非用于限定本发明的范围。下述实施例中所使用的试验方法如无特殊说明,均为常规方法;所使用的材料、试剂等,如无特殊说明,为可从商业途径得到的试剂和材料。
实施例1鸡枞菌多糖的提取
1、鸡枞菌(T.albuminosus)原料采购于云南。经洗涤和晾干后用于后续实验。
2、制备鸡枞菌粗多糖
(1)将新鲜鸡枞菌样品洗净后,冻干至恒重,粉碎成粉末。将鸡枞菌粉末经索氏提取法进行脱脂,得到脱脂后的鸡枞菌。具体脱脂方法为:将装有鸡枞菌粉末的滤纸包放入抽提筒中,注入一次虹吸量2倍的无水乙醚,使样品包完全浸没在乙醚中。在65℃恒温水浴中进行抽提,冷凝下滴的乙醚成连珠状(回流3次/h以上),抽提至抽取筒内的乙醚用滤纸点滴检查无油迹为止。抽提完毕后取出滤纸包,置于通风橱内待乙醚挥发。乙醚挥发后,将滤纸包置于50℃烘箱中干燥12h,得脱脂鸡枞菌粉末。
(2)配置提取液:
水提液:水(蒸馏水,85℃);酸提液:1M HCl溶液(室温);碱提:将0.01M NaBH4溶于1M NaOH溶液中,得到碱提液。
(3)1)分别混合步骤(2)各提取液和步骤(1)脱脂后的鸡枞菌,各组固液比都为1:15(w/v),以500rpm速率磁力搅拌混匀3h,分别经3000g离心15min,得上清液,旋转蒸发浓缩,得浓缩产物;旋蒸方法为将上清液在50℃温度下恒温水浴,在真空度0.08Mpa条件下浓缩至原体积三分之一;
2)醇沉:向步骤1)浓缩产物中加入95%(v/v)的乙醇溶液,使乙醇的最终浓度达到80%(v/v),4℃静置过夜(12h),得沉淀物。
3)用蒸馏水溶解沉淀物,所用蒸馏水质量为沉淀物的20倍,重复醇沉1次(步骤1)离心浓缩和步骤2))。收集醇沉沉淀,用水复溶,得复溶液。用8000~12000Da透析袋透析复溶液48h,得透析产物。冷冻干燥透析产物,获得三种鸡枞菌多糖,分别为:水提鸡枞菌多糖(Hot water extraction polysaccharide,HWEP)、酸提鸡枞菌多糖(Hydrochloric acidextract polysaccharide,HAEP)和碱提鸡枞菌多糖(Sodium hydroxide extractpolysaccharide,SHEP)。
实施例2实施例1鸡枞菌多糖的性质
一、实验方法
1、实施例1HWEP、HAEP和SHEP多糖得率计算公式为:
多糖得率(%)=提取所得鸡枞菌多糖质量/鸡枞菌粉末干重×100%。
2、测定总糖含量:参照苯酚-硫酸法测定样品的总糖含量。
(1)绘制葡萄糖标准曲线
精密称取干燥的无水葡萄糖标准品,溶解于超纯水配置成150mg/L标准葡萄糖溶液备用。于试管中分别加入10、20、30、40、50和60μL 150μg/mL葡萄糖标准溶液,再依次分别加入190、180、170、160、150和140μL 2.5%苯酚溶液,震匀。分别加入0.5mL浓硫酸,涡旋震匀,待溶液冷却至室温。蒸馏水作为空白对照,平行三次。于490nm处测量吸光度。以浓度x为横坐标、吸光度值A为纵坐标,建立标准曲线回归方程。
(2)测定样品总糖含量
分别取10mg实施例1的HWEP、HAEP和SHEP多糖样品到K-max玻璃管,冰浴,边震荡边分别加入0.45mL 72%浓硫酸,30℃水浴加热1h,充分混匀。待试管冷却后,分别加入4.95mL蒸馏水,涡旋震匀,100℃金属浴加热3h,每隔20min震荡反应管一次,冷却至室温得HWEP、HAEP和SHEP水解后样品溶液。于试管中加入各样品溶液40μL,再分别加入160μL 2.5%苯酚溶液,震匀。加入0.5mL浓硫酸,涡旋震匀,待溶液冷却至室温。蒸馏水作为空白对照,于490nm处测量吸光度。于标准曲线内计算出HWEP、HAEP和SHEP多糖样品对应的总糖浓度,计算总糖含量。
3、分析单糖组成
(1)水解:分别取10mg实施例1的HWEP、HAEP和SHEP多糖于K-max管,分别加入1mg肌醇,溶于0.45mL 72%的硫酸,30℃下水解1h。加入4.95mL蒸馏水,置于100℃条件下继续水解3h,得HWEP、HAEP和SHEP水解液。
(2)还原:分别取1mL HWEP、HAEP和SHEP水解液于K-max管,加入0.30mL 25%氨水溶液至溶液呈弱碱性(pH<7)。随后加入0.1mL 3M氨水溶液(含150mg/mL硼氢化钠)于30℃水浴加热1h,冰浴冷却。随后加入0.05mL醋酸至溶液pH<5,得HWEP、HAEP和SHEP还原样品。
(3)乙酰化:分别取0.2mL还原样品于K-max管,加入0.3mL N-甲基咪唑和2mL乙酸酐,涡旋混合。30℃水浴30min,冰浴冷却。随后加入3mL蒸馏水和2mL二氯甲烷,涡旋混合,1500g离心5min,弃上层。继续加入2mL蒸馏水和1.33mL二氯甲烷,涡旋混合,离心弃上层。继续加入2mL蒸馏水,涡旋混合,离心弃上层,重复3次。随后于40℃水浴加热蒸发二氯甲烷。加入0.67mL丙酮,涡旋混合后于40℃水浴加热蒸发丙酮,重复2次。加入0.27mL丙酮,充分混合后转移到气相小瓶用于气相色谱分析。
(4)分析:用气相色谱-氢火焰离子化检测器(GC-FID,Shimadzu G2010Plus,Kyoto,Japan)搭配DB-225色谱柱(Agilent Technologies Inc.,Calif.USA.15m×0.53mm×1μm)进行定性和定量分析。初始柱温设定在200℃保持1min,然后以10℃/min的速率逐渐升高至210℃,保持20min。载气为氮气(3mL/min)。进样量为2μL,进样温度为250℃。以鼠李糖(Rha)、岩藻糖(Fuc)、阿拉伯糖(Ara)、木糖(Xyl)、甘露糖(Man)、半乳糖(Gal)、葡萄糖(Glc)为标准品分析实施例1HWEP、HAEP和SHEP多糖样品中单糖组成。单糖标品的GC色谱图如图1所示。
4、测定分子量
用超纯水分别配置实施例1HWEP、HAEP和SHEP多糖样品,浓度都为2.5mg/mL。用高效凝胶排阻色谱测定多糖样品的分子量(Mw)。色谱柱用1根TSK Super AW-L(35×4.6mm)柱前保护柱和3根串联的TSK-GEL色谱柱(Super AW4000,3000,2500;150×6mm),结合RefractoMax520型示差折光检测器(RI)进行检测。以0.2M NaNO3为流动相,流速0.6mL/min。洗脱温度55℃;进样量20μL;用葡聚糖标准液(10、40、70、500和2000kDa)为标准品建立标准曲线,计算多糖样品分子量。以横坐标表示分子量,纵坐标表示出峰时间。标准曲线方程为:y=-1.4031x+18.365,R2=0.9959。
5、测定蛋白含量
