Abstract

本发明背景技术
本发明涉及制备口香糖的方法。更具体地说,本发明涉及含有效量活性药物的口香糖的制备方法。优选,将该活性药物添加至口香糖涂层,以改善其从口香糖的释放速率,并通过使用亲水胶基组合物改善其释放。
近年来,已经作出了许多努力用以控制口香糖中的多种成分的释放特性。最为特别的方面在于,已经对各种口香糖制品中的甜味剂和风味剂的延迟释放进行了尝试,从而延长了令人满意的口香糖咀嚼时间。甜味剂和风味剂释放的延迟,也避免了在初始的咀嚼期间,发生令人不快的甜味剂或风味剂的突然性的大规模涌现。在另一方面,一些成分已经被处理,这样就提高了其在口香糖中的释放速率。

本发明提供了用于向人体输送药物或活性剂的改善方法。为此,提供了被涂敷的口香糖制品,其中包括了药物或活性剂。该药物或活性剂存在于口香糖组合物的涂层中。通过向口香糖涂层中添加活性剂,结果发现该药物或活性剂从口香糖中被快速释放进唾液中。连续地咀嚼该口香糖可以在口腔前庭制造压力,并可以迫使药物或活性剂通过口腔中的口腔粘膜直接进入人体的体循环。然而,这样的亲脂活性剂可以被部分地束缚在口香糖胶基和口香糖基质中,并不能被完全释放。
在一方面,本发明提供了一种亲水胶基,包括:
a)约20%至约90%的亲水聚合物;
b)约5%至约35%的亲水软化剂/乳化剂;和
c)约4%至约50%填料;
d)该口香糖胶基并没有疏水聚合物、弹性体溶剂、蜡、和疏水软化剂。
本发明还提供了改善了的口香糖制品,其中在口香糖涂层和亲水胶基组合物中包括了药物和活性剂。
本发明中,亲脂活性剂也包括在口香糖芯中。

为此,本发明提供了一种药物输送方法,包括以下步骤:提供一种亲水口香糖芯和一种口香糖涂层,其中在口香糖涂层中包含有药物;且咀嚼该口香糖以使药物从口香糖涂层中释放进咀嚼者的口腔中。
该活性药物可以是任何亲脂的制剂,并且在传统上就作为药物,且通过口腔使其被摄入。这样的活性剂可以是维生素、癌症化疗药、抗真菌药、口服避孕药、镇痛剂、抗酸剂、骨骼肌松弛药、抗组胺药、减充血剂、抗菌剂、麻醉剂、镇咳剂、利尿剂、消炎药、抗生素、AIDS治疗药、神经病药物、抗病毒药、精神治疗剂、治疗糖尿病药、心血管治疗药、营养药品和营养添加剂。
因此,本发明实施方案的一个优点是提供了向人体输送亲脂药物或活性剂的新方法。
而且,本发明实施方案的一个优点是提供了一种人体服用亲脂药物或制剂的方法,服用量比传统的口服量低,但仍能实现同样的效果。
另外,本发明实施方案的一个优点是提供了一种服用亲脂药的方法,该方法比目前的方法感觉更可口。
还有,本发明实施方案的一个优点是提供了一种输送药的改善方法。
优选实施方案的详细描述
本发明提供用于向人体输送药物和其它活性剂的改善方法,以及改善的胶基和口香糖制品。如本文所使用的,术语口香糖也包括泡泡糖等。根据本发明,药物或活性剂被包含在口香糖制品的涂层中,与现有的一些这样的制品形成对照,这些现有的制品中药物或活性剂被直接包含在口香糖组合物中。
因此,随着口香糖被咀嚼,活性剂被更快地释放进唾液中。在持续不断的咀嚼期间,由于口香糖制造的压力迫使唾液中的药物或活性剂接着就通过了口腔中的口腔粘膜。
当将活性剂例如,维生素E添加进口香糖涂层时,活性剂具有提高的水分散能力,并从口香糖涂层中快速地释放进口中。根据活性剂,这些活性剂一般是非水溶的但却是油溶性的或亲脂的,将该活性剂添加至口香糖涂层,则会提高活性剂从口香糖中的释放。
