技术领域
本发明涉及睡眠质量改善剂。
背景技术
在现代社会,由于压力、食物偏食、环境等因素,患失眠症或睡眠不足的人正在增加。据称睡眠不足也成为肥胖、高血压、抑郁症、糖尿病等的原因,消除睡眠不足很重要。近年来,不仅关注睡眠时间,还关注睡眠质量。
作为睡眠质量,包括入睡的好坏、睡眠的深度、清醒后心情的好坏等,特别是若白天频繁地感到睡意则集中力会下降,工作和学习的表现会下降,同时也可能引起工作中的事故等,所以防止非睡眠中的睡意很重要。
迄今为止,作为具有改善睡眠质量的作用的有效成分,例如报道有均二苯代乙烯系化合物和/或橄榄提取物(专利文献1)、呈现出植物玫瑰的花瓣香味的挥发性有机化合物(专利文献2)、3-羟基丁酸单体(专利文献3)、来自秋葵种子的酶解物的秋葵种子提取物(专利文献4)等。

发明内容
发明所要解决的课题
但是,上述有效成分很难说能够充分改善睡眠质量、如抑制非睡眠时的睡意等。因此,本发明的课题在于提供一种非睡眠时睡意改善等优异的睡眠质量改善剂。
用于解决课题的手段
本发明人为了发现非睡眠时睡意改善等睡眠质量改善效果优异的成分进行了各种研究,结果发现,库页冷杉的减压水蒸气蒸馏物的油性馏分具有优异的睡眠质量改善效果,如能够显著抑制非睡眠时的睡意等,从而完成了本发明。
即,本发明涉及一种睡眠质量改善剂,其特征在于,其含有库页冷杉的减压水蒸气蒸馏物油性馏分作为有效成分。
另外,本发明涉及一种睡眠质量改善方法,其通过挥发来应用上述睡眠质量改善剂。
发明效果
通过应用本发明的睡眠质量改善剂,能够迅速入睡,中途不醒来而能够长时间维持睡眠到早上,睡醒也能感到清爽和疲劳感的恢复。并且,由于能够抑制白天感到睡意,因此能够在工作等中维持集中力,能够稳定地实现高表现。

具体实施方式
本发明的睡眠质量改善剂以库页冷杉的减压水蒸气蒸馏物油性馏分作为有效成分。
作为原料的库页冷杉的部位没有特别限制,从被间伐、剪枝的树木的有效利用的方面、睡眠质量改善效果优异的方面出发,适合使用木质部和/或叶的部分。
本发明中使用的有效成分是对库页冷杉进行减压水蒸气蒸馏而得到的蒸馏物的油性馏分(精油)。该减压水蒸气蒸馏物油性馏分即使不花费浓缩、纯化等工夫,也以高浓度含有单萜类,因此具有清爽的香气,香气的嗜好性高,而且睡眠质量改善效果也优异。
需要说明的是,蒸馏时的加热也可以是利用一般的加热器等进行的加热,但从加热效率的方面出发,优选利用微波进行的加热,特别优选不加水而照射微波进行加热。通过照射微波,植物中包含的水分子被直接加热而产生水蒸气,其作为移动相发挥作用而使植物中的挥发性成分被蒸馏,因此该蒸馏方法被认为包含基于挥发性成分的沸点的减压蒸馏的要素和水蒸气蒸馏的要素。
这种基于微波照射的蒸馏物例如可以通过使用图1所示的装置来得到。图中,1表示微波蒸馏装置,2表示蒸馏槽,3表示微波加热装置,4表示搅拌叶片,5表示气流流入管,6表示蒸馏物流出管,7表示冷却装置,8表示加热控制装置,9表示减压泵,10表示压力调节阀,11表示压力控制装置,12表示蒸馏对象,13表示蒸馏物。
