癌症疫苗需要诱导能够打破耐受性并克服免疫抑制性肿瘤环境的强效免疫反应以便有效。最近已经强调了CD4 T细胞在介导肿瘤破坏方面的重要性,然而已经证实诱导自身特异性CD4反应较为困难。相比之下,识别经修饰的自身表位的CD4 T细胞已经显示在一些自身免疫疾病的病理生理学方面发挥作用,如类风湿性关节炎(RA)、胶原蛋白II诱导的关节炎、肉状瘤病、乳糜泻和银屑病(Choy 2012;Grunewald和Eklund 2007;Coimbra等人2012;Holmdahl等人1985)。这些常见修饰之一是精氨酸的瓜氨酸化,其涉及精氨酸的带正电荷的醛亚胺基(=NH)转化为瓜氨酸的带中性电荷的酮基(=O)。瓜氨酸化经由肽酰精氨酸脱亚胺酶(PAD)介导,该酶是见于各种组织中的钙依赖性酶家族。Ireland等人的最近的报告(Ireland和Unanue 2011)表明,瓜氨酸化的T细胞表位在抗原提呈细胞(APC)上的提呈也依赖于自噬和PAD活性。这种过程也被证明是使得能够加工内源性抗原以在专职APC以及上皮细胞的MHC II类分子上提呈的有效机制(Munz 2012;Schmid,Pypaert,和Munz 2007)。自噬在APC中是组成性的,但是在其它细胞中只在应激下才被诱导(Green和Levine 2014)。识别瓜氨酸化表位的T细胞不会针对不表达瓜氨酸化肽的正常健康细胞。自噬是由应激(如低氧和缺营养)引发的,并且被上调以促进肿瘤的存活(Green和Levine2014)。

NPM与核仁的核糖核蛋白的结构有关,并且可以与单链核酸和双链核酸结合,但是优先结合G-四联体形成的核酸(由富含鸟嘌呤的序列在核酸中形成的二级结构)。NPM还可以作为蛋白质和核酸的分子伴侣(Hingorani,Szebeni,和Olson 2000;Szebeni和Olson1999;Okuwaki等人)。NPM属于细胞核伴侣家族蛋白质,被称为核仁磷酸蛋白(Np),它们都共有保守的N端区域。使用各种蛋白质底物的体外实验已经表明,NPM在预防蛋白质在细胞环境中聚集上具有活性(Szebeni和Olson 1999),并且NPM作为组蛋白伴侣起功能,其能够组装组蛋白、组装核小体和增加乙酰化依赖性转录(Okuwaki等人,2001;Swaminathan等人,2005)。
NPM最普遍的形式是NPM1.1(也叫B23.1)。还存在两种替代性剪接的同种型,称为NPM1.3(也叫B23.2)和NPM1.2。目前尚未发现NPM1.2的功能。NPM1.1是所有组织中最普遍的形式(Chang和Olson 1990;Wang,Umekawa和Olson 1993)。在原生环境下,NPM作为寡聚体存在(Herrera等人,1996),但是也可以形成五聚体和十聚体(Namboodiri等人,2004)。
NPM所参与的细胞活动的多样性使得其既是潜在的致癌基因又是潜在的肿瘤抑制物。NPM表达和/或定位的失调可以通过不同的机制以促成肿瘤发生。NPM蛋白在各种肿瘤中过表达,并且已经被提出作为胃癌(Tanaka等人,1992)、结肠癌(Nozawa等人,1996)、卵巢癌(Shields等人,1997)和前列腺癌(Subong等人,1999)的标志物。在一些情况下,NPM的表达水平已经与肿瘤进展阶段相关联。例如,NPM mRNA的过表达与膀胱癌的复发和疾病进展到更高阶段独立地相关(Tsui等人,2004)。蛋白质组学分析确定NPM作为雌激素调节蛋白,与人乳腺癌症细胞的获得性雌激素独立性相关(Skaar等人,1998年),表明NPM的表达状态可以与特定的病理生理特征关联。
NPM能够与不同细胞区室中的许多配偶体结合,包括核仁因子、转录因子、组蛋白、参与细胞增殖的蛋白质以及对致癌压力的反应。蛋白质的翻译后修饰发生在细胞压力环境下,其中一种修饰涉及瓜氨酸化,即由肽酰精氨酸脱亚胺酶(PAD)将精氨酸残基转化为瓜氨酸。瓜氨酸化的发生是作为在应激细胞中诱导的降解和再循环进程(自噬)的结果(Ireland和Unanue 2011)。瓜氨酸化的表位随后可以被提呈在MHC II类分子上,以由CD4 T细胞识别。响应于瓜氨酸化蛋白所释放的强效免疫反应可以被利用并且重新定向以摧毁癌症细胞。这种免疫反应是由杀伤性CD4 T细胞介导的,其随后分泌大量的IFNγ。这增加了MHC II类的表达,然后直接地杀伤肿瘤细胞,而不需要CD8 T细胞的参与(Brentville等人,2016;Durrant,Metheringham,和Brentville,2016)。肿瘤识别取决于瓜氨酸化和自噬。已经表明,用源自细胞骨架蛋白(即波形蛋白)的两种瓜氨酸化肽免疫接种小鼠后,诱导出大量的分泌IFNγ的CD4 T细胞。在离体情况下,这些CD4 T细胞识别由饥饿或雷帕霉素诱导自噬的肿瘤细胞。波形蛋白的功能和在肿瘤中表达先前已经在WO2014023957中详述。
自从发现NPM1基因的末端外显子中的杂合突变后,对NPM产生了很大兴趣。NPM1基因映射于染色体5q35,并且通过替代性剪接以三种同种型进行表达。NPM1.1(P06748-1)(294个残基)是最丰富的一种,并且显示在核仁中定位。同种型NPM1.2(P06748-2)缺少框内外显子(外显子8),产生相对于缺少内部区段(残基195-223)的NPM1.1更短的蛋白质。NPM1.3(P06748-3)在3'端使用替代性外显子,其导致产生相对于NPM1.1缺少最后35个氨基酸的更短的蛋白质结构(Wang,Umekawa,和Olson 1993);这种同种型的表达水平低,并且具有核质定位。最丰富的NPM1.1同种型(NPM1)在所有组织中表达。
NPM1基因在许多肿瘤类型中上调、突变且染色体易位;已经在患有非霍奇金淋巴瘤、急性早幼粒细胞白血病、骨髓增生异常综合征、和急性髓系白血病的患者中发现了染色体畸变(Falini等人,2007)。大多数NPM突变最终影响294个氨基酸的NPM蛋白中的在第288位氨基酸或第290位氨基酸的色氨酸残基(Duployez等人,2018)。超过一半的突变还与正常的核型有关(Duployez等人,2018)。然而,在占2%至8%的成人型非霍奇金淋巴瘤的间变性大细胞淋巴瘤(ALCL)中以及在较少的AML病例中,NPM基因与其它基因发生易位(Jairajpuri等人,2014)。在ACL中,2号染色体上的ALK与5号染色体上的NPM融合,然而在<1%的AML病例中,由于3号染色体和5号染色体的融合,易位产生被称为NPM MLF1(骨髓异常增生/髓系白血病因子1)的嵌合基因(Falini等人,2007)。急性早幼粒细胞白血病(APL)中迄今仅仅影响三个病例的极其罕见的易位涉及NPM1基因与维甲酸受体-α基因(RARα)的融合(Falini等人,2007)。NPM1的杂合小鼠很容易发生肿瘤。NPM也经常在不同组织学来源的各种实体瘤(前列腺(Leotoing等人,2008年)、肝脏(Yun等人,2007年)、甲状腺(Pianta等人,2010年)、结肠(Nozawa等人,1996年)、胃(Tanaka等人,1992年)、胰腺(Zhu等人,2015)、胶质瘤和胶质母细胞瘤(Chen等人,2015;Holmberg Olausson等人,2015)、星形细胞瘤(Kuo等人,2015)等)中过表达,并且在许多情况下,其过表达与有丝分裂指数和转移相关。因此,由于一系列实体癌和血液病癌症表达NPM,NPM是非常有吸引力的疫苗靶标。此外,存在针对突变形式的特异性抗体,使得诊断性测定得以实施,以鉴定具有从NPM靶向疗法受益的可能性的患者。
NPM已经显示与AKT1(Lee等人,2008)、BARD1(Sato等人,2004)、BRCA1(Sato等人,2004)和核仁素(nucleolin)(Li等人,1996)相互作用,并且具有多个结合配偶体(Lindstrom,2011)。人们认为NPM1可以通过肿瘤抑制因子p53/ARF途径的失活来促进肿瘤生长,尽管NPM1在以低水平表达时可以通过抑制中心体复制来抑制肿瘤生长。