其它的制剂或药物可以包括在本发明中。本发明的术语“活性剂”指的是一旦摄取和/或代谢具有所需的治疗或生理上的效果的化合物。该治疗效果可以是降低了生物体内异物或其它肠内菌群或动物群的生长,使疾病状况减轻(例如,疼痛、酸回流或其它不适减轻),对确定心情和行为的脑化学或分子有影响,或有益于营养的提高。当然这些仅仅是说明治疗效果的例子。本领域普通技术人员将能容易地识别具体的制剂具有的治疗效果或与给定的治疗效果有关。
该活性剂可以是任何在传统上就作为药物的制剂,且通过口腔使其被摄入。这样的活性剂可以是维生素、癌症化疗药、抗真菌药、口服避孕药、镇痛剂、抗酸剂、骨骼肌松弛药、抗组胺药、减充血剂、麻醉剂、镇咳剂、利尿剂、消炎药、抗生素、抗病毒药、精神治疗剂、治疗糖尿病药、心血管治疗药、生物工程药、营养药品和营养添加剂。使用本发明可以被输送的具体的维生素包括,且主要限于脂溶维生素例如,维生素B1、核黄素、吡哆辛、泛酸、胆碱、肉毒硷、维生素D和其类似物、维生素A和类胡萝卜素、视黄酸、维生素E和维生素K。
可以配制成合适口香糖制品的组合物描述于例如,US5858423、US5858413、US5858412和US5858383中。另外,Goodman和Gilman的“治疗的药物基础(The Pharmaceutical Basis of Therapeutics)”(Hardman等人编辑,McGraw Hill,NY出版)提供了有用的药物和其作用机理的全面指导。含药物的口香糖在输送制剂例如,尼古丁方面特别有效,描述于例如US5866179、US5889028、US5846557中,其中描述的一般的口香糖组合物含咳嗽抑制剂。这些专利在此引入作为参考,提供了将药物制剂混入口嚼制品的教导。应该理解本发明的口香糖制品和涂层并不限于上述所列的制剂,甚至于可以是任何亲脂药用的或其它活性制剂,这些制剂可以被配制进口香糖涂层使其摄取,并用于本发明。
营养药品和营养添加剂也可以被添加进入口香糖以及口香糖涂层作为活性剂。其中是亲脂草药和植物性药材,包括但不仅限于辣椒、洋甘菊、猫爪木、紫锥花、大蒜、姜、银杏、各种人参、绿茶、北美黄连、醉椒、荨麻、西番莲、沙巴棕、小连翘属植物和缬草。其它也可以被添加至口香糖涂层作为活性剂的营养药品是安息香、氨基葡萄糖、葡萄籽提取物、瓜拉那、磷脂酰丝氨酸、植物甾醇、植物化学制剂、异黄酮、卵磷脂、番茄红素和多元酚以及失重剂例如,去甲麻黄硷。
口香糖制品和涂层中药物和制剂的含量是这样进行选择的,当咀嚼口香糖时,唾液中应含有足够高浓度的药物或制剂。
根据本发明,根据制剂或药物的不同,对剂量的控制就会是不同的。例如,如果药物是镇痛剂,则口香糖产品则会以“所需”为基础服用。当然,类似于口服镇痛剂,对咀嚼的口香糖块的数量就会有限制,例如经常是不超过每四小时一小丸和每天不超过四至五次。如果该制剂是维生素,例如用维生素E来增强机能或其它有益的营养,则该口香糖产品将根据需要咀嚼。
现在说到口香糖,根据本发明的口香糖可以基于多种公知的不同的口香糖。例如,该口香糖可以是低或高水分含量的、有糖或无糖的,低卡路里(通过高胶基或低卡路里填充剂)的和/或可以含有牙制剂。
亲脂活性剂可以与甜味剂一起被添加进口香糖涂层,更具体地说是高效甜味剂例如祝马丁、二氢查耳酮、双氧恶噻嗪钾、阿斯巴甜、N-取代APM衍生物例如新甜(neotame)、氯化蔗糖衍生物、缩二氨酸基酰胺、糖精、环己氨磺酸盐。这些甜味剂也可以具有降低令人不快的味道,例如苦味的作用。也可以将苦味抑制剂或口味掩蔽剂与活性剂和甜味剂一起添加,用以使令人不快的口味降低。