该装置1中,将成为蒸馏对象12的原料库页冷杉放入蒸馏槽2中,一边用搅拌叶片4搅拌,一边从设置于蒸馏槽2的上表面的微波加热装置3放射微波,加热原料。该蒸馏槽2与气流流入管5和蒸馏物流出管6连通。气流流入管5将空气或氮气等惰性气体导入蒸馏槽2中,该气流从蒸馏槽2的下部导入。另外,蒸馏物流出管6将来自原料的蒸馏物从蒸馏槽2的上部导出到外部。
上述蒸馏槽2内部通过安装在其上的温度传感器和压力传感器(均未图示)测定温度和压力,分别通过加热控制装置8和压力控制装置11、压力调节阀10进行调整。
另外,经由蒸馏物流出管6从蒸馏槽2流出的气体状的蒸馏物通过冷却装置7变为液体,作为蒸馏物13获得。该蒸馏物13中包含油性馏分13a和水性馏分13b,其中将油性馏分13a分取作为有效成分使用。
在蒸馏时,将上述蒸馏槽2内的压力设为3~95千帕、优选设为3~40千帕、进一步优选设为3~20千帕的程度进行即可,此时的蒸气温度为40℃~100℃。在压力为3千帕以下时,植物中的挥发性成分的蒸气压上升被抑制,另外,相较于水蒸气蒸馏的要素,基于各成分的沸点的减压蒸馏的要素为主,从沸点低的物质依次流出,因此从不能有效进行沸点高于水的成分的提取这点、以及大量提取回避效率低且香气嗜好性差的高沸点成分这点出发,有时不优选。另外,在95千帕以上时,由于原料的温度升高,能量损耗大,原料的氧化也被促进,从这点出发,有时不优选。
另外,蒸馏时间为0.2~8小时的程度、优选为0.4~6小时的程度即可。在0.2小时以下时,植物中的未提取成分大量残留,在8小时以上时,原料为接近干固的状态,因此提取效率有时降低。
进而,作为导入蒸馏槽2内的气体,可以为空气,但优选氮气、氦气、氩气等惰性气体,作为其流量,每分钟的流量为蒸馏槽2的0.001~0.1容量倍的程度即可。
作为本发明的睡眠质量改善剂中的有效成分,也可以直接使用如上所述得到的库页冷杉的减压水蒸气蒸馏物油性馏分,根据需要也可以通过常规方法进一步浓缩或纯化而使用。
本发明的睡眠质量改善剂的剂型没有特别限定,例如可以为液剂、凝胶剂、固体制剂、泵式喷雾、气溶胶等剂型。
在作为液剂使用的情况下,本发明的睡眠质量改善剂可以直接使用库页冷杉的减压水蒸气蒸馏物油性馏分,也可以用适当的溶剂稀释。从睡眠质量改善效果和经济性的方面出发,用溶剂稀释时的库页冷杉的减压水蒸气蒸馏物油性馏分的含量优选为0.1~30质量%、更优选为0.5~10质量%、进一步优选为1.0~5质量%。
作为溶剂的示例,可以举出例如水、甲醇、乙醇、异丙醇、异丁醇、仲丁醇、叔丁醇、甲基溶纤剂、乙基溶纤剂、甘油等醇类;乙二醇二甲醚、3-甲氧基-1-丁醇、3-甲氧基-3-甲基-1-丁醇、1,4-二噁烷、四氢呋喃等醚类;丙酮、甲基乙基酮等酮类;N,N-二甲基甲酰胺、N,N-二甲基乙酰胺等酰胺类、二甲基亚砜等。它们可以单独使用或将2种以上组合使用。
为了使库页冷杉的减压水蒸气蒸馏物油性馏分溶解在上述溶剂中,也可以使用现有公知的阴离子系表面活性剂、阳离子系表面活性剂、非离子系表面活性剂或两性表面活性剂。
