Abstract
The invention relates to the technical field of preparation of repairing active ingredients and repairing products, and particularly discloses a hyaluronic acid-containing composition and application thereof in preparation of products with repairing effects. The hyaluronic acid-containing composition comprises hyaluronic acid and lavender extract; wherein the weight ratio of the hyaluronic acid to the lavender extract is 1. The composition formed by mixing the lavender extract and hyaluronic acid has a certain repairing effect; particularly, the repairing effect of the composition formed by mixing the lavender extract obtained by extracting the lavender ethanol extract by using the organic solvent and the hyaluronic acid is further enhanced; therefore, the compound has important application value when being used as an active ingredient for preparing skin care products, cosmetics or medicaments with repairing effect.
Description
Technical Field
The invention relates to the technical field of preparation of repairing active ingredients and repairing products, in particular to a hyaluronic acid-containing composition and application thereof in preparation of products with repairing effects.
Background
The skin is the largest organ of the human body; for adults, the total skin area is about 7600 square centimeters, which accounts for 15% of the total body weight, and about 1/3 of the circulating blood volume is supplied to the skin. The skin is the first line of defense of the human body and is easily damaged by the injury factors to form a wound surface. Wound healing refers to the process of regeneration, repair and reconstruction of local tissues after skin is subjected to the action of a wound-causing factor to cause tissue loss. The repair of the wound surface of the skin depends on the repair capability of the skin, and the repair product is matched to realize faster and better repair.
Hyaluronic acid, is a disaccharide glycosaminoglycan composed of D-glucuronic acid and N-acetylglucosamine. The prior art fills the scar into the crater of some acne scars, the scar caused by trauma or operation, thereby realizing the filling of the scar. But it does not have obvious effect of promoting the repair of the skin; moreover, the medicine is easy to be absorbed and metabolized by human body, and the maintaining time is relatively short.
Therefore, the hyaluronic acid raw material is added with other active ingredients, so that a composition with good repairing effect is developed, and the hyaluronic acid composition has important application value.
Disclosure of Invention
In order to overcome at least one of the technical problems of the prior art, the invention provides a hyaluronic acid-containing composition.

extracting the dried lavender whole plant with ethanol, concentrating and drying the extracting solution after the extraction is finished to obtain the lavender ethanol extract, namely the lavender extract.
Preferably, the ethanol adopts 60-80% ethanol water solution by volume fraction.
Most preferably, the ethanol is 75% ethanol water solution by volume fraction.
The inventor surprisingly finds that the composition formed by mixing the lavender extract extracted by using the ethanol water solution with the volume fraction of 60-80% and the hyaluronic acid has a certain repairing effect.
Preferably, the weight amount of the ethanol is 8 to 15 times of that of the dried lavender whole plant;
most preferably, the amount of ethanol is 12 times the weight of the dried whole lavender herb.
Preferably, the extraction is heating reflux extraction; the extraction time is 1-2 h.
Preferably, the lavender ethanol extract is further extracted.
The specific extraction method comprises the following steps: adding the lavender ethanol extract into water, stirring uniformly to obtain a suspension, and then adding an organic solvent for extraction; separating the organic solvent layer, concentrating and drying the organic solvent layer to obtain the lavender extract.
The inventor further finds in research that the repairing effect of the composition formed by mixing the lavender extract obtained by extracting the ethanol extract of the lavender with the hyaluronic acid through the organic solvent is further enhanced.
Preferably, the amount of water is 20-30 times of the weight of the ethanol extract of lavender.
Preferably, the volume ratio of water to organic solvent is 1-2.
Most preferably, the volume ratio of water to organic solvent is 1.
Preferably, the organic solvent is a mixed organic solvent.
Preferably, the mixed organic solvent consists of n-hexane, chloroform and ethyl acetate in a volume ratio of 2-4.
Most preferably, the mixed organic solvent consists of n-hexane, chloroform and ethyl acetate in a volume ratio of 3.
The inventor surprisingly discovers in research that when the composition is prepared by mixing the lavender extract obtained by extracting the lavender extract with the organic solvent consisting of normal hexane, chloroform and ethyl acetate and hyaluronic acid, the repairing effect is greatly improved, and the composition has excellent repairing effect; the repairing effect is far higher than that of a composition which is prepared by mixing a lavender extract obtained by adopting single n-hexane, chloroform or ethyl acetate extraction and hyaluronic acid; the composition is also far higher than the composition which is prepared by mixing the lavender extract obtained by extracting any two of normal hexane, chloroform and ethyl acetate with hyaluronic acid. The lavender extract extracted by an organic solvent consisting of normal hexane, chloroform and ethyl acetate can synergistically improve the repairing effect of a composition formed by mixing the lavender extract with hyaluronic acid.