用BCA蛋白质定量试剂盒测定鸡枞菌子实体干粉末和实施例1的HWEP、HAEP和SHEP多糖样品的蛋白含量。称取标准蛋白粉末5.00mg,0.9% PBS充分溶解后配置成5mg/mL的蛋白标准溶液。按照50:1的体积比分别加入BCA试剂A液和B液,充分混匀,24h内使用。调整蛋白标准溶液浓度,建立标准曲线使样品浓度落在标准曲线范围内,用吸光度计算蛋白质含量。
6、测定氨基酸含量
将鸡枞菌子实体干粉末以及提取的鸡枞菌粗多糖样品进行氨基酸组成测定。前处理方法:分别称取0.1g不同的鸡枞菌多糖样品于10mL 6M盐酸中水解22h(干燥箱105℃)。打开水解管,定容至25mL。过滤,吸取1mL,蒸发至干,残留物用5mL去离子复溶水再次蒸干,重复两次。最后用2mL pH 2.2柠檬酸钠缓冲溶液溶解,过0.45μm滤膜备用。用Biochrom 30全自动氨基酸分析仪(瑞士华嘉公司)分析氨基酸组成。
7、FT-IR分析
分别称取3mg实施例1的HWEP、HAEP和SHEP多糖样品,分别与100mg KBr粉末混合。研磨并用压片机压成薄片进行傅里叶变换-红外光谱分析。通过Nicolet iS50+iN10光谱仪(Thermo Fisher 114Scientific,MA,USA)进行FT-IR分析。波长范围为4000-500cm-1。
8、数据处理
用GraphPad Prism 9.3.1(GraphPad Software,San Diego,CA,USA)对本实施例实验结果进行图表绘制。用Statistix 8分析数据,用单因素方差分析和LSD多重比较分析法。结果以均值±标准差(Mean±SD)表示。p<0.05表示具有统计学意义。
二、实验结果
1、鸡枞菌多糖和单糖组成分析
(1)多糖:实施例1的HWEP、HAEP和SHEP多糖得率分别为3.90±0.51%、2.70±0.36%和3.34±0.42%(表1)。可见水提、酸提和碱提都能得到较高得率的溶出多糖。在酸或碱性条件下,细胞壁多糖之间或多糖与蛋白质之间的天然相互作用更容易被破坏,使多糖更好溶出。实施例1的HWEP、HAEP和SHEP的总糖含量分别为57.57±4.30%、48.98±1.12%和40.00±5.10%。
(2)单糖:
如表1所示,实施例1的HWEP、HAEP和SHEP多糖都由Rha、Fuc、Ara、Xyl、Man、Gal和Glc组成。不同提取方式鸡枞菌粗多糖中的单糖组成摩尔比各不相同。HWEP中Rha、Fuc、Ara、Xyl、Man、Gal和Glc的摩尔比分别为0.59:6.71:0.16:1.41:4.22:22.14:64.77;HAEP为0.48:5.97:0.56:2.88:6.89:23.78:59.44;SHEP为0.65:1.96:0.20:1.54:0.93:5.12:89.60。HWEP、HAEP和SHEP的单糖摩尔占比最多的为Glc,分别为64.77%、59.44%和89.60%,其次是Gal,分别占22.14%、22.78%和5.12%。SHEP的Glc占比最高,说明碱液提取增加了鸡枞菌细胞壁中葡聚糖的溶出。明鸡枞菌(T.albuminosus)多糖为杂多糖,单糖组成之间有明显差异,不同方法提取的多糖在结构上存在较大差异。

2、测定鸡枞菌多糖分子量
如图2所示,实施例1的HWEP、HAEP和SHEP多糖都具有两个峰。HWEP组具有两个分子量分别为673.85kDa和19.88kDa的组分,其中673.85kDa的组分均较HAEP组(412.37kDa和21.87kDa)和SHEP(510.40kDa和29.24kDa)组中分子量更大(表2)。可能酸和碱的作用导致鸡枞菌细胞壁多糖进一步裂解为小分子聚合物且部分小分子在透析步骤被去除,导致HAEP和SHEP的分子量以及总糖含量比HWEP低。酸碱处理促进鸡枞菌细胞壁中小分子多糖的释放。
3、鸡枞菌多糖FT-IR分析
如图3所示,在3420、2920、1620、1400、1250和1100cm-1处的信号是多糖的典型特征峰。3420cm-1附近的强吸收峰来自于O-H的拉伸,其中HWEP在3405.32cm-1,HAEP在3405.82cm-1,SHEP在3414.07cm-1。在2920cm-1附近的波段吸收峰归因于C-H伸缩振动,其中HWEP在2929.3cm-1,HAEP在2919.26cm-1,SHEP在2919.67cm-1。在1620cm-1(HWEP在1639.79cm-1,HAEP在1647.96cm-1,SHEP在1639.42cm-1)和1400cm-1(HWEP为1421.70cm-1,HAEP在1412.36cm-1,SHEP在1422.26cm-1)的吸收峰分别由不对称和对称的C=O基团拉伸所至,表明实施例1的鸡枞菌多糖中存在羧基。近1250cm-1(HWEP在1239.55cm-1,HAEP在1242.65cm-1)的信号峰是由于S=O的拉伸振动,表明HWEP和HAEP-含有硫酸盐基。在1200~1000cm-1处的强特征吸收是由于C-O-C糖苷键的振动。在1100cm-1附近的具有吸收峰(HWEP在1080.02cm-1处,HAEP在1079.49cm-1,SHEP在1150.29cm-1处)表明HWEP、HAEP和SHEP多糖的糖环为吡喃环。
4、测定鸡枞菌多糖蛋白质和氨基酸
如表2所示,鸡枞菌子实体中蛋白含量为25.82%,含有16种氨基酸。HAEP(25.012%)的蛋白含量较高于HWEP(7.177%)和SHEP(5.953%)。HWEP和HAEP组的鸡枞菌多糖具有17种氨基酸,相比而言SHEP中缺少胱氨酸。表2中“-”表示,低于检测限,未检出。

实施例3鸡枞菌多糖对模拟体外消化和结肠发酵的影响
一、实验方法
1、体外消化
按表3提前24h配置好模拟唾液(SSF模拟液)、模拟胃液(SGF模拟液)和模拟肠液(SIF模拟液)的模拟液,分别取实施例1的HWEP、HAEP和SHEP各0.5g,加入10mL PBS溶液配成0.05g/mL多糖溶液样品,以不加任何样品的PBS溶液为空白组(BC),每组三个平行。以最终消化体积80mL,进行口腔、胃和小肠连续消化。体外消化后,将不同多糖的肠消化样品在3500Da透析袋中透析2d,模拟肠道吸收。最后,将残液冻干,得HWEP、HAEP和SHEP多糖消化冻干粉,低温保存以用于后续体外发酵。具体口腔、胃、肠的消化步骤如下所示。
口腔消化:将α-淀粉酶(10U/mg,250mg/样)加入SSF模拟液,混匀,得SSF消化液。于各组多糖溶液样品中加入10mL SSF模拟液。将pH调至7.0,在37℃搅拌孵育2min,得不同多糖的口腔消化样品。各取0.5mL样品留样。
胃消化:将胃蛋白酶(3000U/mg,52mg/样),加入SGF模拟液,混匀,得SGF消化液。各取20mL不同多糖的口腔消化样品,分别加入SGF消化液中,混匀,将pH调至3.0,在37℃搅拌孵育2h,得不同多糖的胃消化样品。