在血清饥饿期间或抗癌药物的治疗期间,NPM可以被易位到核质,在核质中被磷酸化。NPM是用于治疗血液恶性肿瘤和实体恶性肿瘤的有希望的靶标。在过去的十年中,已经发现了一些靶向NPM1的分子,并且对其效果和治疗潜力进行了研究。在NPM靶向化合物与不同的化疗剂或放疗组合施用时,已经在许多案例中观察到惊人的协同作用,这表明干扰NPM可以使癌症细胞敏化。NSC348884、Rev37-47、1A1RNA适配体、CIGB-300、avrainvillamide、全反式维甲酸(ATRA)、YTR107和NucAnt 6L(N6L)是已经在体外或临床上经受测试的NPM结合化合物(Di Matteo等人,2016)。一些化合物(如NSC348884和1A1 RNA适配体)阻止NPM的寡聚化,因此使得该蛋白质不稳定(Qi等人,2008;Jian等人,2009),而Rev37-47和avrainvillamide与NPM的直接结合阻碍NPM的功能(Wulff,Siegrist,和Myers 2007;Szebeni,Herrera,和Olson 1995)。ATRA是APL的主要治疗方案之一(Lo-Coco等人,2013年),并且体外研究表明NPM的突变形式在与ATRA结合后会发生蛋白酶体降解(Martelli等人,2015年;Di Matteo等人,2016年)。YTR107可以经由NPM通过干扰DNA修复机制来诱导癌症细胞的辐射敏化(Di Matteo等人,2016;Sekhar等人,2014)。虽然机理途径不同,但是这些化合物与NPM结合的最终结果是在体外癌症细胞的凋亡。迄今为止,有两种化合物(CIGB-300,NPM磷酸化抑制剂和Nucant N6L(富含赖氨酸和精氨酸残基))已经进入临床试验。通过靶向NPM使癌症细胞凋亡的详细机制仍有待阐明,然而,迄今为止的证据表明靶向NPM的化合物是有前途的癌症治疗靶标。
在细胞压力和DNA损伤下,P53基因被激活。Tanikawa等人表明,P53诱导的PAD4反式激活导致NPM瓜氨酸化(Tanikawa等人,2009)。PAD4和NPM通常是基于细胞核的,用PAD4转染HEK293T细胞导致NPM从核仁定位到核质,表明PAD4介导的NPM瓜氨酸化发生在细胞核内(Tanikawa等人,2009)。同时,还将NPM从整个细胞裂解物中免疫沉淀出来并且进行质谱分析。鉴定出含有在197位的瓜氨酸的B细胞表位。然后用它来产生抗体(Tanikawa等人,2009)。我们的结果表明,含有在197位残基的瓜氨酸的肽不能诱导出T细胞反应,因此证实其是B细胞表位。相比之下,在本发明中,包含新的瓜氨酸化位置的NPM肽示出强烈的T细胞反应,从而产生抗肿瘤免疫力。
根据本发明的第一方面,提供了一种瓜氨酸化T细胞抗原,其包括以下、基本上由以下组成或由以下组成:
i)氨基酸序列AKFINYVKNCFRMTD,其中精氨酸(R)残基被瓜氨酸取代,或者
ii)i)的氨基酸序列,但是在非精氨酸位置上具有1、2或3个氨基酸置换,和/或1、2或3个氨基酸插入,和/或1、2或3个氨基酸缺失。
本发明人已经出乎意料地发现,可以针对来自表达在肿瘤细胞上的NPM的某些抗原(在该抗原中的精氨酸已经被瓜氨酸取代)产生T细胞反应。此外,含瓜氨酸的肽允许开发包括但不限于肿瘤疫苗的基于T细胞的疗法,以及T细胞受体(TCR)疗法和过继性T细胞转移疗法。
本发明的T细胞抗原可以是MHC II类抗原,即其与MHC II类分子形成复合物并且在MHC II类分子上提呈。技术人员可以通过确定在存在给定多肽下MHC是否可以重新折叠,来确定该给定多肽是否与该MHC分子形成复合体。如果多肽不与MHC形成复合物,则该MHC将不能合适地重新折叠。重新折叠通常用仅识别折叠态MHC的抗体来证实。可以在(Garboczi,Hung,和Wiley 1992)中查询更多的细节。
抗原中的所有精氨酸氨基酸残基可以被转化为瓜氨酸。或者,抗原中的1、2、3或4个精氨酸氨基酸残基可以被转化为瓜氨酸,其余的精氨酸氨基酸残基不被转化。因此,本发明的抗原可以具有1、2、3或4个瓜氨酸残基。本发明的抗原的长度可以至多为25个氨基酸。它们的长度可以是至少5个氨基酸,并且可以不超过18、19、20、21、22、23或24个氨基酸。本发明的T细胞抗原可以与肿瘤相关,并且可以刺激针对肿瘤的免疫反应。
本发明人已经表明,在正常健康供者和HLA转基因小鼠中,识别瓜氨酸化NPM肽的T细胞产生IFNγ,并且在用NPM肽刺激后可以被检测到。本发明人还已经表明,某些瓜氨酸化NPM肽在体内产生T细胞反应,因而可以作为用于癌症治疗的疫苗靶标。本发明人已经表明,在B16F1cDP4PAD2KO荷瘤小鼠中,抗肿瘤反应消失。这证明,PAD2对体内肿瘤细胞中精氨酸277的瓜氨酸化以及抗肿瘤效果至关重要。这些结果表明,PAD4在细胞核内使不同的残基瓜氨酸化以调控NPM的表达,但是PAD2(其主要在细胞质中表达)负责对NPM残基瓜氨酸化以用于MHC-II提呈。

这两种肽与小鼠同源,因此不被识别为外来物。
已知瓜氨酸化肽在具有共同HLA-DR4基序的自身免疫患者中刺激T细胞反应。相比之下,本发明人首次表明,瓜氨酸化NPM肽(如在277位瓜氨酸化的NPM 266-285和在277位瓜氨酸化的NPM 261-280)可以在HLA-DP4和DR4转基因小鼠中刺激强效的T细胞反应。显示对在277位瓜氨酸化的NPM 266-285产生反应的所有健康供者都表达HLA-DP4。此外,其中的两个供者还表达HLA-DR4。这使得该肽成为在更多群体中治疗血液肿瘤和实体肿瘤的有希望的疫苗。对在277位瓜氨酸化的NPM 266-285的反应是最强烈的,对未修改的野生型序列的反应性最小。识别这种瓜氨酸化肽抗原的T细胞可以靶向肿瘤细胞,并且在体内引起强烈的抗肿瘤效果,从而为瓜氨酸化NPM 266-285cit作为疫苗靶标以用于癌症治疗提供了首次证据。在277位瓜氨酸化的NPM 261-280针对表达MHC-II的肿瘤产生强烈的抗肿瘤反应,但是其针对不能表达MHC-II的肿瘤产生微弱的反应。这表明,在MHC-II阴性的肿瘤中,CD4 T细胞可以通过对CD8和/或NK反应的旁效应(bystander effect)来诱导抗肿瘤反应,但是在肿瘤表达MHC-II时,抗肿瘤反应更强烈,表明CD4 T细胞也可以介导直接的肿瘤杀伤。
MHC II类抗原加工途径可以受到许多因素的影响,如外源性抗原的内化和加工、每种MHC II类分子的肽结合基序以及MHC II类:肽复合物的输送和稳定性。MHC II类肽结合槽的两端是打开的,并且与MHC I类分子相比,它受肽长度的限制较少。与MHC II类分子结合的肽具有在13个至25个氨基酸的范围内的长度并且通常突出于MHC分子之外(Kim等人,2014;Sette等人,1989)。这些肽含有九个氨基酸的连续段,被称为核心区域。这些氨基酸中的一些氨基酸直接与肽结合槽相互作用(Andreatta等人,2017)。核心肽的任一侧的氨基酸突出于肽结合槽之外;这些氨基酸被称为肽侧翼区域。它们也可以影响肽结合和随后与T细胞的相互作用(Arnold等人,2002;Carson等人,1997;Godkin等人,2001)。MHC II类肽的长度允许将长肽(例如15至20聚体)用于筛选。例如,在NPM中,这将需要71种15聚体的重叠性肽;这些肽覆盖完整的294个氨基酸并且有11个氨基酸产生重叠(或者,如果使用15个氨基酸重叠的20聚体,则将需要56种20聚体的肽)。在这71种肽中,28种肽含有精氨酸残基。为了筛选28种肽,可以将这些肽组合成较小的肽库(peptide pool),并且将其纳入体外试验或用于体内鼠免疫接种研究。在设计新表位个性化疫苗以筛选数百种肽方面,这种类型的筛选是标准的。例如,Liu等人研究了上皮性卵巢癌患者对新抗原的反应(Liu等人,2019)。他们筛选了75种肽并且发现27种肽刺激T细胞反应。Bobisse等人筛选了776种肽,并且发现15种(2%)肽刺激T细胞反应(Bobisse等人,2018)。
这种方法还是鉴定MHC I类肽的可行途径,因为较长的20聚体肽也可以含有嵌入的MHC I限制性表位,并且已经被用于鉴定MHC II类和MHC I类限制性CD4和CD8 T细胞反应。鉴于使用这种用于鉴定个别癌症患者的癌症疫苗新抗原靶标的方法,其对于单一抗原是同样可行且合理的途径,以便开发疫苗来治疗其体内的肿瘤表达瓜氨酸化抗原的大量癌症患者。这将需要在多个供者中进行测试,以确保鉴定出与不同MHC II类分子和MHC I类分子结合的表位。将在每个供者中单独地或集合地使用相同的71种15聚体重叠肽或56种20聚体重叠肽。
MHC II类分子具有高度的多态性,肽结合基序是高度退化的,许多混合肽已经被鉴定为可以与多种MHC II类分子结合(Consogno等人,2003)。已经从MHC II类:肽复合物的晶体学研究中鉴定出对肽结合至关重要的氨基酸(Corper等人2000;Dessen等人1997;Fremont等人1996;Ghosh等人1995;Latek等人2000;Li等人2000;Lee,Wucherpfennig和Wiley 2001;Brown等人1993;Smith等人1998;Stern等人1994;Scott等人1998;Fremont等人1998)。这些研究已经表明,P1、P4、P6和P9总是指向MHC,然而P-1、P2、P5 P8和P11总是指向TCR。表1列出了HLA-DR和HLA-DP等位基因的频率(Thomsen和Nielsen 2012;Gonzalez-Galarza等人2015)。

相比之下,MHC I类分子示出更受限制的肽结合特性。通过预测算法分析已知的天然结合肽,也确定了对MHC I类结合至关重要的氨基酸(Jurtz等人,2017年),表明P2和P9(HLA-B*0801除外)指向MHC,充当结合锚定残基。