在上述表面活性剂中,作为阴离子系表面活性剂,可以使用例如高级脂肪酸皂、皂用坯料、金属皂、N-酰基-L-谷氨酸三乙醇胺、N-酰基-L-谷氨酸钠、烷基硫酸钠、烷基磺酸钠、聚氧乙烯烷基醚硫酸钠、聚氧乙烯烷基醚磷酸、聚氧乙烯烷基苯基醚磷酸、椰子油脂肪酸甲基牛磺酸钠(N-椰油酰基-N-甲基牛磺酸钠)、月桂基硫酸三乙醇胺、月桂基硫酸钠、月桂酰肌氨酸钠、月桂酰甲基β-丙氨酸钠液、月桂酰甲基牛磺酸钠等中的1种或将2种以上混合使用。
另外,作为阳离子系表面活性剂,可以使用乙基硫酸羊毛脂脂肪酸氨基丙基乙基二甲基铵、烷基三甲基氯化铵、二烷基二甲基氯化铵、二硬脂基二甲基氯化铵、硬脂基二甲基苄基氯化铵、硬脂基三甲基氯化铵、苯扎氯铵、苄索氯铵等中的1种或将2种以上混合使用。
进而,作为非离子系表面活性剂,可以举出聚氧乙烯烷基醚、聚氧化烯烷基醚、聚氧乙烯氢化蓖麻油醚、聚氧乙烯脂肪酸酯、聚氧乙烯烷基苯基醚、山梨聚糖脂肪酸酯、聚氧乙烯山梨聚糖脂肪酸酯、蔗糖脂肪酸酯、甘油脂肪酸酯、聚氧乙烯聚氧丙烯嵌段聚合物、聚氧乙烯烷基胺醚、脂肪酸烷醇酰胺、叔胺氧化物等。该聚氧乙烯烷基醚的聚氧乙烯链为3~18、优选为7~12,烷基链可以为直链或支链,烷基链长为8~22、优选为12~14。另外,上述脂肪酸烷醇酰胺可以举出椰子油脂肪酸、硬脂酸、月桂酸的单乙醇酰胺、二乙醇酰胺等,作为叔胺氧化物,可以举出月桂基二甲基氧化胺、椰子油脂肪酸二甲基氧化胺、月桂酰氨基丙基二甲基氧化胺、辛基二甲基氧化胺、肉豆蔻基二甲基氧化胺等。
本发明的睡眠质量改善剂中也可以添加其他香料成分。作为香料成分,可以举出例如l-薄荷醇、dl-樟脑、乙酸冰片酯、龙脑、母菊兰烯、4-萜烯醇、香叶醇、香茅醛、香叶醛、橙花醛、扁柏醇、薄荷油、桉树油、丝柏油、肉桂油、香柠檬油、松节油、玫瑰油、茴香、鼠尾草、杉、扁柏、桉树等的精油等。
在制成凝胶剂的情况下,可以使用现有公知的凝胶化剂,可以举出例如角叉菜胶、结冷胶、琼脂、明胶、瓜尔胶、果胶、刺槐豆胶、黄原胶、藻酸钠、纤维素衍生物、二亚苄基-D-山梨糖醇、羟丙基化多糖类、硬脂酰菊粉、丙烯酸钠等高吸水性树脂、辛酸铝等,可以使用它们中的1种或将2种以上混合使用。
在制成固体制剂的情况下,优选使用小烛树蜡、蜂蜡、纯白地蜡、微晶蜡、合成烃蜡和乙烯-丙烯共聚物等蜡剂;橄榄油、霍霍巴油、澳洲坚果油等植物油;异壬酸异十三烷基酯、异硬脂酸辛基十二烷基酯、苹果酸二异硬脂酯等脂肪酸酯;二辛酸乙二醇酯、二油酸乙二醇酯、二油酸丙二醇酯、二癸酸丙二醇酯、二(辛酸/癸酸)丙二醇酯、二辛酸新戊二醇酯、二癸酸新戊二醇酯等脂肪酸二醇酯;二异硬脂酸甘油酯、三异硬脂酸甘油酯等脂肪酸甘油酯、椰子油烷基酯、辛基十二醇等高级脂肪酸醇、液体石蜡、液体异链烷烃、聚丁烯、聚异丁烯、脂肪酸聚甘油酯、二聚亚油酸酯、松香、松香衍生物、异硬脂酸氢化蓖麻油、(PPG-7/琥珀酸)共聚物、(己基癸酸/癸二酸)双甘油低聚酯、烯烃低聚物等制成油性固体制剂。其形状没有特别限定,但优选可涂布在应用对象上来应用的棒状。
作为其他任选成分的示例,可示例出增稠剂、紫外线吸收剂、抗氧化剂、除臭剂等。