亲脂活性剂也可以被结合进涂敷的口香糖产品中。多种活性剂,例如维生素可以被添加进口香糖涂层中,但不被添加进口香糖芯中,除非是被包胶的用于以后的释放。如果活性剂对胶基亲合力,它自然不需要包胶就可以缓慢地释放。如果活性剂正常情况下快速释放,则它就不得不被包胶或包裹住,使其在所需的时间释放。
在许多情况下,如果添加进口香糖涂层则活性药物具有低质量的异味或苦味。在大多数情况中,该异味可以用高效甜味剂遮盖,但在其它情况下,可能需要苦味抑制剂用以降低药物的苦味。
有许多种苦味抑制剂可以与活性剂用于食品中。一些优选的苦味抑制剂是钠盐,这在Philadelphia,Pennsylvania的Monell ChemicalSenses Center的R.A.S.Breslin和G.K.Beceuchenp的文章
钠对苦味 的抑制:苦味刺激中的变化(Suppression of Bitterness by Sodium:Variations Among Bitter Taste Stimuli)中有所论述。所论述的钠盐是醋酸钠和葡萄糖酸钠。其它同样有效的钠盐是甘氨酸钠、抗坏血酸钠和甘油磷酸钠。其中,最优选的是葡萄糖酸钠和甘氨酸钠,因为其具有低盐味道,且对于降低大多数活性药物的苦味是有效的。
大多数钠盐是非常易溶于水的,且易于从口香糖涂层中释放出来,起苦味抑制剂的作用。在大多数的情况下,易于从口香糖芯释放的钠盐通过包胶得以改进,使其从口香糖中的释放更快。然而,在一些情况下钠盐会被包胶或包裹住使其从口香糖中的释放延迟。一般,苦味抑制剂应与活性药物一起释放以达到最佳的效果。
由口香糖涂层中的药物的释放也受药物的粒径影响。小粒径的释放较快,而大粒径的释放较慢。通过将药物溶解于液体中来制备口香糖涂层同样可以实现快速的释放。
大多数情况下,口香糖组合物一般包括水溶性填充部分,水不溶性可咀嚼胶基部分和一般的水不溶风味剂。水可溶部分与一部分风味剂在长时间的咀嚼中散失。该胶基部分在整个咀嚼过程中保持在口中。
本发明独特的不溶性胶基将包括亲水聚合物(也称为极性聚合物),包括多种分子量的聚醋酸乙烯酯,短和中链聚酯或聚酰胺,和短和中侧链的聚(乙烯基酯)(例如,聚丁酸乙烯酯、聚丙酸乙烯酯)。不溶性胶基实质上就没有疏水聚合物,例如天然和合成的橡胶弹性体,特别是丁基弹性体、聚异丁烯、苯乙烯-丁二烯橡胶弹性体。该不溶胶基实质上也没有弹性体溶剂,例如萜烯树脂、酯胶和甘油松香酯。该不溶的胶基将含有亲水的软化剂/乳化剂,而实质上没有疏水软化剂。该不溶胶基构成口香糖重量的约5%至约95%,更通常是构成约10%至约50%或约25%至35%。
在具体的实施方案中,本发明的口香糖胶基含约20至约90wt.%的亲水聚合物,约4至约50wt.%的填料,约5至约35wt.%的亲水软化剂/乳化剂,和可选的较少量(约1或更少wt.%)的各种成分例如,色素和抗氧化剂等。
填料/组织形成剂可以包括碳酸镁和碳酸钙、重质碳酸钙、硅酸盐例如硅酸镁和硅酸铝、粘土、矾土、滑石粉、二氧化钛、磷酸一、二和三钙、纤维素聚合物例如燕麦纤维,和其组合。
亲水软化剂/乳化剂可以包括一硬脂酸甘油酯、三醋酸甘油酯、卵磷脂、一和二脂酸甘油酯,和短和中链三脂酸甘油酯、乙酰化一脂酸甘油酯和其组合。
色素和增白剂可以包括FD&C型染料和色淀、水果和蔬菜提取物、二氧化钛和其组合。
除了水溶胶基部分以外,一般的口香糖组合物包括水溶填充部分和一种或多种风味剂。该水溶部分可以包括填充甜味剂、高效甜味剂、风味剂、软化剂、乳化剂、色素、酸化剂、填料、抗氧化剂和提供所需属性的其它成分。