核仁磷酸蛋白的最普遍形式是NPM1.1。还存在两种替代性剪接同种型(被称为NPM1.3和NPM1.2),其中NPM1.1是所有组织中普遍存在的形式。肽LPKVEAKFINYVKNCFRMTD(261-280)和AKFINYVKNCFRMTDQEAIQ(266-285)只存在于NPM1.1(B23.1)和NPM1.3(B23.2)。因此,在277位瓜氨酸化的NPM 261-280和266-285以及编码其的核酸可以用于靶向NPM的最普遍形式(NPM1.1)。在NPM1.3中,相应的肽位于在248位瓜氨酸化的232-251和237-256。
NPM在已经克隆该基因的物种(鸡、小鼠、狗、羊、牛、马、猪和人)之间具有高度的保守性。因此,本发明的抗原(任选地与包含编码这种抗原的序列的核酸组合)可以用于治疗非人哺乳动物的癌症。
本发明在其范围内还包括具有上述氨基酸序列和与上述氨基酸序列具有显著同一性(例如,至少70%、至少80%、至少85%、至少90%、至少95%或至少99%的同一性)的序列的肽,以及肽在医药中的用途,特别是在治疗癌症的方法中的用途。这类肽优选地能够产生针对肿瘤的免疫反应,该肿瘤包括但不限于AML、肺肿瘤、结肠直肠肿瘤、肾肿瘤、乳腺肿瘤、卵巢肿瘤和肝脏肿瘤。两个氨基酸序列或两个核酸序列的同一性百分比通常通过以下来确定:为最佳比较的目的将序列对齐(例如,可以在第一个序列中引入空位以与第二个序列进行最佳对齐),并且比较在相应位置处的氨基酸残基或核苷酸。“最佳对齐”是指两个序列产生最高同一性百分比的对齐。同一性百分比是通过比较序列中相同的氨基酸残基或核苷酸的数目来确定(即,同一性%=相同位置数/位置总数×100)。

本发明的抗原可以在其C端和/或N端包括一个、两个或三个额外的氨基酸。本发明的抗原可以包括具有一个氨基酸置换和一个氨基酸插入、一个氨基酸置换和一个氨基酸缺失、或一个氨基酸插入和一个氨基酸缺失的上述氨基酸序列。本发明的抗原可以包括具有一个氨基酸置换、一个氨基酸插入和一个氨基酸缺失的上述的氨基酸序列。
插入的氨基酸和取代的氨基酸可以是天然存在的氨基酸,也可以是非天然存在的氨基酸,例如,可以包含非天然的侧链。这种改变的肽配体在(Douat-Casassus等人,2007;Hoppes等人,2014和其中的参考文献)中进一步讨论。如果置换和/或插入多于一个氨基酸残基,则取代/插入的氨基酸残基可以彼此相同或彼此不同。每种取代的氨基酸可以具有与被取代的氨基酸不同的侧链。
优选地,本发明的抗原在MHC分子的肽结合槽中与MHC结合。通常,上述的氨基酸修饰不会损害肽与MHC结合的能力。在优选的实施方式中,氨基酸修饰提高肽与MHC结合的能力。例如,可以在将肽锚定在MHC上的位置进行突变。这种锚定位置和这些位置的优选残基是本领域已知的。
本发明的抗原可以用于引起免疫反应。如果是这种情况,重要的是免疫反应对预期的靶标具有特异性,以避免可能与“脱靶”免疫反应有关的不期望的副作用的风险。因此,优选的是本发明的多肽的氨基酸序列不与来自任何其它蛋白质(尤其是另一种人蛋白质)的肽的氨基酸序列匹配。本领域的技术人员将了解如何搜索已知蛋白质序列的数据库以确定根据本发明的抗原是否存在于另一种蛋白质中。
本发明的抗原可以很容易地通过梅里菲尔德合成法(Merrifield synthesis,也称为固相合成法)或任何其它多肽合成方法来合成。GMP级多肽是由Multiple PeptideSystems(San Diego,CA)通过固相合成技术产生的。可选择地,如果需要的话,可以按照本领域已知的方法来重组生产肽。这种方法通常涉及使用包含编码要表达的多肽的核酸序列的运载体(vector),以在体内(例如,在细菌、酵母、昆虫或哺乳动物细胞中)表达多肽。可选择地,也可以使用体外无细胞系统。这种系统在本领域是已知的并且可以商业购买,例如从Life Technologies(Paisley,UK)商业购买。抗原可以被分离出来和/或可以被提供为基本纯的形式。例如,它们可以被提供为基本上不含其它多肽或蛋白质的形式。本发明的肽可以使用Fmoc化学或本领域技术人员已知的其它标准技术来合成。
在第二方面,本发明提供了第一方面的抗原与MHC分子的复合物。优选地,抗原与MHC分子的肽结合槽结合。该MHC分子可以是MHC II类。MHC II类分子可以是DP、DR或DQ等位基因,如HLA-DR4、DR1、DP4、DP2、DP5、DQ2、DQ3、DQ5和DQ6。优选是HLA-DR4和DP4。
本发明的抗原和复合物可以是分离的和/或以基本纯的形式。例如,抗原和复合物可以被提供为基本上不含其它多肽或蛋白质的形式。值得注意的是,在本发明的上下文中,术语“MHC分子”包括重组的MHC分子、非天然存在的MHC分子和功能等效的MHC片段,包括其衍生物或变体,前提是保持肽结合。例如,MHC分子可以与治疗性部分融合,附着在固体支撑物上,具有可溶性形式和/或多聚体形式。
生产可溶性重组MHC分子的方法在本领域是已知的,其中本发明的抗原可以与该可溶性重组MHC分子形成复合物。适合的方法包括但不限于从大肠杆菌细胞或昆虫细胞中进行表达和纯化。可选择地,MHC分子可以通过合成的方式或使用无细胞系统来生产。
本发明的抗原和/或抗原-MHC复合物可以与能够引起治疗性效果的部分缔合。这种部分可以是已知具有免疫原性的载体蛋白。钥孔戚血蓝素(KLH)是用于疫苗组合物的合适载体蛋白的实例。可选择地,本发明的抗原和/或抗原-MHC复合物可以与融合配偶体缔合。融合配偶体可以用于检测目的,或用于将所述抗原或MHC附着在固体支撑物上,或用于MHC低聚化。MHC复合物可以包含可将生物素加入其中(例如,使用BirA酶)的生物素化位点。其它合适的融合配偶体包括但不限于,荧光标签或发光标签、放射性标签、核酸探针和对比试剂、抗体、或产生可检测产物的酶。检测方法可以包括流式细胞术、显微术、电泳或闪烁计数。
本发明的抗原-MHC复合物可以被提供为可溶性形式,也可以通过附着至合适的固体支撑物而被固定化。固体支撑物的实例包括但不限于珠粒、膜、琼脂糖凝胶、磁珠、板、管、柱。可以将抗原-MHC复合物附着至ELISA板、磁珠或表面等离子体共振生物传感器芯片。将抗原-MHC复合物附着在固体支撑物上的方法是技术人员已知的,并且包括例如使用亲和结合配对,例如生物素和链霉亲和素,或抗体和抗原。在优选的实施方式中,抗原-MHC复合物用生物素标记并且附着至涂覆链霉亲和素的表面。
本发明的抗原-MHC复合物可以是多聚体形式,例如二聚体、或四聚体、或五聚体、或八聚体,或更大的多聚体。用于生产多聚体肽MHC复合物的合适方法的实例在(Greten和Schneck 2002)和其中的参考文献中进行描述。通常,抗原-MHC多聚体可以使用以生物素残基标记并且以荧光标记的链霉亲和素复合的抗原-MHC来生产。可选择地,多聚体抗原-MHC复合物可以通过使用免疫球蛋白作为分子骨架来形成。在该系统中,MHC分子的细胞外结构域与免疫球蛋白重链的恒定区融合,由短氨基酸接头分隔。通过利用载体分子(如葡聚糖)还已经生产出抗原-MHC多聚体(WO02072631)。多聚体抗原-MHC复合物可以用于改善对结合部分(如T细胞受体,由于亲和力效应,其与所述复合物结合)的检测。
本发明的抗原可以与MHC复合而提呈在细胞表面上。因此,本发明还提供了一种细胞,在该细胞的表面上提呈本发明的复合物。这种细胞可以是哺乳动物的细胞,优选地是免疫系统的细胞,尤其是专职抗原提呈细胞(如树突状细胞或B细胞)。其它优选的细胞包括T2细胞(Hosken和Bevan 1990)。提呈本发明的抗原或复合物的细胞可以被分离出来,优选地是以群体的形式,或被提供为基本纯的形式。所述细胞在天然状态下可能不会提呈本发明的复合物,或者所述细胞提呈该复合物的水平可以高于该细胞在自然界中的提呈水平。这类细胞可以通过用本发明的抗原脉冲(pulsing)所述细胞而获得。脉冲法包括使用通常在10-5M至10-12M范围的多肽浓度将细胞与抗原孵育几个小时。可以重组生产细胞。提呈本发明抗原的细胞可以用于分离T细胞和T细胞受体(TCR),该T细胞和T细胞受体被所述细胞激活或与所述细胞结合,如下文更详细地描述。
本发明的肽可以用Fmoc化学或本领域技术人员已知的其它标准技术来合成。
生产根据本发明的肽的另一种便利的方法是,通过使用表达系统中的核酸,来表达编码该肽的核酸。这类核酸构成本发明的另一个方面。
技术人员将能够确定对这类核酸进行的置换、缺失和/或添加,其仍将提供本发明的肽。核酸可以是DNA、cDNA或RNA,如通过克隆获得的或通过化学合成产生的mRNA。对于治疗性用途,核酸优选地是以能够在要治疗的对象中表达的形式。本发明的肽或本发明的核酸可以被提供作为分离和/或纯化的形式的分离物,或不含或基本不含天然地与其缔合的物质。在核酸的情形下,它可以不含或基本不含侧翼带有人基因组基因的核酸,但是可以含有一种或多种用于表达的调控序列。核酸可以是全部或部分合成的,并且可以包括基因组DNA、cDNA或RNA。在根据本发明的核酸包括RNA时,提及所示序列应理解为对RNA等同物(用U代替T)的提及。
