Evaluation of Effect

1. Effect on gastrointestinal motility in young rats
1. Experimental Material
1.1 Experimental animals
Weaning (3 weeks old) SD young rats, male, SPF grade, body weight 60-70g; each squirrel cage is hung with an identity card with information such as project numbers, experimental groups, names of experimenters, animal species, sexes and the like, and the tail root of the rat is marked with a line;
the young rats are raised in an animal room at the raising temperature of 23 +/-2 ℃, the relative humidity of 40-70 percent and 12-hour light and shade alternation; animals are raised in 5 cages each time, and padding is replaced twice each week; feeding SPF rat growth and reproduction feed in the adaptation period (3 days); water for experimental animals is filtered by autoclaving; animals used in the experiment will remain healthy; animals ingest and drink water freely during the experiment;
1.2 Experimental drugs
The tested drugs are: the traditional Chinese medicine composition (source: jilin Huakang pharmaceutical industry Co., ltd.) in each embodiment of the invention;
positive drug: jianweining oral liquid for children (source: jilin Huakang pharmaceutical industry GmbH; national Standard B20020507; specification: 10 ml/count);
blank control: sterilizing water for injection;
1.3 Experimental reagents
Sodium carboxymethylcellulose (CMC-Na) (viscosity: 800-1200mpa.s, USP grade; specification: 500 g/bottle);

2.1 Experimental groups and dosing regimens
2.1.1 basis for dose selection
Clinical dosage: the children's stomachic oral liquid is 10ml (containing 16g crude drug) once, 3 times a day;
the high dose and the low dose of the rat are respectively increased and decreased by 2 times by taking 50g crude drug/kg as a medium dose;
2.1.2 grouping
After the adaptation period, 110 rats with good health condition are selected and divided into 11 groups according to a random digital table method, wherein each group comprises 10 rats, and the rats are respectively a blank control group, a model control group, a positive control group, a comparative example 1-3 group and an example 1-3 group;
2.1.3 preparation of medicinal liquid
The traditional Chinese medicine composition in each embodiment of the invention comprises the following components: the composition of example 3 was administered by gavage at 1mL/100g, wherein the composition was administered in three dose groups of 100g crude drug/kg (large dose), 50g crude drug/kg (medium dose) and 25g crude drug/kg (small dose) at 0.5% CMC-Na, and the remaining examples were administered in the medium dose group, stored at 4 ℃ and mixed before use every day;
oral liquid for invigorating stomach of children: taking an appropriate amount of the extract for invigorating stomach of children, preparing with 0.5% of CMC-Na, preserving at 4 deg.C, and mixing well before use every day;
2.2 Molding
Except the blank control group, the other groups adopt a method of chronic restraint stress, excessive fatigue and diet loss to mold, namely an FD model; placing the molded rat in a binding box for binding for 3h every morning, swimming for 10min in the afternoon, fasting every other day, feeding enough food every other day, and holding tail to irritate the rat every other day for 30min for 3 weeks; observing general conditions of animals every day, including hair color, mental state, emotional reaction, defecation and the like for 1 time, and measuring the food intake and the weight of the animals for 1 time every week;
2.3 preparation of a nutritional semi-solid paste
Mixing 16g of milk powder, 10g of sodium carboxymethylcellulose, 8g of sugar, 8g of starch, 6g of activated carbon and 300ml of sterilized water for injection, and uniformly stirring to prepare black semisolid paste;
2.4 gastric emptying Rate, intestinal Pushing Rate

the calculation formula of the gastric emptying rate and the small intestine propulsion rate is as follows:
gastric emptying rate = (mass of 1- (S1-S2)/nutritional semisolid paste) × 100%
Small intestine propulsion rate = carbon tip propulsion length/total length × 100%
2.5 gastric secretion measurement
Taking out the content in the stomach, centrifuging the content and recording the total amount of the gastric juice; taking 1mL of centrifuged supernatant, adding 1 drop of phenol red indicator, carrying out neutralization titration by using 20mmol/L NaOH, recording the consumed NaOH solution amount, and finally calculating the total acidity and the total acid discharge amount;
the total acidity and total acid output are calculated as follows:
total acidity (mmol/L) = NaOH volume used for titration x 2%
Total acid output (mmol/L) = total acidity x amount of gastric acid per hour
2.6 data statistics
Statistical analysis is carried out on experimental data by adopting SPSS22.0 software, the measured data is represented by Mean value plus or minus standard deviation (Mean plus or minus SD), the difference between groups is analyzed by adopting one-factor variance, and the difference p is less than 0.05 and is significance difference;

2.1, molding: feeding common feed to the blank group, feeding self-made high-protein and high-heat feed (biscuit made by mixing dried meat floss, bean flour, flour and milk powder) to the model group rat, and freely drinking water for 7 days continuously; recording the body mass and abdominal circumference of each group of rats and the average food intake and excrement amount of each rat in the current day after the model building is successful;
2.2 grouping: after the model building is successful, the rats are randomly divided into a model group, a positive drug control group, an example 1-3 group and a comparative example 1-3 group, wherein each group comprises 10 rats; taking another 10 rats in the blank group as a blank control group;