肠消化:将胰蛋白酶(250U/mg,64mg/样)加入SIF模拟液,混匀,得SIF消化液。各取40mL不同多糖的胃消化样品,分别加入SIF消化液中,混匀,将pH调至7.0,在37℃搅拌孵育3h,得不同多糖的肠消化样品。

2、体外发酵
(1)制备培养基
配制pH 6.0的2-(N-吗啉)乙磺酸缓冲液(MES),用MES缓冲液配制模拟回肠环境培养基(SIEM)。SIEM培养基的配置方法为:取氯化钠4.5g、磷酸氢二钾2.5g、氯化钙0.34g、水合硫酸镁0.5g、水合硫酸铁、牛胆粉0.05g、半胱氨酸0.4g、细菌蛋白胨3g、酸水解酪蛋白3g、氯化血红素0.01g、维生素混合液(AIN 76混合维生素(MP Biomedicals))1mL和1%刃天青1mL混合溶解,加MES缓冲液定容至1L,pH调至5.8~6.0,过0.22μm无菌滤膜。
(2)制备人体粪便菌液
粪便样品贡献者为健康、无肠道疾病、3个月未摄入抗生素的24~28岁的志愿者。将粪便样品用PBS缓冲溶液按1:10(w/v)比例稀释,4℃,500g离心10min取上清菌液备用。将3~5人的粪便上清液各取相同体积混匀,得粪便接种液。
(3)接种和培养
用本实施例步骤2配置的培养基,将本实施例的HWEP、HAEP和SHEP多糖消化冻干粉和低聚果糖(FOS)分别配制成10mg/mL的溶液作为实验组和阳性对照组,本实施例步骤2配置的空白培养基组作为空白对照组(BC)。在厌氧箱中将粪便接种液按10%的接种量分别接入到各实验组和对照组中,充分混匀,总体系为5mL进行体外发酵。将发酵管放入培养箱振荡器培养(120rpm,37℃)。发酵管密封后上端插入注射器以计量发酵过程产气情况。设置5个发酵时间点,分别在0、6、12、24和48h收集发酵液样品,每个样品设置3个平行。
3、测定总糖、单糖含量与实施例2相同,将鸡枞菌多糖样品换成鸡枞菌多糖体外发酵液样品。
4、测定pH值:用微量pH计测量不同时间点发酵液样品的pH值。
5、测定短链脂肪酸(SCFAs)
取300μL本实施例步骤2的发酵样品,分别与含有300μL 0.15mg/mL二乙基乙酸(溶解于0.2M HCl)和75μL 0.15M草酸的混合液充分混匀。12000g离心10min,取上清液用于气相色谱仪(GC)分析。标准样品为乙酸、丙酸、丁酸、戊酸、异丁酸和异戊酸。标品处理方法与样品处理方法一致。GC搭配氢火焰离子检测器(FID)以及CP-FFAP CB色谱柱(25m×0.53mm×1.00μm,Agilent)。GC参数:氮气为载气,初始温度100℃,5℃/min程序升温至160℃,保持4min。
6、16S rDNA测序
取本实施例步骤2的0h以及48h发酵样品进行16S rDNA基因测序。用DNA提取试剂盒(Life Technologies)从小鼠的结肠内容物中提取核酸。用Qbiut(厂家invitrogen,型号Qubit3.0,试剂Qubit TM dsDNA HS Assay Kit)对提取的核酸进行浓度检测,并使用琼脂糖电泳对完整性进行检测。用343F(5’-TACGGRAGGCAGCAG-3’)和798R(5’-AGGGTATCTAATCCT-3’)通用引物对结合适配序列和条形码序列扩增细菌16S rDNA基因的V3-V4区域。PCR产物用Quant-iT dsDNA HS试剂定量。用Illumina NovaSeq 6000 2*250对纯化后的汇总样本进行细菌rRNA基因高通量测序分析。用QIIME(version 1.9.1)数据分析包分析16S rDNA数据。原始数据用FLASH(version 1.2.11)拼接,拼接后的序列用Trimmomatic(version 0.33)进行批量滤波。用聚类程序USEARCH(version 10.0)将有效序列分组为操作分类单位(OTUs)。进行生物信息分析。
7、数据处理与实施例2相同。
二、实验结果
1、体外消化液分子量变化
HWEP(图4A)、HAEP(图4B)和SHEP(图4C)的分子量分布在口腔、胃和小肠消化阶段均相似,没有显著变化,表明HWEP、HAEP和SHEP都几乎不被口腔消化酶、胃消化酶以及肠消化酶降解。经体外吸收后,其小分子物质被透析去除,其分子量分布与消化之前分子量无显著差异,说明实施例1的HWEP、HAEP和SHEP多糖属于非消化性多糖。
2、体外发酵液pH变化
发酵过程中pH值变化如图5所示,BC组pH值与初始值保持一致,无明显变化,实施例1的HWEP、SHEP、HAEP和FOS组在发酵过程中pH持续下降,且FOS组的下降幅度最大,其次是HWEP组。发酵6~12h之间,FOS组pH值显著下降。SHEP组和HAEP组pH值下降趋势基本一致。HWEP组发酵48h后,相比BC组pH值显著降低。在体外发酵过程中,多糖组的pH值下降,显示其被微生物利用。
3、体外发酵液总糖及单糖组成变化
体外发酵液总糖及单糖组成变化速率反映了微生物利用多糖的速度。如图6A所示。实施例1的HWEP、HAEP和SHEP多糖在体外发酵过程中总糖含量都不断减少,表明其被被肠道微生物酵解利用。HWEP组在整个发酵过程中总糖含量下降最大,下降了初始糖含量的42.37%;其次是HAEP组、SHEP组。HWEP组和HAEP组均在前24h发酵过程中下降较快,随后缓慢下降。而SHEP组的总糖含量下降主要集中在12~24h之间。体外发酵过程中HWEP(图6B)、HAEP(图6C)、和SHEP(图6D)组的半乳糖(Gal)、葡萄糖(Glc)、甘露糖(Man)和阿拉伯糖(Ara)的含量均随发酵时间延长而不断下降,下降程度最大的为Gal和Glc。经48h体外发酵,HWEP、HAEP和SHEP组Gal含量分别下降了初始含量的46.99%、60.14%和65.65%,HWEP、HAEP和SHEP组Glc含量分别下降了初始含量的74.16%、77.6%和67.45%,说明在体外发酵过程中,实施例1的HWEP、HAEP和SHEP多糖被肠道微生物利用,主要集中在发酵前24h;肠道微生物主要利用多糖中Gal、Glc、Man和Ara单糖,其中Gal和Glc的利用程度最大,且HWEP和HAEP在体外发酵体系中具有较好的可发酵性。
4、体外发酵液SCFAs变化
