Description
Technical Field
The invention belongs to the technical field of daily chemicals for pets, and relates to a tear stain removing gel for pets, and a preparation method and application thereof.
Background
Although the eye structure of dogs and cats is similar to that of human eyes and is composed of cornea, iris, crystalline lens, retina and the like, cats also have a third eyelid structure which is not possessed by human eyes and have the functions of secreting tears and protecting eyeballs. The lacrimal gland is an accessory organ of eyes and mainly has the functions of secreting tears to moisten eyeballs, and the tears secreted by the lacrimal glands of dogs and cats normally moisten the eyeballs and then enter a nasal cavity through a nasolacrimal duct so as not to flow out of canthus. If the tear water of dogs and cats can not be normally discharged from nasolacrimal ducts, because the tear water contains porphyrin pigment, if the pigment is accumulated below canthus, dark brown stain can be formed, namely tear stains seen by pet owners at ordinary times; besides water, the tears also contain some components such as protein, inorganic salt, lysozyme, oleamide and the like, wherein the lactoferrin component is easily oxidized to form reddish brown, so that the appearance of the pet is affected by the rusty color.
The factors causing the formation of the tear stains of the pets are many, if the pets are careless to eat at ordinary times, people often eat meals, excessive salt is ingested or the diet is too greasy, the salt in the pets is too high, the lacrimal gland of the pets can be exuberantly secreted, and meanwhile, harmful bacteria in the intestinal tract can be abnormally fermented, so that the pets have the tear stains. If the lacrimal gland of the pet develops and discharges too many tears in a short time or the nasolacrimal duct is blocked, the tears cannot be discharged from the lacrimal duct, the tears only flow out of the canthus, and tears are formed in places where the tears flow over time. In addition, the pets are infected with eye inflammation or fire, and the inflammation around the eyes, the secretion of the eyes and the tear stains are also serious.
When a pet develops tear stains, the owner generally cleans the pet by using eye drops or cleaning agents special for the pet. However, the special cleaning agent for tear stains disclosed in the prior art is less, and the common cleaning agent is weak in tear stain removing capability and strong in irritation, is not suitable for sensitive parts of eyes, or can only temporarily clean off the tear stains, and cannot radically reduce the generation of the tear stains of pets, so that the development of a product which is used for pets, has an obvious instant effect of removing the tear stains, can radically reduce the generation of the tear stains of pets, is mild, has no irritation and is high in safety is significant.

Preparation of erythrocyte suspension: selecting healthy rabbit, taking 9mL of blood from heart, adding 1mL of 2% potassium oxalate solution, centrifuging, discarding supernatant, diluting the precipitate to 20mL with 20mmol/L PBS solution, and storing at 4 ℃ for later use. Each set of samples was selected to be diluted to different concentrations with PBS solution, with 5 concentration gradients set for each sample. Taking 10mL of a diluent of a sample to be tested, adding 200 μ L of the erythrocyte suspension (controlling the final concentration of the sample to be 5, 10, 20, 50 and 100mg/mL respectively), taking distilled water as a total blood-dissolving control, taking a PBS solution as a negative control, gently mixing, incubating for 30min at 37 ℃, centrifuging for 10min at the rotating speed of 2000r/min, taking a supernatant, testing the absorbance (A560) of the supernatant at 560nm by using a spectrophotometer, and calculating the hemolysis rate according to the following formula;
hemolysis rate = [ (experimental group) A560 Negative control group A560 ) /(Whole blood control group) A560 Negative control group A560 )]×100%
A standard curve of the hemolysis rate versus the concentration of the sample was plotted, and the concentration of the sample at which 50% of erythrocytes were hemolyzed (HD 50) was calculated.

Diluting each group of samples to 10g/L by PBS (phosphate buffer solution), taking 10mL of diluent of a sample to be detected, adding 200 mu L of the erythrocyte suspension, taking distilled water as a blank control and 1mg/mL of Sodium Dodecyl Sulfate (SDS) solution as a positive control, gently mixing, incubating at 37 ℃ for 30min, centrifuging at the rotating speed of 2000r/min for 10min, taking supernate, respectively testing the absorbances A540 and A575 at 540nm and 575nm by a spectrophotometer, and calculating the protein Denaturation Index (DI) according to the following formula;
DI=[(R 1 -R 2 )/(R 1 -R 3 )]×100%
wherein R is 1 = blank control group a 575 Blank control group A 540 ,R 2 = Experimental group A 575 Experimental group A 540 ,R 3 = positive control group a 575 Positive control group A 540 。