考虑到可用的核酸序列和克隆,编码本发明的肽的核酸序列可以由技术人员轻易地制备,例如使用本文中包含的信息和参考文献以及本领域已知的技术(例如,参见(Sambrook 1989;Ausubel 1992))。这些技术包括:(i)使用聚合酶链式反应(PCR)来扩增例如来自基因组源的这种核酸的样本;(ii)化学合成;或(iii)制备cDNA序列。编码多肽的DNA可以以本领域技术人员已知的任何合适的方式进行产生和使用,包括通过获取编码DNA,鉴定要表达部分的任一侧的合适的限制性酶识别位点,并且从DNA中切出所述部分。然后,可以将该部分与标准的市售表达系统中的合适启动子可操作地连接。另一种重组方法是用合适的PCR引物来扩增DNA的相关部分。可以对序列进行修饰,例如使用位点定向诱变,以导致经修饰的肽的表达或考虑到用于表达核酸的宿主细胞中的密码子偏好性。
本发明还提供了以质粒、运载体、转录盒或表达盒形式的构建体,其包含至少一种如上所述的核酸。本发明还提供一种重组宿主细胞,其包含一种或多种如上所述的构建体。如前所述,编码本发明的肽的核酸构成本发明的一个方面,生产组合物的方法也构成本发明的一个方面,该方法包括表达编码核酸。可以通过在适当的条件下培养含有该核酸的重组宿主细胞来方便地实现表达。在通过表达进行生产之后,可以使用任何合适的技术来分离和/或纯化组合物,然后进行适当使用。

在一个实施方式中,将本发明的核酸整合到宿主细胞的基因组(如染色体)中。根据标准技术,可以通过包含促进与基因组重组的序列来促进整合。
本发明还提供了一种方法,该方法包括在表达系统中使用如上所述的构建体,以表达如上所述的多肽。

在提及与本发明复合物结合的结合部分使用时,在本文中使用的"特异性"通常是指结合部分不对本发明抗原-MHC复合物外的一种或多种替代性抗原-MHC复合物显示出任何显著结合的情况。

TCR也可以用成像化合物(例如适用于诊断目的的标签)进行标记。这种标记的高亲和力TCR在检测选自CD1-抗原复合物、细菌超抗原和MHC-肽/超抗原复合物的TCR配体的方法中是有用的,该方法包括使TCR配体与对TCR配体具有特异性的高亲和力TCR(或多聚体高亲和力TCR复合物)接触;并且检测与TCR配体的结合。在多聚体高亲和力TCR复合物中,如Zhu等人(Zhu等人,2006年)所述,(例如使用生物素化的异二聚体形成的)荧光链霉素(市售)可以用于提供可检测的标签。荧光标记的多聚体适用于FACS分析,例如以检测携带高亲和力TCR特异针对的肽的抗原提呈细胞。
根据本发明,本发明的含有瓜氨酸的NPM肽可以通过T细胞受体(TCR)用作癌症免疫治疗的靶标。TCR被设计用来识别在与MHC分子复合的APC的表面上展示的短肽抗原(Davis等人,1998)。鉴定含瓜氨酸的特定肽对于开发新的免疫疗法是有利的。例如,这种治疗性TCR可以用作可溶性靶向剂,用于向肿瘤递送细胞毒素剂或免疫效应剂的目的(Boulter等人,2003年;Liddy等人,2012年;McCormack等人,2013年),或者它们可以被用于对T细胞进行工程化以用于过继性疗法(June等人,2014年)。
本发明的TCR(或其多价复合物)可以替代性或额外地与治疗剂缔合(例如,共价地或以其它方式),例如,治疗剂可以是用于杀伤细胞的毒性部分,或免疫刺激剂(如白细胞介素或细胞因子)。与非多聚体野生型或高亲和力T细胞受体异二聚体相比,本发明的多价高亲和力TCR复合物可以具有增强的与TCR配体结合的能力。因此,根据本发明的多价高亲和力TCR复合物对于在体外或在体内追踪或靶向提呈特定抗原的细胞特别有用,并且也可用于作为生产具有此类用途的其它多价高亲和力TCR复合物的中间体。因此,高亲和力TCR或多价高亲和力TCR复合物可以被提供在药学上可接受的制剂中,以在体内使用。
高亲和力的TCR可以用于生产可溶性双特异性试剂。优选的实施方式是包含可溶性TCR(其通过接头与抗CD3特异性抗体片段融合)的试剂。WO10/133828中描述了进一步的细节,包括如何生产这种试剂。本发明的TCR可以作为治疗性试剂使用。在这种情况下,TCR可以是可溶形式,并且可以优选地与免疫效应物融合。合适的免疫效应物包括但不限于:细胞因子,如IL-2和IFN-α;超抗原及其突变体;趋化因子,如IL-8、血小板因子4、黑色素瘤生长刺激蛋白;抗体,包括与免疫细胞(如T细胞或NK细胞)上的抗原结合的其片段、衍生物和变体(如抗CD3、抗CD28或抗CD16);和补体激活剂。
本发明的结合部分可以是抗体。本文所用的术语"抗体"是指免疫球蛋白分子和免疫球蛋白分子的免疫活性部分,即含有与抗原特异性结合的抗原结合位点的分子,无论是天然的还是部分或全部经合成产生的。术语"抗体"包括抗体片段、衍生物、功能等效物和抗体的同源物、人源化抗体,包括包含免疫球蛋白结合结构域的任意多肽,无论是天然的还是全部或部分经合成产生的,以及具有为抗体结合结构域或与抗体结合结构域同源的结合结构域的任意多肽或蛋白质。因此,包含与另一种多肽融合的免疫球蛋白结合结构域或等效物的嵌合分子包括在内。嵌合抗体的克隆和表达描述于EP-A-0120694和EP-A-0125023中。人源化抗体可以是具有非人(例如鼠)抗体的可变区和人抗体的恒定区的经修饰的抗体。制备人源化抗体的方法描述于例如美国专利第5225539号中。抗体的实例是免疫球蛋白同种型(例如IgG、IgE、IgM、IgD和IgA)及它们的同种型亚类;包含抗原结合结构域的片段,如Fab、scFv、Fv、dAb、Fd;和双体抗体。抗体可以是多克隆的或单克隆的。单克隆抗体在本文中可被称为"mab"。
可以采用抗体,例如单克隆抗体,并且使用重组DNA技术来生产保持原始抗体的特异性的其它抗体或嵌合分子。这种技术可以涉及将编码免疫球蛋白可变区或抗体的互补决定区(CDR)的DNA引入到不同免疫球蛋白的恒定区或恒定区加框架区(例如,参见EP-A-184187,GB 2188638A或EP-A-239400)。杂合瘤(或产生抗体的其它细胞)可以经受基因突变或其它变化,这可能会或可能不会改变产生的抗体的结合特异性。
已经表明,整个抗体的片段可以发挥结合抗原的功能。结合片段的实例是:(i)由VL、VH、CL和CH1结构域组成的Fab片段;(ii)由VH和CH1结构域组成的Fd片段;(iii)由单个抗体的VL和VH结构域组成的Fv片段;(iv)由VH结构域组成的dAb片段(Ward等人1989);(v)分离的CDR区;(vi)F(ab')2片段,其是包含两个连接的Fab片段的二价片段;(vii)单链Fv分子(scFv),其中VH结构域和VL结构域经由肽接头连接,该接头使得两个结构域缔合以形成抗原结合位点(Bird等人,1988;Huston等人,1989);(viii)双特异性单链Fv二聚体(PCT/US92/09965);和(ix)"双体抗体",其是通过基因融合而构建的多价片段或多特异性片段(WO94/13804;(Holliger和Winter 1993))。双体抗体是多肽的多聚体,每个多肽包括第一结构域(其包含免疫球蛋白轻链的结合区)和第二结构域(其包含免疫球蛋白重链的结合区),这两个结构域连接在一起(例如,通过肽接头),但是不能彼此结合以形成抗原结合位点:抗原结合位点是由多聚体中一个多肽的第一结构域与该多聚体中另一个多肽的第二结构域结合形成的(WO94/13804)。在使用双特异性抗体时,这些抗体可以是传统的双特异性抗体,其可以用各种方法进行制备(Holliger和Winter 1993),例如用化学方法或由杂合瘤进行制备;或者这些抗体可以是上述的任何双特异性抗体片段。使用scFv二聚体或双体抗体而不是整个抗体可以是优选的。双体抗体和scFv可以被构建为不含Fc区,只使用可变结构域,从而潜在地减少抗独特型反应的影响。双特异性抗体的其它形式包括(Traunecker,Lanzavecchia,和Karjalainen 1991)中描述的单链"Janusins"。双特异性双体抗体,与双特异性整个抗体相对的,也可以是有用的,因为它们可以容易地在大肠杆菌中构建并表达。使用噬菌体展示(WO94/13804)可以容易地从文库中选择适当的结合特异性的双体抗体(和许多其它多肽,如抗体片段)。如果双体抗体的一个臂保持恒定的,例如,具有针对抗原X的特异性,那么在另一个臂变化时可以制备文库,并且选择适当特异性的抗体。“抗原结合结构域"是抗体的一部分,其包括与抗原的部分或全部特异性结合且互补的区域。在抗原大时,抗体可以只与抗原的某一特定部分结合,该部分被称为表位。抗原结合结构域可以由一种或多种抗体可变结构域提供。抗原结合结构域可以包括抗体轻链可变区(VL)和抗体重链可变区(VH)。