图7A表明,各组总SCFAs含量都随发酵时间的延长而不断增加;与BC组相比,发酵12h~48h,多糖组和FOS组总SCFAs含量都显著升高(p<0.05),FOS组在48h达到最高值,其次是HAEP组、SHEP组和HWEP组。从不同SCFAs含量来看,经体外发酵48h后,与BC组相比,各多糖组的乙酸(图7B)和丙酸(图7C)含量均显著增高(p<0.05)。发酵48h后,与BC组相比,HWEP组异丁酸(图7D)水平显著升高(p<0.05),HAEP的异丁酸(图7D)、丁酸(图7E)和异戊酸(图7F)含量均显著升高(p<0.05),SHEP组的丁酸(图7E)和异戊酸(图7F)含量显著增高(p<0.05)。此外,与FOS组相比,各多糖组的丙酸(图7C)含量均增高;HAEP组的异丁酸(图7D)和异戊酸(图7F)含量升高,SHEP组的丁酸(图7E)、异戊酸(图7F)含量增高。各组戊酸(图7G)没有明显差异。在发酵液中添加实施例1的HAEP、SHEP和HWEP多糖促进SCFAs的产生,三种多糖对丙酸盐和丁酸盐的产生具有较好的促进效果,HAEP多糖效果最佳。
5、体外发酵液微生物菌群变化分析
(1)体外发酵液微生物Alpha多样性分析
各组样品Good’s coverage值均接近1,表明微生物样本充分,测序达到深度要求(图8A)。从Chao1指数结果可见,经48h体外发酵后,各组的Chao1值均降低,暗示物种丰富度降低(图8B)。发酵48h后,与BC组相比,HAEP组显著下降(p<0.05),其他组无显著差异(p>0.05);与FOS组相比,各组Chao1值均显著减小(p<0.05)。经Simpon分析(图8C),发酵48h后,HWEP组较FOS组以及BC组均显著下降(p<0.05),其余组之间无显著差异。表明HAEP和SHEP组对微生物的作用与FOS组相似,而HWEP组的物种多样性显著降低。
(2)体外发酵液微生物Beta多样性分析
基于weighted_unifrac算法和进行主坐标(PCoA)分析,绘制3D图。3个主成分分别解释了68.12%、7.03%和2.20%的微生物多样性的变化。相同颜色点代表同一组的不同平行样本,样本间距离越远则表示微生物群落构成差异越大。如图9,各组平行之间距离相近,表明可信度高。发酵48h后,与BC组相比,FOS组以及HWEP组、HAEP组和SHEP组均可见明显分离,相隔较远,表明其微生物群落构成具有较大差异。HAEP组与SHEP组之间相互交错在一起,未见明显分离,表明两组之间微生物群落构成较为相似。说明FOS和HWEP、HAEP和SHEP在体外发酵过程中都对微生物群落构成造成影响,且影响因素各不相同。
(3)多糖在门水平对体外发酵液微生物的影响
将0h的空白对照组(BC-0)以及经48h的各组进行门水平的相对丰度分析,结果如图10和表4所示。0h发酵液的优势菌群主要为Bacteroidota以及Firmicutes(BC-0),经48h体外发酵之后,各组的微生物菌群组成发生改变,BC组中的优势门为Proteobacteria、Bacteroidota和Firmicutes。

如图10A所示,与BC组相比,HWEP、HAEP和SHEP组的Bacteroidota相对丰度显著提高(p<0.05),且占比最高;与BC组相比,HAEP组和SHEP组Firmicutes的相对丰度显著降低(p<0.05);HWEP组Firmicutes的相对丰度下调,但无显著性差异(p>0.05)。HWEP组、HAEP组和SHEP组均极显著降低了F/B值(图10B)。此外,经过体外发酵48h后,与BC组相比,HWEP、HAEP和SHEP组Proteobacteria的相对丰度无显著差异(p>0.05);而Actinobacteriota和Fusobacteria的相对丰度显著下降(p<0.05)。
(4)多糖在属水平对体外发酵液微生物的影响
图11A所示为相对丰度排名前30的菌属。0h发酵液中占比最多的菌属为Bacteroides和Muribaculaceae。经48h体外发酵之后,各组之间菌属结构组成明显改变。图11B单独对各组48h发酵液优势菌属进行了分析统计。与BC组相比,各多糖组都上调了Enterobacter和Prevotella的相对丰度(p<0.05),下调了Escherichia-Shigella、Fusobacterium和Sutterella的相对丰度。与FOS组相比,Escherichia-Shigella、Fusobacterium和Muribaculaceae相对丰度显著减少,Enterobacter、Prevotella和Dialister的相对丰度显著增加。此外,HWEP和HAEP组极显著下调了Sutterella的相对丰度(p<0.01)。HAEP和SHEP组极显著提高了Bacteroides的相对丰度(p<0.001)。HAEP组显著下调了Muribaculaceae的相对丰度(p<0.05)。
多糖组中Prevotella显著上调,说明多糖的加入促进了Prevotella生长以及其对多糖的利用。HWEP、HAEP和SHEP组中Escherichia-Shigella和Fusobacterium均显著下调,说明多糖对Escherichia-Shigella和Fusobacterium具有明显的抑制作用,且对肠道黏膜具有保护作用。
实施例4鸡枞菌多糖对DSS诱导的IBD小鼠的影响
一、实验方法
1、建立DSS诱导小鼠结肠炎模型
将40只雄性小鼠(C57BL/6J)随机分为5个实验组,每组8只。分别为:空白对照组(CON)、造模对照组(DSS)、水提鸡枞菌多糖干预组(DSS+HWEP)、酸提鸡枞菌多糖干预组(DSS+HAEP)和碱提鸡枞菌干预组(DSS+SHEP)。共2个周期造模,分别为12天和14天,第一周期和第二周期造膜组和干预组分别自由饮用1.9%和2.3%的DSS溶液建立IBD模型。两个周期之间进行4天恢复,恢复期间各实验组均饮用无菌水。CON组在整个周期正常饮无菌水。其中DSS+HWEP、DSS+HAEP和DSS+SHEP组在造模期间分别给50mg/kg/d的HWEP、HAEP和SHEP灌胃(鸡枞菌粗多糖由生理盐水均匀溶解),CON组给予等体积生理盐水灌胃。DSS溶液现配现用,每2天换一次。造模共30天,期间每天各实验组和对照组都进行灌胃。HWEP、HAEP和SHEP多糖为实施例1制备得到。
2、小鼠体重记录与疾病活动指数评分
从造模第0天开始,每2d记录造模组DSS溶液以及对照组无菌水的消耗量。每日称量小鼠体重并记录。体重变化以目标天数与第一天小鼠体重质量增减量除以第一天小鼠体重并以百分比表示。对各组小鼠的一般活动情况(精神状态、食欲等)进行观察记录。当天灌胃结束后,收集新鲜粪便,对各种小鼠粪便形状、硬度、血便情况等进行记录评分。使用隐血试剂盒检测各小鼠隐血状况。按照试剂盒说明书滴加显色液A(邻联甲苯胺溶液)和B(过氧化氢),记录显色时长,结合显色时长与显色结果判定隐血状况。综合体重变化、粪便形态、隐血试剂盒粪便显色时长及显色结果,对各组小鼠进行疾病活动指数(DAI)评分。根据小鼠体重下降百分比所得分数、粪便性状、隐血状况的评分总和并除以3,得出每只小鼠DAI,判断各组小鼠造模情况以便及时对造模进行调整。DAI评分细则如表5所示。正常粪便应为成型粪便:长短、软硬适中,椭圆形结实粪便;松散粪便:易散粪便,不黏附于肛门呈糊状、半成形粪便。稀便:可黏附于肛门的稀水样便。