本发明的睡眠质量改善剂优选通过挥发或喷雾到空间中来应用。通过这些方法,从鼻腔等吸入有效成分,发挥其效果。作为使液剂剂型的睡眠质量改善剂挥发的方法,可以举出:直接或在浸渍到适当浸渍体中的状态下使其挥发的方法;将液剂吸起并由挥发体使其挥发的方法;利用藤条香熏的方法;等。此外,也可以使用:使用常温挥发装置、利用风扇等的送风挥发装置、利用火或热源等的加热挥发装置等适当的挥发装置使其挥发的方法;使用泵式喷雾器、气溶胶、超声波振子、加压液喷雾器、加压空气雾化喷雾装置等进行喷雾的方法。作为凝胶剂或固体制剂的使用方法,可以举出直接静置使其挥发,或者涂布到枕头等寝具或睡衣的衣领等上,使其从该涂布部挥发的方法等。这些方法中,从由鼻腔等吸入的效率高、经济性及效果的持续性、效果的稳定性的方面出发,优选通过涂布到枕头等寝具上的方法使其挥发。在将本发明的睡眠质量改善剂涂布或喷雾到寝具等的情况下,例如每1m2以库页冷杉的减压水蒸气蒸馏物油性馏分计应用20~200mg的程度即可。另外,在向空间中喷雾的情况下,每1m3以库页冷杉的减压水蒸气蒸馏物油性馏分计应用1~10mg的程度即可。
另外,本发明的睡眠质量改善剂也可以作为外用剂来应用。作为外用剂,可以举出霜剂、外用液剂、湿布剂、软膏剂、洗液剂、走珠等,可以使用通常混配在外用剂中的任意成分按照常规方法来制备。用量等没有特别限制,可以根据年龄、体重等适当设定,例如对于成人每次涂布以库页冷杉的减压水蒸气蒸馏物油性馏分计为10~300mg的程度的量的外用剂等即可,该外用剂含有1~10质量%的程度的库页冷杉的减压水蒸气蒸馏物油性馏分。
这样,通过从鼻腔等吸入有效成分或经皮吸收,发挥出改善睡眠质量的效果。作为睡眠质量的改善,可包括入睡潜伏期(从清醒状态到进入睡眠的所需时间)的缩短、防止半夜醒来的中途清醒抑止、消除清醒时的疲劳感的疲劳恢复改善、维持睡眠状态的睡眠时间增加、消除清醒时感觉到的睡眠时间不足的睡眠时间不足感改善、抑制在非睡眠中感到睡意的非睡眠时睡意改善等。特别是若在白天引发睡意则集中力降低,工作等的表现降低,同时还有可能引起工作中的事故等,因此防止非睡眠时的睡意很重要。
实施例
下面,举出实施例、制造例等来说明本发明,但本发明不受这些实施例等的任何限制。
制造例1
用压碎式粉碎机(KYB制作所制;图1)粉碎库页冷杉的叶,将其约50kg投入微波水蒸气蒸馏装置的蒸馏槽中。一边搅拌一边将蒸馏槽内的压力保持在约20KPa的减压条件下(蒸汽温度约67℃),照射微波1小时而得到蒸馏物,分取其油性馏分(以下称为“库页冷杉精油”)。得到的库页冷杉精油的量为180mL,相对于投入试样的比例为0.34%。
实施例1
利用下述表1所示的配方和下述制造方法制作棒状的油性固体制剂。

(制造方法)
将制造例1的库页冷杉精油以外的全部成分混合、加热熔解后,添加制造例1的库页冷杉精油并混合,浇注到模具中进行冷却,成型为棒状。如图3所示,将该棒状油性固体制剂(22)安装到供给容器(23)上,制成棒状制剂(21)。
试验例1
睡眠质量改善试验(1):
使用上述实施例1中制造的棒状制剂,对睡眠质量改善效果进行了试验。
(试验方法)
对象:入睡困难的老年人(60岁以上)