本发明还包括基于工程化的蛋白骨架的结合部分。蛋白质骨架来自于稳定的、可溶性、天然的蛋白质结构,这些结构已经被修改以提供感兴趣的靶标分子的结合位点。工程化的蛋白骨架的实例包括,但不限于:亲和体(affibody),它是基于葡萄球菌蛋白A的Z结构域,在其两个α-螺旋上提供了结合界面(Nygren 2008);Anticalin,源自脂钙蛋白,其在β-桶状折叠的开口端整合了针对小配体的结合位点(Skerra 2008);纳米抗体;和DARPin。工程化的蛋白质骨架通常旨在结合与抗体相同的抗原蛋白,并且是潜在的治疗剂。它们可以用于作为抑制剂或拮抗剂,或作为递送工具以将分子(如毒素)靶向到体内的特定组织(Gebauer和Skerra 2009)。短肽也可以用于与靶标蛋白质结合。Phylomer是来自细菌基因组的天然结构肽。这类肽代表蛋白质结构折叠的各种排列,并且可以用于抑制/破坏体内的蛋白质-蛋白质相互作用(Watt 2006)。
如所讨论的,本发明人已经发现,一些经修饰的NPM抗原与肿瘤有关,并且瓜氨酸化肽刺激T细胞反应,这可以用于产生针对肿瘤的免疫反应。本发明提供了用于医药的、第一方面的抗原、第二方面的复合物和/或第三方面的结合部分。第一方面的抗原、第二方面的复合物和/或第三方面的结合部分可以在用于治疗癌症的方法中使用。还提供了第一方面的抗原、第二方面的复合物和/或第三方面的结合部分在制备用于治疗癌症的药物中的用途;以及治疗癌症的方法,该方法包括向需要这种治疗的对象施用本发明的第一方面的抗原、第二方面的复合物和/或第三方面的结合部分。根据本发明的抗原可以单独使用,也可以作为库(pool)组合使用。此外,它们可以与其它治疗剂组合使用,如抗癌剂,包括但不限于检查点阻断药物,如伊匹单抗(ipilimumab)、派姆单抗(pembrolizumab)和纳武单抗(Nivolumab)。
本发明人首次表明,瓜氨酸化NPM肽可以刺激强效的T细胞反应。本发明提供了用于局部刺激针对对象的肿瘤组织的免疫反应的合适手段。针对这些NPMcit肽具有特异性的T细胞可以靶向肿瘤细胞,以在体内引发强烈的抗肿瘤效果,从而为NPMcit表位作为癌症治疗的疫苗靶标提供了首次证据。刺激针对疫苗靶标的免疫反应包括患者的天然免疫反应和旨在引导针对肿瘤的免疫反应的免疫治疗性治疗(例如,检查点抑制剂、针对肿瘤抗原的CAR-T和其它肿瘤免疫治疗)。在各种临床情况下,免疫反应的这种支持或诱导可以是有益的,以启动和维持免疫反应,并且避免往往阻碍这种激活的肿瘤介导的免疫抑制。这些反应可以耐受自身免疫性疾病的治疗。
在一些实施方式中,细胞免疫反应对应激诱导的翻译后修饰的肽具有特异性,其中免疫反应包括激活表达TCRαβ或γδ的T细胞。本发明还涉及TCR、单个TCR亚基(单独或组合)及其亚结构域、可溶性TCR(sTCR),例如可溶性αβ二聚体TCR,其在恒定结构域残基之间具有至少一个在原生TCR中不存在的二硫化物链间键,和克隆的TCR,所述TCR被工程化到自体或异体T细胞中或T细胞祖细胞中,以及制备其的方法,以及携带所述TCR的其它细胞。
癌症可以是乳腺癌(包括雌激素受体阴性乳腺癌)、结肠直肠癌、肺癌、卵巢癌、肾癌、肝癌和AML。
本发明提供了药物组合物,该药物组合物包含本发明的抗原、复合物和/或结合部分,该药物组合物用佐剂或其它药学上可接受的疫苗组分进行配制。在特定的实施方式中,佐剂是TLR配体,如CpG(TLR9)、MPLA(TLR4)、咪喹莫特(TLR7)、poly I:C(TLR3)或AMPLIVANTTLR1/2配体、GMCSF、油乳剂、细菌产品或完全失活的细菌。
抗原可以是T细胞抗原或B细胞抗原。可以将根据本发明的肽单独地使用或组合地使用。此外,它们可以与其它治疗剂组合使用,如抗癌剂,包括但不限于检查点阻断药物,如伊匹单抗。
根据本发明的抗原可以作为肽在体内被递送,可选地以WO02/058728中公开的肽的形式。本发明人已经出乎意料地发现,本发明的抗原在以肽的形式施用时引起强烈的免疫反应。这种肽可以只作为肽序列,或作为含有抗原的多肽,甚至作为全长蛋白质来进行施用。可选择地,根据本发明的抗原可以作为编码抗原的核酸、编码含有抗原的多肽的核酸或甚至编码全长蛋白质的核酸在体内进行施用。这类核酸可以是小基因的形式,即编码前导序列和抗原或前导序列和全长蛋白质。
如本文所用,术语"治疗"包括可以使人或非人动物受益的任何方案。抗原和/或核酸和/或复合物和/或结合部分可以与药学上可接受的一种或多种载体组合使用以形成药物组合物。这种载体可以包括但不限于生理盐水、缓冲盐水、右旋糖、脂质体、水、甘油、乙醇及其组合。
设想的是注射将是用于本发明组合物的治疗性施用的主要途径,尽管也可以使用导管或其它手术管进行递送。一些合适的施用途径包括静脉内、皮下、真皮内、腹膜内和肌肉内施用。可以在重构粉末制剂后再使用液体制剂。
对于静脉内注射或在疾病部位的注射,活性成分将是肠胃外可接受的水溶液的形式,该水溶液不含热原,具有合适的pH值,等渗并且保持稳定性。本领域的那些相关技术人员能够很好地制备合适的溶液,例如使用等渗溶媒,如氯化钠注射液、林格氏注射液或乳酸林格氏注射液。防腐剂、稳定剂、缓冲剂、抗氧化剂和/或其它添加剂可根据需要加入。
用于口服施用的药物组合物可以是片剂、胶囊剂、粉末剂或液体剂的形式。片剂可以包含固体载体,如明胶或佐剂。液体药物组合物通常包含液体载体,如水、石油、动物油或植物油、矿物油或合成油。生理盐水溶液、右旋糖或其它糖类溶液或二醇(如乙二醇、丙二醇或聚乙二醇)可以包括在内。在制剂是液体的情况下,它可以是例如含有pH为6.8-7.6的非磷酸盐缓冲液或冻干粉末的生理盐溶液。
组合物可以以局部方式施用于肿瘤部位或其它所需部位,也可以以靶向肿瘤或其它细胞的方式进行递送。在一些实施方式中,在不使用佐剂的情况下施用抗原,用于包括激活表达TCRαβ或γδ的T细胞的细胞免疫反应。
组合物优选地以"治疗有效量"施用给个体,该治疗有效量足够显示出对个体的益处。实际施用量以及施用的速度和时间进程将取决于所治疗的疾病的性质和严重程度。治疗的处方,例如关于剂量的确定等,属于全科医生和其它医生的职责范围内,并且通常要考虑到要治疗的病症、单个患者的病况、递送部位、施用方法和医生已知的其它因素。本发明的组合物与癌症治疗以及在最初的治疗或手术后预防此类疾病的复发特别相关。上面提到的技术和方案的实例可以在雷明顿药物科学(Remington's Pharmaceutical Sciences)(雷明顿1980)中找到。组合物可以单独施用,也可以同时地或依次地与其它治疗组合使用,这取决于要治疗的病况。其它癌症治疗方法包括本领域已知的其它单克隆抗体、其它化疗药剂、其它放疗技术或其它免疫疗法。本发明组合物的一种特殊应用是作为手术的辅助手段,即帮助降低肿瘤切除后癌症复发的风险。本发明的组合物可以全部地或部分地通过化学合成进行生产。可以按照公认的标准液相或优选的固相肽合成方法来容易地制备该组合物,这些方法的一般描述可广泛地获得(例如,参见《固相肽合成》第二版(Stewart1984),《肽合成的实践》(Bodanzsky 1984)和《应用生物系统430A用户手册》(ABI公司);或者组合物可以在溶液中进行制备,通过液相方法或通过固相、液相和溶液化学的任何组合,例如通过首先完成相应的肽部分,然后如果需要且适当,在去除任何存在的保护基团后,通过相应的碳酸或磺酸或其反应性衍生物的反应来引入残基X。
本发明的抗原、复合物、核酸分子、运载体、细胞和结合部分可以是非天然存在的和/或纯化的和/或工程化的和/或重组的和/或分离的和/或合成的。
本发明还提供了一种识别与本发明复合物结合的结合部分的方法,该方法包括使结合部分候选物与复合物接触,并且确定该结合部分候选物是否结合该复合物。

1.1.商业性mAb
抗IFNγ抗体(克隆XMG1.2)、抗小鼠CD4(克隆GK1.5)、抗小鼠CD8(克隆2.43)和抗人CD4(克隆OKT-4)购自BioXcell(美国)。抗人CD134(克隆REA621)和抗人CD8(克隆REA734)购自Miltenyi(德国)。抗人CD4(克隆RPA-T4)和抗人颗粒酶B(克隆GB11)购自ThermoFisher Scientific(美国),抗人IFNγ(克隆E780)购自eBioscience(美国)。
1.2.细胞系
先前已经描述过用功能性MHC II类DR4(DRB1*0401;T2 DR4)稳定地转染T细胞/B细胞杂合细胞系T2(Kovats等人,1997)。除非另有说明,否则鼠黑色素瘤B16F1细胞系、鼠胰腺pan02细胞系获得自美国典型培养物保藏中心(ATCC),并且在补充有10%胎牛血清(FCS)、L-谷氨酰胺(2mM)和碳酸氢钠缓冲液的RPMI培养基1640(GIBCO/BRL)中进行培养。鼠转基因TRAMP细胞获得自ATCC,并且在杜氏改良的Eagle培养基(DMEM)中进行培养,该培养基含有4mM L-谷氨酰胺,经调节以含有1.5g/L碳酸氢钠和4.5g/L葡萄糖,补充有0.005mg/ml牛胰岛素和10nM脱氢异雄酮,90%;胎牛血清,5%;Nu-Serum IV,5%。鼠乳腺癌细胞系PY8119和PY230获得自ATCC并且在Ham's F12 Kaighn培养基(含5%FBS)中进行培养,PY230细胞系还在存在0.1%的MITO+Serum Extender(Corning)的情况下培养。人细胞系HeLa和小鼠细胞系LLC2获得自ATCC,并且在补充有10%胎牛血清的DMEM培养基中培养。ID8细胞系由美国堪萨斯大学医学中心(KUMC)的K.Roby博士提供,并且在补充有10%胎牛血清的DMEM中培养。