3、小鼠解剖以及血液及组织取样和储存
两个周期的造模结束后,隔天解剖各组小鼠,此前12h内禁水不禁食。记录小鼠体重。用异氟烷麻醉小鼠,收集小鼠眼球外周血于抗凝管中,净置30min,4℃,1000g离心15min,取上层血浆置于无菌无酶离心管中。相同条件重复离心一次,收集上清液置于-80℃冰箱冻存备用。
外周血收集完毕,颈椎脱臼法处死小鼠。解剖取出小鼠整个肠道(包括盲肠和结肠)。测量结肠长度并统一拍照记录。首先从距回肠末端与盲肠互相交接处约1cm处取结肠组织约0.4~0.6cm(选取粪便稀少段),轻轻剪取一段结肠环,直接通过4%甲醛固定。留样备于结肠组织蜡块包埋以及H&E染色和阿利新蓝染色。然后截取回盲部结肠并进行剖解,收取粪便内容物,液氮速冻。剪开余留的结肠,收集粪便内容物并液氮速冻。置于-80℃存样。剩于解剖的结肠部分均分成2份,一部分置于Trizol中用于RNA的提取,一份速冻后置于-80℃存样。
解剖小鼠肝脏及脾脏,同时观察其有无损伤,形态是否有异样,称量记录。
计算方法:脾脏指数(%)=脾脏重量(g)/体重(g)×100%
4、小鼠结肠组织病理学观察
结肠组织病理学评分均为盲阅打分,结肠炎症相关组织学评分参考国际标准,如表6。

5、小鼠结肠组织中相关基因的表达
(1)用Magen生物(美基生物),IVD3020(通用)型RNA提取试剂盒,对结肠组织RNA进行提取,得结肠组织RNA。
(2)RNA逆转录
1)去DNA反应(40μL体系)
根据表7,进行去DNA反应。先将5×gDNA Cleaner Buffer,RNase Free dH2O混匀。然后加入结肠组织RNA。置于PCR仪,42℃条件下反应2min。

2)用Green Pro Taq HS预混型qPCR试剂盒(含Rox)对去DNA的结肠组织RNA进行逆转录,得cDNA。
3)荧光定量PCR检测
将本实施例RNA逆转录提取的cDNA用无酶水稀释5倍。准备384孔PCR板,每个孔含有5.0μL SYBR Premix Ex TaqⅡ、0.2μL ROX Reference DyeⅡ、各0.4μL 10mM正、反链引物和3.0μL DEPC水(试剂混合液)。在加样时,先加试剂混合液,每孔9μL。最后加cDNA 1μL。每个样品都以β-Actin为内参。目的基因和内参均平行三次。于PCR仪(QuantStudio 6Flex)进行扩增。预变性(Repeat:1):95℃,30s;PCR反应(Repeat:40):95℃,5s;60℃,30s;Dissociation。引物信息如表8所示。反应结束后,根据ΔΔCt法以及结果中的Ct值进行目的基因表达的计算。公式为ΔCt=Ct(目的基因)-Ct(内参基因)。ΔΔCt=ΔCt(实验组样品)-ΔCt(对照组样品)。相对表达量=2-ΔΔCt。计算方法如下:

5、小鼠结肠内容物16S rDNA测序
选取对照组和鸡枞菌多糖干预组的结肠内容物样品进行16S rDNA基因测序。每组随机挑选3只。方法同实施例3。
6、小鼠盲肠内容物SCFAs测定
配置0.15mg/mL二乙基乙酸(溶于0.2M HCl中)做为内标。按4:1(v/v)将其与0.15M草酸混合成混合液。准确称取小鼠盲肠内容物约50mg,按照1:10(w/v)加入约500μL混合液。于4℃预冷,并加入两颗无菌钢珠,涡旋混匀至无肉眼可见的固体。4℃,10000g离心10min。取上清液,过0.22μm滤膜并注入气相小瓶中。采用GC测定盲肠内容物中的SCFAs。GC参数同实施例3。

二、实验结果
1、鸡枞菌粗多糖对小鼠炎症表现的影响
实验中使用的鸡枞菌多糖剂量为50mg/kg/d,相当于成年人(60kg)每天约330mg的鸡枞菌粗多糖摄入。如图12A所示,在实验期间,CON组初期由于环境适应期导致体重波动。从第6d起,CON组小鼠食欲正常,精神状态较佳,体重持续增加且趋势稳定(图12A)。其大便正常,多为成椭圆形软硬适中粪便,无血便稀便,DAI评分趋于0且趋势平缓稳定(图12B)。相比之下,DSS组(阳性对照组)在第一个DSS造模周期中,体重持续显著下降,直到第14d(恢复期第2天)降到最低值(p<0.05)(图12A)。DSS组在第4d出现不成型稀便、隐血。DAI评分显著高于CON组(p<0.05),在第14d达到最高。经4d恢复期后,其体重回升,在第20d恢复至初始值,后在第二个DSS造模周期中体重重新持续下降。与DSS组相比,鸡枞菌多糖干预组体重变化以及DAI变化总趋势与其一致。经多糖干预之后,DSS诱导的小鼠的体重下降在第8~10d左右开始出现,相比DSS组均有延缓;第二个DSS造模周期中,体重没有出现持续降低。HWEP和SHEP组小鼠在24d开始体重回升,HAEP组在28d体重出现明显增加。此外,各多糖组的便血症状较DSS组减轻,在恢复期第1d起,便血情况有明显改善,各多糖组第14d的DAI值相较于DSS组明显降低(p<0.05)(图12B)。且在第二个DSS造模周期中,其DAI值较DSS组都降低,稀便减少,腹泻、便血症状减轻。表明实施例1的HWEP、HAEP和SHEP多糖对DSS诱导的IBD小鼠体重下降、隐血及腹泻有缓解的作用,且50mg/kg/d剂量的HAEP较优。
从图13A可见,与CON组相比,DSS组结肠长度明显缩短(p<0.05);与DSS组相比,三种多糖组小鼠结肠长度增加,但无显著性差异(p>0.05)。从结肠解剖状态(图13B)来看,DSS组小鼠结肠有不同程度的肿胀及肠内出血损伤,肠后端、末端粪便呈稀状或不成形松散状。DSS组在结肠在摘取时较其他组更易断裂损伤。CON组结肠组织无异常,肠后端、末端粪便呈梭状或椭圆状。HWEP、HAEP及SHEP组结肠组织状态优于DSS组。表明50mg/kg/d HWEP、HAEP及SHEP多糖改善DSS诱导的结肠长度减少,缓解DSS诱导的IBD小鼠结肠肿胀、出血等炎症。
DSS组脾脏指数高于CON组,但无显著差异(p>0.05)(图13C)。但各多糖组的脾脏指数高于DSS组(p<0.05)。如图13D所示,除SHEP组肝脏指数高于DSS组外(p<0.05),其余各组相较于DSS组均无显著性差异,实施例1的HWEP及HAEP多糖对IBD小鼠肝功能有保护作用。
2、鸡枞菌多糖对IBD小鼠炎症反应的调节作用