为了使口香糖的可咀嚼性和口感最佳化,向口香糖中添加了软化剂。软化剂也就是公知的增塑剂,一般占口香糖重量的约0.5至约15%。该软化剂可以包括甘油、卵磷脂和其组合。甜味剂水溶液例如含山梨醇、氢化淀粉水解产物、玉米糖浆和其组合的那些水溶液,也可以用作口香糖中的软化剂和粘合剂。
填充甜味剂包括糖成分和无糖成分。填充甜味剂一般占口香糖重量的约5至约95%,更一般是约20%至约80%,且更通常是约30%至约60%。糖甜味剂一般包括口香糖领域公知的含糖成分,包括但不仅限于蔗糖、葡萄糖、麦芽糖、糊精、转化糖干粉、果糖、左旋糖、半乳糖和玉米糖浆固体等,单独使用或组合使用。无糖甜味剂包括但不仅限于糖醇,例如山梨糖醇、甘露糖醇、木糖醇、氢化淀粉水解产物和麦芽糖醇等,可以单独使用或组合使用。
也可以使用高效人工甜味剂,与上述一样可以单独使用或组合使用。优选的甜味剂包括但不仅限于氯化蔗糖衍生物、阿斯巴甜、N-取代APM衍生物例如新甜(neotame)、双氧恶噻嗪盐、缩二氨酸基酰胺、糖精和其盐、环己氨磺酸及其盐、甘草甜素、二氢查耳酮、祝马丁和莫尼林等,可以单独使用或组合使用。为了得到更持久的甜味和风味感觉,优选采用包胶或别的使人工甜味剂的至少一部分释放的控制方法。这样的技术如湿式制粒法、蜡式成粒法、喷雾干燥、喷雾冷却、流化床涂敷、凝聚和纤维挤压可以用来实现所需的释放特性。
糖和/或无糖甜味剂的组合可以用于口香糖。另外,软化剂也可以提供另外的甜味,例如和糖或糖醇水溶液一起。
如果需要低卡路里口香糖,可以使用低卡路里填充剂。低卡路里填充剂的实例,包括聚葡萄糖、棉子糖、棉子素(Raftilin)、果寡糖(NutraFlora)、巴糖寡糖、瓜尔豆胶水解产物(Sun Fiber)、或不消化的糊精(Fibersol)。当然,也可以使用其它低卡路里填充剂。
如果需要也可以使用多种风味剂。风味剂的用量是口香糖重量的约0.1至约15%,且优选约0.2至约5%。风味剂可以包括精油、合成风味剂或其混合物,包括但不仅限于源自植物和水果的油例如柑桔油、水果香精、薄荷油、留兰香油、其它薄荷油、丁香油、冬青油和茴香油等。也可以使用人工风味剂和风味成分。天然和人工风味剂可以以任何感觉可接受的方式组合。
在大多数的情况下,通过连续地向市售的本领域公知的混合器中添加各种口香糖成分而制备出口香糖。在这些成分已经被彻底混合之后,从混合器排出胶块,并定形为所需的形状,例如辊压成片和切割成条,挤压成大块或铸成小丸,接着涂敷或挂糖衣。
一般,通过第一次熔化胶基将各成分混合,并添加进运行的混合器。该胶基也可以在混合器中熔化。色素或乳化剂也可以同时被添加。软化剂例如甘油也可以与糖浆和一部分填充剂同时添加。填充剂其它的部分被添加进混合器中。风味剂一般与最后部分的填充剂一起添加。其它可选的成分以一般的方式添加进配料中,这是本领域普通技术人员所公知的。
全部混合步骤一般花费5至15分钟,但有时也需要更长的混合时间。本领域普通技术人员能够识别出上述步骤的许多变化。
在本发明中,亲脂药物或活性剂用于颗粒口香糖的涂层/糖衣。颗粒或球形口香糖如常规口香糖的方法制备,但形成枕头形颗粒或形成球形。该颗粒/球形可以接着被涂敷糖或通过常规的挂糖衣技术挂糖衣,制成独特的涂敷颗粒口香糖。该亲脂活性剂将溶于风味剂中,或可以与其它经常用于一些常规挂糖衣工艺中的粉末混合。将亲脂活性剂与其它口香糖成分分离,将改变其从口香糖的释放速率。活性剂的量可以是口香糖涂层重量的约10ppm至30%。涂层重量可以是终产品重量的约20至约50%,但可以是总口香糖产品的差不多75%。活性剂的用量一般是约12微克至约250毫克每克涂敷的口香糖产品。活性剂的量一般基于一或二个颗粒的剂量。