1.3.免疫原
1.3.1.肽
纯度大于90%的肽由Peptide Synthetics(费勒姆,英国)进行合成,并且以0.2mg的等分试样冻干储存在-80℃中。在使用当天,以10%的二甲基甲酰胺将其重构至适当的浓度。
1.4.质粒和转染
先前已经描述过pVitro 2嵌合性和诱导性HLA-DR4质粒的构建(Brentville等人,2016;Metheringham等人,2009)。为了生成HHDII质粒,由从EL4-HHD细胞中分离出的总RNA合成cDNA。将该cDNA用作模板,使用正向和反向引物来扩增HHD,并且将HHD亚克隆到pCR2.1中。然后将该HHD链(其包括人HLA-A2前导序列、通过甘氨酸丝氨酸接头与人HLA-A*0201MHCI类分子的α1和α2结构域共价连接的人β2-微球蛋白(β2M)分子、以及鼠H-2Db I类分子的α3结构域、跨膜结构域和胞浆结构域)插入至哺乳动物表达运载体pCDNA3(获得自Invitrogen)的EcoRV/HindIII位点。
使用无内毒素的Qiagen maxiprep试剂盒(Qiagen,克劳利)来生成无内毒素的质粒DNA。
利用先前描述的方案(Brentville等人,2016年),使用Lipofectamine转染试剂(Invitrogen),对细胞系进行转染。使用ZFN技术(Sigma)将B16F1细胞的鼠MHC-I和/或MHC-II敲除,并且使用pVitro 2嵌合质粒来转染组成型HLA-DP4。还用HHDII质粒来转染细胞,该HHDII质粒包括人HLA-A2前导序列、通过甘氨酸丝氨酸接头与人HLA-A*0201MHC I类分子的α1和α2结构域共价连接的人β2-微球蛋白(β2M)分子、以及小鼠H-2Db I类分子的α3结构域、跨膜结构域和胞浆结构域,相关部分如前所述(Xue等人,2016年)。还用pVITRO2人HLA-DP4质粒和IFNγ诱导性质粒pDCGAS来转染B16F1 HHDII细胞,人HLA-DP4如前所述(Brentville等人,2019)。
1.6蛋白质印迹
将细胞裂解液配制于含有蛋白酶抑制剂混合物(Sigma)的RIPA缓冲液中,在4%至12%的NuPAGE Bis-Tris凝胶(Invitrogen)上分离出蛋白质,然后将蛋白质转移到PVDF膜上。用3%BSA封闭膜1小时,然后用1:1000的针对人NPM的抗体(克隆FC82291,Abcam)和1:15000的针对β肌动蛋白的抗体(克隆AC-15,Sigma)进行探测。使用荧光二抗IRDye 800RD和IRDye 680RD抗小鼠二抗(用于β肌动蛋白)将蛋白质可视化。使用Licor Odyssey扫描仪对膜进行成像。将NPM蛋白用作阳性对照(ab114194,Abcam)。
1.7体外瓜氨酸化
在0.1M Tris-HCl pH 7.5(Fisher)、10mM CaCl2(Sigma)和5mM DTT(Sigma)中进行NPM的瓜氨酸化。溶液的终浓度为376mM Tris-HCl(pH 7.5),3.76mM CaCl2,1.88mM DTT。将样本与PAD酶在37℃下孵育2小时,然后在-80℃下储存过夜或直到使用。使用终浓度为148mU的PAD2酶和终浓度为152mU的PAD4。PAD酶购自Modiquest,hPAD2为37mU/μl和hPAD4为38mU/μl。
1.8质谱分析法
以比率为1:50的胰蛋白酶与蛋白质,通过胰蛋白酶消化在37℃过夜来制备样本。然后,将样本在真空下干燥,重新悬浮于0.1%甲酸/5%乙腈的LCMS级水中,随后进行MS分析。对于MS分析,通过自动进样器(Eksigent Ekspert nanoLC 425LC系统,采用1-10μl/min的泵模块,以5μl/min运行),以2min的洗涤阱/洗脱(wash trap/elute)配置,将样本注射至35℃的柱温箱中的YMC Triart C18柱(300μm内径,3μm颗粒大小,15cm)中。在87分钟的运行时间内,对样本进行梯度洗脱并通过电极为50μm的Duospray(TurboV)源进入SCIEX6600TripleT质谱仪中。6600被设置为IDA模式(独立数据采集/数据依赖采集),以用于每个循环碎裂的30个离子。总循环时间为1.8s,TOFMS扫描为250ms累积;每个产物离子扫描为50ms。
使用PEAKS Studio 8.0(Bioinformatic Solutions Inc.滑铁卢,加拿大)进行分析数据,搜索SwissProt human(Uniprot人工注释/整理)数据库,25ppm母体质量误差容限,0.1Da片段质量误差容限,搜索针对瓜氨酸化(R)、脱酰胺化(NQR)、氧化(M)的修饰。在5%的最低离子强度下,将位点鉴定为确信的修饰位点。
1.7免疫接种
1.7.1.免疫接种方案
使用HLA-DR4小鼠(Taconic,美国)和专利WO2013/017545A1中描述的HHDII/HLA-DP4转基因品系小鼠(欧洲突变体小鼠保藏库(EMMA),法国),小鼠在8周龄至12周龄之间,并且由诺丁汉特伦特大学的工作人员照顾。所有工作都是在内政部项目许可(Home Officeproject licence)下进行。用10%的二甲基甲酰胺将肽溶解至1mg/mL,然后用佐剂CpG和MPLA(Invivogen,英国)进行乳化(一系列稀释度),每种佐剂6μg/小鼠。将肽(25μg/每只小鼠)经皮下注射至尾巴底部。
对于肿瘤挑战实验,在初次免疫接种前3天,用1x105的B16 HHDII/iDP4细胞、B16F1HHDIIMHCIIKO细胞或B16 HHDII/PAD2KOcDP4细胞在右侧经皮下方式以挑战小鼠,然后按上述方法进行免疫接种。以3至4天的时间间隔来监测肿瘤生长,一旦肿瘤直径达到≥10毫米,则人道地对小鼠实施安乐死。
1.8免疫反应的分析
1.8.1动物组织的分离和分析
将脾脏分解并用红细胞裂解缓冲液处理2分钟。收获肿瘤并且进行机械分解。
1.8.2外周血单核细胞(PBMC)的分离
将外周血样本抽入到锂肝素管(Becton Dickinson)中并且在静脉穿刺后立即处理。使用Ficoll-Hypaque通过密度梯度离心以分离出PBMC。分离之后立即进行PBMC的增殖和培养的ELISpot测定。
1.8.3体外ELISpot测定
根据制造商的说明(Mabtech,瑞典),使用鼠IFNγ捕获和检测试剂来进行ELISpot测定。简言之,将抗IFNγ抗体涂覆在96孔Immobilin-P板的孔中。将合成肽(具有多种浓度)和每孔5x105个脾细胞以一式三份加入到板的各孔中。将5μg/mL的LPS作为阳性对照。以一式三份在相关处每孔添加5x104个肽脉冲的靶细胞,并在37℃下孵育板40小时。孵化完成后,用生物素化的抗IFNγ抗体来检测捕获的IFNγ,并且用链霉素碱性磷酸酶和发色底物进行显影。将脂多糖(LPS;5μg/mL)用作阳性对照。对于阻断研究,使用20μg/mL的抗CD4阻断抗体(RPA-T4)和抗CD8阻断抗体(2.43)(来自Bioxcell公司)。使用自动化酶标仪(CellularTechnologies Ltd)对斑点进行分析和计数。
1.9增殖测定
将外周血样本(约50mL)抽入至锂肝素管(Becton Dickinson)中。将样本在室温下保存,并且在静脉穿刺后立即处理。使用Ficoll-Hypaque通过密度梯度离心来分离出PBMC。在PBMC分离之后,立即进行PBMC的增殖测定。常规地源自健康供者样本的PBMC的中位数为1.04×106个PBMC/mL全血(范围:0.6×106/mL至1.48×106/mL)。通过台盼蓝排除法所评估的活力中位数为93%(范围为90-95%)。
以羧基荧光素琥珀酰亚胺酯(CFSE)(ThermoFisher)承载新鲜分离出的PBMC。简言之,用5mM母液的DMSO溶液制备在50μM储备溶液的温热PBS溶液。将CFSE快速地加入到PBMC(5×106个细胞/mL的承载缓冲液(PBS加5%v/v热灭活胎牛血清))中,以达到5μM的终浓度。将PBMC在室温下在黑暗中孵育5分钟,然后用过量的(x10 v/v体积)承载缓冲液洗涤(300gx 10mins)两次,以去除无细胞结合的CFSE。在完全培养基中使细胞达到1.5×106/mL,然后用含有溶媒(阴性对照)、PHA(阳性对照,终浓度为10μg/mL)或上述肽(10μg/mL)的培养基进行铺板和刺激。