选定条件:
·没有治疗中的睡眠障碍
·日常没有服用安眠药
·每天几乎在同一时刻就寝
·匹兹堡睡眠质量问卷得分6-8分的人
·没有嗅觉障碍的人
以满足以上条件的7名男性、4名女性共计11名为对象实施了试验。
对于对象的枕头,将实施例1中制造的棒状制剂1g(精油成分10mg)涂布到1张面积为7cm2的贴纸的贴附面的相反面上,将其在枕头上贴附2张。使用该库页冷杉精油处理枕,凌晨0点左右就寝,第二天起床后上午7点到8点之间实施基于OSA睡眠调查表(山本由华吏、田中秀树、高濑美纪、山崎胜男、阿住一雄、白川修一郎:以中老年人、老年人为对象的OSA睡眠感调查表(MA版)的开发和标准化.脑和精神医学10:401-409,1999)的问卷调查。作为对照,使用未贴附涂布有实施例1中制造的棒状制剂的贴纸的枕头进行了同样的试验。实验间隔设置了2天以上的间隔。
(非睡眠时的睡眠自我检查评价)
对于对象的提问回答适用下述表2的数值,按每个提问项目计算出所有对象的平均值。结果一并示于表2。并且,基于表2的结果,对各心理标准中的因子进行评价。结果示于表3。
(香气的印象(好恶·强度等)评价)
利用VAS(Visual Analogue Scale,直观模拟标度尺),对于“愉快不愉快”、“平静”、“强度”的各评价项目,将左端设为0、右端设为100,示出线段上的位置,作为其平均值求出VAS测定值。结果示于表4。

由表2、3表明,通过将库页冷杉精油应用于枕头,在“非睡眠时睡意”、“入睡和睡眠维持”、“疲劳恢复”和“睡眠时间”的各因子方面可改善睡眠质量。通过改善“入睡和和睡眠维持”,能够实现“入睡潜伏期”的缩短和“中途清醒”的抑止。另外,如表4所示,确认到库页冷杉精油的香气的嗜好性优异。
试验例2
睡眠质量改善试验(2):
对于16名试验例1的对象,将实施例1中制造的棒状制剂1g(精油成分10mg)涂布到1张面积为7cm2的贴纸的贴附面的相反面上,将其在枕头上贴附2张。在使用该库页冷杉精油处理枕就寝的状态下,利用Alice PDx(Phillips公司)实施了多导睡眠监测。根据文献(野田明子、古池保雄:整夜多导睡眠、生体医工学46(2):134-143,2008)的记载,进行了基于多导睡眠的清醒、入睡等睡眠阶段的判定。通过下述式分

别求出总睡眠时间、睡眠效率、入睡时间、中途清醒时间的平均值。另外,作为对照,使用未贴附涂布有实施例1中制造的棒状制剂的贴纸的枕头实施了同样的试验。结果示于表5。

由表5确认到,通过将库页冷杉精油应用于枕头,与对照相比,总睡眠时间变长,睡眠效率也得到改善。另外,由于到入睡为止的时间变短,中途清醒时间也变短,因此确认到睡眠质量综合性地显著改善。
制剂例1
在90质量%的二丙二醇中混配10质量%的制造例1的库页冷杉精油,制造空间喷雾用睡眠质量改善剂。将所得到的空间喷雾用睡眠质量改善剂用超声波雾化装置(MikuniCorporation制造)在卧室内进行喷雾,结果能够良好地进入睡眠,中途不醒来而能够长时间维持睡眠到早上,睡醒也清爽,能够改善睡眠质量。
制剂例2