正常的结肠组织形态良好,具有完整的黏膜层,隐窝结构完整,其中的杯状细胞排列有序,且无炎症浸润异样,与CON组的结肠切片结果一致(图14A)。DSS组结肠上皮细胞损伤严重,小鼠结肠黏膜有多处炎症浸润且程度较深,隐窝结构受到破坏(图14A)。DSS组小鼠炎症处可见弥漫性中性粒细胞、单核细胞等炎性细胞浸润,其结肠轮廓变化以及组织病理学变化与UC患者的临床表现相似,说明了模型的可靠性。如图14B所示,DSS组组织病理评分显著高于CON组(p<0.001)。与DSS组相比,HAEP、HWEP和SHEP组的组织病理学评分降低。其中,HAEP和HWEP组与DSS组相比具有显著性差异(p<0.05)。多糖干组的IBD小鼠结肠中,肠上皮细胞排列整齐且结构完整,仅有少量表现皮损,隐窝结构明显,少量炎症细胞浸润。表明实施例1的HWEP、HAEP和SHEP多糖改善结肠组织损伤,减少炎症细胞浸润。对DSS诱导的结肠组织损伤等炎症反应的具有改善作用。
与CON组相比,DSS组IL-6(图15A)水平上调,鸡枞菌多糖干预后IL-6表达水平较DSS组下调(p>0.05)但均无显著性差异(p>0.05)。与CON组相比,DSS组IL-1β(图15B)及TNF-α(图15C)的mRNA表达水平非常显著提高(p<0.01)。与DSS组相比,在鸡枞菌多糖干预后,各干预组IL-1β及TNF-α促炎症细胞因子均显著下调(p<0.05),其中对于TNF-α的下调作用显著(p<0.01),对IL-6下调但不显著。
3、鸡枞菌粗多糖对IBD小鼠肠道屏障的影响
1)、结肠组织切片阿利新蓝染色和肠黏膜变化情况
CON组小鼠结肠含有大量杯状细胞且分布均匀,杯状细胞内富含粘液。DSS诱导的IBD小鼠结肠组织中杯状细胞数量显著减少,粘液减少。各多糖干预组中杯状细胞数量、形态及分布(图16A)和粘液分泌情况(图16B)优于DSS组。与CON组相比,DSS组的黏液占比显著降低(p<0.05)。而与DSS组相比,HWEP组的黏液占比显著增加。具有显著差异(p<0.05)。
2)、鸡枞菌粗多糖对结肠组织中紧密连接蛋白表达水平的影响
如图17A和图17B所示,与CON组相比,DSS组的Claudin-1与Claudin-2表达水平显著增加(p<0.05)。相较于DSS组,各多糖组Cluadins的表达水平都下降;其中,HAEP干预后Cluadin-1表达水平显著下降(p<0.05),HWEP干预后Cluadin-2表达水平显著下降(p<0.05)。如图17C所示,相较于CON组,DSS组中Occludin的表达水平极显著增加(p<0.01)。各多糖组表达水平相较于DSS组下降(p<0.05)。其中SHEP组Occludin下降最多。显示实施例1的HWEP、HAEP和SHEP多糖对于DSS诱导的TJ蛋白上调具有缓解作用。
4、鸡枞菌粗多糖促进IBD小鼠盲肠内容物SCFAs的生成
如图18A所示,与CON组相比,DSS组小鼠盲肠内容物中总SCFAs含量明显减少(p<0.05)。由于DSS组小鼠因炎症反应导致肠道菌群紊乱,产生SCFAs的菌群减少。与DSS组相比,多糖组都提高了小鼠盲肠内容物中总SCFAs含量,其中HAEP组与DSS组之间具有显著差异(p<0.05)。与DSS组相比,除戊酸(图18G)外,HAEP组的其他SCFAs均显著提高(p<0.05),达到了CON组小鼠相同的水平。HWEP组的异丁酸(图18D)、异戊酸(图18F)以及戊酸(图18G)水平显著高于DSS组(p<0.05),SHEP组的丙酸(图18C)和戊酸(图18G)水平均显著高于DSS组(p<0.05)。鸡枞菌样品HWEP与SHEP组的乙酸(图18B)和丁酸(图18E)均高于DSS组,但无显著性差异。实施例1的HWEP、HAEP和SHEP多糖对DSS诱导的IBD小鼠结肠SCFAs减少缓解作用,HAEP具有最佳效果。
5、鸡枞菌粗多糖对IBD小鼠结肠微生物的影响
(1)鸡枞菌多糖对肠道微生物多样性和丰富度的影响
小鼠结肠内容物的微生物Alpha及Beta多样性分析如图19所示。从图19A看出,各组曲线一开始快速上升,随后逐渐趋于平缓,表示该样本测序量充足,足以进行数据分析。与CON组相比,DSS组的Chao1指数显著降低(p<0.05);与DSS组相比,HWEP和HAEP组Chao1指数无显著差异(p>0.05),SHEP组的Chao1指数显著提高(p<0.05)(图19B),表明DSS组小鼠结肠微生物物种丰富度相较于正常小鼠显著减少,SHEP多糖干预逆转了这一现象。如图19C和图19D所示,DSS组的Shannon和Simpson指数均较CON组均降低,显示其物种多样性下降。与DSS组相比,HWEP组和HAEP组的Shannon和Simpson指数均无显著差异(p>0.05),而SHEP组的Simpson指数显著提高(p<0.05)。SHEP组的微生物丰度以及其多样性均显示最高值。
通过PCoA分析搭配加权weighted UniFrac算法计算了Beta多样性距离矩阵。如图19E,3个主成分分别解释了54.86%、8.38%和7.04%的微生物多样性的变化。与CON组相比,DSS组明显与其距离较远,说明其微生物结构存在明显差异。三个干预组空间分布交错在一起,且都和DSS组结肠微生物物种分布有分离。说明实施例1的HWEP、HAEP和SHEP多糖改变了微生物结构。
(2)鸡枞菌粗多糖对肠道微生物门水平相对丰度的影响
如图20A所示,各组小鼠的优势门都为Bacteroidota和Firmicutes。与CON组相比,DSS组的Bacteroidota的相对丰度降低;多糖组的Bacteroidota相对丰度都有上调,接近CON组水平。各组之间Firmicutes的相对丰度无显著差异(p>0.05)。如图20B所示,DSS组的F/B值比CON组显著增加(p<0.001),多糖组与DSS组相比,F/B值均显著降低(p<0.05),其中HAEP组极显著降低(p<0.001)。表明鸡枞菌多糖对DSS诱导的小鼠结肠微生物菌群紊乱有较好调节作用。由图20D~G所示,多糖组与DSS组相比,上调了Bacteroidota,下调了Firmicutes,HWEP和SHEP组下调了Proteobacteria,HAEP和SHEP组下调了Actinobacteria的相对丰度,但无显著差异。多糖组对Verrucomicrobia的相对丰度也无显著差异。
(3)鸡枞菌粗多糖对肠道微生物属水平相对丰度的影响