在第7天至第11天,从培养物中取出500μL细胞,在PBS中洗涤,用1:50稀释度的抗CD4(PE-Cy5,克隆RPA-T4,ThermoFisher)、抗CD8 efluor 450(克隆RPA-T8,ThermoFisher)和抗CD134(PE-Cy7,克隆REA621,Miltenyi)染色。根据制造商的说明,使用细胞内固定/透化缓冲液(ThermoFisher),对细胞进行洗涤、固定和透化。使用1:50稀释度的抗IFNγ(克隆4S.B3,ThermoFisher)或抗颗粒酶B(PE,克隆GB11,Thermofisher)进行细胞因子的细胞内染色。在MACSQuant 10流式细胞仪(其配备有MACSQuant软件2.8.168.16380版本)上对染色的样本进行分析,使用染色的溶媒刺激的对照来确定合适的门。
1.10 FACS细胞分选
在第10天,将培养孔中的内含物轻轻地混合,汇集在一起(根据肽刺激)并且在PBS中洗涤(300gx 10mins)。将沉淀物轻轻地重新悬浮在500μL的含有10μL的抗CD4 eFluo450(克隆RPA-T4,ThermoFisher,货号48-0049-42)和10μL的抗CD8 APC(克隆RPA-T8,ThermoFisher,货号17-0088-41)的PBS中。将细胞在4℃下染色30分钟,然后以1.0ml的PBS洗涤(5min x 300g),并且重新悬浮在300μl的FACS分选缓冲液中(补充有1mM EDTA、25mMHEPES和1%v/v HI FCS的PBS)。从每个染色的样本中取出10μl的样本,并且加入90μl的FACS分选缓冲液。在MACSQuant Analyser 10流式细胞仪上收集10000个事件,以确定增殖。将剩余的细胞用于批量FACS分选。
在MoFlo XDP高速细胞分选机中使用无菌条件对细胞进行分选。将所有样本分选到1.0ml的RNA保护液(5份Qiagen的保护液:1份Sigma的FACS分选缓冲液)中,分离出CD4+ve/CFSE高群体和CD4+ve/CFSE低群体。将分选的细胞(混合地)储存在-80℃。
对识别含有瓜氨酸的NPM肽的TCR的α链和β链配对进行测定。将在RNA保护液中的来自CD4+ve/CFSE高群体和CD4+ve/CFSE低群体的分选出的细胞(混合地)运送到iRepertoire Inc(Huntsville,AL,USA)以对TCRA链和TCRB链进行NGS测序,以证实在CD4+ve/CFSE低细胞(其针对肽进行增殖)中的TCR的扩增,与不增殖的CD4+ve/CFSE高群体进行比较。简言之,对分选的细胞的RNA进行纯化,实施RT-PCR,然后使用人TCRα和β250PER引物对(iRepertoire Inc.,Huntsville,AL,USA)对cDNA进行扩增子拯救多重PCR(ARM-PCR)。引物对的信息可以获得自美国专利和商标局(专利号:7,999,092和9,012,148B2)。对PCR/DNA样本进行评估后,将10个样本文库汇集起来,并且使用Illumina MiSeq平台(Illumina,SanDiego,CA,USA)进行测序。使用IRweb软件(iRepertoire)分析原始数据。使用iRweb工具对V基因、D基因和J基因的使用和CDR3序列进行鉴定和分配并且生成树状图。树状图将每个独特的CDR3显示为一个彩色矩形,每个矩形的大小对应于储库中每种CDR3的丰度,并且位置根据V区的使用来确定。
为了阐明TCRα链和TCRβ链的同源配对和测序,IRepertoire使用其iPairTM技术,将CD4+ve/CFSE低的细胞群体(批量分选,同时地进行批量测序)以1个细胞/孔接种到iCapture 96孔板中。进行RT-PCR并且可以用扩增子拯救多重PCR(arm-PCR)从单个细胞中扩增出TCRα链和β链。可以利用iPairTM软件程序来分析数据,用于特定链配对的频率,并且基于与大量数据的比较将序列进行排序。
1.11敲除PAD2
通过Sigma-Aldrich Cell Design StudioTM进行B16F1细胞的PAD2敲除。CompoZr锌指核酸酶(ZFN)技术用于靶向具有配对序列NM008812-r649a1:CTGCAGCCGCACGGTCCGTTCCCGCAGC和NM008812-656a1:TGGGAGCCGCGTGGAGGCGGTGTACGTG的NPM外显子1。经过几轮重定向,达到89%的切割,然后进行单细胞克隆,以建立稳定的克隆。通过Sigma-Aldrich Cell Design StudioTM使用ddPCR和流式细胞术(ab50257和ab150063,Abcam)来评估克隆的PAD2的敲除。用于ddPCR的引物和探针来自Thermo fisher自营(Mm00447012_m1&Mm00447020_m1)。
1.12统计方法
数据以每百万个脾细胞的斑点数表示。平均数和标准差(SD)是由一式四份的读数计算出来的。还计算了每组三只小鼠的平均数和SD。在适当的情况下,使用GraphPad Prism6软件进行方差分析。
实施例1-核仁磷酸蛋白的序列比对和同源性
在哺乳动物中,NPM最普遍的形式是NPM1.1,存在两种替代性剪接的同种型(NPM1.2和NPM1.3),这两种是更短的NPM版本,但是具有高度的同源性(图1)。
实施例2-HHDII/DP4和HHDII/DR4小鼠对核仁磷酸蛋白表位的T细胞反应
由于胸腺内的耐受性和T细胞缺失,针对肿瘤相关表位的T细胞反应往往很弱或不存在。在HLA-DR4和HHDII/DP4转基因小鼠中筛选瓜氨酸化的NPM肽以用于它们刺激IFNγ反应的能力。选择每个含有精氨酸的肽,并且将精氨酸残基置换为瓜氨酸(cit)。表2总结了所选择的肽。
筛选核仁磷酸蛋白肽的反应
进行筛选以鉴定可以在小鼠体内产生免疫反应的潜在瓜氨酸化的NPM表位。用4至6种人瓜氨酸化肽的库对小鼠进行免疫接种。为了减少可能的交叉反应的影响,挑选每个库中的肽,以使得它们不包含任何重叠的氨基酸序列。将每个肽库作为单次免疫接种经皮下施用,每周给与一次,持续三周。每个肽库包含25μg的每种肽,与作为佐剂的CpG/MPLA组合使用。在第三次免疫接种后第7天处死小鼠,通过体外IFNγELISpot来评估对免疫库中每种肽的免疫反应(图2)。我们先前已经表明,瓜氨酸化肽可以在转基因DR4小鼠品系中诱导出反应。鉴于不同的小鼠品系具有不同的MHC储库,使用了两种不同的转基因品系(DR4和DP4)进行筛选。
在HHDII/DP4和HLA-DR4转基因小鼠中检测到针对人NPM瓜氨酸化肽的显著IFNγ反应(图2)。在HHDII/DP4小鼠中(A),NPM瓜氨酸化肽261-280和266-285诱导了显著的免疫反应(分别为p=<0.0001和p=<0.0001)。相比之下,肽186-205和肽191-210所包含的在氨基酸197(B细胞表位)的已知瓜氨酸化位置不能诱导免疫反应。在HLA-DR4小鼠中,仅在响应于NPM瓜氨酸化肽266-285中才检测到显著的IFNγ反应(B)(P=<0.0001),在这些小鼠中没有检测到针对任何其它NPM肽的反应。也检测不到针对含有在197位的瓜氨酸化残基的所述B细胞表位所产生的免疫反应(Tanikawa等人,2009)。

用25μg的NPM 261-280cit肽或266-285cit肽对小鼠进行免疫接种,每周一次,持续三周,来确认核心表位。用单个肽对HHDII/DP4小鼠进行免疫接种,而用NPM 261-280cit和266-285cit的组合对HLA-DR4小鼠进行免疫接种,所有这些肽都与CpG/MPLA一起给予。在第三次免疫接种后第7天将小鼠处死。通过体外IFNγELISpot来评估针对NPM 261-280cit和NPM 266-285cit的免疫反应,以及针对NPM 266-280cit(建议的核心表位)的反应(图3)。在HHDII/DP4中检测到针对人NPM 261-280cit肽和NPM 266-285cit肽的显著IFNγ反应,以及在HLA-DR4转基因小鼠中检测到针对NPM 266-285cit肽的显著IFNγ反应。在HHDII/DP4转基因小鼠中,针对核心肽NPM 266-280cit的反应和针对NPM 261-280cit或NPM 266-285cit的反应之间没有显著的差异。在HLA-DR4转基因小鼠中,针对NPM 266-280cit的反应和针对NPM 266-285cit的反应之间存在显著的差异,这表明在HLA-DR4小鼠中,核心肽是不同的并且可能包括氨基酸QEAIQ(位置281-285)。
266-285cit肽在HLA-DR4和HHDII/DP4小鼠中都产生了最强烈的免疫反应(图2A和2B)。将进一步的探究集中在该肽上。
