Abstract
The invention belongs to the technical field of traditional Chinese medicines, and in particular relates to a composition for regulating uric acid to treat gout and a preparation method thereof. The composition is prepared from the following raw materials, by weight, 10-20 parts of coix seed, 10-20 parts of liquorice, 10-20 parts of radix puerariae, 10-20 parts of lily, 10-20 parts of chicory, 10-20 parts of mulberry leaf, 10-20 parts of cape jasmine, 10-20 parts of poria cocos and 10-20 parts of dandelion. The invention selects organic mixed solvent for extraction, and the obtained organic solvent extract has much stronger uric acid reducing effect than the extract obtained by adopting conventional ethanol and water extraction. Experiments prove that the composition has stronger uric acid reducing effect; compared with the common uric acid reducing medicines, the composition provided by the invention can not cause liver and kidney injury, and has milder uric acid reducing effect and safer effect.

Example 1
1) The preparation method comprises the following steps of: 18 parts of coix seed, 15 parts of liquorice, 15 parts of kudzuvine root, 16 parts of lily, 18 parts of chicory, 13 parts of mulberry leaf, 13 parts of cape jasmine, 15 parts of poria cocos and 18 parts of dandelion, and cleaning;
2) Pulverizing Coicis semen, glycyrrhrizae radix, radix Puerariae, bulbus Lilii, herba Cichorii, folium Mori, fructus Gardeniae, poria, and herba Taraxaci with pulverizer, and making granule diameter smaller than 1cm to obtain mixture;

1) The preparation method comprises the following steps of: 20 parts of coix seed, 10 parts of liquorice, 14 parts of kudzuvine root, 12 parts of lily, 15 parts of chicory, 18 parts of mulberry leaf, 16 parts of gardenia, 20 parts of poria cocos and 20 parts of dandelion;

Comparative example 1
The extraction in step 3) was performed by directly selecting an ethanol solvent, and the rest was the same as in example 3.
Comparative example 2
The extraction in step 3) was directly carried out by steam extraction with water, the remainder being as in example 3.
Comparative example 3
Mixing the organic mixed solvent in the step 3), and mixing n-butanol and ethyl acetate according to a volume ratio of 10:1, and the rest is the same as in example 3.
The uric acid lowering effect was evaluated for the compositions of examples 1 to 4 and comparative examples 1 to 3.
Taking male Kunming mice with the weight of 20+/-2 g, and continuously perfusing the stomach with the suspension of the potassium oxazinate for 10 days under the condition of guaranteeing normal diet and drinking water to establish a hyperuricemia animal model of the mice.
Mice with uric acid content of 160-165 mu mol/L in serum are selected as high uric acid model mice and are used as experimental objects. 21 male hyperuricemia model mice are randomly divided into 7 groups of 3 mice; the extracts with uric acid reducing effect prepared in examples 1 to 4 and comparative examples 1 to 3 are respectively added with water to prepare suspension; then, the stomach-lavage administration is carried out to the mice in 1 to 7 groups of experimental groups according to the amount of 100 mg/kg.d; the model control group was perfused with an equal amount of distilled water. The administration is continued for 10 days; 1h after the end of the administration on day 10, taking blood of the mice, and measuring the uric acid content in serum of the mice; the test results are shown in Table 1.
Table 1 shows the results of uric acid lowering experiments with the extracts of the present invention.

As can be seen from Table 1, the examples 1 to 4 of the present invention obtained a good uric acid lowering effect. In contrast, the comparative examples 1 and 2 have limited uric acid reducing effect due to the selection of conventional ethanol and water solvents, while the comparative example 3 has a better effect due to the selection of organic solvents with different proportions, but is less remarkable than the example 3.