常规挂糖衣工艺一般用蔗糖涂敷,但近年来在挂糖衣方面,已经允许使用其它糖类来代替蔗糖。这样的一些成分包括,但不仅限于葡萄糖、麦芽糖、巴糖、木糖醇、乳糖醇、氢化异麦芽糖、赤藓醇和其它新的糖醇或其组合。这些物料可以与糖衣改性剂混合,改性剂包括但不仅限于阿拉伯树胶、麦芽糖糊精、玉米糖浆、明胶、纤维素类型的物料例如羧甲基纤维素或羟甲基纤维素、淀粉和变性淀粉、植物胶例如藻酸盐、槐树豆胶、瓜尔豆胶和黄芪胶、不溶的碳酸盐例如碳酸钙或碳酸镁和滑石。也可以添加防粘着剂作为糖衣改性剂,可以使用多种糖类和糖醇用于开发新的挂糖衣的或涂敷的口香糖产品。风味剂也可以与糖或无糖涂层和活性剂一起添加以产生出独特的产品特性。
另一种类型的挂糖衣涂敷也可以将亲脂活性剂与口香糖成分分离。该技术称为涂膜,且更通常用于药品而不是口香糖,但工艺类似。膜例如虫胶、玉米醇溶蛋白或纤维素类型的物料被涂敷在颗粒型产品上,形成产品表面上的薄膜。通过将聚合物、增塑剂和溶剂(颜料是可选择的)混合,并将该混合物喷涂到颗粒表面而完成涂膜。在常规的挂糖衣设备中进行,或在更高级的旁透气管(side-vended)涂料盘中进行。因为,亲脂活性剂是醇溶的,它们可以容易地与这种类型的膜一起添加。当使用溶剂例如,醇时,对防止火灾和爆炸,需要额外的预防,并且必须使用专门的设备。
一些膜聚合物可以用水作为膜涂料中的溶剂。在聚合物研究和涂膜技术领域中近年来的发展,排除了在涂料中使用溶剂有关的问题。这些进展使对颗粒或口香糖制品涂敷水性膜成为可能。一些亲脂活性剂可以被悬浮于这种水性膜中,甚至于醇溶剂膜中,其中活性剂是高度可溶的。该膜也可以含有与聚合物和增塑剂一起的风味剂。该活性剂也可以被悬浮于水性或非水性溶剂中,并与水性膜一起涂敷与于该表面。在一些情况下,膜和糖或多元醇涂料的组合是有用的,特别是如果活性剂与膜涂料一起添加的情况。另外,膜涂料可以在涂敷过程的早期、中期或晚期涂敷。这将提供一种独特的源自涂膜产品的活性剂释放。
在具有亲脂活性药物的涂膜涂敷到口香糖制品之后,硬壳糖或多元醇涂层被接着涂敷到该涂膜的产品上。在一些情况下,软壳糖或多元醇涂层也可以被用来涂敷到涂膜的产品上。涂敷到颗粒口香糖的膜涂层的量一般是口香糖产品的约0.5至约3%。硬壳或软壳的保护涂层的量可以是约20%至约75%。当活性剂与膜涂料一起添加时,其中没有糖/多元醇涂层,可以获得产品中较好地对活性剂量的控制。另外,该糖/多元醇保护涂层使产品中活性剂的稳定性得到了改善。
如上所述,涂料可以含有的成分例如是风味剂,以及人工甜味剂和分散剂、着色剂、膜成形剂和粘合剂。本发明的风味剂包括本领域公知的那些风味剂,例如精油、合成风味剂或其混合物,包括但不仅限于源自植物和水果的油例如柑桔油、水果香精、薄荷油、留兰香油、其它薄荷油、丁香油、冬青油和茴香油等。风味剂的用量是涂层中含约0.2至约3%的风味剂,且优选约0.7至约2.0%的风味剂。活性剂可以与用于涂层中的风味剂预混合。
被考虑用于涂层中的人工甜味剂包括但不仅限于合成物质、糖精、祝马丁、缩二氨酸基酰胺、糖精盐、阿斯巴甜、N-取代APM衍生物例如新甜(neotame)、氯化蔗糖衍生物和双氧恶噻嗪钾。该人工甜味剂可以被添加至涂料糖浆中的量,应能使涂层中的含量为约0.01至约0.5%,且优选约0.1至约0.3%的人工甜味剂。