在50质量%的3-甲氧基-3-甲基-1-丁醇中混配50质量%的制造例1的库页冷杉精油,制造空间喷雾用睡眠质量改善剂。使用加热蒸发装置(S.T.CORPORATION制除臭插头)使该睡眠质量改善剂在卧室蒸发,结果能够良好地进入睡眠,中途不醒来而能够长时间维持睡眠到早上,睡醒也清爽,能够改善睡眠质量。
制剂例3
将2质量%的制造例1的库页冷杉精油用7质量%的表面活性剂(聚氧乙烯烷基醚)溶解于水中,制造空间喷雾用睡眠质量改善剂。使用市售的泵式喷雾器将该睡眠质量改善剂在空间中进行喷雾而进入睡眠,结果能够良好地进入睡眠。
制剂例4
使0.1质量%的制造例1的库页冷杉精油分散于99.9质量%的水中,制造空间喷雾用睡眠质量改善剂。使用超声波雾化装置(Echo Tech株式会社制)将该睡眠质量改善剂在卧室进行喷雾,结果能够良好地进入睡眠,中途不醒来而能够长时间维持睡眠到早上,睡醒也清爽,能够改善睡眠质量。
制剂例5
使作为凝胶化剂的3gκ-角叉菜胶分散到制造例1的库页冷杉精油3.0g、丙二醇10g和水84g的混合物中,加热至约60℃而分散后,填充到上面开放的杯型容器中,冷却固化,制造凝胶状的空间挥发用睡眠质量改善剂。将其设置于卧室的枕边,一边使其挥发一边进入睡眠,结果能够良好地进入睡眠,中途不醒来而能够长时间维持睡眠到早上,睡醒也清爽,能够改善睡眠质量。
制剂例6
将2质量%的制造例1的库页冷杉精油用5质量%的表面活性剂(聚氧乙烯烷基醚)溶解于水中,制造空间挥发睡眠质量改善剂。将该睡眠质量改善剂收纳于房间的消臭力(注册商标、S.T.CORPORATION制)的容器中,一边使其在卧室的枕边挥发一边进入睡眠,结果能够良好地进入睡眠,中途不醒来而能够长时间维持睡眠到早上,睡醒也清爽,能够改善睡眠质量。
制剂例7
将制造例1的库页冷杉精油填充到带定量阀的气溶胶容器中,设置于自動でシュパッと(注册商标、S.T.CORPORATION制),每隔90分钟在卧室内进行喷雾而进入睡眠,结果能够良好地进入睡眠,中途不醒来而能够长时间维持睡眠到早上,睡醒也清爽,能够改善睡眠质量。
工业实用性
本发明的睡眠质量改善剂具有优异的睡眠质量改善效果,如能够抑制非睡眠时的睡意等,安全性也优异,因此是有用的。

Claims (7)
1.一种睡眠质量改善剂,其特征在于,其含有库页冷杉的减压水蒸气蒸馏物油性馏分作为有效成分。
2.如权利要求1所述的睡眠质量改善剂,其使用库页冷杉的木质部和/或叶。
3.如权利要求1或2所述的睡眠质量改善剂,其中,睡眠质量改善剂为选自由非睡眠时睡意改善剂、入睡潜伏期缩短剂、中途清醒抑止剂、疲劳恢复感改善剂和睡眠时间不足感改善剂组成的组中的1种以上。
4.如权利要求3所述的睡眠质量改善剂,其中,睡眠质量改善剂为非睡眠时睡意改善剂。
5.如权利要求1~4中任一项所述的睡眠质量改善剂,其为棒状油性固体制剂。
6.一种睡眠质量改善方法,其通过挥发来应用权利要求1~5中任一项所述的睡眠质量改善剂。
7.一种睡眠质量改善方法,其通过从入睡时开始在睡眠中持续挥发来应用权利要求1~5中任一项所述的睡眠质量改善剂。