如图21A所示,CON组中,uncultured_bacterium_f_Muribaculaceae、uncultured_bacterium_f_Lachnospiraceae、Lactobacillus、Alistipes和Akkermansia占据了约60%的相对丰度。如图21B所示,与CON组相比,DSS组的uncultured_bacterium_f_Muribaculaceae、Akkermansia、Lactobacillus和Alistipes的相对丰度都下降,uncultured_bacterium_f_Lachnospiraceae相对丰度上升,其中Alistipes显著减少。相比于DSS组,HWEP、HAEP和SHEP组中uncultured_bacterium_f_Muribaculaceae和Lactobacillus相对丰度均上调,接近CON组,Akkermansia的相对丰度都降低,但无显著性差异。表明实施例1的HWEP、HAEP和SHEP多糖通过促进抗炎细菌的增殖进而抑制炎症因子的表达以缓解IBD小鼠的炎症。与DSS组相比,HAEP组下调了uncultured_bacterium_f_Lachnospiraceae的相对丰度、上调了Bifidobacterium的相对丰度(p<0.05)的相对丰度。
实施例5鸡枞菌多糖对模拟IBD小鼠体外发酵的影响
一、实验方法
(1)体外发酵:培养基的制备方法与实施例3相同。
(2)IBD小鼠粪便菌液的制备
收集12只的IBD小鼠的盲肠粪便(造模过程同实施例4),用PBS稀释成0.1~0.2g/mL的粪便溶液,涡旋充分混匀,2000g离心5min取上清菌液备用,用于下一步样品接种。
(3)接种和培养
用培养基将实施例3步骤1的3种多糖消化冻干粉(HWEP、HAEP和SHEP),分别配制成2.5mg/mL溶液作为实验组,用培养基将低聚果糖(FOS)配置成相同糖含量溶液作为阳性对照组,以空白培养基组作为空白对照组(DSS)。在厌氧箱中将准备好的粪便匀浆接种液按10%的接种量分别接入到含有底物和培养基的溶液中,总体系为2mL进行体外发酵,充分混匀。分别在0h、24h和48h收集样品,每个样品3个平行。将发酵管放入培养箱振荡器培养(120rpm,37℃)。发酵管密封后上端插入注射器以计量发酵过程产气情况。
(4)样品总糖以及单糖含量测定、pH测定、产气测定、SCFAs测定、16S rDNA测序和数据处理与实施例3相同。
二、实验结果
1、体外发酵液产气和pH变化
如图22A所示,各组随着发酵时间的延长,产气量逐渐增多。DSS组和FOS组的产气量最多。发酵48h后,与DSS组相比,各多糖组产气量都下降,大概产气量在0.6~0.9mL之间。如图22B所示。发酵24h之后,DSS组的pH值与0h无明显变化,但在48h有明显下降。发酵48h之后,与DSS组相比,鸡枞菌多糖组pH值无显著差异,FOS组的pH值有明显降低。在整个发酵过程中,HWEP和HAEP组的pH值略微下降,在5.3~5.8之间,SHEP组的pH值无明显变化,pH值下降最少,水平基本保持在初始值附近。
2、体外发酵液中总糖和单糖组成变化
如图23A所示,发酵过程中鸡枞菌多糖组和FOS组的总糖均呈下降趋势,表明鸡枞菌多糖和FOS都能被微生物有效利用。三组鸡枞菌多糖(图23B、图23C和图23D)在0h~24h之间下降最多,在24h~48h多糖的利用减缓。其中HAEP和HWEP组的总糖下降率高于SHEP组。从单糖组成来看,HWEP、HAEP和SHEP组的Man、Gal和Glc的含量都随发酵时间的延长而不断下降。其中Glc下降最为明显,HWEP组、HAEP组和SHEP组均分别下降了初始糖含量的68.26%、51.53%和56.83%,显示微生物对Glc的高利用率;其次为Gal,分别下降了39.31%、20.83%和36.70%。
3、体外发酵液SCFAs变化
与DSS组相比,发酵24~48h后,鸡枞菌多糖组和FOS组的总SCFAs含量都降低,但无显著差异(p>0.05)(图24A),DSS组表现出最高的总SCFAs水平。从不同SCFAs的含量来看,除戊酸(图24G)外,发酵48h后HWEP组、HAEP组和SHEP组的乙酸(图24B)、丙酸(图24C)、丁酸(图24E)、异丁酸(图24D)和异戊酸(图24F)都达不到DSS组的产酸水平。此外,FOS组除丁酸(图24E)外,其他五种SCFAs的含量也比DSS组下降(p>0.05)。这与上文关于鸡枞菌多糖在正常人结肠体外发酵中的结果相反。说明在发酵液添加鸡枞菌多糖或FOS抑制了SCFAs的产生。推测是由于DSS组无碳源加入,其中的微生物菌群主要利用IBD小鼠盲肠内容物中原有的可溶性碳源进行代谢产生SCFAs。当鸡枞菌粗多糖和FOS加入后,微环境发生改变,而产酸微生物菌群对原本的微环境具有更好的适应性,导致鸡枞菌多糖组和FOS组的SCFAs含量低于DSS组。对0h的空白发酵液进行了单糖分析,发现0h发酵液中含有低浓度的Ara、Xyl、Rha、Glc和Gal,证实了以上猜想。
4、体外发酵液微生物分析
(1)Alpha多样性与Beta多样性分析
各组样品Good’s coverage值均接近1,微生物样本测序达到深度要求(图25A)。经48h体外发酵之后,各组的Chao1值都无明显变化,在体外发酵过程中,碳源变化不影响其群落丰富度(图25B)。发酵48h后,与DSS-48h组相比,HWEP、HAEP和SHEP组的shannon值均显著下降(p<0.05)(图25C),表明鸡枞菌多糖显著降低了群落多样性。通过Beta多样性分析,主坐标(PCoA)分析3D图中3个主成分分别解释了79.33%、12.18%、4.74%的微生物多样性的变化(图25D)。各组平行之间距离相近,表明各平行之间结果具有较强可信度。在发酵48h后,所有实验组都显示出与DSS-48h组相比距离较远,表明加入鸡枞菌粗多糖改变了IBD小鼠盲肠体外发酵液微生物群落构成。
2、鸡枞菌粗多糖在门水平对体外发酵液微生物的影响
如图26A,0h发酵液的优势菌群主要为Bacteroidota以及Firmicutes(DSS-0h)。由图26B~G所示,发酵48h后,空白底物组(DSS-48h)中Proteobacteria是优势门,其次是Firmicutes和Bacteroidota。鸡枞菌粗多糖发酵液中Firmicutes、Bacteroidota和Actinobacteriota的相对丰度相比DSS组都下调;HAEP和SHEP的Verrucomicrobiota的相对丰度增加。从图26G结果可见,发酵48h后,HWEP组和HAEP组的F/B值比DSS组下降(p>0.05)。
3、鸡枞菌粗多糖在属水平对体外发酵液微生物的影响