分散剂经常被添加到糖浆涂料中,用于增白和防粘。本发明的用于涂料糖浆的分散剂包括二氧化钛、滑石或任何其它抗粘的化合物。二氧化钛是本发明目前优选的分散剂。添加到涂料糖浆中的分散剂的量应能使涂层含约0.1至约1%,且优选约0.3至约0.6%的该制剂。
着色剂优选直接以染料或色淀的形式被添加至糖浆中。本发明的着色剂包括食品级染料。优选添加至糖浆的膜成形剂包括甲基纤维素、明胶、羟丙基纤维素、乙基纤维素、羟乙基纤维素、羧甲基纤维素等和其组合。粘合剂既可以被添加作为口香糖芯的初始涂层,也可以被直接添加进糖浆。本发明的粘合剂包括阿拉伯树胶、塔哈胶(阿拉伯树胶的另一种类型)、藻酸盐、纤维素制品和植物胶等。
最初作为液体糖浆的涂料含约30%至约80%或85%的如前所述的涂料成分和约15%或20%至约70%的溶剂例如水。在大多数情况下,在转盘中进行涂敷工艺。要涂敷的糖或无糖口香糖芯片被放置于转盘中,形成转动的块。
最后形成涂层的物料或糖浆被涂敷或散布在口香糖芯片上。在将糖浆涂敷到口香糖芯的之前,期间和之后,可以添加风味剂。一旦涂层被干燥形成硬壳,添加另外的糖浆就产生出多个涂层或多层硬涂层。
在硬涂层的挂糖衣步骤中,向口香糖芯片添加糖浆,糖浆的温度是约100°F至约240°F。在多数情况下,在整个过程中,糖浆温度是约130°F至200°F,这样以便于防止糖浆中的多元醇或糖形成结晶。该糖浆可以以喷涂、倾泻或添加到口香糖芯片上的任何本领域普通技术人员公知的方式被混合。
在大多数情况下,通过涂敷单一涂料得到多层,使这些层干燥,并接着重复该工艺。由主要依赖于涂料糖浆浓度的各涂敷步骤确定添加的量。口香糖芯片可以涂敷任何数量的涂层。一般,涂敷到口香糖芯片的涂层不超过约75-100。本发明涂敷的糖浆的量足以产生涂敷的、可食的含约10%至约75%的涂层。其中,需要较高剂量的活性剂,终产品可以是高于75%的涂层。
本领域普通技术人员能够想到,为了得到多个涂层,将多个预测的等分的涂料糖浆涂敷到口香糖芯片上。然而值得注意的是在整个涂敷步骤中,涂敷到口香糖芯片的等分糖浆体积是可以变化的。
一旦将糖浆涂料涂敷到口香糖芯片,则本发明就会考虑在惰性介质中将湿糖浆干燥。优选的干燥介质包括空气。迫使干燥空气与湿糖浆涂料在约70°F至约115°F的温度范围内接触。一般,干燥空气处于约80°F至约100°F的温度范围内。本发明也考虑到干燥空气拥有的相对湿度小于约15%。优选,干燥空气的相对湿度小于约8%。
干燥空气可以以本领域任何公知的方式吹拂过糖浆涂敷的口香糖芯,并与其混合。一般,干燥空气以一定的流速吹拂并围绕或通过糖浆涂敷的口香糖芯的床,对于大规模的操作,流速是约2800立方尺每分钟。如果,加工的是较低量的物料或如果使用的是较小的设备,将采用较低的流速。
多年来,一直是通过向颗粒口香糖的糖涂层添加风味剂,来增强口香糖总体的风味。这些风味剂包括薄荷香精、欧薄荷香精、冬青香精和水果香精。这些风味剂一般是在即刻将涂料糖浆对口香糖芯实施涂敷之前,与涂料糖浆进行预混合的,或以一次或多次涂敷方式,在含口香糖芯的包糖衣锅中与糖浆一起添加。一般,该涂料糖浆是非常热的,约130°F至200°F,且如果与涂料糖浆过早的预混合,则风味剂可能挥发。
该浓缩的涂料糖浆以热液体的形式被涂敷到口香糖芯上,容许糖或多元醇结晶,且接着该涂料用温暖的干燥空气干燥。重复约30至100次以得到重量增加约40%至75%的硬壳涂敷的产品。风味剂涂敷采用一、二、三或甚至四次或更多次这样的涂敷方式。每次添加风味剂,就涂敷几层非风味涂料,用以在涂敷下一风味剂涂料之前,覆盖该风味剂。这降低了在涂敷期间风味剂的挥发。