Preferably, the hyaluronic acid-containing composition also contains alpha-mangostin.
Preferably, the weight ratio of the hyaluronic acid to the lavender extract to the alpha-mangostin is 1:1.
The inventor further surprisingly discovers in research that the alpha-mangostin has excellent repairing effect; the alpha-mangostin is added into the composition consisting of hyaluronic acid and the lavender extract, so that the repairing effect of the composition consisting of hyaluronic acid and the lavender extract can be further greatly improved.
Meanwhile, the inventor also finds that the repairing effect of the composition consisting of hyaluronic acid, lavender extract and alpha-mangostin is better than that of the composition consisting of hyaluronic acid and lavender extract which is independent of the alpha-mangostin; this demonstrates that the addition of alpha-mangostin to a composition consisting of hyaluronic acid and lavender extract synergistically improves the reparative effect of the prepared composition.
The invention also provides application of the hyaluronic acid-containing composition in preparation of products with repairing effects.
Preferably, the product is a skin care product, a cosmetic product or a medicament.

EXAMPLE 1 preparation of hyaluronic acid-containing compositions
The hyaluronic acid-containing composition is prepared by mixing hyaluronic acid and lavender extract according to the weight ratio of 1;
the lavender extract is prepared by the following method: heating the dried lavender whole plant with 75% ethanol water solution with 12 times weight of volume fraction to boiling state, extracting for 1.5h, concentrating the extractive solution, and drying to obtain lavender ethanol extract.
EXAMPLE 2 preparation of hyaluronic acid-containing compositions
The hyaluronic acid-containing composition is prepared by mixing hyaluronic acid and lavender extract according to the weight ratio of 1;
the lavender extract is prepared by the following method: heating dried lavender whole plant with ethanol water solution with volume fraction of 60% 15 times of the weight of the lavender whole plant to boiling state, extracting for 2h, and concentrating and drying the extracting solution after extraction is finished to obtain lavender ethanol extract, namely the lavender extract.
EXAMPLE 3 preparation of hyaluronic acid-containing compositions
The hyaluronic acid-containing composition is prepared by mixing hyaluronic acid and lavender extract according to the weight ratio of 1;
the lavender extract is prepared by the following method: heating the dried lavender whole plant with an ethanol water solution with volume fraction of 80% of 8 times of the weight of the lavender whole plant to boiling state for extraction for 1 hour, and concentrating and drying the extracting solution after the extraction is finished to obtain the lavender ethanol extract, namely the lavender extract.
EXAMPLE 4 preparation of hyaluronic acid-containing compositions
The hyaluronic acid-containing composition is prepared by mixing hyaluronic acid and lavender extract according to the weight ratio of 1;
the lavender extract is prepared by the following method:
(1) Heating dried lavender whole plant with 75% ethanol water solution in an amount which is 12 times the weight of the lavender whole plant to boiling state, extracting for 1.5h, and concentrating and drying the extracting solution after extraction is finished to obtain lavender ethanol extract;
(2) Adding the lavender ethanol extract into 25 times of water by weight, stirring uniformly to obtain a suspension, and then adding an organic solvent for extraction; separating the organic solvent layer, concentrating and drying the organic solvent layer to obtain the lavender extract;
wherein, the volume ratio of the water to the organic solvent in the step (2) is 1;
the organic solvent in the step (2) is n-hexane.
EXAMPLE 5 preparation of hyaluronic acid-containing composition
Example 5 differs from example 4 in the choice of organic solvent extracted during the preparation of lavender extract, which is otherwise the same as example 4;

Examples of the experiments
Taking 130 female KM mice of 6 weeks old, adaptively feeding for 1 week, and randomly dividing into 13 groups of 10 mice each; wherein 12 groups are experimental groups and 1 group is a control group. After grouping, the backs of the mice in each group were shaved, the straight back skin (circular shape with a diameter of 1 cm) was cut under anesthesia, and then the wounds were sutured to prevent contraction; then the experimental group smears vaseline containing the sample to be tested with the weight percentage of 1 percent on the wound; smearing vaseline without the sample to be detected on the control group; the medicine is applied in the morning and at night every day; after 7 days, the new skin of each group of mice is observed, and the wound healing rate is calculated; the test results are shown in Table 1.

As can be seen from the experimental data in table 1, the healing rate of the wound surface of the experimental groups 1 to 3 using the composition containing hyaluronic acid described in examples 1 to 3 is significantly higher than that of the control group. This indicates that: the lavender extract extracted by using an ethanol water solution with the volume fraction of 60-80% is mixed with hyaluronic acid to form a composition, and the composition has a certain repairing effect.
