Description
Technical Field
The invention relates to the technical field of skin care products, in particular to a centella asiatica compound with a safety acne removing effect before puberty.
Background
The intrinsic cause of acne in prepubertal skin is mainly due to the massive secretion of sebum, abnormal keratinization of the pilo-sebaceous duct orifice, propionibacterium acnes colonization, immune inflammatory response and genetic correlation; the external factors are mainly caused by factors such as emotion, diet, improper use of skin care products, sun drying and the like. The symptoms are mainly acne, the early stage related to microorganisms is mainly caused by streptococcus and the middle and later stage is mainly caused by propionibacterium acnes, and the treatment scheme can use anti-acne skin care products with keratolytic or exfoliating functions, anti-inflammatory and antibacterial anti-acne skin care products and other treatment means through external and oral antibacterial drugs.
Madecassoside (MC) is the highest content pentacyclic triterpene compound of centella asiatica, and has the formula C 48 H 78 O 20 The appearance shape of the polymer was white crystals with respect to the molecular mass 975.13. Madecassoside is the secondary plant metabolite of saponins synthesized by centella asiatica by the isoprene pathway. Studies show that madecassoside plays a very good role in moisturizing, resisting oxidation, promoting wound healing and the like in solving skin problems.

3. Cell oil control efficacy test
The method comprises the following steps: human sebaceous gland cell oil control efficacy test method
The method comprises the following steps: research on active ingredient separation and inhibition of human sebaceous gland cell androgen synthesis mechanism of coptis chinensis
System and sample size:
test system: human sebaceous gland cells (SZ 95).
Cell algebra: at least 2 generations.
Cell mass per well: 1X 105 cells/well.
The principle method comprises the following steps: sebaceous glands are the source and target tissues of androgens, in which dehydroepiandrosterone is produced, which are converted, catalyzed by a series of enzymes, to the most active testosterone and 5α -dihydrotestosterone, which stimulate the synthesis of lipids and the proliferation and differentiation of sebaceous gland cells. In this proliferation and differentiation process, undifferentiated naive cells first proliferate, and part of the daughter cells form lipid droplets and move toward the center of glandular cells, gradually developing and differentiating into mature sebaceous gland cells capable of accumulating and secreting lipids. The experiment detects the amount of grease secreted by sebaceous gland cells by a nile red staining method, so as to judge whether a sample has the oil control effect.
The experimental steps are as follows:
1. cells were seeded in six well plates (1×105 cells/well), incubated at 37 ℃ with 5% CO2 for 24h.
2. After the incubation, the normal control group is replaced by fresh medium, the model control group contains testosterone medium, the sample control group contains sample with corresponding concentration and testosterone medium, and the sample control group is incubated for 24 hours at 37 ℃ with 5% CO 2.
3. After the incubation, the medium was removed, pre-chilled D-Hank's washed 3 times, fixed with 60% isopropyl alcohol, D-Hank's washed, nile red stained, photographed with a fluorescence microscope, and Image J analyzed the fluorescence intensity of each group.
A: fluorescence intensity
Applicability and limitations:
the method is suitable for testing the oil control efficacy of cosmetics and raw materials thereof, and requires that a sample can be dissolved in water or prepared into a suspension which can be uniformly dispersed in water.
Judging the basis:
statistical analysis p <0.05, judged significant differences.
Example, sample number complex plus 4 (3-1.0%) formulation addition concentration 1.0%
Test results:
a typical graph of the oil control efficacy of this sample is shown in fig. 7 below.
The fluorescence intensity of the sample numbered complex plus 4 (3-1.0%) was observed to be significantly reduced compared to the model control group, revealing that the sample had oil control efficacy.
Conclusion:
under the experimental conditions, the sample complex plus 4 has the oil control effect.