图27A是相对丰度排名前30的菌属。可见0h发酵液中占比最多的菌属为unclassified_Muribaculaceae、unclassified_Lachnospiraceae和Lachnospiraceae_NK4A136_group。图27B单独对各组48h发酵液优势菌属(TOP10)进行了分析统计。DSS-48h相比DSS-0h显著下调了unclassified_Muribaculaceae、unclassified_Lachnospiraceae、Alistipes、Rikenellaceae_RC9_gut_group和Lachnospiraceae_NK4A136_group的相对丰度(p<0.05),显著上调了Parasutterella、Enterobacter和Ligilactobacillus的相对丰度(p<0.05)。在IBD小鼠中DSS组的uncultured_bacterium_f_Muribaculaceae、Lactobacillus、Alistipes的相对丰度都相对DSS-0h组下降。显示在体外发酵过程中,IBD小鼠的肠道菌群的一些特定优劣势菌属在体外与体内有相似的变化趋势。
与DSS-48h组相比,鸡枞菌多糖组显著上调了Enterobacter的相对丰度(p<0.05),上调了Lachnospiraceae_NK4A136_group的相对丰度;下调了Ligilactobacillus、unclassified_Muribaculaceae、unclassified_Lachnospiraceae、Alistipes和Rikenellaceae_RC9_gut_group的相对丰度(p<0.05)。三组鸡枞菌粗多糖对肠道菌群的结构影响趋于一致。DSS-48h组与FOS组相比,Parasutterella、Enterobacter、unclassified_Muribaculaceae、Desulfovibrio和unclassified_Lachnospiraceae相对丰度显著减少(p<0.05),Ligilactobacillus、Alistipes、Bacteroides和Bifidobacterium的相对丰度显著增加(p<0.05)。
最后所应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范围的限制,对于本领域的普通技术人员来说,在上述说明及思路的基础上还可以做出其它不同形式的变化或变动,这里无需也无法对所有的实施方式予以穷举。凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,均应包含在本发明权利要求的保护范围之内。

1.一种鸡枞菌多糖,其特征在于,具有两个分子量分别为673.85 kDa和19.88 kDa的组分,其由Rha、Fuc、Ara、Xyl、Man、Gal和Glc组成,其摩尔比为0.59:6.71:0.16:1.41:4.22:22.14:64.77。
2.根据权利要求1所述的鸡枞菌多糖,其特征在于,其制备方法包括以下步骤:
S1:将鸡枞菌脱脂,得到脱脂后的鸡枞菌;
S2:混合步骤S1脱脂后的鸡枞菌和提取剂,其比例为1g:14.8~15.2 mL,搅拌混匀,固液分离,得粗提液;
S3:对步骤S2的粗提液进行醇沉,固液分离,得粗提沉淀;
S4:用8000~12000 Da透析袋透析步骤S3的粗提沉淀,得鸡枞菌多糖;

S3:对步骤S2的粗提液进行醇沉,固液分离,得粗提沉淀;
S4:用8000~12000 Da透析袋透析步骤S3的粗提沉淀,得鸡枞菌多糖;
所述提取剂为酸溶液。
5.一种鸡枞菌多糖,其特征在于,具有两个分子量分别为510.40 kDa和29.24 kDa的组分,其由Rha、Fuc、Ara、Xyl、Man、Gal和Glc组成,其摩尔比为0.65:1.96:0.20:1.54:0.93:5.12:89.60。
6.根据权利要求1所述的鸡枞菌多糖,其特征在于,其制备方法包括以下步骤:
S1:将鸡枞菌脱脂,得到脱脂后的鸡枞菌;
S2:混合步骤S1脱脂后的鸡枞菌和提取剂,其比例为1g:14.8~15.2 mL,搅拌混匀,固液分离,得粗提液;
S3:对步骤S2的粗提液进行醇沉,固液分离,得粗提沉淀;
S4:用8000~12000 Da透析袋透析步骤S3的粗提沉淀,得鸡枞菌多糖;
所述提取剂为碱溶液。
7.权利要求1到6任一所述枞菌多糖在制备治疗肠道炎症和/或肠道菌群失调的药物中的应用。
8.根据权利要求7所述的应用,其特征在于,所述肠道菌群为Bacteroidota、Enterobacter、Bacteroides、uncultured_bacterium_f_Muribaculaceae、Lactobacillus、Bifidobacterium、Verrucomicrobiota、Lachnospiraceae_NK4A136_group、Firmicutes、Actinobacteriota、uncultured_bacterium_f_Lachnospiraceae、Ligilactobacillu、unclassified_Muribaculaceae、Rikenellaceae_RC9_gut_group、Proteobacteria、Actinobacteria、Bacteroidota和/或unclassified_Lachnospiraceae。
9.根据权利要求7所述的应用,其特征在于,所述治疗肠道菌群失调的药物为升高Bacteroidota、Enterobacter、Bacteroides、uncultured_bacterium_f_Muribaculaceae、Lactobacillus、Bifidobacterium、Verrucomicrobiota、和/或Lachnospiraceae_NK4A136_ group的丰度,和/或降低Firmicutes、Actinobacteriota、uncultured_bacterium_f_ Lachnospiraceae、Ligilactobacillus、unclassified_Muribaculaceae、Rikenellaceae_ RC9_gut_group、Proteobacteria、Actinobacteria、Bacteroidota和/或unclassified_ Lachnospiraceae的丰度的药物。
10.根据权利要求7所述的应用,其特征在于,所述治疗肠道炎症的药物为治疗溃疡性肠炎的药物。
11.根据权利要求10所述的应用,其特征在于,所述治疗肠道炎症的药物为治疗稀便、腹泻、便血、结肠长度减少、肿胀和/或出血的药物。
12.根据权利要求10所述的应用,其特征在于,所述治疗肠道炎症的药物为治疗结肠炎症细胞浸润、肠黏膜粘蛋白减少、肠道紧密连接蛋白表达升高、肠道促炎症细胞因子表达升高、肠道杯状细胞数量减少和/或肠道短链脂肪酸减少的药物。
13.根据权利要求12所述的应用,其特征在于,所述炎症细胞为中性粒细胞和/或单核细胞,所述促炎症细胞因子为IL-6、IL-1β和/或TNF-α,所述紧密连接蛋白为Claudin-1、Claudin-2和/或Occludin,所述短链脂肪酸为乙酸、丙酸、异丁酸、丁酸、异戊酸和/或戊酸。