为了确定针对NPM 266-285肽的免疫反应是否对瓜氨酸化肽而不是对野生型(wt)版本具有特异性,用NPM 266-285cit肽或NPM 266-285wt肽对小鼠进行免疫接种。HHDII/DP4小鼠经皮下接受25μg肽(NPM 266-285cit或NPM 266-285wt),每周一次,持续三周。在第三次免疫接种后第7天将小鼠处死,通过体外ELISpot来评估对每种肽的免疫反应(图4A和4B)。在用NPM 266-285wt免疫接种的小鼠中检测到低到中度的IFNγ反应,并且该反应与瓜氨酸化肽发生交叉反应(图4B),这些反应对NPM 266-285wt肽不具有特异性,因为用NPM266-285cit肽刺激细胞时也出现类似的反应。相比之下,在用NPM 266-285cit免疫接种的小鼠中检测到强烈的IFNγ反应,这些反应相比于针对NPM 266-285wt肽的反应(P=0.0002)和对照(P=<0.0001)具有显著性。这证实在HHDII/DP4小鼠中产生的免疫反应是针对NPM 266-285肽的瓜氨酸化版本的反应。
实施例3-在肿瘤细胞上呈现的Cit核仁磷酸蛋白肽可以是用于肿瘤治疗的靶标
本发明人已经通过蛋白印迹法确定,黑色素瘤B16F1细胞系组成性地表达NPM并且NPM的体外瓜氨酸化在277位上产生瓜氨酸(图5A和5B)。接下来,在体内评估了NPM 266-285cit肽免疫接种的抗肿瘤效果。评估了NPM 266-285(cit和wt)免疫接种对转染有IFNγ诱导性人DP4(iDP4)的小鼠B16黑色素瘤细胞系生长的影响。用B16 HHDII/iDP4肿瘤细胞挑战小鼠,3天后,用NPM 266-285wt或NPM 266-285cit进行免疫接种。用NPM 266-285cit肽免疫接种的小鼠比仅用CpG/MPLA免疫接种的对照小鼠具有显著的存活优势(图6A)。对照小鼠在第50天示出0%的存活,而NPM 266-285cit免疫接种的小鼠示出65%的存活(p=<0.0001),用266-285wt免疫接种的小鼠也示出显著的30%存活(p=0.0018),这再次表明经野生型肽刺激的T细胞可以与瓜氨酸化表位交叉反应并且识别肿瘤。不存在相关的毒性。与对照组(中位数为1425mm3)相比,NPM 266-285cit免疫接种的小鼠在肿瘤植入后第24天的肿瘤体积也显著更低(图6B,中位数为0mm3)(p=<0.0001)。与对照组(中位数为1425mm3)相比,NPM 266-285wt免疫接种的小鼠的肿瘤体积也显著更低(中位数为34mm3)(p=0.0152)。
实施例4--健康人供者和癌症患者中对NPM的反应
在HHDII/iDP4小鼠中,针对NPM 266-285cit肽的反应不能在免疫接种后第2天检测到,但是可以在免疫接种后第12天检测到。这表明这些反应是原生的反应,并且在这些小鼠中没有预先存在的免疫力。这就提出了一个问题,即人是否具有或能否产生针对NPM266-285cit肽的免疫反应。为了研究这个问题,将PBMC从10个健康供者身上分离出来,并且在存在NPM 266-285cit肽的情况下进行培养。九个供者是HLA-DP4阳性,其中两个供者还是HLA-DR4阳性,另外一个供者(第10个供者)是HLA-DP4和HLA-DR4阴性(阴性对照)。
将来自10个健康供者的PBMC用羧基荧光素琥珀酰亚胺酯(CFSE)进行标记,随后在NPM 266-285cit肽存在下进行体外培养。在第7天和第10天,用抗CD4和抗CD8荧光体偶联抗体对细胞进行染色,然后用流式细胞术来评估增殖(图7A)。在第7天,在10个供者(9个供者是HLA-DP4阳性)中的4个供者中可以检测到CD4 NPM 266-285特异性增殖(CFSE低)群体,在第10天增加,其中10个供者(9个供者是HLA-DP4阳性)中的7个供者示出特异性反应(图7B)。在第10天,对示出良好CD4 NPM 266-285特异性增殖反应的7个供者进行功能分析。测定了所有供者中的IFNγ、颗粒酶B和CD134的表达,将增殖型CD4 T细胞和非增殖型CD4 T细胞进行比较(图7C、D和E)。所有这7个供者的增殖性CD4 NPM 266-285特异性T细胞表达出颗粒酶B。此外,7个供者中的6个供者还表达出IFNγ和CD134,这种表达仅与大多数供者(7个供者中的6个供者)中的增殖性T细胞有关。

这些结果表明,大多数健康供者能够对NPM 266-285cit肽产生CD4增殖性反应,这也与细胞毒活性相关性功能标志物的上调有关。来自一些癌症患者的PBMC能够针对NPM266-285cit肽产生CD4反应,但是其数量低于反应性健康供者产生的数量。这种较低的频率可能是由于这些患者中的药物或某种程度的肿瘤介导的免疫抑制。
实施例5--健康人供者中原生T细胞群对NPMcit肽反应
将PBMC从两个健康供者中分离出来并且分成两部分,耗尽一部分中的CD45RO细胞,另一部分保持CD45RO细胞不被耗尽。将PBMC用羧基荧光素琥珀酰亚胺酯(CFSE)进行标记,随后在NPM 266-285cit肽存在下进行体外培养。在第11天,用抗CD4和抗CD8荧光色素偶联抗体对细胞进行染色,然后用流式细胞术来评估增殖(图8)。在第11天,在CD45RO耗尽群体和CD45RO未耗尽群体中都可以检测到CD4 NPM 266-285cit特异性增殖(CFSE低)群体。在CD45RO耗尽群体中,增殖性CD4 T细胞的百分比更高(23.6%),这表明原生T细胞对NPMcit肽产生反应。
实施例6--不同物种间核仁磷酸蛋白的同源性
核仁磷酸蛋白在小鼠、狗、羊、牛、马、猪和人之间具有高度的保守性(图9)。由于该疫苗在人和小鼠中诱导出T细胞反应并且在小鼠中诱导出抗肿瘤反应,则可以设想在其它物种中也将出现类似的反应。
实施例7--PAD2负责体内肿瘤中的精氨酸277的瓜氨酸化
为了确定是PAD2酶还是PAD4酶对体内瓜氨酸化负责,产生了缺乏PAD2的B16肿瘤细胞系(B16F1cDP4PAD2KO)。敲除PAD4酶是不成功的,因为敲除PAD4后细胞不能生长。将缺乏PAD2酶的B16F1cDP4PADKO肿瘤细胞植入转基因HLA-DP4小鼠中。在用NPM266-285cit与CPG/MPLA的组合免疫接种后对肿瘤生长进行评估,并且与CPG/MPLA对照组的肿瘤生长进行比较(图10)。总体上,在与未免疫接种的CPG/MPLA对照组比较时,用NPM266cit免疫接种的B16F1cDP4PADKO荷瘤小鼠不具有显著的存活优势(P=0.6826,图10A)。在第28天计算肿瘤体积(图10B),并且CpG/MPLA对照小鼠(中位数为190.5mm3)和NPM266-285cit免疫接种的小鼠(中位数为696.6mm3)之间不存在显著差异(P=0.2050)。B16F1cDP4PADKO荷瘤小鼠中的抗肿瘤反应消失,这表明PAD2对体内肿瘤细胞中精氨酸277的瓜氨酸化和抗肿瘤效果至关重要。
实施例8--MHC II类对用NPM 266-285cit免疫接种后的抗肿瘤反应必不可少
为了确定MHC II类对在用NPM 266-285cit免疫接种后的荷瘤小鼠中观察到的抗肿瘤反应是否是必不可少的,将B16细胞系工程化为敲除MHC-II(B16F1HHDIIMHCIIKO)。将缺乏MHC-II的B16F1HHDIIMHCIIKO肿瘤细胞植入至转基因HHDII小鼠中。在用NPM266-285cit与CPG/MPLA组合进行免疫接种后评估肿瘤生长,并且与CPG/MPLA对照组的肿瘤生长进行比较(图11)。总体上,在与CPG/MPLA对照组比较时,用NPM266-285cit免疫接种的B16F1HHDIIMHCIIKO荷瘤小鼠的总存活提高了10%(P=0.0144,图11A)。在28天计算肿瘤体积(图11B),并且CpG/MPLA对照小鼠(中位数为1027mm3)和NPM266-285cit免疫接种小鼠(中位数为904.3mm3)之间不存在显著差异(P=0.2050)。针对MHC-II KO肿瘤的稍微提高的存活率表明CD4 T细胞可以通过增强CD8和/或NK反应的旁效应来诱导抗肿瘤反应,但是肿瘤表达MHC-II时更优的抗肿瘤反应表明CD4 T细胞也可以介导直接杀伤肿瘤。

Claims (12)

3.根据权利要求1或权利要求2所述的抗原与MHC分子的复合物,可选地其中,所述MHC分子是MHC II类,可选地选自HLA-DR4和DP4。

5.根据权利要求4所述的结合部分,在所述结合部分与MHC复合时,所述结合部分结合所述多肽。

10.根据权利要求9所定义使用的所述抗原、所述复合物和/或所述结合部分,其中,所述癌症是AML、肺肿瘤、结肠直肠肿瘤、肾肿瘤、乳腺肿瘤、卵巢肿瘤和肝脏肿瘤。
11.一种药物组合物,包含权利要求1或权利要求2所定义的抗原、权利要求3所定义的复合物、和/或权利要求4-7中任一项所定义的结合部分,以及药学上可接受的载体。
12.一种用于鉴定与权利要求3所述的复合物结合的结合部分的方法,所述方法包括使结合部分候选物与所述复合物接触,并确定所述结合部分候选物是否结合所述复合物。