对于薄荷香精,例如薄荷香精、欧薄荷香精和冬青香精,一些风味剂成分被挥发,但仍保留了足够的风味剂,以使产品具有浓的、强的风味冲击。水果香精,可能含有酯,更容易挥发,且是易燃和/或易爆炸的,因此为了能够将其添加进口香糖涂层,一般这些类型的水果香精可以进行预处理。
在本发明的一个实施方案中,亲脂活性剂与阿拉伯树胶溶液预混合,成为糊状物,并接着被涂敷到芯上。为了降低粘度,在被涂敷之前,将该预混合物与少量的涂料糖浆混合。接着继续驱动干燥空气,使阿拉伯树胶将活性剂粘合到芯上。然后涂敷另外的涂层以覆盖活性剂,将活性剂包埋入涂层中。
口香糖制品实施例
以解释和举例说明的方式,提供本发明的下述实施例和对比实施例。
如前所述,以糖或无糖型配方的形式制备口香糖制品。这些制品被制成丸形或枕形颗粒的或圆球形的或任何其它形状的涂敷/挂糖衣的产品。然而,对于丸形芯的口香糖制品一般被调节成比条形口香糖具有更高量的胶基,使口香糖丸成为更易被消费者接受的大小。
照着这样的思路,如果添加到丸形芯的总产品的约25%的涂层是糖或多元醇,则丸形芯中的胶基也应增加25%。同样地,如果涂敷33%的涂层,则胶基量也应提高33%。结果,口香糖芯通常被配制成具有约25%至约40%的胶基,而除风味剂以外的其它成分,则相应地降低。当活性剂被添加至丸形涂层时,可以采用更高量的胶基。一般,风味剂随着胶基的量增加,因为胶基趋向于将风味剂粘合进口香糖,且需要更多的风味剂得到良好的具有浓郁风味的产品。然而,也可以将风味剂添加到涂层,使风味的冲击提高,更具有风味。
实施例:
常规的涂敷的口香糖组合物可以在口香糖涂层中,含有亲脂活性剂。当对该组合物咀嚼约30分钟时,约50%的亲脂活性剂从咀嚼的口香糖中释放,如通过对口香糖丸分析所确定的。相信该亲脂活性剂被束缚进亲脂胶基组合物中,并没有被释放。结果,对常规口香糖胶基和从常规的非涂敷口香糖制品的亲脂活性剂的释放进行比较测试。

每块口香糖芯重量是1.25g,被涂敷后成每块1.91g,得到了34.5%的涂层。虽然几乎所有的醋酸酯在10分钟内就会被释放,但涂敷的口香糖制品被咀嚼30分钟,对口香糖丸的醋酸维生素E进行分析,则得到80.8%的释放率。比前面的亲脂活性剂从常规涂敷的口香糖制品释放的量高很多。
应该理解,本发明的组合物和方法能够被应用于多种形式的实施方案中,上述只是进行了少量的举例说明。本发明也可以其它形式被具体化,并不脱离其精神或实质。所述的实施方案,无论从哪方面看都只是举例说明,而不是限制,因此本发明的范围通过附加的权利要求书来说明,而不是根据前面的描述。该权利要求书的相同的意思和范围内的所有变化,均包含在其范围内。
Claims (30)
1.一种亲水胶基,包括:
a)约20%至约90%的亲水聚合物;
b)约5%至约35%的亲水软化剂/乳化剂;和
c)约4%至约50%的填料;
d)该口香糖胶基并没有疏水聚合物、弹性体溶剂、蜡和疏水软化剂。
2.根据权利要求1的亲水胶基,其中亲水聚合物选自聚醋酸乙烯酯,短和中链聚酯、短和中链聚酰胺,和短和中侧链的聚(乙烯基酯)和其组合。
3.根据权利要求1的亲水胶基,其中亲水聚合物选自高分子量聚醋酸乙烯酯、低分子量聚醋酸乙烯酯、聚丁酸乙烯酯、聚丙酸乙烯酯和其组合。
4.根据权利要求1的亲水胶基,其中亲水软化剂/乳化剂选自一硬脂酸甘油酯、三醋酸甘油酯、卵磷脂、一和二脂酸甘油酯、短和中链三脂酸甘油酯、乙酰化一脂酸甘油酯和其组合。