As can be seen from the experimental data in table 1, the compositions containing hyaluronic acid of examples 4 to 9 in experimental groups 4 to 9 have improved wound healing rate, but the improvement is not particularly large, compared with the composition containing hyaluronic acid of example 1. This indicates that: the repairing effect of the composition consisting of the lavender extract obtained by extracting the ethanol extract of lavender with an organic solvent and hyaluronic acid is enhanced, but the enhancement is not particularly remarkable. As can be seen from the experimental data in table 1, after the composition containing hyaluronic acid described in example 10 is adopted in the experimental group 10, the wound healing rate is greatly improved compared with the composition containing hyaluronic acid described in example 1; the same is much higher than the hyaluronic acid-containing compositions described in examples 4 to 9. This indicates that: the repairing effect of the composition can be greatly improved only when the composition is prepared by mixing the hyaluronic acid and the lavender extract obtained by extracting the lavender extract by using an organic solvent consisting of normal hexane, chloroform and ethyl acetate.
In addition, as can be seen from the wound healing rates of the experimental groups 4 to 10, in example 10, the composition prepared by mixing the lavender extract extracted by the organic solvent consisting of normal hexane, chloroform and ethyl acetate with hyaluronic acid is selected, and the repairing effect of the composition is far higher than that of the composition prepared by mixing the lavender extract extracted by the normal hexane, chloroform or ethyl acetate with hyaluronic acid; the composition is also far higher than the composition which is prepared by mixing the lavender extract obtained by extracting any two of normal hexane, chloroform and ethyl acetate with hyaluronic acid; this also shows that the lavender extract extracted with an organic solvent consisting of n-hexane, chloroform and ethyl acetate synergistically improves the repairing effect of the composition consisting of it mixed with hyaluronic acid.
As can be seen from the experimental data in Table 1, the wound healing rate of the alpha-mangostin reaches 61.3%, and the alpha-mangostin has very excellent repairing effect; the hyaluronic acid-containing composition of example 11, which was prepared by adding hyaluronic acid and lavender extract to the composition, had a wound healing rate that was further improved by a large amount as compared to example 10. This indicates that: the alpha-mangostin is added into the composition consisting of hyaluronic acid and the lavender extract, so that the repairing effect of the composition consisting of hyaluronic acid and the lavender extract can be further greatly improved; so that the composition containing hyaluronic acid has excellent repairing effect. In addition, the healing rate of the wound surface of the experimental group 11 with the hyaluronic acid-containing composition of example 11 is also higher than that of alpha-mangostin; this demonstrates that the combination of alpha-mangostin with hyaluronic acid and lavender extract synergistically enhances the healing benefits of the resulting compositions.
Claims (10)
1. A hyaluronic acid-containing composition comprises hyaluronic acid and lavender extract;
wherein the weight ratio of the hyaluronic acid to the lavender extract is 1.
2. The hyaluronic acid-containing composition according to claim 1, wherein the weight ratio of hyaluronic acid to lavender extract is 1.
3. The hyaluronic acid-containing composition according to claim 1, wherein said lavender extract is obtained by the following process:
extracting the dried lavender whole plant with ethanol, concentrating and drying the extracting solution after the extraction is finished to obtain the lavender ethanol extract, namely the lavender extract.
4. The hyaluronic acid-containing composition according to claim 3, wherein the ethanol is 60-80% ethanol water solution by volume fraction;
most preferably, the ethanol is 75% ethanol water solution by volume fraction.
5. The hyaluronic acid-containing composition according to claim 3, characterised in that the amount by weight of ethanol is 8-15 times that of the whole plant of dried lavender;
most preferably, the amount of ethanol is 12 times the weight of the dried whole lavender herb.
6. The hyaluronic acid-containing composition according to claim 3, wherein the extraction is a heated reflux extraction; the extraction time is 1-2 h.
7. Hyaluronic acid-containing composition according to claim 3, characterised in that the lavender ethanol extract is further subjected to extraction;
the specific extraction method comprises the following steps: adding the lavender ethanol extract into water, stirring uniformly to obtain a suspension, and then adding an organic solvent for extraction; separating the organic solvent layer, concentrating and drying the organic solvent layer to obtain the lavender extract.
8. The hyaluronic acid-containing composition of claim 7, wherein the amount of water by weight is 20-30 times the weight of the ethanol extract of lavender.
9. The hyaluronic acid-containing composition according to claim 7, characterised in that the volume ratio of water to organic solvent is between 1 and 2;
most preferably, the volume ratio of water to organic solvent is 1.
10. Use of a hyaluronic acid-containing composition according to any of claims 1-9 for the preparation of a product having a reparative effect.