5.根据权利要求1的亲水胶基,其中填料选自碳酸镁、碳酸钙、重质碳酸钙、硅酸镁、硅酸铝、粘土、矾土、滑石粉、二氧化钛、磷酸一、二和三钙、纤维素聚合物和其组合。
6.根据权利要求1的亲水胶基,其中胶基没有丁基弹性体、聚异丁烯和苯乙烯-丁二烯橡胶弹性体。
7.根据权利要求1的亲水胶基,其中胶基没有萜烯树脂、甘油松香酯和酯胶。
8.根据权利要求1的亲水胶基,其中当将亲脂活性剂混合进非涂敷的口香糖制品中时,在咀嚼30分钟内,该胶基从口香糖释放至少10%的亲脂活性剂。
9.一种口香糖制品是使用权利要求1-8中任意一种胶基制备的。
10.一种涂敷的口香糖制品,包括:
a)由亲水胶基制备口香糖芯;和
b)在该芯上涂敷,涂料中包括亲脂活性剂。
11.权利要求10的涂敷的口香糖制品,其中亲脂活性剂选自维生素、癌症化疗药、抗真菌药、口服避孕药、镇痛剂、抗酸剂、骨骼肌松弛药、抗组胺药、减充血剂、麻醉剂、镇咳剂、利尿剂、消炎药、抗生素、抗病毒药、精神治疗剂、治疗糖尿病药、心血管治疗药、生物工程药、营养药品和营养添加剂。
12.一种在涂层中含至少一种亲脂活性剂的涂敷的口香糖制品的制备方法,包括以下步骤:
a)提供口香糖制品芯,其中口香糖由亲水胶基制得;
b)提供一种涂料溶液;
c)用该涂料溶液涂敷该口香糖制品芯,用以提供涂敷的口香糖制品,该涂料包括的亲脂活性剂的量是每克涂敷的口香糖产品约12微克至约250毫克。
13.根据权利要求12的方法,其中在涂敷所述芯之前,将活性剂在涂料溶液中混合。
14.根据权利要求13的方法,其中在添加到涂料溶液之前,该活性剂也与溶剂混合,且得到的混合物被添加到口香糖涂料。
15.根据权利要求14的方法,其中溶剂是水、醇或风味剂。
16.根据权利要求12的方法,其中高效甜味剂选自阿斯巴甜、缩二氨酸基酰胺、双氧恶噻嗪盐、环己氨磺酸及其盐、糖精和其盐、新甜、祝马丁、莫尼林、二氢查耳酮、氯化蔗糖衍生物和其组合被混合进涂料溶液。
17.根据权利要求12的方法,其中所述的亲脂活性剂选自维生素、镇痛剂、抗酸剂、抗组胺药、镇咳药、抗菌剂、减充血剂和麻醉剂。
18.根据权利要求12的方法,其中活性剂是营养药品。
19.根据权利要求12的方法,其中所述的活性剂是维生素E。
20.根据权利要求12的方法,其中涂敷操作包括将涂料溶液多次涂敷,和在涂料溶液涂敷之间涂敷粉料。
21.根据权利要求20的方法,其中活性剂被包括在粉料中。
22.根据权利要求20的方法,其中活性剂被包括在涂料溶液和粉料中。
23.根据权利要求12的方法,其中亲脂活性剂也包括在口香糖芯中。
24.根据权利要求23的方法,其中在口香糖芯和涂层中活性剂是相同的。
25.根据权利要求23的方法,其中在口香糖芯中的活性剂与涂层中的活性剂是不同的。
26.根据权利要求12的方法,其中使用至少两种不同的涂料溶液制备涂层。
27.根据权利要求26的方法,其中活性剂与至少两种不同的涂料中的第一溶液混合,并涂敷形成膜,在该涂膜芯上涂敷没有活性剂的第二涂料溶液。
28.根据权利要求12的方法,其中涂层中的活性剂可以是口香糖涂层重量的约10ppm至30%。
29.一种输送亲脂活性剂的方法,包括以下步骤:
a)提供一种口香糖制品,其具有i)使用亲水胶基制备的口香糖芯,和ii)在涂层中包括亲脂活性剂的涂层;以及
b)在口腔中咀嚼该口香糖产品至少10分钟。
30.根据权利要求29的方法,其中活性剂选自维生素、镇痛剂、骨骼肌松弛药、抗生素、抗病毒药、抗组胺药、减充血剂、消炎药、抗酸剂、精神治疗剂和心血管治疗药。
